Enhanced biological phosphorus removal (EBPR) from wastewater relies on the enrichment of activated sludge with phosphorus-accumulating organisms (PAOs). The presence and proliferation of glycogen-accumulating organisms (GAOs), which compete for substrate with PAOs, may be detrimental for EBPR systems, leading to deterioration and, in extreme cases, failure of the process. Therefore, from both process evaluation and modeling perspectives, the estimation of PAO and GAO populations in activated sludge systems is a relevant issue. A simple method for the quantification of PAO and GAO population fractions in activated sludge systems is presented in this paper. To develop such a method, the activity observed in anaerobic batch tests executed with different PAO/GAO ratios, by mixing highly enriched PAO and GAO cultures, was studied. Strong correlations between PAO/GAO population ratios and biomass activity were observed (R2 > 0.97). This served as a basis for the proposal of a simple and practical method to quantify the PAO and GAO populations in activated sludge systems, based on commonly measured and reliable analytical parameters (i.e., mixed liquor suspended solids, acetate, and orthophosphate) without requiring molecular techniques. This method relies on the estimation of the total active biomass population under anaerobic conditions (PAO plus GAO populations), by measuring the maximum acetate uptake rate in the presence of excess acetate. Later, the PAO and GAO populations present in the activated sludge system can be estimated, by taking into account the PAO/GAO ratio calculated on the basis of the anaerobic phosphorus release-to-acetate consumed ratio. The proposed method was evaluated using activated sludge from municipal wastewater treatment plants. The results from the quantification performed following the proposed method were compared with direct population estimations carried out with fluorescence in situ hybridization analysis (determining Candidatus Accumulibacter Phosphatis as PAO and Candidatus Competibacter Phosphatis as GAO). The method showed to be potentially suitable to estimate the PAO and GAO populations regarding the total PAO-GAO biomass. It could be used, not only to evaluate the performance of EBPR systems, but also in the calibration of potential activated sludge mathematical models, regarding the PAO-GAO coexistence.  相似文献

A sequencing batch reactor was used to study the possibility of harvesting polyhydroxyalkanoate (PHA) from enhanced biological phosphorus removal (EBPR) processes without compromising treatment quality. Because, in EBPR, the highest PHA concentrations are observed after exposure of the sludge to anaerobic conditions, PHA accumulation was evaluated with collection of waste activated sludge (WAS) at the end of the anaerobic stage, in addition to the traditional removal after the aerobic stage. The system achieved good phosphorus removal, regardless of the point of WAS collection. When sludge was harvested at the end of the anaerobic stage, the PHA content of the sludge ranged from 7 to 16 mg PHA/100 mg mixed liquor volatile suspended solids. Although this level of PHA production is below levels obtained with pure cultures, the demonstrated ability to harvest PHA, while simultaneously satisfying phosphorus removal in an EBPR process, is a key initial step towards of the use of wastewater treatment plants for PHA production.  相似文献

Wastewater in Shaoxing wastewater treatment plant (SWWTP) is composed of more than 90% dyeing and printing wastewater with high pH and sulfate. Through a combination process of anaerobic acidogenic [hydraulic retention time (HRT) of 15h], aerobic (HRT of 20h) and flocculation-precipitation, the total COD removal efficiency was up to 91%. But COD removal efficiency in anaerobic acidogenic unit was only 4%. As a comparison, the COD removal efficiency was up to 35% in the pilot-scale upflow anaerobic sludge bed (UASB) reactor (HRT of 15h). GC-MS analysis showed that the response abundance of these wastewater samples decreased with their removal of COD. A main component of the raw influent was long-chain n-alkanes. The final effluent of SWWTP had only four types of alkanes. After anaerobic unit at SWWTP, the mass percentage of total alkanes to total organic compounds was slightly decreased while its categories increased. But in the UASB, alkanes categories could be removed by 75%. Caffeine as a chemical marker could be detected only in the effluent of the aerobic process. Quantitative analysis was given. These results demonstrated that GC-MS analysis could provide an insight to the measurement of organic compounds removal.  相似文献

