A physiologically based pharmacokinetic and pharmacodynamic (PBPK/PD) models were developed for arsenic (As) in tilapia Oreochromis mossambicus from blackfoot disease area in Taiwan. The PBPK/PD model structure consisted of muscle, gill, gut wall, alimentary canal, and liver, which were interconnected by blood circulation. We integrate the target organ concentrations and dynamic response describing uptake, metabolism, and disposition of As and the associated area-under-curve (AUC)-based toxicological dynamics following an acute exposure. The model validations were compared against the field observations from real tilapia farms and previously published uptake/depuration experimental data, indicating that predicted and measured As concentrations in major organs of tilapia were in good agreement. The model was utilized to reasonably simulate and construct a dose-dependent dynamic response between mortality effect and equilibrium target organ concentrations. Model simulations suggest that tilapia gills may serve as a surrogate sensitive biomarker of short-term exposure to As. This integrated As PBPK/PD/AUC model quantitatively estimates target organ concentration and dynamic response in tilapia and is a strong framework for future waterborne metal model development and for refining a biologically-based risk assessment for exposure of aquatic species to waterborne metals under a variety of scenarios.  相似文献

We studied the bioaccumulation of lead in selected tissues of Prussian carp Carassius gibelio (Bloch, 1782) during 12 and 24 months exposure to different doses of this metal in feed and the elimination of lead from tissues during the following 12-month depuration period. Lead concentration was determined using atomic absorption spectrometry method. The highest lead concentrations were observed at 2.0?±?0.54 to 7.4?±?1.1 mg?kg?1 in the kidney, 3.0?±?0.13 to 5.2?±?0.17 mg?kg?1 in the bone, and 4.5 (±0.4)?mg?kg?1 in the hepatopancreas of fish from groups exposed to lead dietary concentration from 8 to 49 mg?kg?1 for 24 months. The rate of accumulation were generally the highest at the beginning of exposure as evidenced by the highest monthly increments of bioaccumulation observed after 3 months of contamination for muscles, hepatopancreatic gland, intestine, and gills. Also analysis of the monthly increments of lead bioaccumulation in bone tissue and the highly significant coefficients of correlation indicate that the dynamics of accumulation are clearly dependent on dose of exposure. Depuration of accumulated lead from the organs depended mainly on tissue and duration of elimination period. Very rapid depuration was observed in soft tissues such as the intestine or muscles. Very low elimination was observed for scales and bones where until the end of the experiment highly significant lead concentration differences were observed in all groups in relation to the control group. Chronic dietary exposure in the range of 8–49 mg Pb?kg?1 resulted in no significant effects on the growth and survival of Prussian carp females.  相似文献

A laboratory study of the cadmium and mercury accumulation and elimination kinetics was conducted on the pelecypod Elliptio complanata (Lightfoot) and the short-lived gastropod Viviparus georgianus (Lea), according to age-classes. Preliminary results (metal concentration vs time of exposure) have demonstrated that uptake of Cd and Hg, in the two molluscs studied, follow a biphasic pattern, whereby a steady state is reached after approximately 16 days' exposure and then accumulation increases again for the rest of exposure period. The elimination of the two metals is also characterized in a biphasic way: fast excretion for the first four days followed by a slower depuration for the rest of exposure time. A two-compartment bioaccumulation model has been used to described the different kinetic parameters: (1) the rate constant for depuration; (2) the rate constant for uptake; (3) the theoretical bioconcentration factor extrapolated to steady-state conditions; and (4) the biological half-life of the metals.  相似文献

Abstract

The use of sewage sludge and effluent as a source of nutrients and water for crop production is increasing worldwide. A study was conducted in 2001 at Pension farm (near Harare) to determine the effect of long term (>30 yrs) application of sewage sludge and effluent on Zn and Cu accumulation in top soil, uptake of these metals by lettuce (Lactuca sativa L.) and mustard rape (Brassica juncea L.), and dry matter yield. Application of sewage sludge/effluent significantly (p < 0.001) increased total Zn (13.7–1563.9 mg kg?1) and Cu (2.5–133.3 mg kg?1) in the top soil (0–20 cm depth) compared to the control. Sewage sludge/effluent addition significantly (p < 0.001) increased Zn uptake by both test crops, while Cu uptake was significant in the first crop of lettuce and the second crop of mustard rape. Based on the dietary patterns of poor urban households in Zimbabwe, the maximum possible intake of Cu will only constitute 40% the Maximum Daily Intake (MDI). The toxicological implications for Zn will however be more severe, exceeding the MDI by 77% through exposure by lettuce consumption and by 251% consumption of mustard rape. It was concluded that long-term addition of sewage sludge/effluent to soil at Pension farm had increased the concentration of Zn and Cu in top soil to levels that pose environmental concern. The consumption of leafy vegetables produced on these soils pose a health risk to poor communities that reside around the study site, especially children, through possible Zn toxicity.  相似文献

