The disruption of mutualisms between plants and mycorrhizal fungi is a potentially powerful mechanism by which invasives can negatively impact native species, yet our understanding of this mechanism's role in exotic species invasion is still in its infancy. Here, we provide several lines of evidence indicating that invasive tamarisk (Tamarix sp.) negatively affects native cottonwoods (Populus fremontii) by disrupting their associations with arbuscular mycorrhizal (AM) and ectomycorrhizal (EM) fungi. At a field site in the early stages of tamarisk invasion, cottonwoods with tamarisk neighbors had reduced EM colonization and altered EM fungal community composition relative to cottonwoods with native neighbors, leading to reductions in EM propagule abundance in the soil beneath tamarisk. Similarly, AM colonization of cottonwoods was reduced with a tamarisk neighbor, but there were no significant changes in AM fungal spore communities or propagule abundance. Root colonization by nonmycorrhizal fungi, including potential pathogens, was higher in cottonwoods with tamarisk neighbors. A greenhouse experiment in which AM and EM inoculation and plant neighbor were manipulated in a fully factorial design showed that cottonwoods benefited from mycorrhizas, especially EM, in terms of shoot biomass when grown with a conspecific, but shoot biomass was similar to that of nonmycorrhizal controls when cottonwoods were grown with a tamarisk neighbor. These results are partially explained by a reduction in EM but not AM colonization of cottonwoods by a tamarisk neighbor. Tamarisk neighbors negatively affected cottonwood specific leaf area, but not chlorophyll content, in the field. To pinpoint a mechanism for these changes, we measured soil chemistry in the field and the growth response of an EM fungus (Hebeloma crustuliniforme) to salt-amended media in the laboratory. Tamarisk increased both NO3- concentrations and electrical conductivity 2.5-fold beneath neighboring cottonwoods in the field. Salt-amended media did not affect the growth of H. crustuliniforme. Our findings demonstrate that a nonnative species, even in the early stages of invasion, can negatively affect a native species by disrupting its mycorrhizal symbioses. Some of these changes in mycorrhizal fungal communities may remain as legacy effects of invasives, even after their removal, and should be considered in management and restoration efforts.

Dispersal limitation plays an important role in a number of equilibrium and nonequilibrium theories about community ecology. In this study we use the framework of island biogeography to look for evidence of dispersal limitation in ectomycorrhizal fungal assemblages on "tree islands," patches of host trees located in a non-host vegetation matrix. Because of the potentially strong effects of island area on species richness and immigration, we chose to control island size by sampling tree islands consisting of a single host individual. Richness on tree islands was high, with estimates ranging up to 42 species of ectomycorrhizal fungi associating with a single host individual. Species richness decreased significantly with increasing isolation of tree islands, with our regression predicting a 50% decrease in species richness when tree islands are located distances of approximately 1 km from large patches of contiguous forests. Despite the fact that fungal fruit bodies produce large numbers of spores with high potential for long-distance travel, these results suggest that dispersal limitation is significant in ectomycorrhizal assemblages. There were no discernible effects of isolation or environment on the species identity of tree island fungal colonists. In contrast to the highly predictable patterns of tree island colonization we observed in a previous study on early successional forests, we suggest that over longer time periods the community assembly process becomes more dominated by stochastic immigration and local extinction events.

The extent to which interspecific plants share mycorrhizal fungal communities depends on the specificity of the symbiosis. For tropical forest tree seedlings, colonization by mycorrhizal fungi associated with established vegetation could have important consequences for survival and growth. I used a novel molecular technique to assess the potential for sharing of mycorrhizas in forest and pasture in southern Costa Rica, by identifying arbuscular mycorrhizal (AM) fungi in roots of the forest canopy tree species Terminalia amazonia, pasture grasses Urochloa ruziziensis and U. decumbens, and seedlings of T. amazonia planted into experimental reforestation plots. I tested the hypotheses that experimental seedlings were colonized either by the AM fungal community of the forest T. amazonia (suggesting host specificity) or of Urochloa (suggesting absence of specificity/importance of local environment). After two years, pasture-grown T. amazonia seedlings were colonized by neither community, but rather by a species of Glomus that was rarely observed on the other plants. These results suggest that conspecific seedlings planted into existing vegetation generate a distinct mycorrhizal community that may influence competitive interactions and the relative costs and benefits of the AM fungal symbiosis at early stages in the life cycle of tropical trees.

