This study compares the accumulation of Cr(VI) and biochemical changes (total chlorophyll, carotenoid, protein, malondialdehyde (MDA) and cysteine contents) and roles of antioxidant enzymes (superoxide dismutase (SOD), guaiacol peroxidase (GPX), ascorbate peroxidase (APX)) in tolerance to metal induced stress in Cucumis utillissimus L. grown in Cr contaminated soil (CS) with garden soil (GS). Furthermore, Cr bioavailability was enhanced by ethylene diamine tetra-acetic acid (EDTA) addition to the soil to forecast the plant’s accumulation pattern at elevated Cr environment. Accumulation of Cr in the leaves of the plant increased with increase in substrate metals concentration. It further increased with the addition of EDTA by 1437% and 487% in GS and CS, respectively at the highest treatment level. The lipid peroxidation increased proportionately with increase in Cr accumulation in the leaves. All the activity of antioxidant enzymes (SOD, GPX and APX) and the level of cysteine increased with dose dependant manner. SOD and cysteine were observed to be higher in the GS than in CS, but APX and GPX were found to be higher in CS than in GS. The increase in GPX and APX activities with the increase in Cr concentration could be assumed that these two enzymes have a major role in the defense mechanism towards stress induced by Cr in C. utillissimus.  相似文献

In order to understand its response towards nickel stress,watercress(Nasturtium officinale R.Br.) was exposed to nickel(1-25 mg/L) for 1,3,5 and 7 days.The accumulation and translocation of nickel were determined and the influence of nickel on biomass,protein content and enzymatic antioxidants was examined for both roots and leaves.It was determined that N.officinale could accumulate appreciable amounts of Ni in both roots and leaves.Nickel accumulated particularly in the roots of plants.Biomass increased at low nickel concentrations but certain measurable change was not found at high concentrations.Under stress conditions the antioxidant enzymes were up-regulated compared to control.An increase in protein content and enzyme activities was observed at moderate exposure conditions followed by a decline at both roots and leaves.The maximum enzyme activities were observed at different exposure conditions.Our results showed that N.officinale had the capacity to overcome nickel-induced stress especially at moderate nickel exposure.Therefore,N.officinale may be used as a phytoremediator in moderately polluted aquatic ecosystems.  相似文献

One purpose of this research is to present accumulation of cadmium (Cd) and copper (Cu) by female Oxya chinensis (Orthopera: Acridoidea) in a simulated soil-plant-insect ecosystem treated with Cd. Fourth-instar nymphs of O. chinensis had been fed on wheat (Triticurn aestivum) seedlings contaminated with Cd and Cu for one month. In the ecosystem, the Cd concentration in wheat seedlings rose greatly with the increasing of Cd in the soil, but the Cu concentration in wheat seedlings was not found elevated. There was a highly significant difference(P〈0.05) in Cd concentrations of wheat seedlings and not any significant difference(P〉0.05) in Cu concentrations of wheat seedlings. The Cd and Cu concentration in different body part-head, thorax, abdomen, and hind femur, varied under different Cd concentrations in soil. There were significant differences (P〈0.05) in the four parts of Cd and Cu accumulations with all treatments. The order of Cd accumulation was thorax 〉abdomen 〉head 〉hind femur and the Cu was abdomen 〉thorax 〉 head〉hind femur. The results indicated that Cd and Cu were accumulated from the soil to grasshoppers through the plant; that is to say, Cd and Cu in environment could be transported to animal or human via food chain.  相似文献

