Estuaries are marine areas at great contamination risk due to their hydrodynamic features. PAH are wide and ubiquitous contaminants with a high presence in these marine environments. Chemical analysis of sediments can provide information, although it does not give a direct measure of the toxicological effect of such contaminants in the biota. Samples of Venerupis pullastra, Cerastoderma edule, and Mytilus galloprovincialis were collected from two locations in Corcubión estuary (Norhwest of Spain). The level of PAH in sediment and biota, and its possible origin were assessed. A moderate level of contamination was observed with a predominance of PAH of a pyrogenic origin. Genotoxic damage, measured as single-strand DNA breaks with the comet assay, was evaluated in gill tissue and in hemolymph. The values of DNA damage obtained showed a higher sensitivity of clams and cockles to the pollution load level. These differences among species make us suggest the use of some other species coupled with mussels as an optimal tool for biomonitoring estuarine environments.  相似文献

In this study, metal and metalloid concentrations and pathogens were measured in shellfish at different locations in a tropical estuary, including sites impacted by sewage and industry. Oyster, mangrove snails and mud snails did not exceed Australian and New Zealand Food Standards maximum levels for copper, lead or estimated inorganic arsenic at any site although copper concentrations in oysters and mud snails exceeded generally expected levels at some locations. Bacterial community composition in shellfish was species-specific regardless of location and different to the surrounding water and sediment. In the snails Telescopium telescopium, Terebralia palustris and Nerita balteata, some bacterial taxa differed between sites, but not in Saccostrea cucullata oysters. The abundance of potential human pathogens was very low and pathogen abundance or diversity was not associated with site classification, i.e. sewage impact, industry impact and reference.  相似文献

Biomonitoring of contaminants (metals, organotins, polyaromatic hydrocarbons (PAHs), PCBs) was undertaken in Milford Haven Waterway (MHW) and a reference site in the Tywi Estuary (St Ishmael/Ferryside) during 2007–2008. Bioindicator species encompassed various uptake routes—Fucus vesiculosus (dissolved contaminants); Littorina littorea (grazer); Mytilus edulis and Cerastoderma edule (suspension feeders); and Hediste (=Nereis) diversicolor (sediments). Differences in feeding and habitat preference have subtle implications for bioaccumulation trends though, with few exceptions, contaminant burdens in MHW were higher than the Tywi reference site, reflecting inputs. Elevated metal concentrations were observed at some MHW sites, whilst As and Se (molluscs and seaweed) were consistently at the higher end of the UK range. However, for most metals, distributions in MH biota were not exceptional. Several metal-species combinations indicated increases in bioavailability upstream, which may reflect the influence of geogenic/land-based sources—perhaps enhanced by lower salinity. TBT levels in MH mussels were below OSPAR toxicity thresholds and in the Tywi were close to zero. Phenyltins were not accumulated appreciably in M. edulis, whereas some H. diversicolor populations appear subjected to localized (historical) sources. PAHs in H. diversicolor were distributed evenly across most of MHW, although acenaphthene, fluoranthene, pyrene, benzo(a)anthracene and chrysene were highest at one site near the mouth; naphthalenes in H. diversicolor were enriched in the mid-upper Haven (a pattern seen in M. edulis for most PAHs). Whilst PAH (and PCB) concentrations in MH mussels were mostly above reference and OSPAR backgrounds, they are unlikely to exceed ecotoxicological thresholds. Bivalve Condition indices (CI) were highest at the Tywi reference site and at the seaward end of MH, decreasing upstream—giving rise to several significant (negative) relationships between CI and body burdens. Despite the possible influence of salinity gradient as a complicating factor, multivariate analysis indicated that a combination of contaminants could influence the pattern in condition (and the biomarkers metallothionein and TOSC). Integrating bioaccumulation data with biological and biochemical endpoints is seen as a useful way to discriminate environmental quality of moderately contaminated areas such as MHW and to prioritise cause and effect investigations.  相似文献

