Nine populations of Aphanius fasciatus Nardo, 1827 from the central Mediterranean were analysed by examining the mitochondrial control region and the morphology of the bony elements of the skull and vertebral column, to study the degree of intraspecific differentiation of A. fasciatus considering the level of isolation of the different populations and the palaeogeographic history of the central Mediterranean area. Both the molecular and morphological analyses differentiate between the populations, even if the topologies of the two trees are different. Molecular and osteological investigations have consistently demonstrated a well-supported differentiation of the south-eastern Sicilian populations both within the same group (Tigano et al. in Ital J Zool 71:1124–1133, 2004a; Tigano et al. in Abstract volume XI European Congress of Ichthyology, Tallin, Estonia, 2004b), and from the populations from western Sicily, Tunisia and the island of Malta. The molecular results show that the nine populations are characterised by haplotypes that are well defined in relation to a probably limited gene flow; while, as regards the morphological data the differentiation found could be explained in terms of the geographic isolation of the various populations, although the influence of environmental factors, which differ greatly between the various sites where the populations live, cannot be ruled out.  相似文献

Molecular systematic analyses of marine taxa are crucial for recording ocean biodiversity, so too are elucidation of the history of population divergence and the dynamics of speciation. In this paper we present the joined phylogeography of the calanoid copepod Calanus helgolandicus (Claus 1863) from the North East (NE) Atlantic and the Adriatic Sea and the closely related C. euxinus (Hulsemann 1991) from the Black Sea based on sequences of a mitochondrial Cytochrome Oxidase subunit I (COI) fragment. Coalescent-based Bayesian methods and minimum spanning networks are used to reconstruct the history of population divergence. Our results reveal that copepod populations from all three basins share a great number of haplotypes and demonstrate a close genetic affinity of C. euxinus with C. helgolandicus. The data do not support significant genetic structuring among samples within seas. Coalescent analyses suggest divergences between NE Atlantic, Mediterranean, and Black Sea populations dating back to the middle Pleistocene, with the NE Atlantic–Mediterranean divergence being the earliest and the Mediterranean–Black Sea divergence the most recent. These middle Pleistocene dates are much older than the estimated dates of colonisation of the Mediterranean and Black Seas based on paleoclimatic scenarios. Our results do not rule out that the assumed colonisations took place but they indicate that the populations colonising the Mediterranean and the Black Sea were already, and have since remained, diverged. The chaetognath Sagitta setosa, which has a comparable distribution pattern and feeds upon the copepods, provides a unique opportunity to compare phylogeographic patterns and distinguish among alternative hypotheses. The dates produced in this paper are in agreement with those estimated elsewhere for S. setosa. We propose that a great deal of the genetic make-up of marine planktonic populations comprises divergences that date back to long before the last glacial maximum. We consider questions on the taxonomic status of C. euxinus to remain open. However, its high genetic affinity to the C. helgolandicus calls for further investigation.  相似文献

