Symbiotic filamentous bacteria thrive in the intestinal caecum of the deposit-feeding echinoid Echinocardium cordatum. Specimens of E. cordatum were collected at Wimereux (Nord Pas-de-Calais, France) in 1991. Their symbiotic bacteria build nodules by forming multilayered mats around detrital particles that enter the caecum. The morphological features of the bacteria are those of Thiothrix, a sulfide-oxidizing genus. The filaments, which may form rosettes, are sheathed and made by a succession of hundreds of rod-shaped bacteria which store elemental sulfur in the presence of external sulfide. Live bacteria are restricted to the outer layers of the nodules. Their sulfide-oxidizing activity was investigated, using a Biological Oxygen Monitor, by measuring the O2-consumption when reduced sulfur compounds are provided. They oxidize thiosulfate and sulfide. Optimal sulfide oxidation occurs at intermediary pO2 (100 to 160 M O2l-1). Spectrophotometry has confirmed that the sulfur content of the filamentous symbiotic sulfideoxidizing bacteria depends on the presence of external sulfide. This is the first report of symbiotic intradigestive Thiothrixspp.-like bacteria; it lengthens the list of symbioses between sulfide-oxidizing bacteria and invertebrates from sulfide-rich habitats.  相似文献

Sulfide tolerance and detoxification in shallow-water marine fishes   总被引:4,自引:0,他引:4  
Hydrogen sulfide is a potent inhibitor of aerobic respiration. Sulfide is produced in sediments, and many species of fish live in association with the bottom. Tolerance tests, enzyme assays, and chromatography of sulfur compounds in thirteen species of shallow-water marine fishes (collected in San Diego, California, USA in 1987–1988) indicate adaptations to sulfide that vary with habitat and lifestyle. Tidal-marsh inhabitants, like Gillichthys mirabilis and Fundulus parvipinnis, have higher tolerance to sulfide (96 h LC50 at 525 to 700 M) relative to outer-bay and open-coast inhabitants (surviving <12 h at much lower concentrations). The cytochrome c oxidase of all species shows high activity and susceptibility to sulfide poisoning, with 50% inhibition at 30 to 500 nM in various tissues. The two marsh species are able to survive at sulfide concentrations already inhibitory to their cytochrome c oxidase and fatal to other species. All species detoxify sulfide by oxidizing it to thiosulfate. All have sulfide-oxidizing activity in the blood, spleen, kidney, liver and gills, which correlates significantly with heme content. Thiosulfate appears in the tissues of sulfide-exposed fish and builds up to high concentrations (up to 2 mM) with stronger and longer exposure. Unexposed fish contain little or no thiosulfate. Sulfide is barely detectable in the tissues, even in high-sulfide exposure tests. We suggest that fish blood, in having high sulfide-oxidizing activity and no cytochrome c oxidase, can act as a short-term first line of defense against sulfide, and thus minimize the amount that reaches the vital organs. The results of this study indicate that sulfide is a significant environmental factor influencing the ecological distribution of marine fishes.  相似文献

