Growth characteristics and nutrient uptake kinetics were determined for zooxanthellae (Gymnodinium microadriaticum) in laboratory culture. The maximum specific growth rate (max) was 0.35 d-1 at 27 °C, 12 hL:12 hD cycle, 45 E m-2 s-1. Anmmonium and nitrate uptake by G. microadriaticum in distinct growth phases exhibited Michaelis-Menten kinetics. Ammonium half-saturation constants (Ks) ranged from 0.4 to 2.0 M; those for nitrate ranged from 0.5 to 0.8 M. Ammonium maximum specific uptake rates (Vmax) (0.75 to 1.74 d-1) exceeded those for nitrate (0.14 to 0.39 d-1) and were much greater than the maximum specific growth rate (0.35 d-1), suggesting that ammonium is the more significant N source for cultured zooxanthellae. Ammonium and nitrate Vmax values compare with those reported from freshly isolated zooxanthellae. Light enhanced ammonium and nitrate uptake; ammonium inhibited nitrate uptake which was not reported for freshly isolated zooxanthellae, suggesting that physiological differences exist between the two. The uptake of nitrate and ammonium was investigated experimentally during early spring 1989 in the Greenland Sea, with particular attention placed on the roles of irradiance, nitrogen concentrations and nitrateammonium interactions. The phytoplankton assemblage was dominated by the colonial prymnesiophyte Phaeocystis pouchetii. Nitrate concentrations ranged from undetectable at the end of the cruise to greater than 10 M, and ammonium levels ranged from less than 0.1 to 1.9M. The uptake of both nitrate and ammonium as a function of irradiance was found to be a saturation response. Photoinhibition occurred and was found to be greater for ammonium uptake. Ammonium uptake also saturated at irradiance levels five times lower than those needed to saturate nitrate uptake. Nitrate and ammonium uptake as a function of nitrogen concentration also was characterized by a saturation response, with the estimated half-saturation constant (K s) value for nitrate uptake being 0.29 M. Elevated ammonium concentrations inhibited nitrate uptake, and the response appeared to be one of exponential decrease with increasing concentrations of ammonium. The most important factor in the Greenland Sea influencing ammonium uptake during the spring was irradiace, while both irradiance and ammonium concentrations played major roles in regulating nitrate uptake and new production.  相似文献

Michaelis-Menten uptake kinetics were observed at all light intensities. With constant illumination, the Vmax and K1 in nitrate uptake over the natural light intensity range of 0 to 2000 E were 0.343 g-at NO3–N(g)-1 at protein-N h-1 and 26 E, respectively. Nitrate uptake was inhibited at higher light intensities. The Ks for nitrate uptake did not vary as a function of light intensity remaining relatively constant at 0.62 g-at NO3–N 1-1. With intermittent illumination, the Vmzx for light intensity in nitrate uptake over a light intensity range of 0 to 5000 E was 0.341 g-at NO3–N(g)-1-at protein-N h-1. No inhibition of nitrate uptake was observed at higher than natural light intensities. Chaetoceros curvisetus will probably never experience light inhibition of nitrate uptake under natural conditions.  相似文献

Fucus distichus L. was collected near Vancouver, Canada, in late fall and early winter, 1981. The effects of the forms of nitrogen (nitrate, ammonium or urea) and periodic exposure to air on growth, rhizoid development and nitrogen uptake in germlings was investigated. Gamete release, fertilization, germination and germling growth had no requirement for a specific form of nitrogen. Periodic exposure to air increased secondary rhizoid development twofold. Nitrate and ammonium uptake rates of the germlings were higher than for the mature thalli (20 to 40 times for nitrate and 8 times for ammonium), while the halfsaturation constant (K s) values for nitrate were similar (1 to 5 M). The germlings showed saturable uptake kinetics but the mature thalli did not. When germlings were exposed to air it caused a 70% decrease in nitrate uptake, but not change in ammonium uptake. Ammonium uptake in the mature thalli was proportional to the ambient ammonium concentration. Nitrate uptake in the mature thalli appeared to follow saturation kinetics at low nitrate concentrations, but showed a non-saturable component at concentrations greater than 10 M. Presence of ammonium inhibited nitrate uptake by the mature plants but not by the germlings.  相似文献

