Livestock manure is suitable for use as a composting material. However, various intestinal microbes, such as Escherichia coli, are significant components of such manures. Thus, it is desirable that the level of intestinal microbes, and particularly opportunistic pathogens, in compost is inspected and counted regularly. The sensitivity and specificity of detection of E. coli in compost have been improved by selective cultivation followed by colony polymerase chain reaction (PCR) using the ECO primer. Indeed, the sensitivity of this method is higher than that of DNA extraction from compost and PCR. In this study, changes in numbers of E. coli present in a field-scale composting process over time was assessed using selective cultivation and colony PCR. Numbers of ECO-positive colonies after 24 h decreased, with a concomitant rise in compost temperature. ECO-positive colonies were not detected from 33 to 48 h. However, ECO-positive colony numbers increased beginning on day 4 and continuing until day 42. Thus, it seems likely that the high temperatures reached during the composting process did not affect E. coli numbers in the final compost. Additionally, selective cultivation followed by colony PCR using specific primers is an appropriate method of determining levels of cultivable pathogens in composted materials.  相似文献

The objectives of this research were to evaluate the potential for sudden increase and/or regrowth of alternative bacteria as either indicators or pathogens after dewatering of thermophilic and mesophilically digested biosolids. The results showed that, in general, for thermophilic processes, even when a statistically significant (p < 0.05) sudden increase and regrowth occurred for fecal coliforms, Escherichia coli, and Enterococci, it did not occur for Salmonella or Aeromonas. For the mesophilic process evaluated, sudden increase did not occur, but regrowth occurred for fecal coliforms, E. coli, Enterococci, and Salmonella. The results have implications for Class A and B biosolids regulations, as both fecal coliform and Salmonella are part of the regulatory limits. The results also suggest that the public health risks are minimal, as a result of the potential sudden increase and regrowth that may occur.  相似文献

This research investigated the effects of extending the holding time of samples for microbial analysis beyond the standard of 24 hours for purposes such as watershed characterization. Experiments were conducted with both sanitary wastewater and stormwater samples. The refrigerated samples (4 degrees C) were held for up to 9 days before being analyzed for two pathogens (Pseudomonas aeruginosa and Staphylococcus aureus) and five indicator organisms (total coliform, fecal coliform, fecal streptococcus, enterococcus, and Escherichia coli) by membrane filtration. The concentrations (as colony-forming units per 100 mL) were normalized by log10(transformation and used in subsequent statistical analysis testing for significant differences. The results suggested that the concentrations of microorganisms in water samples analyzed on days 1 and 2 did not vary significantly in 8 of 13 analyses. The results of a field study concluded that the concentration of fecal coliform did not change significantly between 7 hours holding time and greater than 24 hours holding time for fecal coliform.  相似文献

Escherichia coli (E. coli), enterococci, and fecal coliform data were collected and compared as potential indicators for swimmablility assessment of a brackish waterbody (Lake Pontchartrain, Louisiana). These indicators were measured during lake background conditions, in stormwater runoff (before dilution with lake water), and in the outfall plume within the lake following storm events. Microbial indicator titers associated with suspended particles and lake-bottom sediments were also investigated. Overall reduction rate constants for fecal coliform, E. coli, and enterococci in lake water and sediment were measured and reported. Attachment of microbial indicators to suspended matter and subsequent sedimentation appeared to be a significant fate mechanism. A slower reduction rate of indicator organisms in sediment further suggested that bottom sediment may act as a reservoir for prolonging indicator organism survival and added concern of recontamination of overlaying waters due to potential solids resuspension. Results indicated that enterococci might be a more stable indicator than E. coli and fecal coliform and, consequently, a more conservative indicator under brackish water conditions.  相似文献

