The effects of reproduction and maternal investment (i.e., milk transfer) on trace element levels remain poorly understood in marine mammals. We examined the blood dynamics of mercury (Hg) and selenium (Se) during lactation in the northern elephant seal (Mirounga angustirostris), a top predator from the North Pacific Ocean. Total Hg and Se levels were measured in whole blood and milk of 10 mother-pup pairs on days 5 and 22 of lactation. Both Hg and Se were transferred to offspring through the milk. Results suggested that the maternal transfer of Se was prominent during lactation, whereas the Hg transfer was larger during gestation. The lactation period affected Hg and Se levels in the blood of elephant seal mothers and pups. Physiological processes and their relationship to body condition should be considered carefully when interpreting trace element levels in the framework of biomonitoring.  相似文献

Serum and blubber of pup harbor (Phoca vitulina), grey (Halichoerus grypus) and harp (Phoca groenlandica) seals from the Estuary and the Gulf of St. Lawrence were analyzed for polybrominated diphenyl ethers (PBDEs). Concentrations of ∑PBDEs (on lipid weight basis) in tissues of harbor seal pups inhabiting the St. Lawrence Estuary were about five times higher than in those from a colony located in the northern Gulf. Harp seal pups have the lowest levels of ∑PBDEs among the seal species born in the Gulf of St. Lawrence. Lipid normalized concentrations of ∑PBDEs in serum and blubber were strongly correlated, suggesting that the blood collection as a less invasive method could be used to monitor ∑PBDEs in pups. This study shows that fluvial inputs of PBDEs are important to the St. Lawrence marine ecosystem and that inhabiting harbor seal pups have a substantial exposure to PBDEs at a critical developmental stage. In addition, the observed difference in PBDE levels between harp and grey or harbor seal pups from the Gulf of St. Lawrence is explained by the difference in diets of their mothers which is linked with their residency time in the Gulf and their seasonal migration pattern.  相似文献

Blubber biopsy and blood samples were collected from weaned grey seal pups and juveniles in 1998 and 1999. The relationship between the concentration of total PDBEs in the blubber and the circulating concentrations of serum proteins, minerals and thyroid hormones was investigated. Fifty-four post-weaned pups were sampled and 55 first year juveniles, of which 13 were recaptured post-weaned pups. The median concentration of total PBDEs in the pups' blubber was 170 and 460 ng g(-1) in the first-year animals. Total blubber PBDEs contributed significantly to the prediction of circulating thyroid hormone concentrations, after controlling for age and condition. Cholesterol and albumin concentrations were also positively related to blubber PBDEs. The results of this study suggest a link between thyroid hormones and exposure to PBDEs in grey seals during their first year of life. Such an association is not alone sufficient evidence for a cause and effect relationship, however it is in accordance with the hypothesis that these compounds are endocrine disrupters in grey seal pups.  相似文献

Pregnant Long Evans rats received 1.0 μg/kg of dioxin toxic equivalents (TEQ) by oral gavage on the 15th gestational day (GD 15), using a dosing mixture that contained two polychlorinated dioxins, four polychlorinated furans and three non-ortho polychlorinated biphenyls (PCBs). Rats were sacrificed on GD 16, GD 21 and postnatal day 4 (PND 4). The lipid content of fetus, pup, placenta and maternal liver, serum and adipose tissue were determined. Treated GD 16 and GD 21 fetuses had identical lipid content to the control group, yet the lipid content of treated pups on PND 4 was 32% higher than that of the control group. On the other hand, the lipid content of placenta, liver, and serum from the treated dams was 44–50%, 24%, and 38% lower than that of the control group, respectively. Polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs) were measured in blubber biopsy samples from 22 live-captured Steller sea lions (Eumetopias jubatus) that had just entered the Strait of Georgia, British Columbia, Canada, for their overwintering feeding season. ∑PBDE ranged from 50 μg kg−1 (lipid weight) in adult females to 3780 μg kg−1 in subadult individuals. ∑PCBs ranged from 272 μg kg−1 in adult females to 14 280 μg kg−1 in subadult individuals. While most PBDE and PCB congeners were transferred through milk to pups, PCBs with log KOW > 7.0 (PCBs 206, 207, 208 and 209) appeared constrained, resulting in a lighter mixture in pups compared to adult females. The ratio of individual PCB congeners by metabolic group (Groups I, II, III, IV and V) to PCB-153 regressed against length of males suggested poor biotransformation of these compounds (slopes did not differ from zero, > 0.05). PBDE congeners 49, 99, 153 and 183 appeared bioaccumulative (slopes of ratio BDE/PCB 153 versus length were higher than zero, < 0.05), but the dominance of the single congener, BDE-47 (64% of total PBDEs), likely due in part to debromination pathways, reduced our ability to explore congener-specific dynamics of PBDEs in these pinnipeds. With 80% of our Steller sea lions exceeding a recent toxicity reference value for PCBs, the fasting-associated mobilization of these contaminants raises concerns about a heightened vulnerability to adverse effects during annual migrations.  相似文献

