A field study was conducted to evaluate the suitability of ethylene diurea (N-[2-(2-oxo-1-imidazolidinyl)ethyl]-N'-phenylurea; EDU) in assessing the impact of O3 on mung bean plants (Vigna radiata L. var. Malviya Jyoti) grown in suburban area of Allahabad city situated in a dry tropical region of India. EDU is a synthetic chemical having anti-ozonant property. Mean monthly O3 concentration varied between 64 and 69 microg m(-3) during the experimental period. In comparison to EDU-treated plants, non-EDU-treated plants showed significant reductions in plant growth and yield under ambient conditions. Significant favourable effects of EDU-application were observed with respect to photosynthetic pigments, soluble protein, ascorbic acid and phenol contents. EDU-treated plants maintained higher levels of pigments, protein and ascorbic acid in foliage as compared to non-EDU-treated ones. The study clearly demonstrated that EDU alleviates the unfavourable effects of O3 on mung bean plants, and therefore can be used as a tool to assess the growth and yield losses in areas having higher O3 concentrations.  相似文献

The present work provides an insight into the development of biochemical adaptations in mung beans against ozone (O3) toxicity. The study aims to explore the O3 stress tolerance potential of mung bean genotypes under exogenous application of growth regulators. The seeds of twelve mung bean genotypes were grown in plastic pots under controlled conditions in the glasshouse. Six treatments, control (ambient ozone level 40–45 ppb), ambient O3 with ascorbic acid, ambient ozone with silicic acid, elevated ozone (120 ppb), elevated O3 with ascorbic acid (10 mM), and elevated ozone with silicic acid (0.1 mM) were applied. The O3 fumigation was carried out using an O3 generator. The results revealed that ascorbic acid and silicic acid application decreased the number of plants with foliar O3 injury symptoms in different degrees, i.e., zero, first, second, third, and fourth degrees; whereas 0–4 degree symptoms represent, no symptoms, symptoms occupying?<?1/4, 1/4–1/2, 1/2–3/4, and?>?3/4 of the total foliage area, respectively. Application of ascorbic acid and silicic acid also prevented the plants from the negative effects of O3 in terms of fresh as well as dry matter production, leaf chlorophyll, carotenoids, soluble proteins and ascorbic acid, proline, and malondialdehyde (MDA) contents. Overall, silicic acid application proved more effective in reducing the negative effects of O3 on mung bean genotypes as compared to that of the ascorbic acid. Three mung bean genotypes (NM 20–21, NM-2006, and NM-2016) were identified to have a better adaptive mechanism for O3 toxicity tolerance and may be good candidates for future variety development programs.

The role of exogenous spermine (0.25 mM Spm, a type of polyamine (PA) in reducing Cd uptake and alleviating Cd toxicity (containing 1 and 1.5 mM CdCl2 in the growing media) effects was studied in the mung bean (Vigna radiata L. cv. BARI Mung-2) plant. Exogenously applied Spm reduced Cd content, accumulation, and translocation in different plant parts. Increasing phytochelatin content, exogenous Spm reduced Cd accumulation and translocation. Spm application reduced the Cd-induced oxidative damage which was reflected from the reduction of H2O2 content, O2 ?– generation rate, lipoxygenase (LOX) activity, and lipid peroxidation level and also reflected from the reduction of spots of H2O2 and O2 ?– from mung bean leaves (compared to control treatment). Spm pretreatment increased non-enzymatic antioxidant contents (ascorbate, AsA, and glutathione, GSH) and activities of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), glutathione S-transferase (GST), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), and glutathione reductase (GR) which reduced oxidative stress. The cytotoxicity of methylglyoxal (MG) is also reduced by exogenous Spm because it enhanced glyoxalase system enzymes and components. Through osmoregulation, Spm maintained a better water status of Cd-affected mung bean seedlings. Spm prevented the chl damage and increased its content. Exogenous Spm also modulated the endogenous free PAs level which might have the roles in improving physiological processes including antioxidant capacity, osmoregulation, and Cd and MG detoxification capacity. The overall Spm-induced tolerance of mung bean seedlings to Cd toxicity was reflected through improved growth of mung bean seedlings.  相似文献

