The effects of varying concentrations of urea, phosphogypsum and paper mill sludge (PMS) on the morphology, histology, tissue protein content, lipid peroxidation (LPX), activities of lactate dehydrogenase (LDH), acetylcholinesterase (AChE) and catalase in earthworms Drawida willsi and Lampito mauritii have been studied over an exposure period of 24?hr. Integumentary lesions, clitellar swelling and loss of pigmentations were found to be major morpho-pathological changes in the worms. Histology indicated cuticular damage, ruptured epithelium and muscle fibres with accumulation of cellular debris. Lowest tissue protein content (57.02?±?4.02?mg/g tissue) and highest LPX (0.113?±?0.04, 0.137?±?0.08?nmol/mg protein) were noticed in D. willsi at a high concentration of PMS, whereas highest tissue protein content was observed in L. mauritii (115.32?±?7.18?mg/g tissue) with the same treatment. In both the species, LDH activity was minimum at a high concentration of urea (0.172?±?0.02; 0.247?±?0.08?U/mg protein). AChE activity was highest (0.099?±?0.002?U/mg protein) at a high concentration of PMS in D. willsi, whereas catalase activity was the maximum (0.338?±?0.02?U/mg protein) at high concentrations of PMS in L. mauritii. The study indicated that morpho-histological and enzymatic alterations in these earthworms exposed to agrochemicals could be useful biomarkers to evaluate soil toxicity.  相似文献

为了探讨马拉硫磷和主要水解产物对圆瘤浮萍的生长抑制作用及可能的氧化损伤机制,采用静态培养法,研究不同浓度的马拉硫磷及其水解产物二甲基二硫代磷酸酯(DMDTP)、二甲基硫代磷酸酯(DMTP)、二甲基磷酸酯(DMP)对圆瘤浮萍生长、叶绿素含量、叶绿体活性、超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性及丙二醛(MDA)含量的影响。结果表明,暴露7 d后,浓度为100 mg a.i.·L-1处理下的浮萍叶状体生长量与空白对照组无显著性差异;DMDTP、DMTP和DMP对圆瘤浮萍叶状体生长量的半数抑制浓度(7 d-IC50)分别为52.9、45.5和98.0 mg a.i.·L-1。随着试验液中DMDTP、DMTP和DMP浓度的升高,圆瘤浮萍叶绿素含量均明显降低,叶绿体活性则分别表现为诱导-抑制、抑制和抑制效应。在DMDTP、DMTP和DMP胁迫下,脂质过氧化产物丙二醛(MDA)含量明显增加;圆瘤浮萍体内的抗氧化酶系统均受到影响。总体表现为随着DMDTP浓度的增加,SOD活性先降低后升高;DMTP和DMP胁迫下,SOD活性呈现先降后升再降的趋势;POD和CAT活性则随3种水解产物浓度的增加呈现先升后降趋势;这表明抗氧化酶系统对外部胁迫的应答很复杂。马拉硫磷对圆瘤浮萍的毒性较小,其水解产物DMDTP、DMTP和DMP均对圆瘤浮萍的生长产生一定的毒性作用。在实际生产中,应多加关注马拉硫磷水解产物对环境生物的风险评估。  相似文献

The objective of this study was to describe the seasonal variations in nucleic acid contents and amino acid profiles in the muscle of juvenile Nephrops norvegicus. RNA and protein contents, and RNA:protein and RNA:DNA ratios varied significantly between seasons, being highest in spring and lowest in autumn/winter ( P<0.05). Though DNA content increased significantly from autumn to summer ( P<0.05), protein:DNA ratio did not show significant seasonal variations ( P=0.05). In respect to protein-bound amino acid content (BAA), a significant increase was observed from winter to summer ( P<0.05). Both essential (EAA) and non-essential amino acids (NEAA) increased significantly (27.6% and 27.8%, respectively; P<0.05), mainly due to the considerable increase in arginine and proline (59.1% and 225.2%, respectively; P<0.05). A significant decrease was observed in the free amino acid content (FAA) from winter to summer ( P<0.05); and a higher percentage decrease occurred in free non-essential (FNEAA; 27.9%) in comparison to free essential amino acids (FEAA; 21.8%). The significant increase in RNA and BAA contents from winter to spring may be related to protein synthesis. On the other hand, the lowest values obtained in winter may be due to a reduction in feeding activity; in this period the muscle protein must be progressively hydrolysed, which is evident with the higher FAA content. The liberated amino acids enter the FAA pool and become available for energy production.  相似文献