Microbial contamination of fresh produce can present a severe risk to public health. By conducting a rigorous survey of irrigation waters, the impacts of fecal contamination on the quality of produce could be assessed. In this study, surface waters were observed to be contaminated with Escherichia coli, Salmonella spp., and somatic coliphages. Culture methods show that out of 373 irrigation water, soil, and vegetable samples collected for a 1-year period, 232 (62.20%) were found positive for E. coli, 213 (57.26%) for somatic coliphages, and 2 (0.53%) for Salmonella spp. Out of 190 water samples, 167 (87.9%) were found to have E.coli, 174 (91.6%) have somatic coliphages, and 1 (0.5%) with Salmonella spp. In soil samples, 36 of 91 (39.6%) have E. coli, 31 (34.0%) have somatic coliphages, and none with Salmonella spp. Lastly, out of 92 vegetable samples, 29 (31.5%), 8 (8.7%), and 1 (1.1%) were found to have E. coli, somatic coliphages, and Salmonella spp., respectively. Molecular analysis confirmed the presence of bacterial contaminants. Seasonal weather conditions were noted to have an effect on the presence and number of these fecal indicator organisms. The observed data suggest that contaminated irrigation water may greatly affect the quality of fresh produce from these agricultural operations.  相似文献

The in situ survival and activity of Streptococcus faecalis and Escherichia coli were studied using membrane diffusion chambers in tropical marine waters receiving oil refinery effluents. Protein synthesis, DNA synthesis, respiration or fermentation, INT reduced per cell, and ATP per cell were used to measure physiological activity. Cell densities decreased significantly over time at both sites for both S. faecalis and E. coli; however, no significant differences in survival pattern were observed between S. faecalis and E. coli. Differences in protein synthesis between the two were only observed at a study site which was not heavily oiled. E. coli was more active in protein synthesis and respiration than S. faecalis at both oiled and unoiled sites, and the percentage of the E. coli population that was respiring was significantly higher than S. faecalis fermenting cells at both sites. However, S. faecalis cells were more active in DNA synthesis and higher in ATP content than E. coli cells at both sites. Although fecal streptococci have been suggested as a better indicator of fecal contamination than fecal coliforms in marine waters, in this study both E. coli and S. faecalis survived and remained physiologically active for extended periods of time. These results suggest that the fecal streptococci group is not a better indicator of fecal contamination in tropical marine waters than the fecal coliform group, especially when that environment is high in long-chained hydrocarbons.  相似文献

Applying manure compost not only results in zinc accumulation in the soil but also causes an increase in zinc mobility and enhances zinc leaching. In this study, the physical and chemical characteristics of zinc, zinc profiles, and zinc balance were investigated to characterise the fate of zinc in fields where the quality and amount of pig manure compost applied have been known for 13 years. Moreover, we determined zinc fractionation in both 0.1 mol L?1HCl-soluble (mobile) and -insoluble (immobile) fractions. Adsorption of zinc in the soil was enhanced with increasing total carbon content following the application of pig manure compost. The 159.6 mg ha?1 year?1manure applied plot (triplicate) exceeded the Japanese regulatory level after only 6 years of applying pig manure compost, whereas the 53.2 mg ha?1 year?1 manure applied plot (standard) reached the regulatory level after 13 years. The zinc loads in the plots were 17.0 and 5.6 kg ha?1 year?1, respectively. However, 5.9 % and 17.2 % of the zinc loaded in the standard and the triplicate pig manure compost applied plots, respectively, were estimated to be lost from the plough layer. Based on the vertical distribution of mobile and immobile zinc content, a higher rate of applied manure compost caused an increase in the mobile zinc fraction to a depth of 40 cm. Although the adsorption capacity of zinc was enhanced following the application of pig manure compost, a greater amount of mobile zinc could move downward through the manure amended soil than through non manure-amended soil.  相似文献

A load model for stormwater constituents is proposed that describes mass retained on the drainage area after previous storms, as a function of a weighted sum of previous antecedent dry periods. It was used to compute the event load for 14 constituents--zinc, copper, cadmium, nickel, chromium, lead, mercury, silver, total suspended solids (TSS), E. coli, fecal coliform, total soluble phosphorus, total phosphorus, and 5-day biochemical oxygen demand. The results show acceptable fits for most constituents based on over 400 rain events (2000 to 2004) in Milwaukee, Wisconsin. Considering the retained pollutant mass after one previous storm rather than assuming the area to be clean after the previous storm gives more accurate results for all constituents, except TSS and silver. The model can be used for mixed land-use areas. The ratios of deposition fluxes of zinc and individual metals were determined at a major parking lot producing a profile characteristic of automobile emissions.  相似文献

