The source water of a drinking water treatment plant prone to blooms, dominated by potential microcystin-producing cyanobacteria, was monitored for two seasons in 2007-2008. In the 2008 season, the median value for potential microcystin-producing cyanobacterial biovolume was 87% of the total phytoplankton biovolume in the untreated water of the plant. Depth profiles taken above the plant's intake identified three sampling days at high risk for the contamination of the plant's raw water with potentially toxic cyanobacteria. Chlorophyceae and Bacillariophyceae caused false positive values to be generated by the phycocyanin probe when cyanobacteria represented a small fraction of the total phytoplanktonic biovolume present. However, there was little interference with the phycocyanin probe readings by other algal species when potential microcystin-producing cyanobacteria dominated the phytoplankton of the plant's untreated water. A two-tiered method for source water monitoring, using in vivo phycocyanin fluorescence, is proposed based on (1) a significant relationship between in vivo phycocyanin fluorescence and cyanobacterial biovolume and (2) the calculated maximum potential microcystin concentration produced by dominant Microcystis sp. biovolume. This method monitors locally-generated threshold values for cyanobacterial biovolume and microcystin concentrations using in vivo phycocyanin fluorescence.  相似文献

Toxic cyanobacteria threaten the water quality of drinking water sources across the globe. Two such water bodies in Canada (a reservoir on the Yamaska River and a bay of Lake Champlain in Québec) were monitored using a YSI 6600 V2-4 (YSI, Yellow Springs, Ohio, USA) submersible multi-probe measuring in vivo phycocyanin (PC) and chlorophyll-a (Chl-a) fluorescence, pH, dissolved oxygen, conductivity, temperature, and turbidity in parallel. The linearity of the in vivo fluorescence PC and Chl-a probe measurements were validated in the laboratory with Microcystis aeruginosa (r(2) = 0.96 and r(2) = 0.82 respectively). Under environmental conditions, in vivo PC fluorescence was strongly correlated with extracted PC (r = 0.79) while in vivo Chl-a fluorescence had a weaker relationship with extracted Chl-a (r = 0.23). Multiple regression analysis revealed significant correlations between extracted Chl-a, extracted PC and cyanobacterial biovolume and in vivo fluorescence parameters measured by the sensors (i.e. turbidity and pH). This information will help water authorities select the in vivo parameters that are the most useful indicators for monitoring cyanobacteria. Despite highly toxic cyanobacterial bloom development 10 m from the drinking water treatment plant's (DWTP) intake on several sampling dates, low in vivo PC fluorescence, cyanobacterial biovolume, and microcystin concentrations were detected in the plant's untreated water. The reservoir's hydrodynamics appear to have prevented the transport of toxins and cells into the DWTP which would have deteriorated the water quality. The multi-probe readings and toxin analyses provided critical evidence that the DWTP's untreated water was unaffected by the toxic cyanobacterial blooms present in its source water.  相似文献

The performance of two field probes (YSI 6600 and TriOS), used for the measurement of in vivo phycocyanin fluorescence, was compared and validated in the laboratory in 2008 and 2009 with cultures of Microcystis aeruginosa and field samples. The background noise of the two probes was low and the detection limits were estimated at 1500 cells mL(-1) for the YSI and 0.69 μg PC L(-1) for the TriOS. The linearity and repeatability of both probes have been excellent. Strong relationships were observed between the in vivo fluorescence and the total cyanobacterial biovolume (R(2) = 0.82 YSI; 0.83 TriOS) or the abundance (R(2) = 0.71 YSI; 0.75 TriOS) of cyanobacteria. However, the difference between cell densities determined by microscopy and measured by the YSI can be very large and has been associated to the variability of cell volume among cyanobacteria. This last observation makes the YSI a qualitative tool if a post-calibration is not done. The analysis of filtrated samples showed that dissolved phycocyanin (extracellular) may represent a significant fluorescence signal. No relationship could be established between the abundance, the total cyanobacterial biovolume or the in vivo fluorescence of phycocyanin and the concentrations of cyanotoxins (R(2) ≤ 0.22).  相似文献

