Ninety Capsicum accessions selected from the USDA Capsicum germplasm collection were screened for their capsaicinoids content using gas hromatography with nitrogen phosphorus detection (GC/NPD). Fresh fruits of Capsicum chinense, C. frutescens, C. baccatum, C. annuum, and C. pubescens were extracted with methanol and analyzed for capsaicin, dihydrocapsaicin, and nordihydrocapsaicin. Mass spectrometry of the fruit crude extracts indicated that the molecular ions at m/z 305, 307, and 293, which correspond to capsaicin, dihydrocapsaicin, and nordihydrocapsaicin, respectively, have a common benzyl cation fragment at m/z 137 that can be used for monitoring capsaicinoids in pepper fruit extracts. Capsaicin and dihydrocapsaicin were the dominant capsaicinoids detected. Capsaicin concentrations were typically greater than dihydrocapsaicin. Concentrations of total capsaicinoids varied from not detectable to 11.2 mg fruit?1. Statistical analysis revealed that accession PI-441624 (C. chinense) had the highest capsaicin content (2.9 mg g?1 fresh fruit) and accession PI-497984 (C. frutescens) had the highest dihydrocapsaicin content (2.3 mg g?1 fresh fruit). Genebank accessions PI-439522 (C. frutescens) and PI-497984 contained the highest concentrations of total capsaicinoids.  相似文献

With the public perception that synthetic pesticides leave harmful residues in crop produce for human consumption, there has been increased interest in using natural products for pest control. The potential of using fruit extracts of hot pepper for controlling the cabbage looper, Trichopulsia ni (Hübner) and spider mite, Tetranychus urticae Koch is explored in this investigation. Crude extracts from fruits of Capsicum chinense, C. frutescens, C. baccatum, and C. annuum, were prepared and tested under laboratory conditions for their insecticidal and acaricidal performance. Mortality was greatest (94%) when fruit extract of accession PI-593566 (C. annuum) was sprayed on larvae of the cabbage looper, while crude extracts of accessions PI-241675 (C. frutescens) and PI-310488 (C. annuum) were repellent to the spider mite. We investigated differences in chemical composition of the crude fruit extracts that may explain the observed differences in mortality and repellency between accessions. Gas Chromatography-Mass Spectrometry spectrometric analysis revealed that capsaicin and dihydrocapsaicin, the pungent components of pepper fruit, were not correlated with toxicity or repellency, indicating that the two capsaicinoids are not likely related to the efficacy of pepper fruit extracts. Major compounds in hot pepper fruit extracts were detected and identified as pentadecanoic acid methyl ester, hexadecanoic acid methyl ester, and octadecanoic acid methyl ester. Spectrometric analysis and toxicity to cabbage looper larvae revealed that pentadecanoic acid methyl ester is likely related to cabbage looper mortality. However, the concentration of pentadecanoic acid methyl ester in some accessions was insufficient to explain the observed mortality of cabbage looper and repellency of spider mite. Fruit extracts of accessions PI-593566 (C. annuum) and PI-241675 (C. frutescens) could be useful for managing populations of cabbage loopers and spider mites, which could reduce reliance on synthetic pesticides. Further study is needed to investigate performance of hot pepper extracts under ultra-violet light and field conditions.  相似文献

