The locus of symbiotic dinoflagellates within host cells provides a habitat which could potentially be exploited by the alga through heterotrophic uptake of host-derived organic substrates. Using zooxanthellae (Symbiodinium sp.) isolated from the tropical sea anemone Aiptasia pulchella collected from Kaneohe Bay, Hawaii, the effect of various potential organic substrates on growth in vitro was assessed in Erdschreiber seawater medium (ES) supplemented with organic compounds. Zooxanthellae maintained at 5 to 7 E m-2 s-1 (below compensation irradiance) grew heterotrophically when supplied with 100 M glycerol, glycolate, acetate, malate, or propionate, and grew in darkness on 100 M propionate. Zooxanthellae exposed to irradiance below compensation were able to utilize carbon sources in the unsupplemented ES medium for slow growth, but generally the growth rate of cultured zooxanthellae was a function of incubation irradiance. Zooxanthellae incubated for 10 wk in unsupplemented ES at 5 to 7 E m-2 s-1 were capable of growth at this low irradiance, but were also capable of net photosynthetic oxygen production at higher irradiances. This suggests that zooxanthellae can be photoautotrophic or facultatively heterotrophic. An estimate for the duration of mitosis (t d ) is made on the basis of growth rate of cultured zooxanthellae in log-phase; this estimate of t d =4.88 h is less than half the estimated t d for zooxanthellae in situ.  相似文献

To determine how the animal and algal components of the symbiotic sea anemone Aiptasia pulchella respond to changes in food availability and culture irradiance, sea anemones from a single clone were maintained at four irradiance levels (320, 185, 115, and 45 E m-2 s-1) and either starved or fed for 5 wk. Changes in protein biomass of sea anemones maintained under these conditions were not related to the productivity of zooxanthellae, since the protein biomass of fed A. pulchella decreased with increase in irradiance and there was no difference in protein biomass among starved sea anemones at the four irradiance levels. Except for the starved high-light sea anemones, the density of symbiotic zooxanthellae was independent of culture irradiance within both starved and fed. A. pulchella. Starved sea anemones contained over twice the density of zooxanthellae as fed sea anemones. Within both starved and fed individuals, chlorophyll per zooxanthella increased with decreasing culture irradiance while algal size remained constant (in fed sea anemones) at about 8.80 m diameter. Chlorophyll a: c 2 ratios of zooxanthellae increased with decreasing culture irradiance in zooxanthellae from starved sea anemones but remained constant in zooxanthellae from fed sea anemones. As estimated from mitotic index data, the in situ growth rates of zooxanthellae averaged 0.007 d-1 and did not vary with irradiance or feeding regime. Photosynthesis-irradiance (P-I) responses of fed A. pulchella indicated an increase in photosynthetic efficiency with decreasing culture irradiance. But there was no consistent pattern in photosynthetic capacity with culture irradiance. Respiration rates of fed sea anemones also did not vary in relation to culture irradiance. The parameter I k , defined as the irradiance at which light-saturated rates of photosynthesis are first attained, was the only parameter from the P-I curves which increased linearly with increasing culture irradiance. The daily ratio of net photosynthesis to respiration for A. pulchella ranged from 1.6 to 2.8 for sea anemones maintained at the three higher irradiances, but was negative for those maintained at 45 E m-2 s-1. Since the final protein biomass was greatest for sea anemones maintained at the lowest irradiance, these results indicate that sea anemone growth cannot be directly related to productivity of zooxanthellae in this symbiotic association.  相似文献