The genetic structure of the flounders Platichthys flesus L. and P. stellatus Pallas was investigated on different spatial scales through analysis of allozyme variation at 7 to 24 polymorphic loci in samples collected from different regions (Baltic Sea, North Sea, Brittany, Portugal, western Mediterranean, Adriatic Sea, Aegean Sea and Japan) in 1984 to 1987. No geographic variation was evident within a region. Some pattern of differentiation by distance was inferred within the Atlantic, while the Mediterranean comprised three geographically isolated populations and was itself geographically isolated from the Atlantic (fixed allele differences at up to three loci were found among P. flesus populations from the Atlantic, the western Mediterranean, the Adriatic Sea, the Aegean Sea and also P. stellatus from the coast of Japan). Sea temperature during the reproductive period probably acts as a barrier to gene flow between populations. Genetic distances among European flounder populations (P. flesus) were higher than, or of the same magnitude as, the genetic distance between Pacific (P. stellatus) and European flounder populations, suggesting that P. flesus is paraphyletic and/or there is no phylogenetic basis to recognising P. stellatus as a different species. The divergence between P. flesus and P. stellatus was thus inferred to be more recent than the divergence between the present P. flesus populations from the NE Atlantic and eastern Mediterranean. The eastern Mediterranean populations are thought to originate from the colonisation of the Mediterranean by a proto-P. flesus/P. stellatus ancestor, whereas the present western Mediterranean population has undergone a more recent colonisation event by P. flesus. Patterns of mitochondrial DNA variation, established on a smaller array of P. flesus samples, were in accordance with the geographic patterns inferred from the allozyme survey. In addition, they supported the hypothesis of a two-step colonisation of the western Mediterranean. These results contribute to our understanding of the biogeography of the Mediterranean marine fauna, especially the group of boreal remnants to which P. flesus belongs. The purpose of this study was to determine the phylogeographic structure of the brackish-hypersaline cyprinodont fish Aphanius fasciatus (Valenciennes, 1821), using sequencing and RFLP analysis of a 1,330 bp mitochondrial DNA segment containing part of the 16S rRNA gene as well as the genes for tRNA-Leu, NADH subunit 1 and tRNA-Ile. Individuals were collected from 13 different sites in Greece and Turkey, while seven published A. fasciatus sequences were also included to cover the area of distribution of the species. Pairwise sequence divergence values ranged from 0 to 4.51%. Congruent phylogenies were recovered with maximum likelihood, maximum parsimony and neighbour-joining methods. All analyses revealed two main groups. The first group consists of populations from almost all localities that drain into the Aegean Sea. The second group comprises the remaining population samples, which in some cases seem to consist of population-specific subgroups. Our results show that vicariant events have predominantly affected the evolution of A. fasciatus, with the Messinian salinity crisis having shaped the present genetic structure of its populations. Additionally, the life-history traits of the species, which determine a low potential for dispersal, coupled with the typical fragmentation of brackish-hypersaline water habitats have led to a high degree of isolation of A. fasciatus populations, even at restricted spatial scales. Analysis of the partitioning of the total amount of polymorphism with analyses of molecular variance (AMOVA) gave a value of F ST = 84.6%. Potential conservation policies concerning A. fasciatus should also consider the low-genetic variability in the majority of its populations and the presence of fixed haplotypes in some of them.  相似文献

Spatial and temporal population genetic structures of the common sole, Solea solea, were studied in Northeastern Atlantic and Mediterranean Sea populations, using three polymorphic exon-primed intron-crossing (EPIC) markers. Results demonstrated significant multilocus differentiation among Eastern Mediterranean and a group composed by Western Mediterranean and Atlantic populations (θ = 0.150, P < 0.001), but also suggested unrecorded genetic differentiation of the Adriatic Sea population. No pattern of isolation-by-distance was recorded across the range covered by sampling, from the Kattegat to the Aegean Sea. Conversely to genetically structured Mediterranean populations, Atlantic populations ranging from Denmark to Portugal could be considered as representative of the same panmictic unit (θ = 0.009, not significant). Results further demonstrated stability of multilocus genetic structure among temporarily replicated cohort samples [0+, 1+, subadults] from several coastal and estuarine locations from Bay of Biscay, excepted for the amylase locus Am2B3-2 at one location (Pertuis d’Antioche). Despite coherence of such observed patterns of multilocus differentiation with previous allozymic surveys in sole, and with patterns generally obtained for other marine fish species, single-locus results from EPICs indicated divergent coalescence schemes supporting a complex response to ecology and history of sole’s populations. Results stress the use of nuclear genes such as EPIC markers to investigate population structure, but also historical, demographic, and possibly selective processes in marine fishes.  相似文献

Sequence variation in the mitochondrial control region was studied in the Mediterranean rainbow wrasse (Coris julis), a species with pronounced pelagic larval phase inhabiting the Mediterranean Sea and the adjacent coastal eastern Atlantic Ocean. A total of 309 specimens from 19 sampling sites were analysed with the aim of elucidating patterns of molecular variation between the Atlantic and the Mediterranean as well as within the Mediterranean Sea. Phylogeographic analyses revealed a pronounced structuring into a Mediterranean and an Atlantic group. Samples from a site at the Moroccan Mediterranean coast in the Alboran Sea showed intermediate frequencies of “Mediterranean” and “Atlantic” haplotypes. We recognised a departure from molecular neutrality and a star-like genealogy for samples from the Mediterranean Sea, which we propose to have happened due to a recent demographic expansion. The results are discussed in the light of previous studies on molecular variation in fish species between the Atlantic and the Mediterranean and within the Mediterranean.  相似文献

