The fatty acid and alcohol composition of the pelagic amphipod, Themisto libellula, was monitored during the 5 first months of its life cycle (4–20 mm length) in an Arctic fjord, Kongsfjorden, Svalbard. Fatty acids of the three major lipid classes, polar lipids (PL), triacylglycerol (TAG), and wax esters (WE), were analyzed to highlight ontogenic changes in their diet and metabolism. The PL composition of T. libellula did not show any strong variations along their growth except during the first month where an important increase of 20:5(n-3) (EPA) and 22:6(n-3) (DHA) was observed. The TAG composition revealed a clear gradient corresponding to a diet shift from omnivorous juveniles toward carnivorous sub-adults and adults. Indeed, fatty acid trophic markers of diatoms were dominant in the juveniles, whereas 20:1(n-9) and 22:1(n-11), the Calanus sp. trophic markers, overwhelmed in the older stages. The WE composition highlighted the same general trend, however, differences were found with the TAG and are discussed as a result of differences in turnover rates and assimilation pathways between the two lipid classes.  相似文献

Changes in the lipid class and fatty acid composition of developing eggs and unfed larvae of cod (Gadus morhua L.) were studied with the objective of determining probable requirements of cod larvae for dietary lipid. The eggs were collected on 24 March 1985 from holding tanks containing cod which had been caught off the northwest coast of Scotland. Phosphatidylcholine (PC) was the only lipid class to decline in absolute terms during embryogenesis. Catabolism of neutral lipid was initiated during the first week after hatching, and the rate of neutral lipid utilisation increased after the larvae had completely absorbed their yolk sacs. The quantity of triacylglycerol (TAG) remained constant during embryogenesis, but the percentage of 22:6(n-3) int TAG increased substantially during this period. It was calculated that ca. 33% of the 22:6(m-3) released during the process of PC catabolism was incorporated into TAG and sterol ester. The results suggest that PC, replete in appropriate essential fatty acids, should represent a major proportion of the lipid in artificial diets for fish and crustacean larvae.  相似文献

The distribution of n-3 highly unsaturated fatty acids (HUFA) over the major neutral and polar lipid classes was determined for two predominant types of live food used in the larviculture of marine fish and shrimp, i.e. freshly hatched and HUFA-enriched Artemia, and compared with data reported in the literature for wild copepods, representing the natural diet of these larvae. Lipid class composition and their content of n-3 HUFA, particularly eicosapentaenoic acid (EPA, 20:5n-3) and docosahexaenoic acid (DHA, 22:6n-3), were assessed in freshly hatched, HUFA-enriched and subsequently starved Artemia franciscana. The n-3 HUFA enrichment was based on feeding Artemia a lipid emulsion in which either fatty acid ethyl esters (EE, diluted with olive oil) or triacylglycerol (TAG) provided a level of 30% n-3 HUFA. Enrichment of Artemia with either type of the lipid emulsions resulted in an increase of total lipid content from 20.0 to 28.2–28.7% of dry matter mainly due to the accumulation of neutral lipid, primarily TAG (from 82 to 158 mg g−1 dry wt in freshly hatched and 24-h enriched Artemia). Enriched brine shrimp utilized up to 27–30% of their TAG content during 72 h of starvation at 12 °C. The absolute tissue concentrations of polar lipids remained constant at 71 to 79 mg g−1 dry wt throughout the enrichment and subsequent starvation. The level of n-3 HUFA increased drastically during enrichment from 6.3% of total fatty acids (8.2 mg g−1 dry wt) in freshly hatched nauplii to between 20.4 and 21.8% (40.4 to 43.2 mg g−1 dry wt) in 24-h enriched Artemia and was not significantly affected by the source of n-3 HUFA. During starvation, 18:0, 20:4n-6 and 20:5n-3 were retained, whereas 18:4n-3, 22:5n-3 and 22:6n-3 were specifically catabolized. The major polar lipids, phosphatidylethanolamine (PE) and phosphatidylcholine (PC), of freshly hatched Artemia showed very low levels of DHA (<0.1% of total fatty acids) and carried about 45% of the total EPA present. Enrichment with either of the emulsions resulted in an increase of the neutral lipid fraction which concentrated >64% of the EPA and >91% of the total DHA present. This is in sharp contrast with the high levels of n-3 HUFA, in particular DHA, in the polar lipid fraction reported for wild