During the 1995 hunting season, 75 harbour porpoises (Phocoena phocoena) were sampled in three locations in West Greenland: Maniitsoq, Nuuk, and Paamiut. Sex, age, morphometrics, reproductive condition, and organochlorine compound (OC) levels in blubber were determined for each individual. OC levels were extremely low and, therefore considered unlikely to affect the population adversely: mean blubber concentrations, expressed on lipid weight basis were 1.98 (S.D.=1.1) mg/kg for PCBs, 2.76 (S.D.=1.66) mg/kg for tDDT and 0.21 (S.D.=0.11) mg/kg for HCB. No statistical differences were observed among individuals caught in the various locations. OC concentrations showed statistically significant positive associations with age in males but negative in females; consequently, mature females presented lower pollutant loads than their male counterparts. Juveniles did not show differences between sexes. A higher proportion of less chlorinated and more metabolizable polychlorinated biphenyls (PCBs) compared to tPCBs was found in calves (age< or =1) than in mature females, indicating that the feeding habits of these two groups differ and that a greater transfer of less chlorinated compounds is passed from females to their pups through lactation and parturition. Harbour porpoises significantly contribute to the dietary intake of OCs by local Inuit populations. This contribution could be reduced if mature males were selectively avoided; however, current hunting procedures make this selection impracticable.  相似文献

We examined concentrations of total mercury (tHg, inorganic and methylated forms) and polychlorinated biphenyls (PCBs) in blood and milk from free-ranging Southern Beaufort-Chukchi Sea polar bears (Ursus maritimus) to assess maternal transfer of contaminants during lactation and the potential health risk to nursing young. Concentrations of contaminants in the blood of dependent and juvenile animals (ages 1-5 years) ranged from 35.9 to 52.2 μg kg(-1) ww for tHg and 13.9 to 52.2 μg kg(-1) ww (3255.81-11067.79 μg kg(-1) lw) for ΣPCB(7)s, similar to those of adult females, but greater than adult males. Contaminant concentrations in milk ranged from 5.7 to 71.8 μg tHg kg(-1)ww and 160 to 690 μg ΣPCB(11)s kg(-1) ww (547-5190 μg kg(-1) lw). The daily intake levels for tHg by milk consumption estimated for dependent young were below the tolerable daily intake level (TDIL) of tHg established for adult humans. Although the daily intake levels of PCBs through milk consumption for cubs of the year exceeded the TDIL thresholds, calculated dioxin equivalents for PCBs in milk were below adverse physiological thresholds for aquatic mammals. Relatively high concentrations of non-dioxin like PCBs in polar bear milk and blood could impact endocrine function of Southern Beaufort-Chukchi Sea polar bears, but this is uncertain. Transfer of contaminants during mid to late lactation likely limits bioaccumulation of dietary contaminants in female polar bears during spring. As polar bears respond to changes in their arctic sea ice habitat, the adverse health impacts associated with nutritional stress may be exacerbated by tHg and PCBs exposure, especially in ecologically and toxicologically sensitive polar bear cohorts such as reproductive females and young.  相似文献

Accumulation and mother-pup transfer of halogenated organic contaminants was studied in hooded seal tissues from eastern Canada. Blubber polychlorinated biphenyl (PCB) and total pesticide concentrations were relatively high, possibly due to their high trophic level and demersal feeding habits. Blood plasma showed the lowest contaminant concentrations compared to blubber and liver, possibly due to a lower affinity of these compounds to lipoproteins in blood plasma. Total contaminant body burden correlated well with blubber, liver, and milk contaminants, but not with blood plasma contaminants, indicating that blood plasma might be less suitable to monitor contaminants in hooded seals. Lactational transfer favored less lipophilic contaminants and was associated with relatively high blood plasma PCB and polybrominated diphenyl ether concentrations in females. Despite lactational transfer, females did not show significantly lower blubber contaminant concentrations or burdens than males. This might be caused by their low blubber, and thus contaminant, loss during lactation compared to other species.  相似文献