This paper reports data on 28 allozyme loci in wild and artificially reared sea bass (Dicentrarchus labrax) samples, originating from either coastal lagoon or marine sites in the Mediterranean Sea. F ST analysis (θ estimator) indicated strong genetic structuring among populations; around 34% of the overall genetic variation is due to interpopulation variation. Pairwise θ estimates showed that, on average, the degree of genetic structuring was much higher between marine populations than between samples from lagoons. Six polymorphic loci showed differences in allele frequencies between marine and lagoon samples. Multivariate analyses of individual allozymic profiles and of allele frequencies suggested that different arrays of genotypes prevail in lagoons compared to marine samples, particularly at those loci that, on the basis of previous acclimation experiments, had been implicated in adaptation to freshwater. On the other hand, variation at “neutral” allozyme loci reflects to a greater extent the geographic location of populations. Allozyme differentiation was also studied in a D. labrax population from the Portuguese coast. Average genetic distance between this population and the Mediterranean populations was quite high (Nei's D = 0.236) and calls into question the taxonomic status of the Portuguese population. Finally, genetic relationships between D. labrax and D. punctatus were evaluated. Average Nei's D was 0.648, revealing high genetic differentiation between the two species, even for two sympatric populations of these species in Egypt; thus gene flow was not indicated between species. The European Raja clavata and the South African R. straeleni are marine skates which exhibit highly conserved morphological and ecological traits. Owing to this, taxonomic and evolutionary relationships between the two taxa have not yet fully elucidated. Here, we have tested the hypothesis that restricted gene flow and genetic divergence between these taxa might be associated with climatic/oceanographic discontinuities by surveying genetic variation in ten geographical samples at control region (CR) and amplified fragment length polymorphism (AFLP) loci. The clustering of CR haplotypes in two reciprocally monophyletic clades consistent with taxon zoogeography and the significant AFLP F values between the European and South African populations indicated the two taxa as recently diverged peripatric sibling species. Within each species, significant spatial genetic heterogeneity among samples at both markers revealed population structuring. We argued that structured populations and isolated sibling species might represent two stages of geographical speciation.  相似文献

Because marine species respond differentially to factors governing survival and gene flow, closely related taxa may display dissimilar phylogeographic histories. New data for the patchily distributed gastropod Nassarius nitidus throughout its Atlantic–Mediterranean range (collected during 2008 and 2009) were used to investigate its phylogeography and recent demography. Results based on mitochondrial COI sequences of 422?N. nitidus individuals from 15 localities revealed contrasting phylogeographic and demographic patterns among N. nitidus populations from each basin. Data suggest the existence of two glacial refugia, one in the Atlantic, around the Iberian Peninsula, and the other in the Paleo-Mediterranean Sea (Adriatic). Bayesian skyline reconstructions suggest that the Adriatic population of N. nitidus remained largely unaffected by the Last Glacial Maximum (LGM), whereas the Iberian Atlantic region experienced dramatic exponential growth after its conclusion. Contemporary North Sea populations of N. nitidus are the endpoint of a leading-edge recolonization process from a southern position. Additionally, a reanalysis of pre-existing material for the continuously distributed close congener N. reticulatus was used to compare both species in the late histories. In contrast to N. nitidus, N. reticulatus prospered during the LGM and experienced an earlier Atlantic expansion during the previous interglacial period. Despite similar life history and dispersal potential, the results here presented suggest that subtle differences in microhabitat requirements between the two species have had important consequences for their particular distribution in response to glacial events.  相似文献