 Free amino acid (FAA) and protein content were measured in various developmental stages of Artemia franciscana, from cysts to Instar III metanauplii. In addition, decapsulated cysts of 15 Artemia populations from different localities were compared with respect to their FAA and protein content. Furthermore, the content and composition of the FAA pool were modulated by hatching the cysts at various salinities, and by enriching the nauplii with algae or a lipid-enrichment emulsion. The FAA content increased threefold from cysts to nauplii, and Instar III metanauplii contained nearly 50% taurine of total FAA. Cysts of A. franciscana were found to contain one-third the amount of FAA compared to the other Artemia species investigated. The content and pool composition of FAA was successfully modulated in 11 of 13 populations, where by the content of FAA was significantly increased when hatched at high salinity. Finally, enrichment elevated the content of FAA and changed the pool composition, thereby showing a dietary effect. Algal enrichment also increased the protein content. The growth, survival, digestive enzyme activity and biochemical composition ofPenaeus japonicus (Bate) larvae and postlarvae were measured under three feeding regimes. Larvae were reared through the protozoeal stages usingChaetoceros gracilis. From the first mysis stage, three feeding regimes were used; (A)C. gracilis plusArtemia sp. nauplii, (B)Artemia sp. nauplii alone or (C)C. gracilis alone. No significant difference was found in growth, survival, protein content or lipid content of postlarvae from the treatments receiving the single-feed type, despite the low protein (7%) and highly unsaturated fatty acid content of the alga. Growth of larvae receiving the mixed diet was significantly higher than in the other treatments. Trypsin activity was more strongly influenced than amylase activity by dietary treatment, and differences in the ratio of these enzymes between treatments suggest independent control of their secretion. Trypsin activity recorded in larvae feeding onC. gracilis was up to six time higher than in larvae feeding onArtemia sp. nauplii, apparently in response to the low protein content of the alga. Larvae receiving the mixed diet exhibited an intermediate level of trypsin activity; it is suggested that the ingestion of algae is necessary for optimal assimilation of the zooplankton component of the diet.  相似文献

An artificial soil method was applied to study the effects of perfluorooctane sulphonate (PFOS) and perfluorooctanoic acid (PFOA) on earthworms (Eisenia fetida). Survival, growth inhibition and damage to DNA of earthworms were detected after 14 d acute exposure. The 14 d-LC50 of PFOS and PFOA was 478.0?mg·kg?1 dw and 759.6?mg·kg?1 dw, respectively, indicating that they were of low toxicity. Both PFOS and PFOA could significantly inhibit the growth of earthworms after 14 d exposure, and growth inhibition rates increased with the greater concentrations of PFOS or PFOA, showing a dose–response relationship (PFOS: r?=?0.951, P r?=?0.962, P?P?50 of PFOS was lower than that of PFOA, the growth inhibition rate of earthworm exposed to PFOS was higher than that exposed to PFOA at the same concentration level, and the median values of TL, CL and OTM in PFOS treatments were also higher than those in PFOA treatments. In conclusion, both these fluorine compounds were moderately toxic to earthworms, but the PFOS effect was greater than that of PFOA.  相似文献