The first objective of this study was to determine the bioaccumulation kinetics of pyrene in the soft tissues of Crassostrea gigas (mantle, muscle, gills, digestive gland, and the remaining soft tissues). As bivalves can biotransform hydrocarbons in more polar compounds (metabolites) that are more easily excreted, the second objective was to investigate the oyster capacity to metabolize pyrene into its metabolite, the 1-hydroxypyrene. To these ends, oysters were exposed 24 h to waterborne 14C-pyrene then placed in depuration conditions for 15 d. Oysters efficiently bioaccumulated pyrene in their soft tissues and equilibrium was reached within the exposure time. The metabolite1-hydroxypyrene was also detected in oyster tissues but represented only 4-14% of the parent pyrene. At the end of the exposure period, the gills and the mantle showed the highest pyrene proportion of total soft tissue content, i.e. 47% and 26%, respectively. After 15 d of depuration, the mantle contained 32% and 30% of the remaining pyrene and 1-hydroxypyrene, respectively. As C. gigas did not display a high capacity for metabolizing pyrene, it can be considered as a good bioindicator species to survey and monitor pyrene contamination in the coastal marine environment.  相似文献

Metal pollution e.g. copper, in water bodies occurs worldwide. Although copper is an essential trace metal, at certain levels it is still considered as pollutant. The aim of this study was to investigate the effect of exposure concentration on copper bioaccumulation in marbled crayfish (Procambarus sp.) by determining uptake and elimination kinetics. Crayfish were exposed to sub-lethal copper concentrations (average measured concentrations of 0.031 and 0.38 mg Cu L−1) for 14 d and transferred to copper-free water for another 14 d. At different time points during the uptake and elimination phases copper concentrations were measured in five organs (exoskeleton, gills, muscle, ovaries and hepatopancreas). At 0.031 mg Cu L−1, copper levels in the crayfish organs were not significantly increased compared to the control animals, suggesting effective regulation. Exposure to 0.38 mg Cu L−1 did lead to not significantly increased copper levels in muscles and ovaries, while the gills and exoskeleton, which are in direct contact with the water, showed significantly higher copper concentrations. In these four organs, copper showed fast uptake kinetics with equilibrium reached within 10 d of exposure. Copper accumulation was highest in the hepatopancreas; uptake in this storage organ steadily increased with time and did not reach equilibrium within the 14-d exposure period. Copper accumulation levels in the marbled crayfish found in this study were hepatopancreas > gills > exoskeleton > muscle.  相似文献

In the current study, the bioaccumulation of essential and nonessential metals and related antioxidant activity were analyzed in three organs (muscle, gills, and liver) of herbivorous (HF) and carnivorous (CF) edible fish of Chenab River. The comparative analysis revealed a more heterogeneous accumulation of metals in the muscles of HF fish than that of CF fish [chromium (Cr, 3.4 μg g?1), cobalt (Co, 1.7 μg g?1), copper (Cu, 3 μg g?1), and iron (Fe, 45 μg g?1) versus Cr (1.3 μg g?1), Co (0.1 μg g?1), Cu (1.1 μg g?1), and Fe (33 μg g?1), respectively, P?<?0.001]. These results implied an organ-specific accumulation of metals at different trophic levels. According to logistic regression analysis, the bioaccumulation of metals had marked differences in HF and CF. The antioxidant activity was significantly related to the tissue type and the metals to which the organs are exposed to. The liver of CF fish had a higher activity of antioxidant superoxide dismutase (SOD), catalase (CAT), glutathione (GSH), and lipid peroxidase (LPO) than that of HF (P?<?0.05). LPO and guaiacol peroxidase (POD) in both groups were associated with a number of metals, but in HF, cadmium (Cd), Cr, Pb, and Zn were more related with the LPO and SOD activities. Moreover, Cd, Co, Fe, Pb, Ni, Cu, and Zn were above the permissible limits set by various agencies. In numerous cases, our results were even higher than those previously reported in the literature. The results provide an insight into the pollution pattern of Chenab River. These results may be helpful in the future to identify biomarkers of exposure in aquatic organisms.

