摘    要:实验证明,亚硒酸钠对水稻外植体(成熟胚、幼穗)、愈伤组织和再生绿芽体细胞的生长都有不同程度的抑制作用,其抑制程度依次为再生绿芽>愈伤组织>外植体,但能促进它们的绿苗分化频率,特别在低硒浓度(1mg/L)处理组,并可改变它们的过氧化物酶同工酶的活性。

Abstract:The effect of Na2SeO3 on callus induction and redfifferentiation of rice somatic cells was studied. The materials used included three indica and two japonica varieties and the explants for culture included young panicles and mature embryos.The effect of selenium on the frequency of callus induction was obvious, but the treatment with Na2SeO3 could improve the callus quality,and produce bright yellow and compact calli,resulting in that 24%-69% of green plantlets were formed. The optimum concentration of Na2SeO3 was 1mg/mL in callus induction medium. The effect of Na2SeO3 on indica varieties was more obvious than that on japonica varietes.It is suggested that Na2SeO3 be worthy to try on these materials which are recalcitrant to redifferentiaton in tissue culture. Changes of peroxidase isozyme gattern of the young panicle derived calli were observed after Na2SeO3 treatment. An addition band (Rf=0.383) was observed in both indica and japonica varieties tested. The effect of Na2SeO3 on semaclonal variation will be investigated.
