In vitro study for the determination of the toxicity of some pesticides (glyphospate and paraquat) and cadmium chloride (CdCl2) on the activities of serum acetylcholinesterase (AChE), lactate dehydrogenase (LDH), aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (AlP), and acid phosphatase (AcP) is described. Changes in electrophoretic patterns of serum proteins were also tested. Results revealed that glyphosate was effective on all enzymes except AcP. Its IC50 values (the concentration of compound that inhibits 50% of the enzyme activity in 1 h at 37 degrees C) were 714.3, 750, 54.2, 270.8, and 71.4 mM for AChE, LDH, AST, ALT, and AlP, respectively. The inhibitory effect of paraquat varied markedly among all enzymes. The IC50 values of paraquat were 321.4 and 750 mM for AST and ALT, respectively. It had mild effect on AChE and LDH; and no effect on the activities of AlP and AcP. The effect of CdCl2 was pronounced with AChE, ALT, AlP, and AcP, and no effect on LDH and AST was found. The corresponding IC50 values were 77.7, 22.2, 33.3, and 83.3 mM for AChE, ALT, AlP, and AcP, respectively. Polyacrylamide gel electrophoretic patterns of serum proteins showed marked differences with glyphosate and CdCl2 but not with paraquat. The results suggest that the in vitro enzyme-activity test seems to have a potential for the assessment of pesticide and heavy metal toxicity.  相似文献

Abstract

A laboratory study was conducted to determine the effect of four experimental insecticides, DOWCO429X, DPX43898, tefluthrin and trimethacarb, on enzyme activities and levels of adenosine 5'‐triphosphate (ATP) in mineral and organic soils. DOWCO429X decreased urease activity in organic soil after 7 days while a stimulatory effect was observed with most treatments after 14 days. No inhibition on acetylene (C2H2) reduction by nitrogenase was evident with any of the insecticides in either soil. With the exception of DOWCO429X and tefluthrin at 7 days in organic soil, none of the insecticide treatments inhibited dehydrogenase activity in either soil. Dehydrogenase activity, measured by formazan formation, was greater in many samples in sandy loam than the control throughout the experiment. No inhibitory effect was observed on amylase activity after 2 or 3 days in sandy soil. A stimulatory effect was apparent in many samples after 2 days in organic soil. All insecticide treatments in sandy soil reduced invertase activity at 2 days. However, none of the experimental insecticides inhibited invertase activity after 3 days. A stimulatory effect in invertase activity was apparent in most cases at 2 days in organic soil and no difference was observed after 3 days. Phosphatase activity in insecticide treated samples was equal to or greater than that of control in sandy soil after 2 h. With the exception of DPX43898, the insecticides depressed phosphatase activity in most organic soil samples. The insecticides did not affect ATP levels in either soil. Results indicated that the chemical treatments at the levels tested did not significantly affect activities of enzymes or level of ATP in both soils.  相似文献

The dissipation of carbendazim and chloramphenicol alone and in combination and their effects on soil fungal:bacterial ratios and soil enzyme activities were investigated. The results revealed that carbendazim dissipation was little affected by chloramphenicol, whereas chloramphenicol dissipation was found to be retarded significantly by the presence of carbendazim. The inhibitory effect of carbendazim on the fungal:bacterial ratios was increased by the presence of chloramphenicol, and the inhibitory effect of chloramphenicol on neutral phosphatase was increased by the presence of carbendazim. Carbendazim increased soil catalase and urease activities, but this increase was partially diminished by the presence of chloramphenicol. Little interaction was observed between carbendazim and chloramphenicol with regard to their influence on soil invertase. The results obtained in this study suggest that combinations of fungicides and antibiotics may alter the compounds’ individual behaviors in soil and their effects on soil enzymes.  相似文献

