The U.S. Department of Agriculture (USDA) pepper (Capsicum spp.) germplasm collection contains several thousand members or accessions. Many of these species and cultivars have not been analyzed for their concentrations of ascorbic acid, capsaicin, and total phenolic compounds, which are important antioxidants having a number of benefits for human health. The objective of this investigation was to select candidate accessions of hot pepper having high concentrations of ascorbic acid, capsaicin, free sugars, and total phenols for use as parents in breeding for these compounds. Seventeen accessions of pepper from the core Capsicum germplasm collection (four accessions of Capsicum chinense; five accessions of C. baccatum; six accessions of C. annuum; and two of C. frutescens) were field grown and their mature fruits were analyzed for their antioxidant composition. Concentrations of these compounds tended to be higher in C. chinense and C. baccatum, than in C. annuum and C. frutescens. Across all accessions the concentration of total phenols was correlated with ascorbic acid (r = 0.97) and free sugars (r = 0.80). Concentrations of total phenols (1.4, 1.3, and 1.3 mg g?1 fruit) and ascorbic acid (1.6, 1.2, and 1.3 mg g?1 fruit) were significantly greater in PI-633757, PI-387833, and PI-633754, respectively, compared to other accessions analyzed. Total capsaicinoids concentrations were greatest (1.3 mg g?1 fruit) in PI-438622 and lowest (0.002 mg g?1 fruit) in Grif-9320. The great variability within and among Capsicum species for these phytochemicals suggests that these selected accessions may be useful as parents in hybridization programs to produce fruits with value-added traits.  相似文献

Elevated concentrations of heavy metals in edible plants could expose consumers to excessive levels of potentially hazardous chemicals. Sixty-three accessions (genotypes) of Capsicum chinense Jacq, collected from 8 countries of origin were grown in a silty-loam soil under field conditions. At maturity, fruits were collected and analyzed for seven heavy metals (Cd, Cr, Ni, Pb, Zn, Cu, and Mo) concentrations. The main objectives of this investigation were: 1) to determine the concentrations of seven heavy metals in the soil and monitor their accumulation in mature fruits, 2) to categorize the pepper accessions as low or high heavy metal accumulators, and 3) to determine if heavy metal content of the pepper fruit was lower than the permitted limits. Concentrations and relative proportions of heavy metals in pepper fruits of C. chinense varied among accessions. Fruits of Plant Introduction (PI) 355820 accumulated significant concentrations of Cd (0.47 μ g g?1dry fruit). PI-260522 accumulated the highest concentration of Pb (2.12 μ g g?1 dry fruit) among the 63 accessions tested. This accession (PI-260522) contained about twice the Pb limit on a fresh weight basis. Among the 63 accessions analyzed, PI-238051 contained the highest levels of Ni (17.2 μ g g?1). We concluded that high accumulator genotypes may be useful for phytoremediation, while, low accumulator accessions might be appropriate selections for growing on Cd-, Pb-, or Ni-contaminated soils to prevent potential human exposure to heavy metals and health hazards through the food chain.  相似文献