Copepods have been widely used to evaluate toxicity of metals present in marine environments. However, a technical difficulty is to understand the possible routes of metal uptake and to identify in which tissues or organs metals are being accumulated. Traditional techniques are hard to be employed once each organ has to be analyzed separately. Autoradiography is an alternative technique to circumvent this limitation, since metal distribution in tissues can be visualized and quantified, even in small organisms like copepods. In the present study, accumulation and distribution of 64Cu in the copepod Calanus hyperboreus was studied using autoradiography. Copepods were exposed for 2 h to copper (2.3 mg L−1; 1.08 MBq 64Cu mg−1 Cu) and then allowed to depurate for 2 h in clean seawater. Total 64Cu was determined by gamma-spectrometry after a metal exposure and a depuration period. 64Cu distribution was determined based on images generated by autoradiography. Metal accumulation was observed on all external surfaces of the copepods, being accumulated mostly on the ventral region, followed by dorsal, urossoma and internal regions. After depuration, radioactivity levels had a decrease in the sum of external body surface. Our results show that copper uptake by C. hyperboreus is fast and that a non-negligible proportion of the accumulated metal can reach internal tissues, which may lead to detrimental physiological effects. Moreover, whole-body autoradiography was demonstrated to be an efficient technique to study copper accumulation and body distribution in a very small organism such as the copepod C. hyperboreus.  相似文献

The purpose of this study was to link toxicokinetics/toxicodynamics (TK/TD) and bioavailability-based metal uptake kinetics to assess arsenic (As) uptake and bioaccumulation in three common farmed species of tilapia (Oreochromis mossambicus), milkfish (Chanos chanos), and freshwater clam (Corbicula fluminea). We developed a mechanistic framework by linking damage assessment model (DAM) and bioavailability-based Michaelis?CMenten model for describing TK/TD and As uptake mechanisms. The proposed model was verified with published acute toxicity data. The estimated TK/TD parameters were used to simulate the relationship between bioavailable As uptake and susceptibility probability. The As toxicity was also evaluated based on a constructed elimination?Crecovery scheme. Absorption rate constants were estimated to be 0.025, 0.016, and 0.175?mL g?1 h?1 and As uptake rate constant estimates were 22.875, 63.125, and 788.318?ng g?1 h?1 for tilapia, milkfish, and freshwater clam, respectively. Here we showed that a potential trade-off between capacities of As elimination and damage recovery was found among three farmed species. Moreover, the susceptibility probability can also be estimated by the elimination?Crecovery relations. This study suggested that bioavailability-based uptake kinetics and TK/TD-based DAM could be integrated for assessing metal uptake and toxicity in aquatic organisms. This study is useful to quantitatively assess the complex environmental behavior of metal uptake and implicate to risk assessment of metals in aquaculture systems.  相似文献

Experiments were carried out to investigate the accumulation and elimination of cadmium (Cd) in tissues (gill, intestine, kidney, liver and muscle) of juvenile olive flounder, Paralichthys olivaceus, exposed to sub-chronic concentrations (0, 10, 50, 100 microg l(-1)) of Cd. Cd exposure resulted in an increased Cd accumulation in tissues of flounder with exposure periods and concentration, and Cd accumulation in gill and liver increased linearly with the exposure time. At 20 days of Cd exposure, the order of Cd accumulation in organs was gill > intestine > liver > kidney > muscle and after 30 days of exposure, those were intestine > gill > liver > kidney > muscle. An inverse relationship was observed between the accumulation factor (AF) and the exposure level, but AF showed an increase with exposure time. During the depuration periods, Cd concentration in the gill, intestine and liver decreased immediately following the end of the exposure periods. No significant difference was found Cd in concentration in the kidney and muscle during depuration periods. The order of Cd elimination rate in organs were decreased intestine > liver > gill during depuration periods.  相似文献

