Bioaccumulation kinetics of five di-, tri- and tetrachlorobiphenyls from water and food were studied in laboratory experiments with goldfish (Carassius auratus). First order rate constants for uptake from water and clearance were determined after simultaneous administration of the five compounds in constant concentration, and were related to bioconcentration factors obtained in a static fish-water equilibration system. Biomagnification by retention of the PCB's from food was studied in a separate experiment.The difference in clearance rates for the chlorobiphenyls is the main reason for the different bioconcentration and biomagnification factors.Absorption efficiencies from water and food are higher than 40%. Clearance half lives vary from 10 days for 2,5-dichlorobiphenyl to 60 days for 2,3′,4′5-tetrachlorobiphenyl, which is correlalated with the decreasing aqueous solubilities of the compounds. Bioconcentration factors are between 0.4 × 106 and 1.5 × 106, biomagnification factors between 0.2 and 1.7, based on extractable lipids. Substitution of chlorine in the position para to the phenyl-phenyl bond influences hydrophobicity and bioaccumulation of the PCB's more strongly than substitution in ortho position.A kinetic model is developed which accounts for the influence of the lipid content of the fish on the clearance rate of a chemical. Reproducible determination of the bioconcentration potential of environmental chemicals is possible by use of an “internal bioaccumulation standard” in a kinetic test system. Food chain accumulation in fish is likely to be an important process only for persistent chemicals with extremely low water solubility.  相似文献

The scientific literature for fate and effects of non-nutrient contaminant concentrations is skewed for reports describing sediment contamination and bioaccumulation for trace metals. Concentrations for at least 22 trace metals have been reported in mangrove sediments. Some concentrations exceed sediment quality guidelines suggesting adverse effects. Bioaccumulation results are available for at least 11 trace metals, 12 mangrove tissues, 33 mangrove species and 53 species of mangrove-habitat biota. Results are specific to species, tissues, life stage, and season and accumulated concentrations and bioconcentration factors are usually low. Toxicity tests have been conducted with 12 mangrove species and 8 species of mangrove-related fauna. As many as 39 effect parameters, most sublethal, have been monitored during the usual 3 to 6 month test durations. Generalizations and extrapolations for toxicity between species and chemicals are restricted by data scarcity and lack of experimental consistency. This hinders chemical risk assessments and validation of effects-based criteria.  相似文献

The impounding of the Three Gorges Reservoir (TGR) at the Yangtze River caused large flooding of urban, industrial, and agricultural areas, and profound land use changes took place. Consequently, substantial amounts of organic and inorganic pollutants were released into the reservoir. Additionally, contaminants and nutrients are entering the reservoir by drift, drainage, and runoff from adjacent agricultural areas as well as from sewage of industry, aquacultures, and households. The main aim of the presented research project is a deeper understanding of the processes that determines the bioaccumulation and biomagnification of organic pollutants, i.e., mainly pesticides, in aquatic food webs under the newly developing conditions of the TGR. The project is part of the Yangtze-Hydro environmental program, financed by the German Ministry of Education and Science. In order to test combinations of environmental factors like nutrients and pollution, we use an integrated modeling approach to study the potential accumulation and biomagnification. We describe the integrative modeling approach and the consecutive adaption of the AQUATOX model, used as modeling framework for ecological risk assessment. As a starting point, pre-calibrated simulations were adapted to Yangtze-specific conditions (regionalization). Two exemplary food webs were developed by a thorough review of the pertinent literature. The first typical for the flowing conditions of the original Yangtze River and the Daning River near the city of Wushan, and the second for the stagnant reservoir characteristics of the aforementioned region that is marked by an intermediate between lake and large river communities of aquatic organisms. In close cooperation with German and Chinese partners of the Yangtze-Hydro Research Association, other site-specific parameters were estimated. The MINIBAT project contributed to the calibration of physicochemical and bathymetric parameters, and the TRANSMIC project delivered hydrodynamic models for water volume and flow velocity conditions. The research questions were firstly focused on the definition of scenarios that could depict representative situations regarding food webs, pollution, and flow conditions in the TGR. The food webs and the abiotic site conditions in the main study area near the city of Wushan that determine the environmental preconditions for the organisms were defined. National and international chemical management programs are assessing thousands of chemicals for their persistence, bioaccumulative and environmental toxic properties; however, data for evaluating the bioaccumulation potential for fish are limited. Computer based models that account for the uptake and elimination processes that contribute to bioaccumulation may help to meet the need for reliable estimates. One critical elimination process of chemicals is metabolic transformation. It has been suggested that in vitro metabolic transformation tests using fish liver hepatocytes or S9 fractions can provide rapid and cost-effective measurements of fish metabolic potential, which could be used to refine bioconcentration factor (BCF) computer model estimates. Therefore, recent activity has focused on developing in vitro methods to measure metabolic transformation in cellular and subcellular fish liver fractions. A method to extrapolate in vitro test data to the whole body metabolic transformation rates is presented that could be used to refine BCF computer model estimates. This extrapolation approach is based on concepts used to determine the fate and distribution of drugs within the human body which have successfully supported the development of new pharmaceuticals for years. In addition, this approach has already been applied in physiologically-based toxicokinetic models for fish. The validity of the in vitro to in vivo extrapolation is illustrated using the rate of loss of parent chemical measured in two independent in vitro test systems: (1) subcellular enzymatic test using the trout liver S9 fraction, and (2) primary hepatocytes isolated from the common carp. The test chemicals evaluated have high quality in vivo BCF values and a range of logK(ow) from 3.5 to 6.7. The results show very good agreement between the measured BCF and estimated BCF values when the extrapolated whole body metabolism rates are included, thus suggesting that in vitro biotransformation data could effectively be used to reduce in vivo BCF testing and refine BCF model estimates. However, additional fish physiological data for parameterization and validation for a wider range of chemicals are needed.  相似文献