The experiment was undertaken to test the efficiency of inoculation of vesicular-arbuscular mycorrhizal (VAM) fungi on the seedling growth of three Pistacia species used as rootstocks. The stratified Pistacia seeds were inoculated with VAM fungi. The highest rate of inoculated roots was 96.7% in P. khinjuck seedlings with G. clarum and G. etunicatum, 83.3% in P. vera seedlings with G. caledonium and 73.3% in P. terebinthus seedlings with G. caledonium. Mycorrhizal inoculations improved seedling height only in P. terebinthus. Certain mycorrhizal inoculations increased the leaf N, but not P and K contents. Seedlings inoculated with G. caledonium had higher reducing sugar contents. It was concluded that pre-inoculated Pistacia seedlings could have a better growth in the harsh field conditions.

Assisted migration of forest trees has been widely proposed as a climate change adaptation strategy, but moving tree populations to match anticipated future climates may disrupt the geographically based, coevolved association suggested to exist between host trees and ectomycorrhizal fungal (EMF) communities. We explored this issue by examining the consistency of EMF communities among populations of 40 year-old Douglas-fir (Pseudotsuga menziesii var. menziesii) trees in a common-garden field trial using four provenances from contrasting coastal climates in southwestern British Columbia. Considerable variation in EMF community composition within test sites was found, ranging from 0.38 to 0.65 in the mean similarity index, and the divergence in EMF communities from local populations increased with site productivity. Clinal patterns in colonization success were detected for generalist and specialist EMF species on only the two productive test sites. Host population effects were limited to EMF species abundance rather than species loss, as richness per site averaged 15.0 among provenances and did not differ by transfer extent (up to 450 km), while Shannon's diversity index declined slightly. Large differences in colonization rates of specialist fungi, such as Tomentella stuposa and Clavulina cristata, raise the possibility that EMF communities maladapted to soil conditions contributed to the inferior growth of some host populations on productive sites. The results of the study suggest locally based specificity in host-fungal communities is likely a contributing factor in the outcome of provenance trials, and should be a consideration in analyzing seed-transfer effects and developing strategies for assisted migration.

Ectomycorrhizal fungal tissues comprise a significant forest-litter pool. Ectomycorrhizal (EM) fungi may also influence the decomposition of other forest-litter components via competitive interactions with decomposer fungi and by ensheathing fine roots. Because of these direct and indirect effects of ectomycorrhizal fungi, the factors that control the decomposition of EM fungi will strongly control forest-litter decomposition as a whole and, thus, ecosystem nutrient and carbon cycling. Some have suggested that chitin, a component of fungal cell walls, reduces fungal tissue decomposition because it is relatively recalcitrant. We therefore examined the change in chitin concentrations of EM fungal tissues during decomposition. Our results show that chitin is not recalcitrant relative to other compounds in fungal tissues and that its concentration is positively related to the decomposition of fungal tissues. Variation existing among EM fungal isolates in chitin concentration suggests that EM fungal community structure influences C and nutrient cycling.