copepods. The contrasting distribution of DHA in the neutral and polar lipid fractions of enriched brine shrimp compared to the natural diet may influence the efficacy of this essential fatty acid for marine fish larvae in aquaculture systems. The pelagic yellowtail kingfish Seriola lalandi has become a target species for aquaculture in Asia and Australasia. Australasian production is reliant on larviculture from eggs of captive brood stock; however, knowledge regarding the nutritional requirements of larvae of this species is still scarce, particularly in relation to lipids. As a first step in establishing these requirements, eggs and larvae from captive S. lalandi brood stock were examined for differences in total protein, total lipid and lipid classes between individual spawning events, over the spawning season, and during larval development from fertilisation to 15 days post hatch. Results indicate that total protein egg−1 varied significantly between individual spawning events within a season, but neither total lipid nor total protein egg−1 varied significantly across the spawning season. Brood stock egg lipids were made up of approximately 60% phospholipid, 25% wax and/or sterol esters (WE), 15% triacylglycerol (TAG), and small amounts of sterols and free fatty acids. During the early larval period, both WE and TAG were utilised concurrently for energy. The larvae experienced very high mortality around 5–7 days post hatch, which coincided with very low levels of all neutral lipid classes. Although many other factors may also influence larval mortality, these results indicate that lipid provisioning may be an important factor in larval survival during the critical period around first-feeding in this species. Examination of ratios of TAG:ST, often used as a condition index in fish larvae, suggested that some of the larvae were suffering from starvation. However, as egg-derived WE appears to provide a significant source of energy during the early larval period in S. lalandi, it is suggested that WE should be included in any index of larval nutritional state.  相似文献

The pteropod Clione limacina (Phipps, 1774) is an arcticboreal, circumpolar species, which is widely distributed in the North Atlantic and Subarctic Oceans; it also occurs in the North Pacific Ocean (in the Oyashio and neighbouring waters) and along the Atlantic coast of North America in the waters of the cold Labrador current to the Cape Hatteras region (35° N). The distribution of C. limacina larvae in the plankton of the Norwegian, Barents and White Seas, the Bear Island-Spitsbergen region of the Greenland Sea, the Newfoundland Grand Bank and the Flemish-Cap Bank region of the North-western Atlantic Ocean, and the Kurile-Kamchatka region of the North-western Pacific Ocean has been studied, and information from literature concerning the reproduction and larval occurrence of the species is summarized. Throughout its distributional are, spawning of C. limacina is characterized by the same general ecological pattern. This species breeds and spawns in all types of water masses occurring within the vertical range which it commonly inhabits — from surface layers to 500 m water depth. In all local populations of the species, the most intensive spawning is correlated with the spring/summer period of annual heating of the local waters, and the highest abundance parallels maximum growth of phytoplankton which serves as food for veligers and early polytrochous larvae. After the end of this period, spawning intensity in all local C. limacina populations declines sharply, but spawning continues at low intensity during the autumn/winter season, being practically continuous throughout the year. Distribution patterns of C. limacina larvae are determined by those of their parental forms (the parental forms spawn in the zones permanently inhabited). The earliest larval stages of C. limacina (veligers) are present predominantly in the upper 100 or 200 m water layer, i.e. in the zone of high phytoplankton abundance. Polytrochous larvae, after becoming predaceous feeders, are distributed throughout the whole water column from the surface to 500 m depth, similar to adult C. limacina. As with the adults, larvae are present (within the species' distribution area) in all types of water masses. Since the beginning of the twentieth century, in the course of the warming of the Arctic Ocean, the southern race of C. limacina (formerly a summer/autumn seasonal invader in the Norwegian Sea) has become a permanent component of the plankton fauna of the Norwegian and Barents Seas in regions influenced by the Norwegian-Northcape Current System.  相似文献