Growth characteristics and nutrient uptake kinetics were determined for zooxanthellae (Gymnodinium microadriaticum) in laboratory culture. The maximum specific growth rate (max) was 0.35 d-1 at 27 °C, 12 hL:12 hD cycle, 45 E m-2 s-1. Anmmonium and nitrate uptake by G. microadriaticum in distinct growth phases exhibited Michaelis-Menten kinetics. Ammonium half-saturation constants (Ks) ranged from 0.4 to 2.0 M; those for nitrate ranged from 0.5 to 0.8 M. Ammonium maximum specific uptake rates (Vmax) (0.75 to 1.74 d-1) exceeded those for nitrate (0.14 to 0.39 d-1) and were much greater than the maximum specific growth rate (0.35 d-1), suggesting that ammonium is the more significant N source for cultured zooxanthellae. Ammonium and nitrate Vmax values compare with those reported from freshly isolated zooxanthellae. Light enhanced ammonium and nitrate uptake; ammonium inhibited nitrate uptake which was not reported for freshly isolated zooxanthellae, suggesting that physiological differences exist between the two. Growth and photosynthetic properties of the marine dinoflagellate Amphidinium carterae Hulbert were examined under continuous illumination in batch cultures at four different irradiances between 2 and 150 E m-2 s-1. The slope of both cell- and Chl a-based photosynthesis versus the irradiance curves was greatest for cells grown at 15 E m-2 s-1. The relative Chl a values cell-1 were 1, 1.5 and 2 for cultures grown at 150, 80 and 15 E m-2 s-1, respectively. A low-temperature (-196°C) fluorescence technique was used to examine cells for photoinhibiton. Photoinhibition was greatest for cells grown at 150 E m-2 s-1. However, significant photoinhibition of this species was noted even at 80 E m-2 s-1. No significant difference in the fluorescence pattern was found between cells grown at 2 and 15 E m-2 s-1. Time course studies indicate that photoinhibition may occur within 2 h following exposure to 350 E m-2 s-1 in cells grown at 15 E m-2 s-1 and is reversible when light levels are lowered within 4 h. The ecological significance of phytoplankton unable to cope with excess photosynthetic excitation energy is discussed.  相似文献

The separate and combined effects of ammonium (10M) and phosphate (2M) on the ultrastructure of zooxanthellae (Symbiodinium sp.) from giant clams, Tridacna maxima, were examined in the field. Nitrogen addition significantly changed the ultrastructure of the zooxanthellae inhabiting the clams. After 9 mo exposure, the cross-sectional area of zooxanthellae from N-treated clams was significantly lower than that from other treatments [N=39.3 m2; C=47.9 m2; P=43.2m2; N+P=44.5 m2; (P=0.001)]. There was also a significant decrease in the size of starch bodies, especially around the pyrenoid of the zooxanthellae from N and N+P treatments [N=1.2 m2; C=2.0 m2; P=1.8 m2; N+P=1.2 m2; (P=2.08E-11)]. This presumably occurs as a result of the mobilization of organic carbon stores in response to stimulated amino acid synthesis under enriched nutrient conditions. These data strongly suggest that the symbiotic zooxanthellae of clams are limited to some extent by the availability of inorganic nitrogen, and that relatively minor changes to the nutrient loading of the water column can have substantial effects on the biochemistry of symbioses such as that which exists between clams and zooxanthellae.  相似文献

Six mediterranean macroalgae were cultivated for more than 2 yr under shade culture conditions, after which light requirements for growth were investigated at 16±2°C. The saturation light levels for growth in the logarithmic phase were related to the bathymetric distribution of the algae on the shore. The eulittoral to supralittoral red alga Bangia atropurpurea was saturated at a photon fluence rate of 71 mol photons m-2 s-1, the upper sublittoral to eulittoral brown algae Scytosiphon lomentaria, Colpomenia peregrina and Kuckuckia spinosa and the sublittoral brown alga Stictyosiphon soriferus at 39 to 71 mol photons m-2 s-1, and the deep-water alga Choristocarpus tenellus at 19 mol photons m-2 s-1. The minimum light requirements for growth of B. atropurpurea and C. tenellus were determined by observing length increase for 56 d under limiting light conditions. The compensation and minimum irradiances required for growth of B. atropurpurea were 0.5 and 1 mol photons m-2 s-1 respectively. The corresponding values for C. tenellus were 0.15 to 0.28 and 0.5 mol photons m-2 s-1 respectively. C. tenellus was the siowest-growing species tested at saturating light conditions, but it grew faster than B. atropurpurea at 1 mol photons m-2 s-1. Both B. atropurpurea and C. tenellus were able to survive 56 d in darkness, but only the latter grew under darkness in the first 14 d.  相似文献

