Sedimentation processes of microcystins (MCs), cyanobacterial toxins, were studied in three reservoirs located in Central Spain in which the cyanobacterial community was dominated by the genus Microcystis. MCs were detected in the sediment traps deployed in all reservoirs. In Santillana reservoir, MCs were identified in sediment traps even though they could not be found in the pelagial samples. In the other reservoirs studied, sedimentation rates for MC-containing particles during the bloom period ranged from 0.43 to 2.53 mg m−2 d−1. Interestingly, this very high sedimentation of toxic biomass is not exclusively related to decaying blooms or autumnal sedimentation due to a drop in water temperature. Instead, it seems that MC-containing colonies may be settling constantly during the bloom period and we were able to estimate that during such a Microcystis dominated bloom, around 4.5% of pelagial MCs may be involved in sedimentation. Further, these settling colonies seem to maintain good cell integrity and MCs seem not to be excreted massively. A certain loss of toxin content along the vertical settling may be attributed to minor losses due to cell lysis or to variations in MC cell quota explained by reduced production or internal consumption. Our results for the first time establish specific settling rates for MC-containing particles in freshwaters and definitely identify sedimentation as a major destination for these toxins. These data may contribute to improve managing strategies concerning risks associated with MCs.  相似文献

It remains unclear why the area of a cyanobacterial bloom increases in a shallow lake after a typhoon passes. In this study, the mechanisms of cyanobacterial bloom expansion were investigated by studying meteorological factors, water quality, algal biomass, and bloom area in Lake Taihu before and after typhoons (2007–2016). Our results showed that typhoon-induced sediment resuspension caused a short-term increase in nutrients, but nutrients returned to pre-typhoon levels after the typhoon passages. The short-term nutrient release during a typhoon did not result in an obvious increase in Microcystis cell density in two bays of Lake Taihu (Gonghu and Meiliang). Under strong winds, Microcystis aggregates were uniformly distributed in the water column downwind and then dispersed into different directions by wind-driven currents. In particular, Microcystis in the surface water were transported to the center of Lake Taihu. After a typhoon, dispersed Microcystis refloated and formed blooms. Thus, the bloom area was enlarged compared with before a typhoon. Several days after a typhoon, the bloom area gradually reduced as a result of a steady breeze on the horizontal accumulation of Microcystis.

Lake Erhai is the second largest lake of Southwest China and an important drinking water source. The lake is currently defined as the preliminary stage of eutrophic states, but facing a serious threat with transfer into intensive eutrophication. The present study examined the dynamics of Microcystis blooms and toxic Microcystis in Lake Erhai during 2010, based on quantitative real-time PCR method using 16S rRNA gene specific for Microcystis and microcystin systhesis gene (mcy), and chemical analysis on microcystin (MC) concentrations. Total Microcystis cell abundance at 16 sampling sites were shown as an average of 1.7?×?107 cells l?1 (1.3?×?102–3.8?×?109 cells l?1). Microcystin LR (MC-LR) and microcystin RR (MC-RR) were the main variants. The strong southwesterly winds, anticlockwise circular flows and geographical characteristics of lake and phytoplankton community succession impacted the distribution patterns of Chl a and MC in the lake. The concentration of Chl a and MC and abundances of total Microsytis and MC-producing Microsystis (MCM) were shown to be positively correlated with pH, DO and TP, negatively correlated with SD, NO3-N, TN/Chl a and TN/TP, and not correlated with NH4-N, TN, dissolved total nitrogen (DTN) and water temperatures. When TN/TP decrease, Microcystis tended to dominate and MC concentrations tended to increase, suggesting that the “TN/TP rule” can be partially applied to explain the correlation between the cyanobacterial blooms and nutrients N and P only within a certain nutrient level. It is speculated that N and P nutrients and the associated genes (e.g., mcy) may jointly drive MC concentration and toxigenicity of Microcystis in Lake Erhai.  相似文献

