The accumulation of cadmium chloride by a living, by an ethanol or potassium cyanide treated marine pseudomonad was studied. The metal uptake by the living strain was linear during the experiment and the amount of accumulated metal was high. The uptake by the treated strain was very fast during the first hours, but the amount of accumulated cadmium was lower.  相似文献

Enumeration of viruses or bacteria on liquid culture media by the Most Probable Number technique (M.P.N.) never gives satistically acceptable results. For accepting or rejecting a count, the authors have established probability tables (95% and 99%) according to microbial densities. The distribution between the aqueous and solid phase of 17 beta-estradiol in the ng-range in waste water was investigated by spiking experiments with a radio-labeled hormone. The distribution was measured by liquid scintillation counting. The major part of 17 beta-estradiol remained in the aqueous phase and did not adsorb to the solids. The fraction in the aqueous phase could be enriched by a conventional solid phase extraction with a recovery rate of up to 100%. Losses were negligible and the compound did not adsorb to the surfaces of glass bottles.  相似文献

The viral contamination of sea-water at different depths and in the sediments in the area near the discharge of treated domestic sewage, was investigated. The isolation frequency of viral particles was low: on 19 samples of sea-water, 4 are positive and on 6 samples of sediment, 1 is positive. Viral concentrations were not substantial: they varied from 2,9 to 5,9 Most Probable Number Cytopathic Unit (M.P.N.C.U.) in 500 ml of sea-water or in 500 g of sediment. No homogeneous distribution of viral particles in sea-water was observed.  相似文献

1-Hydroxypyrene (1-HP) is a carcinogenic and slightly water-soluble polycyclic aromatic hydrocarbon. Ecotoxicity and mutagenicity of 1-HP and its photoproducts, and the effect of Mn2+ and Cu2+ on their mutagenicity were measured with microbial assay in this study. The assay includes spread plate counting, direct counting, microbial mineralization of 14C-UL-D-glucose and Mutatox Test. At the concentration examined (0.8 microM), the photoproducts (after 1.5 h solar irradiation) of 1-HP inhibited microbial glucose mineralization activity (by 64%) after microbial assemblages of a local reservoir site were exposed for 1 day. However, heterotrophic bacteria were able to utilize 1-HP photoproducts as the growth substrates and increase viability counts by up to 4.75-folds. 1-HP exhibited positive response to Mutatox Test in both direct medium and S-9 medium, with the lowest observable effective concentration of 0.625 microM in the test with direct medium. After photolysis, 1-HP decreased its mutagenicity. Mn2+ (312.5 microM-5 mM) and Cu2+ (6.25-100 microM) themselves are not mutagenic. However, addition of the metal ions before or after photolysis modifies the light readings of 1-HP during the test. Therefore, presence of metal ions could affect the genotoxicity of 1-HP in aquatic environments, depending on timing of the addition.  相似文献

The technique of the M.P.N. involving with a large number n of inoculum per dilution (e.g.96) gives a precise estimation of the microbial density. As no table can be used when n is greater than 10, the authors are proposing a computer program to calculate the M.P.N. with its confidence limits. They consider the most common case where number of dilution is greater than 3, dilution factor is 10 or other, and number of inoculated tubes (or wells) per dilution is different from one dilution to another.  相似文献

Abstract

Radiolabelled end‐use mixtures of glyphosate with and without a cationic surfactant (Ethomeen® T/25) and an organosilicone surfactant (Silwet® L‐77) were applied onto trembling aspen (Populus tremuloids Michx.) leaves at the rate of 1.0 kg of AE (acid equivalent) in 35 L/ha area of foliage. A 5‐mm rainfall with an intensity of 10 mm/h was applied at intervals of 0.5, 8, 24, 36, 48, 72, and 96 h after treatment. Glyphosate washoff was determined by liquid scintillation counting of radioactivity in the rain‐washing. At 36 h post‐treatment, both the adjuvants significantly reduced glyphosate washoff (Ethomeen by 69.6% and Silwet by 59.7%) from foliage, compared to the washoff (82.6%) when Vision alone was applied without the adjuvants. Results on the rate of plant growth indicated that with a rain‐free period of 8 h or more, the growth of most seedlings was stunted within 1 or 2 d. Percentage of foliar browning 20 d after treatment with rain‐free period of 8 to 48 h ranged from 8 to 80% for Vision alone, 75 to 100% for Vision with Ethomeen, and 85 to 100% for Vision with Silwet, respectively. Physical properties of the end‐use mixtures were measured with and without the two adjuvants to examine droplet spreading and drying rates in relation glyphosate rainfastness. The Silwet adjuvant lowered the surface tension of the end‐use mixture, but Ethomeen did not. Droplets containing Silwet were spread more than those containing Ethomeen. However, the greater area of contact caused by Silwet did not contribute to a significant increase in the translocation rate of glyphosate into untreated parts of the seedlings, and showed no relationship with rainfastness of glyphosate deposits on trembling aspen.  相似文献