Blubber, liver and kidney samples of harbor seals (Phoca vitulina) from the northern Gulf of Alaska were collected during 2000-2001 for the analysis of polychlorinated naphthalenes (PCNs) and coplanar polychlorinated biphenyls (CoplPCBs). On the lipid weight (lw) base, the total concentrations of PCNs (Sigma PCNs) ranged from 0.3 to 27 ng/g lw, and the total concentrations of CoplPCBs (Sigma CoplPCBs) were 3.6-546 ng/g lw in all the tissue samples. Di-ortho PCBs and mono-ortho PCBs were dominant followed by non-ortho PCBs and PCNs. Sigma Mono-ortho PCBs and Sigma di-ortho PCBs in nursing seals were apparently lower than those in male adult seals, but Sigma PCNs and Sigma non-ortho PCBs in female adults were not significantly different from those in male adults. Differences in PCNs and CoplPCBs congener profiles in female and male adult seals are apparently related to their chemical structure and properties, animal's developmental stages and physiological conditions. A large quantity of mono-ortho and di-ortho PCBs might be transferred to newborns from the nursing seals during lactation, while non-ortho PCBs and PCNs were relatively accumulative in the mother seals. Sigma PCNs and Sigma CoplPCBs in the harbor seals correlated with ages, gender, body weight and blubber thickness, but the blubber Sigma PCNs and Sigma CoplPCBs in Kodiak Island and Southern Alaska Peninsula (KIAP) did not significantly differ from those in Prince William Sound (PWS). In addition to the new PCB data, this first report on PCN contamination in Alaskan harbor seal tissues is useful for the wildlife and ecosystem management and human health protection.  相似文献

Baltic grey seals (Halichoerus grypus) alternate between land and ice breeding, depending on ice conditions. We show that the fitness of grey seal females in terms of pup mortality and quality is reduced when breeding on land as compared with ice. The mean preweaning mortality rate on land was 21.1% (range 0% to 31.6%), and correlated with birth density (range 0.5-5.2 pups 100 m(-2)). The mean mortality rate on ice was 1.5%, where the highest density was 0.2 pups 100 m(-2) in particularly dense breeding groups. Mean weights of pups born on ice were significantly greater (48.3 +/- 8.1 kg) at the onset of moult as compared with pups born on land (37.4 +/- 7.8 kg). Because indices of life-time net reproductive rate (pup survival) and pup quality (weaning weight and health) were more auspicious on ice as compared with land, diminishing ice fields will lower the fitness of Baltic grey seal females and substantially increase the risk for quasi-extinction.  相似文献

Fish and seafood are important contributions to a healthy diet, but also contain persistent organic pollutants (POPs) like polybrominated diphenylethers (PBDEs) and polychlorinated biphenyls (PCBs). Discrepancies have been found between intake and accumulated levels of POPs, where fish consumers have had similar levels of POPs to the general population. Similarly fish oil consumption has been found to reduce accumulation of POPs. This study examined the accumulation of BDE47 or PCB153 in mice fed diets with different nutritional composition, using female mice with pre-weanling pups exposed through gestation and lactation. A fish-based diet was compared to a standard casein-based rodent diet. All diets had low background levels of environmental contaminants and were spiked with BDE47 or PCB153 to levels representing a realistic (∼0.004 μmol kgbw−1 d) or a high dietary exposure (∼1.3 μmol kgbw−1 d). Accumulation of BDE47 or PCB153 in offspring tissues after 18 d lactation reflected the maternal exposure levels. However, the pups of dams fed a fish-based diet had consistently lower BDE47 accumulation in liver, fat and stomach than pups from casein-fed dams. Similarly the pups of dams fed a high dose of PCB153 in a fish diet also accumulated less PCB153 than pups of the dams fed a casein diet, although not significant. In conclusion, the fish based diets seemed to reduce transfer of BDE47 and PCB153 from dams to pups. The study highlights that in-depth knowledge about nutritional impact on toxicokinetics is of great interest to vulnerable consumers.  相似文献

A prospective birth cohort of 1022 participants was established in the Faroe Islands over a 21-month period during 1986-1987. We collected questionnaire data on potential persistent organic pollutant (POP) concentration predictors, such as duration of breastfeeding and blubber consumption. To assess the participants' exposure from in utero to 14 years of age to polychlorinated biphenyls (PCBs) and the insecticide p,p'-DDT and its primary degradate p,p'-DDE, we measured 37 PCB congeners and pesticides in 316 umbilical cord samples taken from participants at birth, in 124 serum samples collected from participants at approximately 7 years of age, and in 795 serum samples collected from participants at 14 years of age. Measurements of higher chlorination PCB congeners made on individuals' serum samples collected at 7 years and 14 years were highly correlated (typically r > 0.5, p > 0.01), although their concentrations at 7 years were generally two to three times higher than at 14 years. Similarly, umbilical cord PCB concentrations were correlated with PCB concentrations in both 7- and 14-year serum samples. Sex-specific differences in higher chlorination PCB and p,p'-DDE concentrations were found at 14 years but not at 7 years, although a sex interaction with blubber consumption and nursing duration was observed at both ages. Both duration of breastfeeding and consumption of blubber were significant predictors of serum summationPCB concentrations at 7 and 14 years. Multivariate analyses showed that breastfeeding duration was the primary contributor to serum summationPCB concentrations at 7 years, and blubber consumption was the primary contributor at 14 years. These data suggest that infant exposures from breastfeeding were sufficiently large so that continued exposures to PCBs, p,p'-DDT, and p,p'-DDE through the diet have not fully diluted their contribution to the summationPCB and p,p'-DDE body burden of the children.  相似文献

