An exposure risk assessment of workers in a refinery production unit was undertaken. Gasoline and its main components were investigated through environmental and biological monitoring. Measured variables were environmental benzene, toluene, pentane and hexane; benzene and toluene in blood and urine; tt-MA (metabolite of benzene) in urine. Multivariate statistical analysis of the data showed that worker's exposure to the above substances fell within the limits specified by organisations such as ACGIH. Also, biological values complied with reference values (RV) for non-occupationally-exposed population. Different values of biological variables were determined by separating smokers from non-smokers: smokers had hematic and urinary benzene values significantly higher than non-smokers. During a 3-yr sampling, it was possible to identify a significant decrease of benzene in the workplace air and of hematic benzene for non-smokers. The most exposed department, one in which tank-lorries were loaded, needs further investigation and extended monitoring.  相似文献

beta-Carboline-3-carboxylic acid methylamide (FG 7142), an anxiogenic agent has been found in cigarette smoke condensate, but not in the cigarette itself. When a cigarette, except its filter portion, was immersed in 20 ml of potassium phosphate buffer, pH 7.4, then heated at 60 degrees C for 2 days with or without presence of methylamine, FG 7142 was detected only in the mixture containing methylamine. Furthermore, when the mixtures of beta-carboline derivatives and various amounts of methylamine hydrochloride were heated at 60 degrees C for 5 days, FG 7142 was formed only in the mixtures containing methylamine and 1-methyl-1,2,3,4-tetrahydro-beta-carboline-3-caroxylic acid (MTCA) or 1,2,3,4-tetrahydro-beta-carboline-3-carboxylic acid (TCCA). FG 7142 was also produced in the mixture of glucose, l-tryptophan and methylamine when heated at 200 degrees C in a dry condition. These observations suggest that FG 7142 is formed through the smoking process and that methylamine in cigarette smoke may play an important role in the formation of FG 7142.  相似文献

beta-Carboline-3-carboxylic acid methylamide (FG 7142), an anxiogenic agent, has been measured in airborne particles, automobile-exhaust particles, incinerator ash, smoke condensate of tree leaves and cigarette-smoke-polluted indoor air by high-performance liquid chromatography. This compound has been detected in indoor as well as outdoor air. The source of this compound in indoor air was determined as cigarette smoke, identified from smoking machine studies. This anxiogenic agent was detected in smoke condensate of tree leaves and incinerator ash from garbage burning plants, but not in diesel-exhaust particles. Considering the present results, together with the previous finding that cigarette smoke contains this compound, FG 7142 is likely to be formed through combustion of plants. Our data also suggest that this compound may be widely distributed in the environment.  相似文献

The categories of urban workers undergoing prolonged exposure to gasoline due to vehicle emissions, evaporation and traffic fumes are considered occupationally exposed to benzene, even if at low concentrations. The aim of this study was to evaluate the specificity of unmetabolized benzene excreted in urine (UBz) and S-phenylmercapturic acid (SPMA) as biomarkers of exposure to very low levels of benzene, and to study the impact of putative individual confounders like smoking and alcohol habits, co-exposure to other toxicants and body mass index on the exposure assessment. Environmental and biological monitoring of exposure to benzene were performed in 114 Urban Policemen. The mean value of UBz in non-smokers was significantly lower than in either groups of light to moderate smokers and heavy smokers (0.24, 1.82 and 2.82 microg L(-1), respectively). On the contrary, SPMA values did not discriminate exposure resulting from smoking habits. Moreover, the concentration of UBz in non-smokers appears to be correlated with environmental benzene concentration (BenzA) (R(2)=0.13, beta=0.37, p=0.002). On the other hand, no significant correlation was found between SPMA concentration (corrected for creatinine excretion and log transformed, LogSPMA) and LogBenzA (R(2)=0.003, beta=0.05, p=0.6). Our findings reinforce previous research on the use of unmetabolized urinary benzene as a specific and sensitive biomarker of low-level exposure to benzene and confirm that smoking habits strongly influence the excretion of UBz.  相似文献