The diet of white-chinned petrels Procellaria aequinoctialis breeding at the Crozet Archipelago (southern Indian Ocean) was studied using two complementary methods: lipid analysis of stomach oils as trophic markers together with the conventional dietary approach (i.e., stomach content analysis). Objectives were (1) to investigate the adult diet when they feed for themselves by analyzing stomach oil lipids, and (2) to compare the lipid signature of chick and adult oils. Stomach oils mainly consisted of triacylglycerols (TAG), diacylglycerol-ethers (DAGE) and wax esters (WE) (66, 14 and 11%, respectively). The dietary origin of TAG and WE was evaluated by linear discriminant analyses with fatty acid and fatty alcohol fractions. Analyses evidenced that stomach oils did not originate from Antarctic krill, but instead from myctophid fish, thus demonstrating the importance of mesopelagic fish in the nutrition of adult petrels. This result was consistent with the identification of digested remains of myctophids recovered from adult stomach contents after long foraging trips. Large amounts of a rare lipid class, DAGE (up to 76% of total lipids), were identified in two stomach oils, together with fresh remains of the squid Gonatus antarcticus (99% by mass), suggesting that DAGE could have the potential to be trophic markers of cephalopods. Moreover, six oils probably originated from Patagonian toothfish, thus confirming strong interactions between white-chinned petrels and fisheries. Comparison between chick and adult stomach oils indicated no major differences in their biochemical composition suggesting an identical dietary origin of oils, mainly myctophids. Both adult and chick oils can therefore be used to determine the feeding ecology of adult birds when they feed far away from their breeding grounds. Finally, food analysis of chick samples and adult samples collected after short and long trips indicated different foraging grounds during the two kinds of trips, and also between long trips performed in subtropical and Antarctic waters.  相似文献

Pleuragramma antarcticum is a key component of the neritic assemblages in the Antarctic coastal waters. Larvae of this species were sampled from 2008 to 2011 in the Dumont d’Urville Sea (East Antarctica). The lipid class composition [triacylglycerols (TAG), cholesterol (Chol) and polar lipids (PL)] of larvae was measured to assess the larval condition. The total amount of lipids was linearly related to the quantity of structural polar lipids, suggesting that growth is favored over lipid storage. The TAG:Chol ratio showed interannual variability in the condition of fish larvae, probably related to prey availability. Nevertheless, the essential fatty acids composition of polar lipids illustrates that larvae with low levels of TAG:Chol could be either growing or under starvation. Only the combination of a low TAG:Chol ratio and low polar lipids content, which can also be mobilized during starvation periods, allowed identification of larvae in poor condition. This lipid condition index should be of great assistance to evaluate the probability of survival of P. antarcticum larvae in long-term monitoring. It has widespread applicability and should also be useful in the diagnosis of nutritional condition in other species.  相似文献

Observations on breeding season, copulation, spawning, and development are described for Paedoclione doliiformis, the only gymnosomatous pteropod which consistently retains external larval features throughout its life. The number of eggs spawned per individual is dependent upon temperature and availability of Spiratella retroversa, its thecosome prey. At 17° to 19°C, planktotrophic veligers hatch from floating egg masses within 3 days, and cast their shells approximately 11 days later. The resultant shell-less veligers could complete metamorphosis to the polytrochous larval stage, in which the body is encircled by 3 ciliary bands, within 12 h. Some polytrochous larvae begin feeding on veligers of S. retroversa within 2 days. Unlike other gymnosome species, there is no further change in external appearance except for growth. A comparison of reproduction and development has been made with Clione limacina, another gymnosome which competes for food with P. doliiformis in Nova Scotian (Canada) coastal waters.  相似文献

