Summary.  Antennae of six sympatric bark and ambrosia beetles (Scolytidae), Dendroctonus valens LeConte, Gnathotrichus retusus (LeConte), Hylastes tenuis Eichhoff, Ips mexicanus (Hopkins), Ips plastographus maritimus Lanier, and Pseudohylesinus sericeus (Mannerheim), and two scolytid predators, Enoclerus sphegeus (F.) (Cleridae) and Lascontonus tuberculatus Kraus (Colydiidae), were analyzed by gas chromatographic-electroantennographic detection (GC-EAD) for their responses to synthetic Ips spp. pheromone components, and host and nonhost volatiles. The beetles emerged from cut logs of pitch canker-infected Monterey pine trees, Pinus radiata D. Don. There were significant disparities in EAD response patterns to the hemiterpene and monoterpene alcohol pheromone components that are typically produced by Ips spp. Antennae of I. p. maritimus responded strongly to ( ± )-ipsdienol, ( ± )-ipsenol, amitinol, and lanierone; antennae of I. mexicanus responded strongly to (1S,2S)-(–)-cis-verbenol, with weaker responses to ( ± )-ipsdienol, ( ± )-ipsenol, and amitinol; antennae of H. tenuis responded to (1S, 2R)-(–)-trans-ver-benol, with less pronounced responses to (–)-cis-verbenol and 2-methyl-3-buten-2-ol; and antennae of D. valens, G. retusus, and P. sericeus generally responded to all Ips spp. pheromone components except 2-methyl-3-buten-2-ol (D. valens and G. retusus) and E-myrcenol (G. retusus and P. sericeus). Ips mexicanus responded only to the (–)-enantiomers of ipsenol and ipsdienol, whereas I. p. maritimus responded to (–)-ipsenol, but to both the (+)- and (–)-enantiomers of ipsdienol. The antennae of the two predaceous insects (E. sphegeus and L. tuberculatus) responded to a range of the Ips spp. pheromone components. Host monoterpenes elicited no antennal responses from E. sphegeus, G. retusus, H. tenuis, and I. mexicanus, but several monoterpenes elicited various levels of responses from D. valens and I. p. maritimus antennae. Interestingly, antennae of female D. valens responded to (–), but not (+)-limonene. α- and β-Pinene elicited weak responses from L. tuberculatus antennae. EAD responses to selected nonhost volatiles were almost identical among the six scolytid species, with trans-conophthorin eliciting the strongest response in most cases, followed by three C6- alcohols and two C8-alcohols. The antennal responses by most of these species to linalool or geranylacetone were very weak; (E)-2-hexenal, (Z)-3-hexenyl acetate, and benzyl alcohol elicited almost no response. The response pattern of P. sericeus to nonhost volatiles differed slightly from the rest of the scolytids: a strong response to linalool, weaker response to the C8-alcohols. The two predaceous Coleoptera generally had weak, but detectable, responses to nonhost volatiles, except for a relatively strong response to trans-conophthorin by L. tuberculatus. No notable differences in EAD responses were observed between males and females of the two Ips spp. Our results provide an electrophysiological baseline for future efforts to identify attractive and repellent semiochemicals (aggregation pheromones, host kairomones, or nonhost interruptants) for this guild of scolytids and their key predators that are associated with moribund and pitch canker- infected P. radiata.  相似文献

Summary Ips duplicatus withI. typographus co-inhabiting Norway spruce (Picea abies (L.) Karst.) would benefit from a pheromone blend distinct from that of the larger competitorI. typographus. GC-MS analysis showed thatI. duplicatus males feeding in the host produced ipsdienol (Id),cis-verbenol (cV),trans-verbenol (tV), myrtenol (Mt), andE-myrcenol (EM) and traces of 2-methyl-3-buten-2-ol (MB).I. duplicatus produced Id in approximately racemic form (48.9-54.5% (+)-(S)-isomer). The amounts of Id and EM released over a 9 day period had a maximum of 250 and 5 ng/h/male, respectively, on day 2. Exposure ofI. duplicatus males to myrcene and -pinene resulted in the production of small amounts of Id, cV, tV, Mt, andtrans-pinocarveol, but not of EM. In laboratory bioassays with walking beetles, the pheromone component Id alone was weakly attractive while EM was inactive, but in binary combination with Id strongly synergized attraction. A combination of EM and Id at a release rate equivalent to 100–200 males was more attractive in the field than 70 unmated males in a spruce log. The addition of myrcene ( a suggested pheromone precursor of Id) to Id did not enhance trap caches, while addition of EM increased catches > 10-fold. Subtracting EM from a blend of Id, EM, cV and MB drastically reduced trap catches while subtraction of cV or MB or both had no significant effect. Addition of EM over a wide concentration range to the synthetic pheromone ofI. typographus did not reduce the