The analysis of mitochondrial DNA in loggerhead sea turtles (Caretta caretta) from eight foraging grounds in the Mediterranean and the adjoining Atlantic revealed deep genetic structuring within the western Mediterranean. As a consequence, the foraging grounds off the North-African coast and the Gimnesies Islands are shown to be inhabited mainly by turtles of the Atlantic stocks, whereas the foraging grounds off the European shore of the western Mediterranean are shown to be inhabited mainly by turtles from the eastern Mediterranean rookeries. This structuring is explained by the pattern of sea surface currents and water masses and suggests that immature loggerhead sea turtles entering the western Mediterranean from the Atlantic and the eastern Mediterranean remain linked to particular water masses, with a limited exchange of turtles between water masses. As the north of the western Mediterranean comprises mostly individuals from the highly endangered eastern Mediterranean rookeries, conservation plans should make it a priority to reduce the mortality caused by incidental by-catch in these areas.  相似文献

Understanding the evolutionary processes from recent demographic history is especially difficult for interstitial organisms due to their poorly known natural history. In this study, the genetic variation and population history of the four Ototyphlonemertes (Diesing in Sitz ber Math Nat Kl Akad Wiss Wien 46:413–416, 1863) species were evaluated from samples collected along the Brazilian coast (between 27°31′S and 13°00′W) in 2006. The mitochondrial region cytochrome c oxidase subunit 3 (COX3) is analyzed to assess the genetic variation of these dioecious species. Although these species have a sympatric distribution along the coast, our data suggest that their levels of differentiation and their demographic histories differ sharply. There is strong evidence of gene flow among demes in O. erneba and O. evelinae, and their level of structuring is much lower than for the other two species. Indeed, the COX3 fragment reveals cryptic lineages in O. lactea and O. parmula. The results seem to contradict the high genetic structuring and low intrapopulational variability expected with the ecological constriction and habitat discontinuity faced by these organisms, meaning that there might be gene flow among populations or their dispersal capability has been underestimated.  相似文献

A large macrogeographic differentiation has been observed among Sotalia guianensis populations along the South American coast. However, no genetic structure has been detected so far in closely distributed populations of this species, even though it has been observed in other cetaceans. Here, we examined the fine scale population structure for the largest populations of S. guianensis inhabiting Sepetiba and Paraty embayments at the south-eastern coast of Rio de Janeiro, Brazil. Analysis of mitochondrial DNA (mtDNA) control region sequences failed to detect variability among sequences. Conversely, evidence of significant male population structure was found on the basis of ten nuclear microsatellite loci. Surprisingly, the microsatellite markers were able to distinguish between individuals from the two embayments located 60 km apart. The results suggest that differences in habitat type and behavioral specializations are likely to explain the patterns of genetic structure. These findings should provide baselines for the management of communities exposed to increasing human-driven habitat loss.  相似文献

The pattern of genetic variability of two species of Mediterranean gobiids was compared, with reference to their different life history traits (Aphia minuta paedomorphic and pelagic; Gobius niger metamorphosed and benthic). The aim was to evaluate how different life histories can affect the genetic structure in these marine teleosts. The study was carried out on populations of both species sampled in the western Mediterranean and in the Adriatic Sea. Seven restriction endonucleases were used for the RFLP analysis of a mitochondrial DNA segment comprising the NADH dehydrogenase subunits 3, 4L and 4. The results highlighted two different patterns of genetic variation, a weak genetic structure in A. minuta and population subdivision in G. niger. These observations may be explained not only in terms of the different dispersal capabilities of these species, but also considering that A. minuta is an abbreviate iteroparous spawner while G. niger is a protracted iteroparous spawner. Because abbreviate iteroparity is a reproductive strategy selected in stable environments with high resource availability, Pliocene and Pleistocene climate oscillations may have represented factors that negatively influenced the reproductive success of A. minuta, producing demographic fluctuations and bottlenecks, as suggested by the mismatch distribution analysis. The weak genetic structure of A. minuta populations seems to be therefore due to a more recent re-colonization of the Mediterranean basin after a severe population decline, rather than to the high vagility of this pelagic goby.  相似文献

