Increasing evidences show that inhalation of indoor bioaerosols has caused numerous adverse health effects and diseases. However, the bioaerosol size distribution, composition, and concentration level, representing different inhalation risks, could vary with different living environments. The six-stage Andersen sampler is designed to simulate the sampling of different human lung regions. Here, the sampler was used in investigating the bioaerosol exposure in six different environments (student dorm, hospital, laboratory, hotel room, dining hall, and outdoor environment) in Beijing. During the sampling, the Andersen sampler was operated for 30 min for each sample, and three independent experiments were performed for each of the environments. The air samples collected onto each of the six stages of the sampler were incubated on agar plates directly at 26 °C, and the colony forming units (CFU) were manually counted and statistically corrected. In addition, the developed CFUs were washed off the agar plates and subjected to polymerase chain reaction (PCR)-denaturing gradient gel electrophoresis (DGGE) for diversity analysis. Results revealed that for most environments investigated, the culturable bacterial aerosol concentrations were higher than those of culturable fungal aerosols. The culturable bacterial and fungal aerosol fractions, concentration, size distribution, and diversity were shown to vary significantly with the sampling environments. PCR-DGGE analysis indicated that different environments had different culturable bacterial aerosol compositions as revealed by distinct gel band patterns. For most environments tested, larger (>3 μm) culturable bacterial aerosols with a skewed size distribution were shown to prevail, accounting for more than 60 %, while for culturable fungal aerosols with a normal size distribution, those 2.1–4.7 μm dominated, accounting for 20–40 %. Alternaria, Cladosporium, Chaetomium, and Aspergillus were found abundant in most environments studied here. Viable microbial load per unit of particulate matter was also shown to vary significantly with the sampling environments. The results from this study suggested that different environments even with similar levels of total microbial cuturable aerosol concentrations could present different inhalation risks due to different bioaerosol particle size distribution and composition. This work fills literature gaps regarding bioaerosol size and composition-based exposure risks in different human dwellings in contrast to a vast body of total bioaerosol levels.  相似文献

Background culturable bacteria aerosols were collected and identified in two large public buildings located in Minneapolis, Minnesota and Seattle, Washington over a period of 5 months and 3 months, respectively. The installed particulate air filters in the ventilation systems were used as the aerosol sampling devices at each location. Both pre and final filters were collected from four air handing units at each site to determine the influence of location within the building, time of year, geographical location and difference between indoor and outdoor air. Sections of each loaded filter were eluted with 10 ml of phosphate buffered saline (PBS). The resulting solutions were cultured on blood agar plates and incubated for 24 h at 36 degrees C. Various types of growth media were then used for subculturing, followed by categorization using a BioLog MicroStation (Biolog, Hayward, CA, USA) and manual observation. Environmental parameters were gathered near each filter by the embedded on-site environmental monitoring systems to determine the effect of temperature, humidity and air flow. Thirty nine different species of bacteria were identified, 17 found only in Minneapolis and 5 only in Seattle. The hardy spore-forming genus Bacillus was the most commonly identified and showed the highest concentrations. A significant decrease in the number of species and their concentration occurred in the Minneapolis air handling unit supplying 100% outdoor air in winter, however no significant correlations between bacteria concentration and environmental parameters were found.  相似文献

Bioaerosol sampling by a personal rotating cup sampler CIP 10-M   总被引:1,自引:0,他引:1
High concentrations of bioaerosols containing bacterial, fungal and biotoxinic matter are encountered in many workplaces, e.g. solid waste treatment plants, waste water treatment plants and sewage networks. A personal bioaerosol sampler, the CIP 10-M (M-microbiologic), has been developed to measure worker exposure to airborne biological agents. This sampler is battery operated; it is light and easy to wear and offers full work shift autonomy. It can sample much higher concentrations than biological impactors and limits the mechanical stress on the microorganisms. Biological particles are collected in 2 ml of liquid medium inside a rotating cup fitted with radial vanes to maintain an air flow rate of 10 l min(-1) at a rotational speed of approximately 7,000 rpm. The rotating cup is made of sterilisable material. The sampled particles follow a helicoidal trajectory as they are pushed to the surface of the liquid by centrifugal force, which creates a thin vertical liquid layer. Sterile water or another collecting liquid can be used. Three particle size selectors allow health-related aerosol fractions to be sampled according to international conventions. The sampled microbiological particles can be easily recovered for counting, incubation or further biochemical analysis, e.g., for airborne endotoxins. Its physical sampling efficiency was laboratory tested and field trials were carried out in industrial waste management conditions. The results indicate satisfactory collection efficiency, whilst experimental application has demonstrated the usefulness of the CIP 10-M personal sampler for individual bioaerosol exposure monitoring.  相似文献