The substrate analogue [14C]-methylammonium was used to study ammonium/methylammonium uptake by Symbiodinium microadriaticum (zooxanthellae). The value of the Michaelis constant (K m) for the uptake system was approximately 35 M with methylammonium as substrate; ammonium was a competitive inhibitor of methylammonium uptake, and the K m for ammonium uptake (determined as the inhibition constant, K i, for methylammonium) was 6.6 M. Methylammonium uptake by zooxanthellae was light-dependent. Methylammonium uptake rates of zooxanthellae which had been freshly isolated from the hermatypic coral Acropora formosa (0.85±0.05x10-10 mol min-1 cell-1) were lower than those of axenic cultures of the zooxanthellae from Montipora verrucosa (Acroporidae) grown under various nitrogen regimes (1.6 to 12x10-10 mol min-1 cell-1). Maximum uptake rates were found for ammonium-starved cultured M. verrucosa zooxanthellae (10.2 to 12x10-10 mol min-1 cell-1); M. verrucosa zooxanthellae growing with ammonium as nitrogen source and zooxanthellae which had been freshly isolated from A. formosa gave similar and considerably lower uptake rates (0.85 to 1.6x10-1 mol min-1 cell-1). These results suggest that either coral tissue contains sufficient ammonium to repress synthesis of the uptake system of the algal symbionts or, alternatively, there are additional barriers to ammonium transport for zooxanthellae in vivo.  相似文献

The locus of symbiotic dinoflagellates within host cells provides a habitat which could potentially be exploited by the alga through heterotrophic uptake of host-derived organic substrates. Using zooxanthellae (Symbiodinium sp.) isolated from the tropical sea anemone Aiptasia pulchella collected from Kaneohe Bay, Hawaii, the effect of various potential organic substrates on growth in vitro was assessed in Erdschreiber seawater medium (ES) supplemented with organic compounds. Zooxanthellae maintained at 5 to 7 E m-2 s-1 (below compensation irradiance) grew heterotrophically when supplied with 100 M glycerol, glycolate, acetate, malate, or propionate, and grew in darkness on 100 M propionate. Zooxanthellae exposed to irradiance below compensation were able to utilize carbon sources in the unsupplemented ES medium for slow growth, but generally the growth rate of cultured zooxanthellae was a function of incubation irradiance. Zooxanthellae incubated for 10 wk in unsupplemented ES at 5 to 7 E m-2 s-1 were capable of growth at this low irradiance, but were also capable of net photosynthetic oxygen production at higher irradiances. This suggests that zooxanthellae can be photoautotrophic or facultatively heterotrophic. An estimate for the duration of mitosis (t d ) is made on the basis of growth rate of cultured zooxanthellae in log-phase; this estimate of t d =4.88 h is less than half the estimated t d for zooxanthellae in situ.  相似文献

Methane mussels (Bathymodiolus sp., undescribed; personal communication by R. Turner to CRF) were collected in September 1989 and April 1990 from offshore Louisiana in the Gulf of Mexico. These mussels contain endosymbiotic methane-oxidizing bacteria and are capable of utilizing environmental methane as a source of energy and carbon. Oxygen consumption, methane consumption, and carbon dioxide production were measured in mussels with intact symbionts, functionally aposymbiotic mussels, and separated symbiont preparations under controlled oxygen and methane conditions, in order to study the roles of the symbionts and the hosts in methane utilization. The association was found to be very efficient in fixing methane carbon (only 30% of CH4 consumed is released as CO2), and to be capable of maximal rates of net carbon uptake of nearly 5 mol g-1 h-1. Rates of oxygen and methane consumption were dependent upon oxygen and methane concentrations. Maximal consumption rates were measured at 250 to 300 M O2 and 200 to 300 M CH4, under which conditions, oxygen consumption by the gill tissues (containing symbionts) had increased more than 50-fold over rates measured in the absence of methane. A model is proposed for the functioning of the intact association in situ, which shows the symbiosis to be capable of achieving growth rates (net carbon assimilation) in the range of 0.003 to 0.50% per day depending upon oxygen and methane concentrations. Under the conditions measured in the seep environment (200 M O2, 60 M CH4), a mussel consuming methane at rates found to be typical (4 to 5 mol g-1 h-1) should have a net carbon assimilation rate of about 0.1% per day. We suggest that the effectiveness of this symbiosis arises through integration of the morphological and physiological characteristics inherent to each of the symbiotic partners, rather than from extensive specialization exhibited by other deep-sea chemotrophic associations.  相似文献