An alkaline comet assay and a micronucleus test were carried out on erythrocytes of the European chub, Squalius cephalus L., collected in spring and autumn in 2005 and 2006 at three sampling sites in River Sava, near Zagreb, Croatia. The results of comet assay showed the lowest genotoxic influence at the least polluted site, while higher DNA damage was observed at the polluted sites. Although the basal levels of DNA damage were elevated, a clear gradation of DNA damage was found due to pollution intensity in all sampling periods. The lowest cytogenetic damage as revealed by the micronucleus test (MNT) was observed as well at the least polluted site. High variations in MN frequency were observed between sampling periods, although the number of micronucleated erythrocytes was consistently the highest one at the polluted site. The comet assay as a biomarker of genotoxic effect exhibited higher sensitivity in discriminating the genotoxic capacity of studied polluted sites while the MNT was less sensitive. However, both tests should be used together in biomonitoring studies because they can reveal different aspects of DNA damage; comet assay, the early event of genotoxic exposure, and MNT, its final result as a mutagenic potential.  相似文献

The amount of the trace elements As, Ba, Cd, Cr, Cu, Hg, Li, Mn, Ni, Pb, Rb, Se, Sr, and Zn was measured in top soils and edible mushrooms, Boletus edulis, Macrolepiota procera, collected at five distinct green microhabitats inside the Lucca province, North-Central Italy (years 2008–2009). Results showed a top soil element content within the Italian statutory limits. Concerning the amount of mushroom elements, we observed significant species-differences obtaining higher levels of Ni, Rb, and Se in B. edulis or As, Pb, Cu in M. procera. Bioaccumulation factors (BCFs: element in mushroom/element in soil) resulted species-dependent and element-selective: in particular, B. edulis preferentially accumulated Se (BCFs varying from 14 to 153), while M. procera mainly concentrated Cu (BCFs varying from 5 to 15). As well, both species displayed between-site BCF differences. By a multivariate principal component approach, cluster analysis (CA), we could resolve two main clusters of soil element composition, corresponding to the most ecologically divergent sites. Besides, CA showed no cluster relating to element contents of B. edulis at the different collection sites, while a separation in groups was found for M. procera composition with respect to harvesting locations, suggesting uptake systems, in this saprotrophic species, sensitive to microhabitat. Regarding consumer safety, Cd, Hg, Pb levels resulted sometime relevant in present samples, never reaching values from current literature on mushrooms collected in urban-polluted areas. Our findings encourage a deeper assessment of the molecular mechanisms of metal intake by edible mushrooms, encompassing genetic biochemical and geo-ecological variables, with particular awareness to element bioavailability in soils and fungi.  相似文献

In Brazil, the state of São Paulo contains both preserved areas (Juréia-Itatins Ecological Station) and extremely impacted ones (Cubatão Municipality). This study evaluated the concentrations of five metals (Cu, Cd, Cr, Pb, and Hg) in two mangroves with different levels of anthropogenic impact and the apparent genotoxicity to Ucides cordatus. Water and sediment samples were obtained, and metal concentrations were determined with an atomic absorption spectrophotometer. The genotoxic impact was quantified based on the number of micronucleated cells per 1,000 analyzed (MN‰), using hemolymph slides stained with Giemsa. Metal concentrations in water were below the detection limit, except for lead, although no significant difference was observed between the areas (P?>?0.05). Sediment from Cubatão had higher concentrations of Cd, Pb, Cr, and Cu than sediment from Juréia-Itatins (P?<?0.05), but no significant differences in metal concentrations were detected among depth strata of the sediment (P?>?0.05). Crabs from Cubatão had a 2.6 times higher mean frequency of micronucleated cells (5.2?±?1.8 MN‰) than those from Juréia-Itatins (2.0?±?1.0 MN‰; P?<?0.0001). The more-polluted conditions found in the mangrove sediments of Cubatão were reflected in the micronucleus assay, demonstrating their genotoxic effect; however, genetic damage should be attributed to a synergistic effect with other kinds of pollutants previously recorded in different environments of Cubatão. U. cordatus proved to be an excellent bioindicator of mangrove pollution. This study established, for the first time, the normal frequency of MN‰ in a population of this species within an ecological station.  相似文献