The objective of this study was to determine filter materials and extraction methods that are appropriate to use for environmental sampling of B. anthracis. Four types of filters were tested: mixed cellulose ester (MCE) with a pore size of 3 microm, polytetrafluoroethylene (PTFE) with pore sizes of 1 and 3 microm, and gelatin with a pore size of 3 microm. Bacillus subtilis var. niger endospores (also known as Bacillus globigii[BG]) were used as a surrogate for B. anthracis. Endospores were collected into Button Inhalable Aerosol Samplers with sampling times of 15 minutes, 1 hour, and 4 hours. Physical collection efficiency was determined by measuring upstream and downstream B. subtilis concentrations with an optical particle counter. Vortexing with ultrasonic agitation and vortexing with shaker agitation extraction methods were evaluated. The MCE, 1 microm PTFE, and gelatin filters provided physical collection efficiencies of 94% or greater. The 3 microm PTFE filter showed inconsistent physical efficiency characteristics between filters. Epifluorescence microscopic analysis of the gelatin filter extraction fluid revealed the presence of contamination by non-culturable bacteria. Mean differences for microbial culturability were not statistically significant for filter materials and extraction methods. However, the vortexing with shaker agitation extraction method resulted in higher total microbial counts in the extraction fluids for MCE and 1 microm PTFE filters when compared to vortexing with ultrasonic agitation. In summary, the MCE and 1 microm PTFE filters in combination with vortexing and shaker extraction demonstrated the best performance for the filter collection and extraction of BG spores.  相似文献

A unique two-stage cyclone bioaerosol sampler has been developed at NIOSH that can separate aerosols into three size fractions. The ability of this sampler to collect infectious airborne viruses from a calm-air chamber loaded with influenza A virus was tested. The sampler's efficiency at collecting aerosolized viral particles from a calm-air chamber is essentially the same as that from the high performance SKC BioSampler that collects un-fractionated particles directly into a liquid media (2.4 × 10(4) total viral particles per liter of sampled air (TVP/L) versus 2.6 × 10(4) TVP/L, respectively, after 15 min) and the efficiency is relatively constant over collection times of 15, 30 and 60 min. Approximately 34% of the aerosolized infectious virus collected after 15 min with the NIOSH bioaerosol sampler remained infectious, and infectious virus was found in all three size fractions. After 60 min of sampling, the infectious virus/liter air found in the NIOSH bioaerosol sampler was 15% of that found in the SKC BioSampler. This preservation of infectivity by the NIOSH bioaerosol sampler was maintained even when the initial infectivity prior to aerosolization was as low as 0.06%. The utility of the NIOSH bioaerosol sampler was further extended by incorporating an enhanced infectivity detection methodology developed in our laboratory, the viral replication assay, which amplified the infectious virus making it more readily detectable.  相似文献

Airborne transmission of bacterial pathogens from point sources (e.g., ranches, dairy waste treatment facilities) to areas of food production (farms) has been suspected. Determining the incidence, transport and viability of extremely low levels of pathogens require collection of high volumes of air and characterization of live bacteria from aerosols. We monitored the numbers of culturable bacteria in urban aerosols on 21 separate days during a 9 month period using high volume cyclonic samplers at an elevation of 6 m above ground level. Culturable bacteria in aerosols fluctuated from 3 CFU to 6 million CFU/L of air per hour and correlated significantly with changes in seasonal temperatures, but not with humidity or wind speed. Concentrations of viable bacteria determined by fluorescence staining and flow cytometry correlated significantly with culturable bacteria. Members of the phylum Proteobacteria constituted 98% of the bacterial community, which was characterized using 16S rRNA gene sequencing using DNA from aerosols. Aquabacterium sp., previously characterized from aquatic environments, represented 63% of all clones and the second most common were Burkholderia sp; these are ubiquitous in nature and some are potential human pathogens. Whole genome amplification prior to sequencing resulted in a substantial decrease in species diversity compared to characterizing culturable bacteria sorted by flow cytometry based on scatter signals. Although 27 isolated colonies were characterized, we were able to culture 38% of bacteria characterized by sequencing. The whole genome amplification method amplified DNA preferentially from Phyllobacterium myrsinacearum, a minor member of the bacterial communities, whereas Variovorax paradoxus dominated the cultured organisms.  相似文献