Synthetic musks are ubiquitous pollutants in aquatic environments. As hydrophobic chemicals, they can accumulate in terrestrial and aquatic organisms. Investigations into the bioaccumulation of these chemicals in aquatic ecosystem have, however, been limited, and previous results were inconsistent among species and ecosystem. Studies on this topic have been carried out in European countries, the USA, and Japan, but very few are known of the situation in China. The aim of this study was to investigate contaminant levels of musks in fish from Taihu Lake, the second largest freshwater lake in China, as well as bioaccumulation and biomagnification of the pollutants in the freshwater food chain. Five polycyclic musks and two nitro musks were determined in 24 fish species and nine surface sediment samples from Taihu Lake. HHCB (1,3,4,6,7,8-hexahydro-4,6,6,7,8,8-hexamethylcyclopenta[γ]-2-benzopyran) and AHTN (7-acetyl-1,1,3,4,4,6-hexamethyl-1,2,3,4-tetrahydronaphthalene) were the predominant contaminants in the fish samples, with concentrations ranging from below the limit of detection (LOD) to 52.9 and from <LOD to 7.5 ng/g lipid weight, respectively. Other contaminants were at low detection frequencies. The results indicated low concentrations of musks yet widespread occurrence of these contaminants in fish from Taihu Lake. Species-specific and lipid-related bioaccumulation characteristics were suggested, but no significant region-specific differences were observed. Normalized biota-sediment accumulation factors for HHCB and AHTN were noted to increase with trophic levels in fish. Trophic magnification factors were estimated at 1.12 for HHCB and 0.74 for AHTN. A biomagnification for HHCB, and probably biodilution for AHTN, in the freshwater food chain are indicated, when trophic magnification factors were concerned. However, the correlations between logarithmic concentrations of the chemicals and trophic levels were not statistically significant. Further study using long food chains in this lake is still needed.  相似文献