Recovering struvite from dairy manure has consistently posed problems for researchers. This study separated solids from anaerobically digested dairy manure using a filtration system. Filtrate was rich in free magnesium (160 to 423 mg/L), ammonium (320 to 1800 mg N/L) and orthophosphate (93 to 332 mg P/L). High concentrations of free calcium (128 to 361 mg/L) and alkalinity (3309 to 6567 mg/L as CaCO3), however, may hinder struvite precipitation. Batch precipitation tests were conducted to identify and overcome factors that interfere with struvite formation. Precipitation tests at pH 9 identified calcium and ionic strength as most probable interferences. Calcium addition did not significantly change phosphorus removal efficiency, but decreased struvite purity because of formation of calcium phosphates when Ca:P activity ratio was greater than 0.5 to 1. Batch tests demonstrated effective calcium removal from anaerobically digested dairy manure through precipitation of calcium carbonate at pH 9 to 10 while retaining magnesium and orthophosphate, lessening hindrance to struvite formation.  相似文献

Two experiments were conducted to evaluate the effect of compost addition to soil on fractionation and bioavailability of Cu, Mn, and Zn to four crops. Soils growing Swiss chard (Beta vulgaris var. cicla L.) and basil (Ocimum basilicum L.) were amended (by volume) with 0, 20, 40, and 60% Source-Separated Municipal Solid Waste (SS-MSW) compost, and dill (Anethum graveolens L.) and peppermint (Mentha X piperita L.) were amended with 0, 20, 40, and 60% of high-Cu manure compost (by volume). The SS-MSW compost applications increased the concentration of Cu and Zn in all fractions, increased Mn in acid extractable (ACID), iron and manganese oxides (FeMnOX), and organic matter (OM) fractions, but decreased slightly exchangeable-Mn. Addition of 60% high-Cu manure compost to the soil increased Cu EXCH, ACID, FeMnOX, and OM fractions, but decreased EXCH-Mn, and did not change EXCH-Zn. Addition of both composts to soil reduced bioavailability and transfer factors for Cu and Zn. Our results suggest that mature SS-MSW and manure composts with excess Cu and Zn could be safely used as soil conditioners for agricultural crops.  相似文献

Escherichia coli and total coliforms in water and sediments at lake marinas   总被引:2,自引:0,他引:2  
Escherichia coli, a fecal coliform, and total coliforms were monitored between September 1999 and October 2001 in five marinas at Lake Texoma, located on the Oklahoma and Texas border. The general trend was that densities of E. coli were lower in the summer season due to the lower loading of fecal material into Lake Texoma and the ecological conditions of the lake, such as more vigorous grazing by protozoa and less viability of E. coli at an elevated temperature. The densities of total coliforms greatly increased in the summer. E. coli levels increased with depth, and the bottom water samples had higher densities of E. coli mainly due to their association with particles. There was a direct relationship between amount of gasoline sold, which was related to recreational boating activity, and the resuspension of E. coli. This indicated that recreational boating activity in lake marinas may have resuspended bottom sediments with bound E. coli, and the presence of E. coli in marinas was not an indication of recent fecal contamination. E. coli were detected in the largest densities at the boat dock points, followed by the gasoline filling station, and marina entrance. In addition, enumeration of bacteria in bottom sediment showed that the densities of E. coli and total coliforms in sediment were much higher compared to those in lake water.  相似文献

