There is little information on the indoor environment in hotels. Analysis of fungal DNA by quantitative PCR (qPCR) is a new method which can detect general and specific sequences. Dust was collected through swab sampling of door frames in 69 hotel rooms in 20 countries in Europe and Asia (2007-2009). Five sequences were detected by qPCR: total fungal DNA, Aspergillus and Penicillium DNA (Asp/Pen DNA), Aspergillus versicolor (A. versicolor DNA), Stachybotrys chartarum (S. chartarum DNA) and Streptomyces spp. (Streptomyces DNA). Associations were analysed by multiple linear regression. Total fungal DNA (GM = 1.08 × 10(8) cell equivalents m(-2); GSD = 6.36) and Asp/Pen DNA (GM = 1.79 × 10(7) cell equivalents m(-2); GSD = 10.12) were detected in all rooms. A. versicolor DNA, S. chartarum DNA and Streptomyces DNA were detected in 84%, 28% and 47% of the samples. In total, 20% of the rooms had observed dampness/mould, and 30% had odour. Low latitude (range 1.5-64.2 degrees) was a predictor of Asp/Pen DNA. Seaside location, lack of mechanical ventilation, and dampness or mould were other predictors of total fungal DNA and Asp/Pen DNA. Hotel ranking (Trip Advisor) or self-rated quality of the interior of the hotel room was a predictor of total fungal DNA, A. versicolor DNA and Streptomyces DNA. Odour was a predictor of S. chartarum DNA. In conclusion, fungal DNA in swab samples from hotel rooms was related to latitude, seaside location, ventilation, visible dampness and indoor mould growth. Hotels in tropical areas may have 10-100 times higher levels of common moulds such as Aspergillus and Penicillium species, as compared to a temperate climate zone.  相似文献

Presence of Stachybotrys chartarum in indoor environments has been linked to building-associated disease, however, the causative agents are unknown. Verrucarol (VER) and trichodermol (TRID) are hydrolysis products of some major S. chartarum mycotoxins, i.e. macrocyclic trichothecenes and trichodermin. We optimized gas chromatography-mass spectrometry (GC-MS) methods for detecting VER and TRID in S. chartarum-contaminated indoor environmental samples. Heptafluorobutyryl derivatives of both VER and TRID exhibited little MS fragmentation and gave much higher detection sensitivity (sub-picogram injected onto the GC column), both in GC-MS and GC-MSMS, than trimethylsilyl derivatives. Optimal detection sensitivity and specificity was achieved by combining chemical ionization and negative ion (NICI) detection with MSMS. With this method, VER and TRID were detected in building materials colonized by S. chartarum and TRID was demonstrated in dust settled in the breathing zone in a house where an inner wall was colonized. In summary, we have shown that NICI-GC-MSMS can be used to demonstrate mycotoxins in house dust in S. chartarum-contaminated dwellings.  相似文献

In this study mould damaged materials, including carpet, concrete, gypsum board, insulation, plastic, sand and wood, from 20 different buildings with moisture problems were collected. To study emissions from these materials both conventional methods for sampling, such as collection on Tenax TA, were used as well as complementary methods for sampling a wider spectrum of compounds, such as more volatile VOCs, amines and aldehydes. Analysis was carried out using gas chromatography and high-performance liquid chromatography. Mass spectrometry was used for identification of compounds. Alcohols and ketones were almost exclusively emitted from the materials after they had been wet for a week. Acids were also emitted in large quantities from wet gypsum board and plastic. No primary or secondary amines could be identified, but two tertiary amines, trimethylamine and triethylamine, were emitted from sand contaminated by Bacillus. The most common moulds found were Penicillium and Aspergillus. A multivariate method (partial least squares, PLS) was used to investigate the emission patterns from the materials. Materials with bacterial growth had a different VOC profile to those with only mould growth.  相似文献