The study of lead exposure among workers in Selangor and the Federal Territory was carried out based on the delta-aminolevulinic acid (ALA) level in urine. Occupations which are expected to have higher lead exposure were chosen in this research. The ALA level in the workers' urine was linked to a few variables which may contribute to the lead level in the body. The result of this study showed that the ALA level of the urine of university students (0.352 +/- 0.038 mg/100 ml) < clerical staff (0.560 +/- 0.043 mg/100 ml) < traffic police (0.612 +/- 0.064 mg/100 ml) < vehicle workshop workers (0.673 +/- 0.099 mg/100 ml) < petrol kiosk workers (0.717 +/- 0.069 mg/100 ml) < bus drivers/conductors (0.850 +/- 0.055 mg/100 ml) which was similar to workers in the printing industry (0.852 +/- 0.110 mg/100 ml). The ALA levels in the urine of the exposed workers were significantly different from the control group (university students). However, results obtained from clerical staff revealed that they were also in the exposed group category. Analysis of variance showed that the exposed groups are in a population which is different from the control population. Correlation tests suggest that there is no significant connection between the ALA level in the urine and the variables tested. Furthermore, Duncan's Multiple Range Test showed no significant differences between the smoking/non smoking group, alcoholic/non-alcoholic group, race and sex (p > 0.05).  相似文献

We studied the concentration of 10 primary aromatic amines (AA), which are classified as suspected carcinogens, in indoor and outdoor air in Italy. The measured AA included: aniline, o-toluidine, m-toluidine, p-toluidine, 2,3-dimethylaniline, 2,4-dimethylaniline, 2,5-dimethylaniline, 2,6-dimethylaniline, 2-naphtylamine and 4-aminobiphenyl. In the indoor environment (homes, offices and public buildings) the level of contamination (expressed as sum of 9 AA, excluding aniline) varied from 3 ng/m3 (hospital ward) to 207 ng/m3 (discotheque). In most indoor environments with no contamination from cigarette smoke the AA levels were below 20 ng/m3, whereas in the presence of smokers higher values were observed. Aniline levels were more erratic (varying from 53 ng/m3 (office of non-smokers) to 1929 ng/m3 (discotheque) and were not related to cigarette smoke. The concentration range of AA (excluding aniline) in the outside air varied from 3 ng/m3 (Siena) to 104 ng/m3 (Brindisi); aniline concentration was extremely variable. Most samples of outdoor air had AA levels lower than 40 ng/m3. In conclusion, AA are widespread air contaminants and attain a high concentration in heavily contaminated indoor environments, due to smoking and poor ventilation. AA occasionally attain a high level in outdoor air as well. Therefore, a strategy of reduction of the exposure to AA should consider the abatement of multiple sources of contamination.  相似文献

Chronic effects of chlornitrofen (CNP) on the reproduction of Brachionus urceolaris (Rotatoria) were investigated by exposure of individuals to CNP from the egg stage, which had been attached to the adult. The survivors of 12 neonates, which had been exposed to CNP100, 70 or 40 microg liter(-1) decreased to 50% at the age of c. 2, 4 and 6 days, respectively, compared to c. 6.5 days for those exposed to 0, 10 and 20 microg liter(-1) CNP. Release of offspring (mostly two individuals per day) started at two days old. At the peak, four days old, a control female produced 8.1+/-0.9 offspring per day compared with 4.5+/-1.3 (mean+/-SD, n=12) at 40 microg liter(-1). The cumulative numbers of offspring produced by a female were 25.8+/-1.2, 24.2+/-2.9, 22.3+/-3.6 and 13.6+/-3.1 (mean+/-SD, n=12) at control, 10, 20 and 40 microg liter(-1) CNP exposure, respectively. The 50% reproductive impairment concentration was calculated to be 37 microg liter(-1). Growth of neonates was barely detectable at 70 microg liter(-1), and the rapid increase in the effect of CNP from 40 to 70 microg liter(-1) was attributed to an increase in tolerance with growth of the neonates. The effects of CNP on reproduction were also tested by CNP exposure through food (CNP-accumulated Chlorella). The 50% reproductive impairment concentration of CNP in the alga was calculated to be c.60 microg g(-1) (wet weight) by the same method used to assess the dissolved CNP, although the effect of CNP which may have been released from the alga to the water could not be estimated precisely.  相似文献