attraction of females of this species in the laboratory. A two-species pheromone interaction field test releasingI. typographus pheromone components (MB + cV) at 10–1000 male equivalents (ME) andI. duplicatus pheromone (Id + EM) at 0, 10–1000 ME in all possible combinations showed both positive intraspecific dose-response effects and an interspecific inhibition. Higher release rates of EM appeared to inhibitI. typographus, especially males. In a tree colonization model, the response of the two competing species to their respective pheromones show a good separation during the mass-attack with a small initial cross-attraction. It remains to be shown whether either of the two pheromone systems have in fact evolved in the present sympatry, or if they are an incidental effect of ancestry of these phylogenetically distantIps.  相似文献

Summary. In a wind tunnel bioassay the effect of three concentrations of natural extracts of (1) Scots pine wood, Pinus sylvestris, and (2) larval frass on the behavioural response of unmated females and males of the old house borer, Hylotrupes bajulus, was tested and compared to the behavioural effects of the male-produced sex pheromone (3R)-3-hydroxy-2-hexanone. The influence on the behaviour of both sexes was found to be equally significant for the two higher concentrated hexane extracts of wood and larval frass. Therefore several synthetic monoterpenes present in the extracts and ethanol were tested at the two higher concentrations (1:100, 1:1000 vol/vol). Among the higher concentrated monoterpenoid hydrocarbons [(+)-α-pinene, (+)-β-pinene, (+)-limonene], only α-pinene increased the activity, orientation towards scent source and interest towards conspecifics. The tests with higher concentrated ethanol and the oxygenated monoterpenes [(-)-verbenone, (-)-trans-pinocarveol, (+)-terpinen-4-ol, (+)-α-terpineol, (-)-myrtenol] revealed that verbenone is the most effective stimulant for the females, followed by trans-pinocarveol, terpinen-4-ol and α-terpineol. For males, terpinen-4-ol was the only mediator significantly inducing attraction and orientation towards the scent source combined with an interest in conspecifics apparent by fighting or courtship behaviour. Males did not respond to verbenone which is a main compound of larval frass. Myrtenol and ethanol were ineffective in both sexes. In fact behavioural observations suggest that the beetles were repelled by the high dose of myrtenol. Using the ten-fold lower dose of the synthetic monoterpenes (1:1000 vol/vol), all semiochemicals except myrtenol lost activity. Myrtenol, however, induced behavioural responses, like increased activity and orientation towards scent source, only at the low concentration. Based on the results, primary attraction of unmated old house borer is probably mediated by monoterpenes of coniferous wood, while secondary attraction to infested wood would occur in response to volatiles of larval frass. To better understand the attractiveness of host tree, Chinese white pines (Pinus armandi Fr.) to Chinese white pine beetle (Dendroctonus armandi Tsai and Li), the antennal responses of D. armandi to the host volatile, as well as the pure enantiomers and racemates of some monoterpenes, were examined using an electroantennogram (EAG). EAG responses of male and female D. armandi to blended volatiles extracted from the host and some synthetic terpenes (α-phellandrene, (−)-β-pinene, (+)-α-pinene, (−)-α-pinene, (−)-camphene, β-myrcene, (S)-(−)-limonene, (+)-camphene and (R)-(+)-limonene) showed significant variation due to different compound concentrations and sex of the beetles. EAG responses to extracted blended volatiles were significantly greater in females than in males, but the EAG response was not always proportional to the volatile concentration. At lower concentrations, females responded strongly to α-phellandrene and males to (−)-β-pinene, while at higher concentrations, females responded most strongly to α-phellandrene and males to (+)-α-pinene. Females were significantly responsive to (−)-α-pinene, α-phellandrene and (−)-camphene, while males were more responsive to (S)-(−)-limonene, (+)-α-pinene and (R)-(+)-limonene. The EAG responses of the female D. armandi to the volatile oil were significantly higher than that of the males, and the infested pine volatiles could evoke higher EAG response. Most of the test compounds elicited similar responses, which suggested that several of the compounds may be used in combination by D. armandi in habitat and/or host community location at the Qinling forest ecosystem.  相似文献