The U.S. Department of Agriculture (USDA) pepper (Capsicum spp.) germplasm collection contains several thousand members or accessions. Many of these species and cultivars have not been analyzed for their concentrations of ascorbic acid, capsaicin, and total phenolic compounds, which are important antioxidants having a number of benefits for human health. The objective of this investigation was to select candidate accessions of hot pepper having high concentrations of ascorbic acid, capsaicin, free sugars, and total phenols for use as parents in breeding for these compounds. Seventeen accessions of pepper from the core Capsicum germplasm collection (four accessions of Capsicum chinense; five accessions of C. baccatum; six accessions of C. annuum; and two of C. frutescens) were field grown and their mature fruits were analyzed for their antioxidant composition. Concentrations of these compounds tended to be higher in C. chinense and C. baccatum, than in C. annuum and C. frutescens. Across all accessions the concentration of total phenols was correlated with ascorbic acid (r = 0.97) and free sugars (r = 0.80). Concentrations of total phenols (1.4, 1.3, and 1.3 mg g-1 fruit) and ascorbic acid (1.6, 1.2, and 1.3 mg g-1 fruit) were significantly greater in PI-633757, PI-387833, and PI-633754, respectively, compared to other accessions analyzed. Total capsaicinoids concentrations were greatest (1.3 mg g-1 fruit) in PI-438622 and lowest (0.002 mg g-1 fruit) in Grif-9320. The great variability within and among Capsicum species for these phytochemicals suggests that these selected accessions may be useful as parents in hybridization programs to produce fruits with value-added traits.  相似文献

The potential of using fruit extracts of hot pepper as alternatives to synthetic acaricides for controlling the two-spotted spider mite, Tetranychus urticae Koch, is explored in this study. This investigation suggests that methanolic extracts of these three accessions may have a great potential for repelling spider mites and should be field-tested on a large-scale to assess their value in managing populations of spider mites, which could reduce reliance on synthetic acaricides. An attempt was made to correlate repellency with chemical constituents of fruit extracts of the most repellent accessions to identify chemical sources of repellency. Capsaicin and dihydrocapsaicin, the pungent components of pepper fruit, were not correlated with toxicity or repellency, indicating that these are not likely related to the toxicity or repellency of the pepper fruit extracts. Elevated concentrations of heavy metals in edible plants could expose consumers to excessive levels of potentially hazardous chemicals. Sixty-three accessions (genotypes) of Capsicum chinense Jacq, collected from 8 countries of origin were grown in a silty-loam soil under field conditions. At maturity, fruits were collected and analyzed for seven heavy metals (Cd, Cr, Ni, Pb, Zn, Cu, and Mo) concentrations. The main objectives of this investigation were: 1) to determine the concentrations of seven heavy metals in the soil and monitor their accumulation in mature fruits, 2) to categorize the pepper accessions as low or high heavy metal accumulators, and 3) to determine if heavy metal content of the pepper fruit was lower than the permitted limits. Concentrations and relative proportions of heavy metals in pepper fruits of C. chinense varied among accessions. Fruits of Plant Introduction (PI) 355820 accumulated significant concentrations of Cd (0.47 μ g g?1dry fruit). PI-260522 accumulated the highest concentration of Pb (2.12 μ g g?1 dry fruit) among the 63 accessions tested. This accession (PI-260522) contained about twice the Pb limit on a fresh weight basis. Among the 63 accessions analyzed, PI-238051 contained the highest levels of Ni (17.2 μ g g?1). We concluded that high accumulator genotypes may be useful for phytoremediation, while, low accumulator accessions might be appropriate selections for growing on Cd-, Pb-, or Ni-contaminated soils to prevent potential human exposure to heavy metals and health hazards through the food chain.  相似文献

The main objective of this investigation was to evaluate fruits of C. chinense accessions for their concentration of β -carotene, ascorbic acid, and phenols for use as parents in breeding for these phytochemicals. Mature fruits of 63 accessions of C. chinense originally acquired from Belize, Brazil, Colombia, Ecuador, Mexico, Peru, Puerto Rico, and the United States were analyzed for their chemical composition. Fruits of C. chinense accessions PI-152452 (Brazil) and PI-360726 (Ecuador) contained the greatest concentrations of ascorbic acid (1.2 and 1.1 mg g?1 fresh fruit, respectively), while PI-438648 (Mexico) contained the greatest concentration of total phenols content (349 μ g g?1 fresh fruit) among the other 63 accessions tested. Accession PI-355817 from Ecuador contained the greatest concentrations of β -carotene (8 mg g?1fresh fruit). These accessions were identified as potential candidates for mass production of antioxidants with health-promoting properties.  相似文献