The lugwormArenicola marina L. oxidizes entering sulfide to thiosulfate. After 8 h of normoxic incubations with sulfide concentrations of 0.2 to 1.0 mmoll-1 thiosulfate in the coelomic fluid amounted up to about 4 mmoll-1 whereas sulfite concentrations were 100-fold lower and no accumulation of sulfate in the coelomic fluid was found. The sulfide oxidation was highly oxygen dependent. An increase of oxygen partial pressure ( ) in the medium was followed by enhanced thiosulfate production and by a decrease of sulfide concentration in the coelomic fluid. Under normoxia, the sulfide oxidation rate was sufficient to compensate the influx of sulfide into the coelomic fluid when the sulfide concentration in the medium was below 0.33 mmoll-1. When external sulfide was raised beyond this level, sulfide up to 5 moll-1 in the coelomic fluid appeared. Succinate in the body wall tissue was low as long as no sulfide appeared in the coelomic fluid, indicating the maintenance of an aerobic metabolism. The oxidation of sulfide to thiosulfate was localized in the mitochondria of the body wall tissue. The oxygen consumption of mitochondria was stimulated by the addition of sulfide. The mitochondrial sulfide oxidation rate depended on the amount of mitochondrial protein and followed a Michaelis-Menten kinetic. An apparentK m of 0.68±0.29 moll-1 and aV max of 41.9±22.3 nmol min-1 mg-1 protein was calculated. Sulfide was stoichiometrically oxidized to thiosulfate with 1 mol sulfide consuming 1 mol oxygen. Sulfide oxidation was not inhibited by sulfide concentrations as high as 100 moll-1. At low concentrations of cyanide or azide, when respiration without sulfide was already inhibited, sulfide oxidation could still be stimulated, tentatively indicating the existence of an alternative terminal oxidase. Vestimentiferan tubeworms, which rely on intracellular sulfide-oxidizing autotrophic bacteria for organic carbon, flourish at deep-sea hydrothermal vents despite the erratic nature of their habitat. To assess the degree to which differences in habitat chemistry (sulfide, pH/CO2) might impact host and symbiont metabolic activity, Riftia pachyptila tubeworms were collected from habitats with low (H2S < 0.0001 mM) and high (up to 0.7 mM) sulfide concentrations. The elemental sulfur content of the symbiont-containing trophosome organ was lower in specimens collected from the low-sulfide site. Symbiont abundance, RubisCO activity, and trophosome carbon fixation rates were not significantly different for individuals collected from low- versus high-sulfide habitats. Carbonic anhydrase activities were higher in the anterior gas exchange organs of R. pachyptila from the low-sulfide habitat. Despite large differences in habitat chemistry, symbiont abundance and autotrophic potential were consistent, while the host appears to tailor carbonic anhydrase activity to environmental CO2 availability.  相似文献

The hydrothermal vent crab Bythograea thermydron is exposed to high environmental concentrations of sulfide and low levels of oxygen for extended periods of time. It has previously been shown that hydrogen sulfide is oxidized to the relatively non-toxic thiosulfate (S2O 3 2– ), which accumulates in the hemolymph. Hemolymph thiosulfate levels in freshly captured crabs vary significantly among crabs from different hydrothermal vent sites as well as between crabs from different microhabitats within the same site. Hemolymph thiosulfate concentrations were not significantly different between crabs captured at the same site 6 mo apart. Hemolymph thiosulfate concentrations ranged from 66 mol 1–1 in a crab captured at a site with relatively low sulfide venting, to 3206 mol 1–1 in an individual that was netted from an active smoker vent with much higher sulfide exposure. The differences in hemolymph thiosulfate between sites and the stability of hemolymph thiosulfate in crabs captured at the same site at different times suggest that sulfide exposure is significantly different between sites and that this exposure may not vary significantly over the course of a few months. B. thermydron experimentally exposed to sulfide had high levels of thiosulfate in their hemolymph and increased abilities to regulate oxygen consumption in conditions of low oxygen. This enhancement of regulatory abilities suggests that the previously demonstrated increased hemocyaninoxygen (Hc–O2) affinity due to elevated thiosulfate may be adaptive in vivo. Average oxygen-consumption rates were much higher in crabs experimentally exposed to sulfide than in unexposed crabs. Crabs injected with isosmotic thiosulfate did not have increased oxygen-consumption rates as did the sulfide-exposed individuals, but did show a similar reduction in P c (the critical partial pressure of oxygen at which crabs can no longer regulate oxygen consumption). This suggests that it is the sulfide exposure and/or detoxification rather than the elimination of thiosulfate that causes the increase in metabolic rate. Thiosulfate diffuses into dead crabs and into live crabs exposed to 15 mmol S2O 3 2- l–1, indicating substantial permeability, and yet live crabs are able to eliminate thiosulfate when incubated in sea water containing 1.5 mmol S2O 3 2- l–1, suggesting a process that has an active component.  相似文献

