We studied the bioaccumulation of lead in selected tissues of Prussian carp Carassius gibelio (Bloch, 1782) during 12 and 24 months exposure to different doses of this metal in feed and the elimination of lead from tissues during the following 12-month depuration period. Lead concentration was determined using atomic absorption spectrometry method. The highest lead concentrations were observed at 2.0?±?0.54 to 7.4?±?1.1 mg?kg?1 in the kidney, 3.0?±?0.13 to 5.2?±?0.17 mg?kg?1 in the bone, and 4.5 (±0.4)?mg?kg?1 in the hepatopancreas of fish from groups exposed to lead dietary concentration from 8 to 49 mg?kg?1 for 24 months. The rate of accumulation were generally the highest at the beginning of exposure as evidenced by the highest monthly increments of bioaccumulation observed after 3 months of contamination for muscles, hepatopancreatic gland, intestine, and gills. Also analysis of the monthly increments of lead bioaccumulation in bone tissue and the highly significant coefficients of correlation indicate that the dynamics of accumulation are clearly dependent on dose of exposure. Depuration of accumulated lead from the organs depended mainly on tissue and duration of elimination period. Very rapid depuration was observed in soft tissues such as the intestine or muscles. Very low elimination was observed for scales and bones where until the end of the experiment highly significant lead concentration differences were observed in all groups in relation to the control group. Chronic dietary exposure in the range of 8–49 mg Pb?kg?1 resulted in no significant effects on the growth and survival of Prussian carp females.  相似文献

In Anabastestudineus, the per cent changes noted in the aspartate aminotransferase (AAT) activities of gill, brain, intestine, liver, muscle and kidney until 6 hr of exposure to lethal concentration (10.5 mg/L disyston) were relatively much less when compared to those of sublethal concentration (4.0 mg/L disyston). The authors investigated the arsenic (As) accumulation in different tissues (muscle, gill, liver, stomach, and intestine) and the possible correlation between tissue concentration and hematological parameters in mullet (Mugil cephalus Linnaeus, 1758) caught in Faro Lake (Messina, Sicily, Italy). On all fish, hematological analyses of blood samples, measurement of biometric indices, and the removal of the muscles, gills, liver, stomach, and intestine for the determination of arsenic concentration were performed. A hemogram was performed to find effects of arsenic concentration in tissues on hematological variables. One-way analysis of variance showed significant differences of arsenic concentration in different tissues, with higher values in the gill. The correlation between hematological parameters and tissue arsenic concentration showed a statistical significance for red blood cell (RBC), hemoglobin concentration (Hb), and hematocrit (Hct) with the liver As concentration. Biometric indices (weight, length, and fork length) showed a significant correlation with As concentration of the muscle and liver also. Our results indicate the role of some hematological parameters as biomarkers useful to monitoring anthropogenic load of arsenic in water and sediment, because variations of these parameters represent one of the effects that arsenic exposure can have on fish.

One of the major challenges in assessing the potential metal stress to aquatic organisms is explicitly predicting the internal dose in target organs. We aimed to understand the main sources of copper (Cu) accumulation in target organs of tilapia (Oreochromis mossambicus) and to investigate how the fish alter the process of Cu uptake, depuration, and accumulation (toxicokinetics (TK)) under prolonged conditions. We measured the temporal Cu profiles in selected organs after single and combined exposure to waterborne and dietary Cu for 14 days. Quantitative relations between different sources and levels of Cu, duration of treatment, and organ-specific Cu concentrations were established using TK modeling approaches. We show that water was the main source of Cu in the gills (>94 %), liver (>89 %), and alimentary canal (>86 %); the major source of Cu in the muscle (>51 %) was food. Cu uptake and depuration in tilapia organs were mediated under prolonged exposure conditions. In general, the uptake rate, depuration rate, and net bioaccumulation ability in all selected organs decreased with increasing waterborne Cu levels and duration of exposure. Muscle played a key role in accounting for the rapid Cu accumulation in the first period after exposure. Conversely, the liver acted as a terminal Cu storage site when exposure was extended. The TK processes of Cu in tilapia were highly changed under higher exposure conditions. The commonly used bioaccumulation model might lead to overestimations of the internal metal concentration with the basic assumption of constant TK processes.  相似文献