Summary. The mountain pine beetle, Dendroctonus ponderosae Hopkins, and the pine engraver, Ips pini (Say), often co-exist in lodgepole pine, Pinus contorta var. latifolia Engelmann. Intra- and interspecific semiochemical communication occurs in both species and their complete semiochemical repertoire and precise dynamics of pheromone production have not been elucidated. Porapak-Q extracts of captured volatiles from beetles of each species aerated at different attack phases (freshly emerged, pioneer sex alone in the log and both sexes paired in new galleries), followed by gas chromatographic-electroantennographic detection (GC-EAD) and GC-mass spectroscopic analyses identified 17 compounds (seven compounds common to both species, six present in D. ponderosae and four present in I. pini) that excited the antennae of either or both species. Seven compounds for D. ponderosae and nine for I. pini had not been assessed for behavioural activity. In field trapping experiments, 2-phenylethanol produced by both species inhibited the response of D. ponderosae to its aggregation pheromones. exo- and endo-Brevicomin produced by D. ponderosae significantly decreased the response of I. pini to its aggregation pheromone ipsdienol. Nonanal, a ubiquitous compound found in the volatiles of lodgepole pine, various nonhosts and in both beetle species deterred the response of I. pini to ipsdienol. The occurrence of cis-verbenol, trans-verbenol and verbenone in emergent I. pini, and verbenone and 2-phenylethanol in emergent D. ponderosae suggests that these compounds may inhibit aggregation and induce dispersal following emergence. Termination of aggregation in D. ponderosae appears to depend on the production of frontalin in combination with changes in the relative ratios of verbenone, exo-brevicomin, trans-verbenol and 2-phenylethanol. In I. pini, the cessation of ipsdienol production by males is probably the main factor in terminating aggregation. Received 16 November 1999; accepted 7 August 2000  相似文献

The bark beetles Pityogenes bidentatus and Pityogenes quadridens (Coleoptera, Curculionidae, Scolytinae) are sibling species that feed and reproduce in bark areas on branches of Scotch pine, Pinus sylvestris. To identify aggregation pheromone components of both species, hindguts and head/thoraxes of males and females of both species feeding in hosts were extracted in pentane and analyzed by gas chromatography and mass spectrometry. Hindguts of male P. bidentatus contained grandisol as the major component along with small amounts of (4S)-cis-verbenol and other monoterpenes. Dose–response bioassays in the laboratory showed the components were attractive at 0.2 ng/min to walking beetles from a distance of ≥25 cm. In the field in southern Sweden, grandisol and (4S)-cis-verbenol were weakly attractive alone when released at rates of 0.05 and 0.5 mg/day, respectively, from a slow-rotating trap pair. Catch increased 3.6- to 13-fold when the two components were released together. The male proportion of the catch was 0.8 early in the flight period, declined to 0.5 on the peak flight day, and then declined further during the next 2 weeks to 0.2 on the last day of the flight period. Hindguts of male P. quadridens contained (2S,5R)- and (2S,5S)-chalcogran, as well as (E)-2-(3,3-dimethylcyclohexylidene)ethanol (E-grandlure 2) and E/Z-mixture of 2-(3,3-dimethylcyclohexylidene)acetaldehyde (grandlures 3 and 4), while female hindguts had only a trace amount of chalcogran. Laboratory studies proved E-grandlure 2 is an essential pheromone component for P. quadridens. Field bioassays with a slow-rotating trap pair in which the attractiveness of blends containing various candidate components were compared with that of less complete mixtures, showed that chalcogran and E-grandlure 2 were synergistic aggregation pheromone components of P. quadridens. Field tests also showed that grandisol (from P. bidentatus) reduced attraction of P. quadridens to its aggregation pheromone, whereas E-grandlure 2 and chalcogran (from P. quadridens) reduced response of P. bidentatus to its aggregation pheromone. Our results suggest that aggregation pheromone components from males of each species not only attract conspecifics but also aid individuals in avoiding interspecific mating and competition for food and spatial resources within the bark phloem layer.  相似文献

