A total of 144 isolates of Pseudomonas spp. (48 each from the Yamuna River water, wastewater irrigated soil and groundwater irrigated soil) were tested for their resistance against certain heavy metals and antibiotics. Minimum inhibitory concentrations (MICs) of Hg2?+?, Cd2?+?, Cu2?+?, Zn2?+?, Ni2?+?, Pb2?+?, Cr3?+? and Cr6?+? for each isolate were also determined. A maximum MIC of 200 ??g/ml for mercury and 3,200 ??g/ml for other metals were observed. The incidences of metal resistance and MICs of metals for Pseudomonas isolates from the Yamuna water and wastewater irrigated soil were significantly different to those of groundwater irrigated soil. A high level of resistance against tetracycline and polymyxin B (81.2%) was observed in river water isolates. However, 87.5% of Pseudomonas isolates from soil irrigated with wastewater showed resistance to sulphadiazine, whereas 79.1% were resistant to both ampicillin and erythromycin. Isolates from soil irrigated with groundwater exhibited less resistance towards heavy metals and antibiotics as compared to those of river water and wastewater irrigated soil. Majority of the Pseudomonas isolates from water and soil exhibited resistance to multiple metals and antibiotics. Resistance was transferable to recipient Escherichia coli AB2200 strains by conjugation. Plasmids were cured with the curing agent ethidium bromide and acridine orange at sub-MIC concentration.  相似文献

The aim of the present study was to determine the level of antibiotic resistance patterns and distribution of heavy metal resistance of bacterial isolates from aquatic animals (Lissotriton vulgaris, Pelophylax ridibundus, Emys orbicularis, Mauremys rivulata, and Natrix natrix) in Turkey (Kavak Delta). A total of 153 bacteria have been successfully isolated from cloaca and oral samples of the aquatic amphibians and reptilians which were found, namely, Aeromonas sp. (n?=?29), Plesiomonas sp. (n?=?7), Vibrio sp. (n?=?12), Citrobacter sp. (n?=?12), Enterobacter sp. (n?=?11), Escherichia sp. (n?=?22), Klebsiella sp. (n?=?22), Edwardsiella sp. (n?=?6), Hafnia sp. (n?=?1), Proteus sp. (n?=?19), Providencia sp. (n?=?8), and Pseudomonas sp. (n?=?4). In terms of antibiotic and heavy metal susceptibility testing, each isolate was tested against 12 antibiotics and 4 metals. There was a high incidence of resistance to cefoxitin (46.40 %), ampicillin (44.44 %), erythromycin (35.29 %), and a low incidence of resistance to gentamicin (6.53 %), kanamycin (8.49 %), chloramphenicol (9.15 %), and cefotaxime (10.45 %). The multiple antibiotic resistance index of each bacterial species indicated that bacteria from raised amphibians and reptiles have been exposed to tested antibiotics, with results ranging from 0 to 0.58. Most isolates showed tolerance to different concentrations of heavy metals, and minimal inhibition concentrations ranged from100 to >3,200 μg/mL. According to these results, a significant occurrence of bacteria in the internal organs of reptiles and amphibians, with a high incidence of resistance against antibiotics and heavy metals, may risk aquatic animals and the public health. These data appoint the importance of epidemiological surveillance and microbiological monitoring and reinforce the need to implement environment protection programs for amphibian and reptile species.  相似文献

A hundred Enterococcus strains were isolated from seawater samples collected from coastal areas of Istanbul. Isolates were identified to the species level using standard biochemical tests specified by Facklam and Collins. The species distribution was as follows Enterococcus faecalis (96%), Enterococcus gallinarum (3%) and Enterococcus solitarius (1%). The resistance of bacteria to both heavy metals (zinc [Zn], iron [Fe], cadmium [Cd], chrome [Cr], cobalt [Co]) and antibiotics (ampicillin 10 μg [AP], penicillin G 10 Units [PG], gentamycin 10 μg [GM], streptomycin 10 μg [S], chloramphenicol 10 μg [C], erythromycin 15 μg [E], kanamycin 30 μg [K], amikacin 30 μg [AK], nalidixic acid 30 μg [NA], and vancomycin 30 μg [VA]) was evaluated. None of the strains was resistant to VA. It was found that among the 100 isolates, those that exhibit resistance to antibiotics, particularly NA, S and K, were also resistant all the heavy metals tested. To our knowledge this is the first report focusing on determination of resistance of environmental enterococci found in Istanbul against heavy metals and antibiotics. Thus, combined expressions of antibiotic and heavy metal resistance may help to reinforce ecological and epidemiological studies and to determine the role of these strains in antibiotic and heavy metal resistance dissemination.  相似文献