This study examined physicochemical parameters to assess their effectiveness as stability and maturity indicators during the process of composting pig manure and fungus residue at different ratios. The results showed that composting mixtures with all ratios of pig manure to fungus residue maintained a temperature exceeding 50 °C for more than 10 days during composting and met the requirement for pathogen destruction. The treatment containing mainly pig manure showed higher nitrogen loss and a shorter thermophilic phase and maturity time than the treatment containing mainly fungus residue. The germination index (GI) values indicated that compost maturity was achieved in the final compost with initial ratios of pig manure to fungus residue of 9:1–7:3 (GIs of 101.4%, 91.2%, and 81.3%); the ratio of 6:4 did not reach compost maturity (GI of 63.8%) and had an inhibitory effect on seed germination. The results of this study suggest that a ratio of pig manure to fungus residue of approximately 8:2 can be considered suitable for the efficient and quality composting of pig manure and fungus residue.

Implications: Co-composting of pig manure and edible fungi residue with appropriate proportion can effectively reduce the risk of environmental pollution caused by agricultural wastes, as well as achieve a safer and high-quality organic fertilizer, which can be used to improve physical and chemical properties of the soil, increase crop yields, and promote agricultural sustainable development. Therefore, technique of co-composting of pig manure and edible fungi residue has a wide prospect of application in practical production all over the world.  相似文献

A laboratory pot experiment was conducted to study the effect of amending soil with four different sources of organic matter on the degradation rate of alpha and beta endosulfan isomers. Poultry by-product meal, poultry manure, dairy manure, and municipal solid waste compost were cured, dried, ground (<1 mm) and thoroughly mixed with a calcareous soil at a rate of 2% and placed in plastic pots. Endosulfan was added at the rate of 20 mg kg(-1). The moisture level was kept near field capacity and the pots were kept at room temperature. Soil sub-samples, 100 g each, were collected from every pot at days 1, 8, 15, 22, 29, 43, and 57 for the measurement of endosulfan isomers. Endosulfan residues were extracted from the soil samples with acetone. The supernatant was filtered through anhydrous sodium sulphate, 5 mL aliquot was diluted to 25 mL with hexane, mixed well, and then two sub-samples from the filtrates were analyzed for alpha and beta endosulfan isomers by gas chromatography. The results indicated that the half-life (T(1/2)) of alpha-endosulfan in the poultry by-product meal treatment was 15 days compared to about 22 days in the other treatments. The T(1/2) of beta-endosulfan was 22 days in the poultry by-product meal treatment and followed a bi-phasic pattern, 57 days in the municipal solid waste compost treatment and the extrapolated T(1/2) was about 115 days for the other three treatments.  相似文献

The goal of this study was to optimize the mixture of swine manure (SM) and cattle manure (CM) used in the vermicomposting process, seeking to increase the manure biodegradation rate and enhance the biomass production of both earthworms and higher plants. To achieve this goal, physico-chemical parameters were determined to assess the final compost quality after 50 days of vermicomposting. The different manure ratios used to produce the composts (C) were as follows (SM:CM, % m/m basis): C1 100:0, C2 (75:25), C3 (50:50), C4 (25:75), and C5 (0:100). In addition, the earthworm biomass and the phytoproductivity of lettuce (Lactuca sativa L.) plants grown in mixtures (1:1) of natural soil and the most viable vermicomposts were investigated. The C1 and C2 compost compositions were associated with high earthworm mortality rates. The C3 compost provided the highest mineral concentrations and C5 showed the highest lettuce yield (wet biomass). The results verify that stabilized cattle manure is an excellent substrate for the vermicomposting process and that fresh swine manure must be mixed with pre-stabilized cattle manure to ensure an optimized vermicomposting process, which must be controlled in terms of temperature and ammonia levels. It is concluded that small livestock farmers could add value to swine manure by applying the vermicomposting process, without the need for high investments and with a minimal requirement for management of the biodegradation process. These are important technical aspects to be considered when circular economy principles are applied to small farms.  相似文献

Two most probable number (MPN) methods-lauryl tryptose broth with Escherichia coli broth confirmation and direct A-1 broth incubation (A-1)--were compared for the enumeration of fecal coliform in lime-treated biosolid. Fecal coliform numbers were significantly higher using the A-1 method. Analysis of positive A-1 tubes, however, indicated that a high percentage of these were false positives. Therefore, the use of A-1 broth for 40 CFR Part 503 Pathogen Reduction (CFR, 1993) compliance testing is not recommended.  相似文献