Carnivorous zooplankton is a key element to the energy transfer through the arctic food web, linking lipid rich herbivores to the top predators. We investigated the growth and lipid dynamic of the Arctic pelagic amphipod Themisto libellula in Kongsfjorden (Svalbard, 79°N) from May to October 2007. Additional samplings were performed in spring and summer 2006 and further north in Rijpfjorden (80°N), in September 2006 and 2007. In Kongsfjorden, the first free-swimming stages (3 mm) appeared early May and reached their adult length (25 mm), in October. During their first year, they grew according to a Von Bertalanffy model and most probably constituted a single cohort. Juveniles had the highest growth rate (0.19 mm day−1) and revealed relatively low total lipid (TL) content (about 2.5% wet weight (WW)) with phospholipids as the major lipid class. Sub-adults showed a distinct decrease of growth rates which coincided with the increase of neutral lipid storage, reflecting a switch in energy allocation, from somatic growth to lipid storage. Indeed wax esters (WE) increased up to 48.5% TL on average in adults in 2006 while triacylglycerols (TAG) remained almost constant below 25.2% TL. The absence of lipid accumulation (in disproportion of the weight) in 2007 could be explained by a higher metabolism of T. libellula or preys of lower quality. In Rijpfjorden, adults in their second year continued accumulating lipid (up to 10% WW) with high and similar proportions of both lipid classes, WE and TAG. We highlighted that T. libellula exhibited a variable lipid metabolism along its life cycle depending on its physiological needs and environmental conditions.  相似文献

During austral summer of 1985 different developmental stages (CIII, CIV, CV, females, males) of the Antarctic copepod Euchaeta antarctica and females of Euchirella rostromagna were collected in the southeastern Weddell Sea to determine their lipid contents and compositions. For E. antarctica the analyses revealed a strong ontogenetic accumulation of lipids towards the older copepodids with highest lipid contents in late CV stages and adults. The females of E. rostromagna had moderate lipid levels. The most striking difference between these two species concerns their lipid class compositions. E. antarctica deposited predominantly wax esters, whereas in E. rostromagna the major lipid class consisted of triacylglycerols, an unusual storage lipid in polar marine copepods. Principal fatty acids in E. antarctica were the monounsaturates 18:1(n-9) and 16:1(n-7), especially in the lipid-rich stages, while the polyunsaturated fatty acids 20:5(n-3) and 22:6(n-3), usually membrane lipids, dominated in the lipid-poor stages. The wax ester moieties in E. antarctica consisted almost entirely of 14:0 and 16:0 fatty alcohols. Major components in E. rostromagna were the fatty acids 18:1(n-9), 16:0, 20:5(n-3) and 22:6(n-3). The potential of fatty acids and alcohols as typical trophic markers is rendered largely insignificant in the two species due to catabolic processes.  相似文献

Many free-living copepods produce and store lipids prior to entering diapause (long-term dormancy). Heteropsyllus nunni Coull is the only marine harpacticoid copepod known to undergo any form of diapause. This study presents the first information on the types of lipids and fatty acids produced for long-term diapause in this benthic species. Sexually immature adults of H. nunni undergo diapause within a pliable self-made cyst. Prior to entering diapause (which lasts 3–4 months), they produce and store large amounts of orange lipid. The lipids apparently are utilized during diapause. Although some residual lipids remain, chiefly around the gonads, after the copepods emerge from their cysts, the lipid stores are visibly reduced. Typically, the copepods mate and produce eggs within 48 h after diapause is terminated. Light level and confocal laser scanning microscopy revealed that the lipid stores are distributed throughout the body in numerous oil droplets and not as a single oil sac, as seen in many marine calanoid copepods prior to overwintering (winter diapause). Transmission electron microscopy showed lipid spheres within the gut epithelium and large droplets of lipids stored extracellularly. Confocal laser scanning microscopy of copepods in pre-diapause, during diapause (encysted), post-diapause (recently excysted), and in reproductive condition, revealed that lipid stores are reduced following diapause, but are not totally absent. Analysis of lipid classes showed that H. nunni store predominantly