Enhanced biological phosphorous removal (EBPR) performance was found to be adequate with reduced return-activated sludge (RAS) flows (50% of available RAS) to the anaerobic tank and smaller-than-typical anaerobic zone volume (1.08 hours hydraulic retention time [HRT]). Three identical parallel biological nutrient removal pilot plants were fed with strong, highly fermented (160 mg/L volatile fatty acids [VFAs]), domestic and industrial wastewater from a full-scale wastewater treatment facility. The pilot plants were operated at 100, 50, 40, and 25% RAS (percent of available RAS) flows to the anaerobic tank, with the remaining RAS to the anoxic tank. In addition, varying anaerobic HRT (1.08 and 1.5 hours) and increased hydraulic loading (35% increase) were examined. The study was divided into four phases, and the effect of these process variations on EBPR were studied by having one different variable between two identical systems. The most significant conclusion was that returning part of the RAS to the anaerobic zone did not decrease EBPR performance; instead, it changed the location of phosphorous release and uptake. Bringing less RAS to the anaerobic and more to the anoxic tank decreased anaerobic phosphorus release and increased anoxic phosphorus release (or decreased anoxic phosphorus uptake). Equally important is that, with VFA-rich influent wastewater, excessive anaerobic volume was shown to hurt overall phosphorus removal, even when it resulted in increased anaerobic phosphorus release.  相似文献

The potential to use the results of biochemical acid potential (BAP) tests to predict the ultimate digestibility of raw and pretreated waste activated sludge (WAS) was investigated. The ultimate methane production from biochemical methane potential (BMP) tests on raw and pretreated samples which spanned a range of biodegradability proved linearly related to the volatile fatty acid (VFA) and soluble chemical oxygen demand (COD) production in corresponding BAP tests. In addition, a linear relationship between NH4-N production in the BMP and BAP tests was observed. Despite the linear nature of the relationships, the ratio of the production of methane in the BMP tests to the production of VFAs in the BAP tests varied with the biodegradability of the sludge samples. Waste Activated Sludge samples with low digestibility had ultimate yields of CH4 that were greater than the VFA yields in BAP tests, whereas sludge samples with high digestibility had lower yields of CH4 than the corresponding VFA yields. This trend contrasted with the NH4 results, in which the yields in the BAP tests were consistently less than those observed in the BMP tests. It was hypothesized that the varying relationship between CH4 and VFA yields was because of the inhibition of anaerobic oxidation of long-chain fatty acids (LCFAs) in the BAP tests. Long-chain fatty acids would be converted to CH4 in BMP tests but produced as digestion intermediates in the BAP tests and were not measured as part of the VFA yield. Hydrogen and acetate were identified as the two most likely intermediates that would accumulate in the BAP tests (which would cause inhibition). A stoichiometric model to facilitate the development of an improved understanding of the biodegradation processes in the BAP and BMP tests was assembled. When the model was applied to the BAP tests the anaerobic oxidation of LCFAs and propionate and methanogenesis were excluded from the model. The model was employed to estimate the extent of degradation of lipids, carbohydrates, and proteins in the batch tests as a function of the ultimate biodegradability of the sludge samples. On the basis of model fitting, it was determined that the degradation of lipids in BMP tests decreased, whereas the degradation of carbohydrates and proteins increased as the digestibility of the sludge samples increased. The varying ratio of lipid to protein and carbohydrate degradability with increasing digestibility of the sludge samples describes the relationship between VFA production and CH4 production in the BAP, and BMP tests, respectively.  相似文献

A membrane-assisted and a conventional activated sludge system, both operated in an enhanced biological phosphorus removal (EBPR) mode and under identical operating conditions, were studied to investigate the effect of the membrane solids-liquid separation on nitrification activity. Both the membrane EBPR (MEBPR) and conventional EBPR (CEBPR) processes achieved stable and complete removal of ammonium-nitrogen from the influent wastewater. However, when the intrinsic nitrification activity was assessed in offline batch tests, the CEBPR mixed liquor exhibited 15 to 75% greater nitrification potential than the MEBPR counterpart. These results were further validated by monitoring nitrification rates of conventional mixed liquor as it evolved toward a membrane mixed liquor. It was also demonstrated that the larger aerobic mass fraction of the MEBPR system could not be the only factor influencing the reduced intrinsic nitrification rate. The present study strongly suggests that the presence of a membrane solids-liquid separation per se may be sufficient to alter the nitrification kinetics of an EBPR mixed liquor and that this possibility should be considered in arriving at an appropriate process design.  相似文献

