DEP、DOP暴露对鲤鱼的酸碱性磷酸酶的影响

摘    要:为了研究酞酸酯类对水生生物的毒性作用,将鲤鱼暴露于DEP与DOP的6个质量浓度组中进行96h-LC50急性毒性试验,测得其半致死质量浓度(96 h-LC50)分别为41.50mg/L与7.95 mg/L。分别设置3个质量浓度和1个对照组,测定DEP与DOP对鲤鱼肝脏及肌肉组织酸性磷酸酶(ACP)与碱性磷酸酶(ALP)活性的影响。结果表明:DEP与DOP对鲤鱼肝脏及肌肉组织ACP活性的影响趋势均表现为低质量浓度诱导,高质量浓度抑制;DEP对鲤鱼肝脏及肌肉组织ALP活性的影响亦表现为低质量浓度诱导,高质量浓度抑制;而DOP对鲤鱼肝脏ALP活性的影响表现为随着暴露质量浓度的增大而活性降低,呈现出明显的剂量-效应关系。酸性磷酸酶和碱性磷酸酶可能在鱼体抵御酞酸酯毒性作用中有重要作用,其机理有待进一步研究。

Abstract:The purpose of this study is to detect and confirm the effect of PAEs on enzyme activity change in the carp.As is known,Dioctyl-phthalate(DOP) and Diethyl phthalate(DEP) are two kinds of artificial plasticizing agent which are widely applied in the world for bettering the plasticity of plastics for the time being.PAEs is also known as products causing serious pollution and detriment to the living environment and eco-system.For its extremely large application,it can hardly ascertain how serious consequences it would bring to the human environment and results of human efforts to degrade its bad effects for its longtime application.This study,however,has made an attempt to evaluate the toxicities of PAEs to aquatic ecosystem.For our research purpose,we have taken as our study focus the industrial application of PAEs and define its potential impact on the environment.As the primary pet fish in general water environment,carp has been found very sensitive to the environment pollutants.In proceeding with our research,we let carps separately exposed to DEP and DOP of six different concentrations for 96 h-LC50.As a result,we found that DEP and DOP may cause distortion of the carp in bodies and make them upside-down and head-up,with obvious scoliosis.It shows that the 96 h-LC50 is 41.50 mg/L and 7.95 mg/L.And,then,we tried three different concentrations of DEP and DOP(1/10 LC50,1/5 LC50,1/2.5 LC50) with a control group(0 mg/L) confected to measure the activities of ACP and ALP in carp’s liver and muscles.The results of our experiments show that the effects of DEP and DOP on carp’s liver and muscle tissue tend to be inhibited by the high concentrations(16.60 mg/L,3.20 mg/L),whereas the low concentrations(4.15 mg/L,0.80 mg/L) were induced.The effects of DEP on the ALP activity of carp liver and muscle tissue also indicate inhibition of high concentrations(16.60 mg/L,3.20 mg/L) and induction of low concentrations(4.15 mg/L,0.80 mg/L).At the same time,the effects of DOP on the carp’s liver reveal that: as the concentration increases,the ALP activity tends to decrease,showing a clear dose-response relationship.Thus,ACP and ALP may imply a significant role in resisting the toxic effects of phthalates,though the reason for it needs further studies.