We determined the length, volume, dry biomass, and density in seeds of five castor bean cultivars and verified notable physicochemical trait differences. Seeds were then subjected to different toxic aluminum (Al) concentrations to evaluate germination, relative root elongation, and the role of root apices’ rhizosphere mucilage layer. Seeds’ physicochemical traits were associated with Al toxicity responses, and the absence of Al in cotyledons near to the embryo was revealed by Al-hematoxylin staining, indicating that Al did not induce significant germination reduction rates between cultivars. However, in the more sensitive cultivar, Al was found around the embryo, contributing to subsequent growth inhibition. After this, to investigate the role of mucilage in Al tolerance, an assay was conducted using NH4Cl to remove root mucilage before or after exposure to different Al concentrations. Sequentially, the roots were stained with hematoxylin and a quantitative analysis of staining intensity was obtained. These results revealed the significant contribution of the mucilage layer to Al toxicity responses in castor bean seedlings. Root growth elongation under Al toxicity confirmed the role of the mucilage layer, which jointly indicated the differential Al tolerance between cultivars and an efficient Al-exclusion mechanism in the tolerant cultivar.  相似文献

Several treatment technologies are available for the treatment of palm oil mill wastes. Vermicomposting is widely recognized as efficient, eco-friendly methods for converting organic waste materials to valuable products. This study evaluates the effect of different vermicompost extracts obtained from palm oil mill effluent (POME) and palm-pressed fiber (PPF) mixtures on the germination, growth, relative toxicity, and photosynthetic pigments of mung beans (Vigna radiata) plant. POME contains valuable nutrients and can be used as a liquid fertilizer for fertigation. Mung bean seeds were sown in petri dishes irrigated with different dilutions of vermicomposted POME-PPF extracts, namely 50, 60, and 70% at varying dilutions. Results showed that at lower dilutions, the vermicompost extracts showed favorable effects on seed germination, seedling growth, and total chlorophyll content in mung bean seedlings, but at higher dilutions, they showed inhibitory effects. The carotenoid contents also decreased with increased dilutions of POME-PPF. This study recommends that the extracts could serve as a good source of fertilizer for the germination and growth enhancement of mung bean seedlings at the recommended dilutions.  相似文献

Attempt has been made to study the accumulation behaviour of a common plant, Mung bean (Vigna radiata) towards Cr(III) and Cr(VI) to have an insight on the migration and bio-magnification of Cr. For this purpose healthy germinated Mung bean seeds were sown in the sand in the presence of Hoagland's nutrient solution containing measured amount of K(2)(51)Cr(2)O(7) and (51)Cr(NO(3))(3).9H(2)O. Growth rate was also studied in the presence and absence of phosphate salts in the medium. It has been found that the transfer of chromium from soil to plant is significantly low (maximum 5% for both Cr(III) and Cr(VI)). Maximum accumulation of Cr occurs in the root with respect to the total chromium accumulation by the plant. Other parts of the Mung bean plant, e.g. cotyledons, shoot and leaves, show negligible accumulation. Therefore, the chance of direct intake of Cr through food as well as through the grazing animals to human body is less.  相似文献

Bush bean (Phaseolus vulgaris L.) lines 'S156' (O3-sensitive)/'R123' (O3-tolerant) and cultivars 'BBL 290' (O3-sensitive)/'BBL 274' (O3-tolerant) were used to study the effects of O3 on stomatal conductance (gs), density, and aperture size on leaf and pod surfaces with the objective of establishing links between the degree of plant sensitivity to O3 and plasticity of stomatal properties in response to O3. Studies in open-top chambers (OTCs) and in continuously stirred tank reactors (CSTRs) established a clear relationship between plant developmental stages, degrees of O3 sensitivity and gs: while 'S156' had higher gs rates than 'R123' earlier in development, similar differences between 'BBL 290' and 'BBL 274' were observed at later stages. Gs rates on the abaxial leaf surfaces of 'S156' and 'BBL 290', accompanied by low leaf temperatures, were significantly higher than their O3-tolerant counterparts. Exposure to O3 in CSTRs had greater and more consistent impacts on both stomatal densities and aperture sizes of O3-sensitive cultivars. Stomatal densities were highest on the abaxial leaf surfaces of 'S156' and 'BBL 290' at higher O3 concentrations (60 ppb), but the largest aperture sizes were recorded on the adaxial leaf surfaces at moderate O3 concentrations (30 ppb). Exposure to O3 eliminated aperture size differences on the adaxial leaf surfaces between sensitive and tolerant cultivars. Regardless of sensitivity to O3 and treatment regimes, the smallest aperture sizes and highest stomatal densities were found on the abaxial leaf surface. Our studies showed that O3 has the potential to affect stomatal plasticity and confirmed the presence of different control mechanisms for stomatal development on each leaf surface. This appeared to be more evident in O3-sensitive cultivars.  相似文献