Photosynthetic responses to irradiance by the photosymbionts of the two Red Sea sponges Theonella swinhoei (Gray) and Clionavastifica (Hancock) growing under dim light conditions were measured in situ (in September 1997) using a newly developed underwater pulse amplitude modulated (PAM) fluorometer. Relative rates of photosynthetic electron transport (ETR) were calculated as the effective quantum yield of photosystem II (Y ) multiplied with the photosynthetic photon flux (PPF). Photosynthesis versus irradiance (P-I ) curves, obtained within minutes, showed that individual specimens of both sponges, growing under very low light conditions, feature lower light saturation points as well as lower maximal ETRs than individuals growing under higher light. Evaluations of such curves using low irradiances of the actinic light source (20 to 130 μmol photons m−2 s−1) showed a general decrease in Y, with a shoulder from the lowest irradiance applied till 20 to 30 μmol photons m−2 s−1. Point measurements yielded ETRs close to what could be estimated from the P-I curves. These point measurements also revealed good correlations between the diurnally changing ambient irradiances (1 to 50 μmol photons m−2 s−1) and average ETR values for both species. Further analysis showed that although Y values varied considerably between the different point measurements, they did not decrease significantly with light under these very low irradiances. Therefore, PPF rather than Y seems to determine the in situ diel photosynthetic performance at the low ambient irradiances experienced by these sponges. Primary production at Antarctic coastal sites is contributed from sea ice algae, phytoplankton and benthic algae. Oxygen microelectrodes were used to estimate sea ice and benthic primary production at several sites around Casey, a coastal area in eastern Antarctica. Maximum oxygen export from sea ice was 0.95 mmol O2 m−2 h−1 (~11.7 mg C m−2 h−1) while from the sediment it was 6.08 mmol O2 m−2 h−1 (~70.8 mg C m−2 h−1). When the ice was present O2 export from the benthos was either low or negative. Sea ice algae assimilation rates were up to 3.77 mg C (mg Chl-a)−1 h−1 while those from the benthos were up to 1.53 mg C (mg Chl-a)−1 h−1. The contribution of the major components of primary productivity was assessed using fluorometric techniques. When the ice was present approximately 55–65% of total daily primary production occurred in the sea ice with the remainder unequally partitioned between the sediment and the water column. When the ice was absent, the benthos contributed nearly 90% of the primary production.  相似文献

Bacterial abundance, production, and extracellular enzyme activity were determined in the shallow water column, in the epiphytic community of Thalassia testudinum, and at the sediment surface along with total carbon, nitrogen, and phosphorus in Florida Bay, a subtropical seagrass estuary. Data were statistically reduced by principle components analysis (PCA) and multidimensional scaling and related to T. testudinum leaf total phosphorus content and phytoplankton biomass. Each zone (i.e., pelagic, epiphytic, and surface sediment community) was significantly dissimilar to each other (Global R = 0.65). Pelagic aminopeptidase and sum of carbon hydrolytic enzyme (esterase, peptidase, and α- and β-glucosidase) activities ranged from 8 to 284 mg N m−2 day−1 and 113–1,671 mg C m−2 day−1, respectively, and were 1–3 orders of magnitude higher than epiphytic and sediment surface activities. Due to the phosphorus-limited nature of Florida Bay, alkaline phosphatase activity was similar between pelagic (51–710 mg P m−2 day−1) and sediment (77–224 mg P m−2 day−1) zones but lower in the epiphytes (1.1–5.2 mg P m−2 day−1). Total (and/or organic) C (111–311 g C m−2), N (9.4–27.2 g N m−2), and P (212–1,623 mg P m−2) content were the highest in the sediment surface and typically the lowest in the seagrass epiphytes, ranging from 0.6 to 8.7 g C m−2, 0.02–0.99 g N m−2, and 0.5–43.5 mg P m−2. Unlike nutrient content and enzyme activities, bacterial production was highest in the epiphytes (8.0–235.1 mg C m−2 day−1) and sediment surface (11.5–233.2 mg C m−2 day−1) and low in the water column (1.6–85.6 mg C m−2 day−1). At an assumed 50% bacterial growth efficiency, for example, extracellular enzyme hydrolysis could supply 1.8 and 69% of epiphytic and sediment bacteria carbon demand, respectively, while pelagic bacteria could fulfill their carbon demand completely by enzyme-hydrolyzable organic matter. Similarly, previously measured T. testudinum extracellular photosynthetic carbon exudation rates could not satisfy epiphytic and sediment surface bacterial carbon demand, suggesting that epiphytic algae and microphytobenthos might provide usable substrates to support high benthic bacterial production rates. PCA revealed that T. testudinum nutrient content was related positively to epiphytic nutrient content and carbon hydrolase activity in the sediment, but unrelated to pelagic variables. Phytoplankton biomass correlated positively with all pelagic components and sediment aminopeptidase activity but negatively with epiphytic alkaline phosphatase activity. In conclusion, seagrass production and nutrient content was unrelated to pelagic bacteria activity, but did influence extracellular enzyme hydrolysis at the sediment surface and in the epiphytes. This study suggests that seagrass-derived organic matter is of secondary importance in Florida Bay and that bacteria rely primarily on algal/cyanobacteria production. Pelagic bacteria seem coupled to phytoplankton, while the benthic community appears supported by epiphytic and/or microphytobenthos production.  相似文献