Peppers, a significant component of the human diet in many regions of the world, provide vitamins A (β-carotene) and C, and are also a source of many other antioxidants such as capsaicin, dihydrocapsaicin, and phenols. Enhancing the concentration of antioxidants in plants grown in soil amended with recycled waste has not been completely investigated. Changes in pepper antioxidant content in relation to soil amendments and fruit development were investigated. The main objectives of this investigation were to: (i) quantify concentrations of capsaicin, dihydrocapsaicin, β-carotene, ascorbic acid, phenols, and soluble sugars in the fruits of Capsicum annuum L. (cv. Xcatic) grown under four soil management practices: yard waste (YW), sewage sludge (SS), chicken manure (CM), and no-much (NM) bare soil and (ii) monitor antioxidant concentrations in fruits of plants grown under these practices and during fruit ripening from green into red mature fruits. Total marketable pepper yield was increased by 34% and 15% in SS and CM treatments, respectively, compared to NM bare soil; whereas, the number of culls (fruits that fail to meet the requirements of foregoing grades) was lower in YW compared to SS and CM treatments. Regardless of fruit color, pepper fruits from YW amended soil contained the greatest concentrations of capsaicin and dihydrocapsaicin. When different colored pepper fruits (green, yellow, orange, and red) were analyzed, orange and red contained the greatest β-carotene and sugar contents; whereas, green fruits contained the greatest concentrations of total phenols and ascorbic acid.  相似文献

Composition of ginger oil prepared from fresh ginger rhizomes, Zingiber officinale Roscoe (Zingiberaceae) was determined by gas chromatography (GC) and GC-mass spectrometric techniques. The main sesquiterpene hydrocarbons identified were alpha-zingiberene (27-30%), alpha-curcumene (8-9%), beta-sesquiphellandrene (4.8%). and bisabolene (3.2%). The function of zingiberene and curcumene as insecticides, repellents, and insect feeding deterrents has been previously reported. Other plant species having similar constituents might be found. Leaves of six wild tomato accessions of Lycopersicon hirsutum f. glabratum (Mull); three accessions of L. hirsutum f. typicum (Humb & Bonpl.); two accessions of L. pennellii Corr. (D'Arcy); one accession of L. pimpinellifolium; and one commercial tomato L. esculentumm cv. Fabulous were analyzed. Analysis of L. hirsutum f. typicum (Solanaceae) accessions indicated the presence of zingiberene, curcumene, and other lipophilic secondary metabolites in the leaves of two accessions (PI-127826 and PI-127827). An average three month old wild tomato plant of accessions PI-127826 and PI-127827 provided 1.93 and 1.30 kg fresh leaves (averaging about 38,307 and 28,130 cm2 exposed leaf surface area, respectively) and produced 19.3 and 10.1 g of zingiberene and curcumene (PI-127826) and 17.2 and 1.8 g of zingiberene and curcumene (PI-127827), respectively. Leaf extracts of the wild tomato L. hirsutum f. typicum (accessions PI-127826 and PI-127827) can be used as a biorational source of zingiberene and curcumene.  相似文献