Sediments and the associated biota constitute an important compartment in the biogeochemical cycle of trace metals in soft substrate megatidal estuaries. The relationship between physicochemical, ecological properties, metal concentrations determined in megatidal estuary sediments from the French coast of the English Channel, the Baie des Veys and the macrobenthic organisms, are analysed, interpreted and reported. Total concentrations of zinc, cadmium, lead and copper were measured in mudflats and saltmarsh sediments using Differential Pulse Voltammetry. Sediment characteristics were obtained by measurement of particle size, water content, total organic carbon and carbonate content, using AFNOR standards. A semi-quantitative scale was used for assessing the density of the macrobenthic flora and fauna at each sampling site. Analysis of data obtained from this study showed a correlation between the concentrations of monitored trace metals and species of the macrobenthic fauna. The results of this study show that the physico-chemical characteristics of the sediments affect the retention of metals in the sediment and this in turn affects the biota.  相似文献

The concentrations of Cd, Cu, Zn, Fe, Pb, Ni, and Co were measured in gastropod mollusks Patella caerulea in the Mediterranean area. The organisms were collected at two coastal sites in Iskenderun Gulf during winter, spring, summer, and autumn 2008. Samples of the digestive gland, gill, and muscle were analyzed for heavy metals. The aim of study is to determine heavy metal levels in tissues of P. caerulea in different locations. Tissues of P. caerulea from the polluted site showed metal concentrations appreciably higher than unpolluted organisms. The highest metal levels were registered in the digestive gland of P. caerulea. Generally, digestive gland and gills showed higher metal concentrations than muscle. The average concentrations of heavy metals analyzed exhibited the following decreasing order: Fe > Zn > Cu > Ni > Cd > Pb > Co for both stations 1 and 2. Seasonal changes in metal concentrations were observed in the tissues of P. caerulea from a polluted and an unpolluted population.  相似文献

The distribution and accumulation of heavy metals in the sediments, especially those nearest of wastewater discharges of south of Spain, were investigated. Sediment samples from 14 locations were collected and characterised for metal content (e.g. Ni, Cu, Zn, Cr, Pb, Mn, Cd and Hg), organic carbon, total nitrogen, total phosphorous, n-hexane-extractable material, carbonates and grain size. Concentration data were processed using correlation analysis and factor analysis. The correlation analysis of concentrations data showed important positive correlations among organic carbon, total phosphorus, Cu, Zn, Cd and Hg, otherwise weak correlations among Mn, Cr, Ni and CO3 2???, indicating that these metals have complicated geochemical behaviours. The use of statistical factor analysis also confirmed these results. Sediments pollution assessment was carried out using geoaccumulation and metal pollution indexes (MPI8). The results revealed that sediments of Cádiz bay and Sancti Petri channel were uncontaminated with the studied metals.  相似文献

Sediment quality data provide essential information for evaluating ambient environmental quality conditions. Sediments are important carriers of trace metals in the environment and reflect the current quality of the system. In the present study, distribution of mercury, lead, cadmium, copper, zinc, chromium and manganese in Cochin estuary were studied. The distribution of oxides of metals and textural quality were also studied in detail. It was found that the concentration of metals in the sediments near the industrial belt was extremely high. Correlation of different metals and metal oxides were analysed. It was found that all the alloys were correlated significantly (α?<?0.01) but in case of metals, correlation was only among certain metals.  相似文献

Soil, rock and water samples were collected from India??s oldest coalfield Raniganj to investigate trace metal contamination from mining activity. Our data reveal that trace metal concentration in soil samples lies above the average world soil composition; especially, Cr, Cu, Ni and Zn concentrations exceed the maximum allowable concentration proposed by the European Commission for agricultural soils. In particular, Cr, Cu and Ni exceed the ecotoxicological limit, and Ni exceeds the typical value for cultivated soils. Mineral dissolution from overburden material and high adsorption capacity of laterite soil are responsible for the elevated concentrations. This is evident from enrichment factor (E f), geoaccumulation index (I geo) and metal pollution index values. Sediment quality guideline index indicates toxicity to local biota although enrichment index suggests no threat from consuming crops cultivated in the contaminated soil.  相似文献