The substrate analogue [14C]-methylammonium was used to study ammonium/methylammonium uptake by Symbiodinium microadriaticum (zooxanthellae). The value of the Michaelis constant (K m) for the uptake system was approximately 35 M with methylammonium as substrate; ammonium was a competitive inhibitor of methylammonium uptake, and the K m for ammonium uptake (determined as the inhibition constant, K i, for methylammonium) was 6.6 M. Methylammonium uptake by zooxanthellae was light-dependent. Methylammonium uptake rates of zooxanthellae which had been freshly isolated from the hermatypic coral Acropora formosa (0.85±0.05x10-10 mol min-1 cell-1) were lower than those of axenic cultures of the zooxanthellae from Montipora verrucosa (Acroporidae) grown under various nitrogen regimes (1.6 to 12x10-10 mol min-1 cell-1). Maximum uptake rates were found for ammonium-starved cultured M. verrucosa zooxanthellae (10.2 to 12x10-10 mol min-1 cell-1); M. verrucosa zooxanthellae growing with ammonium as nitrogen source and zooxanthellae which had been freshly isolated from A. formosa gave similar and considerably lower uptake rates (0.85 to 1.6x10-1 mol min-1 cell-1). These results suggest that either coral tissue contains sufficient ammonium to repress synthesis of the uptake system of the algal symbionts or, alternatively, there are additional barriers to ammonium transport for zooxanthellae in vivo.  相似文献

The shortterm (10–22 d) effect of Zn, Hg, Cu, Cd, Pb, and Ni on the length growth of Mytilus edulis is studied. Significant reductions of growth rate was found at 0.3 g Hgl-1, 3 g Cul-1, 10 g Znl-1, and 10 g Cdl-1 added to the local sea water, while concentrations of up to 200 gl-1 of Pb and Ni had no effect on the growth. With exposure to Cu and Zn, there was a linear reduction in growth rate with increasing metal concentration up to about 6 g Cul-1 and 100 g Znl-1. Above these levels, growth stopped with Cu, while with Zn it was stabilized at about 20% of control growth. When Hg and Cd were added, a curvilinear relationship between growth and metal concentration is indicated. With Hg, growth rate is nearly zero above 3–4 g Hgl-1, while the growth rate was 50% of control after 10 d of exposure to 100 g Cdl-1. At 2 g Cdl-1 there was a significant stimulation of length increase. Observed EC50-values for growth were 0.3–0.4 g Hgl-1, 3–4 g Cul-1, 60 g Znl-1, and 100 g Cdl-1.  相似文献

