自制便携式振荡装置/间接竞争酶联免疫吸附法在水库微囊藻毒素-LR快速应急监测中的应用

摘    要:应用自制便携式振荡装置,运用间接竞争酶联免疫吸附法(icELISA)建立快速测定水中微囊藻毒素(MC)-LR的现场初筛方法。结果表明:(1)自制便携式振荡装置的检测结果与手工振荡不存在差异,可用自制便携式振荡装置替代手工来进行振荡孵育操作。(2)icELISA建立了快速测定MC-LR的现场监测方法,MC-LR的标准曲线线性关系较好(r=-0.997 0),满足定性定量分析的要求,检出限为0.03μg/L,测定下限为0.12μg/L,空白溶液与实际水样加标回收率为96.2%~113.0%,方法精密度、准确度均较好。

Abstract:An icELISA method for the determination of microcystin-LR(MC-LR)in water was developed by using a self-made portable oscillator.The results showed that:(1)there was no difference between the results of selfmade portable oscillator and manual vibration.The self-made portable oscillator could be used to replace manual vibration.(2)A rapid method for on-site monitoring of MC-LR was established by icELISA.A good linear relationship(r=-0.997 0)was obtained,which met the requirements of qualitative and quantitative analysis.Detection limit and low limit of determination of MC-LR were 0.03μg/L and 0.12μg/L separately.With good precision and accuracy,the recovery of blank and actual water sample was 96.2%-113.0%.
