从开发天然功能保健食(药)品的角度,总结了分布于我国南方各省的资源植物余甘子(滇橄榄,Phyllanthus emblica)的生物学、植物地理学、传统草药学、药理学、植物化学等的研究与开发进展,包括作者实验室近年开展的有关余甘子果汁澄清和果酒"去沉淀"的微生物酶法加工技术部分内容,以及引人关注的余甘子果植物多酚提取物及其在美白护肤品新用途的开发,讨论了金沙江干热河谷地域余甘子农业栽培的可行性.最后展望了余甘子这种药食两用资源植物的应用前景及研究与开发趋势.  相似文献

Protective efficacy of MPG (2-mercaptopropionyl glycine) was studied against the toxic effects of lead acetate in Swiss albino mice. The animals were treated with single dose of lead acetate @ 180, 200 and 250 mg/kg b.wt. in presence and absence of MPG. The results indicated that the body weight was slightly higher in MPG treated groups on day 10 as compared to only respective lead treated groups in all the three dose level. However, significantly lower body weight was observed in both lead treated and lead along with MPG treated groups as compared to control. Patten of mortality is similar in both lead treated and lead plus MPG treated groups. Conspicuous degenerative changes in testicular tissues and elevation in sperm head shape abnormality were observed in both lead treated and lead along with MPG treated groups but the sperm head shape abnormality and damage were more in lead treated groups as compared to lead plus MPG treated groups. But this difference was non-significant between the two groups. These observations suggest that MPG may not be significantly effective against lead induced damage in testicular tissues at cellular level. However, MPG is able to maintain slightly lower level of sperm abnormality in all the three dose level as compared to their respective lead treated groups. Further, studies are needed to find out the optimum dose of MPG for protection against the lower doses of lead induced lethality as MPG is not significantly effective against the higher doses of lead.  相似文献

Spermatozoa are among the most variable animal cell types, and much research is currently directed towards explaining inter- and intraspecific variation in sperm form and function. Recent comparative studies in passerine birds have found associations between the level of sperm competition and both sperm length and sperm velocity. In species with sperm competition, postcopulatory sexual selection may shape the morphology of sperm as adaptations to the female environment. The speed of evolutionary change in sperm morphology at the species level is largely unknown. In this study, we analysed variation in sperm morphology among morphologically distinct and geographically isolated bluethroat subspecies in Europe. Consistent with previous studies, our analyses of mtDNA and nuclear introns suggest recent divergence and lack of lineage sorting among the subspecies. We found significant divergence in total sperm length and in the length of some sperm components (i.e. head and midpiece). There was a significantly positive relationship between pairwise divergences in sperm morphology and mitochondrial DNA, suggesting a role for genetic drift in sperm divergence. The magnitude of sperm length divergence was considerably higher than that in other geographically structured passerines, and even higher than that observed between several pairs of sister species. We hypothesize that the rapid sperm evolution in bluethroats is driven by sperm competition, and that strong postcopulatory sexual selection on sperm traits can lead to rapid speciation through reproductive incompatibilities.  相似文献

Sperm competition exerts strong selection on males to produce spermatozoa with an optimal morphology that maximizes their fertilization success. Long sperm were first suggested to be favored because they should swim faster. However, studies that investigated the relationship between sperm length and sperm competitive ability or sperm swimming velocity yielded contradictory results. More recently, ratios of the different sections of a spermatozoon (the head, midpiece, and flagellum) were suggested to be more crucial in determining swimming velocity. Additionally, sperm ability to remain and survive in the female storage organs may also influence fertilization success, so that optimal sperm morphology may rather maximize sperm longevity than velocity. In this study, we investigated how sperm morphology is related to sperm velocity and sperm longevity in the house sparrow Passer domesticus. Sperm velocity was found to be correlated with head/flagellum ratio. Sperm with small heads relative to their flagellum showed higher swimming velocity. Additionally, shorter sperm were found to live longer. Finally, we found sperm morphological traits to vary substantially within males and the head/flagellum ratio to be unrelated to total sperm length. We discuss the hypothesis that the substantial within-male variation in sperm morphology reflects a male strategy to produce a diversity of sperm from long, fast-swimming to short, long-living sperm to maximize their fertilization success in a context of sperm competition.  相似文献