Monthly variation in photosynthesis, dark respiration, chlorophyll a content and carbon: nitrogen (C:N) ratios in different lamina sections of adult plants of Ascoseira mirabilis Skottsberg from King George Island, Antarctica, was investigated between September 1993 and February 1994. Light saturated net photosynthesis (P max) showed maximum values in September (12 to 25 mol O2 g-1 fr wt h-1), and decreased towards the summer to values ranging between 2.0 and 5.0 mol O2 g-1. In the distal section, however, a second optimum occurred in December (25 mol O2 g-1 fr wt h-1). Dark respiration rates were also highest in October and November and decreased strongly in December to February (6.0 and 1.0 mol O2 g-1 fr wt h-1, respectively). Gross photosynthesis exhibited high values between September and December. Concomitant with the seasonal decrease of photosynthetic efficiency () from mean values of 1.2 mol O2 g-1 fr wt h-1 (mol photons cm-2 s-1)-1 in September to 0.3 mol O2 g-1 fr wt h-1 (mol photons cm-2 s-1)-1 in January, the initial light saturating point (I k) gradually increased from 19 to 60 mol photons m-2 s-1. Likewise C:N ratios were low in spring (12 to 13) and increased in summer (20). In general, the photosynthetic parameters P max, gross photosynthesis, and Chl a concentrations were significantly higher in the distal section of the thallus. In contrast, C:N ratios were lower in the distal section of the lamina. The results show that photosynthesis obviously strongly supports growth of the alga in late winter to spring, as it does in some morphologically related brown algae from temperate and polar regions. Batch cultures of the marine unicellular centric diatom Chaetoceros calcitrans (Paulsen) Takano were maintained by serial subculturing every 4 d into nutrient-enriched natural sea-water medium supplemented with 350, 950 and 1 400 g-at Si l-1. The diatom cultures removed initial silica concentrations of 350 and 950 g-at l-1 from the medium within 2 and 3 d, respectively. About 30 g-at l-1 of the highest initial concentration remained in the medium after 4 d. The mean final cell density with an enrichment of 350 g-at Si l-1 was 3.43±0.26×104 cells l-1 (median cell volume = 77.5±5.0 m3); with 950 g-at Si l-1, 8.55±0.55×104 cells l-1 (50.0±4.5 m3); and with 1 400 g-at Si l-1, 9.72±0.48×104 cells l-1 (37.3±5.0 m3). There was no significant difference in the final total organic weight of cells produced, which was in the range of 170 to 190 mg per 250 ml culture. This consisted of proportionately more lipid and carbohydrate and less protein from the treatment with 350 g-at Si l-1 than from the 1 400 g-at Si l-1 treatment.  相似文献

Specimens of the oceanic decapod Systellaspis debilis were collected from six sites in the East Atlantic Ocean between 1970 and 1984, and were analysed for Mn, Fe, Cu, Zn and Cd. The data confirm that there are small but significant differences in mean metal concentrations from some sites which showed no obvious pattern in relation to geographic location of the samples. As a result, ranges of site means are quoted as baseline levels for each metal (g g-1 dry wt): 2.3 to 2.9 g Mn g-1, 31.2 to 77.8 g Fe g-1, 25.9 to 83.4 g Cu g-1, 41.9 to 92.9 gZn g-1, 11.1 to 31.8 g Cd g-1. The concentration of cadmium in S. debilis from all sites was raised relative to cadmium concentrations reported for coastal decapods, perhaps as a result of dietary enrichment. Metal accumulation may provide useful information for understanding the complex feeding behaviour of many oceanic animals.  相似文献

The reef coral Pocillopora damicornis (Linnaeus) was grown for 8 wk in four nutrient treatments: control, consisting of ambient, unfiltered Kaneohe Bay seawater [dissolved inorganic nitrogen (DIN, 1.0 M) and dissolved inorganic phosphate (DIP, 0.3 M)]; nitrogen enrichment (15 M DIN as ammonium); phosphorus enrichment (1.2 M DIP as inorganic phosphate); and 15 M DIN+1.2 M DIP. Analyses of zooxanthellae for C, N, P and chlorophyll a after the 8 wk experiment indicated that DIN enrichment increased the cellular chlorophyll a and excess nitrogen fraction of the algae, but did not affect C cell-1. DIP enrichment decreased both C and P cell-1, but the decrease was proportionally less for C cell-1. the response of cellular P to both DIN and DIP enrichment appeared to be in the same direction and could not be explained as a primary effect of external nutrient enrichment. The observed response of cellular P might be a consequence of in situ CO2 limitation. DIN enrichment could increase the CO2 (aq) demand by increasing the net production per unit area. Mussels, Mytilus edulis planulatus, were collected from Port Phillip Bay, Victoria, Australia, in February 1984, to test the assumption that they are integrators of cadmium pollution. Groups of mussels were subjected to the same average dose of cadmium (21 g l-1), administered according to different dosing regimes over 4 wk (Regimes 1, 2, 3, 5); other groups received twice the average dose (42 g l-1) in half the time (Regime 4). During each regime, the mussels were exposed to the different cadmium concentrations for one week at each concentration. Nominally, the regimes were: (1) 36 g (Wk 1) to 26 g (Wk 2) to 16 g (Wk 3) to 6 g (Wk 4) Cd l-1; (2) 6 to 16 to 26 to 36 g Cd l-1; (3) 21 to 21 to 21 to 21 g Cd l-1; (4) 42 to 42 g Cd l-1; (5) 42 to 42 g Cd l-1 to background (<0.5 g) to background. Differences in cadmium accumulation by mussels from Regimes 1 and 4 were not statistically significant, nor were differences in accumulation between mussels from Regimes 2 and 3. However, mussels from Regimes 1 and 4 had accumulated significantly more cadmium than had mussels from Regimes 2 and 3. Accumulation by mussels from Regime 5 was not significantly different from that by mussels from any of the other regimes. These results suggest that, at least for cadmium, the assumption that mussels are integrators of pollution should be treated with caution. They also have implications with regard to the quantitative biological monitoring of pollution. For example, even in a carefully controlled monitoring program, using mussels of standard size and condition, significant differences in cadmium content between mussels need not indicate exposure to different levels of contamination. Rather, these differences could reflect differences in the regime by which the contamination was received.  相似文献