Residue levels of the chlorinated hydrocarbons polychlorinated biphenyls (PCBs), total DDT, alpha-, beta- and gamma-hexachlorocyclohexane (HCH), hexachlorobenzene (HCB), and oxychlordane in blubber, and the elements mercury, cadmium, copper, selenium, arsenic, and zinc in liver, of 82 harbour seals, Phoca vitulina, were determined. The seals were found dead or dying in Norwegian waters during the disease outbreak caused by a morbilli virus in 1988. Of the chlorinated hydrocarbons, the highest concentrations were found of PCBs, which were 2-4 times higher than the total DDT concentrations. P,p'-DDE was the main contributor to the total DDT, and constituted about 80%. The PCB and total DDT concentrations ranged from 0.4-38 and 0.1-8.8 mg kg(-1), respectively. The mercury concentrations ranged from 0.1-89 mg kg(-1). Significantly higher mean levels of PCBs (13 mg kg(-1) and mercury (16 mg kg(-1)) were found in blubber and liver, respectively, of seals from the Southern coast of Norway, as compared to the corresponding mean levels in seals from the Oslofjord (8.8 and 4.1 mg kg(-1)), and at the Northwestern coast (5.8 and 7.9 mg kg(-1)), respectively. A significant positive correlation was found between the concentrations of selenium and mercury. When the seals were grouped according to sex and age, females of ageclass > 1 and pups of both sexes had significantly lower PCB and total DDT levels than males ageclass > 1. Significantly higher hepatic mercury levels were found in seals ageclass > 1 as compared to pups. Only low levels of the other organochlorines, cadmium and arsenic, were found. Copper and zinc were considered to be present at normal physiological levels. The present organochlorine and heavy metal concentrations gave no support to suggestions that organochlorines and heavy metal pollution may be directly involved in the observed seal deaths.  相似文献

Accumulated maternal dioxins are passed onto the fetus and neonate via the placenta and maternal milk. In Japan in 1968, an accidental human exposure to rice oil contaminated with polychlorinated biphenyls (PCBs) and other dioxin-related compounds, such as polychlorinated dibenzofurans (PCDFs), led to development of Yusho oil disease. We investigated differences in blood dioxin concentrations in mother-children pairs affected by the Yusho incident. From 2002 to 2008, blood samples were collected from 26 pairs of Yusho mothers and their children (19 mothers, 26 children). Specific congeners of seven polychlorinated dibenzo-p-dioxins (PCDDs), ten PCDFs, and four non-ortho PCBs were analyzed. The children had significantly lower TEQ concentrations of PCDDs, PCDFs, and coplanar PCBs compared to their mothers. The mother-child difference in blood concentrations varied with the congeners; the largest for 2,3,4,7,8-pentaCDF and the smallest for 1,2,3,4,6,7,8-heptaCDD. The level for 2,3,4,7,8-pentaCDF, which characterizes Yusho oil disease, was approximately 17-30 times higher in the mothers than in the general population, whereas there were no significant differences between children in the formula-fed group and the general population. In contrast, the mean level for 2,3,4,7,8-pentaCDF in the breast-fed group was approximately 1.5 times, (range 0.5-6.5 times) higher than that in the general population. Over 30 years after the Yusho incident, the mean blood dioxin levels in the offspring were only a fraction of the levels in their mothers. This is more consistent with exposure via breast milk than via transplacental transfer in the Yusho incident.  相似文献