China’s freshwater subtropical shallow lakes are increasingly eutrophic and susceptible to production of heavy growths or water blooms of cyanobacteria. One example was the heavy water bloom that occurred for the first time in Lake Xuanwu, in 2005, an urban lake located in Nanjing city. The aim of the present study was to determine dynamics of water bloom dominating Microcystis in this lake. The environmental occurrence and concentrations of cyanobacterial toxins (cyanotoxins) were investigated in the Czech Republic. Concentrations of microcystins (MCs), cylindrospermopsin (CYN) or saxitoxins (STXs) were determined immunochemically by ELISA assays in 30 water samples collected from the surface layers of 19 reservoirs during the summer season of 2010. MCs were detected in 18 reservoirs and 83 % of samples, with median and maximal concentration being 1.5 and 18.6 μg/L, respectively. The high frequency of MC occurrence coincided with prevalence of cyanobacterium Microcystis sp., which was detected in 87 % samples, followed by Dolichospermum (Anabaena) sp. observed in 33 % samples. CYN was detected by ELISA only in one sample at a concentration of 1.2 μg/L. STXs presence was indicated for the first time in Czech water reservoirs when the toxins were found at low concentrations (0.03–0.04 μg/L) in two samples (7 %) collected from two different reservoirs, where STXs co-occurred with MCs and eventually also with CYN. In both STX-positive samples, the phytoplankton community was dominated by Microcystis sp., but Dolichospermum sp. and/or Aphanizomenon sp. were also present as putative producers of STX and/or CYN. Cyanotoxins commonly occurred in Czech water reservoirs, and MCs frequently at concentrations possibly associated with human health risks. MCs were the most prevalent and abundant cyanotoxins, but also other cyanotoxins were detected, though sporadically. Further research and regulatory monitoring of cyanotoxins other than MCs is therefore required.  相似文献

Short-term hydrodynamic fluctuations caused by extreme weather events are expected to increase worldwide because of global climate change, and such fluctuations can strongly influence cyanobacterial blooms. In this study, the cyanobacterial bloom disappearance and reappearance in Lake Taihu, China, in response to short-term hydrodynamic fluctuations, was investigated by field sampling, long-term ecological records, high-frequency sensors and MODIS satellite images. The horizontal drift caused by the dominant easterly wind during the phytoplankton growth season was mainly responsible for cyanobacterial biomass accumulation in the western and northern regions of the lake and subsequent bloom formation over relatively long time scales. The cyanobacterial bloom changed slowly under calm or gentle wind conditions. In contrast, the short-term bloom events within a day were mainly caused by entrainment and disentrainment of cyanobacterial colonies by wind-induced hydrodynamics. Observation of a westerly event in Lake Taihu revealed that when the 30 min mean wind speed (flow speed) exceeded the threshold value of 6 m/s (5.7 cm/s), cyanobacteria in colonies were entrained by the wind-induced hydrodynamics. Subsequently, the vertical migration of cyanobacterial colonies was controlled by hydrodynamics, resulting in thorough mixing of algal biomass throughout the water depth and the eventual disappearance of surface blooms. Moreover, the intense mixing can also increase the chance for forming larger and more cyanobacterial colonies, namely, aggregation. Subsequently, when the hydrodynamics became weak, the cyanobacterial colonies continuously float upward without effective buoyancy regulation, and cause cyanobacterial bloom explosive expansion after the westerly. Furthermore, the results of this study indicate that the strong wind happening frequently during April and October can be an important cause of the formation and expansion of cyanobacterial blooms in Lake Taihu.  相似文献

It is well known that several morphospecies of Microcystis, such as Microcystis aeruginosa (Kützing) Lemmermann and Microcystis viridis (A. Brown) Lemmermann can produce hepatotoxic microcystins. However, previous studies gave contradictory conclusions about microcystin production of Microcystis wesenbergii (Komárek) Komárek. In the present study, ten Microcystis morphospecies were identified in waterblooms of seven Chinese waterbodies, and Microcystis wesenbergii was shown as the dominant species in these waters. More than 250 single colonies of M. wesenbergii were chosen, under morphological identification, to examine whether M. wesenbergii produce hepatotoxic microcystin by using multiplex PCR for molecular detection of a region (mcyA) of microcystin synthesis genes, and chemical analyses of microcystin content by ELISA and HPLC for 21 isolated strains of M. wesenbergii from these waters were also performed. Both molecular and chemical methods demonstrated that M. wesenbergii from Chinese waters did not produce microcystin.  相似文献