Ninety Capsicum accessions selected from the USDA Capsicum germplasm collection were screened for their capsaicinoids content using gas hromatography with nitrogen phosphorus detection (GC/NPD). Fresh fruits of Capsicum chinense, C. frutescens, C. baccatum, C. annuum, and C. pubescens were extracted with methanol and analyzed for capsaicin, dihydrocapsaicin, and nordihydrocapsaicin. Mass spectrometry of the fruit crude extracts indicated that the molecular ions at m/z 305, 307, and 293, which correspond to capsaicin, dihydrocapsaicin, and nordihydrocapsaicin, respectively, have a common benzyl cation fragment at m/z 137 that can be used for monitoring capsaicinoids in pepper fruit extracts. Capsaicin and dihydrocapsaicin were the dominant capsaicinoids detected. Capsaicin concentrations were typically greater than dihydrocapsaicin. Concentrations of total capsaicinoids varied from not detectable to 11.2 mg fruit?1. Statistical analysis revealed that accession PI-441624 (C. chinense) had the highest capsaicin content (2.9 mg g?1 fresh fruit) and accession PI-497984 (C. frutescens) had the highest dihydrocapsaicin content (2.3 mg g?1 fresh fruit). Peppers, a significant component of the human diet in many regions of the world, provide vitamins A (β-carotene) and C, and are also a source of many other antioxidants such as capsaicin, dihydrocapsaicin, and phenols. Enhancing the concentration of antioxidants in plants grown in soil amended with recycled waste has not been completely investigated. Changes in pepper antioxidant content in relation to soil amendments and fruit development were investigated. The main objectives of this investigation were to: (i) quantify concentrations of capsaicin, dihydrocapsaicin, β-carotene, ascorbic acid, phenols, and soluble sugars in the fruits of Capsicum annuum L. (cv. Xcatic) grown under four soil management practices: yard waste (YW), sewage sludge (SS), chicken manure (CM), and no-much (NM) bare soil and (ii) monitor antioxidant concentrations in fruits of plants grown under these practices and during fruit ripening from green into red mature fruits. Total marketable pepper yield was increased by 34% and 15% in SS and CM treatments, respectively, compared to NM bare soil; whereas, the number of culls (fruits that fail to meet the requirements of foregoing grades) was lower in YW compared to SS and CM treatments. Regardless of fruit color, pepper fruits from YW amended soil contained the greatest concentrations of capsaicin and dihydrocapsaicin. When different colored pepper fruits (green, yellow, orange, and red) were analyzed, orange and red contained the greatest β-carotene and sugar contents; whereas, green fruits contained the greatest concentrations of total phenols and ascorbic acid.  相似文献

Composting and land application of municipal sewage sludge (SS) and yard waste (YW) compost are increasingly popular ways for using organic waste as a source of organic matter, while decreasing the amount of waste being diverted into landfills. Researchers have largely ignored the effect of SS and SS mixed with YW (SS+YW) compost on the antioxidant contents of vegetables grown under this practice. Accordingly, the main objective of this investigation was to monitor the impact of SS and SS+YW on the nutritional composition of pepper and melon fruits at harvest. Total phenols and ascorbic acid contents of pepper and melons (determined by the Folin-Ciocalteau and the dichlorophenol–indophenol methods, respectively) were greater in pepper than melon fruits. Soil amended with SS or SS+YW significantly elevated the concentrations of ascorbic acid and total phenols in melon fruits compared to no-mulch native soil. The application of SS did not modify the concentration of β-carotene in melon fruits at three harvests. Regardless of soil treatments, ascorbic acid and total phenols concentrations were greater in melon fruits collected at the first harvest compared to harvest three. Pepper fruits collected at harvest three contained the greatest concentrations of ascorbic acid and total phenols; whereas, pepper fruits collected at the second harvest contained the greatest concentrations of soluble sugars.  相似文献

Production of methyl ketones as naturally occurring insecticides from wild tomato accessions is explored in this study. Density of two glandular trichomes (type IV and type VI) on the leaves of six wild tomato accessions of Lycopersicon hirsutum f. glabratum (Mull); three accessions of L. hirsutum f. typicum (Humb & Bonpl.); two accessions of L. pennellii Corr. (D'Arcy); and density of type VI glandular trichomes of the commercial tomato Lycopersicon esculentum cv. Fabulous are reported. Densities of type IV and VI glandular trichomes varied among the accessions tested. Type IV trichomes occurred at much higher densities than type VI. Over all accessions and counting trichome densities on both abaxial and adaxial leaflet surfaces, type IV densities averaged 96,378 trichomes.g?1 leaflets while type VI densities averaged 67,350 trichomes.g?1 leaflets. Concentrations of four methyl ketones (2-tridecanone, 2-dodecanone, 2-undecanone, 2-pentadecanone) were determined per unit leaf surface area (mm2) and per g fresh leaflets. Concentrations of total methyl ketones ranged from 81.3 μg. g?1 fresh leaflets on L. esculentum cv. Fabulous to 5.5 mg. g?1 on L. hirsutum f. glabratum (PI 134417). Two methyl ketones, 2-undecanone and 2-tridecanone, predominated trichome secretions in five of the L. hirsutum f. glabratum accessions tested (PI 251304, PI 126449, PI 134417, PI 134418, and LA 407).  相似文献