Blooms of toxic cyanobacteria in Algerian reservoirs represent a potential health problem, mainly from drinking water that supplies the local population of Ain Zada (Bordj Bou Arreridj). The objective of this study is to monitor, detect, and identify the existence of cyanobacteria and microcystins during blooming times. Samples were taken in 2013 from eight stations. The results show that three potentially toxic cyanobacterial genera with the species Planktothrix agardhii were dominant. Cyanobacterial biomass, phycocyanin (PC) concentrations, and microcystin (MC) concentrations were high in the surface layer and at 14 m depth; these values were also high in the treated water. On 11 May 2013, MC concentrations were 6.3 μg/L in MC-LR equivalent in the drinking water. This study shows for the first time the presence of cyanotoxins in raw and treated waters, highlighting that regular monitoring of cyanobacteria and cyanotoxins must be undertaken to avoid potential health problems.  相似文献

The occurrence and severity of harmful cyanobacterial or blue-green algal blooms (HABs) have increased in recent decades, posing a serious threat of illness to humans. In some countries, water contaminated with cyanotoxins that is used for drinking or haemodialysis has posed a particularly serious risk. However, it is now recognized that recreational exposure to natural toxins by skin contact, accidental swallowing of water or inhalation can also cause a wide range of acute or chronic illnesses. In this review, we focus on the importance of cyanotoxin management in recreational waters. The symptoms related with HAB poisonings, the recommended safety concentrations limit for cyanobacteria and cyanotoxins in such waters, as well as early health hazard indicators of their presence and their monitoring are all discussed. We also present in this review an overview of the methods developed in recent decades for eliminating cyanobacteria and the toxic compounds that they produce.  相似文献

A monitoring program of cyanobacteria and cyanotoxins in the framework of the surveillance of the Water Treatment Plant efficiency of the municipality of Santa Comba D?o (Portugal) was conducted from 1994 until 2007. With these data we aimed to answer the question, are MCs produced evenly over the years in a single water body? Samples were taken by the local health authorities in the site of the Water treatment plant and analyzed for total phytoplankton, cyanobacteria and the hepatotoxic cyanotoxins microcystins (MCs). Apart from 1999 and 2000, cyanobacteria represented a high proportion of total phytoplankton, attaining during several months of the year more than 90% of total phytoplankton density. A total of 24 cyanobacteria species were identified and Microcystis aeruginosa, Anabaena flos-aquae and Aphanizomenon flos-aquae were the main potentially toxic cyanobacteria species present throughout the period. MC concentration varied between 0.3 μg MC-LR eq l?1 in October 98 and 87.0 μg MC-LR eq l?1 in September 2001. The evolution of the average cell quota does not reveal any especial trend, although in 2001 the quota was the highest but not significantly different from the other years. The threshold limit of 5000 cells per ml of Microcystis aeruginosa should be taken into consideration in monitoring programs because the 10,000 cells per ml proposed by several other national programs might not be enough to prevent human health risks. Due to high annual variation in MC cell quota, monitoring programs of cyanobacteria and MC should be extended in time, since short term studies do not provide us the data needed for a safe management of a water body used for human purposes.  相似文献

An unusual bloom of Chrysosporum ovalisporum (basionym Aphanizomenon ovalisporum) occurred for the first time in the Murray River and distributary rivers in New South Wales, Australia, from mid-February to early June 2016. At its greatest extent, it contaminated a combined river length of ca. 2360 km. Chrysosporum ovalisporum usually comprised >99% of the total bloom biovolume at most locations sampled, which at times exceeded 40 mm3 l?1. The origins of the bloom were most likely reservoirs on the upper Murray River, with cyanobacterial-infested water released from them contaminating the river systems downstream. An integrated approach using three analytical methods: (1) identification and enumeration by microscopy, (2) multiplex quantitative polymerase chain reaction (qPCR), and (3) toxin analysis, was used to obtain data for the assessment of risk to water users and management of the bloom. qPCR indicated some cyrA and stxA genes responsible for cylindrospermopsin and saxitoxin biosynthesis respectively were present, but mostly below the level of quantification. No mcyE genes for microcystin biosynthesis were detected. Toxin analysis also revealed that cylindrospermopsin, saxitoxin and microcystin were all below detection. Lack of measurable toxicity in a species usually considered a cylindrospermopsin producer elsewhere meant the possibility of relaxing management guidelines; however, high (Red) alerts needed to be maintained due to risk to water users from other biohazards potentially produced by the cyanobacteria such as contact irritants. A three-tiered monitoring strategy is suggested for monitoring cyanobacterial blooms to provide enhanced data for bloom management.  相似文献