Mutualistic interactions with fungal endophytes and dinitrogen-fixing bacteria are known to exert key biological influences on the host plant. The influence of a fungal endophyte alkaloid on the toxicity of a plant has been documented in Oxytropis sericea. Oxytropis sericea is a perennial legume responsible for livestock poisoning in western North America. Livestock poisoning is attributed to the alkaloid swainsonine, which is synthesized inside the plant by the fungal endophyte Embellisia sp. In this study, the ability of Oxytropis sericea to form a dinitrogen-fixing symbiosis with Rhizobium and the effects of this symbiosis on the production of swainsonine by Embellisia sp. were evaluated in a greenhouse environment. Seeds of O. sericea were grown in plastic containers. Twenty-week-old O. sericea seedlings were inoculated with four strains of Rhizobium. Twenty weeks after inoculation, plant growth and root nodulation by Rhizobium were measured. Dinitrogen fixation was confirmed using an acetylene reduction assay (ARA) on excised root nodules. Dry leaves were analyzed for swainsonine content. A second set of plants was treated with fungicide to evaluate the effect of reduced fungal endophyte infection on plant growth and swainsonine production. All inoculated plants produced indeterminate nodules. The ARA indicated that 98% of the excised nodules were fixing dinitrogen. Rhizobium-treated plants had greater swainsonine concentrations than the non-inoculated controls. Plants that established from seeds treated with fungicide had lower biomass than non-fungicide-treated controls and plants treated with foliar fungicide. Plants treated with foliar fungicide and the controls had greater swainsonine concentrations than the plants that received seed fungicide. This greenhouse study is the first report of nodulation and dinitrogen fixation in O. sericea. It also demonstrates that dinitrogen fixation increases the production of swainsonine in O. sericea plants infected with Embellisia sp. Results from this study suggest that dinitrogen fixation affects swainsonine production and has the potential to support the symbiosis between Embellisia sp. and O. sericea when soil nitrogen is limited. Oxytropis sericea competitiveness appears to be facilitated by an ability to simultaneously associate with Rhizobium and a fungal symbiont.

Ectomycorrhizal fungi form symbioses with most temperate and boreal tree species, but difficulties in measuring carbon allocation to these symbionts have prevented the assessment of their importance in forest ecosystems. Here, I surveyed allocation patterns in 14 culture studies and five field studies of ectomycorrhizal plants. In culture studies, allocation to ectomycorrhizal fungi (NPPf) was linearly related to total belowground net primary production (NPPb) by the equation NPPf = 41.5% x NPPb - 11.3% (r2 = 0.55, P < 0.001) and ranged from 1% to 21% of total net primary production. As a percentage of NPP, allocation to ectomycorrhizal fungi was highest at lowest plant growth rates and lowest nutrient availabilities. Because total belowground allocation can be estimated using carbon balance techniques, these relationships should allow ecologists to incorporate mycorrhizal fungi into existing ecosystem models. In field studies, allocation to ectomycorrhizal fungi ranged from 0% to 22% of total allocation, but wide differences in measurement techniques made intercomparisons difficult. Techniques such as fungal in-growth cores, root branching-order studies, and isotopic analyses could refine our estimates of turnover rates of fine roots, mycorrhizae, and extraradical hyphae. Together with ecosystem modeling, such techniques could soon provide good estimates of the relative importance of root vs. fungal allocation in belowground carbon budgets.

Investigating how arbuscular mycorrhizal fungi (AMF)-plant interactions vary with edaphic conditions provides an opportunity to test the context-dependency of interspecific interactions. The relationship between AMF and their host plants in the context of other soil microbes was studied along a gradient of heavy metal contamination originating at the site of zinc smelters that operated for a century. The site is currently under restoration. Native C3 grasses have reestablished, and C4 grasses native to the region but not the site were introduced. Interactions involving the native mycorrhizal fungi, non-mycorrhizal soil microbes, soil, one C3 grass (Deschampsia flexuosa), and one C4 grass (Sorghastrum nutans) were investigated using soils from the two extremes of the contamination gradient in a full factorial greenhouse experiment. After 12 weeks, plant biomass and root colonization by AMF and non-mycorrhizal microbes were measured. Plants from both species grew much larger in soil from low-contaminated (LC) origin than high-contaminated (HC) origin. For S. nutans, the addition of a non-AMF soil microbial wash of either origin increased the efficacy of AMF from LC soils but decreased the efficacy of AMF from HC soils in promoting plant growth. Furthermore, there was high mortality of S. nutans in HC soil, where plants with AMF from HC died sooner. For D. flexuosa, plant biomass did not vary with AMF source or the microbial wash treatment or their interaction. While AMF origin did not affect root colonization of D. flexuosa by AMF, the presence and origin of AMF did affect the number of non-mycorrhizal (NMF) morphotypes and NMF root colonization. Adding non-AMF soil biota reduced Zn concentrations in shoots of D. flexuosa. Thus the non-AMF biotic context affected heavy metal sequestration and associated NMF in D. flexuosa, and it interacted with AMF to affect plant biomass in S. nutans. Our results should be useful for improving our basic ecological understanding of the context-dependency of plant-soil interactions and are potentially important in restoration of heavy-metal-contaminated sites.