In this continuing study on photosynthesis of the marine red alga Gracilaria conferta, it was found that ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) in crude extracts had a K m (CO2) of 85 M. Since seawater contains only ca. 10 M CO2, it appears that this alga must possess a CO2 concetrating system in order to supply sufficient CO2 to the vicinity of the enzyme. Because this species is a C3 plant (and thus lacks the C4 system for concentrating CO2), but can utilize HCO3 - as an exogenous carbon source, we examined whether HCO3 - uptake could be the initial step of such a CO2 concetrating system. The surface pH of G. conferta thalli was 9.4 during photosynthesis. At this pH, estimated maximal uncatalyzed HCO3 - dehydration (CO2 formation) within the unstirred layer was too slow to account for measured phostosynthetic rates, even in the presence of an external carbonic anhydrase inhibitor. This observation, and the marked pH increase in the unstirred layer following the onset of light, suggests that a HCO3 - transport system (probably coupled to transmembrane H+/OH- fluxes) operates at the plasmalemma level. The involvement of surface-bound carbonic anhydrase in such a system remains, however, obscure. The apparent need of marine macroalgae such as G. conferta for CO2 concentrating mechanisms is discussed with regard to their low affinity of Rubisco to CO2 and the low rate of CO2 supply in water. The close similarity between rates of Rubisco carboxylation and measured photosynthesis further suggests that the carboxylase activity, rather than inorganic carbon transport and intercoversion events, could be an internal limiting factor for photosynthetic rates of G. conferta.  相似文献

Photosynthetic performance in the kelp Laminaria solidungula J. Agardh was examined from photosynthesis irradiance (P-I) parameters calculated from in situ 14C uptake experiments, using whole plants in the Stefansson Sound Boulder Patch, Alaskan Beaufort Sea, in August 1986. Rates of carbon fixation were determined from meristematic, basal blade, and second blade tissue in young and adult sporophytes. Differences in saturating irradiance (I k, measured as photosynthetically active radiation, PAR), photosynthetic capacity (P max), and relative quantum efficiency () were observed both between young and adult plants and between different tissue types. I k was lowest in meristematic tissue (20 to 30 E m–2 s–1) for both young and adult plants, but consistently 8 to 10 E m–2 s–1 higher in young plants compared to adults in all three tissues. Average I k for non-meristematic tissue in adult plants was 38 E m–2 s–1. Under saturating irradiances, young and adult plants exhibited similar rates of carbon fixation on an area basis, but under light limitation, fixation rates were highest in adult plants for all tissues. P max was generally highest in the basal blade and lowest in meristematic tissue. Photosynthetic efficiency () ranged between 0.016 and 0.027 mol C cm–2 h–1/E m–2 s–1, and was highest in meristematic tissue. The relatively lower I k and higher exhibited by L. solidungula in comparison to other kelp species are distinct adaptations to the near absence of light during the eight-month ice-covered period and in summer when water turbidity is high. Continuous measurement of in situ quantum irradiance made in summer showed that maximum PAR can be less than 12 E m–2 s–1 for several days when high wind velocities increase water turbulence and decrease water transparency.The Univeristy of Texas Marine Science Institute Contribution No. 695  相似文献