Understanding on the bioaccumulation and depuration of PAHs (polycyclic aromatic hydrocarbons) in Penaeus monodon is important in seafood safety because it is one of the most popular seafood consumed worldwide. In this study, we used anthracene as the precursor compound for PAHs accumulation and depuration in the shrimp. Commercial feed pellets spiked with anthracene were fed to P. monodon. At 20 mg kg−1 anthracene, P. monodon accumulated 0.1% of the anthracene from the feed. P. monodon deputed the PAH two times faster than its accumulation. The shrimp reduced its feed consumption when anthracene content in the feed exceeded 20 mg kg−1. At 100 mg kg−1 anthracene, P. monodon started to have necrosis tissues on the posterior end of their thorax. The bioaccumulation factor (BAF), uptake rate constant (k1) and depuration rate constant (k2) of anthracene in P. monodon were 1.15 × 10−3, 6.80 × 10−4 d−1 and 6.28 × 10−1 d−1, respectively. The depuration rate constant is about thousand times higher than the uptake rate constant and this indicated that this crustacean is efficient in depurating hydrocarbons from their tissue.  相似文献

Compared to traditional chemical or physical treatments, phytoremediation has proved to be a cost-effective and environmentally sound alternative for remediation of contaminated dredged sediment. A field study was conducted in a sediment disposal site predominantly colonized by Typha angustifolia under different sediment moisture conditions to estimate the phytoremediation effects of dredged sediment. The moisture content was 37.30 % and 48.27 % in aerated and waterlogged sediment, respectively. Total nitrogen (TN) content was higher in the waterlogged sediment than in the aerated sediment. The total Cd contents were lower in aerated sediment, which was mainly resulted from the lower exchangeable fraction of Cd. The bioaccumulation of P, Cu and Pb in T. angustifolia was promoted by waterlogging, and the belowground tissue concentrations and accumulation factors (AFs) of Cu were higher than that of other metals, which can be explained by that Cu is an essential micronutrient for plants. Consistent with many previous studies, T. angustifolia showed higher metal levels in roots than in above-ground tissues at both the sediment conditions. Due to the improved biomass produced in the aerated sediment, the removals of nutrients and the metals by plant harvest were higher from aerated sediment than from waterlogged sediment. It was indicated that maintaining the dredged sediment aerated can avoid release risk and plant uptake of metals, while the opposite management option can promote phytoextraction of these contaminants.  相似文献

Tissue residues of the PAHs, anthracene, fluorene, fluoranthene and pyrene were determined in Lumbriculus variegatus at four time intervals during both a 96-h exposure period to monitor uptake, and a 96-h clean water period to assess depuration. Mean BCFs were 2390, 1210, 452 and 1920 for fluoranthene, anthracene, fluorene and pyrene, respectively. BCFs were positively correlated with the octanol/water partition coefficient values of the four PAHs. Depuration occurred most rapidly for fluorene, followed by anthracene and fluoranthene. No apparent depuration of pyrene was observed during the 96-h depuration period. Because of rapid depuration of some PAHs, it appears that the 24 h clean water gut clearance period recommended in conjunction with sediment bioaccumulation tests with L. variegatus has potential to result in an under-estimation of bioaccumulation of some chemicals.  相似文献

Hypoxic events frequently occur in the aquatic environment in association with micro pollutants, including heavy metals. Only a few studies are however available on the uptake and biological responses of heavy metals under hypoxic conditions. To elucidate the phenomenon, mirror carp Cyprinus carpio L. (16.13-16.22 g) were exposed chronically to dietary copper (Cu; 250 and 500 mg kg dry wt.−1) for 30 d under normoxic (8.25 mg O2 L−1) and hypoxic (∼3 mg O2 L−1) conditions and adopting an integrated approach, sub-lethal biomarker responses were determined at different levels of biological organisation. Level of oxidative DNA damage (as determined by modified Comet assay) showed strong significant difference following exposure to dietary Cu level under normoxic (1.6-fold) as well as under hypoxic condition at both Cu levels (2.1 and 2.5-folds respectively). Significant difference was also observed for haematological parameters (i.e. increased red and white blood cells, haematocrit value and haemoglobin concentration). Quantitative histology revealed alterations in tissues (i.e. liver and gills) for hypoxic and all dietary Cu treatment groups under both normoxic and hypoxic conditions suggesting a compensatory response to these organs (< 0.05). The order of Cu accumulation in tissues (as determined by ICP-OES) was liver > intestine > kidney > gill. Interestingly, SGR under both normoxic and hypoxic conditions reduced with elevating Cu levels (p = 0.019). Overall, the results provide evidence for enhanced toxicological responses in fish following exposure to Cu either alone or in combination with hypoxic condition and lends support to the evolving viewpoint that many water quality guidelines should be revisited in terms of new ecotoxicological criteria.  相似文献