Phytoplankton and water quality long term trends are presented from a 20-year monitoring program of Chesapeake Bay and several of its major tributaries. Increasing phytoplankton biomass and abundance are ongoing within this estuarine complex, with diatoms the dominant component, along with chlorophytes and cyanobacteria as sub-dominant contributors in the tidal freshwater and oligohaline regions. Diatoms, dinoflagellates, and cryptomonads are among the major flora downstream in the tributaries and within the Chesapeake Bay. Water quality conditions within the three tributaries have remained rather stable over this time period; while there are long term trends of reduced nutrients, increasing bottom oxygen, and decreasing water clarity for the lower Chesapeake Bay. Of note is an increasing trend of cyanobacteria biomass at 12 of the 13 stations monitored at tributary and Chesapeake Bay stations, plus the presence of 37 potentially harmful taxa reported for these waters. However, the overall status of the phytoplankton populations is presently favorable, in that it is mainly represented and dominated by taxa suitable as a major food and oxygen source within this ecosystem. Although potentially harmful taxa are present, they have not at this time exerted profound impact to the region, or replaced the diatom populations in overall dominance.  相似文献

Molecular techniques have clear value for community characterization; however, almost all previous datasets are based upon non-molecular measurements and it is hard to compare "old" data with "new" data because few correlations have been made. Therefore, the purpose of this evaluation was to simultaneously use molecular and non-molecular methods within the same sampling program to determine how data compare. Three methods were used for characterizing microbial populations in Lake 260 (L260) at Experimental Lakes Area (Ontario, Canada) during a whole-lake exposure study. Methods included whole-cell microscopic counts (for bacteria, cyanobacteria, algae and zooplankton), chlorophyll a, and small sub-unit (ssu)-rRNA hybridization using EUB-338, EUB-785, CYAN-785-a/b, EUCA-1379 and UNIV-1390 gene probes. Strong correlations were found between the EUB-785 probe signal and "bacteria minus cyanobacteria" direct counts, and the EUB-338 probe signal and "bacteria plus cyanobacteria" counts. Furthermore, the difference in probe signal between EUB-338 and EUB-785 (a presumptive signature for cyanobacteria and plastids) correlated with cyanobacterial direct counts and also with chlorophyll a. However, EUCA-1379 probe signal did not correlate with algae counts, and UNIV-1390 probe signal only correlated with total bacteria counts. Results suggest that, although ssu-rRNA methods are fast, reproducible, and specifically detect "viable" organisms, their use may be limited to non-eukaryotic populations unless new probes are developed that are more specific.  相似文献

Toxic cyanobacteria (TCB) are known worldwide for the adverse impacts on humans and animals. Species composition and the seasonal variation of TCB in water bodies depend on interactions between physical and chemical factors. The present investigation delineates temporal variations in physico-chemical water quality parameters, viz. nutrients and density, diversity, and distribution of toxic cyanobacteria and cyanotoxins in Lake Ambazari (21°7′52″N, 79°2′22″E) and Lake Phutala (21°9′18″N, 79°2′37″E) at Nagpur (Maharashtra State), India. These lakes are important sources of recreational activities and fisheries. Toxic cyanobacterial diversity comprised Anabaena, Oscillatoria, Lyngbya, Phormidium, and Microcystis, a well-known toxic cyanobacterial genus, as dominant. Chlorophyll-a concentrations in the lakes ranged from 1.44 to 71.74 mg/m3. A positive correlation of Microcystis biomass existed with orthophosphate-P (p?<?0.05) and nitrate-N (p?>?0.05). Identification and quantification of microcystin variants were carried out by high performance liquid chromatography equipped with photodiode array detector. Among all the tested toxin variants, microcystin-RR (arginine–arginine) was consistently recorded and exhibited a positive correlation (p?<?0.05) with Microcystis in both the water bodies. Microcystis bloom formation was remarkable between post-monsoon and summer. Besides nutrient concentrations governing bloom formation, the allelopathic role of microcystins needs to be established.  相似文献