The effects of phenanthrene (Phe) on hepatosomatic index (HSI) and hepatic enzymatic activities in hybrid tilapia (Oreochromis niloticus O. aureus ) were investigated via the static freshwater exposure at dosage of 50, 100, and 400 g/L for 4–14 d. Compared with the control group, HSI was significantly decreased (P < 0.05) at 400 g/L at day 14. Increased enzymatic activities (P < 0.05) were observed for catalase (CAT), glutathione peroxidase (GPx) and superoxide dismutase (SOD) at either 100 or 400 g/L at day 8 and 14, as well as for CAT at 50 g/L at day 14, except for GPx at 400 g/L at day 8. Ethoxyresorufin O-deethylase (EROD) activity was significantly increased (P < 0.05) at all dosage at day 4 as well as at 50 g/L at day 8, but significantly decreased at either 100 or 400 g/L at day 14 (P < 0.05). Glutathione-S-transferase (GST) activity was not a ected. The results suggest that CAT, GPx, SOD and EROD, as well as HSI in tilapia may be used as the biomarkers or indexes for evaluating or monitoring the pollution of polycyclic aromatic hydrocarbons (PAHs) such as Phe.  相似文献

Investigation on Cr( Ⅵ ) reduction was conducted using Pseudomonas aeruginosa. The study demonstrated that the Cr(Ⅵ) canbe effectively reduced to Cr( Ⅲ ) by Pseudomonas aeruginosa. The effects of the factors affecting Cr( Ⅵ ) reduction rate including carbon source type, pH, initial Cr(Ⅵ) concentration and amount of calls inoculum were thoroughly studied. Malate was found to yield maximum biotransformation, followed by succinate and glucose, with the reduction rate of 60.86%, 43. 76% and 28.86% respectively. The optimum pH for Cr( Ⅵ ) reduction was ?.0, with reduction efficiency of 61.71 % being achieved. With the increase of initial Cr(Ⅵ) concentration,the rate of Cr(Ⅵ) reduction decreased. The reduction was inhibited strongly when the initial Cr(Ⅵ) concentration increased to 157 mg/L. As the amount of cells inoculum increased, the rate of Cr( Ⅵ ) reduction also increased. The mechanism of Cr( Ⅵ ) reduction and final products were also analysed. The results suggested that the soluble enzymes appear to be responsible for Cr (Ⅵ) reduction by Pseudomonas aeruginosa, and the reduced Cr( Ⅲ ) was not precipitated in the form of Cr(OH)3.  相似文献

To assess the sublethal toxicity of the herbicide acetochlor to earthworms and to find out biomarkers possible inducted under acetochlor exposure, Eiseniafetida was exposed to artificial soils supplemented with different concentrations of acetochlor(5, 10, 20, 40 and 80 mg/kg soil). Effects of the acetochlor on cytochrome P450 monooxygenases p-nitroanisole O-demethylase(ODM), aldrin epoxidase(AE) and glutathione-S-transferases (GSTs) activities were determined. The results revealed cytochrome P450 monooxygenases were elevated with increasing concentrations of acetochlor, and the AE activity increased significantly compared with control at the concentration of 80 mg/kg (P〈0.05). However, ODM activity from E. fetida was not induced significantly by acetochlor at all treatments(P〉0.05). Sodium dodecyl sulfate-polyacryamide gel electrophoresis(SDS-PAGE) showed that one protein band was visualized and no evident differences were found in protein profiles between treatments and control. The GST activity increased significantly with longer duration(P〈0.05) and increasing concentrations of acetochlor exposure(P〈0.05). This study showed that the monooxygenases and GSTs activities in E. fetida could be induced by acetochlor, and thus, the AE and GST could be used in sublethal assays for soil contamination surveys and GST could be used as biomarkers of acetochlor exposure in E. fetida.  相似文献