The objective of this study was to investigate the sorption capacity and the fractionation of sorbed nickel and cobalt onto anaerobic granular sludges. Two different anaerobic granular sludges (non-fed, pH=7) were loaded with nickel and cobalt in adsorption experiments (monometal and competitive conditions). The combination of sequential extraction with the sorption isotherm analysis allowed the assessment of the sorption capacity of individual fractions present in the anaerobic granular sludges. The operational fractionation of the sorbed heavy metals was determined using a modified Tessier sequential extraction procedure. The sorption characteristics of each extracted fraction (exchangeable, carbonates, organic matter/sulfides and residual fractions) fitted well to the Langmuir model. The organic matter/sulfides fraction showed the highest affinity for cobalt and nickel in both sludges investigated compared to the other operationally defined fractions. The presence of iron negatively affected cobalt and nickel accumulation in this organic matter/sulfides fraction. The trace metals-iron sulfide interactions are likely to be the key process in controlling the distribution of cobalt and nickel during sorption onto non-fed methanogenic granules due to the high affinity of iron sulfides towards the metals studied.  相似文献

剩余污泥发酵既能实现污泥减量,又能获得污水生物脱氮除磷所需的挥发性脂肪酸VFAs等。采用序批式污泥发酵实验,在(30±1)℃的条件下,研究剩余污泥投配比对序批式污泥发酵系统污泥减量、产酸以及基质释放过程的影响,探索VFAs生成最大化的条件。实验结果表明:投加接种污泥不能明显提高序批式污泥发酵系统剩余污泥的减量效果,但可以改善污泥的产酸特性;当污泥投配比大于50%时,发酵过程中将出现2个VFAs浓度峰,且第二次峰浓度更大;较为理想的序批式污泥发酵投配比是60%~80%、发酵时间为5 d或25 d,前者VFAs表观转化率最大(约24%),后者发酵液中VFAs浓度最高(约600 mg/L);发酵过程中,NH4+-N总体呈上升趋势,磷浓度发酵5 d时达到最大值。  相似文献

Goals, Scope and Background

From 2005, deposition of organic waste will be banned in Sweden. Likewise, in Germany and Austria, similar bans are being planned, and further countries will probably follow. Thus, there is a need to develop new methods and to refine established techniques for sludge management in the whole of the European Union. For this end, there is also an urgent need for appropriate ecotoxicological approaches to elucidate and assess the hazard potential of sewage sludge. Therefore, the present study was designed to assess the capacity of various established sludge treatment methods using different oxygen regimes to degrade recalcitrant nitro-substituted organic compounds and reduce their toxicity. Sewage sludge samples from a wastewater treatment plant in Sweden (Cambrex Karlskoga AB, industrial area Björkborn) receiving wastewater from industries manufacturing pharmaceutical substances, chemical intermediates and explosives were processed with different sludge treatment methods. Among other treatment methods, bioreactors (for anaerobic and aerobic sludge treatment) were used. In the present investigation, a battery ofin vitro bioassays was employed to compare the cytotoxic and genotoxic potentials of different fractions of sludge samples in order to elucidate whether the treatments were suitable to reduce the toxicity of the sludge.

Methods

In order to investigate the cytotoxicity of the extracts of treated and untreated sludge samples, the acute cytotoxicity test with the permanent cell line RTL-W1 was used. Genotoxicity was tested by means of the comet assay (single cell gel electro-phoresis) with RTL-W1cells, and mutagenicity was assessed with the Ames test using the Salmonella typhimurium strains TA98, TA98NR and TA100. Sludge toxicity was tested in different fractions of organic extracts produced by acetone and hexane extractions. The subsequent clean-up procedure (silica gel chro-matography and elution with hexane and dichloromethane) resulted in two fractions, a lipophilic hexane-fraction and a semi-lipophilic dichloromethane-fraction. For the genotoxicity and mutagenicity tests, these fractions were reunited at equal ratios.