The grey corn weevil, Tanymecus (Episomecus) dilaticollis, is an economically important polyphagous pest in Eastern and Central Europe. The present research is the first published electrophysiological study of olfactory sensitivity of adult T. dilaticollis to plant volatiles. The electrophysiological responses of male and female antennae were recorded to 20 synthetic volatile organic compounds (VOCs), previously identified to be released from the preferred host plants, maize and sunflower. Antennae of both sexes responded to a wide range of volatiles from different chemical classes: fatty acid derivatives, aromatic compounds and terpenoids. Two green leaf volatiles, (E)-2-hexen-1-ol and (Z)-3-hexen-1-ol, and one terpenoid, (±)-linalool, elicited the strongest electrophysiological responses by male and female antennae. Relatively high electrophysiological activity by T. dilaticollis antennae was also recorded to benzaldehyde, eugenol and caryophyllene. In the field, (E)-2-hexen-1-ol and (Z)-3-hexen-1-ol, when presented individually, caught significantly more male and female T. dilaticollis than unbaited controls. These compounds could be developed into an attractive trap for the detection and monitoring of T. dilaticollis.  相似文献

From May 2002 to October 2003, a fortnightly sampling programme was conducted in a restricted macrotidal ecosystem in the English Channel, the Baie des Veys (France). Three sets of data were obtained: (1) physico-chemical parameters, (2) phytoplankton community structure illustrated by species composition, biovolume and diversity, and (3) primary production and photosynthetic parameters via P versus E curves. The aim of this study was to investigate the temporal variations of primary production and photosynthetic parameters in this bay and to highlight the potential links with phytoplankton community structure. The highest level of daily depth-integrated primary production Pz (0.02–1.43 g C m−2 d−1) and the highest maximum photosynthetic rate P B max (0.39–8.48 mg C mg chl a −1 h−1) and maximum light utilization coefficient αB [0.002–0.119 mg C mg chl a −1 h−1 (μmol photons m−2 s−1)] were measured from July to September. Species succession was determined based on biomass data obtained from cell density and biovolume measurements. The bay was dominated by 11 diatoms throughout the year. However, a Phaeocystis globosa bloom (up to 25 mg chl a m−3, 2.5 × 106 cells l−1) was observed each year during the spring diatom bloom, but timing and intensity varied interannually. Annual variation of primary production was due to nutrient limitation, light climate and water temperature. The seasonal pattern of microalgal succession, with regular changes in composition, biovolume and diversity, influenced the physico-chemical and biological characteristics of the environment (especially nutrient stocks in the bay) and thus primary production. Consequently, investigation of phytoplankton community structure is important for developing the understanding of ecosystem functioning, as it plays a major role in the dynamics of primary production.  相似文献

 Short-term effects of temperature and irradiance on oxygenic photosynthesis and O2 consumption in a hypersaline cyanobacterial mat were investigated with O2 microsensors in a laboratory. The effect of temperature on O2 fluxes across the mat–water interface was studied in the dark and at a saturating high surface irradiance (2162 μmol photons m−2 s−1) in the temperature range from 15 to 45 °C. Areal rates of dark O2 consumption increased almost linearly with temperature. The apparent activation energy of 18 kJ mol−1 and the corresponding Q 10 value (25 to 35 °C) of 1.3 indicated a relative low temperature dependence of dark O2 consumption due to mass transfer limitations imposed by the diffusive boundary layer at all temperatures. Areal rates of net photosynthesis increased with temperature up to 40 °C and exhibited a Q 10 value (20 to 30 °C) of 2.8. Both O2 dynamics and rates of gross photosynthesis at the mat surface increased with temperature up to 40 °C, with the most pronounced increase of gross photosynthesis at the mat surface between 25 and 35 °C (Q 10 of 3.1). In another mat sample, measurements at increasing surface irradiances (0 to 2319 μmol photons m−2 s−1) were performed at 25, 33 (the in situ temperature) and 40 °C. At all temperatures, areal rates of gross photosynthesis saturated with no significant reduction