Effects of cadium, zinc and lead on soil enzyme activities   总被引:5,自引:0,他引:5  
Heavy metal (HM) is a major hazard to the soil-plant system. This study investigated the combined effects of cadium (Cd), zinc (Zn) and lead (Pb) on activities of four enzymes in soil, including calatase, urease, invertase and alkalin phosphatase. HM content in tops of canola and four enzymes activities in soil were analyzed at two months after the metal additions to the soil. Pb was not significantly inhibitory than the other heavy metals for the four enzyme activities and was shown to have a protective role on calatase activity in the combined presence of Cd, Zn and Pb; whereas Cd significantly inhibited the four enzyme activities, and Zn only inhibited urease and calatase activities. The inhibiting effect of Cd and Zn on urease and calatase activities can be intensified significantly by the additions of Zn and Cd. There was a negative synergistic inhibitory effect of Cd and Zn on the two enzymes in the presence of Cd, Zn and Pb. The urease activity was inhibited more by the HM combinations than by the metals alone and reduced approximately 20%--40% of urease activity. The intertase and alkaline phosphatase activities significantly decreased only with the increase of Cd concentration in the soil. It was shown that urease was much more sensitive to HM than the other enzymes. There was a obvious negative correlation between the ionic impulsion of HM in soil, the ionic impulsion of HM in canola plants tops and urease activity. It is concluded that the soil urease activity may be a sensitive tool for assessing additive toxic combination effect on soil biochemical parameters.  相似文献

It has been reported that there is an interaction between Benzo[a]pyrene (BaP), a widespread carcinogenic polycyclic aromatic hydrocarbon, and tributyltin (TBT), an organometal used as an antifouling biocide. This study was therefore designed to examine the potential in vivo influence of BaP, TBT and their mixture on splenic antioxidant defense systems of Sebastiscus marmoratus. The fish were exposed to water containing environmentally relevant concentrations of BaP, TBT and their mixture. Spleens were collected for biochemical analysis after exposure for 7, 25, 50 d and after recovery for 7, 20 d. Cotreatment with BaP and TBT for 7 d potentiated the induction of glutathione peroxidase (GPx) activity by BaP or TBT alone. The cotreatment for 25 and 50 d resulted in inhibition of GPx activity, which was similar to the effect of TBT. Splenic glutathione S-transferase (GST) activities were significantly elevated in S. marmoratus exposed to BaP starting from 7 d and remained high up to 25 d. However, no further activity change was found with prolonged exposure. Cotreatment of BaP and TBT primarily inhibited the GST activity, which was similar to the effect of TBT. Cotreatment with BaP and TBT for 25 or 50 d potentiated the depletion of GSH (glutathione) by BaP or TBT alone. MDA (malondialdehyde) contents in spleen of S. marmoratus were not significantly altered compared with the control during the test period. Spleen, as an immune organ, is sensitive to exposure of BaP or TBT. It should have an effective mechanism to counteract oxidative damage. Antioxidative defense systems in spleen of S. marmoratus should be considered as potential biomarkers. Short-term exposure of BaP or TBT could result in induction of antioxidant defense system. A significant decrease of these indices, such as GSH, GST, GPx might indicate more severe contamination.  相似文献

The toxic effects of Cr(Ⅵ) on antioxidant enzymes of Oxya chinensis(Orthoptera: Acridoidae) were determined. Changes in the activities of the antioxidant enzymes superoxide dismutase(SOD), catalase(CAT), and guaiacol peroxidase(GPx) were measured in O. chinensis insects injected with Cr(Ⅵ). Fifth-nymphs of O. chinensis insects were injected with Cr(Ⅵ) with different concentrations (0, 75, 150, 225, 300, 375, 450 mg/kg of body weight). The results showed that Cr(Ⅵ) led to the change of SOD, CAT, and GPx activities at different concentrations, which revealed that: (1) The oxidative stress of SOD increased with the increase of Cr (Ⅵ)concentration. (2) With the increase of Cr (Ⅵ) concentrations, CAT activities for females increased at lower concentrations, but decreased at higher concentration range, which indicated that antioxidant system of O. chinensis was not influenced by the presence of Cr (Ⅵ). A very similar response to Cr(Ⅵ) effect for males indicated that Cr(Ⅵ) concentrations were not high enough to damage O. chinensis in terms of CAT. (3) The GPx activity for females increased in all treatments, which revealed that the damage power of Cr(Ⅵ)was increased with the increase of Cr(Ⅵ) concentrations in terms of GPx, but the effect was not so remarkable. There was not a consistent trend of GPx activities for males in all treatments of Cr(Ⅵ). Cr(Ⅵ)-induced changes in antioxidant enzymes were different for SOD, CAT and GPx, of which the tendency was that activities generally changed with increase of concentrations of Cr(Ⅵ) suggesting SOD, CAT, and GPx could serve as indices of oxidative stress to some extent.  相似文献