Effects of food availability and season on the free amino acid (FAA) and total protein content of the copepod Calanus finmarchicus females were investigated in two mesocosm experiments on the Norwegian west coast in spring and autumn. Starved C. finmarchicus females showed no change in total FAA content, but the FAA pool composition changed drastically. During the first 10 days of starvation the protein content showed a moderate decline (<2 µg ind -1); however, during the following 21 days the total content was drastically reduced, from 63 to 9 µg ind -1. This supports the notion of a sequential catabolism of endogenous nutrients during starvation. In females at high food concentrations, the body protein content increased during spring, but decreased during autumn. The FAA pool composition of females differed between spring and autumn in 14 of the 18 FAA investigated. Reduced body protein content and increased proportion of essential free amino acid were observed during starvation. Similar changes were observed in females sampled at the end of the mesocosm experiments in the autumn. The results suggest that mature C. finmarchicus females are in a negative protein balance during autumn, despite high food concentrations, contributing to a lower fitness than in females maturing during the spring.  相似文献

研究了不同浓度铀(0.1、1.0、10.0和30.0mg·L-1)胁迫对满江红鱼腥藻(Anabaena azollae)和爪哇伪枝藻(Scytonemaj avanicum)生长及抗氧化酶活性的影响。结果表明,低浓度铀(0.1和1.0mg·L-1)条件下,两种蓝藻的超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性及丙二醛(MDA)含量均无明显变化;叶绿素a含量和可溶性蛋白质含量均较对照上升,表明低浓度铀对A.azollae和S.javanicum均未形成明显胁迫,对生长表现为刺激和促进作用。10.0mg·L-1浓度的铀对A. azollae的生长影响不大,对S. javanicum却产生了明显的抑制作用。30.0mg·L-1铀胁迫下,两种蓝藻中的MDA显著增加,抗氧化酶系统受到破坏,生长严重受阻。实验结果显示,A. azollae和S. javanicum的耐铀极限分别在10.0~30.0mg·L-1和1.0~10.0mg·L-1之间,A. azollae的抗铀性比S. javanicum强。  相似文献

The present study is a comprehensive comparison of the biochemical composition (protein, lipid, glycogen and cholesterol contents, and amino acid and fatty acid profiles) of 14 species of cephalopods with different life strategies (benthic, nektobenthic, benthopelagic and pelagic) in distinct habitats (neritic, demersal, oceanic and deep sea), with special emphasis placed on a male Architeuthis sp. The giant squid showed a significantly lower protein and total amino acid content in the gonad and digestive gland. The major essential amino acids were leucine, lysine and arginine. The major non-essential amino acids were glutamic acid, aspartic acid and proline. The benthopelagic cirrate octopus Opisthoteuthis sp. showed a lower content of these nitrogen compounds in the muscle. In respect to lipid and fatty acid contents, the giant squid showed the highest values in the gonad and digestive gland and the lowest in the muscle. Most of saturated fatty acid content was presented as 16:0; monounsaturated fatty acid content, as 18:1 and 20:1; and polyunsaturated fatty acid content, as 20:4n-6, 20:5n-3 and 22:6n-3. The highest cholesterol content in the gonad was detected in Opisthoteuthis sp. and Architeuthis sp.; in the digestive gland. in Todarodes sagittatus; and in the muscle, in Sepia elegans. The highest glycogen value in the gonad was detected in Octopus vulgaris; in the digestive gland and muscle, the highest values were attained in Opisthoteuthis sp. The relationships between life strategies and biochemical composition were investigated and principal component analysis (PCA) was performed. The different cephalopod life strategies could be well separated on the basis of the first two principal components. The nektobenthic and pelagic strategies were clearly separated from the benthic, suggesting that these groups of species are characterized by lower lipid content and higher protein content in the gonad. A rather similar life-strategy distinction was obtained for the digestive gland. The benthopelagic strategy was also well separated from benthic and pelagic strategies and from Architeuthis sp. In the muscle, the results indicated lower nitrogen and carbon compound contents in Architeuthis sp. and Opisthoteuthis sp. The environmental conditions that possibly explain the Architeuthis sp. stranding, the relationships between biochemical compositions and the life strategies of the different cephalopod species studied are discussed.Communicated by S.A. 为了研究铅与纳米SiO2联合染毒所致的细胞损伤特征,并从氧化应激方面探讨其可能的作用机制。用铅和SiO2处理A549细胞,采用四唑盐(MTT)比色法检测细胞存活率,评价铅和SiO2联合染毒所致的细胞损伤特征;采用硫代巴比妥酸(TBA)比色法检测细胞内丙二醛(MDA)含量,评价铅与SiO2联合染毒所致细胞的氧化应激状态;检测了细胞内抗氧化物还原型谷胱甘肽(GSH)含量以及细胞内抗氧化酶的活性,以评价铅与SiO2联合染毒对细胞抗氧化系统的影响。将实验数据进行ANOVA分析。结果表明,铅、SiO2单独染毒组各指标没有明显改变;而联合染毒能造成细胞氧化损伤,表现为细胞存活率、GSH水平、超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH-Px)活性显著低于对照组及2个单独染毒组(P<0.05),细胞内MDA含量显著高于对照组及各单独染毒组(P<0.05)。可见,联合染毒可引起明显的细胞毒性,氧化损伤可能是铅与SiO联合染毒致肺细胞毒性损伤的作用机制之一。  相似文献