The levels of six perfluorocarboxylates (PFCAs), four perfloroalkylsulfonates (PFASs), and one sulfonamide were measured in paired samples of maternal serum, umbilical cord serum, and breast milk. The maternal and cord sera were strongly correlated with each other for all measured compounds (r > 0.5 and p < 0.01). Nevertheless, there was a significant difference in compound composition profile between the two sera matrices, with a more depletion of the longer chain compounds in cord serum. The transfer efficiency values from maternal to cord serum (TFCS/MS) decreased by 70% with each increasing unit of -CF2 chain within a PFCA group, and for perfluorooctanesulfonate (PFOS), by a half compared to perfluorooctanoate (PFOA). In contrast to the strong correlation in concentrations between the two sera matrices, the pattern of compounds in breast milk differed considerably with those in sera. Accordingly, compound- and matrix-specific transfer must be considered when assessing prenatal and postnatal exposure.  相似文献

Breast milk is a noninvasive specimen to assess maternal and infant exposure to polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs). In this study, 206 breast milk samples were collected from 87 participants during lactation, at <7, 15, 30, or 90 days postpartum in four cities in Korea. The total concentrations of PCBs (ΣPCB) and OCPs (ΣOCP) ranged from <LOQ to 84.0 (median: 12.1) ng g−1 lipid weight and from <LOQ to 559 (median: 144) ng g−1 lipid weight, respectively. The residue levels of these contaminants measured in our study were relatively lower than those reported for European, African and Asian populations. Within a month postpartum typically after day seven the levels of ΣPCB and ΣOCP significantly increased. Some OCP compounds were correlated with maternal age, BMI, parity, and delivery mode. Certain types of dietary habits such as seafood and noodle consumption were significantly associated with ΣPCB and ΣOCP. The estimated daily intakes (EDIs) of ΣPCB and ΣOCP were 45.2–127 ng kg−1 bw day−1 and 625–1259 ng kg−1 bw day−1 during lactation, respectively, which are lower than the threshold values proposed by the US EPA and Health Canada. The exposure of Korean infants to chlordanes via breast milk had a potential health risk which deserves further investigation.  相似文献

Blubber samples from grey seal (Halichoerus grypus) pups were collected in 1998 through to 2000. Twenty four newly weaned pups were sampled in November 1998, 13 of which were re-sampled during their first year of life, between March and December 1999, and an additional 48 and 25 samples (new cohorts) were obtained in 1999 (November-December) and 2000 (May-September) respectively. SimgaPBDE concentrations (sum of Di-HxBDEs) ranged from 45 to 1500 ng/g lipid, with a geometric mean of 290 ng/g lipid. BDE 47 dominated the congener profile, followed by BDEs 100, 99, 153 and 154. SimgaPCB concentrations (sum of 41 congeners) ranged from 100 to 93000 ng/g lipid, with a geometric mean of 4600 ng/g lipid, with CB congeners 153, 138 and 180 making up 84% of the total PCB concentrations. The predominant OC pesticides were p,p'-DDE (geometric mean=860 ng/g lipid, range=160-3800 ng/g lipid) and p,p'-DDT (geometric mean=120 ng/g lipid, range=30-670 ng/g lipid). No significant differences were found between males and females, probably due to the young age of the animals. Significant decreases (P<0.01 for PBDEs and P<0.05 for PCBs/OCs) were observed between newly weaned seals in 1998 and 1999, but not for the majority of juveniles in 1999 and 2000. Significant differences (P<0.01) were also found between the different seasons in 1999 newly weaned and juvenile seals, reflecting changes in blubber mass.  相似文献

We measured the concentrations of polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), dioxin-like polychlorinated biphenyls (PCBs), and non-dioxin-like PCBs in paired samples of blood and breast milk collected from 67 secundiparas in Sapporo City, Japan, and combined this data with those of the 30 secundiparas previously measured. The arithmetic mean total toxic equivalents (TEQ-WHO) concentrations of PCDDs, PCDFs, non-ortho PCBs, and mono-ortho PCBs in blood and breast milk of the 97 secundiparous subjects were 3.0-23 (mean: 13, median: 14) and 2.7-20 (mean: 8.6, median: 8.5) pg TEQ g−1 lipid, respectively. The sums of the concentrations of 56 non-dioxin-like PCB congeners that were measured in the subjects’ blood and breast milk were 16-326 (mean: 107, median: 100) and 12-252 (mean: 73, median: 67) ng g−1 lipid, respectively. The partitioning ratios of individual congeners of PCDDs, PCDFs, dioxin-like PCBs, and non-dioxin-like PCBs from blood to breast milk in secundiparas were almost the same as those of primiparas that have been recently reported, suggesting that the partitioning ratios of these compounds from maternal blood to breast milk in women is little affected by delivery. Furthermore, the partition of PCB congeners with chlorine at the 2-, 3-, 4′-, and 5-positions or the 2-, 4-, 4′-, and 5-positions of the biphenyl ring from the blood to the breast milk tended to occur at a higher level than that of other congeners.