Experiments were conducted to investigate the degradation of aldicarb, an oxime carbamate insecticide, in sterile, non-sterile and plant-grown soils, and the capability of different plant species to accumulate the pesticide. The degradation of aldicarb in soil followed first-order kinetics. Half lives (t1/2) of aldicarb in sterile and non-sterile soil were 12.0 and 2.7 days, respectively, which indicated that microorganisms played an important part in the degradation of aldicarb in soil. Aldicarb disappeared more quickly (p< or =0.05) in the soil with the presence of plants, and t1/2 of the pesticide were 1.6, 1.4 and 1.7 days in the soil grown with corn, mung bean and cowpea, respectively. Comparison of plant-promoted degradation and plant uptake showed that the enhanced removal of aldicarb in plant-grown soil was mainly due to plant-promoted degradation in the rhizosphere.  相似文献

The impact of UV-B radiation on 10 genotypically different barley and tomato cultivars was tested in a predictive study to screen for potentially UV-tolerant accessions and to analyze underlying mechanisms for UV-B sensitivity. Plant response was analyzed by measuring thermoluminescence, fluorescence, gas exchange and antioxidant status. Generally, barley cultivars proved to be much more sensitive against UV-B radiation than tomato cultivars. Statistical cluster analysis could resolve two barley groups with distinct differences in reaction patterns. The UV-B sensitive group showed a stronger loss in PSII photochemistry and a lower gas-exchange performance and regulation after UV-B radiation compared to the more tolerant group. The results indicate that photosynthetic light and dark reactions have to play optimally in concert to render plants more tolerant against UV-B radiation. Hence, measuring thermoluminescence/fluorescence and gas exchange in parallel will have much higher potential in identifying tolerant cultivars and will help to understand the underlying mechanisms.  相似文献

Copper tolerant fungi are known for more than 60 years but the complete mechanisms of copper tolerance by these fungi are still not fully understood. Copper tolerance has previously been linked to oxalic acid excretion by copper tolerant brown rot fungi. The oxalic acid then reacts with copper in the wood to form an insoluble and therefore less toxic copper oxalate. It has been suggested that copper tolerance could be due to lowering of the pH of the medium rather than the low solubility of copper oxalate. In order to elucidate this presumption, copper/chromium/boron (CCB) treated wood specimens were acidified with organic (oxalic, acetic, lactic, formic) and inorganic (sulphuric) acids and exposed to copper tolerant (Antrodia vaillantii, Leucogyrophana pinastri) and copper sensitive (Poria monticola, Gloeophyllum trabeum) brown rot fungal strains according to the mini block procedure. After eight weeks of exposure, the wood specimens were isolated and their mass losses determined. Additionally, electron paramagnetic resonance (EPR) measurements on the exposed specimens were performed. The EPR spectra of the specimens decayed by A. vaillantii were very similar to the EPR spectra of the specimens acidified with oxalic acid. Additionally, acidification of the CCB impregnated specimens made them significantly more susceptible to decay by both the copper tolerant and copper sensitive brown rot fungi.  相似文献

The effects of various ozone exposures in predisposing bean leaves (Phaseolus vulgaris L.) to Botrytis cinerea have been investigated under laboratory conditions. Seedlings of two bean cultivars were exposed to incremental ozone concentrations (120, 180 and 270 microg m(-3) for 8-h day(-1)) for five days and primary leaves were subsequently inoculated with conidia suspended in water or in an inorganic phosphate solution (Pi), and with mycelium. Ozone injury increased with increasing ozone concentration and was much higher in the ozone-sensitive cultivar 'Pros' than in the ozone-insensitive 'Groffy'. Ozone only increased the number of lesions on leaves of Pros after inoculation with either of the conidial suspensions. The Pi-stimulated infection in Groffy was reduced by the lower ozone concentrations. Ozone decreased lesion expansion after inoculation with mycelium. In a chronic fumigation experiment, plants of the two cultivars were exposed to 90 microg m(-3) (7-h day(-1)) and the primary and the oldest tree trifoliate leaves were inoculated after five and seven weeks of exposure. Ozone enhanced the senescence-related injury only in Pros. The number of lesions was not influenced by ozone for either cultivar, conidial suspension or inoculation date. Lesion expansion after inoculation with mycelium was generally reduced in exposed plants. Thus, contrasting effects of ozone on the susceptibility of bean leaves to B. cinerea were observed depending on the cultivar, the conidial suspension, the disease parameter and the ozone exposure pattern. In extrapolating the laboratory results to the field, it is suggested that episodic and chronic exposures to ambient ozone are of minor importance in increasing the susceptibility of bean leaves to B. cinerea.  相似文献