Samples of soil, water, and sediments from industrial estates in Lagos were collected and analyzed for heavy metals and physicochemical composition. Bacteria that are resistant to elevated concentrations of metals (Cd2?+?, Co2?+?, Ni2?+?, Cr6?+?, and Hg2?+?) were isolated from the samples, and they were further screened for antibiotic sensitivity. The minimum tolerance concentrations (MTCs) of the isolates with dual resistance to the metals were determined. The physicochemistry of all the samples indicated were heavily polluted. Twenty-two of the 270 bacterial strains isolated showed dual resistances to antibiotics and heavy metals. The MTCs of isolates to the metals were 14 mM for Cd2?+?, 15 mM for Co2?+? and Ni2?+?, 17 mM for Cr6?+?, and 10 mM for Hg2?+?. Five strains (Pseudomonas aeruginosa, Actinomyces turicensis, Acinetobacter junni, Nocardia sp., and Micrococcus sp.) resisted all the 18 antibiotics tested. Whereas Rhodococcus sp. and Micrococcus sp. resisted 15 mM Ni2?+?, P. aeruginosa resisted 10 mM Co2?+?. To our knowledge, there has not been any report of bacterial strains resisting such high doses of metals coupled with wide range of antibiotics. Therefore, dual expressions of antibiotics and heavy-metal resistance make the isolates, potential seeds for decommissioning of sites polluted with industrial effluents rich in heavy metals, since the bacteria will be able to withstand in situ antibiosis that may prevail in such ecosystems.  相似文献

The concentrations of Cd, Cu, Zn, Fe, Pb, Ni, and Co were measured in gastropod mollusks Patella caerulea in the Mediterranean area. The organisms were collected at two coastal sites in Iskenderun Gulf during winter, spring, summer, and autumn 2008. Samples of the digestive gland, gill, and muscle were analyzed for heavy metals. The aim of study is to determine heavy metal levels in tissues of P. caerulea in different locations. Tissues of P. caerulea from the polluted site showed metal concentrations appreciably higher than unpolluted organisms. The highest metal levels were registered in the digestive gland of P. caerulea. Generally, digestive gland and gills showed higher metal concentrations than muscle. The average concentrations of heavy metals analyzed exhibited the following decreasing order: Fe > Zn > Cu > Ni > Cd > Pb > Co for both stations 1 and 2. Seasonal changes in metal concentrations were observed in the tissues of P. caerulea from a polluted and an unpolluted population.  相似文献

δ-Aminolevulinic acid dehydratase (ALAD; E.C. 4.2.1.24) is a metalloprotein and plays a crucial role in heme synthesis. Its sensitivity to toxic metals depends on the metallic co-factors. In this study the effects of some heavy metals on ALADs activity of five Pseudomonas isolates from Akar?ay stream (Afyonkarahisar) have been studied in order to determine whether their ALADs could be used as biosensor for lead and other heavy metals contamination. The data obtained from the study were analysed statistically by using SPSS 10.0 software for Windows. According to the results, Ni(II) increased the ALAD activity of Pseudomonas putida, Pseudomonas pseudoalcaligenes and Pseudomonas aureginosa ATTC 27853. Mn(II) also increased the enzymic activity in all strains examined except P. pseudoalcaligenes. These were found not to be statistically meaningful. P. aeruginosa 2’s enzymic activity was inhibited by Mg(II) and Zn(II), significantly (p’lt;’.05). There was a statistically meaningful relation between enzymic activity of both P. pseudoalcaligenes, P. putida and increasing Pb(II) concentration (p’lt;’.05). In addition, a formula was also deviced in order to determine the doses of metals in the environment of the organisms. As a result of the study, we can suggest that Pseudomonas ALADs can be used as a biosensor for lead and some other heavy metal exposure in aquatic environments.  相似文献