wax esters/sterol esters (83% of total lipids) during diapause. The predominant lipid is most likely wax esters, as sterol esters typically are found only in small amounts in copepods. Fatty acid (FA) profiles of the copepods in diapause showed 16:0 to be most abundant followed by 16:1n-7 and 18:0; other FA occurred at concentrations <10% of total FA. Three polyunsaturated fatty acids (PUFA), 20:5n-3, 18:2n-6 and 20:4n-6, were found at concentrations <2% of total FA. These PUFA are "essential fatty acids" in H. nunni, obtained through dietary sources. The lipid classes and fatty acids present in H. nunni during diapause are compared to those of other copepods, some in a state of diapause and others not. It appears that lipid class and FA profiles are indicative of genetic makeup, type of diet or amount of food consumed prior to dormancy. The present study investigates amino and fatty acid dynamics of embryos of different-sized simultaneous hermaphrodite shrimp (SH) (Lysmata seticaudata) during early (ERS) and late reproductive seasons (LRS). A significant relative decrease in total amino acids and essential amino acids (EAA) was recorded (P<0.05) during the development of embryos produced by shrimp collected during ERS and LRS. The content of non-essential amino acids (NEAA) showed a smaller variation, without a marked decrease. During the last embryonic stage, the major EAAs of embryos were, in decreasing magnitude, lysine and arginine, while the major NEAAs were glutamic acid and valine. A substantial decrease in lipid content (P<0.05) was observed, and the quantitatively more important fatty acids were the saturates 16:0 and 18:0, the monounsaturates 18:1n-9 and 18:1n-7 and the polyunsaturates 20:4n-6 (arachidonic acid, ARA), 20:5n-3 (eicosapentaenoic acid, EPA) and 22:6n-3 (docosahexaenoic acid, DHA). Monounsaturates were used at a higher rate, and embryos produced by SH shrimp displayed similar consumption rates of saturated and polyunsaturated fatty acids. Considering individual fatty acids, no clear utilization pattern between different-sized SH shrimp in ERS and LRS was recorded. The inexistence of consistent differences between amino and fatty acid utilization during embryogenesis among different-sized SH shrimp in ERS and LRS emphasizes the variability affecting offspring in decapod crustaceans.Communicated by S.A. The lipid/fatty acid composition of marine fish eggs and larvae is linked with buoyancy regulation, but our understanding of such processes is largely restricted to species with pelagic eggs. In this study, we examined developmental changes in the lipid/fatty acids of eggs and embryos of Pacific cod (Gadus macrocephalus), a species that spawns demersal eggs along coastal shelf edges, but as larvae must make a rapid transition to the upper reaches of the water column. Adult Pacific cod were collected in the Gulf of Alaska during the spawning season and eggs of two females were artificially fertilized with sperm from three males for each female. The eggs were subsequently reared in the laboratory to determine (1) how lipids/fatty acids were catabolized during egg and larval development, and (2) whether lipid/fatty acid catabolism had measurable effects on egg/embryo density. Eggs incubated at 4°C began hatching after 3-weeks and continued to hatch over a 10-day period, during which there was a distinct shift in lipid classes (phospholipids (PL), triacyglycerols (TAG), and sterols (ST)) and essential fatty acids (EFAs: 22:6n-3 (DHA), 20:5n-3 (EPA), and 20:4n-6 (AA)). In the egg stage, total lipid content steadily decreased during the first 60% of development, but just prior to hatch we observed an unexpected 2–3-fold lipid increase (~6–9 μg individual−1) and a significant drop in egg density. The increase in lipids was largely driven by PL, with evidence of long-chained fatty acid synthesis. Late-hatching larvae had progressively decreasing lipid and fatty acid reserves, suggesting a shift from lipogenesis to lipid catabolism with continued larval development. Egg density measures suggest that lipid/fatty acid composition is linked to buoyancy regulation as larvae shift from a demersal to a pelagic existence following hatch. The biochemical pathway by which Pacific cod are apparently able to synthesize EFAs is unknown, therefore representing a remarkable finding meriting further investigation.  相似文献