Personal aerosol samplers are widely used to monitor human exposure to airborne materials. For bioaerosols, interest is growing in analyzing samples using molecular and immunological techniques. This paper presents a personal sampler that uses a two-stage cyclone to collect bioaerosols into disposable 1.5 ml Eppendorf-type microcentrifuge tubes. Samples can be processed in the tubes for polymerase chain reaction (PCR) or immunoassays, and the use of multiple stages fractionates aerosol particles by aerodynamic diameter. The sampler was tested using fluorescent microspheres and aerosolized fungal spores. The sampler had first and second stage cut-off diameters of 2.6 microm and 1.6 microm at 2 l min(-1)(geometric standard deviation, GSD = 1.45 and 1.75), and 1.8 microm and 1 microm at 3.5 l min(-1)(GSD = 1.42 and 1.55). The sampler aspiration efficiency was >or=98% at both flow rates for particles with aerodynamic diameters of 3.1 microm or less. For 6.2 microm particles, the aspiration efficiency was 89% at 2 l min(-1) and 96% at 3.5 l min(-1). At 3.5 l min(-1), the sampler collected 92% of aerosolized Aspergillus versicolor and Penicillium chrysogenum spores inside the two microcentrifuge tubes, with less than 0.4% of the spores collecting on the back-up filter. The design and techniques given here are suitable for personal bioaerosol sampling, and could also be adapted to design larger aerosol samplers for longer-term atmospheric and indoor air quality sampling.  相似文献

The aim of the work presented here is to study the effectiveness of building air handling units (AHUs) in serving as high volume sampling devices for airborne bacteria and viruses. An HVAC test facility constructed according to ASHRAE Standard 52.2-1999 was used for the controlled loading of HVAC filter media with aerosolized bacteria and virus. Nonpathogenic Bacillus subtilis var. niger was chosen as a surrogate for Bacillus anthracis. Three animal viruses; transmissible gastroenteritis virus (TGEV), avian pneumovirus (APV), and fowlpox virus were chosen as surrogates for three human viruses; SARS coronavirus, respiratory syncytial virus, and smallpox virus; respectively. These bacteria and viruses were nebulized in separate tests and injected into the test duct of the test facility upstream of a MERV 14 filter. SKC Biosamplers upstream and downstream of the test filter served as reference samplers. The collection efficiency of the filter media was calculated to be 96.5 +/- 1.5% for B. subtilis, however no collection efficiency was measured for the viruses as no live virus was ever recovered from the downstream samplers. Filter samples were cut from the test filter and eluted by hand-shaking. An extraction efficiency of 105 +/- 19% was calculated for B. subtilis. The viruses were extracted at much lower efficiencies (0.7-20%). Our results indicate that the airborne concentration of spore-forming bacteria in building AHUs may be determined by analyzing the material collected on HVAC filter media, however culture-based analytical techniques are impractical for virus recovery. Molecular-based identification techniques such as PCR could be used.  相似文献