Remote sensing has been used from the 1980s to study inland water quality. However, it was not until the beginning of the twenty-first century that CHRIS (an experimental multi-angle sensor with good spectral and spatial resolutions) and MERIS (with good temporal and spectral resolutions) started to acquire imagery with very good resolutions, which allowed to develop a reliable imagery acquisition system so as to consider remote sensing as an inland water management tool. This paper presents the methodology developed, from the field data acquisition with which to build a freshwater spectral library and the study of different atmospheric correction systems for CHRIS mode 2 and MERIS images, to the development of algorithms to determine chlorophyll-a and phycocyanin concentrations and bloom sites. All these algorithms allow determining water eutrophic and ecological states, apart from generating surveillance maps of toxic cyanobacteria with the main objective of Assessment of the Water Quality as it was used for Monitoring Ecological Water Quality in smallest Mediterranean Reservoirs integrated in the Intercalibration Exercise of European Union Water Framework Directive (WFD). We keep on using it to monitor the Ecological Quality Ratio (EQR) in Spain inland water.  相似文献

An innovative framework for optimising investments in water quality monitoring has been developed for use by water and environmental agencies. By utilising historical data, investigating the accuracy of monitoring methods and considering the risk tolerance of the management agency, this new methodology calculates optimum water quality monitoring frequencies for individual water bodies. Such information can be applied to water quality constituents of concern in both engineered and natural water bodies and will guide the investment of monitoring resources. Here we present both the development of the framework itself and a proof of concept by applying it to the occurrence of hazardous cyanobacterial blooms in freshwater lakes. This application to existing data demonstrates the robustness of the approach and the capacity of the framework to optimise the allocation of both monitoring and mitigation resources. When applied to cyanobacterial blooms in the Swan Coastal Plain of Western Australia, we determined that optimising the monitoring regime at individual lakes could greatly alter the overall monitoring schedule for the region, rendering it more risk averse without increasing the amount of monitoring resources required. For water resources with high-density temporal data related to constituents of concern, a similar reduction in risk may be observed by applying the framework.  相似文献

The increasing incidence of toxic cyanobacterial blooms, together with the difficulties to reliably predict cyanobacterial toxin (e.g. microcystins) concentration, has created the need to assess the predictive ability and variability of the cyanobacterial biomass–microcystin relationship, which is currently used to assess the risk to human and ecosystems health. To achieve this aim, we assessed the relationship between cyanobacterial biomass and microcystin concentration on a spatiotemporal scale by quantifying the concentration of cyanobacterial biomass and microcystin in eight lakes over 9 months. On both a temporal and spatial scale, the variability of microcystin concentration exceeded that of cyanobacterial biomass by up to four times. The relationship between cyanobacterial biomass and microcystin was weak and site specific. The variability of cyanobacterial biomass only explained 25 % of the variability in total microcystin concentration and 7 % of the variability of cellular microcystin concentration. Although a significant correlation does not always imply real cause, the results of multiple linear regression analysis suggest that the variability of cyanobacterial biomass and cellular microcystin concentration is influenced by salinity and total phosphorus, respectively. The weak cyanobacterial biomass–microcystin relationship, coupled with the fact that microcystin was present in concentrations exceeding the WHO drinking water guidelines (1 μg?L?1) in most of the collected samples, emphasizes the high risk of error connected to the traditional indirect microcystin risk assessment method.  相似文献