due to photoinhibition at high irradiances. The initial slope and the onset of saturation (E k = 148 to 185 μmol photons m−2 s−1) estimated from P versus E d curves showed no clear trend with temperature, while maximal photosynthesis increased with temperature. Gross photosynthesis was stimulated by temperature at each irradiance except at the lowest irradiance of 54 μmol photons m−2 s−1, where oxygenic gross photosynthesis and also the thickness of the photic zone was significantly reduced at 40 °C. The compensation irradiance increased with temperature, from 32 μmol photons m−2 s−1 at 25 °C to 77 μmol photons m−2 s−1 at 40 °C, due to increased rates of O2 consumption relative to gross photosynthesis. Areal rates of O2 consumption in the illuminated mat were higher than dark O2 consumption at corresponding temperatures, due to an increasing O2 consumption in the photic zone with increasing irradiance. While it is known that Antarctic sea ice biomass and productivity are highly variable over small spatial and temporal scales, there have been very few measurements from eastern Antarctic. Here we attempt to quantify the biomass and productivity and relate patterns of variability to sea ice latitude ice thickness and vertical distribution. Sea ice algal biomass in spring in 2002, 2003 and 2004 was low, in the range 0.01–8.41 mg Chl a m−2, with a mean and standard deviation of 2.08 ± 1.74 mg Chl a m−2 (n = 199). An increased concentration of algae at the bottom of the ice was most pronounced in thicker ice. There was little evidence to suggest that there was a gradient of biomass distribution with latitude. Maximum in situ production in 2002 was approximately 2.6 mg C m−2 h−1 with assimilation numbers of 0.73 mg C (mg Chl a)−1 h−1. Assimilation numbers determined by the 14C incubations in 2002 varied between 0.031 and 0.457 mg C (mg Chl a)−1 h−1. Maximum fluorescence quantum yields of the incubated ice samples in 2002 were 0.470 ± 0.041 with E k indices between 19 and 44 μmol photons m−2 s−1. These findings are consistent with the shade-adapted character of ice algal communities. In 2004 maximum in situ production was 5.9 mg C m−2 h−1 with an assimilation number of 5.4 mg C (mg Chl a)−1 h−1. Sea ice biomass increased with ice thickness but showed no correlation with latitude or the time the ice was collected. Forty-four percent of the biomass was located in bottom communities and these were more commonly found in thicker ice. Surface communities were uncommon.  相似文献

The diatom Cylindrotheca closterium was exposed to transient light- and osmotic conditions as occur during its tidal emersion. The objective was to analyze how this simulated emersion contributes to the production of active oxygen species (AOS) and via this, to oxidative cell damage. Light- and salinity conditions were varied in factorial combination: low light (no UVB) or high light (unweighted UVB-dose rates of respectively 0.01; 0.07; 0.24; 1.03 W m−2) at normal (30 psu) or high salinity (60 psu). UVB (0.01–0.24 W m−2) and high salinity had a significant, negative effect on the photosynthetic efficiencies ΔF/F m’ (steady-state quantum yield) and F v/F m (maximum yield). UVB at 1.03 W m−2 (15 kJ m−2 d−1) almost arrested electron transport. At ecologically relevant UVB levels, i.e. below 0.24 W m−2 (≈3.4 kJ m−2 d−1) with UVB:PAR<0.4:100 (PAR photosynthetically active radiation) only dynamic photoinhibition was observed (protection via heat dissipation). Non-photochemical quenching was positively correlated with the de-epoxidation of diadinoxanthin (DD) to diatoxanthin (DT). A decreasing ratio DT/(DD+DT) after 4 h of UVB at >0.07 W m−2 and at 60 psu indicated a reversal of the diatom xanthophyll cycle (diminished photoprotection) which may be caused by an enhanced AOS production. Oxidative stress and -damage to C. closterium cells were assessed applying fluorescent indicator dyes, via confocal microscopy and quantitative image analysis. AOS production rates (cellular DCF fluorescence) were stimulated by UV, and were ~50% higher at 60 psu. AOS production decreased with an increasing pre-exposure (0–4 h) to normal UVB (0.24 W m−2), which indicated a stimulation of the antioxidative defence. Non-protein thiols (indicator CMF) and glutathione pools (HPLC-analyzed) decreased with UVB-dose rates (0.01–0.24 W m−2), most likely due to AOS-mediated thiol oxidation. Hypersalinity (60 psu) and UVB (0.01–0.24 W m−2) caused membrane depolarization (dye DIBAC4(3)) and phospholipid hydrolysis (phospholipase A2 dye: bis-BODIPY FL-C11-PC). AOS production may have diminished the membrane polarity, and peroxidized the membrane lipids (HPLC-analyzed malondialdehyde) which enhanced PLA2 activity. The dyes indicated an increased oxidative (lipid) damage at a 15% inhibition of photosynthesis in this diatom, at UVB levels and salinities that can be expected in situ during its periodic tidal emersion.  相似文献