 Various developmental stages (early larvae to adults) of Euphausia superba have been collected in different seasons in the Weddell Sea, the Lazarev Sea and off the Antarctic Peninsula to investigate the role of lipids and fatty acids in the life cycle of the Antarctic krill. The total-lipid data for E. superba exhibited seasonal variations, with low lipid levels in late winter/early spring and the highest levels in autumn. Seasonal changes were most pronounced in the immature and adult specimens, increasing from about 10% lipid of dry mass to more than 40%. The fatty-acid compositions of the younger stages were dominated by 20:5(n-3), 22:6(n-3) and 16:0. These are typical phospholipid fatty acids, which are major biomembrane constituents. The phospholipid composition was similar in the older stages. With increasing storage of triacylglycerols in the lipid-rich immature and adult stages, the fatty acids 14:0, 16:0 and 18:1(n-9) prevailed, comprising about 70% of total triacylglycerol fatty acids. The trophic-marker fatty acids 16:1(n-7) and 18:4(n-3), indicating phytoplankton ingestion, were less abundant. They reflected, however, the dependence of the larvae on phytoplankton as well as the seasonal changes in algal composition. The generally close linear relationships between fatty acids and lipid suggest that the fatty-acid compositions of the collected specimens were largely independent of the respective developmental stage, season and region. The linear fit indicates that triacylglycerol accumulation started at a level of about 5% of total lipid. Considering the various overwintering scenarios under discussion, the life cycle and reproductive strategies of krill are discussed in the context of the lipid metabolism and fatty-acid composition of E. superba. Lipid production is effective enough to accumulate large energy reserves for the dark season, but E. superba does not exhibit the sophisticated biosynthetic pathways known from other Antarctic euphausiids and copepods. Although important, lipid utilisation appears to be just one of several strategies of E. superba to thrive under the extreme Antarctic conditions, and this pronounced versatility may explain the success of this species in the Southern Ocean. Four species of microalgae (Chaetoceros muelleri, Tetraselmis suecica, Tahitian Isochrysis sp. (T-iso) and Dunaliella tertiolecta) with distinctly different fatty acid profiles were grown in continuous culture and fed to prawn larvae (Penaeus japonicus, P. semisulcatus and P. monodon) as monospecific diets. The best two diets (C. muelleri and T. suecica) were also fed as a mixed diet. Experiments were run until the larvae fed the control diet of C. muelleri metamorphosed to Mysis 1. The survival and development (i.e. performance) of the larvae were affected by algal diet, and the diets were ranked in the order of decreasing nutritional value: C. muelleri ≥ T. suecica > T-iso > D. tertiolecta. Larvae fed a mixed diet of C. muelleri and T. suecica (2:3 by dry weight) performed as well or better than those fed C. muelleri, and the performance of both these groups of larvae was better than those fed T. suecica. The lipid and carbohydrate compositions of the algae had little or no effect on the lipid and carbohydrate compositions of the larvae or their performance. However, the larvae that performed best (i.e. those fed C. muelleri) had significantly more lipid and carbohydrate than those that performed worst (i.e. those fed D. tertiolecta). Larvae fed C. muelleri or the mixed-algae diet had higher proportions of the essential fatty acids eicosapentaenoic acid [EPA, 20:5(n-3)] and arachidonic acid [ARA, 20:4(n-6)] than the larvae fed on other diets. Furthermore, the larvae fed T. suecica, which showed intermediate performance between larvae fed C. muelleri and T-iso or D. tertiolecta, also had higher proportions of EPA and ARA. Both C. muelleri and T. suecica contained EPA and ARA, but T-iso and D. tertiolecta did not, except for trace amounts of EPA in T-iso. The fatty acid ARA appears to be much more important in the diet of larval prawns than has so far been considered. The level of the essential fatty acid docosahexaenoic acid [DHA, 22:6(n-3)] in the algal diet and the larvae was not related to the performance of the larvae; only C. muelleri and T-iso contained DHA. However, the nauplii contained large proportions of DHA, suggesting that these were sufficient to meet the larval requirements for DHA during their development to Mysis 1. Antarctic fishes contain large quantities of lipid in adipose tissue, blood and oxidative muscle. In this study, the fatty acyl compositions