The purpose of this study was to evaluate the effect of traffic volume on ambient black carbon (BC) concentration in an inner-city neighborhood "hot spot" while accounting for modifying effects of weather and time. Continuous monitoring was conducted for 12 months at the Baltimore Traffic Study site surrounded by major urban streets that together carry over 150,000 vehicles per day. Outdoor BC concentration was measured with an Aethalometer; vehicles were counted pneumatically on two nearby streets. Meteorological data were also obtained. Missing data were imputed and all data were normalized to a 5-min observational interval (n = 105,120). Time-series modeling accounted for autoregressively (AR) correlated errors. This study found that outdoor BC was positively correlated at a statistically significant level with neighborhood-level vehicle counts, which contributed at a rate of 66 +/- 10 (SE) ng/m3 per 100 vehicles every 5 min. Winds from the SW-S-SE quarter were associated with the greatest increases in BC (376-612 ng/m3). These winds would have entrained BC from Baltimore's densely trafficked central business district, as well as a nearby interstate highway. The strong influence of wind direction implicates atmospheric transport processes in determining BC exposure. Dew point, mixing height, wind speed, season, and workday were also statistically significant predictors. Background exposure to BC was estimated to be 905 ng/m3. The optimal, statistically significant representation of BC's autocorrelation was AR([1:6]) x 288 x 2016, where the short-term AR factor (lags 1-6) indicated that BC concentrations are correlated for up to 30 min, and the AR factors for lags 288 and 2016 indicate longer-term autocorrelations at diurnal and weekly cycles, respectively. It was concluded that local exposure to BC from mobile sources is substantially modified by meteorological and temporal conditions, including atmospheric transport processes. BC concentration also demonstrates statistically significant autocorrelation at several time scales.  相似文献

In this study we investigated the accumulation, tissue-specific distribution and possible debromination of BDE 209 in a terrestrial songbird species, the European starling, using silastic implants as a method of exposure. BDE 209 accumulated in the blood of the exposed starlings to a mean peak concentration of 16+/-4.1 ng/ml on day 10. After this peak, there was a decline to 3.3+/-0.4 ng/ml blood at the end of the exposure period of 76 days, which suggests elimination of BDE 209. In the exposed group, the muscle concentrations (461 ng/g lipid weight [lw], 430 ng/g lw) were about twofold those in liver (269 ng/g lw, 237 ng/g lw). In addition to BDE 209, other PBDE congeners, particularly octa- and nonaBDEs, were also present in the muscle and liver, suggesting bioformation from BDE 209. To our knowledge, these results are the first indications for the debromination of BDE 209 in birds.  相似文献

We examined the relationship between cotinine measures in follicular fluid (FF) and urine to inform our exposure assessment strategy for an ongoing epidemiological study of secondhand tobacco smoke (STS) exposure and early pregnancy loss. Among subjects undergoing in vitro fertilization (IVF), we compared cotinine levels in paired urine and FF samples from the same women and examined FF cotinine levels over time. We found a weak rank-order relationship (Spearman r < 0.2) and poor agreement for classifying nonsmoking individuals as exposed to STS (sensitivity = 0.29-0.71; specificity = 0.35-0.72) between cotinine concentrations in FF and urine. We observed fair reliability (ICC = 0.42-0.52) in FF cotinine concentrations from women undergoing multiple IVF cycles. If available, FF cotinine concentrations may be desired as a biomarker of low-level tobacco smoke exposure over urinary cotinine in studies of early reproduction. Collection of multiple FF samples for cotinine analysis may be needed to accurately represent long-term STS exposure.  相似文献

We investigated whether exposure to environmentally relevant polybrominated diphenyl ethers (PBDEs) causes immunomodulation in captive nestling American kestrels (Falco sparverius). Eggs within each clutch, divided by laying sequence, were injected with safflower oil or penta-BDE congeners-47, -99, -100, and -153 dissolved in safflower oil (18.7 microg sigmaPBDEs/egg) approximating Great Lakes birds. For 29 days, nestlings consumed the same PBDE mixture (15.6+/-0.3 ng/g body weight per day), reaching sigmaPBDE body burden concentrations that were 120x higher in the treatment birds (86.1+/-29.1 ng/g ww) than controls (0.73+/-0.5 ng/g ww). PBDE-exposed birds had a greater PHA response (T-cell-mediated immunity), which was negatively associated with increasing BDE-47 concentrations, but a reduced antibody-mediated response that was positively associated with increasing BDE-183 concentrations. There were also structural changes in the spleen (fewer germinal centers), bursa (reduced apoptosis) and thymus (increased macrophages), and negative associations between the spleen somatic index and sigmaPBDEs, and the bursa somatic index and BDE-47. Immunomodulation from PBDE exposure may be exacerbated in wild birds experiencing greater environmental stresses.  相似文献