The presence of heavy metals in composts is a main cause of adverse effects on animal and human health, transmitted through the food chain from the soil, groundwater and plants. In this study, the contents of Zn, Cd, Pb and Cu present in co-composts of poultry manure (liquid or solid) with a co-composting material (barley wastes or chestnut burr/leaf litter) were assessed. A compost of solid manure was used as control because a compost cannot be obtained from the liquid manure. The original solid poultry manure showed a Zn content of 2134+/-75 mg/kg, exceeding the current legal limit in Spain of 1100 mg/kg. In the solid poultry manure co-compost with chestnut burr/leaf litter and barley wastes, Zn content decreased to 813+/-25 mg/kg and 883+/-37 mg/kg, respectively. The contents in heavy metals (Zn, Cd, Pb and Cu) of the co-composts were under the maximum limit permitted under the Spanish legislation, excepting for the Zn level in liquid poultry manure co-composted with chestnut burr/leaf litter.  相似文献

Yard waste compost provides an organic amendment useful for improving soil structure and nutrient status. The activities of the enzymes hydrolyzing urea (urease) and sucrose (invertase) in the rhizosphere of potato plants were determined under field conditions. Soil urease and invertase activities were monitored in compost amended soil, grass buffer strips, and in adjacent bare soils during 35 d following soil treatment. Soil urease activity was increased by application of yard waste compost compared to untreated soil which provide evidence of increased soil microbial population following application of compost. Some transitional effects on urease activities were observed following Pyrethrins and Neemix-4E application, these effects were neither drastic nor prolonged enough to be considered deleterious to the soil microorganisms and their activities important to soil fertility. No relationship was found between invertase activity and the three management practices or soil organic matter content. It is suggested that application of botanical insecticides like pyrethrins and Neemix-4E may be useful in delaying urea fertilizer mineralization to maintain N in a form less leachable, so that the duration of N availability to plants is prolonged. The present study may also provide information on urease activity as a sensitive bioindicator of soil quality that reflects the effects of land management on soil quality and may assist land managers in monitoring long-term productivity and sustainability of agricultural lands.  相似文献

Land productivity can decline when top soil is lost. In Kentucky, limited resource farmers often produce vegetable crops on erodible lands. The objectives of this study were 1) to quantify the impact of three soil management practices (SMPs) on quantity of potato produced on erodible land, 2) to evaluate the impact of pyrethrin and azadirachtin insecticides on potato tuber quality, and 3) to assess the impact of yard waste compost on the chemical composition (ascorbic acid, free sugars, phenol contents) of potato tubers. Potatoes (Solanum tuberosum L. cv. Kennebec) were grown in a silty loam soil of 10% slope. Plots (n= 18) were 3.7 m wide and 22 m long (10% slope), with metal borders of 20 cm above ground level. Two botanical insecticides, Multi-Purpose Insecticide (containing pyrethrin 0.2%) and Neemix 4EC (containing 0.25% azadirachtin) were sprayed twice on potato foliage during each of two growing seasons (1997 and 1999) at the recommended rates of 6 lbs and 2 gallons.acre(-1), respectively. The SMPs were tall fescue strips (FS) intercropped between each two potato rows, soil mixed with yard waste compost (COM) and no-mulch (NM) treatment (roto-tilled bare soil). The experimental designwas a 2 x 3 x 3 factorial with main factors of two insecticides and three SMPs replicated three times. Average potato yield was lowest in NM and FS and highest in COM treatments. Yield obtained from the bottom of the plots was greater than that obtained from the top of plots. Tuber defects (rot, scab, sun green, hollow heart, necrosis, and vascular discoloration) were significantly different between the two growing seasons. The two insecticidal treatments did not have much influence on potato yield or tuber defects. Tubers obtained from tall fescue treatments had low levels of ascorbic acid and reducing sugars compared to compost treatments.  相似文献