研究了苯并[a]芘暴露对鲫鱼(Carassius auratus)的几种生物标志物的影响.在鲫鱼接受3个不同质量分数苯并[a]芘(BaP)的腹腔注射14 d后,分析了其肝脏7-乙氧基-3-异吩噁唑酮-脱乙基酶(EROD)活性,以及超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)等抗氧化酶活性和红细胞核异常(NA)率变化.结果显示:w(BaP)为1 mg/kg处理组的EROD活性显著升高(P<0.001).w(BaP)为0.1和1 mg/kg处理组SOD活性显著低于对照组(P<0.001),在暴露结束时,w(BaP)为1 mg/h处理组CAT活性显著低于对照组(P<0.01),而w(BaP)为0.1和1 mg/kg处理组GPx活性被显著抑制(P<0.001).表明BaP对EROD活性有诱导作用,而对SOD,CAT和GPx等抗氧化酶活性的抑制作用明显.所有处理组NA率与对照组均无显著差异.因此,鲫鱼EROD,SOD,CAT和GPx等酶活性的改变适于作为生物标志物,可以用来显示暴露于多环芳烃有机污染物鱼类的生活状态.   相似文献

为研究城市污水处理厂(STPs)尾水是否会引起海洋生物的氧化逆境,将取自青岛市李村河污水处理厂的尾水用海水稀释到不同浓度(0%、1%、5%、10%、20% V/V), 对文蛤(Meretrix meretrix)暴露培养15d, 分析其内脏中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)、谷胱甘肽还原酶(GR)的活性. 结果表明:SOD活性随尾水暴露时间延长基本表现为“抛物线型”的响应特征,体积比10%、20%的尾水暴露6~12d期间,文蛤体内SOD活性均受到诱导,最大增幅为15.88%;CAT和GPx对尾水的主要响应特征是抑制效应,分别在暴露9d、12d时最为明显,最大抑制率分别为64.38%和52.39%;较高浓度(20%)的尾水能够持续抑制GR活性,而其他处理组中,GR对尾水表现为“抛物线型”的响应特征,即:暴露3d或6d时活性增加,而后显著降低.GR活性的最大诱导出现在暴露3d和6d时,分别比对照组上升81.47%和80.91%.研究结果进一步表明STPs不能有效降解有害化学物质以消除尾水的毒性, 文蛤内脏SOD、CAT、GPx和GR活性能够敏感指示尾水诱导的氧化逆境.  相似文献

选择幼龄鲫鱼为材料 ,研究锌(Zn2+)及其配合物(Zn-EDTA)低浓度长期暴露(40d)对鲫鱼(Carassius auratus)肝脏锌富集和抗氧化防御系统的影响 .结果表明 ,鲫鱼肝脏中锌的积累量随锌暴露浓度的升高而增加,且Zn2+处理明显高于Zn EDTA处理 .2种形态锌对鲫鱼肝脏超氧化物歧化酶 (SOD)、过氧化氢酶 (CAT)和谷胱甘肽过氧化物酶 (GPx)均表现出抑制作用 ,其中Zn2+暴露浓度与酶活性之间存在良好的剂量效应关系;Zn2+在低浓度时对谷胱甘肽转硫酶(GST)有诱导作用 ,高浓度时为抑制作用 .Zn EDTA对GST活性始终产生抑制作用 .CAT与GPx对锌暴露比GST和SOD更为敏感 ,适合作为水环境锌污染的早期监测指标 .  相似文献