We evaluated the use of the gas exchange rate as an ecologically relevant indicator of chemical stress in avian embryos/eggs. Northern bobwhite quail (Colinus virginianus) were exposed to octahydro-1,3,5,7-tetranitro-1,3,5,7-tetrazocine (HMX) via feed containing nominal concentrations of 0, 12.5, 50.0, and 125.0 mg kg(-1). Metabolic rates (oxygen consumption) of developing quail eggs were then measured via respirometry to examine potential effects of HMX exposure. Metabolic rates were examined on 5, 9, and 21 d of incubation. Next, concentrations of HMX in embryos/eggs were determined by liquid chromatography-mass spectrometry. Mean (+/-SE) concentrations of HMX in eggs were 21.0+/-5.9, 1113+/-79.0, 3864+/-154.0, and 7426+/-301.1 ng g(-1) in control, low, medium and high dose groups, respectively. There were significant differences in oxygen consumption among the three embryo ages, however differences among the ages were not consistent among dose groups (age x dose group interaction p<0.0001). Oxygen consumption rates did not vary as a function of HMX in embryos (p=0.18). No evidence was observed for alterations of in ovo metabolic rates associated with HMX exposure.  相似文献

Real-time measurement of outdoor tobacco smoke particles   总被引:1,自引:0,他引:1
The current lack of empirical data on outdoor tobacco smoke (OTS) levels impedes OTS exposure and risk assessments. We sought to measure peak and time-averaged OTS concentrations in common outdoor settings near smokers and to explore the determinants of time-varying OTS levels, including the effects of source proximity and wind. Using five types of real-time airborne particle monitoring devices, we obtained more than 8000 min worth of continuous monitoring data, during which there were measurable OTS levels. Measurement intervals ranged from 2 sec to 1 min for the different instruments. We monitored OTS levels during 15 on-site visits to 10 outdoor public places where active cigar and cigarette smokers were present, including parks, sidewalk cafés, and restaurant and pub patios. For three of the visits and during 4 additional days of monitoring outdoors and indoors at a private residence, we controlled smoking activity at precise distances from monitored positions. The overall average OTS respirable particle concentration for the surveys of public places during smoking was approximately 30 microg m(-3). OTS exhibited sharp spikes in particle mass concentration during smoking that sometimes exceeded 1000 microg m(-3) at distances within 0.5 m of the source. Some average concentrations over the duration of a cigarette and within 0.5 m exceeded 200 microg m(-3), with some average downwind levels exceeding 500 microg m(-3). OTS levels in a constant upwind direction from an active cigarette source were nearly zero. OTS levels also approached zero at distances greater than approximately 2 m from a single cigarette. During periods of active smoking, peak and average OTS levels near smokers rivaled indoor tobacco smoke concentrations. However, OTS levels dropped almost instantly after smoking activity ceased. Based on our results, it is possible for OTS to present a nuisance or hazard under certain conditions of wind and smoker proximity.  相似文献

A study was undertaken to examine the uptake of the organophosphate insecticide pyridaphenthion in the chlorophyta Chlorella saccharophila. Algae cultures were exposed to the initial nominal concentration 10.0 mg l(-1) pyridaphention during seven days. The insecticide bioconcentrates in the biomass to the highest level of 441.5 +/- 25.9 mg kg(-1) on the fifth day of exposure and was followed by a decrease to 76.6 +/- 5.1 mg kg(-1) on the seventh day. A model was constructed to describe the dynamic process, which estimated a bioconcentration factor (BCF) equal to 28. The study demonstrates the potential of accumulation of pyridaphenthion in aquatic organisms and helps to expand the pyridaphenthion toxicity database. The replacement of fenitrothion by pyridaphenthion concerning their use in rice flooded cultures is discussed.  相似文献

The level of mercury exposure in Slovenian dental practice was studied by measurement of air mercury levels in 63 surgeries and by analysis of the mercury content in blood and urine samples of professionally exposed and control groups (total of 77 participants).