Adult male Swiss albino mice were administered ip. suspension solution of cypermethrin in 0.15% DMSO at the doses of 30 mg, 60 mg and 90 mg/kg b. wt. daily for 5 days. Another group of animals was injected cyclophosphamide ip. (60 mg/kg b. wt.) in similar manner which served as positive control. Effect of cypermethrin on body and testes weight and sperm head morphology was studied. Clastogenic potential of cypermethrin was studied by using modified Allium test. The cytological changes were studied in the root tip cells of Allium cepa after 3 days treatment with three different concentration of cypermethrin (0.1, 1.0 and 10.0 microg/ml). The results revealed that body weight gain was considerably reduced in higher dose groups, but the testicular weight did not change significantly in any of the cypermethrin treated groups. However, a significant elevation in the number of abnormal shape of sperm head was noticed in higher dose groups as compared to control. It was observed that the abnormality in the shape of sperm head was dose-dependent. The cytological changes in the root tip cells of Allium cepa indicated that cypermethrin is having toxic effects on the root tip cells in the form of stickiness of chromosomes and also affect the mitotic activity. This study suggest that cypermethrin may have the potential to induce adverse effects on sperm head shape morphology of mouse as well as clastogenic effects on root tip cells of Allium cepa.  相似文献

For the first time in a marine bivalve species, geographic intraspecific variation in sperm morphology in the Chilean mussel, Perumytilus purpuratus, is reported. Samples were obtained in December 2010 from the rocky intertidal at three locations: Antofagasta (23o44′01.08′′S, 70o26′26.11′′W), Valparaíso (32o57′23.07′′S, 71o33′01.86′′W), and Valdivia (39°50′44.26′′S, 73°25′51.99′′W). Scanning electron microscopy was used to describe sperm traits, and genetic data were obtained by sequencing a 16S mtDNA 427-bp fragment. The results showed geographic intraspecific variation with two statistically significant patterns of genetically differentiated sperm morphologies. One genetic clade was found for Antofagasta and Valparaíso, displaying a unique sperm morphotype with a short acrosome accounting for 26 % of the head length, and the other from Valdivia, with a larger acrosome occupying 47 % of the head length. These results will lead to new research to distinguish among alternative hypotheses: the presence of cryptic species, incipient speciation, or a polymorphism in sperm traits of P. purpuratus.  相似文献

The structure of the spermatozoa of 11 species from five families of archaeogastropod (Haliotidae, Fissurellidae, Trochidae, Turbinidae, Phasianellidae) has been examined using transmission electron microscopy. All sperm are of the primitive, or ect-aquasperm type and each species has a sperm head (nucleus and acrosome) with unique morphology. Furthermore, the results from the present study together with those published in the literature reveal that, although there are a few exceptions, the size (length to breadth ratio) and spape of the nucleus and acrosome of sperm of species within each family are similar. It is therefore possible to identify members of a family and differentiate between families using sperm morphology. The intrafamilial similarities and interfamilial differences in sperm structure indicate that a spermiocladistic study of the suborder Vetigastropoda could provide interesting insights into the phylogenetic relationships of this group.  相似文献

The sperm of Acanthodoris pilosa were examined in fresh preparations, histochemically stained samples, electronmicroscope sections, and in paraffin sections of the whole reproductive mass. Sperm are morphologically the same in the ampulla (vas deferens) and the spermatotheca and spermatocyst (bursae for sperm received during copulation). The electron micrographs show sperm with a long spiralled, fibrous, nuclear head with non-fibrous tongue-like acrosome. The tail, attached directly to the posterior of the head, has a 9+2 filamentous arrangement surrounded by a spiralled sheath which contains mitochondria and granular thread running the length of the tail. The thin-walled ampulla contains non-motile sperm and no energy sources such as polysaccharides and lipids. The bursae hold motile sperm and, with histochemical stains, were shown to contain polysaccharide droplets in walls and lumen. The lack of self-fertilization in nudibranchs appears to be associated with a physiological immaturity of the sperm in the hermaphroditic duct, where egg and sperm meet as they leave the ovotestis.  相似文献