The increasing usage of antibiotics in the animal farming industry is an emerging worldwide problem contributing to the development of antibiotic resistance. The purpose of this work was to investigate the prevalence and antibiotic resistance profile of bacterial isolates collected from animal farming aquatic environments and meats in a peri-urban community in Daejeon, Korea. In an antibacterial susceptibility test, the bacterial isolates showed a high incidence of resistance (~26.04%) to cefazolin, tetracycline, gentamycin, norfloxacin, erythromycin and vancomycin. The results from a test for multiple antibiotic resistance indicated that the isolates were displaying an approximately 5-fold increase in the incidence of multiple antibiotic resistance to combinations of two different antibiotics compared to combinations of three or more antibiotics. Most of the isolates showed multi-antibiotic resistance, and the resistance patterns were similar among the sampling groups. Sequencing data analysis of 16S rRNA showed that most of the resistant isolates appeared to be dominated by the classes Betaproteobacteria and Gammaproteobacteria, including the genera Delftia, Burkholderia, Escherichia, Enterobacter, Acinetobacter, Shigella and Pseudomonas.  相似文献

Pathogenic species of Aeromonas produce a range of virulence factors, including aerolysin, cytotonic enterotoxins, and serine protease, to cause acute gastroenteritis and wound infections in humans and animals. Recognizing that not all Aeromonas strains are pathogenic, in this study, we proposed to evaluate Aeromonas removal effectiveness based on the presence of virulence gene-containing Aeromonas as a proper means to assess microbial risk of Aeromonas. We developed and applied real-time PCR assays to quantify serine protease (ser) gene- and heat-labile cytotonic enterotoxin (alt) gene-containing Aeromonas in water samples. Among 18 Aeromonas isolates from the source water, only three isolates possessed all three genes (aer, ser, and alt). A higher percent of isolates has either ser gene (89%) or alt gene (72%) compared to the percent of isolates containing aer gene (44%). Results of this study suggested that several different conventional and unconventional drinking water treatment processes could effectively remove Aeromonas from source water. As the comprehensive knowledge of the distribution of virulence factors in different Aeromonas species is currently not available, using real-time PCR to quantify various virulence factor genes in water samples and/or isolates can be a practical means for better assessment of microbial risks in water.  相似文献

The aim of this study was to monitor the heavy metal contents and fecal pollution in Tapes decussatus (carpet shell clam) from Izmir Bay (Eastern Aegean). Bivalve mollusks were sampled on January, March, July, and October 2007 in the Izmir Bay. Izmir Bay is one of the great natural bays of the Mediterranean. Concentrations of heavy metals were determined in the clams from the different seasons. Fecal coliform densities were determined to evaluate the degree of water pollution and clams’ microbiological accumulation of the classical microbial pollution indicators. The concentration of heavy metals in T. decussatus from Izmir Bay were Hg 0.044–0.13; Cd 0.026–0.24; Pb 0.38–1.2; Cr 2.3–3.7; Cu 6.4–8.4; Zn 56.0–81.8, and Ni 8.1–9.6 μg/g (dry weight). The maximum values were generally obtained in July and March except Ni. This study found that the levels of heavy metals except Cr in T. decussatus were below Spanish and European Communities legislations for shellfish as food. Microbial pollution indicators (heterotrophic bacteria and fecal coliform) were measured in T. decussatus. Maximum heterotrophic bacteria and fecal coliforms were recorded in the winter while the lowest were detected in summer.  相似文献

Contamination of surface waters has a direct impact on the public health of entire communities. Microorganisms inhabiting contaminated surface waters have developed mechanisms of coping with a variety of toxic metals and drugs. Investigations were carried out to isolate and identify lead-resistant bacteria from the river K?z?l?rmak along the city of K?r?kkale, Turkey. Of the 33 lead-resistant isolates, one isolate with a minimal inhibitory concentration of 1,200 mg?L?1 was isolated and identified as Enterococcus faecalis by using biochemical tests and 16S rRNA sequencing. Lead-resistant E. faecalis isolate was found out to be resistant to other heavy metals like aluminum, lithium, barium, chromium, iron, silver, tin, nickel, zinc, and strontium and to drugs like amikacin, aztreonam, and gentamicin. E. faecalis harbored four plasmids with the molecular sizes of 1.58, 3.06, 22.76, and 28.95 kb. Plasmid profile analyses of cured derivatives revealed that the lead resistance ability of E. faecalis was still existing despite the elimination of all the plasmids. Moreover, the antibiotic resistance pattern of the cured derivatives did not demonstrate any change from the parental strain. Our findings indicated that the lead resistance genes of E. faecalis were located on the chromosomal DNA rather than the plasmid.  相似文献