对湘潭锰矿尾渣库地区商陆根际和非根际土壤酶的活性特征进行了研究,结果表明,商陆根际土壤各种酶的活性显著大于非根际土壤。商陆根际环境对土壤酶活性的影响表现为:蔗糖酶>脲酶>脱氢酶>酸性磷酸酶>过氧化氢酶,根际效应值(R/S)分别为:1.622、1.598、1.586、1.485和1.328。除过氧化氢酶活性外,土壤各种酶活性与重金属复合污染程度显著负相关,表现出重金属复合污染对土壤酶活性的抑制效应。商陆可有效改善土壤环境,提高土壤各种酶的活性,是锰污染土壤植物修复的理想植物。  相似文献

Yard waste compost provides an organic amendment useful for improving soil structure and nutrient status. The activities of the enzymes hydrolyzing urea (urease) and sucrose (invertase) in the rhizosphere of potato plants were determined under field conditions. Soil urease and invertase activities were monitored in compost amended soil, grass buffer strips, and in adjacent bare soils during 35 d following soil treatment. Soil urease activity was increased by application of yard waste compost compared to untreated soil which provide evidence of increased soil microbial population following application of compost. Some transitional effects on urease activities were observed following Pyrethrins and Neemix-4E application, these effects were neither drastic nor prolonged enough to be considered deleterious to the soil microorganisms and their activities important to soil fertility. No relationship was found between invertase activity and the three management practices or soil organic matter content. It is suggested that application of botanical insecticides like pyrethrins and Neemix-4E may be useful in delaying urea fertilizer mineralization to maintain N in a form less leachable, so that the duration of N availability to plants is prolonged. The present study may also provide information on urease activity as a sensitive bioindicator of soil quality that reflects the effects of land management on soil quality and may assist land managers in monitoring long-term productivity and sustainability of agricultural lands.  相似文献

Intensive remediation of abandoned former organochlorine pesticides (OCPs) manufacturing areas is necessary because the central and surrounding soils contaminated by OCPs are harmful to crop production and food safety. Organochlorine and its residues are persistent in environments and difficult to remove from contaminated soils due to their low solubility and higher sorption to the soils. We performed a comprehensive study on the remediation of OCPs-contaminated soils using thermal desorption technique and solvent washing approaches. The tested soil was thermally treated at 225, 325, 400, and 500 °C for 10, 20, 30, 45, 60, and 90 min, respectively. In addition, we tested soil washing with several organic solvents including n-alcohols and surfactants. The optimal ratio of soil/solvent was tested, and the recycling of used ethanol was investigated. Finally, activities of polyphenol oxidase (PPO), urease (URE), alkaline phosphatase, acid phosphatase (ACP), and invertase (INV) were assayed in the treated soils. The tested soil was thermally treated at 500 °C for 30 min, and the concentration of contaminants in soil was decreased from 3,115.77 to 0.33 mg kg?1. The thermal desorption in soil was governed by the first-order kinetics model. For the chemical washing experiment, ethanol showed a higher efficiency than any other solvent. Using a 1:20 ratio of soil/solvent, the maximum removal of OCPs was achieved within 15 min. Under this condition, approximately 87 % of OCPs was removed from the soils. More than 90 % of ethanol in the spent wash fluid could be recovered. Activities of some enzymes in soils were increased after ethanol treatment. But ALP, ACP, and INV activities were decreased and PPO and URE showed slightly higher activities following remediation by thermal treatment. Both heating temperature and time were the key factors for thermal desorption of OCPs. The n-alcohol solvent showed higher removal of OCPs from soils than surfactants. The highly efficient removal of OCPs from soil was achieved using ethanol. More than 90 % of ethanol could be recovered and be reused following distillation. This study provides a cost-effective and highly efficient way to remediate the OCPs-contaminated soils.  相似文献