Artemia franciscana was grown on Isochrysis galbana Green (clone T. Iso) at saturated food concentrations (13 to 20 mg C l−1) for 11 d at 26 to 28 °C, and 34 ppt salinity. Three groups of brine shrimp were used in the feeding experiments: metanauplius III and IV (Group 1), post-metanauplius II and III (Group 2) and post-metanauplius VIII (Group 3), corresponding to 4-, 7- and 11-d-old animals, respectively. The ingestion rate, clearance rate and carbon balance were estimated for these stages at different concentrations of 14C-labeled I. galbana ranging from 0.05 to 30 mg C l−1. The handling time of algae was determined for all three groups. The ingestion rate (I, ng C ind−1 h−1) increased as a function of animal size and food concentration. In all three groups, the ingestion rate increased to a maximum level (I max) and remained constant at food concentrations ≥10 mg C l−1 (saturated food concentrations). The clearance rate (CR, μl ind−1 h−1) increased with increasing food concentration up to a maximum rate (CR max), after which it decreased for even higher food concentrations. The functional response of A. franciscana was most consistent with Holling's Type 3 functional response curve (sigmoidal model), which for the two oldest groups (Group 2 and 3) differed significantly from a Type 2 response (p < 0.05). The gut passage time for the three groups of A. franciscana, feeding on saturated food concentration (20 mg C l−1), varied between 24 and 29 min. As the nauplii developed to pre-adult stage the handling time of the algae increased as a function of animal size. The assimilation rate (ng C ind−1 h−1) in the youngest stages (Group 1 and 2) increased with increasing food concentrations, reaching a maximum level close to 10 mg C l−1. At higher food concentrations the assimilation rate decreased, and the proportions of defecated carbon increased, reaching 60 to 68% in the post-metanauplius stages (Group 3). The assimilation efficiency (%) was high at the lowest food concentrations in all three groups (89 to 64%). The effect of irradiance, prey concentration and pH on the growth and grazing responses of the mixotrophic prymnesiophyte Chrysochromulina ericina under N-and P-replete conditions was studied using the pedinophyte Marsupiomonas pelliculata as prey. The two organisms were inoculated in monocultures and in mixed cultures at different predator: prey ratios at three irradiances and allowed to grow for 4–7 days. All cultures were non-axenic. Algal densities and pH were monitored throughout the experiments and growth and grazing rates were measured. An increase in growth of C. ericina cultures at irradiances of 25 and 70 μmol photons m−2 s−1 was observed after the addition of prey, while growth of C. ericina cultures at the high irradiance (150 μmol photons m−2 s−1) was unaffected by the addition of prey. However, although the growth of C. ericina increased at low irradiance (25 μmol photons m−2 s−1), it did not reach the same level as monocultures at the high irradiance (150 μmol photons m−2 s−1), suggesting that phagotrophy can only partly replace photosynthesis in C. ericina. Maximum growth rates of C. ericina at irradiances of 25 and 70 μmol photons m−2 s−1 were obtained at concentrations of > 0.15–0.3×105 M. pelliculata ml−1, corresponding to 50–100 μg C 1−1. Ingestion of M. pelliculata cells by C. ericina did not generally follow Michaelis—Menten kinetics. Deviation from the expected saturation kinetics was especially pronounced at irradiances of 70 and 150 μmol photons m−2 s−1. At these irradiances ingestion of M. pelliculata cells by C. ericina decreased at high concentrations of M. pelliculata, indicating an increased uptake of bacterial prey in these cultures. The growth rate of C. ericina was affected in both monocultures and in mixed cultures when pH increased above 8.6, and growth stopped around pH 9. The prey alga M. pelliculata tolerated high pH better and, consequently, took over in the mixed cultures when pH exceeded 9. An investigation to characterize the causes of Pinna nobilis population structure in Moraira bay (Western Mediterranean) was developed. Individuals of two areas of the same Posidonia meadow, located at different depths (A1, −13 and A2, −6 m), were inventoried, tagged, their positions accurately recorded and monitored from July 1997 to July 2002. On each area, different aspects of population demography were studied (i.e. spatial distribution, size structure, displacement evidences, mortality, growth and shell orientation). A comparison between both groups of individuals was carried out, finding important differences between