While several methods are available for bioaerosol monitoring, impaction remains the most common one, particularly for collecting fungal spores. Earlier studies have shown that the collection efficiency of many conventional single-stage bioaerosol impactors falls below 50% for spores with an aerodynamic diameter between 1.7 and 2.5 microm because their cut-off size is 2.5 microm or greater. The cut-off size reduction is primarily done by substantially increasing the sampling flow rate or decreasing the impaction jet size, W, to a fraction of a millimetre, with both measures often impractical to implement. Some success has recently been reported on the utilization of an ultra-low jet-to-plate distance, S (S/W < 0.1), in circular impactors. This paper describes a laboratory evaluation and some field testing of two single-stage, single-nozzle, slit bioaerosol impactors, Allergenco-D and Air-O-Cell, which feature the same jet dimensions and flow rate but have some design configuration differences that were initially thought to be of low significance. The collection efficiency and the spore deposit characteristics were determined in the laboratory using real-time aerosol spectrometry and different microscopic enumeration methods as the test impactors were challenged with the non-biological polydisperse NaCl aerosol and the aerosolized fungal spores of Cladosporium cladosporioides, Aspergillus versicolor, and Penicillium melinii. The tests showed that a relatively small reduction in the jet-to-plate distance of a single-stage, single-nozzle impactor with a tapered inlet nozzle, combined with adding a straight section of sufficient length, can significantly decrease the cut-off size to the level that is sufficient to efficiently collect spores of all fungal species. Furthermore, it appears that the slit jet design may improve the application of partial spore counting methodologies with respect to those applied to circular deposits. Data from a demonstration field study, conducted with the two samplers in environments containing a variety of fungal species, supported the laboratory findings.  相似文献

In this study, glycerol was tested as a collection substrate for passive bioaerosol sampling. Filters (mixed cellulose acetate and nitrate) were soaked in glycerol and exposed for an aerosol from three different fungal species: Penicillum commune, Aspergillus versicolor and Paecilomyces variotii. The passive sampling method was compared with a closed-face polycarbonate filter sampling method. Exposure was performed in an exposure chamber. The total number of spores was determined by microscopic techniques, and the cultivable number was determined by cultivation on Malt Extract Agar dishes. The glycerol soaked filter demonstrated a good correlation with the closed-face sampler with regard to the total count. Spores stored in a pumped filter cassette were not affected by storage for up to 7 days. On the other hand, the culturability of the spores was markedly decreased after 1 day when stored on glycerol soaked filters.  相似文献

The need to determine occupational exposure to bioaerosols has notably increased in the past decade, especially for microbiology-related workplaces and laboratories. Recently, two new cyclone-based personal bioaerosol samplers were developed by the National Institute for Occupational Safety and Health (NIOSH) in the USA and the Research Center for Toxicology and Hygienic Regulation of Biopreparations (RCT & HRB) in Russia to monitor bioaerosol exposure in the workplace. Here, a series of wind tunnel experiments were carried out to evaluate the physical sampling performance of these two samplers in moving air conditions, which could provide information for personal biological monitoring in a moving air environment. The experiments were conducted in a small wind tunnel facility using three wind speeds (0.5, 1.0 and 2.0 m s(-1)) and three sampling orientations (0°, 90°, and 180°) with respect to the wind direction. Monodispersed particles ranging from 0.5 to 10 μm were employed as the test aerosols. The evaluation of the physical sampling performance was focused on the aspiration efficiency and capture efficiency of the two samplers. The test results showed that the orientation-averaged aspiration efficiencies of the two samplers closely agreed with the American Conference of Governmental Industrial Hygienists (ACGIH) inhalable convention within the particle sizes used in the evaluation tests, and the effect of the wind speed on the aspiration efficiency was found negligible. The capture efficiencies of these two samplers ranged from 70% to 80%. These data offer important information on the insight into the physical sampling characteristics of the two test samplers.  相似文献

As a complement to traditional exposure assessment, monitoring deposition of aerosols can be a simple and quick screening method for identifying deposited aerosols. In this presentation examples of screening studies, based on wipe sampling in combination with adequate analytical techniques, are described. These screening methods are rapid, simple and easy to carry out. The examples given in this presentation show a broad applicability and the methods are proven useful for assessing aerosol distribution in the workplace as well as to identify target spots for more extensive assessment of a worker's exposure situation.  相似文献