Laboratory experiments were conducted to determine the effect of 32 pesticides applied at 2 levels on populations of microorganisms, activities of urease, dehydrogenase, phosphatase and nitrogenase in a clay loam incubated for 1 week. Results indicated that a decrease in bacterial number was observed with thiram for 2 days and stimulation with chlorpyrifos after 7 days. Some fungicides and fumigants inhibited fungal numbers for 2 days. The recovery was rapid and stimulatory effects on microbial numbers were evident in many samples. None of the pesticides inhibited soil urease drastically. Formazan formation was not suppressed vigorously by the treatments. With the exception of DD and Vorlex at a high level, none of the treatments inhibited phosphatase in the hydrolysis of p-nitrophenyl disodium orthophosphate. A temporary decrease in nitrogenase activity in acetylene (C2H2) reduction was observed with many pesticides. The low amount of pesticides applied to the clay loam is unlikely to have detrimental effects on soil microbes and the enzymes important to soil fertility.  相似文献

采用室内培养实验,通过将外源Cd添加到同一母质、全镉含量相近的不同利用方式的红壤中(林地、水稻土和菜园土),研究了外源Cd污染对不同利用方式红壤脲酶活性的影响。结果表明:整个培养过程中,Cd污染对3种红壤脲酶活性都有抑制作用,且随重金属浓度的增强而增强。同剂量Cd污染对3种红壤脲酶活性的抑制效应不同,大小为林地>水稻土>菜园土。实验设定的Cd处理水平下,对林地、水稻土和菜园土脲酶活性产生显著抑制作用(p<0.05)的Cd浓度分别为5、30和50 mg/kg土。  相似文献

The aim of this study was to investigate the behavior of the association between atrazine and glyphosate in the soil through mineralization and degradation tests. Soil treatments consisted of the combination of a field dose of glyphosate (2.88 kg ha?1) with 0, ?, 1 and 2 times a field dose of atrazine (3.00 kg ha?1) and a field dose of atrazine with 0, ?, 1 and 2 times a field dose of glyphosate. The herbicide mineralization rates were measured after 0, 3, 7, 14, 21, 28, 35, 42, 49, 56 and 63 days of soil application, and degradation rates after 0, 7, 28 and 63 days. Although glyphosate mineralization rate was higher in the presence of 1 (one) dose of atrazine when compared with glyphosate alone, no significant differences were found when half or twice the atrazine dose was applied, meaning that differences in glyphosate mineralization rates cannot be attributed to the presence of atrazine. On the other hand, the influence of glyphosate on atrazine mineralization was evident, since increasing doses of glyphosate increased the atrazine mineralization rate and the lowest dose of glyphosate accelerated atrazine degradation.  相似文献

采用外源添加重金属和露天盆栽实验研究了Cd、Ni对含羞草、三叶草根际土壤脲酶、过氧化氢酶和蔗糖酶活性的影响。结果表明,Cd、Ni单一污染条件下,低浓度的Cd、Ni对含羞草组、三叶草组的脲酶、过氧化氢酶有激活作用,高浓度的Cd、Ni对土壤脲酶、过氧化氢酶有一定的抑制作用;对蔗糖酶有较强的抑制作用。在Cd、Ni复合污染条件下,对含羞草组、三叶草组的脲酶、过氧化氢酶和蔗糖酶产生抑制作用;Cd、Ni单一及复合污染对土壤酶活性的抑制大小顺序为:脲酶>蔗糖酶>过氧化氢酶,其中土壤脲酶可以作为Cd、Ni污染的预警指标。含羞草能显著提高土壤脲酶、过氧化氢酶和蔗糖酶的活性,修复能力大于三叶草,在Cd、Ni污染修复方面有很好的应用前景。  相似文献

The rate of degradation of forchlorfenuron, a cytokinin-based plant growth regulator (PGR) was explored in typical grapevine soils of India with simultaneous evaluation of its effect on biochemical attributes of the test soils in terms of the activities of specific soil microbial enzymes. In all the test soils, namely clay, sandy-loam and silty-clay, the dissipation rate was faster at the beginning, which slowed down with time, indicating a non-linear pattern of degradation. Degradation in soils could best be explained by two-compartment 1st + 1st order kinetics with half-life ranging between 4–10 days. The results suggest that organic matter might be playing a major role in influencing the rate of degradation of forchlorfenuron in soil. The rate of degradation in sandy-loam soil was fastest followed by clay and silty-clay soils, respectively. Comparison of the rate of degradation in natural against sterilized soils suggests that microbial degradation might be the major pathway of residue dissipation. Changes in soil enzyme activities as a consequence of forchlorfenuron treatment were studied for extra-cellular enzymes namely acid phosphatase, alkaline phosphatase and β -glucosidase and intracellular enzyme-dehydrogenase. Although small changes in enzyme activities were observed, forchlorfenuron did not have any significant deleterious effect on the enzymatic activity of the test soils. Simple correlation studies between degradation percentage and individual enzyme activities did not establish any significant relationships. The pattern and change of enzyme activity was primarily the effect of the incubation period rather than the effect of forchlorfenuron itself.  相似文献