In order to assess the intake of lead and cadmium by consumers of home grown vegetables in urban areas, replicated experimental plots of uniform size, comprising summer and winter crops, were established in 94 gardens and allotments in nine towns and cities in England.The geometric mean lead and cadmium concentrations for the soils (n = 94) were 217 g g–1 (ranging from 27 to 1,676 g g–1) and 0.53 g g–1 (<0.2–5.9 g g–1), respectively. Compared with agricultural soils, the garden and allotment soils contained elevated levels of lead but not cadmium.Lead concentrations in the vegetables ranged from <0.25 g g–1 to 16.7 g g–1 dry weight and cadmium concentrations ranged from <0.025 g g–1 to 10.4 g g–1 dry weight. Lead concentrations were higher than reported background levels, although <1% exceeded the statutory limit for saleable food in the UK (1 g g–1 fresh weight). The morphological and reproductive development of gametophytes of Ecklonia maxima (Osbeck) Papenf. has been studied in vitro under a wide range of light and temperature conditions, and the results related to the natural environment over the range of the distribution of the species. Initial vegetative growth was light saturated at 20 E m-2 s-1, and maximal at 17.5° and 20°C. Most rapid egg production corresponded to low cell number of female gametophytes, and this process was light saturated at 60 E m-2 s-1, and optimised at 15° and 17.5°C. Cell number was also low at lower temperatures, with reduced fertility rates. Sub-saturating irradiances and supraoptimal temperatures caused females to become filamentous, producing many more cells, and reducing reproductive rates. The final egg production per female was, however, greater in these sub-optimal conditions, and this phenomenon is interpreted as an ecological adaptation which improves survival prospects in conditions prevailing at the fringes of the depth and geographical distribution of the species. The optimal, and maximum (22.5°C) temperatures for reproduction are far higher than those of northern hemisphere Laminarians. E. maxima is very much a warm temperate organism, and this could have implications for marine phytogeographical studies on the west coast of southern Africa.  相似文献

Michaelis-Menten uptake kinetics were observed at all light intensities. With constant illumination, the Vmax and K1 in nitrate uptake over the natural light intensity range of 0 to 2000 E were 0.343 g-at NO3–N(g)-1 at protein-N h-1 and 26 E, respectively. Nitrate uptake was inhibited at higher light intensities. The Ks for nitrate uptake did not vary as a function of light intensity remaining relatively constant at 0.62 g-at NO3–N 1-1. With intermittent illumination, the Vmzx for light intensity in nitrate uptake over a light intensity range of 0 to 5000 E was 0.341 g-at NO3–N(g)-1-at protein-N h-1. No inhibition of nitrate uptake was observed at higher than natural light intensities. Chaetoceros curvisetus will probably never experience light inhibition of nitrate uptake under natural conditions.  相似文献