The mean ambiental mercury concentration for all investigated surgeries was 2.8 ug Hg/m3 (range: 0.4–8.2), which is considerably below the health-based occupational exposure limit of 25 ug/m3. No significant relationships between age or type of surgeries, or the time during the working day and the air mercury levels were found.

The mean value for mercury in blood was 3.0 ng Hg/g (range: 0.9–7.7), which can be considered as the normal range for the general population. The occupational profile of the workers, years in the profession, as well as sex, showed a nonsignificant effect on blood mercury.

Urinary mercury levels were also low. Only 3 of 44 values exceeded 15 ng Hg/g. No influence of sex or occupational profile was found; however, a significant negative linear relationship (r=−0.473; P=0.0012) was observed between the concentration of mercury in urine and years in the profession.

The data indicate good mercury hygiene in Slovenian dentistry.  相似文献

One hundred and ninety-six randomly selected nonsmoking subjects collected air samples close to their breathing zone by wearing personal monitors for 24 h. The study was centred in Basel, Switzerland, and comprised housewives in one group, primarily for assessing exposures in the home, and office workers in a second group to assess the contribution of the workplace to overall exposure. Samples collected were analysed for respirable suspended particles (RSP), nicotine, 3-ethenylpyridine and environmental tobacco smoke (ETS) particles by using ultraviolet absorbance, fluorescence and solanesol measurements. Saliva cotinine analyses were also undertaken to confirm the nonsmoking status of the subjects. Based upon median 24 h time weighted average concentrations, office workers who live and work with smokers were exposed to 39 μg m-3 RSP, 6.6 μg m-3 ETS particles and 0.90 μg m-3 nicotine. Housewives living with smokers were exposed to median concentrations of 34 μg m-3 RSP, 1.4 μg m-3 ETS particles and 0.60 μg m-3 nicotine. Workplaces where smoking occurred were estimated, on average, to contribute between 34 and 46% to annual exposure of ETS particles and nicotine. Based upon 90th percentile values the most highly exposed housewives, those living with smokers, would potentially inhale 18 cigarette equivalents per year whilst the most highly exposed office workers, both living and working with smokers, might inhale 61 cigarette equivalents. The rate at which subjects misreported their nonsmoking status varied between 9.7 and 12.2%.  相似文献

The biological defence mechanism called MXR or MXD for multixenobiotic resistance or defence protects cells against the entry and the accumulation of xenobiotics. As the defence is modulated by man made chemicals, MXR is used as a biomarker of organisms' exposure to environmental contamination. However, to reliably assess and evidence MXR induction, the use of a reference level is required. In this context, we focused on MXR background level in a freshwater bivalve, the zebra mussel Dreissena polymorpha, in order to propose its use as a reference during MXR evaluation. We monitored the MXR transport activity in mussels collected either in a natural population or in a caged population and then transplanted to clean water in the laboratory. The results showed that MXR activity was decreased to its baseline level after an eight to nine day depuration period (13.1+/-3.1; 7+/-2.6; 13.7+/-3.9 pmol RB min(-1)org(-1) after three experiments of laboratory depurations). Moreover, significant MXR induction was measured in depurated zebra mussels exposed to contaminated sites (39.6+/-3.7; 59.2+/-20.3 pmol RB min(-1)org(-1) after two experiments of field exposure), showing that the laboratory depuration did not affect the induction potential. The MXR responses (decrease as well as increase) occurred in few days and were highly significant, highlighting its reactivity in zebra mussels. Finally, this paper confirms the usefulness of MXR as a tool in biomonitoring studies and provides a protocol for field experiments that enables to establish and use the background level of MXR activity as a reference.  相似文献