Increasing concern about persistence and environmental impact of synthetic pesticide residues require development of biodegradable and environmentally safe alternatives. The potential of using fruit extracts of hot pepper as alternatives to synthetic acaricides for controlling the two-spotted spider mite, Tetranychus urticae Koch, is explored in this study. Twenty-four Capsicum accessions (Solanaceae) were screened for their toxicity and repellency to the spider mites. Crude extracts from fruits of C. chinense, C. frutescens, C. baccatum, C. annuum, and C. pubescens were prepared in methanol and tested for their acaricidal properties. Spider mite mortality was greatest (45%) when fruit extract of accession Grif-9169 (C. annuum) was used. Results from diving board bioassays indicated that mites avoided filter paper strips treated with hot pepper extracts from accessions PI-596057 (C. baccatum), PI-195299 (C. annuum), and Grif- 9270 (C. annuum). This investigation suggests that methanolic extracts of these three accessions may have a great potential for repelling spider mites and should be field-tested on a large-scale to assess their value in managing populations of spider mites, which could reduce reliance on synthetic acaricides. An attempt was made to correlate repellency with chemical constituents of fruit extracts of the most repellent accessions to identify chemical sources of repellency. Capsaicin and dihydrocapsaicin, the pungent components of pepper fruit, were not correlated with toxicity or repellency, indicating that these are not likely related to the toxicity or repellency of the pepper fruit extracts. Municipal sewage sludge (MSS) used for land farming typically contains heavy metals that might impact crop quality and human health. A completely randomized experimental design with three treatments (six replicates each) was used to monitor the impact of mixing native soil with MSS or yard waste (YW) mixed with MSS (YW +MSS) on: i) sweet potato yield and quality; ii) concentration of seven heavy metals (Cd, Cr, Mo, Cu, Zn, Pb, and Ni) in sweet potato plant parts (edible roots, leaves, stem, and feeder roots); and iii) concentrations of ascorbic acid, total phenols, free sugars, and β-carotene in sweet potato edible roots at harvest. Soil samples were collected and analyzed for total and extractable metals using two extraction procedures, concentrated nitric acid (to extract total metals from soil) as well as CaCl2 solution (to extract soluble metals in soil that are available to plants), respectively. Elemental analyses were performed using inductively coupled plasma mass spectrometry (ICP-MS). Overall, plant available metals were greater in soils amended with MSS compared to control plots. Concentration of Pb was greater in YW than MSS amendments. Total concentrations of Pb, Ni, and Cr were greater in plants grown in MSS+YW treatments compared to control plants. MSS+YW treatments increased sweet potato yield, ascorbic acid, soluble sugars, and phenols in edible roots by 53, 28, 27, and 48%, respectively compared to plants grown in native soil. B-carotene concentration (157.5 μg g?1 fresh weight) was greater in the roots of plants grown in MSS compared to roots of plants grown in MSS+YW treatments (99.9 μg g?1 fresh weight). Concentration of heavy metals in MSS-amended soil and in sweet potato roots were below their respective permissible limits.  相似文献

Organic matter and nutrients in municipal sewage sludge (SS) and chicken manure (CM) could be recycled and used for land farming to enhance fertility and physical properties of soils. Three soil management practices were used at Kentucky State University Research Farm, Franklin County, to study the impact of soil amendments on kale (Brassica oleracea cv. Winterbar) and collard (Brassica oleracea cv. Top Bunch) yields and quality. The three soil management practices were: (i) SS mixed with native soil at 15 t acre?1, (ii) CM mixed with native soil at 15 t acre?1, and (iii) no-mulch (NM) native soil for comparison purposes. At harvest, collard and kale green plants were graded according to USDA standards. Plants grown in CM and SS amended soil produced the greatest number of U.S. No. 1 grade of collard and kale greens compared to NM native soil. Across all treatments, concentrations of ascorbic acid and phenols were generally greater in kale than in collards. Overall, CM and SS enhanced total phenols and ascorbic acid contents of kale and collard compared to NM native soil. We investigated the chemical and physical properties of each of the three soil treatments that might explain variability among treatments and the impact of soil amendments on yield, phenols, and ascorbic acid contents of kale and collard green grown under this practice.  相似文献