Polychlorinated biphenyls (PCBs) are typical organic contaminants in the environment. It is indicated that plants and soil microorganisms have a positive synergistic effect on the remediation of PCB-contaminated soil. To investigate the effect of intercropping on arbuscular mycorrhizal (AM) fungal colonization and PCB remediation, a pot-cultivation experiment with two intercropping treatments, corn (Zea mays L.) / ryegrass (Lolium perenne L.) and corn/alfalfa (Medicago sativa L.), and a corn monoculture was conducted in a greenhouse. All treatments were inoculated with Funneliformis mosseae M47V. Plant biomass, root mycorrhizal colonization rate, concentration of PCBs and their homologs in soil, 16S rDNA gene abundance, and community composition measured by Terminal Restriction Fragment Length Polymorphism (T-RFLP) were determined after harvesting the plants. Intercropping significantly increased the root mycorrhizal colonization rate and plant biomass of corn (P < 0.05), as well as the available N content of the soil. A significant difference of the bacterial community composition was found among different treatments (P < 0.05). Compared with corn monoculture, corn/alfalfa intercropping significantly increased soil bacteria abundance (P < 0.05). The dissipation rates of total PCBs, as well as that of penta-chloro biphenyls were significantly increased in the intercropping treatments, when compared to the corn monoculture treatment. Moreover, corn/ryegrass intercropping has a significantly positive effect on the dissipation of tri-chloro biphenyls. Non-metric multidimensional scaling (NMDS) analysis indicated that the PCBs homologues composition were significantly correlated with the relative abundance of 128 bp and 148 bp T-RFs. Corn intercropping with ryegrass or alfalfa has a positive effect on root mycorrhizal colonization rate and plant biomass of corn. Inoculation of AM fungi in intercropping treatments significantly improved the efficiency of PCB remediation by promoting bacterial abundance and shifting the bacterial community composition. In conclusion, intercropping combined with AM fungi have positive synergistic effects on the remediation of PCB-contaminated soils. © 2018 Science Press. All rights reserved.

Inoculation of azotobacter has significant positive effects on the growth characteristics and pigments in mangrove seedlings of Avicennia marina and Ceriops decandra. The bacterial inoculation significantly increased the root dry biomass at the maximum of 75.8% at 30 gl(-1) salinity in Ceriops decandra. But in Avicennia marina, the shoot dry biomass was increased significantly at the maximum of 56.12% at 30 gl(-1) salinity in general, the Azotobacter beijerinkii improved the growth characteristics better in both species of mangroves preferably at higher salinity levels in A. marina and at a range of salinity in C. decandra. The results recommend this forraising vigorous seedlings under nursery conditions.