of free fatty acids (FFA) and triacylglycerol (TAG) in serum, adipose tissue and oxidative muscle were determined in two species of Antarctic fishes, Trematomus newnesi Boulenger, 1902, and Notothenia gibberifrons Lönnberg, 1905, collected off the Antarctic Peninsula in February and March 1987. Total lipid contents of serum and oxidative muscle in each species also were measured. T. newnesi, a mesopelagic species, has significantly more lipid in the serum than N. gibberifrons, a demersal species (17.6±2.4 and 10.4±1.0 mg lipid g-1 serum, respectively). The oxidative muscle of T. newnesi also contains more lipid than does that of N. gibberifrons (12.5 and 9.3% by dry weight, respectively). Fatty acids comprising greater than 5% of one or more of the lipid pools assayed in both species include 14:0, 16:0, 16:1, 16:2, 18:0, 18:1, 20:5 and 22:6. The percentages of 16:1 were lower (p<0.05) in oxidative muscle FFA than in all other assayed lipid pools in both species, suggesting a catabolic preference for some monoenes in oxidative muscle of these fishes. The percentage of 14:0 was two-to three-fold higher in adipose tissue TAG than in all other lipid pools in both species. Differences in fatty acid compositions between lipid pools indicate than proteins involved in the mobilization, transport or catabolism of fatty acids exhibit differential recognition of fatty acids in T. newnesi and N. gibberifrons.  相似文献

Little is known about the changes in composition of brain lipids and fatty acids at different stages of development in fish. Wild Atlantic herring (Clupea harengus L.) were collected from Loch Linnhe and the Firth of Clyde, Scotland, from August 1990 to March 1991. Lipid class and fatty acid compositions of brain lipids were studied at four different stages of development: larvae at the end of the yolk sac stage, two juvenile stages and sexually mature adults. The total lipid content in brains increased during development, and larval brains contained higher proportions of neutral lipids and lower proportions of polar lipids than the brains of juvenile or adult herring. Increased proportions of polar lipids in juvenile and adult herring brains were mainly due to increased percentages of phosphatidylcholine (PC), phosphatidylethanolamine (PE), cerebrosides and sulphatides. The increase in the proportions of the glycolipid classes suggested increasing levels of myelination with development. In total lipids, saturated fatty acids generally decreased and monounsaturated fatty acids and dimethyl acetals (derived from PE-plasmalogen) increased from larvae to adults. However, the proportions of polyunsaturated fatty acids in individual phosphoglycerides were generally highest in juvenile stages, due mainly to increased 22:6n-3, and were lowest in adult fish. Relatively high percentages of 24:1 isomers were found in all the phosphoglycerides, but primarily PC, and these increased during development from larvae to adult. Fatty acids were distributed between individual phosphoglycerides with a characteristic pattern that did not change with development, although the relative amounts of individual fatty acids were altered. The variations and roles of the different lipid components of herring brain are discussed with respect to lipid compositions and functions in brains of other fishes and vertebrates.  相似文献

The impact of supplementing lipid emulsions rich in eicosapentaenoic acid (EmEPA), docosahexaenoic acid (EmDHA) or saturated fatty acids (EmCOCO) to a standard algal diet [3:1 mixture of Isochrysis galbana (T-iso) and Chaetoceros neogracile, St-diet] on Argopecten purpuratus broodstock was evaluated. Broodstock fecundity was compared as well as the egg quality in terms of lipid content, fatty acid composition and lipid class distribution. Fecundity was defined as the number of eggs released in the spawning process, since spawning was virtually complete. Results indicated that the total lipid content of the eggs of A. purpuratus was diet independent. A greater energy reserve was spent on a larger number of oocytes and not on bigger sized oocytes with a higher lipid content. The lipids supplied through the emulsions were at least partially allocated to the eggs, demonstrating that the fatty acid composition of the eggs could be manipulated, especially the neutral lipid fraction. Levels of EPA changed more rapidly than DHA levels, supporting the observation that they fulfilled an energetic and structural role, respectively. The St-diet supplemented with 50%EmCOCO