Mould in buildings constitutes a threat to health. Present methods to determine the moulds comprise counting of spores or determination of viable moulds which give imprecise measures of total mould cell biomass. Analysis of ergosterol and β-glucan as markers of mould cell biomass is expensive and cumbersome. To evaluate if airborne enzyme activity was related to mould in buildings air samples were taken using an impinger technique or cellulose filters in 386 rooms in 141 buildings. The samples were analysed for the activity of N-acetylhexosaminidase (NAHA) and expressed as enzyme units per m(3) (EU per m(3)). The highest value found in a building was used for the classification of the building and was related to the results from the subsequent technical inspection. In buildings without mould damage, the NAHA activity was generally below 20 EU per m(3). In buildings with mould damage, almost all the buildings had activities above 20 EU per m(3) (specificity 85%). At 30 EU per m(3) the specificity was 100%. Measurements of airborne enzyme activity have a high sensitivity and specificity to identify buildings with mould problems. The method can be used in the investigations of building related symptoms or for home exposure characteristics when investigating diseases such as asthma that can be related to mould exposure.  相似文献

Prolonged moisture on building materials can lead to microbial growth on them. Microbes can emit spores, metabolites and structural parts into the indoor air and thus, cause adverse health effects of people living and working in these buildings. So far, culture methods have been used for assessment of microbial contamination of building materials. In this work, we used quantitative PCR (qPCR) for the detection of selected fungal and bacterial groups in 184 building materials of different types and compared the results with culture-based analysis. Nine either commonly found species, genera or groups of fungi, or those considered as moisture damage indicators, and one bacterial genus, Streptomyces, were determined using qPCR. Fungi and mesophilic actinomycetes were also cultivated using standard media and conditions of the routine analysis. The bacterial genus Streptomyces and the fungal group Penicillium/Aspergillus/Paecilomyces were the most prevalent microbial groups in all building material types, followed by Stachybotrys chartarum and Trichoderma viride/atroviride/koningii. The highest prevalences, concentrations and species diversity was observed on wooden materials. In general, the results of the two methods did not correlate well, since concentrations of fungi and streptomycetes were higher and their occurrence more prevalent when determined by qPCR compared to culture-based results. However, with increasing concentrations, the correlation generally increased. The qPCR assay did not detect Aspergillus versicolor and Acremonium strictum as often as culture.  相似文献

Growth of different microorganisms is often related to dampness in buildings. Both fungi and bacteria produce complicated mixtures of volatile organic compounds that include hydrocarbons, alcohols, ketones, sulfur- and nitrogen-containing compounds etc. Microbially produced substances are one possible explanation of odour problems and negative health effects in buildings affected by microbial growth. A mixture of five fungi, Aspergillus versicolor, Fusarium culmorum, Penicillium chrysogenum, Ulocladium botrytis and Wallemia sebi were grown on three different humid building materials (pinewood, particle board and gypsum board) and on one synthetic medium. Six different sampling methods were used, to be able to collect both non-reactive volatile organic compounds and reactive compounds such as volatile amines, aldehydes and carboxylic acids. Analysis was performed using gas chromatography, high-performance liquid chromatography and ion chromatography, mass spectrometry was used for identification of compounds. The main microbially produced metabolites found on pinewood were ketones (e.g. 2-heptanone) and alcohols (e.g. 2-methyl-1-propanol). Some of these compounds were also found on particle board, gypsum board and the synthetic medium, but there were more differences than similarities between the materials. For example, dimethoxymethane and 1,3,5-trioxepane and some nitrogen containing compounds were found only on particle board. The metabolite production on gypsum board was very low, although some terpenes (e.g. 3-carene) could be identified as fungal metabolites. On all materials, except gypsum board, the emission of aldehydes decreased during microbial growth. No low molecular weight carboxylic acids were identified.  相似文献

We know little about the health consequences of human exposure to microorganisms in the environment. This may largely be due to the lack of universally accepted, sensitive, reproducible, non-discriminatory, and health relevant methods for characterizing the exposure. In this review, some of the different mass spectrometry methods that have been developed for measuring endotoxin and peptidoglycan from bacteria and mycotoxins from moulds in samples from-mainly damp-indoor environments are discussed.  相似文献

以常州市典型城市黑臭水体神童浜为对象,采用小球藻和大型溞作为急性毒性测试手段,对整治前、后神童浜4个点位的上覆水和孔隙水的毒性效应进行了对比和评价。结果表明,整治后神童浜上覆水水质明显改善;上覆水在整治前、后对小球藻生长均无抑制作用,反而呈促进作用;对黑臭水体的整治降低了上覆水和孔隙水的生物毒性,但整治后的上覆水仍存在富营养化问题,且孔隙水对水生生物仍具有一定的毒性风险。提出,利用生物毒性测试能反映出黑臭水体整治前、后的毒性变化,可应用于对黑臭水体整治效果的评价。  相似文献