Karasu River, which is the only river in the Erzurum plain, is the source of the Euphrates River (Eastern Anatolia of Turkey). The river is in a serious environmental situation as a result of pollution by agricultural and industrial sewage and domestic discharges. The present study aims to evaluate genotoxic effects of toxic metals in chub, Leuciscus cephalus, and transcaucasian barb, Capoeta capoeta, collected from contaminated site of the Karasu River, in comparison with fish from an unpolluted reference site. Heavy metal concentrations in surface water of the river were determined. The condition factor (CF) was taken as a general biomarker of the health of the fish, and genotoxicity assays such as micronucleus (MN) and other nuclear abnormalities (NA) were carried out on the fish species studied. MN and NA such as kidney-shaped nucleus, notched nucleus, binucleated, lobed nucleus, and blebbed nucleus were assessed in peripheral blood erythrocytes, gill epithelial cells, and liver cells of the fish. A significant decrease in CF values associated with a significant elevation in MN and NA frequencies was observed in fish collected from the polluted sites compared with those from the reference site. Results of the current study show the significance of integrating a set of biomarkers to identify the effects of anthropogenic pollution. High concentrations of heavy metals have a potential genotoxic effects, and the toxicity is possibly related to industrial, agricultural, and domestic activities.  相似文献

The presence of pharmaceuticals in the environment is now a major concern given their potential adverse effects on organisms, particularly human beings. Because the feeding style and habitat of the crab Carcinus maenas make this species vulnerable to organic contaminants, it has been used previously in ecotoxicological studies. Lysosomal membrane stability (LMS) in crabs is a general indicator of cellular well-being and can be visualized by the neutral red retention (NRR) assay. LMS in crab hemolymph has been evaluated as a cellular biomarker of adverse effects produced by exposure to pharmaceutical compounds. Crabs were exposed in the laboratory to four different pharmaceuticals for 28 days in a semistatic 24-h renewal assay. Filtered seawater was spiked every 2 days with various concentrations (from 0.1 to 50 μg·L?1) of caffeine, ibuprofen, carbamazepine, and novobiocin. Results showed that NRR time, measured at day 28, was significantly reduced (p?<?0.05) after exposure to environmental concentrations of each pharmaceutical (caffeine?=?15 μg·L?1; carbamazepine?=?1 μg·L?1; ibuprofen?=?5 μg·L?1; and novobiocin?=?0.1 μg·L?1) when compared with control organisms. The predicted “no environmental effect” concentration/measured environmental concentration results showed that the selected pharmaceuticals are toxic at environmental concentrations and need further assessment. LMS monitoring in crabs is a sensitive tool for evaluating exposure to concentrations of selected drugs under laboratory conditions and provides a robust tier 1 testing approach (screening biomarker) for rapid assessment of marine pollution and environmental impact assessments for analyzing pharmaceutical contamination in aquatic environments.  相似文献