2013年6—10月进行了阳澄湖和滆湖的水样采集及分析,对水体中胞内和胞外3种微囊藻毒素(MC-LR,MCRR,MC-YR)和TN、TP、Chl-a等富营养化指标在两湖的分布情况及关系进行了研究。结果表明,阳澄湖的微囊藻毒素及富营养化因子浓度在不同点位的差异小,而滆湖从北部到南部呈下降趋势,两湖相比,滆湖的浓度远远高于阳澄湖;富营养化因子影响微囊藻毒素的浓度分布和变化;相关性分析表明MC-LR、MC-RR和MC-YR与CODMn、TN、TP、PO3-4-P、Chl-a分别呈极显著正相关性(P0.01),MC-LR、MC-YR与NH+4-N呈显著负相关(P0.05);逐步回归性分析显示Chl-a是影响3种微囊藻毒素浓度的关键因子,可以通过Chl-a对水体中MCs的浓度进行预测,为微囊藻水华和微囊藻毒素污染的预警提供重要的科学参考。  相似文献

To clarify the possible influence of Microcystis blooms on the exchange of phosphorus (P) between sediment and lake water, an enclosure experiment was conducted in the hypereutrophic subtropical Lake Donghu during July-September 2000. Eight enclosures were used: six received sediment while two were sediment-free. In mid-August, Microcystis blooms developed in all the enclosures. There was a persistent coincidence between the occurrence of Microcystis blooms and the increase of both total P (TP) and soluble reactive P (SRP) concentrations in the water of the enclosures with sediments. In sediment-free enclosures, TP and SRP concentrations remained rather stable throughout the experiment, in spite of the appearance of Microcystis blooms. The results indicate that Microcystis blooms induced massive release of P from the sediment, perhaps mediated by high pH caused by intense algal photosynthesis, and/or depressed concentrations of nitrate nitrogen (NO3-N).  相似文献

Colony is a key to Microcystis becoming a dominant population and forming blooms. To find the mechanism of colony formation, we investigated cell wall structures of colonial and unicellular strains. Results showed that colonial strains had significant surface layer protein (S-layer) on the surface of cells than unicellular strains by transmission electron microscopy. Western blot showed colonial strains had more S-layer than the unicellular strains. When the S-layer gene (GenBank accession number CAO89090.1) of Microcystis aeruginosa PCC7806 was expressed in Synechocystis sp. PCC6803, PCC6803 aggregated into colonial morphology. The results indicated that the S-layer could promote colony formation in Microcystis. Based on the S-layer sequences of PCC6803 and PCC7806, nine S-layer genes in other Microcystis strains were screened from the GenBank. Sequence comparing showed that the S-layers conserved regions were all located in N-terminal. The S-layers contain repeats-in-toxin (RTX) sequences with Ca2+-binding site, and their amino acid composition, hydrophobicity, isoelectric point, etc. were consistent with the characteristics of RTX-type S-layer in bacteria.

Purpose

In the reservoir created in the reclaimed land in Isahaya Bay, Japan, Microcystis aeruginosa, which produces microcystins (MCs), bloomed every year, and the water with high levels of MCs in the reservoir has been often drained to Isahaya Bay to adjust the water level. The principal aims of this study are to clarify the water conditions suitable for blooming of M. aeruginosa in the reservoir, to follow the amount of distribution of MCs inside and outside the reservoir, and to discuss how blooming of M. aeruginosa is controlled in the reservoir and how MCs produced by Microcystis spread or accumulate in the aquatic environment.