Aerosols of water contaminated with Legionella bacteria constitute the only mode of exposure for humans. However, the prevention strategy against this pathogenic bacteria risk is managed through the survey of water contamination. No relationship linked the Legionella bacteria water concentration and their airborne abundance. Therefore, new approaches in the field of the metrological aspects of Legionella bioaerosols are required. This study was aimed at testing the main principles for bioaerosol collection (solid impaction, liquid impingement and filtration) and the in situ hybridization (FISH) method, both in laboratory and field assays, with the intention of applying such methodologies for airborne Legionella bacteria detection while showering. An aerosolization chamber was developed to generate controlled and reproducible L. pneumophila aerosols. This tool allowed the identification of the liquid impingement method as the most appropriate one for collecting airborne Legionella bacteria. The culturable fraction of airborne L. pneumophila recovered with the liquid impingement principle was 4 and 700 times higher compared to the impaction and filtration techniques, respectively. Moreover, the concentrations of airborne L. pneumophila in the impinger fluid were on average 7.0 x 10(5) FISH-cells m(-3) air with the fluorescent in situ hybridization (FISH) method versus 9.0 x 10(4) CFU m(-3) air with the culture method. These results, recorded under well-controlled conditions, were confirmed during the field experiments performed on aerosols generated by hot water showers in health institutions. This new approach may provide a more accurate characterization of aerobiocontamination by Legionella bacteria.  相似文献

Atmospheric aerosols were collected during the scientific expedition to Mt. Qomolangma (Everest) in May–June, 2005. The elemental concentrations of the aerosols were determined by inductively coupled plasma mass spectrometry. This yielded data for the concentration of 14 elements: Na, Mg, Al, K, Ca, Ti, V, Cr, Mn, Fe, Ni, Cu, Zn, and Pb. The mean elemental concentrations were generally comparable with those from central Asia and the Arctic, while much higher than those from Antarctic. Size, morphology, and chemical composition of 900 individual aerosol particles were determined by scanning electron microscopy and energy-dispersive X-ray microanalysis. Based on morphology and elemental composition, the particles were clustered into eight groups: soot (8%), tar ball (3%), alumosilicates/silica (55%), calcium sulfate (16%), Ca/Mg carbonate (2%), Fe/Ti-rich particles (3%), Pb-rich particles (1%), and biological particles (12%). The sampling site, located at 6,520 m in the Himalayas, is particularly remote and located at high altitude. Nonetheless, high aerosol enrichment factors for copper, chromium, lead, nickel, vanadium, and zinc all suggest the influence of long-range transported pollution, while enrichment in calcium and the presence of alumino-silicates in individual particle analyses indicates a distinct mineral dust influence. The backward air mass trajectories showed that the northwestern part of India may contribute to the atmospheric aerosol in the central high Himalayas.  相似文献

Direct-reading aerosol measurement usually uses the optical properties of airborne particles to detect and measure particle concentration. In the case of occupational hygiene, mass concentration measurement is often required. Two aerosol monitoring methods are based on the principle of light scattering: optical particle counting (OPC) and photometry. The former analyses the light scattered by a single particle, the latter by a cloud of particles. Both methods need calibration to transform the quantity of scattered light detected into particle concentration. Photometers are simpler to use and can be directly calibrated to measure mass concentration. However, their response varies not only with aerosol concentration but also with particle size distribution, which frequently contributes to biased measurement. Optical particle counters directly measure the particle number concentration and particle size that allows assessment of the particle mass provided the particles are spherical and of known density. An integrating algorithm is used to calculate the mass concentration of any conventional health-related aerosol fraction. The concentrations calculated thus have been compared with simultaneous measurements by conventional gravimetric sampling to check the possibility of field OPC calibration with real workplace aerosols with a view to further monitoring particle mass concentration. Aerosol concentrations were measured in the food industry using the OPC GRIMM? 1.108 and the CIP 10-Inhalable and CIP 10-Respirable (ARELCO?) aerosol samplers while meat sausages were being brushed and coated with calcium carbonate. Previously, the original OPC inlet had been adapted to sample inhalable aerosol. A mixed aerosol of calcium carbonate and fungi spores was present in the workplace. The OPC particle-size distribution and an estimated average particle density of both aerosol components were used to calculate the mass concentration. The inhalable and respirable aerosol fractions calculated from the OPC data are closely correlated with the results of the particle size-selective sampling using the CIP 10. Furthermore, the OPC data allow calculation of the thoracic fraction of workplace aerosol (not measured by sampling), which is interesting in the presence of allergenic particles like fungi spores. The results also show that the modified COP inlet adequately samples inhalable aerosol in the range of workplace particle-size distribution.  相似文献