The aim of this study was to analyze the temporal distribution of phytoplanktonic cyanobacteria in a site located in the freshwater tidal zone near the extraction point for the drinking water supply. Samples were taken considering three timescales as follows: hours, days, and weeks, during the period of highest development of cyanobacteria. The phytoplankton density, microcystin concentration (LR, RR, YR), and chlorophyll-a were related to meteorological variables (wind and temperature), tidal high, and physical-chemical variables (nutrients, pH, conductivity, light penetration). The results obtained in this study showed that the variables that primarily modulate the temporal distribution of cyanobacteria were temperature, pH, light penetration, conductivity, and nutrients (particularly NO3 ? and NH4 +), while the winds and tide had a secondary effect, only evidenced at an hourly scale. Therefore, this timescale would be the most suitable for monitoring cyanobacterial populations, when the amount of cyanobacterial cells exceeds the alert I level proposed by the World Health Organization. This recommendation is particularly important for the water intake zones in Río de la Plata, which are vulnerable to the damage generated by cyanobacteria on the water quality.  相似文献

Metals have been central to the development of human civilisation from the Bronze Age to modern times, although in the past, metal mining and smelting have been the cause of serious environmental pollution with the potential to harm human health. Despite problems from artisanal mining in some developing countries, modern mining to Western standards now uses the best available mining technology combined with environmental monitoring, mitigation and remediation measures to limit emissions to the environment. This paper develops risk screening and prioritisation methods previously used for contaminated land on military and civilian sites and engineering systems for the analysis and prioritisation of chemical risks from modern metal mining operations. It uses hierarchical holographic modelling and multi-criteria decision making to analyse and prioritise the risks from potentially hazardous inorganic chemical substances released by mining operations. A case study of an active platinum group metals mine in South Africa is used to demonstrate the potential of the method. This risk-based methodology for identifying, filtering and ranking mining-related environmental and human health risks can be used to identify exposure media of greatest concern to inform risk management. It also provides a practical decision-making tool for mine acquisition and helps to communicate risk to all members of mining operation teams.  相似文献

Environmental health monitoring and surveillance include activities such as collection of information on the production and use of chemicals; preparation of inventories of waste discharges; measurement of physical, chemical and biological agents in air, water and food, at work place and at home; epidemiological investigations, and collection and analysis of environmental, and health statistical data. There are two main objectives of these activities: estimation of human exposure to potentially harmful environmental factors and timely detection of adverse health effects; and the assessment of environmental conditions in relation to established guidelines and standards. Environmental health monitoring and surveillance projects initiated, organized and implemented by the Specialized Agencies and other bodies of the United Nations system include monitoring of air and water quality and of food and animal feed contamination; pilot projects on air pollution exposure assessment and biological monitoring; and ionizing radiation surveillance. Principles of environmental and health monitoring in occupational environment, and of monitoring and surveillance of environmental health effects are outlined. The effectiveness of different monitoring methods in detecting temporal changes in water quality depends on the achievable sampling intervals, and how these relate to the extent of temporal variation. However, water quality sampling frequencies are rarely adjusted to the actual variation of the monitoring area. Manual sampling, for example, is often limited by the level of funding and not by the optimal timing to take samples. Restrictions in monitoring methods therefore often determine their ability to estimate the true mean and variance values for a certain time period or season. Consequently, we estimated how different sampling intervals determine the mean and standard deviation in a specific monitoring area by using high frequency data from in situ automated monitoring stations. Raw fluorescence measurements of chlorophyll a for three automated monitoring stations were calibrated by using phycocyanin fluorescence measurements and chlorophyll a analyzed from manual water samples in a laboratory. A moving block bootstrap simulation was then used to estimate the standard errors of the mean and standard deviations for different sample sizes. Our results showed that in a temperate, meso-eutrophic lake, relatively high errors in seasonal statistics can be expected from monthly sampling. Moreover, weekly sampling yielded relatively small accuracy benefits compared to a fortnightly sampling. The presented method for temporal representation analysis can be used as a tool in sampling design by adjusting the sampling interval to suit the actual temporal variation in the monitoring area, in addition to being used for estimating the usefulness of previously collected data.  相似文献