Results and Discussion

The acute cytotoxicity test with RTL-Wl cells revealed a high cytotoxic potential for the semi-li-pophilic DM-fractions of all sludge samples with NR50 values (= effective concentration for 50% cell death in the neutral red test) from 8.9 up to 20 mg sludge d.w./ml medium. A low cytotoxic potential for the hexane fractions of the untreated sludge samples (NR50 400 to < 400 mg sludge d.w./ml medium) was observed, whereas the hexane fractions of the treated sludge samples showed elevated cytotoxicity increasing further with treatment in the bioreactors. The comet assay indicated that three out of eight of the reunited fractions had a significant genotoxic potential. Whereas the genotoxic potential of one sample treated anaerobically was very high with an induction factor of 11.6, a similar sample (taken from the same anaerobic reactor four months later) and one untreated sample showed lower potentials. The samples treated in another anaerobic bioreactor as well as the samples treated aerobically showed no genotoxic potential. Results indicate that aerobic treatment was basically adequate for reducing the genotoxicity of the sludge, whereas anaerobic treatment was only partly useful for reduction of genotoxicity. The Ames test revealed a very high mutagenic potential for the reunited fractions of the untreated sludge samples with strain TA98 (maximum induction factors (IFmax up to 45) and a relatively high potential for one of the samples treated aerobically (S2, IFmax = 18 (TA98, S9-)), thus documenting the suitability of both anaerobic and aerobic treatments to reduce the mutagenicity of the samples, however, with the aerobic treatment being less effective. Conclusions. Overall, none of the microbiological treatments for wastewater sludge in bioreactors was found to be ideal for general toxicity reduction of the sludge samples. Whereas cytotoxicity of the sludge increased or levelled off in most cases following either treatment, genotoxicity both increased or decreased after anaerobic treatment, depending on the specific sample. However, mutagenicity could generally be reduced by anaerobic treatment and, to a lesser degree, by aerobic treatment. Recommendationsand Perspectives. The complex modification of the diverse damage potentials of sludge sample extracts by use of anin vitro biotest battery following treatment for toxicity reduction in bioreactors showed that considerations of different toxicological endpoints is essential for an adequate hazard assessment. Whereas in the case of cytotoxicity reduction, the reactors proved ineffective, mutagenicity could be reduced significantly at least in some cases in this case study.

In this study, the combined effects of temperature and solids retention time (SRT) on enhanced biological phosphorus removal (EBPR) performance and the mechanism of EBPR washout were investigated. Two pilot-scale University of Cape Town (South Africa) systems fed with synthetic wastewater were operated at 5 and 10 degrees C. The results showed that the phosphorus removal performance was optimum at total SRT ranges of 16 to 24 days and 12 to 17 days for 5 and 10 degrees C, respectively, and steady-state phosphorus removal was greater at the lower temperature. Higher SRT values of up to 32 days at 5 degrees C and 25 days at 10 degrees C slightly reduced EBPR performance as a result of increased extent of endogenous respiration, which consumed internally stored glycogen, leaving less reducing power for poly-hydroxy alkanoate (PHA) formation in anaerobic stages. The phosphorus-accumulating organism (PAO) washout SRTs of the systems were determined as 3.5 days at 5 degrees C and 1.8 days at 10 degrees C, considerably less than the washout SRTs of nitrifiers. Polyphosphorus, the main energy reserve of the EBPR bacterial consortium, was not completely depleted, even at washout points. The inability of EBPR biomass to use glycogen to generate reducing power for PHA formation was the major reason for washout. The results not only suggest that glycogen mechanism is the most rate-limiting step in EBPR systems, but also that it is an integral part of EBPR biochemistry, as proposed originally by Mino et al. (1987), and later others (Pereira et al., 1996, Erdal et al., 2002; Erdal, Z. K., 2002). The aerobic washout SRT values (2.1 and 1.2 days for 5 and 10 degrees C, respectively) of this study did not fit the linear line for PAO washout developed by Mamais and Jenkins (1992). Perhaps this was because the feeds used during this study were chemical-oxygen-demand-limited (acetate-based synthetic feed), whereas the feeds used for their study were phosphorus-limited (external acetate added to domestic wastewater), resulting in different ratios of PAOs and nonPAOs in the biomass.  相似文献