Post-copulatory sexual selection (PCSS) is thought to be one of the evolutionary forces responsible for the rapid and divergent evolution of sperm design. However, whereas in some taxa particular sperm traits are positively associated with PCSS, in other taxa, these relationships are negative, and the causes of these different patterns across taxa are poorly understood. In a comparative study using New World blackbirds (Icteridae), we tested whether sperm design was influenced by the level of PCSS and found significant positive associations with the level of PCSS for all sperm components but head length. Additionally, whereas the absolute length of sperm components increased, their variation declined with the intensity of PCSS, indicating stabilising selection around an optimal sperm design. Given the diversity of, and strong selection on, sperm design, it seems likely that sperm phenotype may influence sperm velocity within species. However, in contrast to other recent studies of passerine birds, but consistent with several other studies, we found no significant link between sperm design and velocity, using four different species that vary both in sperm design and PCSS. Potential reasons for this discrepancy between studies are discussed. Male genitalia show rapid and divergent evolution. It is rarely determined whether variation in male genital morphology influences male reproductive success. Male damselflies possess a unique aedeagus with a re-curved head and spiny lateral processes, and most females have two sperm storage organs, a spherical bursa copulatrix and a tubular spermatheca. Previous studies have indicated that the re-curved head may remove bursal sperm, whereas the lateral processes remove spermathecal sperm. However, we need more direct evidence of these functions. We compared sperm number in female sperm storage organs by interrupting copulation to examine sperm removal by the male. In Calopteryx cornelia, males removed almost all bursal sperm but only partially removed spermathecal sperm. In contrast, females of Mnais pruinosa store sperm primarily in the bursa, and males removed only bursal sperm. To examine the functions of male spiny lateral processes, we compared mating behaviour between control and experimental males from which we removed (cut) the lateral processes. In C. cornelia, cutting of the lateral processes resulted in a decreased number of abdominal movements during copulation and no removal of spermathecal sperm. The amount of bursal sperm removed during copulation also decreased in experimental males compared to the unmanipulated males. However, in M. pruinosa, the experimental removal of male lateral processes did not decrease the abdominal movements and little affected the removal of bursal sperm. Inter-specific differences between C. cornelia and M. pruinosa may be caused by variation in the strategies of female sperm storage.  相似文献

Human exposure to electronic waste (e-waste) is increasing, especially in developing nations. This has potential short and chronic adverse health risks. Information is currently scarce on potential genotoxicity of e-waste leachate. In this study, the potential mutagenic and genotoxic effects of leachates from e-waste dumpsites at two electronic markets in Lagos, Nigeria, were investigated. The assays utilized were: bone marrow micronucleus (MN) and chromosome aberration (CA), spermatogonia CA, sperm morphology, and sperm count in mice. Experiments were carried out at concentrations of 1%, 5%, 10%, 25%, and 50% (v/v; leachate: distilled water) of leachate samples. MN analysis showed a concentration-dependent induction of micronucleated polychromatic erythrocytes (MNPCE) across the treatment groups. In the CA tests (bone marrow and spermatogonia), there was concentration-dependent significant reduction in mitotic index and induction of different types of CA. Assessment of sperm shape showed a significant increase in sperm abnormalities with significant decrease in mean sperm count in treated groups. Heavy metals analyzed in the tested samples are believed to contribute significantly to the observed genetic damage. This indicates that e-waste contains potential genotoxic agents; and constitutes a genetic risk in exposed human population.  相似文献