Method

We monitored the water quality (temperature, salinity, dissolved inorganic nitrogen (DIN), and dissolved inorganic phosphorus) in the reservoir with seasonal blooming of microalgae including phytoplankton and M. aeruginosa using the concentrations of chlorophyll ?? and MCs, respectively, and collected the surface sediment in the reservoir and the bay to determine the MC content using the ELISA method.

Result

M. aeruginosa bloomed in extremely low DIN conditions of the water in warm seasons (spring and late summer to autumn). The year-mean standing stock of MCs was approximately 34.5?kg in the water and 8.4?kg in the surface sediment in the reservoir. Approximately 64.5?kg of MCs was discharged with the effluent to the bay in a year.

Conclusion

Since a large amount of MCs always suspends in the water in the reservoir and it has been discharged to the bay, suspension-feeding animals are exposed most seriously to the high levels of MCs occurring in these areas. We need to pay attention to the danger of widespread dispersal of MCs and biological concentration of MCs by fish and clam inside and outside the reservoir.  相似文献

Massive blooms of the harmful benthic dinoflagellate Ostreopsis cf. ovata are of growing environmental concern in the Mediterranean, having recently caused adverse effects on benthic invertebrates and also some intoxication episodes to humans.The toxicological potential of produced palytoxin-like compounds was investigated in the present study on a typical marine sentinel species, the mussel Mytilus galloprovincialis. Organisms were sampled during various phases of a O. cf. ovata bloom, in two differently impacted sites. The presence of the algal toxins was indirectly assessed in mussels tissues (mouse test and hemolysis neutralization assay), while biological and toxicological effects were evaluated through the measurement of osmoregulatory and neurotoxic alterations (Na+/K+-ATPase and acetylcholinesterase activities), oxidative stress responses (antioxidant defences and total oxyradical scavenging capacity), lipid peroxidation processes (level of malondialdehyde), peroxisomal proliferation, organelle dysfunctions (lysosomal membrane stability, accumulation of lipofuscin and neutral lipids), immunological impairment (granulocytes percentage).Obtained results demonstrated a significant accumulation of algal toxins in mussels exposed to O. cf. ovata. These organisms exhibited a marked inhibition of the Na+/K+-ATPase activity and alterations of immunological, lysosomal and neurotoxic responses. Markers of oxidative stress showed more limited variations suggesting that toxicity of the O. cf. ovata toxins is not primarily mediated by an over production of reactive oxygen species. This study provided preliminary results on the usefulness of a multi-biomarker approach to assess biological alterations and toxicological events associated to blooms of O. cf. ovata in marine organisms.  相似文献

Ponds play an important role in urban areas. However, cyanobacterial blooms counteract the societal need for a good water quality and pose serious health risks for citizens and pets. To provide insight into the extent and possible causes of cyanobacterial problems in urban ponds, we conducted a survey on cyanobacterial blooms and studied three ponds in detail. Among 3,500 urban ponds in the urbanized Dutch province of North Brabant, 125 showed cyanobacterial blooms in the period 2009–2012. This covered 79 % of all locations registered for cyanobacterial blooms, despite the fact that urban ponds comprise only 11 % of the area of surface water in North Brabant. Dominant bloom-forming genera in urban ponds were Microcystis, Anabaena and Planktothrix. In the three ponds selected for further study, the microcystin concentration of the water peaked at 77 μg l?1 and in scums at 64,000 μg l?1, which is considered highly toxic. Microcystin-RR and microcystin-LR were the most prevalent variants in these waters and in scums. Cyanobacterial chlorophyll-a peaked in August with concentrations up to 962 μg l?1 outside of scums. The ponds were highly eutrophic with mean total phosphorus concentrations between 0.16 and 0.44 mg l?1, and the sediments were rich in potential releasable phosphorus. High fish stocks dominated by carp lead to bioturbation, which also favours blooms. As urban ponds in North Brabant, and likely in other regions, regularly suffer from cyanobacterial blooms and citizens may easily have contact with the water and may ingest cyanobacterial material during recreational activities, particularly swimming, control of health risk is of importance. Monitoring of cyanobacteria and cyanobacterial toxins in urban ponds is a first step to control health risks. Mitigation strategies should focus on external sources of eutrophication and consider the effect of sediment P release and bioturbation by fish.  相似文献

