燃煤PM2.5不同组分对血管内皮细胞的毒性

摘    要:采用MTS法评价燃煤PM2.5不同组分对血管内皮细胞EA.hy926的细胞毒性.以大同散煤为样品煤,在实验室采用固定源稀释通道采集燃煤PM2.5,提取燃煤PM2.5全颗粒物、无机组分及有机组分,分别对人脐静脉内皮细胞EA.hy926进行染毒,采用MTS法检测PM2.5不同组分对EA.hy926细胞增殖的影响.结果表明:燃煤PM2.5全颗粒物、无机组分及有机组分均可抑制血管内皮细胞增殖,且具有剂量-反应关系;在相同染毒剂量组内,有机组分的活性抑制影响显著高于全颗粒物和无机组分,其差异具有统计学意义,但全颗粒物和无机组分之间并无统计学差异.PM2.5的来源和组分是影响其细胞毒性的重要因素,燃煤PM2.5不同组分均可抑制血管内皮细胞增殖,血管内皮损伤是PM2.5致心血管毒性的可能机制之一.

Abstract:The cytotoxicity of different compositions of PM2.5(PM2.5 suspension and its organic and inorganic extracts) on human umbilical vein endothelial cell EA.hy926 was evaluated using the MTS method.The coal sample taken from Datong coal mine,coal-fired PM2.5 was sampled by fixed source dilution channel in the laboratory.All particles,as well as isolated inorganic and organic compositions,were extracted to contaminate EA.hy926 respectively,and the MTS method was used to measure the effect of different compositions of PM2.5 on cell proliferation of EA.hy926.The results showed that different compositions from coal-fired PM2.5(all particles,inorganic and organic compositions) all decreased the viability of vascular endothelial cells in a dose-dependent manner.Compared with other preparations(all particles and inorganic composition),the organic extraction of PM2.5 had significantly higher inhibiting effect in the same contamination dose group,with statistically significant differences.However,the difference between all particles and the inorganic composition was not statistically significant.In conclusion,composition and source of PM2.5 are important factors which influence cytotoxicity.Different compositions of coal-fired PM2.5 can inhibit vascular endothelial cell proliferation,and vascular endothelial injury is one of the possible mechanisms of cardiovascular toxicity caused by PM2.5.