北部湾文蛤对石油烃和多氯联苯的氧化应激响应   总被引:1,自引:0,他引:1  
为探讨海域现场双壳类动物对有机污染物积累的抗氧化响应,在北部湾潮间带11个采样点采集文蛤(Meretrix meretrix)样品,测定其软组织中w(TPHs)(TPHs为石油烃)、w(PCBs)(PCBs为多氯联苯)以及鳃、内脏中GSH(还原型谷胱甘肽),GSSG(氧化型谷胱甘肽)、SOD(超氧化物歧化酶)、CAT(过氧化氢酶)、GPx(谷胱甘肽过氧化物酶)、GST(谷胱甘肽转硫酶)、TBARS(硫代巴比妥酸反应物)7种氧化应激物的水平,分析其空间分布特征,并进行有机污染物含量与氧化应激物响应值之间的相关性分析. 结果表明:文蛤软组织中w(TPHs)和w(PCBs)分别为78.22~300.71μg/g和4.23~26.68ng/g,最大值均出现在S10采样点(防城港西湾);内脏中CAT活性较高,其他氧化应激物均在鳃中有较高水平. 与对照采样点S1(湛江流沙湾)相比,S2、S3、S6、S10、S11等5个采样点文蛤组织内SOD、CAT、GST、GPx、GSH水平较低;大多数采样点文蛤鳃中w(GSSG)较低,而S3、S4、S6、S7、S9、S11等6个采样点文蛤内脏中w(GSSG)较高;大多数采样点文蛤组织中TBARS含量较低,表明其抗氧化防御机能尚未丧失. TPHs显著抑制文蛤鳃中的w(GSSG) (R=-0.64,P<0.05),PCBs显著抑制文蛤鳃中的GPx(R=-0.72,P<0.05)和GST(R=-0.72,P<0.05)的活性,表明w(GSSG)及GPx和GST的活性可作为指示北部湾有机污染的生物标志物.   相似文献

通过人工土壤法,研究0.1, 1, 10, 100, 1000μg/kg类二 多氯联苯暴露浓度对赤子爱胜蚓 (Eisenia fetida)抗氧化酶(SOD酶、CAT酶和GST酶)、抗氧化分子(GSH)、氧化损伤物(MDA)等生理活性指标在第2, 7, 14, 28d时的影响.结果显示,类二 多氯联苯刺激赤子爱胜蚓生理活性显著增加(P<0.05),可诱导蚯蚓抗氧化系统应答启动;不同暴露时间下,受试物浓度与蚯蚓生理活性的相关性分析显示,蚯蚓不同生理活性指标对受试物的最佳响应时间不同, Cu-Zn SOD响应最早(7d), SOD, CAT, GSH, MDA次之(14d), GST响应最迟(28d);在最佳响应时间下,受试物暴露浓度对数与生理活性值对数均存在显著的剂量-效应关系(P <0.001)  相似文献

采用水培试验,研究了镉胁迫下喷施细胞分裂素类物质6-苄氨基嘌呤(6-BA)和激动素(KT)对玉米幼苗生长、叶片叶绿素含量、脯氨酸含量、植株镉含量和活性氧清除酶活性的影响.结果表明,与单纯镉胁迫相比,喷施6-BA和KT提高了玉米幼苗的生物量、根和地上部镉含量、叶绿素和脯氨酸含量,降低了叶片丙二醛(MDA)含量,促进了叶片过氧化物酶(POD)和过氧化氢酶(CAT)活性的升高,但对超氧化物歧化酶(SOD)活性的影响不明显.说明细胞分裂素类物质6-BA和KT可缓解镉胁迫对玉米幼苗的伤害,其机制可能与细胞分裂素物质能维持叶绿素稳定、刺激细胞内H2O2清除酶类活性及促进脯氨酸积累有关.当6-BA浓度在0~20mg·L-1之间时,其对玉米幼苗镉胁迫的缓解效应随喷施浓度的提高而增强,而KT的最适喷施浓度为70mg·L-1.  相似文献