The degradation of cyanobacterial blooms often causes hypoxia and elevated concentrations of ammonia, which can aggravate the adverse effects of blooms on aquatic organisms. However, it is not clear how one stressor would work in the presence of other coexistent stressors. We studied the toxic effects of elevated ammonia under hypoxia using a common yet important cladoceran species Daphnia similis isolated from heavily eutrophicated Lake Taihu. A 3?×?2 factorial experimental design was conducted with animals exposed to three un-ionized ammonia levels under two dissolved oxygen levels. Experiments lasted for 14 days and we recorded the life-history traits such as survival, molt, maturation, and fecundity. Results showed that hypoxia significantly decreased survival time and the number of molts of D. similis, whereas ammonia had no effect on them. Elevated ammonia significantly delayed development to maturity in tested animals and decreased their body sizes at maturity. Both ammonia and hypoxia were significantly detrimental to the number of broods, the number of offspring per female, and the number of total offspring per female, and significantly synergistic interactions were detected. Our data clearly demonstrate that elevated ammonia and hypoxia derived from cyanobacterial blooms synergistically affect the cladoceran D. similis.  相似文献

Degradation of three sulfonamides (SAs), namely sulfamethoxazole (SMX), sulfamethazine (SMZ), and sulfadimethoxine (SDM) in surface water and sediments collected from Taihu Lake and Dianchi Lake, China was investigated in this study. The surface water (5–10 cm) was collected from the east region of Taihu Lake, China. Two sets of degradation experiments were conducted in 3-L glass bottles containing 2 L of fresh lake water and 100 μg/L of individual SAs aerated by bubbling air at a rate of approximately 1.2 L/min, one of which was sterilized by the addition of NaN3 (0.1 %). Sediment samples were taken from Taihu Lake and Dianchi Lake, China. For the sediment experiment, 5 g of sediment were weighed into a 50-mL glass tube, with 10 mg/kg of individual SAs. Different experimental conditions including the sediment types, sterilization, light exposure, and redox condition were also considered in the experiments. The three SAs degraded in lake water with half-lives (t 1/2) of 10.5–12.9 days, and the half-lives increased significantly to 31.9–49.8 days in the sterilized water. SMZ and SDM were degraded by abiotic processes in Taihu and Dianchi sediments, and the different experimental conditions and sediments characteristics had no significant effect on their declines. SMX, however, was mainly transformed by facultative anaerobes in Taihu and Dianchi sediments under anaerobic conditions, and the degradation rate of SMX in non-sterile sediment (t 1/2 of 9.6–16.7 days) were higher than in sterilized sediment (t 1/2 of 18.7–135.9 days). Under abiotic conditions, degradation of SMX in Dianchi sediment was faster than in Taihu sediment, probably due to the higher organic matter content and inorganic photosensitizers concentrations in Dianchi sediment. High initial SAs concentration inhibited the SAs degradation, which was likely related to the inhibition of microorganism activities by high SAs levels in sediments. Results from this study could provide information on the persistence of commonly used sulfanomides antibiotics in lake environment.  相似文献

The buoyancy of Microcystis colonies determines the occurrence and dominance of bloom on the water surface. Besides the cell density regulation and the formation of larger size aggregates, increases in cell volume per colony (Vcell) and the colony’s compactness (i.e., volume ratio of cells to the colony, VR) may promote Microcystis colony buoyancy. Yet only a few studies have studied the relationship between the internal structure variation of colonies and their buoyancy, and the co-regulatory role of Vcell and VR of Microcystis colonies in the floating velocity (FV) remains largely unexplored. In the present study, we optimized a method for measuring the compactness of Microcystis colonies based on the linear relationship between total Vcell and chlorophyll a. Different relationships between the VRs and FVs were observed with different colony size and Vcell range groups. Both field and laboratory experiments showed that FV/(D50, median diameter)2 had a significant linear relationship with VR, indicating that the cell density and extracellular polysaccharides were unchanged over a short time period and could be estimated via the slope and intercept of a fitted line. We also constructed a functional relationship between FV, VR, and Vcell and found that high VR and Vcell can promote Microcystis buoyancy. This means that increasing cell compactness or Vcell may be an active regulation strategy for Microcystis colonies to promote buoyancy. Therefore, quantifying the internal structure of Microcystis colonies is strongly recommended for the assessment of Microcystis bloom development and their management.