The sea bream??s nematode and Sparus aurata, sampled from the Iskenderun Bay, Mediterranean, in March 2008 were analyzed by inductively coupled plasma?Catomic emission spectrometry for their some heavy metal (Cd, Cr, Cu, Fe, Hg, Mn, Mg, Pb, and Zn) levels. The metal concentrations of the parasites were compared to different organs (liver, muscle, gill, intestine, and skin) of the fish hosts. There were significant differences in Cd, Cr, Cu, Fe, Mn, Zn, Hg, Mg, and Pb concentrations in tissues of fish and its parasite. The parasite Cd, Cu, and Pb concentration was higher than the other tissues. Furthermore, significant differences were detected in the heavy metal accumulations between the parasitized and unparasitized fish tissues in Cd, Cu, Hg, and Pb concentrations. The Cd, Hg, and Pb concentrations were found in fish muscle at mean concentrations over the permissible limits proposed by the Food and Agriculture Organization.  相似文献

Vibrio species isolated from four different sampling stations in the west coast of Peninsular Malaysia were screened for their antimicrobial resistance and plasmid profiles. A total of 138 isolates belonging to 15 different species were identified. Vibrio campbellii, V. parahaemolyticus, V. harveyi, and V. tubiashii were found to predominance species at all stations. High incidence of erythromycin, ampicillin, and mecillinam resistance was observed among the Vibrio isolates. In contrast, resistance against aztreonam, cefepime, streptomycin, sulfamethoxazole, and sulfonamides was low. All the Vibrio isolates in this study were found to be susceptible to imipenem, norfloxacin, ofloxacin, chloramphenicol, trimethoprim/sulfamethoxazole, and oxytetracycline. Ninety-five percent of the Vibrio isolates were resistant to one or more different classes of antibiotic, and 20 different resistance antibiograms were identified. Thirty-two distinct plasmid profiles with molecular weight ranging from 2.2 to 24.8 kb were detected among the resistance isolates. This study showed that multidrug-resistant Vibrio spp. were common in the aquatic environments of west coast of Peninsular Malaysia.  相似文献

Concentrations of heavy metals (Cu, Zn, Cd, and Pb) in surface water (including total recoverable, dissolved, suspended solids) and in aufwuchs encrusted on Moerella iridescens Benson from seven selected sites and two reference sites in Maluan Bay were investigated in order to understand current metal contamination due to industrialization and urbanization in Xiamen, China. The muscle tissues of the study species (Penceus penicillatus, Scylla serrata Forskal, Harengula zunasi Bleeker, Tillapia nilotica) from a trawling area within Maluan Bay were also analyzed in order to evaluate its safety as seafood. Based on the obtained data, metal concentrations in surface water were compared with Marine Seawater Quality Standards of China and the US EPA acute and chronic criteria, which showed that Maluan Bay may be subjected to different levels of contamination by the metals. Metal concentrations under study in the edible parts (muscle) of the investigated biota species were within the safety permissible levels for human consumption. Through Pearson??s correlation analysis, the relationships between metal concentrations in surface water and in M. iridescens were evaluated. Copper concentrations in M. iridescens were more strongly positively correlated with particulate copper in suspended solids and total recoverable copper in water rather than with dissolved copper at the sampling sites. The data suggested that copper-rich suspended solids contributed substantially to copper accumulation by M. iridescens and played a critical role in the pathway of copper into the food chain. The conclusions of this investigation are likely to be applicable to other relevant scenarios.  相似文献

Morphological mentum deformities which represent sublethal effect of exposure to different types of pollutants were evaluated in Chironomus spp. larvae inhabiting three polluted rivers of Juru River Basin in northwestern peninsular Malaysia. Using mentum deformity incidences, the modified toxic score index (MTSI) was developed based on Lenat??s toxic score index (TSI). The suggested MTSI was compared with TSI in terms of its effectiveness to identify different pollutants including heavy metals. The MTSI showed stronger relationship to total deformity incidence expressed as percentage. Additionally, the multivariate RDA model showed higher capability of MTSI to explain the variations in heavy metal contents of the river sediments. The MTSI was recommended in bioassessment of water and sediment quality using the mentum deformities of Chironomus spp. larvae from aquatic ecosystems receiving anthropogenic, agricultural, or industrial discharges.  相似文献