Crassostrea ariakensis is an important aquacultured oyster species in Asia, its native region. During the past decade, consideration was given to introducing C. ariakensis into Chesapeake Bay, in the United States, to help revive the declining native oyster industry and bolster the local ecosystem. Little is known about the ecology and biology of this species in Asia due to confusion with nomenclature and difficulty in accurately identifying the species of wild populations in their natural environment. Even less research has been done on the population genetics of native populations of C. ariakensis in Asia. We examined the magnitude and pattern of genetic differentiation among 10 wild populations of C. ariakensis from its confirmed distribution range using eight polymorphic microsatellite markers. Results showed a small but significant global θ ST (0.018), indicating genetic heterogeneity among populations. Eight genetically distinct populations were further distinguished based on population pairwise θ ST comparisons, including one in Japan, four in China, and three populations along the coast of South Korea. A significant positive association was detected between genetic and geographic distances among populations, suggesting a genetic pattern of isolation by distance. This research represents a novel observation on wild genetic population structuring in a coastal bivalve species along the coast of the northwest Pacific.  相似文献

Mitochondrial d-loop sequences were analyzed to characterize the phylogeographic and population genetic structure of the northern clingfish (Gobbiesox maeandricus Girard). Sequence analysis of 378 bp from 111 individuals sampled in 14 localities along the northeast Pacific coast and within the Strait of Georgia from 1996 to 1999 revealed marked genetic differentiation (Φct=0.247) among regional population groupings. The gene genealogy distinguished two major clades of haplotypes separated by at least 1.1% sequence divergence. One clade with very low haplotype diversity (h=0.2095, n=18) occurred only within the recently unglaciated Strait of Georgia. The other clade had high haplotype diversity (h=0.8808, n=93) and was found in all populations. High haplotype diversity was found in open coastal populations, both north and south of the maximum extant of the Wisconsin ice sheet, suggesting that the clingfish range was not pushed to a southern refugium during the last glacial maximum. A nested clade analysis also did not detect a large northward expansion from a single southern refugium. The level of sequence divergence and coalescent-based analyses suggest that the observed patterns of polymorphism are the result of Pleistocene diversification within multiple refugia, followed by population expansion and asymmetrical lineage introgression. In a previous study on the kuruma shrimp Penaeus japonicus from the South China Sea, we detected high genetic divergence between two morphologically similar varieties (I and II) with distinct color banding patterns on the carapace, indicating the occurrence of cryptic species. In the present study, we clarify the geographical distribution of the two varieties in the western Pacific by investigating the genetic differentiation of the shrimp from ten localities. Two Mediterranean populations are also included for comparison. Based on the mitochondrial DNA sequence data, the shrimps are separated into two distinct clades representing the two varieties. Variety I comprises populations from Japan and China (including Taiwan), while variety II consists of populations from Southeast Asia (Vietnam, Singapore and the Philippines), Australia and the Mediterranean. Population differentiation is evident in variety II, as supported by restriction profiles of two mitochondrial markers and analysis of two microsatellite loci. The Australian population is genetically diverged from the others, whereas the Southeast Asian and Mediterranean populations show a close genetic relationship. Variety I does not occur in these three localities, while a small proportion of variety II is found along the northern coast of the South China Sea and Taiwan, which constitute the sympatric zone of the two varieties. The present study reveals high genetic diversity of P. japonicus. Further studies on the genetic structure of this species complex, particularly the populations in the Indian Ocean and Mediterranean, are needed not only to understand the evolutionary history of the shrimp, but also to improve the knowledge-based fishery management and aquaculture development programs of this important biological resource.  相似文献

Rhizophora mangle L. is a widespread mangrove species in the Western Hemisphere. Mangrove habitat loss and their importance to coastal and reef ecosystems make greater understanding of their genetic structure useful for conservation and management. An amplified fragment polymorphism (AFLP) analysis was performed on samples from Florida and the Caribbean to discover the genetic structure present. R. mangle had variable genetic diversity not related to latitude; P ranged 7 %–92 %. Some other factor, perhaps human impact, has caused low genetic diversity in some populations. Across Florida R. mangle populations varied in genetic diversity with less diversity (Gst?=?0.195) and greater gene flow on the Atlantic coast (Nm =2.07) than on the Gulf coast (Gst?=?0.717, Nm?=?0.197). Gene flow between Caribbean islands was low (Nm?=?0.386) compared to continental populations (Nm?=?1.40), indicating that long distance dispersal is not common between islands. Analysis of molecular variance (AMOVA) analysis showed significant deviations from Hardy-Weinberg expectations at the level of region among subpopulations and overall genetic difference among subpopulations for R. mangle. One implication for management is that small continental populations and island populations may be genetically isolated and distinct from each other.  相似文献