The stomatal resistance, measured with a ventilated diffusion porometer at various times before, during, and after exposure to 20–25 pphm ozone, was followed in water-stressed or well-watered beans, beans exposed at either low (37%) or high (73%) atmospheric humidity, and two tobacco cultivars exposed at the same two humidities. The two tobacco cultivars that were compared were the 03-susceptible Bel W-3 and the 03-resistant Consolidated L. The stomata of the water-stressed but unwilted bean plants closed quickly from a resistance of 2.9 ± 0.3 sec/cm to 8.4 ± 1.0 sec/cm when exposed to O3 whereas those in the unstressed plants closed slowly from a resistance of 2.5 ± 0.6 sec/cm to 5.2 ± 0.8 sec/cm after exposure to O3 for 10 min. Exposure to 03 for 30 min in the moist atmosphere caused no change in stomatal resistance of the bean plants whereas in the dry atmosphere the stomata closed from a resistance of 3.7 ± 0.4 sec/cm to 6.7 ± 0.6 sec/cm, but opened again when ozonation was terminated. With tobacco exposed to O3 in a dry atmosphere the stomata of the 03-resistant cultivar closed more rapidly than the 03-susceptible variety, whereas in a moist atmosphere the stomata of both cultivars closed slowly and equally during the 60 min of ozonation.  相似文献

Purpose

Electroplating industries are the main sources of heavy metals, chromium, nickel, lead, zinc, cadmium and copper. The highest concentrations of chromium (VI) in the effluent cause a direct hazards to human and animals. Therefore, there is a need of an effective and affordable biotechnological solution for removal of chromium from electroplating effluent.

Methods

Bacterial strains were isolated from electroplating effluent to find out higher tolerant isolate against chromate. The isolate was identified by 16S rDNA sequence analysis. Absorbed chromium level of bacterium was determined by inductively coupled plasma-atomic emission spectrometer (ICP-AES), atomic absorption spectrophotometer (AAS), scanning electron microscope (SEM), transmission electron microscope (TEM) and energy dispersive X-ray analysis (EDX). Removal of metals by bacterium from the electroplating effluent eventually led to the detoxification of effluent confirmed by MTT assay. Conformational changes of functional groups of bacterial cell surface were studied through Fourier transform infrared spectroscopy.

Results

The chromate tolerant isolate was identified as Bacillus cereus. Bacterium has potency to remove more than 75% of chromium as measured by ICP-AES and AAS. The study indicated the accumulation of chromium (VI) on bacterial cell surface which was confirmed by the SEM-EDX and TEM analysis. The biosorption of metals from the electroplating effluent eventually led to the detoxification of effluent. The increased survivability of Huh7 cells cultured with treated effluent also confirmed the detoxification as examined by MTT assay.

Conclusion

Isolated strain B. cereus was able to remove and detoxify chromium (VI). It would be an efficient tool of the biotechnological approach in mitigating the heavy metal pollutants.  相似文献

Effect of air pollution on peri-urban agriculture: a case study   总被引:10,自引:0,他引:10  
Peri-urban agriculture is vital for the urban populations of many developing countries. Increases in both industrialization and urbanization, and associated air pollution threaten urban food production and its quality. Six hour mean concentrations were monitored for SO(2), NO(2) and O(3) and plant responses were measured in terms of physiological characteristics, pigment, biomass and yield. Parameter reductions in mung bean (Vigna radiata), palak (Beta vulgaris), wheat (Triticum aestivum) and mustard (Brassica compestris) grown within the urban fringes of Varanasi, India correlated directly with the gaseous pollutants levels. The magnitude of response involved all three gaseous pollutants at peri-urban sites; O(3) had more influence at a rural site. The study concluded that air pollution in Varanasi could negatively influence crop yield.  相似文献