Riftia pachyptila Jones, 1981, lives in association with a chemoautotrophic, sulfide-oxidizing -Proteobacterium that occurs in a specialized organ, the trophosome. Ultrastructurally different bacterial subpopulations occur in different regions of the trophosome lobules (central rods, median small cocci, peripheral large cocci) and contain vesicles, which have been proposed to be sites of elemental sulfur storage. Differently colored trophosomes have been suggested to reflect different amounts of elemental sulfur in the tissue. In this study, the presence of elemental sulfur (S0) was confirmed in the vesicles of the symbionts of R. pachyptila by electron energy loss spectrography (EELS). The proportion of (two-dimensional) area occupied by sulfur vesicles in the cells was found to be strongly correlated with trophosome color, both in individuals with uniformly colored trophosomes and individuals that exhibited a gradual color change along the length of their trophosomes. Elemental sulfur content was highly variable between individuals from a single collection, suggesting a high degree of microhabitat heterogeneity within vestimentiferan aggregations.Communicated by O. Experiments were conducted in order to specify the reductants responsible for the carbon dioxide fixation of the symbiotic sulfur bacteria in the gutless marine oligochaete Phallodrilus leukodermatus (Annelida) from shallow calcareous sediments in Bermuda. Carbon dioxide-uptake rates were suppressed by S= and stimulated by S2O3 =. Individuals which hosted bacteria containing reserve energy substances maintained a high short-term CO2-uptake activity, while bacteria in worm homogenates and in worms treated with an antibiotic (Baypen) did not show any significant metabolic activity. Absolute uptake rates in P. leukodermatus were usually considerably higher than those reported for other animals harbouring prokaryotic sulfuroxidizing symbionts. Utilization of thiosulfate rather than sulfide is compatible with the preferred occurrence of the worms around the redox discontinuity layer and has been confirmed in other thiobiotic animals. Sulfur stored in the symbiotic bacteria appears to be oxidized to sulfate and be excreted when the worms are held under energy-limited conditions. The data emphasize the complexity of the possible metabolic pathways involved in the oxidation of reduced-sulfur compounds by bacterial symbionts in marine invertebrates.  相似文献

We examined the growth rate (µ) ofUlva lactuca L. (collected from Roskilde Fjord, Denmark in 1987) at different levels of dissolved inorganic carbon (DIC), pH and oxygen in two culture facilities. Growth was faster in Facility A (µ max ca 0.3 d–1) than in B (µ max ca 0.2 d–1), probably because of more efficient stirring and higher light intensity. The growth-DIC response curve exhibited low half-saturation constant (K 1/2) values (0.35 mM DIC in A, 0.55 mM in B) and growth rates close toµ max at natural seawater concentration of 2 mM DIC. Growth rate showed a low sensitivity to oxygen over a wide range of DIC and oxygen concentrations. Collectively, the results demonstrated an efficient mechanism for DIC use, unaffected by acclimatization to DIC concentrations between 0.2 and 3 mM. The growth rate decreased little between pH 7.5 and 9 at 2 mM DIC, but steeply above pH 9 approaching zero just above pH 10. The decline of growth at high pH may result from direct pH effects on cell pH, reduced HCO 3 - availability and impaired operation of the carbon uptake process. The growth responses ofU. lactuca to DIC, pH and oxygen resembled those observed in previous short-term photosynthetic experiments. This similarity is probably due to the fast growth ofU. lactuca which means that photosynthetic products are rapidly converted into cell growth. Based on the culture experiments we argue that field plants ofU. lactuca not exposed to stagnant water and DIC depletion are likely to be limited in growth by environmental factors other than DIC (e.g. light and nutrients). Dense mats ofU. lactuca, however, may show reduced growth as a result of DIC depletion, high pH and self-shading.  相似文献