The contribution of bioconcentration and biomagnification of 14C-terbutryn and 14C-benzo[a]pyrene via food and water to the bioaccumulation in Gammarus fossarum and Asellus aquaticus was investigated in single species-tests. In this investigation the uptake of 14C-terbutryn and 14C-benzo[a]pyrene via food and water by G. fossarum (L.) and A. aquaticus (L.) was examined. Bioconcentration factors (BCFs) and biomagnification factors (BMFs) were determined. Calculated BCFs were clearly higher than BMFs, indicating that water is the primary route of uptake. The uptake of terbutryn and benzo[a]pyren via water in G. fossarum and A. aquaticus is faster than uptake via food. The elimination and metabolism of the two chemicals by G. fossarum and A. aquaticus were studied. Terbutryn was eliminated almost completely in both species. In general, the elimination of terbutryn from G. fossarum and A. aquaticus was fast with half-life of 5 h. The elimination of terbutryn by G. fossarum after biomagnification is slower than after bioconcentration. No difference was found in elimination of terbutryn by A. aquaticus after biomagnification and after bioconcentration. Metabolites of terbutryn in G. fossarum and A. aquaticus were analyzed by HPLC. After the bioconcentration experiment a higher percentage of metabolites was found in G. fossarum than in A. aquaticus. This was confirmed for the experiment with uptake via food. The spectrum of metabolites was similar in both species, with hydroxyterbutryn being the major metabolism product in water. 14C-B[a]P could nearly completely be eliminated by G. fossarum (rest of activity 2%) after uptake via water. 14C-B[a]P could not be eliminated by G. fossarum and A. aquaticus after uptake via food. Concentrations of organochlorine pesticides and stable isotope ratios of nitrogen and carbon were measured in a tropical freshwater ecosystem to evaluate the contamination level of biota and examine the bioaccumulation patterns of pollutants through the food web. Chemical analyses showed a general and heavy contamination of the entire food web. They revealed the strong accumulation of pollutants by juveniles of diadromous fishes and shrimps, as they re-enter the river. The role of ecological factors in the bioaccumulation of pesticides was evaluated. Whereas the most persistent pollutants (chlordecone and monohydro-chlordecone) were related to the organisms diet and habitat, bioaccumulation of β-HCH was only influenced by animal lipid content. The biomagnification potential of chlordecone through the food chain has been demonstrated. It highlighted the importance of trophic transfer in this compound bioaccumulation process. In contrast, bioconcentration by passive diffusion from water seemed to be the main exposure route of biota to β-HCH.  相似文献

Traditionally, regulatory approaches to the bioaccumulation of hydrophobic organic chemicals (HOCs) have emphasized the direct accumulation of these chemicals from solution across biological membranes, leading to the development of the bioconcentration factor as a measure of direct uptake of freely dissolved HOCs. However, an often larger fraction of the total amount of many HOCs in the water column is not freely dissolved, but is partitioned among suspended sediments and particulate matter in the water column. Partitioned HOCs are available for accumulation by organisms ingesting the contaminated particulate matter. The net accumulation of HOCs from water through consumption and direct uptake of dissolved HOC is termed bioaccumulation, quantified using a bioaccumulation factor. In order to develop recommendations designed to close the gap between current knowledge concerning bioaccumulation and regulations, the Institute of Evaluating Health Risks organized a working conference, 'The Bioaccumulation of Hydrophobic Organic Chemicals by Aquatic Organisms'. This paper reflects the view of workshop participants that the bioaccumulation paradigm can be used in a number of practical applications.  相似文献

Several models of varying complexity have been used to predict pollutant concentrations in the higher levels of the food web from those in lower levels, but the role of the biomagnification process in aquatic food chains is still controversial. We used the fugacity-based approach to verify the transfer of PCBs through the pelagic food chain of Lake Iseo (N. Italy), sampling several zebra mussel specimens and some fish belonging of different trophic levels. The zebra mussel seems to be a suitable starting species for modelling the bioaccumulation process through the trophic web, not only because its physiological characteristics and population size do not change much with time (as do algae and zooplankton) but also because it takes up toxicants exclusively from the water, as shown by the application of two predictive trophic models commonly used. The data provided by one of those models were in good agreement with our experimental data on fish in Lake Iseo, that show a not negligible uptake from food for the top predator species (pike and perch) with an increase of about three times in comparison with the PCB levels measured in the zebra mussel specimens.  相似文献