Plasma PBDE and thyroxine levels in rats exposed to Bromkal or BDE-47   总被引:2,自引:0,他引:2
In experimental models, polybrominated diphenyl ethers (PBDEs), a group of brominated flame retardants, have caused effects in a number of biological end-points, including neurobehavioural effects, disturbances in thyroid and steroid hormone homeostasis, and other steroid-related effects. Almost exclusively, only external dose metrics (dose per body weight basis) have been studied in connection to the observed effects. In this study we report on new analyses of plasma PBDE levels in surplus samples from earlier studies on thyroid hormones (TH) in exposed rodents. Female, 7-week old Sprague-Dawley rats were given either Bromkal 70-5 DE (Study I; 18 or 36 mg/kg bw/day) or BDE-47 (Study II; 1, 6 or 18 mg/kg bw/day) daily by gavage for two weeks. At an external dose of 18 mg/kg bw/day significant TH effects (decreased plasma free thyroxin levels) were observed in both studies, corresponding to an internal (plasma) dose of 463 microg sumPBDE/g lipid (Study I) or 421 microg BDE-47/g lipid (Study II). If we compare the contribution of different BDE congeners to the total BDE level in rat plasma after Bromkal exposure (Study II), and in the Bromkal mixture itself, the most important congener in the Bromkal mixture were also found in plasma. However, the relative concentration of BDE-99 was lower, and that of BDE-153 was higher, than that of the mixture, indicating selectivity in uptake, metabolism and/or excretion of the individual BDE congeners. Explicitly, the possible in vivo conversion of BDE-99 to BDE-47, and of BDE-154 to BDE-153 could not be excluded. The internal dose in the present rat study could be compared to reported human serum doses of PBDE. Human serum/blood levels have a wide range, from 3 to 6 ng sumPBDE/g lipid in background samples from Europe, about 10 times higher in US sample, and up to 100 times higher (300-600 ng/g lipid) in upper-end levels in collected samples from USA. As a consequence, the margin between effects levels in the rat and exposure levels in man varies widely, with a quotient roughly from 1000 to 100,000. Generally, it could be expected that this margin is lower than if external dose metrics would be used. An even lower margin could be expected as recent studies have shown effects in offspring at lower doses than those giving effects in our studies. Lastly, it should be noted that humans are already exposed to a mixture of chemicals in daily life, a fact that complicates this kind of comparison.  相似文献

Persistent organochlorine pesticides in human milk samples from Australia   总被引:2,自引:0,他引:2
Human milk has been used as a surrogate for the assessment of body burden and exposure to persistent lipophilic organochlorine pesticides (OCPs). With the exception of restricted use of mirex, the use of persistent, lipophilic OCPs has been banned in Australia since the 1980s. The decline of human body burden of OCPs following their ban in many industrialised countries is well recorded worldwide from the 1970s until the 1990s though little is known on whether these trends are continuing. In this study, 157 human milk samples collected during 2002 and 2003 as well as 24 samples collected in 1993 were analysed as 20 regional pools for 17 OCPs. OCPs were detected in all pooled human milk samples from 2002/03 typically with highest concentrations of p,p'-dichlorodiphenyldichloroethylene (p,p'-DDE) (mean+/-standard deviation; median concentration 311+/-174; 279 ng g(-1) lipid) followed by beta-hexachlorocyclohexane (beta-HCH) (80+/-173; 21 ng g(-1) lipid). Other OCPs consistently detected included dieldrin (16+/-6; 17 ng g(-1) lipid), hexachlorobenzene (HCB) (18+/-16; 14 ng g(-1) lipid), transnonachlor (11+/-5; 9 ng g(-1) lipid) and p,p'-dichlorodiphenyltrichloroethane (p,p'-DDT) (9+/-6; 7 ng g(-1) lipid). The results from this study indicated that following a substantial decline of OCP concentrations from the early 1980s to the 1990s, little decline could be observed since then. Furthermore the use of multivariate statistics indicated some regional trends with slightly higher levels of the broadly used insecticides DDT and HCH in both historic and recent samples from Melbourne, whereas, sample pools collected from mothers that lived in rural Queensland and New South Wales as well as Adelaide and Sydney showed comparatively higher levels of heptachlor and dieldrin - both of which have been used for termite treatment. These results indicate that even 20 years after the discontinuation of usage, historical use of OCPs rather than exposure via global transport of OCPs is responsible for continuous low exposure in Australia.  相似文献