Sulfur content and fine structure were studied for tissues of three species of clams, Lucinoma annulata, Calyptogena elongata and Lucina floridana, which inhabit sulfide-rich environments and whose gills harbor symbiotic sulfur bacteria. Lucinoma annulata and C. elongata were dredged from the Santa Barbara basin, California, USA, at a depth of 480 to 490 m, and Lucina floridana were dug from below the roots of seagrasses in Saint Joseph Bay, Florida, at a depth of 0.25 to 2m. Foot tissue of Lucinoma annulata, without symbionts, had a total sulfur content of 1.4±0.1 (SD) mg 100 mg-1 dry weight of tissue (%DW). The symbiont-containing gill tissue of different individuals of L. annulata varied in color from dark red to pale yellow, and the total sulfur content was 2.5±0.4% DW in red gills and was 5.6±3.3 % DW in the yellowest gills. Maintenance of L. annulata in the laboratory for 21 d in the absence of sulfide resulted in the loss from the gill of yellow deposits which were elemental sulfur in the form of liquid-crystalline sulfur globules rather than solid orthorhombic sulfur crystals. The foot tissue did not contain elemental sulfur. When examined by freeze-etch microscopy, sulfur globules were found only within bacteria and not in the animal host cytoplasm. Sulfur globules were confined to the periplasmic space of the bacteria. C. elongata and Lucina floridana resembled Lucinoma annulata in the physical form and distribution of elemental sulfur. The absence of elemental sulfur in the animal cytoplasm suggests that its formation from sulfide is not a detoxification scheme to protect animal tissue from sulfide toxicity. The sulfur deposits probably represent inorganic energy reserves that permit the symbiotic bacteria to function even during the temporary absence of external sulfide.  相似文献

Specimens of the hydrothermal vent pogonophoran Riftia pachyptila Jones were collected by submersible at a depth of 2 600 m at the 21°N hydrothermal vent site on the East Pacific Rise (20°50N, 109°06W) in April and May of 1982. The worms were maintained in pressurized aquaria for up to 45 d for metabolic studies. Consumption of O2 was regulated down to low PO 2 (oxygen partial pressure) values; O2 consumption rates were 0.63 and 1.12 mol g-1 wet wt h-1 at 2.5° and 8°C, respectively; such rates were comparable to those previously measured for other pogonophorans. Intact specimens of R. pachyptila (including bacterial symbionts) did not consume significant amounts of CH4 from the environment. The respiratory quotients, in the absence of added sulfide, indicated that metabolism was mainly heterotrophic. High rates of uptake of dissolved amino acids were recorded for one specimen. The total [CO2] in the vascular blood and the Hb-containing coelomic fluid were high. Under anaerobic conditions, there were equilibrium distributions of pH, total [CO2] and sulfide concentrations between the vascular blood and the coelomic fluid, apparently because these metabolites were readily exchanged between the two compartments. The vascular blood bound neither CH4 nor H2. However, sulfide was reversibly bound by both the vascular blood and coelomic fluid; because this binding depended strongly on pH (with a maximum at about 7.5), HS- was probably the molecular species bound. Under anaerobic, but not aerobic conditions, the trophosome bound substantial amount of sulfide; thus, the high concentrations of sulfide in the trophosome may have resulted mainly from sulfide bound to sulfide oxidases under anaerobic conditions. The occurrence of sulfide and thiosulfate-oxidizing bacteria was examined in waters and sediments of the Black Sea and the Caraco Trench. Isolates obtained by enrichment-culture techniques exhibited facultative autotrophic-growth characteristics and oxidized thiosulfate or sulfide. Acid was not formed during growth in mineral medium. Most of the isolates were facultatively anaerobic, using nitrate as the terminal hydrogen acceptor. Strictly aerobic bacteria were found only in oxygenated waters. Comparison of the metabolic capabilities of the isolates during growth in sulfide and thiosulfate media with chemical parameters measured in the Black Sea and Cariaco Trench waters suggests that the biological oxidation of thiosulfate is the rate-limiting step in the oxidative turnover of sulfur compounds in the basins. Evidence for the oxidation of sulfide in the anoxic waters is presented, and possible explanations for the occurrence of this process in the absence of suitable electron acceptors (O2. NO3) are discussed. A maximum dark assimilation of carbon dioxide was located just below the oxygen-sulfide interface in the Cariaco Trench. Cells ofPhaeodactylum tricornutum Bohlin develop the ability to take up L-lysine when they are deprived of nitrogen (illuminated in nitrogen-free medium), carbon (incubated in darkness) or both. Cells with a developed uptake system take up and accumulate lysine in an unchanged form. Uptake occurs under either aerobic or anaerobic conditions and is dependent on the presence of sodium+ ions (K s Na +=,ca. 10 mM). Some potassium+ ions are necessary for uptake, presumably within the cells, but with potassium+-replete cells, increasing K+ concentration depresses lysine uptake. The lysine-uptake porter also transports L-arginine.K s values are about 1.5 M for lysine and 0.5 M for arginine. It is, however, possible that the uptake system developed by incubating cells in darkness differs from that produced in light; it shows a pronounced pH optimum at pH 8.5, whereas the activity of the light-developed system declines from pH 6.5 to pH 9.0 and correlates well with the concentration of lysine+. The uptake system developed in darkness may also have a higher affinity for lysine. Lysine uptake is not inhibited by 1 mM concentrations of nitrate, nitrate, ammonium, or urea nor by similar concentrations of amphoteric or acidic amino acids.  相似文献