Inorganic nitrogen metabolism in blue light was studied for the green algaUlva rigida C. Agardh collected in the south of Spain (Punta Carnero, Algeciras) in the winter of 1987. NH4 + has been reported to inhibit NO3 - uptake; however,U. rigida showed a net NO3 - uptake even when the NH4 + concentration of the external medium was three or four times greater than the concentration of NO3 -. NO3 - uptake rates were similar in both darkness and in blue light of various photon fluence rates (PFR) ranging from 17 to 160 mol m-2 s-1. Since NO3 - uptake is an active mechanism involving the consumption of ATP, respiratory metabolism can provide enough ATP to maintain the energetic requirement of NO3 - transport even in darkness. In contrast, NO3 - reduction inU. rigida was highly dependent on the net photosynthetic rate. After 7 h in blue light, intracellular NO3 - concentrations ([NO3 -] i ) were higher in specimens exposed to intensities below the light compensation point (LCP) than in those incubated at a PFR above the LCP. When PFR is below the light compensation point, NO3 - reduction is low, probably because all the NADH produced by the cells is oxidized in the respiratory chain in order to produce ATP to maintain a steady NO3 - transport rate. The total nitrogen (TN) and carbon (TC) contents decreased from darkness to 33 mol m-2 s-1 in blue light. In this range, catabolic processes prevailed over anabolic ones. In contrast, increases in TN and TC contents were observed above the light compensation point. The C : N ratio increased with light intensity, reaching a stable value of 17 at 78 mol m-2 s-1 in blue light. Intracellular NO3 - concentration and NO3 - reduction appear to be directly controlled by light intensity. This external control of [NO3 -]i and the small capacity ofU. rigida to retain incorporated NO3 -, NO2 - and NH4 + ions may explain its nitrophilic character.  相似文献

The bioavailability of iron in seawater filtered through a 0.025-m filter was investigated using 59Fe-labeled iron uptake by the macroalga Laminaria japonica (Areschoug: Phaeophyta) (collected in the northern Japan Sea 1993) as an assay. About 80% of the iron in the 0.025-m filtered coastal seawater was soluble and/or small colloidal organically bound iron, associated with natural organic ligands forming complexes with ferric ion. After decomposition of the organic matter by ultraviolet (UV) irradiation, 55% of the iron addition [or 0.6 nM, nearly the concentration of Fe(OH) 2 + in equilibrium with amorphous hydrous ferric oxide in seawater at pH 8.0] in the 0.025-m filtered coastal seawater was taken up by the macroalga. Since the iron concentrations in the 0.025-m filtered coastal seawater are 0.1 to 2.0 nM and only 0.6 nM of the iron is likely available to biota over 1 to 2 d, we suggest that only small amounts of bioavailable iron exist in coastal seawater not affected by inflow from land and that a significant fraction of dissolved (<0.025 ) iron occurs in forms, such as organic iron complexes, other than the simple hydroxo-complex species predicted by thermodynamic models.  相似文献

Rainwater concentrations of either ammonium or nitrate were sufficient to stimulate chlorophyll a (chl a) production in bioassay experiments using Gulf Stream surface water collected off North Carolina during the summer of 1991. Previous studies primarily examined inshore waters and did not address the impact of rainwater ammonium. An increase in chl a occurred within 1 d of the addition of synthetic rainwater (2 or 5% rainwater, 98 or 95% seawater) containing up to 10 M ammonium; this increase was followed by a decrease in chl a the following day. A similar response to nitrate addition (5% addition of 20 M nitrate rain) was observed. In separate experiments, natural rainwater having nitrate and ammonium concentrations less than those in the experimental synthetic rain yielded a greater chl a response than synthetic rain when added at similar dilutions (0.5 to 5.0% rain). The maximum dissolved inorganic nitrogen concentration in the enriched seawater in these bioassays was 1.8 M; prior to enrichment the maximum was < 0.4 M. Bioassay experiments begun 2 d after a major storm event (sustained NE winds with gusts to 13 m s-1 and ca. 390 mol m-2 inorganic nitrogen deposition from rain) showed a chl a increase in response to addition of natural rainwater, but not to synthetic rainwater with similar dissolved inorganic nitrogen concentration. These results suggest that phytoplankton stimulants, in addition to nitrate and ammonium, exist in natural rain but not in the synthetic rain used in these experiments.  相似文献