Organic extracts of marine sediments from the North Sea and the Baltic Sea were investigated with two toxicity assays. The comet assay based on the fish cell line Epithelioma papulosum cyprini (EPC) was applied to determine the genotoxic potential; zebrafish embryos (Danio rerio) were used to quantify the teratogenic potential of the samples. EC(50) values were calculated from dose-response curves for both test systems. Highest teratogenic and genotoxic effects normalised to total organic carbon (TOC) content were detected in sediment samples of different origins. Polychlorinated biphenyls (PCBs) and polycyclic aromatic hydrocarbons (PAHs) are not likely to be the causes of the observed effects, as demonstrated by a two-step fractionation procedure of selected extracts. The toxic potential was more pronounced in fractions having polarity higher than those possessed by PAHs and PCBs. The suitability of the two in vitro test systems for assessing genotoxic and teratogenic effects of marine sediment extracts could be demonstrated.  相似文献

The anaerobic biodegradability of wastewaters from different chemical plants was determined by an integrated test strategy consisting of an anaerobic toxicity test, a static anaerobic biodegradation test and a continuously operated fixed bed reactor test. This integrated procedure offered a useful tool for screening wastewaters for anaerobic biodegradability. Four different wastewaters were tested for anaerobic toxicity and biodegradability. A more detailed characterization of particulate organic matter in wastewater streams is needed to improve solid-liquid separation and biological processes for wastewater treatment. The objective of this paper was to evaluate particle size distributions and the associated chemical composition for municipal, industrial, and agricultural waste streams. Most of the organic matter in these wastewaters was larger than a molecular weight of 10(3) amu and therefore would require extracellular hydrolysis before any bacterial metabolism. Particle size distributions were significantly different for the studied waste streams. In municipal wastewater, the organic matter was evenly distributed in all eight size fractions ranging from 10(3) amu up to 63 microm. The industrial and agricultural wastewaters, however, contained mainly soluble organic matter (<10(3) amu) and larger particles (>1.2 microm for the industrial and >10 microm for the agricultural waste) leaving a gap in the size range of large macromolecules and colloids. The relative protein and carbohydrate concentrations varied for the different size fractions compared to the measured chemical oxygen demand (COD) in the corresponding size fraction. Thus, the design of the solid-liquid separation at a treatment plant could be used to purposefully modify the overall chemical composition of the organic matter before further biological treatment. Particle size distributions will influence design and operation of biological nutrient removal processes such as denitrification or biological phosphorus removal that may be carbon limited if a large fraction of the organic matter is composed of large particles with slow hydrolysis rates. Measured particle size distributions for the different waste streams in this study (municipal, industrial, agricultural) were significantly different requiring specific approaches for treatment plant design.  相似文献

A preliminary bench-scale study of parallel University of Cape Town (UCT) biological nutrient removal systems showed improvement in anoxic denitrification rates resulting from prefermentation of a septic (i.e., high volatile fatty acid [VFA] content), phosphorus-limited (i.e., total chemical oxygen demand/total phosphorus [TP] ratio < 40:1) wastewater. Net phosphorus removals due to enhanced biological phosphorus removal (EBPR) were only improved marginally by prefermentation in spite of significant increases in anaerobic phosphorus release, polyhydroxyalkanoate formation, and higher anoxic and aerobic uptakes. This probably was due to the high VFA/TP ratio in the raw influent relative to the VFA requirements for EBPR because enough VFAs were already present for phosphorus removal prior to prefermentation. An additional assessment of prefermentation using parallel UCT systems with step feed of 50% of the influent to the anoxic zone was completed. This second phase quantified the effect of prefermentation in a step-feed scenario, which prioritized prefermentation use to enhance denitrification rather than EBPR. While specific denitrification rates in the anoxic zone were significantly improved by prefermentation, high denitrification in the clarifiers and aerobic zones (simultaneous denitrification) made definitive conclusions concerning the potential improvements in total system nitrogen removal questionable. The prefermented system always showed superior values of the zone settling velocity and sludge volume index and the improvement became increasingly statistically significant when the prefermenter was performing well.  相似文献