We report the first study on the occurrence of high-level aminoglycoside-resistant (HLAR) Enterococci in coastal bathing waters and beach sand in Malaysia. None of the encountered isolates were resistant to high levels of gentamicin (500 μg/mL). However, high-level resistance to kanamycin (2,000 μg/mL) was observed in 14.2 % of tested isolates, the highest proportions observed being among beach sand isolates. High-level resistance to kanamycin was higher among Enterococcus faecalis and Enterococcus faecium than Enterococcus spp. Chi-square analysis showed no significant association between responses to tested antibiotics and the species allocation or source of isolation of all tested Enterococci. The species classification of encountered Enterococci resistance to vancomycin was highest among Enterococcus spp. (5.89 %) followed by E. faecium (4.785) and least among E. faecalis. A total of 160 isolates were also tested for virulence characteristics. On the whole, caseinase production was profoundly highest (15.01 %) while the least prevalent virulence characteristic observed among tested beach Enterococci was haemolysis of rabbit blood (3.65 %). A strong association was observed between the source of isolation and responses for each of caseinase (C?=?0.47, V?=?0.53) and slime (C?=?0.50, V?=?0.58) assays. Analysis of obtained spearman’s coefficient showed a strong correlation between caseinase and each of the slime production (p?=?0.04), gelatinase (p?=?0.0035) and haemolytic activity on horse blood (p?=?0.004), respectively. Suggestively, these are the main virulent characteristics of the studied beach Enterococci. Our findings suggest that recreational beaches may contribute to the dissemination of Enterococci with HLAR and virulence characteristics.  相似文献

Manila clams (Ruditapes philippinarum) and sediments were collected bimonthly during 2007 at five locations in Jiaozhou Bay near Qingdao, China, to determine heavy metal concentrations and to assess the validation of R. philippinarum as a metal biomonitor. Concentrations of heavy metals in clam soft tissues ranged between 0.75 and 3.31, 0.89 and 15.20, 5.70 and 26.03, 52.12 and 110.33, 10.30 and 72.34, 9.64 and 28.60, and 3.15 and 52.75 μg g???1 dry weight for Cd, Pb, Cu, Zn, Mn, Cr, and Ni, respectively. Most of the highest values occurred at the northeast bay and the lowest values occurred at the western part. Regarding seasonal variation, relatively high tissue metal concentrations were observed during October or December. A similar pattern was also found in habitat sediments. There was a strong correlation between the concentrations of Cd, Pb, Zn, Mn, Cr, and Ni in soft tissues and surrounding sediments. It is indicated that R. philippinarum could be used as a biomonitor for heavy metal contamination in Jiaozhou Bay.  相似文献

In this study, bioaccumulation of the heavy metals (Fe, Zn, Cd) in the liver, gill, gonad and muscle tissues of Dicentrarchus labrax, Mugil cephalus and Sparus aurata were investigated seasonally. Camlik is the important fisheries lagoon located in The Iskenderun Bay, Mediterranean coast of Turkey. Due to heavy industrial and agricultural activities in the region, the bay has the polluted coastal waters of Turkey. Therefore, the levels of cadmium, zinc, iron were determined in the muscle in each species because of its importance for human consumption and also the liver, gill, gonad were analysed since these organs tend to accumulate metals. Heavy metal levels were found generally higher in the liver and gill than the gonad and muscle tissues in three species; the levels of all metals in a given tissue were generally higher in Mugil cephalus than the other two species. Data collected in this study suggest that Camlik lagoon is affected by a relatively metal pollution. The concentrations of Zn concentrations in the muscle tissues exceeded the acceptable levels for a food source for human consumption and other metals in the edible parts of the investigated fish were in the permissible safety levels for human uses.  相似文献