The potential of using phytochemicals from leaves of wild tomato for controlling the two-spotted spider mite, Tetranychus urticae Koch, is explored in this study as a promising alternative to the use of synthetic pesticides. Wild tomato accessions of Lycopersicon hirsutum plants that are not consumed by humans were planted under greenhouse conditions for mass production of leaves. Crude extracts from leaves of three accessions of L. hirsutum, six accessions of L. hirsutum f. glabratum, and one accession each of L. pennellii and L. pimpinellifolium were prepared in chloroform, ethanol and hexane. Two spider mite bioassays, one a measure of antibiosis and the other a measure of repellency, were utilized to determine the acaricidal performance of the crude extracts. The bioassay for antibiosis was a 6-h no-choice test. The bioassay for repellency utilized a ring bioassay. Chloroform leaf extracts of L. hirsutum f. glabratum accessions (PI-251304, PI-134417, PI-134418, and PI-126449) exhibited greatest antibiotic activity on two-spotted spider mites; the hexane extracts exhibited greatest repellency. Extracts from PI-251304, PI-126449, PI-134417, and PI-134418 were especially lethal (chloroform) or repellent (hexane). We investigated differences in chemical composition of the crude leaf extracts that may explain the observed differences in mortality and repellency among the different accessions. Major chemical compounds (α -curcumene, α -zingiberene, trans-caryophyllene, 2-undecanone, and 2-tridecanone) known to have pesticidal efficacy were detected and quantified in the crude leaf extracts using a gas chromatograph (GC) equipped with a mass spectrometer (GC/MS). Lethality of extracts was mainly associated with the presence of high concentrations of 2-tridecanone; repellency of extracts was mainly associated with the presence of trans-caryophyllene. Leaf extracts of L. hirsutum f. glabratum accessions that contain significant quantities of 2-tridecanone and/or trans-caryophyllene could be useful for managing populations of spider mites, which could reduce reliance on synthetic pesticides.  相似文献

Two plant species, arugula (Eruca sativa) and mustard (Brassica juncea) were field-grown under four soil management practices: soil mixed with municipal sewage sludge (SS), soil mixed with horse manure (HM), soil mixed with chicken manure (CM), and no-mulch bare soil (NM) to investigate the impact of soil amendments on the concentration of glucosinolates (GSLs) in their shoots. GSLs, hydrophilic plant secondary metabolites in arugula and mustard were extracted using boiling methanol and separated by adsorption on sephadex ion exchange disposable pipette tips filled with DEAE, a weak base, with a net positive charge that exchange anions such as GSLs. Quantification of GSLs was based on inactivation of arugula and mustard myrosinase and liberation of the glucose moiety from the GSLs molecule by addition of standardized myrosinase (thioglucosidase) and spectrophotometric quantification of the liberated glucose moiety. Overall, GSLs concentrations were significantly greater (1287 µg g?1 fresh shoots) in plants grown in SS compared to 929, 890, and 981 µg g?1 fresh shoots in plants grown in CM, HM, and NM soil, respectively. Results also revealed that mustard shoots contained greater concentrations of GSLs (974 µg g?1 fresh shoots) compared to arugula (651 µg g?1 fresh shoots).  相似文献

The main aim of this study was to assess the impact of pesticidal residues on soil microbial and biochemical parameters of the tea garden soils. The microbial biomass carbon (MBC), basal (BSR) and substrate induced respirations (SIR), β -glucosidase activity and fluorescein diacetate hydrolyzing activity (FDHA) of six tea garden soils, along with two adjacent forest soils (control) in West Bengal, India were measured. The biomass and its activities and biochemical parameters were generally lower in the tea garden soils than the control soils. The MBC of the soils ranged from 295.5 to 767.5 μ g g? 1. The BSR and SIR ranged from 1.65 to 3.08 μ g CO2-C g? 1 soil h? 1 and 3.08 to 10.76 μ g CO2-C g? 1h? 1 respectively. The β -glucosidase and FDHA of the soils varied from 33.3 and 76.3 μ g para-nitrophenol g? 1 soil h? 1 and 60.5 to 173.5 μ g fluorescein g? 1h? 1respectively. The tea garden soils contained variable residues of organophosphorus and organochlorine pesticides, which negatively affected the MBC, BSR, SIR, FDHA and β -glucosidase activity. Ethion and chlorpyriphos pesticide residues in all the tea garden soils varied from 5.00 to 527.8 ppb and 17.6 to 478.1 ppb respectively. The α endosulfan, β endosulfan and endosulfan sulfate pesticide residues in the tea garden soils ranged from 7.40 to 81.40 ppb, 8.50 to 256.1 ppb and 55 to 95.9 ppb respectively. Canonical correlation analysis shows that 93% of the total variation was associated with the negative impact of chlorpyriphos, β and α endosulfan and endosulfan sulfate on MBC, BSR and FDHA. At the same time ethion had negative impact on SIR and β -glucosidase. Data demonstrated that the pesticide residues had a strong impact on the microbial and biochemical components of soil quality.  相似文献