Diplosoma listerianum differs from most ascidians in that, at ovulation, eggs are emitted at the bottom of the ovary and segregated into the tunic, so that fertilization occurs far from seawater. A fertilization canal, a hollow extension of the ovary, conducts sperm towards the egg. In the present paper, ultrastructural evidence is reported on the morphological relationship between the ovary, egg envelopes and oocyte and on the mechanism by which sperm-egg interaction is established. In the ovary, the very complex sperm, equipped with a spiral dense groove, undergo metamorphosis as the first step in a sperm reaction and then pass through the ovary epithelium insinuating themselves between the intercellular junctions which appear to be mouldable, although able to maintain the egg-ovary barrier. Sperm then reach the vitelline coat, where a further step in the sperm reaction occurs. Before the egg abandons the ovary, the sperm head is incorporated into the oocyte by a process recalling phagocytosis, with the formation of an engulfing pocket. Sperm-egg contact and incorporation in D. listerianum occur in a way, never previously reported for other ascidians, in which fusion of plasma membranes takes place immediately after sperm-egg contact. Unlike other cytoplasmic components, the dense groove persists until the sperm enters the egg. It gives a corkscrew-like configuration to the sperm head and allows close adhesion to cell membranes, facilitating sperm movement. Expulsion of numerous cortical granules and features of a cortical reaction were observed in the egg penetrated by the sperm. The mode of internal fertilization of this species in comparison with that of other tunicates and phylogenetic aspects are discussed. Ripe colonies of D. listerianum collected in the Lagoon of Venice, Italy in 1986 and 1991 and colonies reared on glass in aquaria were used for our investigations.  相似文献

Some theoretical models of sperm competition make the assumption that in fish species with external fertilisation, sperm length relates positively to swimming speed at the expense of sperm longevity. Few studies have tested this assumption. We used the three-spined stickleback, Gasterosteus aculeatus L., to study functional sperm morphology. In this study, the relationship between males’ mean sperm length and fertilisation rate was investigated in vitro in a non-competitive situation. Fertilisation at different time points after sperm release was taken into account, and sperm morphology was quantified from scanning electron microscopy images. The time series of artificial fertilisations demonstrated that males which produced sperm with a longer tail fertilised faster, but their sperm had a shorter lifespan (or activity period). It was further suggested that males that produced sperm with a larger midpiece had greater fertilisation chances later on in the fertilisation process. Thus, in sticklebacks, there exists functional variation in sperm morphology, and sperm tail length is traded off against sperm longevity (or activity).  相似文献

Males are limited in sperm and seminal fluid, which can lead to prudent sperm allocation across consecutive matings. While sperm depletion is usually considered a population characteristic, we investigated an individual size-related variation in sperm depletion in the promiscuous sea slug Chelidonura sandrana. We found that sperm counts substantially declined across four successive copulations, indicating sperm depletion. Sperm depletion occurred at a disproportionally fast rate in larger sperm donors. Based on our finding of body size-dependent sperm depletion, we further investigated whether this translates into prudent sperm allocation across ejaculates. We found no evidence that slugs adjusted the number of delivered sperm in response to partner body weight, a tight proxy of fecundity. Instead, sperm counts increased with sperm donor body weight. Sperm donation independent of partner body size and—as found in previous work—partner novelty leaves scope for currently unidentified traits like the sperm competition environment to affect male sperm allocation. Alternatively, we propose that, in C. sandrana, a low optimal male mating rate close to the female optimum as indicated by our data may ultimately render indiscriminate sperm allocation the most beneficial male mating strategy.  相似文献

In the colonial ascidian Diplosoma listerianum (collected in the Lagoon of Venice in 1986 and 1991), ovulating eggs detach themselves from the ovary wall and segregate in the tunic without exposure to seawater. With the aim of finding evidence of the pathway followed by spermatozoa to approach the oocyte, histological and ultrastructural observations were made. The results showed that the hollow ovary elongates in a fertilization canal which flanks the sperm duct and opens externally, next to the anus. Intercellular extended tight junctions isolate the lumen of this canal from the blood. Sperm were found in the fertilization canal, both free and in intracellular vacuoles of wandering cells (phagocytes). These sperm showed differences in comparison to those from the sperm duct, which are possible signs of a sperm reaction. The main modifications were shown by the dense groove, a narrow invagination of the plasmalemma bound to the nuclear envelope by dense material. In non-modified sperm, the groove runs spirally all along the head, while in those in the fertilization canal, it coils in the anterior half of the head, pressing back the long mitochondrion and endoplasmic tubules. These modifications were interpreted to be result of the release of the dense groove, considered to be a sort of stretched, contractile spring. The presence of sperm in the fertilization canal and particularly in phagocytes is discussed in relation to the capacity for exogenous sperm storage by zooids and to the necessity of waste clearance in order for successive waves of fresh sperm to be able to move towards fully grown oocytes. The origin of the female fertilization canal, the role of its tight junctions, and the complex sperm morphology are discussed as adaptations to internal fertilization in D. listerianum.  相似文献