The distribution of cyanobacteria in the surface waters of the North Sea was measured during July 1987. Numbers of cyanobacteria ranged from 2.5x106 to 1.7x108 cells 1-1. In the majority of stations, cyanobacterial numbers were highest in the near-surface water and a subsurface maximum was found at only one station. The distribution of 14C among the end-products of photosynthesis was determined for picoplankton (<1 m) and other phytoplankton >1 m throughout the North Sea. The majority of label was found in the protein fraction of both picoplankton and >1 m phytoplankton; incorporation into lipids and polysaccharides plus nucleic acids was much lower. We interpret the large incorporation into protein to be a consequence of nutrient limitation of these natural assemblages. Photosynthetic parameters of the two size fractions were also determined. Assimilation number (P m B ) and initial slope were greater for the picoplankton fraction than for phytoplankton >1 m but there was no evidence of significant photoinhibition of either fraction at irradiances up to 1 000 E m-2 s-1.  相似文献

The possible modification of mercury toxicity by selenium in embryos of the Pacific oyster Crassostrea gigas and the larvae of the crab Cancer magister was investigated. Mercury concentration eliciting abnormal development in 50% of the oyster embryos (EC50) was 5.7 g l-1 (48 h) and mortality in 50% of the crab larvae (LC50) occurred with 6.6 g l-1 (96 h). The 48 h EC50 for selenium was greater than 10,000 g l-1 for oyster embryos and the 96 h LC50 for crab zoeae was 1040 g l-1. The response from each species, when exposed to both toxicants, revealed, that a high level of selenium (5,000 g l-1) increased mercury toxicity. Moderate selenium concentrations (10 to 1,000 g l-1) tended to decrease mercury toxicity, although no statistical verification could be made. The order of administration of toxicants had no effect on the response of Crassostrea gigas embryos. Early developmental stages (8 h) of C. gigas embryos were most sensitive to dissolved Hg; toxicant administration 24 h after fertilization resulted in no apparent abnormalities in development.  相似文献

Analyses of individual content of carbon (C), nitrogen (N), and hydrogen (H) were carried out for all larval stages of Pagurus bernhardus and Carcinus maenas, and for newly metamorphosed crabs. Maximum range in total larval development is 12.8 to 165.8 g C, 3.2 to 35.1 g N, and 1.9 to 24.9 g H in P. bernhardus and 3.1 to 43.2 g C, 0.7 to 10.1 g N, and 0.4 to 6.3 g H in C. maenas. From these data energy equivalents were calculated. Maximum range in total larval life is 0.43 to 6.38 J ind. -1 in P. bernhardus and 0.1 to 1.49 J ind. -1 in C. maenas. There is a 32.4% mean loss of energy in P. bernhardus megalopa development; this seems to describe the normal developmental pattern in this stage. Biomass was determined as fresh and dry weight respectively. Photosynthetic performance in the kelp Laminaria solidungula J. Agardh was examined from photosynthesis irradiance (P-I) parameters calculated from in situ 14C uptake experiments, using whole plants in the Stefansson Sound Boulder Patch, Alaskan Beaufort Sea, in August 1986. Rates of carbon fixation were determined from meristematic, basal blade, and second blade tissue in young and adult sporophytes. Differences in saturating irradiance (I k, measured as photosynthetically active radiation, PAR), photosynthetic capacity (P max), and relative quantum efficiency () were observed both between young and adult plants and between different tissue types. I k was lowest in meristematic tissue (20 to 30 E m–2 s–1) for both young and adult plants, but consistently 8 to 10 E m–2 s–1 higher in young plants compared to adults in all three tissues. Average I k for non-meristematic tissue in adult plants was 38 E m–2 s–1. Under saturating irradiances, young and adult plants exhibited similar rates of carbon fixation on an area basis, but under light limitation, fixation rates were highest in adult plants for all tissues. P max was generally highest in the basal blade and lowest in meristematic tissue. Photosynthetic efficiency () ranged between 0.016 and 0.027 mol C cm–2 h–1/E m–2 s–1, and was highest in meristematic tissue. The relatively lower I k and higher exhibited by L. solidungula in comparison to other kelp species are distinct adaptations to the near absence of light during the eight-month ice-covered period and in summer when water turbidity is high.