With the public perception that synthetic pesticides leave harmful residues in crop produce for human consumption, there has been increased interest in using natural products for pest control. The potential of using fruit extracts of hot pepper for controlling the cabbage looper, Trichopulsia ni (Hübner) and spider mite, Tetranychus urticae Koch is explored in this investigation. Crude extracts from fruits of Capsicum chinense, C. frutescens, C. baccatum, and C. annuum, were prepared and tested under laboratory conditions for their insecticidal and acaricidal performance. Mortality was greatest (94%) when fruit extract of accession PI-593566 (C. annuum) was sprayed on larvae of the cabbage looper, while crude extracts of accessions PI-241675 (C. frutescens) and PI-310488 (C. annuum) were repellent to the spider mite. We investigated differences in chemical composition of the crude fruit extracts that may explain the observed differences in mortality and repellency between accessions. Gas Chromatography-Mass Spectrometry spectrometric analysis revealed that capsaicin and dihydrocapsaicin, the pungent components of pepper fruit, were not correlated with toxicity or repellency, indicating that the two capsaicinoids are not likely related to the efficacy of pepper fruit extracts. Major compounds in hot pepper fruit extracts were detected and identified as pentadecanoic acid methyl ester, hexadecanoic acid methyl ester, and octadecanoic acid methyl ester. Spectrometric analysis and toxicity to cabbage looper larvae revealed that pentadecanoic acid methyl ester is likely related to cabbage looper mortality. However, the concentration of pentadecanoic acid methyl ester in some accessions was insufficient to explain the observed mortality of cabbage looper and repellency of spider mite. Fruit extracts of accessions PI-593566 (C. annuum) and PI-241675 (C. frutescens) could be useful for managing populations of cabbage loopers and spider mites, which could reduce reliance on synthetic pesticides. Further study is needed to investigate performance of hot pepper extracts under ultra-violet light and field conditions.  相似文献

Indoxacarb was applied at 75 and 150 g a.i. ha? 1 for two years to an eggplant (Solanum melongena L.) crop grown in the field plots in order to evaluate its efficacy for management of the lepidopteron pest, shoot and fruit borer. The residues of the insecticide were quantified by high-pressure liquid chromatography (HPLC). The mean initial deposits of indoxacarb on eggplant fruits were found to be 2.60–2.634 mg kg? 1 and 3.64–3.68 mg kg? 1 from the two rates of applications, respectively. They declined with time and reached to non-detectable (< 0.02 mg kg? 1) after 15–20 d. Residues dissipated with a half-life of 3.0–3.8 d from both first and second-year application. A 3 d waiting period for harvest of fruits after insecticide application and processing resulted in the residue levels that were below the Codex maximum residue limit (MRL) of 0.5 mg kg? 1 thereby achieving a maximum safety and minimum risk to consumers. The best combination of chemicals for decontamination of indoxacarb was found to be by washing with a mixture of alkali and potassium permanganate (KMnO4) thereby resulting in the removal of 67.5% and 59.2 % residues for 5 and 10 μ g g? 1 spiking doses, respectively. Major products formed on reaction of indoxacarb with alkali were identified by electron spray ionization chromatography/mass spectrometry (ESI/MS). Abstract