Five medicinal plants viz. Abelmoschatus moschatus Linn., Clitoria tematea L., Plumbagozeylanica L., Psorolea corylifolia L. and Withania sominifera L. were grown in a polypot experiment in five soils representing coal mine soil, coppermine soil, fly ash, skeletal soil and forest soil with and without mycorrhizal inoculations in a completely randomized block design. Dry matter yield and mycorrhizal root colonization of plants varied both in uninoculated and inoculated conditions. The forest soil rendered highest dry matter due to higher yield of A. moschatus, P. zeylanica and P corylifolia while fly ash showed lowest dry matter without any inoculants. P. cematea were best in coalmine soil and W. sominifera in copper mine soil without mycorrhizal inoculation. The mycorrhiza was found to enhance the dry matter yield. This contributed minimum 0.19% to maximum up to 422.0% in different soils as compared to uninoculated plants. The mycorrhizal dependency was noticed maximum in plants grown in fly ash followed by coal mine soil, copper mine soil, skeletal soil and forest soil. The mycorrhizal response was increased maximum in W. sominifera due to survival in fly ash after inoculation followed by P corylifolia and P cematea. Percent root colonization in inoculated plant was increased minimum of 1.10 fold to maximum of 12.0 folds in comparison to un-inoculated plants . The native mycorrhiza fungi were also observed to colonize 4.0 to 32.0% roots in plants understudy. This study suggests that mycorrhizal inoculation increased the dry matter yield of medicinal plants in all soils under study. It also helps in survival of W. sominifera in fly ash.

A treatability study was conducted to determine the potential of white rot fungi to remediate soil from a Superfund site that had been contaminated with DDT. A tiered approach was utilized, starting with simple laboratory studies to screen the potential of white rot fungal strains to degrade DDT and culminating with a soil pan study that simulated land farming. Results from early tiers of the study indicated that Phanerochaete sordida had the best potential for remediating the soil. In the soil pan study, the fungus ultimately grew very well after second inoculation. However, the good growth did not translate to higher DDT removal compared to removal in pans that were not inoculated. 14[DDT] fate studies indicated the small amount of removal that did occur for both fungal inoculated and non‐inoculated conditions could be partially attributed to incorporation into humic material. Addition of a surfactant to the soil enhanced the removal of DDT in both inoculated and non‐inoculated soil. Consequently, under simulated land fanning conditions, this strain of fungus was not successful in remediating this soil.

Human activities release tremendous amounts of nitrogenous compounds into the atmosphere. Wet and dry deposition distributes this airborne nitrogen (N) on otherwise pristine ecosystems. This eutrophication process significantly alters the species composition of native grasslands; generally a few nitrophilic plant species become dominant while many other species disappear. The functional equilibrium model predicts that, compared to species that decline in response to N enrichment, nitrophilic grass species should respond to N enrichment with greater biomass allocation aboveground and reduced allocation to roots and mycorrhizas. The mycorrhizal feedback hypothesis states that the composition of mycorrhizal fungal communities may influence the composition of plant communities, and it predicts that N enrichment may generate reciprocal shifts in the species composition of mycorrhizal fungi and plants. We tested these hypotheses with experiments that compared biomass allocation and mycorrhizal function of four grass ecotypes (three species), two that gained and two that lost biomass and cover in response to long-term N enrichment experiments at Cedar Creek and Konza Long-Term Ecological Research grasslands. Local grass ecotypes were grown in soil from their respective sites and inoculated with whole-soil inoculum collected from either fertilized (FERT) or unfertilized (UNFERT) plots. Our results strongly support the functional equilibrium model. In both grassland systems the nitrophilic grass species grew taller, allocated more biomass to shoots than to roots, and formed fewer mycorrhizas compared to the grass species that it replaced. Our results did not fully support the hypothesis that N-induced changes in the mycorrhizal fungal community were drivers of the plant community shifts that accompany N eutrophication. The FERT and UNFERT soil inoculum influenced the growth of the grasses differently, but this varied with site and grass ecotype in both expected and unexpected ways suggesting that ambient soil fertility or other factors may be interacting with mycorrhizal feedbacks.

The present investigation was carried out to study the toxicity of heavy metals (Zn, Cd, Cu, Ni, Pb and Al) on the dehydrogenase activity of ectomycorrhizal (Suillus leutus, Scleroderma aurantium, Cenococcum graniforme and Boletus spp.) and non-mycorrhizal rhizospheric soil of pine seedlings. The treatment of heavy metals adversely affected the dehydrogenase activity. Inoculation of mixed ectomycorrhizal fungi harbored increased activity of dehydrogenase. It was observed that in absence of ectomycorrhizae, heavy metals drastically reduced the enzyme activity at higher concentration of metals.