An understanding of the scaling laws governing aerosol sampler performance leads to new options for testing aerosol samplers at small scale in a small laboratory wind tunnel. Two methods are described in this paper. The first involves an extension of what is referred to as the "conventional" approach, in which scaled aerosol sampler systems are tested in a small wind tunnel while exposed to relatively monodisperse aerosols. Such aerosols are collected by test and reference samplers respectively and assessed gravimetrically. The new studies were carried out for a modified, low flowrate version of the IOM personal inhalable aerosol sampler. It was shown that such experiments can be carried out with a very high level of repeatability, and this supported the general validity of the aerosol sampler scaling laws. The second method involves a novel testing system and protocol for evaluating the performances of aerosol samplers. Here, scaled aerosol samplers of interest are exposed to polydisperse aerosols, again in a small wind tunnel. In this instance, the sampled particles are counted and sized using a direct-reading aerodynamic particle sizer (the APS). A prototype automated aerosol sampler testing system based on this approach was built and evaluated in preliminary experiments to determine the performance of another modified version of the IOM personal inhalable aerosol sampler. The design of the new test system accounts for the complex fluid mechanical coupling that occurs near the sampler inlet involving the transition between the external flow outside the sampler and the internal airflow inside the sampler, leading in turn to uncontrolled particle losses. The problem was overcome by the insertion of porous plastic foam plugs. where the penetration characteristics are well understood, into the entries of both the test and the reference samplers. Preliminary experiments with this new system also supported the general validity of the aerosol sampler scaling laws. In addition, they demonstrated high potential that this approach may be applied in a standardised aerosol testing method and protocol.  相似文献

Three sampling and analytical methods have been developed and evaluated for ortho-phthalaldehyde (OPA): (1) an HPLC-UV method for OPA in air, (2) a fluorimetric method for OPA on surfaces, and (3) a colorimetric method for OPA on surfaces. (1) The air sampler contains 350 mg of silica gel coated with 1 mg of acidified 2,4-dinitrophenylhydrazine (DNPH). Air sampling may be conducted at 0.03 to 1.0 L min(-1) for periods up to 8 h. Samples were eluted with ethyl acetate, and the eluents were allowed to stand for 72 h. Analysis was by high performance liquid chromatography (HPLC) with a UV detector set at 369 nm. An unusual phenomenon was the observation that the stability of the sample on a sampler at 3 degrees C tends to decrease as the total quantity of OPA collected on the sampler decreases. Elution of the samples within 24 h of air sampling is required. The detection limit (LOD) is approximately 0.02 microg of OPA per sample. OPA on surfaces may be collected with strips cut from a sheet of polyvinyl alcohol (PVA wipe). (2) In the surface wipe method with analysis by fluorescence measurement, the strips of PVA wipe were placed into dimethyl sulfoxide. An aliquot was treated with aqueous N-acetyl-l-cysteine and ethylenediamine. Analysis was performed with a portable fluorometer (excitation and emission wavelengths = 365 nm and 438 nm, respectively). The LOD is 0.2 microg per sample. (3) In the surface wipe method with visual colorimetric detection, the strips of PVA wipe were placed into 30 : 70 acetonitrile : water. An aliquot was treated with N-(1-naphthyl)ethylenediamine in 0.1 m sulfuric acid. After color development, the LOD is approximately 48 microg per sample. These methods have been field tested in a hospital.  相似文献

One hundred workers carried personal air sampling equipment during three days to assess exposure to inhalable and respirable Mn. A novel four-step chemical fractionation procedure developed for the speciation of Mn in workroom aerosols was applied for selected aerosol filters. Blood and urine samples were analysed for Mn. The geometric mean (GM) concentrations of inhalable (n = 265) and respirable (n = 167) Mn determined in all filters were 254 microg m(-3) and 28 microg m(-3) respectively. Only 10.6% (95% CI 8.9-12.5) respirable Mn was found in the inhalable fraction when inhalable and respirable samples collected in parallel were considered (n = 153 pairs). There was a high correlation (Pearson's r = 0.70; p < 0.001) between respirable and inhalable Mn. The largest amounts of Mn in the inhalable aerosol fraction were found as Mn0 and Mn2+ (47.4%), whereas 28% was practically "insoluble". The associations between B-Mn and aerosol concentrations of Mn were weak, but an association was found between U-Mn and respirable Mn; Pearson's r being 0.38 between "soluble" respirable Mn and U-Mn. No significant association was found between the "insoluble" components (probably SiMn) and Mn in biological samples.  相似文献