为研究五氯酚钠(PCP-Na)对雄性小鼠的生殖毒性作用,将100只昆明雄性小鼠随机分为5组:3个染毒组、1个阴性对照组(蒸馏水)和1个阳性对照组(环磷酰胺40mg·kg-1).以不同剂量(13.44、26.88、53.76mg·kg-1)PCP-Na经口染毒,连续染毒5d,每天1次.染毒结束后第2d和第23d分两批处死动物,检测精子数量、精子畸形率以及睾丸、附睾重量及其脏器系数的变化.结果表明,PCP-Na染毒结束后第2d,与阴性对照组相比,高剂量组(53.76mg·kg-1)精子畸形率显著增加(p<0.05),睾丸、附睾重量及附睾系数显著下降(p<0.05),其他指标无显著变化(p>0.05).中、低剂量组(26.88、13.44mg·kg-1)各检测指标与阴性对照组均无显著性差异(p>0.05);PCP-Na染毒结束后第23d,与阴性对照组相比,高剂量组(53.76mg·kg-1)精子畸形率显著增加、精子密度显著减少、睾丸和附睾重量显著下降(p<0.05),中剂量组(26.88mg·kg-1)附睾重量显著下降(p<0.05),其他指标均无显著变化(p>0.05);PCP-Na引起的精子畸形以头部畸形为主,主要为无定形,其次为无钩形、香蕉形和尾折叠.以上结果提示,在本实验条件下,PCP-Na对雄性小鼠具有明显的生殖毒性.  相似文献

Spermatozoa vary greatly in size and shape among species across the animal kingdom. Postcopulatory sexual selection is thought to be the major evolutionary force driving this diversity. In contrast, less is known about how sperm size varies among populations of the same species. Here, we investigate geographic variation in sperm size in barn swallows Hirundo rustica, a socially monogamous passerine with a wide Holarctic breeding distribution. We included samples from seven populations and three subspecies: five populations of ssp. rustica in Europe (Czech, Italy, Norway, Spain, and Ukraine), one population of ssp. transitiva in Israel, and one population of ssp. erythrogaster in Canada. All sperm traits (head length, midpiece length, tail length, and total length) varied significantly among populations. The variation among the European rustica populations was much lower than the differences among subspecies, indicating that sperm traits reflect phylogenetic distance. We also performed a test of the relationship between the coefficient of between-male variation in total sperm length and extrapair paternity levels across different populations within a species. Recent studies have found a strong negative relationship between sperm size variation and extrapair paternity among species. Here, we show a similar negative relationship among six barn swallow populations, which suggests that the variance in male sperm length in a population is shaped by the strength of stabilizing postcopulatory sexual selection.  相似文献