Skeletonema costatum was grown at different steady-state growth rates in ammonium or silicate-limited chemostats. The culture was perturbed from its steady-state condition by a single addition of the limiting nutrients ammonium or silicate. The transient response was followed by measuring nutrient disappearance of the liliting perturbation experiment indicate that three distinct modes of uptake of the limiting nutrient can be distinguished; surge uptake (V s ), internally controlled uptake (V i ), and externally controlled uptake (V e ). An interpretation of these three modes of uptake is given and their relation to control of uptake of the limiting nutrient is discussed. The uptake rates of the non-limiting nutrients were shown to be depressed during the surge of the uptake of the limiting nutrient. Kinetic uptake parameters, K s and V max, were obtained from data acquired during the externally controlled uptake segment, V e . The same V max value of 0. 12 h-1, was obtained under either silicate or ammonium limitation. Estimates of K s were 0.4 g-at NH4-N l-1 and 0.7 g-at Si l-1. Short-term 15N uptake-rate measurements conducted on nitrogen-limited cultures appear to be a combination of V s or V i , or at lower substrate concentrations V s and V e . The effects of total dissolved inorganic carbon (DIC), free carbon dioxide [CO2(aq)], and bicarbonate (HCO 3 - ) concentrations on net photosynthetic oxygen evolution of the marine angiosperm Thalassia testudinum Banks ex König collected from Biscayne Bay (1988) and from Tampa Bay (1990), Florida, USA, were examined. Rates of photosynthesis declined by 85% from pH 7.25 to 8.75 in buffered seawater media with constant DIC concentration (2.20 mM), suggesting a strong influence of CO2(aq) concentration. A plateau in the pH-response curve between pH 7.75 and 8.50 indicated possible utilization of HCO 3 - . Responses of photosynthesis measured in buffered seawater media of varying DIC concentrations (0.75 to 13.17 mM) and pH (7.8 to 8.61) demonstrated that photosynthesis is rate-limited at ambient DIC levels. Photosynthesis increased in media with increasing HCO 3 - concentrations but near-constant CO2(aq) levels, confirming HCO 3 - assimilation. Calculated half-saturation constants (K s )for CO2(aq) and HCO 3 - indicated a high affinity for the former [K s (CO2)=3 to 18 M] and a much lower affinity for the latter [K s (HCO 3 - )=1.22 to 8.88 mM]. Calculated V max values for HCO 3 - were generally higher than those for CO2(aq), suggesting relatively efficient HCO 3 - utilization, despite the apparent low affinity for this carbon form.  相似文献

The dinoflagellate symbionts (zooxanthellae) present in many reef corals aid in the survival of the symbiotic unit in nitrogen deficient tropical waters by providing additional routes of nitrogen uptake and metabolism. The enzymatic pathway of ammonia assimilation from seawater and the re-assimilation of coral ammonium waste by zooxanthellae was studied by examining the affinity of glutamine synthetase for one of its substrates, ammonia. Glutamine synthetase activity was measured in dinoflagellates of the species Symbiodinium microadriaticum found in symbiotic association with various marine coelenterates. Michaelis-Menten kinetics for the substrate ammonia were determined for freshly isolated dinoflagellates from Condylactis gigantea (apparent NH3 Km=33 M) and for cultured dinoflagellates from Zoanthus sociatus (apparent NH3 Km=60 M). On the basis of the low apparent Kms for NH3, it appears that ammonia assimilation by these symbiotic dinoflagellates occurs via the glutamine synthetase/glutamate synthase pathway. Additionally, the uptake of exogenous ammonium by an intact coelenterate-dinoflagellate symbiosis was strongly inhibited by 0.5 mM methionine sulfoximine, and inhibitor of glutamine synthetase.  相似文献