A number of new vestimentiferan species occur at northeast Pacific hydrothermal vent sites. The trophosome and bacterial symbionts of three species, collected from the Juan de Fuca and Explorer Ridges between 1984 and 1986, were studied by transmission electron microscopy (TEM). As in Riftia pachyptila, trophosome tissue is organised into lobules each having an axial blood vessel, and intracellular bacterial symbionts are contained in membrane vacuoles. The bacteria have many cytoplasmic inclusions including tubular membrane systems, glycogen-like particles and poly--hydroxybutyrate (PHB) or sulfur bodies. Glycogen production may be quantitatively important to both the symbionts and the host. Glycogen-like granules appear to first accumulate in the bacterial cells and then be released into the bacteriocyte cytoplasm as bacteria are degraded. Although various stages of bacterial growth and degradation are observed, data are insufficient to verify any across-lobule progression of these processes. Morphological comparison of the symbionts reveals that similar symbionts are found in different vestimentiferan species and that one to two bacterial types exist within single individuals. Rates of sulfate reduction were measured during the spring dry and summer wet seasons of 1992 in shallow coastal sediments of the central Great Barrier Reef lagoon. In the dry season, sulfate reduction rates, integrated to 18–20 cm depth, ranged from 23.8 to 30.8 mmol S m-2 d-1. In the wet season, heavy monsoonal rains and wind-induced waves caused severe disturbance of sediments leading to less anoxic conditions, and to a 2- to 3-fold decline in rates of sulfate reduction (10.2 to 12.8 mmol S m-2 d-1). The percentage of reduced 35S recovered as acid-volatile sulfide (% AVSred) ranged from 15.8 to 44.9% in spring, and was significantly reduced at each station in summer (range 14.5 to 31.4%). An analysis of variance indicated that seasonality accounted for 31.5% of the total variance in total rates of sulfate reduction, with only 7.5% of the variance accounted for by depth into the sediment; there were no station differences. In both seasons, there were no clear patterns in the proportion of radiolabel incorporated into the AVS and chromium-reducible sulfur (CRS) pools with depth into the sediment. These results are contrary to the seasonal patterns and pathways of sulfate reduction in temperate coastal sediments.  相似文献