them. In A1, the individuals were more aggregated and mean and maximum size were higher (A1, 10.3 and A2, 6 individuals/100 m2; A1, x = 47.2 ± 9.9; A2, x = 29.8 ± 7.4 cm, P < 0.001, respectively). In A2, growth rate and mortality were higher, the latter concentrated on the largest individuals, in contrast to A1, where the smallest individuals had the higher mortality rate [A1, L = 56.03(1 − e−0.17t ); A2, L = 37.59(1 − e−0.40t ), P < 0.001; mean annual mortality A1: 32 dead individuals out of 135, 23.7% and A2: 16 dead individuals out of 36, 44.4%, and total mortality coefficients (z), z A1(−30) = 0.28, z A1(31–45) = 0.05, z A1(46−) = 0.08; z A2(−30) = 0.15, z A2(31–45) = 0.25]. A common shell orientation N–S, coincident with the maximum shore exposure, was observed in A2. Spatial distribution in both areas showed not enough evidence to discard a random distribution of the individuals, despite the greater aggregation on the deeper area (A1) (A1, χ 2 = 0.41, df = 3, P > 0.5, A2, χ 2 = 0.98, df = 2 and 0.3 < P < 0.5). The obtained results have demonstrated that the depth-related size segregation usually shown by P. nobilis is mainly caused by differences in mortality and growth among individuals located at different depths, rather than by the active displacement of individuals previously reported in the literature. Furthermore, dwarf individuals are observed in shallower levels and as a consequence, the relationship between size and age are not comparable even among groups of individuals inhabiting the same meadow at different depths. Summary. Across North America, populations of the pine engraver, Ips pini, differ in their expressed ratios of the two enantiomers of ipsdienol, the main component of its aggregation pheromone. We confirm previous studies, showing that the percentage of (+)-ipsdienol ranged from approximately 40 to 70% for New York (NY) males and less then 5% for California (CA) males. We performed line crosses including the F1, F2, and backcross generations between these populations. These line crosses showed that most F1 hybrids produced an intermediate enantiomeric blend of ipsdienol that was closer to the CA blend, with a frequency distribution peak near 15% (+)-ipsdienol. There was also strong segregation to either parental type in the F2 and backcross generations, but not in a pattern that could be clearly described by a single autosomal locus. There was also an X-linked effect that caused some individuals in the F1 to have phenotypes more characteristic of NY populations. Generation means analysis confirms this X-linked effect, and also suggests a complicated autosomal dominance by dominance epistatic interaction. Despite the appearance of segregation, these results suggest a more complicated system of pheromone blend control than the single major gene previously found between divergent lines for high and low (+)-ipsdienol blends from the hybrid zone in British Columbia.   相似文献

The central California coast is a highly productive, biodiverse region that is frequently affected by the toxin-producing dinoflagellate Alexandrium catenella. Despite the consistent presence of A. catenella along our coast, very little is known about the movement of its toxins through local marine food webs. In the present study, we investigated 13 species of commercial finfish and rock crabs harvested in Monterey Bay, California for the presence of paralytic shellfish toxins (PSTs) and compared them to the presence of A. catenella and PSTs in sentinel shellfish over a 3-year period. Between 2003 and 2005, A. catenella was noted in 55% of surface water samples (n = 307) and reached a maximum concentration of 17,387 cells L−1 at our nearshore site in Monterey Bay. Peak cell densities occurred in the month of July and were associated with elevated shellfish toxicity in the summers of 2004 and 2005. When A. catenella was present, particulate PSTs were detected 71% of the time and reached a maximum concentration of 962 ng STXeq L−1. Of the 13 species tested, we frequently detected PSTs in Pacific sardines (Sardinops sagax; maximum 250 μg STXeq 100 g−1), northern anchovies (Engraulis mordax; maximum 23.2 μg STXeq 100 g−1), brown rock crabs (Cancer antennarius; maximum 49.3 μg STXeq 100 g−1) and red rock crabs (C. productus; 23.8 μg STXeq 100 g−1). PSTs were also present in one sample of Pacific herring (Clupea pallas; 13.3 μg STXeq 100 g−1) and one sample of English sole (Pleuronectes vetulus; 4.5 μg STXeq 100 g−1), and not detected in seven other species of flatfish tested. The presence of PSTs in several of these organisms reveals that toxins produced by A. catenella are more prevalent in California food webs than previously thought and also indicates potential routes of toxin transfer to higher trophic levels.