The kinetic response of ammonium- or silicate-limited and ammonium- or silicatestarved populations of Chaetoceros debilis, Skeletonema costatum, and Thalassiosira gravida was determined by a single addition of the limiting nutrient to a steady-state culture and subsequent monitoring of the nutrient disappearance of the limiting and non-limiting nutrients at frequent time intervals. The kinetic response of nonlimited (nutrient) populations of these three species was also determined. Three distinct modes of the uptake of the limiting nutrient were observed for ammonium-or silicate-limited populations of these three species, surge uptake (V s ), internally (cellular) controlled uptake (V i ), and externally (ambient limiting nutrient concentration) controlled uptake (V e ). Non-limited populations did not exhibit the three distinct segments of uptake, V s , V i and V e . Estimates of the maximal uptake rate (V max) and the Michaelis constant (K s ) were obtained from nutrient-limited populations during the V e segment of the uptake curve. Pooled values of V e for the three ammonium-limited populations yielded V max and K s estimates of 0.16 h-1 and 0.5 g-at NH4–N l-1. Kinetic data derived from the V e segment of the uptake curve for silicate-limited populations yielded different values of V max and K s for each of the three species. In a number of parameters that were measured, T. gravida was clearly different from C. debilis and S. costatum and its recovery from nutrient starvation was the slowest. Recovery of all species from silicate limitation or starvation was slower than from ammonium limitation or starvation. Ammonium-starved populations maintained a maximal uptake rate at a substrate concentration an order of magnitude lower (0.1 g-at NH4–N l-1) than that observed for NH4-limited populations (1.0 g-at NH4–N l-1). Adaptation to the severity of the nutrient limitation occurred as changes in the magnitude of cellular characteristics, such as short-term uptake potential (V s ) and affinity for the substrate (K s ). For valine uptake by the polychaete Nereis virens Sars, the kinetic constants were: V max=355 nmol g-1 fresh weight h-1, K m=20 M. Leucine and some other amino acids acted as partial inhibitors of valine uptake. Valine uptake rate was 78% higher at 21.5 S than at 14 S. The major portion of valine absorbed by the polychaete could be extracted as free valine, with 6.5 to 15.6% being respired, and 3.6 to 9.5% incorporated into proteins. Calculations indicate that 7 to 12% of the metabolism of N. virens may be sustained by uptake of glycine and aspartic acid from natural concentrations. It is suggested that uptake of amino acids by this worm is important in the nitrogen cycling of marine sediments.  相似文献

Light intensity within the range of 40 to 210 E · m-2 · s-1 had relatively little effect on the abundance, length or distribution of Ceramium rubrum unicellular, hyaline hairs. External NH 4 + concentrations less than 0.5 M stimulated hair initiation and growth in apical regions of the thalli. Ammonium concentrations in excess of 20 M inhibited hair formation on all regions of the thalli. Ammonium uptake velocities of plants with hairs were approximately twice those of plants without hairs. Oyster (Ostrea edulis L.) seed was reared on five rations of Isochrysis galbana Parke: 10, 30, 100, 200 and 300 cells l-1, in an open-flow system. Physiological parameters such as clearance, ingestion, absorption and respiration rates were measured, and the scope for growth (SFG) calculated from these parameters was compared with actual growth over 20 d. Actual growth was negligible at 10 cells l-1 (daily maintenance ration=2.5% organic wt) and maximum (growth rate=13.64 d-1) at 200 cells l-1 (daily ration=43% organic wt), which also supported the maximum gross and net growth efficiencies (K 1=50%, K 2=85%). Ingestion rate was directly correlated to cell concentration up to maximum of 100 cells l-1, but further increases failed to support higher ingestions. Absorption efficiency decreased with increasing algal concentration from 95% down to 60%. Due to the costs of growth, respiration rate significantly increased when food was added above maintenance levels; however, metabolic costs associated with feeding activity were undetectable. The scope for growth calculated from these physiological parameters agreed with the long-term growth performance, validating the physiological energetics method as a valuable tool for predicting long-term growth performance under constant environmental conditions.  相似文献

Six mediterranean macroalgae were cultivated for more than 2 yr under shade culture conditions, after which light requirements for growth were investigated at 16±2°C. The saturation light levels for growth in the logarithmic phase were related to the bathymetric distribution of the algae on the shore. The eulittoral to supralittoral red alga Bangia atropurpurea was saturated at a photon fluence rate of 71 mol photons m-2 s-1, the upper sublittoral to eulittoral brown algae Scytosiphon lomentaria, Colpomenia peregrina and Kuckuckia spinosa and the sublittoral brown alga Stictyosiphon soriferus at 39 to 71 mol photons m-2 s-1, and the deep-water alga Choristocarpus tenellus at 19 mol photons m-2 s-1. The minimum light requirements for growth of B. atropurpurea and C. tenellus were determined by observing length increase for 56 d under limiting light conditions. The compensation and minimum irradiances required for growth of B. atropurpurea were 0.5 and 1 mol photons m-2 s-1 respectively. The corresponding values for C. tenellus were 0.15 to 0.28 and 0.5 mol photons m-2 s-1 respectively. C. tenellus was the siowest-growing species tested at saturating light conditions, but it grew faster than B. atropurpurea at 1 mol photons m-2 s-1. Both B. atropurpurea and C. tenellus were able to survive 56 d in darkness, but only the latter grew under darkness in the first 14 d.  相似文献