Total sulfur (STOT), elemental sulfur (S°) and sulfur-isotope compositions (δ34S) of marine animals were analyzed to determine whether these chemical characteristics could help distinguish animals with a sulfur-based, thiotrophic nutrition from animals whose nutrition is based on methanotrophy or on more normal consumption of phytoplankton-derived organic matter. The presence of S° was almost entirely confined to the symbiont-containing tissues of thiotrophs, but was sometimes undetectable in thiotrophic species where sulfide availability was probably low. When S° contents were subtracted, the remaining tissue-sulfur concentrations were similar for all nutritional groups. δ34S values were typically lower for thiotrophs than for other groups, although there was overlap in methanotroph and thiotroph values at some sites. Field evidence supported the existence of small to moderate (1 to 10‰)34S fractionations in the uptake of sulfides and metabolism of thiosulfate. In general, a total sulfur content of >3% dry weight, the presence of elemental sulfur, and δ34S values less than +5‰ can be used to infer a thiotrophic mode of nutrition. Certain hydrothermal vent invertebrates, e.g. Riftia pachyptila and Calyptogena magnifica, are clearly established as harboring dense populations of chemoautotrophic sulfur bacteria in specialized tissues. By contrast, the physiological characteristics of the abundant intracellular gill symbiont of the vent mussel Bathymodiolus thermophilus have been questioned. The low activities of enzymes diagnostic for CO2 fixation (Calvin cycle) and for sulfur-driven energy generation, as measured by other investigators, have been attributed to bacterial contamination of the gill surface. Based on research at the Galápagos Rift hydrothermal vents in 1988 and subsequent laboratory experiments, the current study confirms that the B. thermophilus symbiont is a psychrophile for which thiosulfate and sulfide stimulate CO2 fixation. It strongly indicates that the symbiont is a chemoautotroph by establishing the following: (1) Sulfide and thiosulfate can stimulate CO2 fixation by partially purified symbionts by up to 43-fold and 120-fold, respectively; (2) the ribulose-1,5-bisphosphate carboxylase/oxygenase activity of the symbiont is sufficient to account for its sulfide- or thiosulfate-stimulated CO2 incorporation; (3) the symbiont's molar growth yield on thiosulfate, as judged by CO2 incorporation, is indistinguishable from that of free-living chemoautotrophs. Due to the high protein-degrading activity of B. thermophilus gill lysate, it is also suggested that host lysis of symbionts plays a more important role in the nutrition of the vent mussel than in R. pachyptila or C. magnifica, for which no comparable protein-degrading activity was found.  相似文献

Coastal sediments receiving different amounts of organic carbon through sedimentation were investigated with respect to sulfate reduction and methanogenic activity. Sampling was carried out at sediment temperatures of 7° and 15°C. Sulfate-reducing and methanogenic bacteria were found at all depths. Sulfate reduction decreased with depth and the highest sulfide concentrations were found a few centimeters below the sediment surface, up to 15 mM at 15°C and pH 7.1. In the same segments a maximum in the methane concentration was also found, 0.91 mM. The high sulfide concentration inhibited the methane formation from acetate but not from carbon dioxide. In the organic rich sediment sulfate reduction was limited by the diffusion of SO 4 + into the sediment and methane production from acetate by sulfide diffusion out of the sediment. When electron acceptor concentration limits sulfate reduction, thermodynamic calculations show that the utilization of electron donors more reduced than acetate is favored. In the sediment with the high carbon-input, acetate predominated at 15°C whereas in the low carbon-input sediment hardly any short chain organic acids were detected. The possibility of a shift in sulfate reduction from acetate oxidation to acetate production is discussed.  相似文献