The influence of the Nyanza Chemical Waste Dump Superfund Site on the Sudbury River, Massachusetts, was assessed by analysis of sediment, fish prey organisms, and predator fish from four locations in the river system. Whitehall Reservoir is an impoundment upstream of the site, and Reservoir #2 is an impoundment downstream of the site. Cedar Street is a flowing reach upstream of the site, and Sherman Bridge is a flowing reach downstream of the site. Collections of material for analysis were made three times, in May, July, and October. Sediment was analyzed for acid-volatile sulfide (AVS), simultaneously-extracted (SEM) metals (As, Cd, Cr, Hg, Pb, Sb, Zn), and total recoverable Hg. The dominant predatory fish species collected at all sites, largemouth bass (Micropterussalmoides), was analyzed for the same suite of metals as sediment. Analysis of stomach contents of bass identified smallfish (yellow perch Perca flavescens, bluegill Lepomismacrochirus, and pumpkinseed Lepomis gibbosus), crayfish,and dragonfly larvae as the dominant prey organisms. Samples of the prey were collected from the same locations and at the sametimes as predator fish, and were analyzed for total and methyl mercury. Results of AVS and SEM analyses indicated that sediments were not toxic to aquatic invertebrates at any site. The SEM concentrations of As, Cd, and Cr were significantly higher at Reservoir #2 than at the reference sites, and SEM As and Cdwere significantly higher at Sherman Bridge than at Cedar St. Sediment total Hg was elevated only at Reservoir #2. Hg washigher at site-influenced locations in all fish speciesexcept brown bullhead (Ameiurus nebulosus). Cd washigher in bluegill, black crappie (Pomoxis nigromaculatus),and brown bullhead, and Cr was higher in largemouth bass filletsamples but not in whole-body samples. There were no seasonal differences in sediment or prey organism metals, but some metalsin some fish species did vary over time in an inconsistent manner. Predator fish Hg concentration was significantly linearlyrelated to weighted prey organism methyl Hg concentration. Largemouth bass Hg was significantly lower at Reservoir #2 in our study than in previous investigations in 1989 and 1990. High concentrations of inorganic Hg remain in river sediment asa result of operation of the Nyanza site, and fish Hg concentrations in river reaches downstream of the site areelevated compared to upstream reference sites. However, thedifferences are relatively small and Hg concentrations inlargemouth bass from the site-influenced locations are nohigher than those from some other, nearby uncontaminatedsites. We hypothesize that this results from burial ofcontaminated sediment with cleaner material, which reducesbioavailability of contaminants and possibly reducesmethylation of mercury.  相似文献

This study investigated dispersion and bioaccumulation of mining-related elements from an open-pit olivine mine at Seqi in Southwest Greenland (64°?N) using lichens (Flavocetraria nivalis), seaweeds (Fucus vesiculosus), mussels (Mytilus edulis) and fish (Myoxocephalus scorpius). The mine operated between 2005 and 2009, and samples were taken every year within a monitoring area 0–17 km from the mine during the period 2004–2011. A total of 46 elements were analysed in the samples. After mining began, highly elevated metal concentrations, especially nickel (Ni), chromium (Cr), iron (Fe) and cobalt (Co), were observed in lichens relative to pre-mining levels (up to a factor of 130) caused by dust dispersion from the mining activity. Elevated metal concentrations could be measured in lichens in distances up to ~5 km from the mine/ore treatment facility. Moderately elevated concentrations of Ni and Cr (up to a factor of 7) were also observed in seaweeds and mussels but only in close vicinity (<1 km) to the mine. Analyses of fish showed no significant changes in element composition. After mine closure, the elevated metal concentrations in lichens, seaweeds and mussels decreased markedly, and in 2011, significantly elevated metal concentrations could only be measured in lichens and only within a distance of 1 km from the mine.  相似文献

This study identifies and quantifies the spatial variations of metal contamination in water, sediment and biota: the common cockle (Cerastoderma edule) and the Mermaid’s glove sponge (Haliclona oculata), within a heavily anthropogenically impacted semi-enclosed estuarine–coastal area with a low ability to disperse and flush contaminants (Poole Harbour, UK). The results showed that metal contamination was detected in all environmental compartments. Water was polluted with As, and Hg sediment metals were mostly within “the possible effect range” in which adverse effects occasionally occurs. Cockles had considerable concentrations of Ni, Ag and Hg in areas close to pollution sources, and sponges accumulate Cu and Zn with very high magnitude. A systematic monitoring approach that includes biological monitoring techniques, which covers all embayments, is needed, and an integrated management of the semi-enclosed coastal zones should be based on the overall hydrological characteristics of these sensitive areas and their ability to self‐restore which is different than open coastal zones.  相似文献