Abstract

The effects of cadmium and lead on chronic mercury accumulation were investigated in O. aureus. After 140 days’ exposure the accumulation of mercury in the liver, brain, gill filaments, intestine, caudal muscle, spleen, trunk kidney and eye was analysed. The exposure concentrations were 0.05, 0.10 and 0.20 mg/L for mercury alone. O. aureus was also exposed to mixtures of 0.05 mg/L mercury with lead (0.05 mg/L and 0.50 mg/L or cadmium (0.05 mg/L) and 0.10 mg/L mercury with 0.10 mg/L cadmium. In food fish, a knowledge of toxic metal accumulation patterns is of great importance because of their contribution to the human diet and, as fishmeal, to the diet of agricultural animals. The trunk kidney consistently accumulated higher concentrations of mercury than any of the other tissues investigated.  相似文献

Specimens of farmed European sea bass (Dicentrarchus labrax L., 1758) were exposed to different cadmium (Cd) concentrations (4.47, 5.63, 7.08 and 8.91 mg l(-1)) for 24 and 48 h. The effects of Cd on numbers of some cell types and structures (i.e., chloride cells, CCs; macrophage aggregates, MAs; rodlet cells, RCs) and on structure and ultrastructure of the main organs (gill, liver, intestine, kidney) were studied with routine process for light and transmission electron microscopy. Following cadmium exposure, the numbers of branchial CCs as well as intestinal and renal RCs increased significantly within 24h. Increase in metal concentration did not affect the magnitude of the numerical increment of the aforementioned cells. Moreover, in treated fish (24 and 48 h) the numbers of MAs in both head kidney and spleen were significantly higher than in control conspecifics, whilst the global area of MAs was less influenced by the acute treatment. In exposed sea bass, all the examined organs exhibited cellular modifications which appeared time- and dose-dependent. The gills showed telangectasia, lamellar fusion, oedema, epithelial lifting and leukocyte infiltration. In the liver, kidney and intestine acute cell swelling and vacuolization were common. Ultrastructurally the alterations observed frequently in hepatocytes, tubular epithelial cells and enterocytes included presence of numerous myelinoid bodies, damaged mitochondria, dilatation of endoplasmic reticulum, high number of lysosomes and autophagolysosomes. In intestinal and kidney tubular epithelia of treated fish, rodlet cells displayed some anomalies like dilatation of nuclear envelope, cytoplasmic vacuolization, presence of myelinoid bodies, rodlets degeneration and extensive discharge activity.  相似文献

A laboratory study of the cadmium and mercury accumulation and elimination kinetics was conducted on the pelecypod Elliptio complanata (Lightfoot) and the short-lived gastropod Viviparus georgianus (Lea), according to age-classes. Preliminary results (metal concentration vs time of exposure) have demonstrated that uptake of Cd and Hg, in the two molluscs studied, follow a biphasic pattern, whereby a steady state is reached after approximately 16 days' exposure and then accumulation increases again for the rest of exposure period. The elimination of the two metals is also characterized in a biphasic way: fast excretion for the first four days followed by a slower depuration for the rest of exposure time. A two-compartment bioaccumulation model has been used to described the different kinetic parameters: (1) the rate constant for depuration; (2) the rate constant for uptake; (3) the theoretical bioconcentration factor extrapolated to steady-state conditions; and (4) the biological half-life of the metals.  相似文献

Concentrations of trace elements (Hg, Cd, Pb, Zn, Cu, Fe, and Se) in different organs and tissues (liver, kidney, muscle tissue, spleen, heart, lung, and fat tissue) of loggerhead turtles Caretta caretta from eastern Mediterranean Sea were determined. The highest levels of mercury and cadmium were found in liver (Hg: 0.43 microg g(-1) wet weight; Cd: 3.36 microg g(-1) wet weight) and kidney (Hg: 0.16 microg g(-1) wet weight; Cd: 8.35 microg g(-1) wet weight). For lead the overall concentrations were low and often below the limit of detection. Copper and selenium tended to be higher in liver than in other tissues and organs, while for zinc the concentrations were quite homogenous in the different organs and tissues, except fat tissue (64.7 microg g(-1) wet weight) which showed a higher accumulation of this element. For iron the greatest concentrations were observed in liver (409 microg g(-1) wet weight) and spleen (221 microg g(-1) wet weight).  相似文献