In 1989, researchers discovered that amphibians, particularly frogs and toads from many parts of the world, appeared to be declining. In many ecosystems amphibians play a central role in ecosystem energy flow and nutrient cycling, and they act as keystone species. The recent increase in solar ultraviolet-B radiation (UVB, 280-320nm) has been thought to be one stressor responsible for the decline in amphibian populations. Along with other stressors, such as habitat destruction, anthropogenic influences and natural causes, UVB radiation could contribute to adverse effects among amphibians. Amphibians provide a good model for examining the effects of environmental stressors, because both lethal and sub-lethal responses are well documented in a range of studied xenobiotics in many species. In this experiment, the effects of UVB radiation on the accumulation and depuration kinetics of bisphenol A (BPA) were studied. Additionally, the accumulation was further modeled with correction for growth dilution. The results indicate that UVB radiation did not affect the toxicokinetics of BPA, and that the applied growth correction had only a negligible influence on the toxicokinetic estimations in this experiment. However, BCFs values calculated as k(u)/k(e) where closer to C(a)/C(w) calculated values when growth dilution was incorporated in the model. This method can be used in other experiments, where the growth dilution can affect toxicokinetic estimations.  相似文献

Bioaccumulation and elimination of 14C-lindane in Enchytraeus albidus was studied in artificial OECD soil and a silty loam from an agricultural field in Central West Portugal. Results showed that enchytraeids were able to bioaccumulate the chemical with a kinetic pattern similar to that of earthworms: fast uptake within a few days and a biphasic elimination pattern. A 10 day period to study uptake was sufficient, but a few more days were probably necessary for elimination. Bioaccumulation was influenced by soil type. The authors suggest that higher organic matter (OM) content and also the higher content on sand particles in the OECD soil may have led to a faster elimination: hydrophobic chemicals tend to adsorb to OM being in this way less bioavailable and therefore less bioaccumulated having bioaccumulation factor value around 6 while in natural soil is 10; the sand could act as abrasive particles (helpers) in the elimination process leading to an elimination of 90% of the chemical in two days while in natural soil 67% was eliminated in the same period of time.  相似文献

Bioaccumulation of persistent organic pollutants (POPs) leads to high levels in human milk and high doses of POPs for nursing infants. This is currently not considered in chemical risk assessment. A coupled model for bioaccumulation of organic chemicals in breast-feeding mother and nursing infant was developed and tested for a series of organic compounds. The bioaccumulation factors (BAFs) in mother, breast milk and child were predicted to vary with log KOW and, for volatile compounds, with KAW and concentration in air. The concentrations of POPs in the infant body increase the first half year to about factor 3 above mother and decline thereafter to lower levels. The predicted results are close to empirical data and to an empirical regression. The new mother–child model is compact due to its easy structure and the analytical matrix solution. It could be added to existing exposure and risk assessment systems, such as EUSES.  相似文献

Predictive correlations of the bioaccumulation factor of persistent organic pollutants in aquatic biota are presented as functions of their octanol/water partition coefficient. The correlations demonstrate the importance of differentiating among the different levels in the food web and of accounting for the pollutant's bioavailability by considering the amount freely dissolved in water instead of the total concentration. They also reveal the significance of the pollutant's octanol/water partition coefficient value on its biomagnification along the levels of the trophic chain. Prediction results, finally, demonstrate that the correlations provide reasonably accurate estimates of bioaccumulation, typically within an order-of-magnitude.  相似文献

BACKGROUND, AIM, AND SCOPE: The use of fish embryos is not regulated by current legislations on animal welfare and is therefore considered as a refinement, if not replacement of animal experiments. Fish embryos represent an attractive model for environmental risk assessment of chemicals since they offer the possibility to perform small-scale, high-throughput analyses. MAIN FEATURES: Beyond their application for determining the acute toxicity, fish embryos are also excellent models for studies aimed at the understanding of toxic mechanisms and the indication of possible adverse and long-term effects. Therefore, we have reviewed the scientific literature in order to indicate alternative applications of the fish embryo model with focus on embryos of the zebrafish. RESULTS AND DISCUSSIONS: The analysis of the mode of action is important for the risk assessment of environmental chemicals and can assist in indicating adverse and long-term effects. Toxicogenomics present a promising approach to unravel the potential mechanisms. Therefore, we present examples of the use of zebrafish embryos to study the effect of chemicals on gene and protein patterns, and the potential implications of differential expression for toxicity. The possible application of other methods, such as kinase arrays or metabolomic profiling, is also highlighted. Furthermore, we show examples of toxicokinetic studies (bioconcentration, ABC transporters) and discuss limitations that might be caused by the potential barrier function of the chorion. Finally, we demonstrate that biomarkers of endocrine disruption, immune modulation, genotoxicity or chronic toxicity could be used as indicators or predictors of sub-acute and long-term effects. CONCLUSIONS: The zebrafish embryo represents a model with an impressive range of possible applications in environmental sciences. Particularly, the adaptation of molecular, system-wide approaches from biomedical research is likely to extend its use in ecotoxicology. RECOMMENDATIONS AND PERSPECTIVES: Challenges for future research are (1) the identification of further suitable molecular markers as indicators of the mode of action, (2) the establishment of strong links between (molecular) effects in short-term assays in embryos and long-term (toxic) effects on individuals, (3) the definition of limitations of the model and (4) the development of tests that can be used for regulatory purposes.  相似文献