A pot experiment was conducted to examine the influence of potassium (K) fertilizer (K2SO4) application on the phytoavailability and speciation distribution of cadmium (Cd) and lead (Pb) in soil. Spring wheat (Triticum aestivum L.) was selected as the test plant. There were seven treatments including single and combined contamination of Cd and Pb. CdCl2·2.5 H2O and Pb(NO3)2 were added to the soil at the following dosages: Cd + Pb = 0.00 + 0.00, 5.00 + 0.00, 25.0 + 0.00, 0.00 + 500, 0.00 + 1000, 5.00 + 500 and 25.0 + 1000 mg kg−1, denoted by CK, T1, T2, T3, T4, T5 and T6, respectively. The K fertilizer had five levels: 0.00, 50.0, 100, 200 and 400 mg K2O kg−1 soil, denoted by K0, K1, K2, K3 and K4, respectively. The results showed that the K fertilizer promoted the dry weight (DW) of wheat in all treatments and alleviated the contamination by Cd and Pb. The application of K2SO4 reduced the uptake of Cd in different parts including roots, haulms and grains of wheat; the optimum dosage was the K2 level. K supply resulted in a significant (P < 0.05) decrease in the soluble plus exchangeable (SE) fraction of Cd and there was a negative correlation (not significant, P > 0.05) between the levels of K and the SE fraction of Cd in soil. The application of the K fertilizer could obviously restrain the uptake of Pb by wheat and there were significant (P < 0.05) negative correlations between the concentrations of Pb in grains and the levels of K in soil. K supply resulted in a decrease in the SE fraction of Pb (except the K1 level) from the K0 to K4 levels. At the same time, the application of the K fertilizer induced a significant (P < 0.05) decrease in the weakly specifically adsorbed (WSA) fraction of Pb and a significant (P < 0.05) increase in the bound to Fe–Mn oxides (OX) fraction of Pb. At different K levels, the concentration of Pb in the roots, haulms and grains had a positive correlation with the SE (not significant, P > 0.05) and WSA (significant, P < 0.05) fractions of Pb in the soil. All the K application levels in this experiment reduced the phytoavailability of Cd and Pb. Thus, it is feasible to apply K fertilizer (K2SO4) to alleviate contamination by Cd and/or Pb in soil. Moreover, the level of K application should be considered to obtain an optimal effect with the minimum dosage.  相似文献