采用亚急性毒性实验方法,研究了铅(Pb2+)对河南华溪蟹(Sinopotamon henanense)卵巢组织抗氧化酶活性与脂质过氧化产物的影响.实验设置了5个Pb2+浓度组(3.675、7.35、14.70、29.40和58.80mg·L-1)和1个空白对照组,在3个染毒时间(5、10和15d)下对超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)活力和丙二醛(MDA)含量进行了测定.结果显示,在不同浓度和不同暴露时间下,铅对河南华溪蟹卵巢SOD、CAT、GPx活力及MDA含量均有明显的影响.SOD、CAT活力表现为低浓度诱导激活,高浓度抑制,即先升后降的变化规律,GPx活力在5d和10d时表现出先升后降的趋势,在15d时活力不断下降,且SOD在10d、14.70mg·L-1Pb2+浓度组,CAT在5d、7.35mg·L-1Pb2+浓度组,GPx在10d、3.675mg·L-1Pb2+浓度组分别升至峰值,而卵巢MDA含量随染毒时间的延长和染毒浓度的升高逐渐增多,且在15d、58.80mg·L-1Pb2+浓度组中较对照组差异极显著。结果表明,Pb2+暴露对河南华溪蟹卵巢产生了明显的氧化胁迫作用,且抗氧化酶和脂质过氧化物水平可以作为重金属对水生生物毒理学研究的生物标志物.  相似文献

为评价十溴联苯醚(BDE-209)对植物的生态毒性效应,探明植物抵抗BDE-209胁迫的反应机制,文章选用紫花苜蓿作为受试植物,考察了不同浓度BDE-209污染土壤对紫花苜蓿种子萌发及幼苗生长的影响,并对幼苗体内的抗氧化酶活性(POD、CAT、SOD)及可溶性蛋白质(SP)对污染物胁迫的响应进行了研究,同时还对植株的根、叶片进行扫描电镜观察。结果表明,BDE-209胁迫处理与各项生长指标呈显著负相关,抑制作用强度依次为根长>鲜重>芽长>发芽率。植株体内的抗氧化酶活性和可溶性蛋白含量随污染物浓度的增加不断上升,符合线性关系,POD、CAT、SOD的活性最大可高达对照的3倍、1.77倍和2.81倍,可溶性蛋白含量可达到对照的1.89倍。扫描电镜结果显示,随着污染物浓度的提高,植物根部发根增多,叶片上气孔密度增大,气孔开度变小。  相似文献

以栉孔扇贝(Chlamys farreri)为受试生物,研究了栉孔扇贝对8:2氟调聚羧酸(8:2FTCA)的代谢转化特征以及代谢过程中靶器官的氧化应激响应.结果发现,栉孔扇贝可将8:2FTCA转化为8:2氟调聚不饱和酸(8:2FTUCA)、7:3氟调聚羧酸(7:3FTCA)、全氟辛酸(PFOA)、全氟壬酸(PFNA)和全氟庚酸(PFHpA)等代谢产物.栉孔扇贝鳃和肝脏中代谢产物总量最高,为8:2FTCA的主要代谢靶器官.与本课题组前期虾夷扇贝的相关研究相比,8:2FTCA在栉孔扇贝与虾夷扇贝体内的生物转化行为主要有3方面相似之处:检测到的代谢产物相同、代谢靶器官相同以及鳃是最终代谢产物PFOA生成和蓄积的主要场所;不同之处主要体现在栉孔扇贝中代谢产物以7:3FTCA占比较高,虾夷扇贝中则是PFOA占比较高.同时在8:2FTCA暴露过程中,栉孔扇贝靶器官的关键抗氧化酶出现了一定的应激效应.丙二醛(MDA)和谷胱甘肽(GSH)等氧化应激指标以及谷胱甘肽过氧化物酶(GSH-Px),超氧化物歧化酶(SOD)和过氧化氢酶(CAT)等抗氧化酶系出现不同程度变化:GSH-Px在整个代谢转化过程中呈现抑制效应;SOD,CAT活性在鳃中呈诱导效应,而在肝脏中呈现明显的剂量-效应关系,低剂量组中均呈抑制效应,高剂量组中大部分时间呈诱导效应.GSH和MDA含量不仅表现出剂量-效应关系,组织间也存在差异.暴露实验结束后,生理指标均有不同程度恢复.  相似文献