An intertidal site in the Clyde Estuary, UK, was selected to evaluate the role of sediment geochemistry on the bioaccumulation of polychlorinated biphenyls (PCBs) by mussels (Mytilus edulis). The area had previously been identified as showing anomalously high levels of PCB contamination (over 1,500 microg kg(-1) total PCB in sediment, 22 congeners). Samples of surface sediment and M. edulis were collected from two closely located sites, one within the anomalous area and another representing typical PCB contamination in the estuary. Sediment samples were separated into grain size fractions and analysed for a range of biomarker compounds, PCBs and sediment mineralogy. The anomalous site showed an atypical association of PCBs with sediment properties, despite both locations showing influence of both petrogenic and pyrogenic organic contamination. Interrogation of data using correlation and principal component analysis showed that sediment mineralogy as well as organic matter composition influenced PCB congener distribution. One sediment source was found to control the PCB concentration in mussels at both locations and clay mineralogy appears to control PCB uptake by biota with preference for higher molecular weight congeners. Overall bioavailability is determined by sediment TOC.  相似文献

Several mutagenic agents may be present in substances released in the environment, which may cause serious environmental impacts. Among these substances, there is a special concern regarding the widespread use of silver nanoparticles (AgNP) in several products due to their widely known bactericidal properties, including in the medical field and the food industry (e.g., active packaging). The assessment of the effects of AgNP released in the environment, having different concentrations, sizes, and being associated or not to other types of materials, including polymers, is therefore essential. In this research, the objective was to evaluate the genotoxic and cytotoxic effects of AgNP (size range between 2 and 8 nm) on root meristematic cells of Allium cepa (A. cepa). Tests were carried out in the presence of colloidal solution of AgNP and AgNP mixed with carboxymethylcellulose (CMC), using distinct concentrations of AgNP. As a result, when compared to control samples, AgNP induced a mitotic index decrease and an increase of chromosomal aberration number for two studied concentrations. When AgNP was in the presence of CMC, no cytotoxic potential was verified, but only the genotoxic potential for AgNP dispersion having concentration of 12.4 ppm.  相似文献

The glyphosate-based herbicide, Roundup®, is one of the most used pesticides worldwide. In concert with the advent of transgenic crops resistant to glyphosate, the use of this pesticide has led to an increase in agricultural yields. The objective of this study was to evaluate the genotoxic effect that the herbicide Roundup® (at a concentration of 6.67 μg/L, corresponding to 3.20 μg/L glyphosate) can have on the fish Corydoras paleatus. Treatment groups were exposed for 3, 6, and 9 days, and effects were analyzed using the piscine micronucleus test (PMT) and comet assay. A group subjected to filtered water only was used as a negative control. The PMT did not show differences between the control and exposed groups for any of the treatment times. In contrast, the comet assay showed a high rate of DNA damage in group exposed to Roundup® for all treatment times, both for blood and hepatic cells. We conclude that for the low concentration used in this research, the herbicide shows potential genotoxic effects. Future research will be important in evaluating the effects of this substance, whose presence in the environment is ever-increasing.  相似文献

The concentrations of six metals (Ag, Cd, Cu, Hg, Pb, and Zn) were investigated and compared in three tissues (arms, digestive gland, and mantle) of three cephalopod species from the Tunisian waters: the common octopus (Octopus vulgaris), the common cuttlefish (Sepia officinalis), and the European squid (Loligo vulgaris). Whatever the species or the sites, the digestive gland displayed the highest concentrations of Ag, Cd, Cu, Pb, and Zn, highlighting its major role in their bioaccumulation and detoxification. This is also true for Hg but only for the digestive gland of O. vulgaris. Muscle from the arms and the mantle contained thus relatively low trace metal concentrations except for Hg in L. vulgaris and S. officinalis. Geographic comparison of metal concentrations in Tunisian cephalopods from three locations indicates that higher concentrations of Ag, Pb, and Hg were observed in cephalopods from northern and eastern coasts, whereas the highest Cd levels were detected in the southeastern, reflecting different conditions of exposure. Comparing the trace element concentrations between species, Ag, Cd, Cu, Hg, and Zn concentrations were the highest in the digestive gland of octopuses. This may be related to the differences in ecological features and swimming behavior among different cephalopod species. Effects of length and sex on metal levels were also considered, indicating a limited influence of sex on metal concentration.  相似文献