Two spectrophotometric assays for protein commonly used in marine research (Coomassie stain, Bradford; alkaline copper, Lowry) and a more recent assay which has not been applied in this field (bicinchoninic acid, Smith) were compared for homogenates of the marine diatom Thalassiosira pseudonona using bovine serum albumin (BSA) as a standard. When homogenates were prepared by precipitating protein with trichloroacetic acid (TCA) and redissolving in 1 N NaOH, the protein content estimated by the Lowry and Smith assays agreed closely, but was consistently 20% higher than that indicated by the Bradford assay. To determine if this difference was due to the choice of a protein standard, protein from T. pseudonana was purified and compared to BSA, bovine gamma-globulin (BGG), and casein. The reactivity of the purified protein (expressed as the slope of the absorbance vs protein concentration curve) did not differ between cultures grown at high or low irradiance. For the Smith and Bradford assays the reactivity of BSA was not significantly different from algal protein, but for the Lowry assay, algal protein was significantly higher in reactivity than BSA. BGG was not significantly different in reactivity from algal protein for the Lowry and Smith assays, but BGG gave significantly lower absorbances than algal protein in the Bradford assay. These results suggest that BSA is a suitable standard for algal protein in the Bradford assays, while BGG is preferable for the Lowry assay. Either protein standard could be used for the Smith assay. Differences in purified algal protein reactivity compared to BSA could not account for the differences among the assays, nor could interference by chlorophyll a. Precipitating protein with TCA prior to analyses gave lower protein than direct analyses of homogenates for the Lowry and Smith assays, but no differences were found for the Bradford assay. As a result, the Lowry and Smith assays indicated up to 60% greater protein than the Bradford if TCA precipitation was not performed. This may be due to removal of free amino acids and small peptides which are less reactive in the Bradford assay. The 20% higher protein found in the Lowry or Smith vs Bradford assays may be due to different assay sensitivity to small peptides or other compounds which are precipitated along with proteins by TCA. Although the Smith assay is substantially simpler to perform than the Lowry, there appear to be no quantitative differences in the results. It remains unclear which spectrophotometric assay is most accurate, but the Bradford assay is faster and simpler, and is less likely to be affected by non-protein compounds found in marine phytoplankton.  相似文献

Spatial variations in the N2O emissions and denitrification potential of riparian buffer strips (RBS) in a polluted river were examined. The river received large pollutant inputs from urban runoff and wastewater discharge, resulting in impaired water quality in the river and downstream reservoir. The potential for nitrogen removal by RBS was evaluated by measuring in situ N2O emission fluxes in static closed chambers and sediment denitrification potentials with acetylene inhibition techniques. The results showed that N2O emission fluxes decreased from the upstream (16.39 μg/(m2·h)) to downstream (0.30 μg/(m2·h)) sites and from the water body to upland sites. The trend in decreasing N2O emission fluxes in the downstream direction was mainly associated with sediment/soil textures (clay loam→sandy soil) and sediment/soil water contents and was also related to the vegetation along the RBS and nutrients in the sediments/soils. The correlation coefficient was highest (r=0.769) between the N2O emission flux and sediment/soil water content. Sediment/soil denitrification potentials under N-amended and ambient conditions were higher (highest 32.86 mg/(kg·h)) for the upstream sites, which were consistent with in situ N2O flux rates.  相似文献

Mussels have been widely used as bioindicators of coastal contamination, and recent reports have demonstrated that metals are accumulated from both the dissolved phase and from ingested food. In the winter and spring of 1995, we examined the influence of the chemical composition of food (protein content, trace element concentrations and ratios in the diatom Thalassiosira pseudomana) on the assimilation of six trace elements (Ag, Am, Cd, Co, Se and Zn) in the mussel Mytilus edulis (L.). Differences of up to 38% in diatom protein content had no major influence on the assimilation of any trace element or carbon. Protein assimilation in M. edulis examined with a 35S radiotracer was also independent of protein content in the diatoms. Similarly, Se assimilation in mussels was not affected by the different Se concentrations in the diatoms. Cd assimilation increased with increasing Cd concentration, presumably due to higher desorption of Cd under acidic conditions typical of the mussel gut. Zn assimilation was inversely related to Zn concentration in the food particles, implying a partial regulation of this metal in the mussels. There was no evidence of any interaction of Cd and Zn in their assimilation by the mussels. These results suggest that mussels are highly responsive, in an element-specific way, to some components of ingested food (e.g., metal concentration), but other food components (such as the biochemical composition of the algae) have little effect on assimilation.  相似文献