Several cesium and strontium bioaccumulation models are used widely in national and international guidance for ecological and human health risk assessments for radiocesium (134Cs and 137Cs) and radiostrontium (90Sr), but have not been used to make predictions of radiological risk from nuclear accidents under variable environmental conditions on broad geographical scales. In this paper, we first present models for predicting the bioaccumulation of cesium and strontium by aquatic biota based on ambient concentrations of dissolved potassium and calcium, respectively, and then test these models using independent data from aquatic ecosystems at Canadian nuclear sites. Secondly, models yielding the best predictions across a wide range of input parameters were selected to estimate bioaccumulation factors (BAFs) for cesium and strontium in aquatic ecosystems across Canada, using trophic level of organisms and dissolved potassium for cesium and calcium concentrations for strontium as predictor variables, and presented as contour maps of radiological risk. The models show that risk from bioaccumulation of cesium and strontium can vary by several orders of magnitude depending on site-specific environmental conditions and trophic ecology. Overall, our results suggest that single-parameter approaches taken by regulatory standards may either over- or under-predict radiological risk at many locations, and are thus not readily suitable to inform siting decisions for new nuclear developments.  相似文献

Dyestuffs generally do not readily undergo aerobic biodegradation during sewage treatment processes and for new products an assessment of their bioaccumulation in fish is a requirement under certain environmental chemicals legislation. The results presented in this paper show that the partition coefficient in n-octanol/water is a useful indicator of the bioaccumulation tendency of dyestuffs, thus supporting its use as a screening test for bioaccumulation as proposed by the OECD Chemicals Testing Programme.  相似文献

In environmental life-cycle assessments (LCA), fate and exposure factors account for the general fate and exposure properties of chemicals under generic environmental conditions by means of 'evaluative' multi-media fate and exposure box models. To assess the effect of using different generic environmental conditions, fate and exposure factors of chemicals emitted under typical conditions of (1).Western Europe, (2). Australia and (3). the United States of America were compared with the multi-media fate and exposure box model USES-LCA. Comparing the results of the three evaluative environments, it was found that the uncertainty in fate and exposure factors for ecosystems and humans due to choice of an evaluative environment, as represented by the ratio of the 97.5th and 50th percentile, is between a factor 2 and 10. Particularly, fate and exposure factors of emissions causing effects in fresh water ecosystems and effects on human health have relatively high uncertainty. This uncertainty is mainly caused by the continental difference in the average soil erosion rate, the dimensions of the fresh water and agricultural soil compartment, and the fraction of drinking water coming from ground water.  相似文献

Bioaccumulation by Hyalella of all metals studied so far in our laboratory was re-evaluated to determine if the data could be explained satisfactorily using saturation models. Saturation kinetics are predicted by the biotic ligand model (BLM), now widely used in modelling acute toxicity, and are a pre-requisite if the BLM is to be applied to chronic toxicity. Saturation models provided a good fit to all the data. Since these are mechanistically based, they provide additional insights into metal accumulation mechanisms not immediately apparent when using allometric models. For example, maximum Cd accumulation is dependent on the hardness of the water to which Hyalella are acclimated. The BLM may need to be modified when applied to chronic toxicity. Use of saturation models for bioaccumulation, however, also necessitates the need for using saturation models for dose-response relationships in order to produce unambiguous estimates of LC50 values based on water and body concentrations. This affects predictions of toxicity at very low metal concentrations and results in lower predicted toxicity of mixtures when many metals are present at low concentrations.  相似文献