To examine the Cd, Hg, Ag, and Zn accumulation in the green mussel Perna viridis affected by previous exposure to Cu, Ag, or Zn, the dietary metal assimilation efficiency (AE) and the uptake rate from the dissolved phase were quantified. The mussel's filtration rate, metallothionein (MT) concentration, and metal tissue burden as well as the metal subcellular partitioning were also determined to illustrate the potential mechanisms underlying the influences caused by one metal pre-exposure on the bioaccumulation of the other metals. The green mussels were pre-exposed to Cu, Ag, or Zn for different periods (1-5 weeks) and the bioaccumulation of Cd, Hg, Ag, and Zn were concurrently determined. Pre-exposure to the three metals did not result in any significant increase in MT concentration in the green mussels. Ag concentration in the insoluble fraction increased with increasing Ag exposure period and Ag ambient concentration. Our data indicated that Cd assimilation were not influenced by the mussel's pre-exposure to the three metals (Cu, Ag, and Zn), but its dissolved uptake was depressed by Ag and Zn exposure. Although Hg assimilation from food was not affected by the metal pre-exposure, its influx rate from solution was generally inhibited by the exposure to Cu, Ag, and Zn. Ag bioaccumulation was affected the most obviously, in which its AE increased with increasing Ag tissue concentration, and its dissolved uptake decreased with increasing tissue concentrations of Ag and Cu. As an essential metal, Zn bioaccumulation remained relatively stable following the metal pre-exposure, suggesting the regulatory ability of Zn uptake in the mussels. Zn AE was not affected by metal pre-exposure, but its dissolved uptake was depressed by Ag and Zn pre-exposure. All these results indicated that the influences of one metal pre-exposure on the bioaccumulation of other metals were metal-specific due to the differential binding and toxicity of metals to the mussels. Such factors should be considered in using metal concentrations in mussel's soft tissues to evaluate the metal pollution in coastal waters.  相似文献

Mercury bioaccumulation and decontamination kinetics in the edible cockle Cerastoderma edule were studied through a mesocosms experiment after a medium-term exposure to the metal.The results revealed that the bivalve presented distinct bioaccumulation kinetics according to the different tissues. While the gills showed a linear accumulation pattern, the digestive gland and the entire organism presented a saturation model, with higher accumulation during the first 7d of exposure and lower during the rest of the time. In addition, the bioaccumulation rate was not proportional to the Hg concentration, since the organisms under lower contamination presented higher bioconcentration factors than the ones under higher contamination. Gills were the tissues with higher mercury accumulation capability.Concerning the decontamination phase, C. edule lost approximately 80% of the mercury after 24 h exposure in clean seawater. Nevertheless, never reached the original condition, showing in the final (20 d detox), Hg levels (>0.5 ppm) higher than those allowed by the legislation regulating human food consumption. Prolonged exposure to inorganic arsenic has been a severe environmental public health issue worldwide in the recent decades. Increasing evidence has suggested a possible role of prolonged arsenic exposure through drinking water in the development of arsenic-induced chronic noncancer diseases, among which hypertension and type 2 diabetes (T2D) are the focus of concern. Although exposure to high levels of arsenic has been reported to be associated with excess risk of hypertension or T2D in a dose-dependent manner, the association has yet to be established, especially low-level exposure. This cross-sectional study was designed to evaluate the potential association between prolonged environmental arsenic exposure through drinking water and the prevalence of hypertension and T2D in Inner Mongolia, China, with emphasis on the assessment of low-level exposure. In this study (a total of 669 men and women), we found that the blood pressure levels were significantly correlated with cumulative arsenic exposure and that the systolic blood pressure of the subjects with arsenic exposure >50 μg/L was significantly higher than those of the subjects with <10 and 10–50 μg/L exposure. Significant prevalence of hypertension was found in the subjects of the >50 μg/L group both before and after adjustment for confounders. In addition, a significant negative relationship was found between urinary arsenic percentage of dimethylated arsenic (DMA%) and the prevalence of hypertension in the >50 μg/L group. However, low-level arsenic exposure (10–50 μg/L) was not statistically associated with hypertension. No significant difference of blood glucose was found among the groups with different arsenic exposure levels. No statistical association was found between arsenic exposure and T2D. Our findings suggested that prolonged arsenic exposure might play a role in the development of hypertension; however, only high-level arsenic was associated with the risk of hypertension. Our findings also indicated that lower DMA% might be related with the increased susceptibility of arsenic-induced hypertension.  相似文献