Two species of salps, Salpa thompsoni and Ihlea racovitzai, were sampled during three cruises to the Lazarev Sea, Southern Ocean, in summer (December–January) 2005/2006, Autumn (April–May) 2004 and Winter (July–August) 2006. Dry weight, carbon, nitrogen, protein, lipid and carbohydrate contents were measured to characterize the potential value of salps as a food source for predators in the Antarctic ecosystem. Biochemical composition measurements showed that despite having a high percentage of water (~94% of wet weight), both species had relatively high carbon and protein contents in their remaining dry weight (DW). In particular I. racovitzai showed high carbon (up to 22% of DW) and protein (up to 32% of DW) values during all seasons sampled, compared to lower values for S. thompsoni (carbon content only about 15% of the DW, protein content about 10% of the DW). At the same time, carbohydrates (CH) and lipids (Lip) only accounted for a small portion of salp DW in both species (1.4% CH and 3.6% Lip for I. racovitzai; 2.1% CH and 2.9% Lip for S. thompsoni). There was little variability in the biochemical composition of either salp species between the seasons sampled. Both biochemical composition and life cycle characteristics suggest that Antarctic salps, especially I. racovitzai, may be important prey items for both cold and warm-blooded predators in an environment where food is often very scarce.  相似文献

纳米Fe3O4作为一种功能材料,在生物医药、生物靶向材料、微波吸收材料和高梯度磁分离器等方面应用前景广阔,其潜在的生物毒性也备受关注。为研究纳米Fe3O4对生物体可能造成的氧化损伤,以昆明小鼠为受试体,设置5、10、20和40mg·kg-14个染毒组,腹腔注射染毒7d后,测定小鼠肺组织中活性氧(reactive oxygen species,ROS)、还原型谷胱甘肽(glutathione,GSH)和丙二醛(malondialdehyde,MDA)的含量。结果显示,随着纳米Fe3O4染毒剂量的升高,肺组织ROS和MDA含量逐渐上升,GSH含量逐渐降低,各指标均呈一定的剂量-效应关系。剂量≥10mg·kg-1,肺组织ROS含量与对照组相比有显著差异(p<0.05);剂量≥20mg·kg-1,肺组织MDA含量与对照组相比有显著差异(p<0.05);剂量≥40mg·kg-1,肺组织GSH含量与对照组相比有显著差异(p<0.05)。研究表明,较高剂量(≥20mg·kg-1)的纳米Fe3O4颗粒材料会引起小鼠肺细胞的氧化损伤。  相似文献

The Jiushanwai channel is located in the Lucheng district of Wenzhou City, China. Four aquatic plant species (Canna indica, Cyperus alternifolius, Myriophyllum verticillatum, and Lythrum salicaria) were planted on floating-beds constructed in a north-south layout along this channel. Five sites were sampled during spring, summer and autumn. The physiological characteristics (content of chlorophyll and soluble protein, activity of peroxidase and catalase) of the plants and the corresponding water quality were determined and correlations between these indices were analysed. The physiological indices of the plants and water quality changed regularly at the five sampling sites. Myriophyllum verticillatum was the most sensitive species to water quality changes, whereas Lythrum salicaria was least sensitive. Of the physiological indices examined, soluble protein content was most sensitive to water quality changes. The physiological indices of M. verticillatum are deemed to be most suitable to indicate river water quality.  相似文献

为了探明磺胺甲恶唑(sulfamethoxazole,SMZ)的生态毒理效应,通过实验室人工控制毒理实验,研究了低浓度SMZ暴露对小麦体内超氧化物歧化酶(SOD)活性、叶绿素(CHL)和蛋白质(SP)含量的影响。结果显示:染毒7d后,各浓度组小麦叶片的SOD活性均被显著诱导(P<0.01),并且染毒浓度的升高增强了SOD的活性,表明SMZ暴露胁迫下,小麦可启动自身的保护机制以最大限度地减少自由基损伤。而小麦叶片的CHL含量,随着SMZ染毒浓度的增加而逐渐降低。并且当SMZ暴露浓度较高时,小麦叶片的CHL含量被显著抑制(P<0.05))。当SMZ染毒浓度为0.05~0.50mg·L-1时,SP的含量被显著诱导(P<0.01));1.00mg·L-1SMZ对SP的含量产生显著(P<0.01)抑制,这说明SMZ染毒剂量将对蛋白质的合成产生严重影响。综上,SOD的活性变化可反映出SMZ暴露对小麦的污染效应及其生态毒性作用,但将其作为评估SMZ污染暴露的生物标志物有待于进一步深入研究。  相似文献