以常州市典型城市黑臭水体神童滨为对象,利用大型溞-固定沉积物毒性实验,成功地对治理前、后污染河流沉积物毒性变化进行监测,获得较浸出液毒性实验更灵敏的结果。结果表明,治理前污染河流4个采样点沉积物的毒性均为中毒级,治理后,3个采样点的毒性下降到微毒级,1个点位毒性为无毒级。根据河道整治前、后的沉积物对水生生物的综合毒性变化,评价河道治理前的危害程度和治理后的生态安全性,为采取合理的整治方法以达到可持续治理效果提供科学依据。  相似文献

Prior to market entry, new chemical substances are assessed for their risk to human health and the environment. Conventional risk assessment (RA) is limited in scope, i.e. it usually does not cover the entire life cycle of a substance, nor does it take into account sustainability aspects such as the amount of raw materials and energy required to produce the substance. Life cycle assessment (LCA) can provide this pivotal information to support an informed decision on the sustainability of a new substance. Unfortunately, LCA has had little regulatory application up to now. We believe that increasing the focus on combined use of LCA and life cycle-based RA could lead to improved regulatory long-term decisions for marketed chemicals. Inclusion of human biomonitoring could increase the robustness of such decisions even further. In addition, the combined use of the three methods allows a robust search for sustainable alternatives of currently marketed chemicals that have an unfavourable risk profile.  相似文献

Nanoscale materials offer new possibilities for the development of novel remediation and environmental monitoring technologies. Different nanoscale materials have been exploited for preventing environmental degradation and pollutant transformation. However, the rapid self-aggregation of nanoparticles or their association with suspended solids or sediments where they could bioaccumulate supports the need for polymeric coatings to improve mobility, allows faster site cleanups and reduces remediation cost. The ideal material must be able to coordinate different nanomaterials functionalities and exhibit the potential for reusability. We hereby describe two novel environmental applications of nanostructured poly (amic acid)-based (nPAA) materials. In the first application, nPAA was used as both reductant and stabilizer during the in situ chemical reduction of chromium(vi) to chromium(iii). Results showed that Cr(vi) species were rapidly reduced within the concentration range of 10(-1) to 10(2) mM with efficiency of 99.9% at 40 °C in water samples and 90% at 40 °C in soil samples respectively. Furthermore, the presence of PdNPs on the PAA-Au electrode was found to significantly enhance the rate of reduction. In the second application, nPAA membranes were tested as filters to capture, isolate and detect nanosilver. Preliminary results demonstrate the capability of the nPAA membranes to quantitatively capture nanoparticles from suspension and quantify their abundance on the membranes. Silver nanoparticles detection at concentrations near the toxic threshold of silver was also demonstrated.  相似文献

We previously developed a simplified yeast two-hybrid assay of estrogenic activity. In the present study, the optimal conditions for sensitivity and stability of this assay were decided. The assay could determine the estrogenic activity of 4 x 10(-11) mol dm(-1) 17beta-estradiol and also anti-estrogenic activity by using 4-hydroxytamoxifen as a control. The assay was used to test various chemicals suspected of estrogenicity. Many kinds of chemical, including alkylphenols, naphthols, xylenols, methoxychlor, phthalates, and bisphenol-A, showed estrogenic activity, and several, such as 2,5-xylenol and di-iso-octyl phthalate, showed anti-estrogenic but no estrogenic activity. Copresence of two estrogenic chemicals produced additive activity. The assay was also used to test leachate and water samples from the sea-based landfill of Osaka North Port in Japan. Estrogenic activity was detected in leachate from a municipal waste-dumping site, but not in that from a dredged soil-dumping site, which did however cause inhibition of cell growth.  相似文献