resulted in a significantly higher fecundity compared to the algal diet supplemented with 25%EmEPA+25%EmDHA and the non-supplemented algal diet. It would seem that saturated fatty acids (SAFA) were more easily or preferentially incorporated in the female gonads of A. purpuratus. The relative content of SAFA and 18:2( n-6) in these eggs rose significantly. The relative content of the highly unsaturated fatty acids, EPA and DHA, on the other hand was substantially lower in the neutral lipid fraction, but hardly affected in the polar lipid fraction. It appeared that the maintenance of an adequate DHA/EPA ratio (approximately 1.2) was more important than the absolute levels of the two fatty acids, as long as a threshold value was reached.  相似文献

During early development in fish, phospholipase A2 (EC 3.1.1.4) regulates membrane lipid modifications, which relates to changes in environmental conditions and provision of fatty acids required for metabolic energy substrates and prostaglandin biosynthesis. A method to analyze phospholipase A2 in rat tissues has been modified to measure its activity in embryonic Atlantic halibut (Hippoglossus hippoglossus L.). Egg and embryo samples were collected during the 1994 spawning season. Enzyme activity was undetectable at fertilization but in 10-d embryos was 230 pmol mg−1 h−1 (at 20 °C) and increased by ∼120% at hatch (17-d). Significant alterations in the fatty acid composition of important phospholipids, phosphatidylcholine (PC) and phosphatidylethanolamine (PE), were also observed. The content of some critical polyunsaturated fatty acids, and the ratio of unsaturated/saturated fatty acids, declined significantly over development. Acyl-chain restructuring mediated through the activity of phospholipase A2, coupled with other observed lipid changes (significant increases in the PC/PE ratio and cholesterol content), would produce a decreased fluidity of membranes during embryonic development, coinciding with the predicted upward movement of larvae in the water column. Arachidonic acid (20:4n-6) removed from PE could serve as a precursor for biosynthesis of 2-series prostaglandins, and eicosapentaenoic acid (20:5n-3) from PC is a likely source for other prostaglandin types. Despite removal of polyunsaturated fatty acids, there was an overall increase in lipid and fatty acid concentration, which can be attributed to amino acid catabolism during early developmental stages. Changes in the chemical composition of developing dolphin (Coryphaena hippurus) eggs and prefeeding yolksac larvae were determined in order to estimate probable dietary requirements of first-feeding larvae. Daily dry matter, protein nitrogen (PN), non-protein nitrogen (NPN), lipid, gross energy content, fatty acid and amino acid profiles from Day 1 to Day 2 eggs and Day 1 to Day 3 larvae were compared. Lipid was the primary endogenous energy source accounting for the daily caloric deficit through both the egg and larval stages, except over the day of hatching. The catabolism of lipid by embryos (0.078 cal d–1) was greater than that by yolksac larvae (0.036 cal d–1). The higher demand for energy by embryos was related to a greater rate of protein synthesis during the egg stage. The ratio of PN:NPN increased during egg development without change in total nitrogen content, but was constant throughout the yolksac larvae period. The lipid content per embryo did not decrease over the hatching period (Day 2 to 3, postspawning). However, there was a loss in amino acid content not totally accounted for by sloughing of the chorion at hatching. This loss, as protein, accounted for 0.053 cal of gross energy, which represented 70% of the total estimated energy needs of the fish over this period. Loss of non-essential amino acids (25%) was higher than that of essential amino acids (13%). Proline and tyrosine accounted for 32% of the total loss of amino acids at this time. The only preferential use of fatty acids over any period was a small but significant drop in the content of C22:6n-3 prior to the onset of feeding (Day 5, postspawning). It is speculated that the pattern of energy-substrate use of first-feeding dolphin larvae will reflect the pattern of endogenous energy use during the egg and prefeeding yolksac larval stages. Diets or feeding regimens with lipid as the primary energy source, and containing a fatty acid profile similar to that of eggs or yolksac larvae, should be useful in culturing this species, at least during the early feeding stages.  相似文献