This paper reports a study of the anaerobic biodegradation of non-ionic surfactants alkyl polyglucosides applying the method by measurement of the biogas production in digested sludge. Three alkyl polyglucosides with different length alkyl chain and degree of polymerization of the glucose units were tested. The influence of their structural parameters was evaluated, and the characteristics parameters of the anaerobic biodegradation were determined. Results show that alkyl polyglucosides, at the standard initial concentration of 100 mgC L?1, are not completely biodegradable in anaerobic conditions because they inhibit the biogas production. The alkyl polyglucoside having the shortest alkyl chain showed the fastest biodegradability and reached the higher percentage of final mineralization. The anaerobic process was well adjusted to a pseudo first-order equation using the carbon produced as gas during the test; also, kinetics parameters and a global rate constant for all the involved metabolic process were determined. This modeling is helpful to evaluate the biodegradation or the persistence of alkyl polyglucosides under anaerobic conditions in the environment and in the wastewater treatment.  相似文献

The simultaneous fate of organic matter and 4 endocrine disruptors (3 polycyclic aromatic hydrocarbons (PAHs) (fluoranthene, benzo(b)fluoranthene, and benzo(a)pyrene) and nonylphenols (NP)) was studied during the anaerobic digestion followed by composting of sludge at lab-scale. Sludge organic matter was characterized, thanks to chemical fractionation and 3D fluorescence deciphering its accessibility and biodegradability. Total chemical oxygen demand (COD) removal was 41% and 56% during anaerobic digestion and composting, respectively. 3D fluorescence highlighted the quality changes of organic matter. During continuous anaerobic digestion, organic micropollutants’ removal was 22?±?14%, 6?±?5%, 18?±?9%, and 0% for fluoranthene, benzo(b)fluoranthene, benzo(a)pyrene, and nonylphenols, respectively. Discontinuous composting allowed to go further on the organic micropollutants’ removal as 34?±?8%, 31?±?20%, 38?±?10%, and 52?±?6% of fluoranthene, benzo(b)fluoranthene, benzo(a)pyrene, and nonylphenols were dissipated, respectively. Moreover, the accessibility of PAH and NP expressed by their presence in the various sludge organic matter fractions and its evolution during both treatments was linked to both the quality evolution of the organic matter and the physicochemical properties of the PAH and NP; the presence in most accessible fractions explained the amount of PAH and NP dissipated.

The abundance and relevance ofAccumulibacter phosphatis (presumed to be polyphosphate-accumulating organisms [PAOs]), Competibacter phosphatis (presumed to be glycogen-accumulating organisms [GAOs]), and tetrad-forming organisms (TFOs) to phosphorus removal performance at six full-scale enhanced biological phosphorus removal (EBPR) wastewater treatment plants were investigated. Coexistence of various levels of candidate PAOs and GAOs were found at these facilities. Accumulibacter were found to be 5 to 20% of the total bacterial population, and Competibacter were 0 to 20% of the total bacteria population. The TFO abundance varied from nondetectable to dominant. Anaerobic phosphorus (P) release to acetate uptake ratios (P(rel)/HAc(up)) obtained from bench tests were correlated positively with the abundance ratio of Accumulibacter/(Competibacter +TFOs) and negatively with the abundance of (Competibacter +TFOs) for all plants except one, suggesting the relevance of these candidate organisms to EBPR processes. However, effluent phosphorus concentration, amount of phosphorus removed, and process stability in an EBPR system were not directly related to high PAO abundance or mutually exclusive with a high GAO fraction. The plant that had the lowest average effluent phosphorus and highest stability rating had the lowest P(rel)/HAc(up) and the most TFOs. Evaluation of full-scale EBPR performance data indicated that low effluent phosphorus concentration and high process stability are positively correlated with the influent readily biodegradable chemical oxygen demand-to-phosphorus ratio. A system-level carbon-distribution-based conceptual model is proposed for capturing the dynamic competition between PAOs and GAOs and their effect on an EBPR process, and the results from this study seem to support the model hypothesis.  相似文献