The use of low-cost and environmental safety amendments for the in situ immobilization of heavy metals has been investigated as a promising method for contaminated soil remediation. Natural materials and waste products from certain industries with high captive capacity of heavy metals can be obtained and employed. Reduction of extractable metal concentration and phytotoxicity could be evaluated and demonstrated by the feasibility of various amendments in fixing remediation. In this review, an extensive list of references has been compiled to provide a summary of information on a wide range of potentially amendment resources, including organic, inorganic and combined organic-inorganic materials. The assessment based on the economic efficiency and environmental risks brought forth the potential application values and future development directions of this method on solving the soil contamination.  相似文献

优先评估化学品名录对推进有毒有害化学品的管控、保护环境和人类健康具有重要意义。综述了国内外优先评估化学品筛选方法的发展、原则和步骤,总结了筛选评估流程中重点关注的多种危害特性,比较了各国优先评估化学品名录及其在各国/组织的禁止/限制情况,并分析其在我国的生产状况。结果表明,各国家/组织共筛选出1 146种优先评估化学品,有239种在至少2个国家/组织中被列为优先评估化学物质,有208种在我国生产使用,但其中164种尚未列入我国优控化学品名录,可作为下一步风险评估的对象。邻苯二甲酸二(2-乙基己基)酯(DEHP)被所有国家/组织列为优先关注化学品,接下来应严格管控塑料制品中的DEHP,或寻找合适的替代物。研究可为我国优先评估化学品的筛选与环境管理提供参考。  相似文献

One hundred unpackaged rice samples, each weighing 500 g, were randomly collected at retail stores and open markets in the largest rice growing area (Thrace) in Turkey and analysed for mould counts, predominant mould genera, moisture content and mycotoxin levels. Mould counts ranged from 1.0 × 101 to 1.5 × 104 cfu/g in 70 of 100 samples, and the correlation between moisture content and mould count was significant (p????0.05). Aspergillus spp. and Penicillium spp., potential mycotoxin producers, were the dominant moulds. In one area from which samples were collected, the mycotoxin content of rice was found to be positively correlated with moisture content; samples with higher moisture also contained higher numbers of moulds. The levels of total aflatoxins, aflatoxin B1 and ochratoxin A were higher than the maximum tolerable limits (4, 2 and 3 ??g/kg, according to the EC Regulation and the Turkish Food Codex) for 32, 14 and 30 of 100 rice samples, respectively. This is the first comprehensive report of ochratoxin A levels in rice grown in Thrace, Turkey.  相似文献

Dwellings showing a presence of moulds are considered to be unhealthy both by the inhabitants and by sanitary authorities. Although the thresholds of pathogenicity have not yet been established, the toxic, allergic and infectious risk of indoor moulds is better understood today. A study on indoor fungi contamination for 128 dwellings was done between October and May in France. It concerned 69 dwellings, the occupants of which either complained to the sanitary authorities about problems of moulds and humidity or consulted a doctor who related their symptoms to housing conditions. Fifty-nine other dwellings, the occupants of which were healthy, constituted the control group. We present the statistical analysis of questionnaires, which aimed to clarify characteristics of dwellings associated with high concentrations of airborne moulds. Air samples were taken with an impactor in 500 rooms. On visiting dwellings, investigators obtained answers to 25 questions concerning characteristics of inhabitants and living space, as well as the presence of mould indicators. Indoor and outdoor temperature and indoor relative humidity of air measurements were taken. The total concentration of fungi in the air was significantly higher in ground floor apartments versus those on other floors (p = 0.047), in small and highly occupied dwellings (p = 0.03 and 0.003), in dwellings with electric heating (p = 0.04), without a ventilation system (p = 0.003), with water damage (p = 0.003), and finally, in those where the investigator noted an odour of moisture or visible moulds (p < 0.001). The efficacy of the latter criteria in the evaluation of insalubrity is discussed.  相似文献

在田间试验条件下,考察不同钝化材料对农田Cd轻度污染水稻修复效果及稻麦轮作后第二年水稻修复后效。结果表明,在轻度Cd污染农田中,各钝化材料处理均能不同程度降低土壤有效态Cd含量和水稻籽粒中Cd含量。其中,在钝化材料施用当季和稻麦轮作后第二季水稻中修复效果最好的处理为中量纳米材料处理和石灰配施中量纳米材料处理,这两种处理对土壤有效态Cd含量降低率分别为50.94%和47.15%,对水稻籽粒中Cd含量降低率分别为73.74%和69.41%。  相似文献