The genetic differentiation of the European lobster (Homarus gammarus) was investigated in 3,283 individuals from 44 population samples throughout its geographical distribution (Norway to Greece) by means of polymerase chain reaction restriction fragment length polymorphism analysis of a 3-kb mitochondrial DNA segment. Ninety composite haplotypes were revealed with the number of haplotypes in each population sample ranging from 4 to 31. Private haplotypes were found at very low frequencies. The global exact test of sample differentiation based on composite haplotype frequencies was statistically significant. FST analyses also showed significant heterogeneity among the European lobster population samples (FST=0.078, P<0.001). This differentiation was mainly due to the population samples from northern Norway, the Netherlands, and the Mediterranean Sea. These samples differentiated from the rest due to reduced gene diversity rather than to unique haplotypes. The relationships among these samples were illustrated with cluster analyses; four major distinct groups were evident: Mediterranean, northern Norway, Netherlands, and the remaining Atlantic samples. Based on the low degree of differentiation revealed in the European lobster and its limited capacity for dispersal, the most probable hypothesis is that all populations have been established from a common refuge after the end of the last Ice Age, that is, within the past 15,000 years. The results of this study show that mitochondrial DNA is a powerful tool for the determination of the genetic structure among lobster samples, which is important for a proper management policy designed to protect and to cultivate this species.Communicated by O. Many meiofaunal species are reported to be cosmopolitan, but due to uncertainties of identification, the affiliation of specimens from geographically distant areas to the same species-taxon is problematic. In this study, we examined morphological and molecular variation in samples of Xenotrichula intermedia Remane (Gastrotricha: Chaetonotida) from the Mediterranean Sea, the northwestern Atlantic and the northern Gulf of Mexico. Univariate analysis of 16 morphological traits was unable to detect differences among populations, except for the length of the pharynx, which was significantly shorter in the Gulf of Mexico specimens. Canonical discriminant analysis separated the Gulf of Mexico specimens from the other two populations, with pharynx length contributing about half of the total discrimination. Molecular analysis based on restriction-fragment length polymorphisms (RFLPs) in a 710-base pair polymerase chain-reaction (PCR) produet representing roughly half of the mitochondrial cytochrome oxidase I (COI) gene detected four haplotypes: one each from the Mediterranean and the Gulf of Mexico populations and two coexisting within the Atlantic population. The estimated nucleotide-sequence divergence calculated for each pairwise combination of haplotypes (based on the proportion of shared fragments) ranged from 5.3 to 11.5%. The high genetic divergence and the inability to clearly separate populations based on morphology suggest that individuals characterized by different haplotypes are genetically isolated sibling species.  相似文献

Marine invertebrates with high larval dispersal capacity typically exhibit low degrees of population differentiation, which reflects both contemporary and historical processes. We sampled 346 individuals from seven populations of the lined shore crab, Pachygrapsus crassipes Randall, along the northeastern Pacific Coast and Korea during summer 2003. DNA sequence analysis of 613 bp of the mitochondrial COI gene showed that overall gene diversity (h) was high (0.92±0.01), whereas overall nucleotide diversity (π) was low (0.009±0.005). A total of 154 mtDNA haplotypes were identified; however, 114 were present in only one individual. Analysis of molecular variance revealed significant genetic structuring at Point Conception, CA, USA, that is likely due to the oceanographic circulation patterns, which result in asymmetrical migration of haplotypes. However, genetic variation among eastern Pacific populations was generally low, probably because of high contemporary gene flow and recent common ancestry of haplotypes. Mismatch analysis and nested clade analysis suggested that the population history of this region is characterized by two contiguous northwards range expansions, which are congruent with Late Pleistocene glacial cycles. Highly significant genetic differentiation was detected between eastern Pacific populations and Korea, indicating transpacific gene flow is restricted. Time of divergence between the two transpacific lineages was estimated between 0.8 and 1.2 Myrs ago. The small, recently founded population of P. crassipes at Bamfield, BC, Canada, did not appear to have undergone a founder effect.  相似文献