Microcystins (MCs) are toxic cyclic heptapeptides produced by various cyanobacteria genera, especially Microcystis. We identified 10 out of 12 MCs produced by three Microcystis aeruginosa strains from cyanobacteria collections, UTEX 2666, UTEX 2670 and UAM 1303, by using two analytical methods: Matrix-assisted Laser Desorption Ionization Time of Flight Mass Spectrometry (MALDI-TOF/MS) and HPLC Photodiode Array Detector coupled to a hybrid Quadrupole Time of Flight Mass Spectrometry (HPLC-PDA-QTOF/MS). MALDI-TOF/MS failed to detect non-polar MCs, such as MC-LY and MC-LW. HPLC-QTOF/MS permitted the accurate identification of most MCs present in methanolic extracts. Besides, three new MCs, namely: [D-Glu(OCH3)6, D-Asp3] MC-LAba, MC-YL and MC-YM were detected by HPLC-QTOF/MS.  相似文献

Microcystis aeruginosa is a species of freshwater cyanobacteria which can form harmful algal blooms in freshwater water bodies worldwide. However, in spite its sporadic occurrences for short periods of time in estuarine waters, their influence on zooplankton populations present in these ecosystems has not been extensively studied. In this work, Artemia franciscana was used as test organism model, studying mortality against several strains of M. aeruginosa with different degrees of toxigenicity, measuring whole-live cells and homogenate extracts. Results were compared with microcystin-LR equivalent content, measured by immunoassay. The results show that there were no significant differences between both exposure models (whole cells and extracts), and there are significant differences respect to the toxigenicity of cyanobacterial blooms depending of the M. aerugionosa strain involved in the process. Analysis of microcystin-LR equivalent concentration test immediately below the lowest significant concentration in all M. aerugionosa strains was used to determine the potential risk associated with the cell densities during a bloom. Comparison among the selected M. aerugionsa strains show that these factors have influence in the results obtained, and thus, several differences have been evidenced depending of the microcystin-LR equivalent production and the strain type involved.  相似文献

This work supports previous studies in the Great Barrier Reef lagoon that show the new nitrogen (N) load introduced by Trichodesmium is similar to or greater than that from riverine discharges. However, the current management programs aimed at improving the chronic eutrophic state of the GBR ignore the N load from Trichodesmium. These programs also ignore the evidence that Trichodesmium blooms could promote the bioavailability of heavy metals and be a source of toxins in the ciguatera food chain. Further work is urgently required to better quantify the potential impacts of Trichodesmium and develop management plans to reduce those impacts. A simple algorithm that uses MODIS imagery is developed for not only monitoring the spatial extent of Trichodesmium blooms but also for quantifying the concentration of those blooms. The algorithm is based on the readily available MODIS L2 data. A management plan that includes the harvesting of Trichodesmium is outlined.  相似文献

Seasonal variations in the level of total microcystins in water samples collected from Donghu Lake and a fish pond in Wuhan, China, were studied between March 1995 and February 1996 using a protein phosphatase inhibition assay involving a radioactive 32P-labelled substrate. The assay is highly reliable and repeatable, and is probably the most sensitive assay for microcystin detection to date. Results of the survey indicated the presence of microcystins in the water samples, and the concentration of microcystins appeared to be related to the degree of eutrophication and water temperature. There is also a correlative relationship between the quantity of microcystins and the abundance of microcystin-producing cyanobacteria (Anabaena and Oscillatoria) in the water bodies over a year cycle. In the present study, the positive detection of microcystins in water bodies having no signs of algal bloom warns of considerable potential threat of these waters to public health.  相似文献