Measurements of metals in Littorina littorea from clean and contaminated sites in Great Britain confirmed that body concentrations of Ag, Cd and Hg vary according to environmental contamination, while the essential elements Cu and Zn are regulated. Investigation of the cytosolic distribution of metals in L. littorea, using Sephadex G-75 gel-permeation chromatography, revealed important distinctions in the partitioning of elements. In contrast to Ag and Hg, which were mainly associated with high molecular weight ligands, Cd was bound predominantly to an intermediate molecular weight, soluble protein (CdBP-I, apparent mol. wt=20 000). However, although the main function of CdBP-I, both in clean as well as contaminated individuals, was sequestration of Cd, significant quantities of other non-essential metals (Ag, Hg) were similarly bound. Laboratory experiments confirmed the role of CdBP-I in complexing cadmium, and also revealed the induction of a second cadmium-binding protein (CdBP-II) in response to high cadmium levels. The apparent molecular weight (10 000), absorbance characteristics and high (inducible)-SH content of CdBP-II suggest similarities with metallothionein. A dose-related increase in the-SH content of very low molecular weight (>3 000) fractions was also observed in cadmium-exposed L. littorea, although no cadmium was associated with these ligands. A detoxifying role is tentatively proposed for the metal-binding proteins CdBP-I and II in L. littorea. However, some spillover of cadmium to the high molecular weight protein pool was observed in individuals exposed to cadmium in the field and laboratory. The use of L. littorea in biochemical assays of environmental contamination is discussed.  相似文献

氨基甲酸酯类农药广泛应用于农业生产中,其在环境中的残留及其对非靶标生物的毒性作用引起关注。以5种氨基甲酸酯类农药包括残杀威、灭多威、抗蚜威、涕灭威和呋喃丹为研究对象,以蛋白核小球藻为受试生物,应用微板毒性分析法系统测定每种农药及其五元混合物对蛋白核小球藻在不同暴露时间(12、24、48、72和96 h)的生长抑制作用,并同步分析农药对蛋白核小球藻的生理特性如叶绿素含量、蛋白质含量、超氧化物歧化酶(SOD)活性和脂质过氧化物丙二醛(MDA)含量的影响。结果表明,5种农药对蛋白核小球藻的浓度-效应均具有明显的时间依赖性,农药抗蚜威在中低浓度促进绿藻生长,呈现非单调J型浓度-效应曲线(CRC)特征,其余4种农药的CRC呈现经典S型;以半数效应浓度的负对数(p EC50)为毒性大小指标,5种农药在96 h时毒性大小为:呋喃丹(p EC50=3.43)残杀威(p EC50=2.76)抗蚜威(p EC50=2.12)灭多威(p EC50=2.11)涕灭威(p EC50=1.89)。浓度为EC50的5种农药处理后的蛋白核小球藻中叶绿素和蛋白质含量随暴露时间的延长而减少,但不同农药处理的绿藻中叶绿素和蛋白质含量减少率随暴露时间延长变化趋势稍有不同; SOD酶活性随着暴露时间延长逐渐下降,MDA含量逐渐增加,这说明藻细胞受到破坏,脂质过氧化的损害程度超过细胞修复能力,SOD活性被抑制,细胞的抗氧化能力下降,藻细胞内的H2O2不断积累,导致MDA含量升高。五元混合物对蛋白核小球藻的毒性也具有一定的时间依赖性,并表现出刺激作用,即hormesis现象,且混合物毒性与组分浓度比具有良好的线性相关性;暴露于混合物的小球藻均在96 h出现了刺激效应,其叶绿素与蛋白质含量随暴露时间延长不断增加,SOD活性不断升高,MDA含量不断减少;五元混合物均在96 h呈现出拮抗作用,且与混合物的浓度和组分浓度比相关。  相似文献