Fourteen Italian cultivars of Phaseolus vulgaris were exposed to a single pulse of ozone (O(3), 150 nl l(-1)) or to filtered air (<3 nl l(-1)) for 3.5 h. O(3) sensitivity was assessed by recording the extent of visible symptoms, effects on chlorophyll (Chl) content and changes in Chl a fluorescence parameters. This paper reports the results of an initial screening of 14 bean cultivars that was used to select a small number of cultivars for further work. Seven cultivars showed visible symptoms of injury in the range of 2-60 h after the end of the O(3) fumigation. O(3) significantly depressed total Chl content in most cultivars and a significant correlation was found between Chl content and visible symptoms. Most cultivars showed a significant change in the F(v)/F(m) ratio, even when there were no visual symptoms. There was no relationship between the extent of visual symptoms and quenching coefficients, indicating that these parameters were of no use in the determination of sensitivity to O(3) stress.  相似文献

The objective of this study was to establish relationship between boron induced oxidative stress and antioxidant system in Vigna radiata plants and also to investigate whether brassinosteroids will enhance the level of antioxidant system that could confer tolerance to the plants from the boron induced oxidative stress. The mung bean (V. radiata cv. T-44) plants were administered with 0.50, 1.0 and 2.0 mM boron at 6 d stage for 7 d along with nutrient solution. At 13 d stage, the seedlings were sprayed with deionized water (control) or 10−8 M of 28-homobrassinolide and plants were harvested at 21 d stage to assess growth, leaf gas-exchange traits and biochemical parameters. The boron treatments diminished growth, water relations and photosynthetic attributes along with nitrate reductase and carbonic anhydrase activity in the concentration dependent manner whereas, it enhanced lipid peroxidation, electrolyte leakage, accumulation of H2O2 as well as proline, and various antioxidant enzymes in the leaves of mung bean which were more pronounced at higher concentrations of boron. However, the follow-up application of 28-homobrassinolide to the boron stressed plants improved growth, water relations and photosynthesis and further enhanced the various antioxidant enzymes viz. catalase, peroxidase and superoxide dismutase and content of proline. The elevated level of antioxidant enzymes as well as proline could have conferred tolerance to the B-stressed plants resulting in improved growth, water relations and photosynthetic attributes.  相似文献

Absorption of nitrogen dioxide (NO(2)) by various broad-leaved tree species was determined by the (15)N dilution method. The tree seedlings were continuously exposed to 0.3 ppm (microl litre(-1)) NO(2) or the mixture of 0.3 ppm NO(2) and 0.1 ppm O(3) for 30 days. The total amount of NO(2)-nitrogen absorbed by a seedling during the 30-day exposure period primarily depended on the size of the seedling. Among the tested tree species, three cultivars of Populus showed the highest rate of NO(2) absorption per unit leaf area, reaching as much as 0.3 mg N per dm(2) per day. The absorption rates for Populus cultivars were more than four times greater than those for Viburnum or Cinnamomum which had the lowest rate. A highly significant correlation was recognised between the rate of NO(2) absorption and the stomatal conductance among the species. Three cultivars of Populus which had the highest rates of NO(2) absorption were most susceptible to the mixture of NO(2) and O(3). On the contrary, Cinnamomum, Viburnum and Quercus, which showed the lowest rate of NO(2) absorption, were very tolerant to the mixed gas. These results indicate that the species difference in susceptibility to the mixture of NO(2) and O(3) was mainly determined by the difference in rate of absorption of these gases. Exposure to NO(2) alone had no detrimental effect on the tested tree species.  相似文献

The effects of three cultivars, two water regimes and two rates of applying nitrogen fertilisers were tested when studying the performance of rice (Oryza sativa L.) near to, and distant from, a fertiliser plant emitting atmospherically dispersed pollutants. The atmosphere near the fertiliser plant had average daily peak concentrations of 144 and 210 microg m(-3) of SO(2) and NO(2), respectively. Growth was less near to, than at a distance from, the fertiliser plant. On average it was decreased by water stress while the effects of different amounts of nitrogen fertiliser were variable. Whilst there were a number of interactions involving nitrogen and water treatments, the most consistent were associated with the responses of the three cultivars at the two locations. The three cultivars performed similarly at the unpolluted control site, but there were major differences at the polluted site. These were most clearly exemplified by changes in the proportion of dry matter allocated to yields of grain and straw. At the unpolluted site, grain accounted for about 30% of the combined yields of grain and straw. At the polluted site, grain accounted for 1, 23 and 31% of the combined grain and straw yields of cultivars CO 43 (the most sensitive), TKM 9 and GR 3 (the most tolerant). Grain yields were closely related to numbers of filled grains per plant. At the polluted site, 98% of grains failed to develop in CO 43, whereas in GR 3, the number of panicles, and therefore the potential number of grains, was significantly enhanced.  相似文献