As part of a research programme on the transfer of several radionuclides along a pelagic trophic chain, two groups of 12 trout were kept for 8 weeks in water contaminated with 30 Bq ml(-1) of (54)Mn. In order to simulate chronic contamination and limit alterations in the physical and chemical characteristics of the medium, the water was renewed every 2 days. The kinetics of the accumulation and elimination of the radionuclide were monitored in one group of fish. The second group was used to study the contamination of the main organs and tissues at the end of the accumulation phase. The dynamics of contamination can be described by a bi-compartmental model, taking into account the fluctuations in the concentration of (54)Mn in the water, as well as the biological dilution resulting from the growth of the fish. The theoretical value of the steady-state concentration factor for zero growth is 13 (w.w.) and the radionuclide release is characterised by two biological half-lives of 6 and 97 days. At the end of the accumulation phase, the (54)Mn is preferentially fixed in the bone, gills, skin and brain. The data obtained at the end of the depuration phase allow one to classify the organs in two groups with different elimination kinetics. The first group consists of organs of penetration or transit, such as the skin, gills, kidneys, liver, primary and secondary gut and viscera, whereas the second group is made up of the receptor and storage organs and tissues such as the bone, head, fins and muscle.  相似文献

This study is to elucidate the specific accumulation of 20 trace elements in tissues/organs of great cormorants from two different colonies (Lake Biwa and Mie) in Japan. In the body distribution of trace elements, some elements revealed tissue-specific accumulation such as most of the burden of Mo, Ag and Cd in liver, Tl and Cd in kidney, Cu, Rb and Cs in muscle, and V, Sr and Ba in bone. Gender-related variation was not observed in both populations for most of the trace elements, except for higher hepatic Sr in males from Lake Biwa. Hepatic V, muscular Hg and Tl, and Cd in liver, kidney and muscle increased with growth. Comparison of trace element levels in tissues between the two colonies showed that Cr, Rb, Sr, Cd, Cs, Ba and Tl levels were higher in Lake Biwa than in Mie, whereas Zn, Co and Hg in Mie samples were greater than Lake Biwa. Variations of elemental levels in stomach contents also showed similar patterns, thus, showing that dietary sources tended to be the main factor for these regional variations. Toxic Hg and Cd concentrations in the liver of cormorants from the two colonies were lower than those from other areas, implying relatively low exposure to these metals in the present study sites. Concentrations of V, Co, Ag, Cd, Cs, Hg, Tl, Pb and Bi in liver remained more or less at the same level between 1993 and 2003, while hepatic Cr, Mn, Cu, Zn, Se, Rb, Sr and Ba showed apparent decrease, which might be related to the biological factors.  相似文献

Histopathological alterations to white seabass, Lates calcarifer aged 3 months in acute and subchronic cadmium exposure were studied by light and scanning electron microscopy. The 96-h LC50 values of cadmium to L. calcarifer was found to be 20.12 +/- 0.61 mg/l and the maximum acceptable toxicant concentration (MATC) was 7.79 mg/l. Fish were exposed to 10 and 0.8 mg/l of Cd (as CdCl,H2O) for 96 h and 90 days, respectively. The study showed that gill lamellae and kidney tubules were the primary target organs for the acute toxic effect of cadmium while in the subchronic exposure, the toxic effect to gills was less than that of kidney and liver. Gill alterations included edema of the epithelial cells with the breakdown of pillar cell system, aneurisms with some ruptures, hypertrophy and hyperplasia of epithelial and chloride cells. The liver showed blood congestion in sinusoids and hydropic swelling of hepatocytes, vacuolation and dark granule accumulation. Lipid droplets and glycogen content were observed in hepatocytes at the second and third month of subchronic exposure. The kidney showed hydropic swelling of tubular cell vacuolation and numerous dark granule accumulation in many tubules. Tubular degeneration and necrosis were seen in some areas.  相似文献