Crude extracts from leaves of three accessions of L. hirsutum, six accessions of L. hirsutum f. glabratum, and one accession each of L. pennellii and L. pimpinellifolium were prepared in chloroform, ethanol and hexane. Two spider mite bioassays, one a measure of antibiosis and the other a measure of repellency, were utilized to determine the acaricidal performance of the crude extracts. The bioassay for antibiosis was a 6-h no-choice test. The bioassay for repellency utilized a ring bioassay. Chloroform leaf extracts of L. hirsutum f. glabratum accessions (PI-251304, PI-134417, PI-134418, and PI-126449) exhibited greatest antibiotic activity on two-spotted spider mites; the hexane extracts exhibited greatest repellency. Extracts from PI-251304, PI-126449, PI-134417, and PI-134418 were especially lethal (chloroform) or repellent (hexane). We investigated differences in chemical composition of the crude leaf extracts that may explain the observed differences in mortality and repellency among the different accessions. Major chemical compounds (alpha -curcumene, alpha -zingiberene, trans-caryophyllene, 2-undecanone, and 2-tridecanone) known to have pesticidal efficacy were detected and quantified in the crude leaf extracts using a gas chromatograph (GC) equipped with a mass spectrometer (GC/MS). Field experiment was conducted to investigate the di-n-butyl phthalate (DBP) and di-2-ethylhexyl phthalate (DEHP) contamination in Capsicum annum fruit grown in DBP and DEHP contaminated soil, and to evaluate the effect of DBP and DEHP on the quality of capsicum fruit. The top layer soil (0-10 cm) of plots was treated with a mixture of DBP and DEHP (1:1 w/w) and capsicum seedlings were transplanted. After 90 days, capsicum fruit, shoot and root samples were collected. DBP and DEHP concentration in various parts of the samples were determined by gas chromatography. Vitamin C and capsaicin contents in fruit were determined using 2,4-dinitrophenylhydrazine colorimetric analysis and sodium nitrite-sodium molybdate colorimetric analysis, respectively. The results showed that DBP concentration in fruit, shoot and root increased with the increase of soil-applied DBP/DEHP concentration, but DEHP was not detected in all samples. When the soil-applied DBP/DEHP concentration was 5, 10, 20, 40, 80 and 160 mg kg(-1) soil, compared with control, vitamin C and capsaicin content in capsicum fruit decreased by 1.6%, 5.9%, 10.6%/o, 18.2%, 19.2%, 22.6% and 1.6%, 2.5%, 12.9%, 20.1%, 22.2%, respectively. Pearson correlation analysis demonstrated that the decrease of vitamin C and capsaicin content was negatively correlated to the increase of DBP concentration in capsicum fruit, which suggested that DBP uptake by the plant might be mainly responsible for quality degradation of capsicum fruit.  相似文献

BACKGROUND: Capsaicin, a non-toxic natural product antifoulant, is an attractive alternative to the currently used toxic metal-based antifoulants for protecting the submerged surfaces. The problem associated with conducting capsaicin experiments is its limited aqueous solubility. METHODS: A procedure was developed to obtain the maximum capsaicin concentration in water. Capsaicin was first dissolved in one of the three organic solvents, methanol, ethanol, and acetone. The organic solvent was replaced by gradually adding distilled water and evaporating the organic solvent. RESULTS: The best solvent for obtaining highest concentration of capsaicin in water was ethanol. A maximum capsaicin in water 6400 ppm was achieved by using 16 mg capsaicin dissolved in 0.8 ml ethanol followed with the addition of 2.5 ml distilled water in 0.2 ml increments and then slow evaporation of the solvent. CONCLUSION: The concentration obtained using our approach is well above the threshold concentration of capsaicin in deterring the marine organism attachments.  相似文献

The toxicity of two methylketones (2-undecanone and 2-tridecanone), the major constituents of the accessions tested, to adults of the sweetpotato whitefly, Bemisia tabaci (Gennadius) and fourth instar larvae of the Colorado potato beetle (CPB), Leptinotarsa decemlineata (Say), was determined using no-choice bioassays. 2-undecanone caused 80% mortality of the fourth instar larvae of the CPB at the highest concentration tested (100 mg 2-undecanone mL(-1) of acetone) while 2-tridecanone caused 72% mortality of whiteflies at 20 mg 2-tridecanone mL(-1) of 65% ethanol. The concentration of 2-undecanone and 2-tridecanone on the leaves of each of the five accessions of L. hirsutum f. glabratum and the mass spectra of 2-undecanone and 2-tridecanone are presented. Overall, the concentration of 2-undecanone on the leaves was significantly higher than 2-tridecanone. Concentration of the two methylketones varied among accessions and sampling season. Plants produced the highest concentration of 2-undecanone during the month of August. Natural product-based pesticides offer advantages that they are specific to the target species and typically have unique modes of action with little mammalian toxicity. Studies conducted at Kentucky State University revealed that the insecticidal and acaricidal performance of crude extracts from the fruits of Capsicum species justified the need for developing a natural pesticide formulation for agricultural use. Analysis of hot pepper extracts indicated the presence of three compounds having pesticidal efficacy: capsaicin, dihydrocapsaicin, and pentadecanoic acid methyl ester. The main objectives of this investigation were to: i) determine the half-live (T1/2 value) of capsaicin, dihydrocapsaicin, and pentadecanoic acid methyl ester in soil sprayed with hot pepper formulation and ii) assess the impact of hot pepper formulation on total soil enzyme activity. Analysis of soil sprayed with pepper extracts indicated that T1/2 values of capsaicin, dihydrocapsaicin, and pentadecanoic acid methyl ester were 7.3, 1.5, and 10.2 d, respectively. Hydrolysis of the fluorescein diacetate [3, 6-diacetylfluorescein (FDA)], used for measuring overall soil enzyme activities, indicated that total enzyme activities was slightly enhanced by 2% in soil sprayed with hot pepper extracts.  相似文献