The efficacy of Common Effluent Treatment Plant (CETP) based on activated sludge process employed for treatment of tannery effluent was analyzed for the efficiency to reduce chromium and other contaminants. Physico-chemical parameters of treated effluent were found within permissible levels of statutory limits. A significant reduction in COD and BOD levels were achieved during the course of treatment in CETP. A reduction of 98.46%, 87.5%, and 96.15% in bacterial counts especially in pathogens like Escherichia coli, Vibrio spp., and Pseudomonas spp. were observed after treatment. Pathogens were not detected in the dried sludge. Complete elimination of fecal streptococci was observed in treated effluent. Around 10.8% of microbial isolates from the effluent showed ability to reduce chromate >90%. In treated effluent chromium level was 5.48 mg/L, which exceeds the statutory limit of Indian standards. Cleaner production options to reduce chromium levels before discharging into water bodies is discussed.  相似文献

The suitability of Mazzaella laminarioides and Sarcothalia crispata as heavy metal biomonitors of Cd, Cu, Hg, Pb, and Zn was assessed by comparing bioaccumulation of these elements in different life stages and frond sizes in samples from three locations, San Vicente Bay (industrial area), Coliumo, and Quidico (the latter as a reference station), where different degrees of heavy metal pollution are recorded. Bioaccumulation and bioconcentration factors of Cd, Cu, Hg, Pb, and Zn were evaluated. The two macroalgae species showed similar patterns, with higher values of Cu, Hg, Pb, and Zn in polluted areas. M. laminarioides bioaccumulated higher concentrations of all metals assessed than S. crispata, independent of life stage and frond size. The results also showed significantly higher Cu, Hg, Pb, and Zn concentrations (p < 0.05) in water samples from San Vicente Bay than those measured in Coliumo and Quidico. Concentrations of Cd, Hg, Pb, and Zn in San Vicente Bay and Cd, Hg, and Pb in Coliumo and Quidico exceed the mean values considered to represent natural concentrations (Cu = 3.00 μg L?1; Zn = 5.00 μg L?1; Pb = 0.03 μg L?1; Cd = 0.05 μg L?1; Hg = 0.05 μg L?1); however, the concentrations recorded do not cause negative effects on the growth and survival of macroalgae. The assessment of heavy metals bioaccumulated in M. laminarioides and S. crispata, particularly Hg, Pb, and Zn, offers a reliable approach for pollution assessment in rocky intertidal environments. Cu and Cd concentrations in seawater samples from San Vicente and Coliumo Bays were significantly higher than in those from Quidico (p value < 0.05); no significant differences in Cd concentrations were observed between San Vicente and Coliumo Bays (p < 0.05). Exceptionally, Cd is bioaccumulated at high levels independent of its availability in the water, thus reaching high concentrations in control areas. High concentrations of metals like Cu and Zn may limit or inhibit Cd uptake in macroalgae, since the transport channels are saturated by some metals, reducing the accumulation of others. These macroalgae species offer good potential for the development of suitable heavy metal pollution survey tools in rocky intertidal environments.  相似文献

We report the first study on the occurrence of antibiotic-resistant enterococci in coastal bathing waters in Malaysia. One hundred and sixty-five enterococci isolates recovered from two popular recreational beaches in Malaysia were speciated and screened for antibiotic resistance to a total of eight antibiotics. Prevalence of Enterococcus faecalis and Enterococcus faecium was highest in both beaches. E. faecalis/E. faecium ratio was 0.384:1 and 0.375:1, respectively, for isolates from Port Dickson (PD) and Bagan Lalang (BL). Analysis of Fisher’s exact test showed that association of prevalence of E. faecalis and E. faecium with considered locations was not statistically significant (p?<?0.05). Chi-square test revealed significant differences (χ 2?=?82.630, df?=?20, p?<?0.001) in the frequency of occurrence of enterococci isolates from the considered sites. Resistance was highest to nalidixic acid (94.84 %) and least for chloramphenicol (8.38 %). One-way ANOVA using Tukey–Kramer multiple comparison test showed that resistance to ampicillin was higher in PD beach isolates than BL isolates and the difference was extremely statistically significant (p?<?0.0001). Frequency of occurrence of multiple antibiotic resistance (MAR) isolates were higher for PD beach water (64.29 %) as compared to BL beach water (13.51 %), while MAR indices ranged between 0.198 and 0.48. The results suggest that samples from Port Dickson may contain MAR bacteria and that this could be due to high-risk faecal contamination from sewage discharge pipes that drain into the sea water.  相似文献