Concentrations of As, Al, Ag, Ba, Bi, Cd, Co, Cr, Cu, Fe, Ga, Mg, Mn, Ni, Pb, Sb, Sn, Sr, Tl, V and Zn were analyzed by inductively coupled mass spectrometry (ICP-MS) in the intestinal helminth Pomphorhynchus laevis and its host Barbus barbus. The fish were caught in the Danube river downstream of the city of Budapest (Hungary). Ten out of twenty one elements analyzed were found at higher concentrations in the acanthocephalan than in different tissues (muscle, intestine, liver and kidney) of barbel. Considering the fish tissues, most of the elements were present at highest concentrations in liver, followed by kidney, intestine and muscle. Spearman correlation analyses indicate that there is competition for metals between the parasites and the host. The negative relationships between parasite number and metal levels in organs of the barbel support this hypothesis. The bioconcentration factors for Ag, As, Ba, Bi, Cu, Ga, Mn, Pb, Sr, Tl, and Zn showed that the parasites concentrated metals to a higher degree than the fish tissues. They accumulated the metals As, Cd, Cu, Fe, Ni, Pb, Sr and Zn even better than established bioindicators such as the mussel Dreissena polymorpha as revealed by data from the literature. The results presented here emphasize that acanthocephalans of fish are very useful as sentinels for metal pollution in aquatic ecosystems. Ratio of metal concentrations in the parasites and the host tissues provide additional information. Not including acanthocephalans in accumulation bioindication studies with fishes (as still customarily done) may lead to false results.  相似文献

Field results have shown that Mytilus californianus is able to release its Cd concentrations significantly in just a few days. The existing paradigm states that Cd elimination from Mytilus soft tissues is a very slow process. This discrepancy was investigated in the laboratory, testing the effect of two Cd levels (10 and 1 microgram l-1) on its release from Mytilus trossulus and M. californianus soft tissues. After exposure to 10 micrograms l-1, both species showed a significant uptake with no elimination after several days of depuration. After exposure to 1 microgram l-1, the responses were different. No significant Cd uptake was seen in M. trossulus while in M. californianus uptake was significant but returned to the background level after just 1 day of depuration. This response of M. californianus is consistent with that reported from field studies. These results are important for environmental monitoring programs since M. californianus has been used as equivalent to other Mytilus species in the assessment of Cd pollution.  相似文献

Mercury (Hg), cadmium (Cd), iron (Fe) manganese (Mn), zinc (Zn) and copper (Cu) concentrations in the liver, kidney and muscle of nine killer whales (including three calves) that stranded together in the northern area of Japan were determined. The Hg and Cd concentrations were found at trace levels in the calf organs, and increased with age. The Fe concentration in the muscle was significantly lower in the calves than in the mature whales and also increased with age. In contrast, Mn and Cu concentrations in the muscle were significantly higher in the calves than in the mature whales, and changes in the Zn concentration relative to age were unclear. These results suggest minimal mother-to-calf transfer of the toxic metals Hg and Cd and accumulation of these metals in the organs with age, while the essential metals Mn and Cu were found at higher concentrations in the muscle of calves than in mature whales.  相似文献

BaP is one of the most studied PAH, due to its ubiquitous presence in aquatic environments and toxicity to aquatic organisms. The main goal of this study was to assess BaP effects in Nile Tilapia after waterborne and dietary exposures, through the evaluation of EROD and GST activities in liver, gills and intestine, and BaP metabolites in bile; and also to evaluate the usefulness of these commonly used biomarkers after two different routes of exposure. Waterborne exposure to BaP led to a significant induction of EROD in all tissues analyzed (644%, 1640% and 2880% in relation to solvent in liver, gill and intestine respectively) while in dietary exposures EROD was induced only in intestine (3143%) after exposure to high BaP concentrations. GST activities with CDNB were slightly induced in liver (40%) and in gill (66%) after water exposure to BaP, and in intestine after dietary exposure to low BaP concentrations (182%). BaP metabolites in bile increased after both exposure routes, and were highly correlated with EROD activity after water exposure. In summary, this work has shown that the effects of BaP on biotransformation pathways depend on the route of exposure. Moreover, barrier tissues like gills and intestine also have an important role in the first-pass metabolism of BaP, reducing the amount of parent compound that reaches the liver to be metabolized. For that reason, EROD activity as a biomarker of exposure should also be applied in extrahepatic organs, like gills and intestine, in monitoring studies. Biliary BaP type metabolites are good reflectors of contamination levels under both exposure routes, while GST activity with CDNB as substrate, as a phase II enzyme, does not seem a reliable biomarker of exposure to BaP regardless the route of exposure.  相似文献