The potential of five nonionic surfactants, Triton X-100, Brij35, Ethylan GE08, Ethylan CD127, and Ethylan CPG660 for enhancing release of carbaryl and ethion from two long-term contaminated soils was evaluated using the batch method. Incorporation of the surfactants into soils enhanced the release of both pesticides to various extents, which could be related to the type of pesticides and type and the amount of surfactants added. Release of ethion was dramatically enhanced by aqueous concentrations of surfactants above their critical micelle concentration values. This was attributed to solubility enhancement through incorporation of the highly hydrophobic compound within surfactant micelles. A concentration of 10 g L(-1) of various surfactants released >70% of the total ethion from the soil irrespective of the surfactant. For carbaryl, the surfactants were effective at low concentrations and dependence on concentration was lower than in the case of ethion. The ethylan surfactants (GE08, CD127, and CPG660) had a higher potential than Triton X-100 and Brij35 for releasing the pesticides. However, there was still a significant portion of carbaryl (11% of the total) and ethion (17% of the total) left in the soil. Our study also showed that there must be an optimal concentration of each surfactant to maximize the mass transfer of pesticides. At some threshold concentration level, additional surfactant started to inhibit the mass transfer of solute from the soil into the water. The results suggested that surfactants could help remediation of soils polluted by pesticides. The choice of surfactant should be made based on the properties of pesticides.  相似文献

This paper reports the release behavior of two triazines (atrazine and simazine) in stabilised soils from a pesticide-contaminated site in South Australia. The soils were contaminated with a range of pesticides, especially with triazine herbicides. With multiple extractions of each soil sample with deionised water (eight in total), 15% of atrazine and 4% of simazine residues were recovered, resulting in very high concentrations of the two herbicides in leachate. The presence of small fractions of surfactants was found to further enhance the release of the residues. Methanol content up to 10% did not substantially influence the concentration of simazine and atrazine released. The study demonstrated that while the stabilisation of contaminated soil with particulate activated carbon (5%) and cement mix (15%) was effective in locking the residues of some pesticides, it failed to immobilise triazine herbicides residues completely. Given the higher water solubility of these herbicides than other compounds more effective strategies to immobilise their residues is needed.  相似文献

Silver nanoparticles (AgNPs) are well known to have antimicrobial ability, but very little is known about the effect of AgNPs on soil exoenzyme activities, which reflect the potential of a soil to support biochemical processes. This study provides evidence of the inhibitory effects of AgNPs on the activities of soil exoenzymes. Six exoenzymes related to nutrient cycles (urease, acid phosphatase, arylsulfatase, β-glucosidase) and the overall microbial activity (dehydrogenase, fluorescein diacetate hydrolase) were tested in soils treated with AgNPs (1, 10, 100 and 1000 μg g(-1)) and silver ion (0.035, 0.175, 0.525, 1 and 1.5 μg g(-1)). AgNPs were capable of inhibiting the activities of all the exoenzymes tested in this study. Especially, the urease and dehydrogenase activities were significantly related to the presence of AgNPs. The effects of silver ions dissolved from the AgNPs were not significant, indicating the adverse effects caused by AgNPs themselves. This study suggested that AgNPs negatively affect soil exoenzyme activities, with the urease activity especially sensitive to AgNPs.  相似文献