In Dolioletta gegenbauri and Doliolum nationalis, collected in 1987 in the bay of Villefranche-sur-mer (French Mediterranean Sea), spermiogenesis is essentially the same. Early spermatids have a round head, a flagellum arising from a single centriole with short microtubules at 45° to its base, several mitochondria, and an acrosome 50 nm thick and 250 nm long with its long axis parallel to the plasma membrane. The acrosomal contents are dense, with a central denser plate. The nuclear envelope next to the acrosome is thickened and concave. In elongating nuclei, strands of chromatin become oriented parallel to the length of the nucleus and then twist helically. Although the mitochondria surround the nucleus, they remain relatively short and do not fuse into a single mitochondrion as in sperm of other tunicates. In very late doliolid spermatids, the acrosome undergoes exocytosis, and exposes fibrous material that stays associated with the tip of the sperm; no acrosomal tubule forms. Exocytosis at this stage may be triggered by fixation. If so, exocytosis probably occurs naturally at some time before fusion of sperm and egg. Sperm have elongate heads (1 m×10 m), with the anterior two-thirds of the nucleus surrounded by mitochondria. Spermiogenesis in doliolids, compared to that in other tunicates, is most like that in solitary members of the class Ascidiacea, except that in the latter the sperm mitochondria fuse and the acrosome appears incapable of exocytosis. In contrast, previous work has shown that salps (class Thaliacea) and colonial didemnid ascidians have an acrosomeless sperm with a spiral mitochondrion, while the class Appendicularia has a sperm with a midpiece, a compact head and an acrosome capable of exocytosis and acrosomal tubule formation. By outgroup comparison with echinoderms and acraniates, appendicularian sperm are plesiomorphic within the Tunicata. Thus, gamete morphology indicates that (1) solitary ascidians and doliolids had a common ancestor, (2) the popular idea that doliolids gave rise to appendicularians is incorrect, and (3) the Thaliacea are polyphyletic, doliolids having arisen very early from the ascidian lineage and salps having arisen later.  相似文献

Sexual selection may operate through either direct selection on preference genes or indirect genetic benefits. However, in many species both direct and indirect selection may operate and can influence female mate and oviposition choice. Adaptations by males for sperm competition can also affect female mate and oviposition choice and can lead to sexual conflict. We investigated the role of direct and indirect benefits and male adaptation to sperm competition for female oviposition decisions in the bitterling, Rhodeus sericeus, a freshwater fish that spawns on the gills of living unionid mussels. Field experiments showed that females chose mussels for spawning on the basis of the number of embryos already in mussels and the body size and/or extent of red coloration of the eyes of territorial males. There was also a significant interaction between these direct and indirect benefits of oviposition choice. Territorial male bitterling avoided leading females to mussels into which rivals had already released sperm. Males also increased their rate of sperm release into mussels in response to sperm release by sneaking males. We discuss the possibility that sexual conflict in oviposition choice occurs in this species.  相似文献

Pronounced sperm dimorphism is reported for the first time in the prosobranch order Vetigastropoda. Using transmission electron microscopy, it is demonstrated thatZalipais laseroni Kershaw (Trochoidea: Skeneidae) produces uniflagellate euspermatozoa (eupyrene, fertile sperm) and multiflagellate paraspermatozoa (oligopyrene, infertile sperm). Euspermatozoa show the following features: (1) a conical acrosomal vesicle; (2) a long tubular, helically coiled nucleus; (3) a short midpiece (mitochondrial sleeve surrounding a 3µm-long electron-dense rod); (4) a chambered body (? fused centrioles) continuous with the dense rod of the midpiece; (5) a flagellum (characterized by an electron-dense sheath surrounding and partly obscuring the central pair of tubules). Paraspermatozoa are composed of an elongate head (lacking an acrosomal complex), a short midpiece (centriolar rods interspersed with mitochondria), and a posterior tuft of flagella. The head consists of a rodshaped anterior body and a condensed nuclear remnant — the latter lodged in a shallow invagination of the anterior body. Multiple flagella are attached via centriolar rods to a layer of dense material lining the nuclear remnant membrane. During paraspermatozoan development, the nucleus partially degenerates, then condenses, while the endoplasmic reticulum (ER), apparently assisted by the Golgi complex, is responsible for production of numerous, electron-dense secretory vesicles. These vesicles subsequently fuse to form the elongate, anterior body of the head region. The ability of at least one line of trochoid gastropods to produce an oligopyrene, multiaxonemal paraspermatozoon, suggests that the Caenogastropoda (with this feature) might have been derived from the Vetigastropoda rather than from any other archaeogastropod source.  相似文献