A comparison of the quantification of a specific microbial group in activated sludge by fluorescent in-situ hybridization, coupled with either direct microscopic counting or flow cytometry, was performed using an enhanced-biological-phosphorus-removal, sequencing-batch reactor. The population dynamics of Candidatus Accumulibacter phosphatis (Cand. A. phosphatis) was evaluated during two separate runs of the reactor. With the operational conditions used, Cand. A. phosphatis was enriched until a failure in the pH controller eliminated its ecological advantage. As a result, the comparison of quantification techniques included Cand. A. phosphatis concentrations as low as 11% and as high as 96% of the total cells in the samples. The analysis demonstrated that, regardless of the particular limitations of each technique, both provided similar results when the activated-sludge flocs were easily dispersed. However, when the activated-sludge samples contained flocs that were difficult to disperse, flow cytometry failed to provide quantitative results.  相似文献

从污染环境分离和筛选得到了9株PAHs降解菌,以其为基本菌种,构建高效修复PAHs复合污染体系的菌群(D)。采用平板稀释涂布计数法对降解体系中菌群(D)的动态结构进行了解析,数据显示菌群内的微生物在降解过程中能相互协作,发挥稳定且高效的降解作用。实验进一步研究了菌群D对单一PAH和混合PAHs的降解特性,结果表明,无论对单一PAH还是混合PAHs,菌群D均具有较强的降解能力。当降解历时6 d,菌群D能使40 mg/L的单一PAH平均降解85.8%,使60 mg/L的混合PAHs平均去除89.4%。菌群D在多环芳烃复合污染体系的生物修复方面具有潜在的应用价值。  相似文献

The body of information presented in this paper is directed to those individuals concerned with the emission of smoke from diesel engines. A series of tests was performed on a Petter Type AA1 diesel engine using barium smoke suppressant additives. An Andersen cascade type sampler was used to collect samples and thus study the effect of the additive upon the total smoke emission, smoke size distribution and smoke composition. For a portion of the tests radioactive barium (133Ba) and scintillation counting techniques were used.

Type 1-D fuel was used for all tests. Baseline tests were performed to determine the smoke emission characteristics using nontreated fuel, 0.5% by volume of Bryton additive, and 0.75% by volume of Lubrizol additive. For the radioactive additive, tests were conducted using dosages of 0.13%, 0.26%, 0.41%, and 0.82% by volume. The results of these tests revealed that the additive does not alter the particle size distribution, but the total mass emission from the engine is reduced by approximately 50% at the dosages tested. The dosage level of the additive does not influence the distribution of barium within the exhaust particles. Also it seems that the soluble percentage (in 0.1 N HCI) is influenced by the dosage level. The results of the solubility tests at low dosage levels seem to imply that the smoke inhibition mechanism occurs within the fuel droplet.  相似文献

TSI Inc. (Shoreview, MN) has introduced three new water-based condensation particle counters (WCPCS) that were designed to detect airborne particles larger than 2.5 nm (model 3786), 5 nm (model 3785), and either 10 or 20 nm (model 3782). These WCPCs are well suited for real-time, environmental monitoring of number concentration of airborne ultrafine particles. Their unique design incorporates the use of water as the working fluid instead of alcohol. Water is odor free, readily available, and eliminates the problem of water condensation and absorption into alcohol working fluids during operation in humid environments. In this study, the performance of three TSI WCPCs was characterized for several aerosol compositions, including sucrose, salt (NaCl), dioctyl sebacate (DOS), dioctyl phthalate (DOP), emery oil (poly-alpha-olefin), silver, impurity residue particles, and ambient aerosol particles. All particles were size selected using a nano differential mobility analyzer (nano-DMA; model 3085, TSI Inc.) to create monodisperse challenge aerosols. The challenge aerosol was mixed uniformly with clean makeup flow and split into a WCPC and a reference instrument to determine the counting efficiency of the WCPC. For the model 3785 WCPC, the D50 (i.e., the particle diameter with 50% counting efficiency) was determined to be 3.1 nm for salt particles, 4.7 nm for sucrose and ambient particles, 5.6 nm for silver particles, and >50 nm for ultrapure oil particles. The sensitivity to oil droplets increased dramatically (D50 < 10 nm) when the oil was slightly contaminated. The D50 of model 3786 ultrafine water-based CPC (UWCPC) was 2.4 nm for impurity residue particles. The D50 of the model 3782 WCPC was 10.8 (with a nominal setting of 10 nm) or 19.8 nm (with a nominal setting of 20 nm) for sucrose particles. All three WCPCs have response times of less than 2 or 3 sec and are therefore able to detect fast-changing events.  相似文献