Freshwater fish Cyprinus carpio was selected for the study of bioaccumulation of organochlorinated pesticides in tissues like gills, muscle, intestine, kidney, and liver in a continuous fed system. The pesticides used were Aldrin, Dieldrin, BHC, and DDT. The bioaccumulation of Dieldrin was maximum of 85.0 microg g(-1) wet weight in liver tissue while minimum of 7.30 microg g(-1) wet weight for DDT at 30 days exposure time. Bioconcentration factor (BCF) has followed the same trend in liver tissue for Dieldrin and DDT. The rate of bioaccumulation was found to be maximum of 4.3879 microg g(-1) wet weight in liver tissue and minimum of 0.0021 microg g(-1) wet weight in gill tissue for 30 days exposure. As evidenced by the increasing values of BCF, pesticide uptake also showed increased trend with the increase in exposure time. A high correlation coefficient ranging between 0.7247 and 0.9616 between the pesticide concentration and exposure time was observed. Based on actual BCF values, log Kow were calculated and the values are well within the reported values of 6.5 indicating efficient relationship between BCF and log Kow because beyond the 6.5 the bioconcentration levels off.  相似文献

The histopathological changes of fenvalerate on the gill, kidney, liver and intestine tissues of the Cirrhinus mrigala were determined by light microscopy. The fish were exposed to two sub-lethal concentrations of fenvalerate (1.5-3.0 ppb). The most common gill changes at all concentrations of fenvalerate were epithelial hyperplasia, epithelial necrosis, desquamation and lamellar fusion. Besides, epithelial lifting, oedema, swelling at the tips of secondary lamellae and curling of secondary lamellae were other histopathological changes. Necrosis of tubular epithelium, pycnotic nuclei in the hematopoietic tissue, hypertrophied epithelial cells of renal tubules, narrowing of the tubular lumen, expansion of space inside the Bowman's capsule and contraction of the glomerulus were observed in kidney tissues of fish. Hepatic lesions in the liver tissues of fish exposed to fenvalerate were characterized by congestion, cloudy swelling of hepatocytes and focal necrosis. Atrophy of epithelial cells, necrosis of epithelial cells, desquamation of mucosal epithelium and infiltration of lymphocytes into the lamina propria were detected in intestine tissues of fish after exposure to fenvalerate.  相似文献

This study determined the metal levels in sediments and tissues of a common estuarine fish, Tetractenos glaber (smooth toadfish), from two metal contaminated and two reference estuaries near Sydney, Australia. Metal levels were highest in sediments and fish from contaminated estuaries. Gonads contained the highest metal levels followed by muscle, gill and liver. Metal accumulation was gender-dependant (e.g. male gonads were >20 times higher in As than females; female gills were >30 times higher than males for Pb). Cadmium, Pb and Ni levels in fish tissues reflected sediment levels, indicating sediment and/or dietary metal uptake. Levels of As, Co, Cd & Pb in gills showed similar patterns to other tissues, suggesting that metals may have been taken up by gills through contaminated water. Similar metal patterns in tissues and sediments suggest more than one uptake pathway. This study indicates that multiple factors influence metal accumulation in fish.  相似文献

The accumulation of two heavy metals (chromium (Cr) and cadmium (Cd)) in the liver, kidney and gut of Rana ridibunda exposed to Cr or to a mixture of Cr and Cd was investigated. The concentration of metallothioneins (MTs) in the same tissues was also studied. Both metals accumulated mainly in the kidney. Cr accumulation in the liver and gut was not affected by the presence of Cd. Furthermore, Cr concentration in the kidney was doubled when Cd was present. MT concentration did not increase after Cr treatment but it increased two- to six-fold over control values in mixture-exposed frogs, the highest value being observed in the gut. MTs in the gut could act as a barrier preventing ingested heavy metals from entering the blood stream. MT concentration correlated positively with Cd concentrations in both the liver and the gut of mixture-exposed animals.  相似文献

Apart from analyses for elemental contaminants in field collected specimens, very little is known about the assimilation, accumulation and toxic effects of inorganic contaminants in reptiles. This study examined the chronic accumulation of Cd in a European lacertid lizard (Podarcis carbonelli) following dietary provision of an environmentally realistic concentration of Cd for 21 weeks. Lizards were provided with Cd that had either been biologically incorporated into crickets, or as Cd(NO3)2 added superficially to crickets just prior to feeding. Among both treatment groups Cd accumulated in tissues in the following order of concentration: gut>liver>kidney>carcass. The majority of the Cd was retained within the gut, and transfer to internal organs was low. Morphological indices, brain and plasma cholinesterase activities, gut and liver metallothionein content, and standard metabolic rate were measured as biomarkers of exposure and effect; however, no differences between control lizards and Cd-treated lizards were observed.  相似文献