Photosynthetic properties of the common red algaGracilaria conferta, collected from the eastern Mediterranean Sea were investigated in 1989, in order to begin evaluating its adaptative strategies with regard to the inorganic carbon composition of seawater, and to test whether the alleged C4 photosynthesis of anotherGracilaria species is common within the genus. Net photosynthetic rates ofG. conferta were, under ambient conditions of inorganic carbon (ca. 10µM, CO2 and 2.2 mM HCO 3 - ), not sensitive to O2 over the range 10 to 300µM, and the CO2 compensation point was low (ca. 0.005µM). Ribulose-1,5-bisphosphate carboxylase/oxygenase was the major carboxylating enzyme, with a crude extract activity of 175µmol CO2 g–1 fresh wt h–1 while phosphoenolpyruvate carboxylase and phosphoenolpyruvate carboxykinase were present at 70 and 20%, respectively, of that activity. No activities of the decarboxylases NAD-and NADP-malic enzyme could be detected. The14C pulse-chase incorporation pattern showed thatG. conferta fixes inorganic carbon via the photosynthetic carbon reduction cycle only, with no evidence for photosynthetic C4 acid metabolism. Photosynthesis at the natural seawater pH of 8.2 was, at 25°C and saturating light, saturated at the ambient inorganic carbon concentration of 2.5 mM. It is proposed that, under ambient inorganic carbon conditions, a CO2 concentrating system other than C4 metabolism provides an internal CO2 concentration sufficient to suppress the O2 effect on ribulose-1,5-bisphosphate carboxylase/oxygenase and, thus, on photorespiration, in a medium where the external free CO2 concentration is lower than theK m(CO2) of the carboxylating enzyme. Since inorganic carbon, under natural saturating light conditions, seems not to be a limiting factor for photosynthesis ofG. conferta, it likely follows that other nutrients limit the growth of this alga in nature.  相似文献

Photosynthetic sulfur bacteria oxidize the sulfide produced by dissimilatory sulfate-reducing bacteria, thus preventing the occurrence of pollution by these compounds in the oxygenic zone of aquatic systems. Zinc is one of the most abundant heavy metals. Its toxic effects have been documented in various organisms that can photosynthesize, but a lack of information prevails in this respect, about photosynthetic sulfur bacteria. The aim of the present study was to evaluate the toxic effects of zinc on green sulfur bacteria (Chlorobiaceae) and purple sulfur bacteria (Chromatiaceae) in a microcosm as well to explore the possible relationships between the bacterial population growths with the physicochemical properties of the metalimnion of Zimapan Reservoir (Mexico) during a 1-year period. Hydrosoluble organic carbon (HOC) and light availability in the metalimnion were the limiting factors for growth in these bacteria. Higher HOC levels occurred in Spring, at the end of the wet season and in Winter. Current zinc levels in the reservoir seem to elicit toxic effects. At lethal zinc concentrations, the best concentration-response relationship was shown. The biomarker for purple sulfur bacteria was the bacteriochlorophyll content, while total reducing sugar was the best biomarker for green sulfur bacteria. At sublethal concentrations, zinc alters adenosine triphosphate levels, total reducing sugar, total protein content, and bacteriochlorophyll content. The observed responses indicate that green sulfur bacteria are more sensitive than purple sulfur bacteria and are suitable target organisms for use in establishing the accepted threshold concentration of zinc (NOEC 0.056–0.097 mg L?1 of Zn) in anoxic deep water.  相似文献

为明确NH_4~+、 NO_3~-、SO_4~(2-)及金属等组分在水溶性提取液对发光细菌的光抑制过程中所起的作用,参照PM_(2.5)样品提取液浓度,模拟配制与3级以上PM_(2.5)样品提取液中主要组分:硫酸盐、硝酸盐、氨盐相同浓度的溶液,同时选取与PM_(2.5)可溶性提取液发光抑制率相关性较强的铅、锌,配制不同浓度级别模拟溶液,测试各单一组分对发光细菌的发光抑制率及其混合溶液对发光细菌的联合影响效应。基于毒性单位法(TU)、相加指数法(AI)和混合毒性指数法(MTI)评价了混合体系联合影响的作用类型。结果表明,与3~6级PM_(2.5)可溶性提取液中硫酸氨、硫酸氢氨、硝酸氨、硫酸锌和硝酸铅浓度相同的模拟溶液对发光细菌的发光没有抑制作用。不同的评价方法对PM_(2.5)主要组分混合体系联合效应评价结果具有较好的一致性,硫酸氨、硝酸氨、硫酸氢氨混合溶液中,对发光细菌的光抑制均为硫酸氢氨的独立作用,硫酸锌与硝酸铅的混合体系,锌和铅对发光细菌的联合影响效应表现为协同,硫酸氨、硝酸氨、硫酸氢氨与硫酸锌、硝酸铅的多元混合体系呈现协同作用。  相似文献