Metal tolerance and phytoextraction potential of two common sorrel (Rumex acetosa L.) accessions, collected from a Pb/Zn contaminated site (CS, Lanestosa) and an uncontaminated site (UCS, Larrauri), were studied in fertilized and non-fertilized pots prepared by combining soil samples from both sites in different proportions (i.e., 0%, 33%, 66% and 100% of Lanestosa contaminated soil). The original metalliferous mine soil contained 20480, 4950 and 14 mg kg(-1) of Zn, Pb and Cd, respectively. The microcosm experiment was carried out for two months under greenhouse controlled conditions. It was found that fertilization increased mean plant biomass of both accessions as well as their tolerance. However, only the CS accession survived all treatments even though its biomass decreased proportionally according to the percentage of contaminated mine soil present in the pots. This metallicolous accession would be useful for the revegetation and phytostabilization of mine soils. Due to its high concentration and bioavailability in the contaminated soil, the highest values of metal phytoextracted corresponded to Zn. The CS accession was capable of efficiently phytoextracting metal from the 100% mine soil, indeed reaching very promising phytoextraction rates in the fertilized pots (6.8 mg plant(-1) month(-1)), similar to the ones obtained with hyperaccumulator plants. It was concluded that fertilization is certainly worth being considered for phytoextraction and revegetation with native plants from metalliferous soils.  相似文献

Abstract

Members of TRP receptor family are involved in response to acidification. Here, we determined the effect of capsaicin, one of the TRP receptor activators, on hemolymph acid-base status in the American cockroach. Periplaneta americana adult individuals were injected with lactic acid (5% or 10%) and exposed to 100?µM capsaicin solution. Hemolymph pH was measured 15?min, 1, 4, 8 and 24?h after lactic acid and capsaicin application with a glass microelectrode. The results demonstrated that cockroaches recover from acidosis within 4?h from acid injection. Capsaicin impaired the buffering capacity of insects’ hemolymph, resulting in significant drop of hemolymph pH observed even 24?h after application. Joint action of capsaicin and acidosis reveals new insight into possible mechanism of capsaicin action on TRP receptors in insects.  相似文献

Two common sorrel (Rumex acetosa) accessions, one from a Zn-Pb contaminated site (CS accession) and the other from an uncontaminated site (UCS accession), were hydroponically exposed to a mixture of heavy metals (Pb2+ + Zn2+ + Cd2+) with and without EDTA at an equimolar rate. The metallicolous CS accession showed a higher tolerance to metal treatment in the absence of the chelating agent, whereas the UCS accession was especially tolerant to EDTA treatment alone. Combination of metal and EDTA treatment resulted in a higher Pb accumulation in shoots of both accessions although plants hardly showed phytotoxic symptoms. Cd and Zn uptake was not augmented by EDTA addition to the polymetallic medium. Chelant-assisted Pb accumulation was 70% higher in the CS accession than in the UCS accession, despite the fact that the former accession evapotranspired less water than the UCS accession. These results support the existence of a non-selective apoplastic transport of metal chelates by R. acetosa roots, not related to transpiration stream.  相似文献