Background Many contaminated sites contain a variety of toxicants. Risk assessment and the development of soil quality criteria therefore require information on the interaction among toxicants. Interactions between heavy metals are relatively well studied, but little is known about those between heavy metals and polycyclic aromatic hydrocarbons (PAHs). Methods 0.1 mg/kg dry soil phenanthrene alone or phenanthrene plus 10 mg/kg cadmium (Cd) were added to soil to determine the influence of phenanthrene on Cd toxicity to soil enzymes (invertase, urease, dehydrogenase and phosphatase) and microorganisms (fungi, bacteria and actinomycete) in paddy soil. Results and Discussion 0.1 mg/kg phenanthrene did not reduce the number of microorganisms. However, the addition of phenanthrene to soil with Cd enhanced the Cd toxicity to soil enzymes and microorganisms. This deleterious effect was seen to mainly affect the growth of fungi and the activity of invertase, urease and dehydrogenase. The order of combined inhibition of Cd and phenanthrene was fungi>bacteria>actinomycete. Conclusion The presence of phenanthrene might enhance the toxicity of Cd to soil microorganisms. Phenanthrene can easily be used by the soil actinomycetes as a source of carbon and energy and the finding may be supportive to the development of bioremediation techniques.  相似文献

The effectiveness of two amendments for the in situ remediation of a Cd- and Ni-contaminated soil in the Louis Fargue long-term field experiment was assessed. In April 1995, one replicate plot (S1) was amended with 5% w/w of beringite (B), a coal fly ash (treatment S1+B), and a second plot with 1% w/w zerovalent-Fe iron grit (SS) (treatment S1+SS), with the aim of increasing metal sorption and attenuating metal impacts. Long-term responses of daily respiration rates, microbial biomass, bacterial species richness and the activities of key soil enzymes (acid and alkaline phosphatase, arylsulfatase, beta-glucosidase, urease and protease activities) were studied in relation to soil metal extractability. Seven years after initial amendments, the labile fractions of Cd and Ni in both the S1+B and S1+SS soils were reduced to various extents depending on the metal and fractions considered. The soil microbial biomass and respiration rate were not affected by metal contamination and amendments in the S1+B and S1+SS soils, whereas the activity of different soil enzymes was restored. The SS treatment was more effective in reducing labile pools of Cd and Ni and led to a greater recovery of soil enzyme activities than the B treatment. Bacterial species richness in the S1 soil did not alter with either treatment. It was concluded that monitoring of the composition and activity of the soil microbial community is important in evaluating the effectiveness of soil remediation practices.  相似文献

Leaching of the strongly sorbing pesticides glyphosate and pendimethalin was evaluated in an 8-month field study focussing on preferential flow and particle-facilitated transport, both of which may enhance the leaching of such pesticides in structured soils. Glyphosate mainly sorbs to mineral sorption sites, while pendimethalin mainly sorbs to organic sorption sites. The two pesticides were applied in equal dosage to a structured, tile-drained soil, and the concentration of the pesticides was then measured in drainage water sampled flow-proportionally.The leaching pattern of glyphosate resembled that of pendimethalin, suggesting that the leaching potential of pesticides sorbed to either the inorganic or organic soil fractions is high in structured soils. Both glyphosate and pendimethalin leached from the root zone, with the average concentration in the drainage water being 3.5 and 2.7 μg L−1, respectively. Particle-facilitated transport (particles >0.24 μm) accounted for only a small proportion of the observed leaching (13-16% for glyphosate and 16-31% for pendimethalin). Drain-connected macropores located above or in the vicinity of the drains facilitated very rapid transport of pesticide to the drains. That the concentration of glyphosate and pendimethalin in the drainage water remained high (>0.1 μg L−1) for up to 7 d after a precipitation event indicates that macropores between the drains connected to underlying fractures were able to transport strongly sorbing pesticides in the dissolved phase. Lateral transport of dissolved pesticide via such discontinuities implies that strongly sorbing pesticides such as glyphosate and pendimethalin could potentially be present in high concentrations (>0.1 μg L−1) in both water originating from the drainage system and the shallow groundwater located at the depth of the drainage system.  相似文献