Organochlorine pesticides (OCPs) are widely used around the world as insecticides, herbicides, fungicides, nematicides, and rodenticides. Despite banned in Brazil, the usage remains occurring in many countries. The persistence and extreme mobility of OCPs contribute to the contamination of the environment and the human body. The OCPs bioaccumulation in adipose tissue triggers the excretion into human milk during breastfeeding. Hence, the present study determined eighteen OCPs residues in the breast milk of mothers from the Western Region of Bahia State, Brazil. Nine different residue species were found, including beta-Hexachlorocyclohexane (9.24?±?0.00?ng g?1 fat), delta- Hexachlorocyclohexane (22.15?±?10.48?ng g?1 fat), Heptachlor (58.08?±?74.13?ng g?1 fat), Aldrin (142.65?±?50.65?ng g?1 fat), Dieldrin (774.62?±?472.68?ng g?1 fat), Endosulfan I (408.44?±?245.51?ng g?1 fat), Dichloro-diphenyl-dichloro-ethylene (29.17?±?22.42?ng g?1 fat), Dichloro-diphenyl-trichloro-ethane (28.87?±?0.00?ng g?1 fat) and Methoxychlor (1699.67?±?797.43?ng g?1 fat). The Methoxychlor presence in all samples may reveal a recent exposure, while Dieldrin and Endosulfan I analyses can point to distant past exposure.  相似文献

A field study was conducted at Kentucky State University (KSU) Research Farm. The soil in five plots was mixed with sewage sludge, five plots were mixed with yard waste compost, five plots were mixed with laying hen manure each at 15 t acre?1, and five unamended plots that never received soil amendments were used for comparison purposes. Plots were planted with onion, Allium cepa L. var. Super Star-F1. The objectives of this investigation were to: 1) determine the concentrations of two organosulfur compounds (dipropyl disulfide and dipropyl trisulfide) in onion bulbs and 2) investigate the effect of mixing soil with three amendments (sewage sludge, yard waste, and chicken manure) on the concentration of dipropyl disulfide and dipropyl trisulfide in onion bulbs. Gas chromatographic/mass spectrometric (GC/MS) analyses of onion oil in chloroform extracts revealed the presence of two major fragment ions that correspond to dipropyl disulfide and – trisulfide. Concentration of these two organic sulfur compounds was greatest (1.5 and 0.8 mg 100 g?1 fresh weight, respectively) in onion bulbs of plants grown in chicken manure and lowest (0.4 and 0.07 mg 100 g?1 fresh weight, respectively) in onion bulbs of plants grown in yard waste compost treatments. We concluded that chicken manure could be exploited in growing onions with health-promoting properties.  相似文献

Free fatty acids and sterols were assessed in fresh manure and anaerobic lagoon sludge from swine production facilities in North Carolina. Eight free fatty acids and five sterols were identified and quantified in both manure and sludge samples. Compound identification was performed by gas chromatography/mass spectroscopy (GC-MS), and compound quantities were determined by gas chromatography after solid phase extraction with a 50:50 mixture of diethyl ether and hexane. The free fatty acids occurring in greatest abundance in both fresh manure and lagoon sludge were palmitic, oleic, and stearic. Free fatty acid content in fresh manure ranged from approximately 3 μ g g?1 dry weight (dw) to over 45 μ g g?1 dw. In lagoon sludge, free fatty acid content ranged from about 0.8 μ g g?1 dw to nearly 4 μ g g?1 dw. Coprostanol and epicoprostanol were the sterols in largest concentrations in fresh manure and lagoon sludge samples. Total sterol content ranged from approximately 0.5 μ g g?1 dw to around 11 μ g g?1 dw in fresh manure and from 3.5 μ g g?1 dw to almost 9 μ g g?1 dw in lagoon sludge. Fresh manure and lagoon sludge both had high levels of inorganic cations (e.g., Ca, Mg, Fe) capable of binding free fatty acids and forming insoluble complexes, thereby potentially reducing fatty acid biodegradation. In anaerobic lagoons, sterols are an organic fraction of sludge that are resistant to bacterial degradation. In the case of fresh manure, fatty acids could represent a potential source of energy via the manufacture of biodiesel fuel, if efficient means for their extraction and transesterification can be devised.  相似文献