利用选择性培养基对氨氧化菌群进行了连续驯化,得到了氨氮去除效率稳定的氨氧化菌群。采用平板菌落计数法结合聚合酶链式反应-变性梯度凝胶电泳(PCR-DGGE)技术考察了氨氧化菌群在连续传代过程中数量及群落结构的动态变化,并考察了pH值、碳源(HCO3-)浓度和氨氮浓度等因素对氨氧化菌群去除氨氮效率的影响。结果表明,通过连续驯化,氨氧化菌的比例由最初的1.8%提高到了31.3%。在碳源浓度为1.5 mg/L,NH4+-N初始浓度为200 mg/L,pH值为8的条件下,菌群对氨氮的去除率达99%以上。  相似文献

Using a combination of radioisotopic counting and chromatographic detection techniques, the kinetics and metabolic fate of deoxynivalenol (DON) in plasma, urine and bile were studied in lactating sheep, as was the transmission of residues to milk. Following intravenous administration, the plasma clearance of 14C-DON-derived radioactivity was rapid and followed a tri-phasic decay curve comprised of a bi-exponential decrease in DON (rapid distribution phase, t1/2 alpha = 16.2 min; slower elimination phase t1/2 beta = 66.5 min) and the formation and elimination (t1/2 beta = 188.0 min) of its major plasma metabolite, DON-glucuronide conjugate, which accounted for 13% of the plasma radioactivity levels. DON was rapidly cleared from the body by metabolism to 7 possible metabolites, which were excreted essentially in the urine (91%) and to a lesser extent in the bile (6%). Most (67%) of the recovered radioactivity was in the form of the glucuronide conjugates of DON (54%) and the de-epoxide metabolite, DOM-1 (13%). Excretion of unmetabolized DON accounted for 11%. The remaining recovered dose (18%) comprised of minor amounts of DOM-1 (6%), DON-sulfate conjugate (2%) and 3 unidentified radioactive components (10%). Studies on the presence of DON-derived residues in milk indicated that, relative to the dose, only trace amounts were transmitted following either oral or iv administration of the toxin.  相似文献

Fluorescence in situ hybridisation (FISH) is a powerful molecular biological tool to detect and enumerate harmful microorganism in the marine environment. Different FISH methods are available, and especially in combination with automated counting techniques, the potential for a routine monitoring of harmful marine microalgae is attainable. Various oligonucleotide probes are developed for detecting harmful microalgae. However, FISH-based methods are not yet regularly included in monitoring programmes tracking the presence of harmful marine microalgae. A limitation factor of the FISH technique is the currently available number of suited fluorochromes attached to the FISH probes to detect various harmful species in one environmental sample at a time. However, coupled automated techniques, like flow cytometry or solid-phase cytometry, can facilitate the analysis of numerous field samples and help to overcome this drawback. A great benefit of FISH in contrast to other molecular biological detection methods for harmful marine microalgae is the direct visualisation of the hybridised target cells, which are not permitted in cell free formats, like DNA depending analysis methods. Therefore, an additional validation of the FISH-generated results is simultaneously given.  相似文献

Growth and yield were reduced but (14)C translocation velocity was not affected by increasing levels of ozone in spring wheat exposed in open top chambers to the following treatments: charcoal filtered air (CF), non-filtered ambient air (NF), or NF with addition of 30 microl litre(-1) ozone, 8 h daily (NFO). Destructive harvests were performed at anthesis and at maturity. Parts of the flag leaf or the second leaf were exposed to (14)CO(2) in small cuvettes for 5 min before, during and after anthesis. The translocation velocity was followed by autoradiography and scintillation counting of the plants frozen and lyophilized at different times after labelling. The label was transported at the same velocity in all the treatments. Ozone induced changes in carbon allocation or partitioning should probably be explained as amounts of carbon transported (mg s(-1)), rather than as transportation velocity (mm s(-1)). The amount translocated may be governed by source conditions under O(3) stress: reduced healthy green biomass and photosynthesis, but perhaps also by impairment of phloem loading because of membrane damage.  相似文献

以洗衣粉中的有机磷为磷源,通过室内模拟的方法研究在不同磷浓度的水体条件下斜生栅藻的生长情况,以及磷对斜生栅藻的生长是否有毒害作用.通过显微计数测定藻细胞数目来探讨洗衣粉中磷对斜生栅藻生长的影响;同时分析了洗衣粉中磷对斜生栅藻96 h生长期中光合色素的影响.结果表明,洗衣粉中的磷除了促进藻生长外,其浓度过低或过高都会抑制藻的生长.由实验结果可得,磷对藻生长的富营养化作用和毒性作用取决于磷浓度的高低.  相似文献
