A bacterial strain capable of degrading carbofuran as the sole carbon source was isolated from carbofuran-phytoremediated rhizosphere soil of rice. A 16S rRNA study identified the strain as Burkholderia sp. (isolate PCL3). Free cells of isolate PCL3 possessed inhibitory-type degradation kinetics with a q max of 0.087 day?1 and S m of 248.76 mg·L?1. Immobilised PCL3 on corncob and sugarcane bagasse possessed Monod-type degradation kinetics with a q max of 0.124 and 0.098 day?1, respectively. The optimal pH and temperature with the highest degradation rate coefficient of carbofuran were pH 7.5 and 35 °C, respectively.  相似文献

Biodegradability of the plant growth retardant paclobutrazol by a microbial consortium in which Pseudomonas was the predominant strain was investigated in batch culture. The consortium which had been isolated from an industrially contaminated sediment was proven to be useful for the treatment of effluents containing paclobutrazol. Paclobutrazol was degraded by the pure isolated strain of Pseudomonas sp. as well as the microbial consortium. Paclobutrazol was utilized as the sole source of carbon and energy. Sixty percent of the paclobutrazol was degraded by the microbial consortium from an initial concentration of 54 mg L?1 within 48 h and more than 98% of an initial concentration of 3.4 mg L?1 was degraded within 36 h. The optimum temperature and pH were determined to be 30°C and 7.0, respectively. A pure strain of a bacterium, isolated from the enrichment culture was identified as Pseudomonas sp. The microbial consortium was tolerant of high pH and could degrade paclobutrazol faster than the pure strain. The degradation rate of this plant growth regulator in an aerobic environment was greater than that under anaerobic conditions.  相似文献

This study was conducted to examine adsorption of pesticides bifenthrin, carbosulfan, λ-cyhalothrin, cypermethrin, endosulfan, parathion methyl, monocrotophos and 4-nitrophenol by sandy clay loam (S.C.L) and sandy loam (S.L) soils (with varying organic content). There was no significant difference between the observed soil water partitioning coefficient values (K d) derived from linear and nonlinear Freundlich isotherms. Adsorption of pesticides on S.C.L soils was higher than those on S.L soils. K d values showed significant correlations (r 2?=?0.8???0.99 and 0.65???0.97) with soil organic carbon content (OC) and weak correlations (r 2?=?0.2???0.29 and 0.1???0.18) with clay contents of S.C.L and S.L soil at p?≤?0.05, respectively for all pesticides (except monocrotophos). Observed K oc values (soil-water partitioning constants based on the organic C fraction of the soil) were in accordance with the literature values of Wauchope and Tomlin with a maximum deviation of less than 0.5 log units. Ten Quantitative Property-Property Relationships (QPPR) among water solubility, n-octanol water coefficient (K ow) and K oc were proposed for studied pesticides except monocrotophos. The models were considered acceptable when predicted-observed difference for log?K ow and log?K oc were ≤?0.3 and ≤?0.5?log units, respectively, during the validation procedure. This work indicates that the log?K oc derived from the log Kow, from some of existing relationships, may be a fair predictor where observed values (i.e., K d and K oc) are not available. Furthermore, predicted leaching potential by groundwater ubiquity scores (GUS) equation was solved by using observed K oc values and literature reported half lives of pesticides. GUS ranked the mobility of nonvolatile compounds i.e., bifenthrin, λ-cyhalothrin, cypermethrin and endosulfan extremely low; methyl parathion very low; 4-nitrophenol low; carbofuran and monocrotophos very high in S.C.L and S.L soils, respectively. Results discussed in this paper provide background to prioritize pesticides or chemical groups that should be evaluated under field conditions with regard to their leaching potential to groundwater in arid climates.  相似文献

A study was conducted on the Chironomus plumosus larvae to determine initiation of resistance to four pesticides – chlorfenvinphos, chlorpyrifos, chlorpyrifos-methyl, and malathion. First generational LC50 values were well within the threshold value for chironomids based upon the literature. Subsequent LC50 values were observed to increase, indicating a lessening of the toxicity of the pesticides to the chironomid. In the case of chlorfenvinphos, the 96 h LC50 for generations 1–23 was 6 µg L?1, in generations 3–7 was 8.57 µg L?1 and 11.14 µg L?1 for generations 8–9. Generations 10–12 had an LC50 value of 22.58 µg L?1 and generation 13 had an LC50 value of 35.08 µg L?1. Generation 14 had an LC50 value of 47.58 µg L?1. Generations 15–19 and 20–24 had 96 h LC50 values of 60.68 µg L?1, 72.58 µg L?1, 85.08 µg L?1, 97.58 µg L?1 and 110.08 µg L?1, respectively. Generations 25, 26 to 30, 31 to 38 and 39 to 45 had 96 h LC50 values of 160.42 µg L?1, 210.7 µg L?1, 262.24 µg L?1 and 274.36 µg L?1, respectively. The variation between LC50 values was found to be statistically significant. This was observed for most pesticides tested. Larval size and life cycle duration was observed to change from generation to generation with the body size decreasing markedly from 1 to 0.3 cm with life cycle increasing from 7 to 39 days.  相似文献

The phenylurea herbicide diuron [N-(3,4-dichlorophenyl)-N,N-dimethylurea] is widely used alone or in a broad range of herbicide formulations. Its degradation in sugarcane-cultivated soils which have been impacted by the herbicide through repeated applications was studied. Liquid culture experiments with diuron as the only carbon source led to the isolation of different bacterial strains capable of degrading diuron. The bacterial species belonging to the genera Bacillus, Vagococcus, and Burkholderia, identified through biochemical and molecular characterization, degraded diuron to different extents. The isolated Bacillus cereus, Vagococcus fluvialis, Burkholderia ambifaria, and Bacillus spp1 degraded diuron by 21%, 25%, 22%, and 19% of the initially applied concentration of 40?mg?L?1, respectively, after 35 days of incubation in liquid culture media. Small amounts of 3,4-dichloroaniline and the de-methylated metabolite N-(3,4-dichlorophenyl)-N-methylurea were detected in liquid culture media. The combination of V. fluvialis and B. ambifaria showed an enhanced degradation of up to 30% of the initially applied concentration of 40?mg?L?1. Degradation by pure isolates was low (18–25%) compared to the capacities of diuron degradation shown by the bacterial communities (58–74%). This study showed the presence of diuron degraders in sugarcane-cultivated soils impacted by diuron due to repeated applications.  相似文献

A highly sensitive competitive ELISA was standardized to determine aflatoxin M1 (AFM1) in milk and dairy samples randomly collected from Faisalabad city, using Riedel-de-Haen, Aflatoxin M1 ELISA Systems, (Art–No. 45169). Standards of 5, 10, 25, 50, and 100 ng L?1 AFM1 in sample/standard buffer were used in optimizing anti-AFM1 antibodies and peroxidase–AFM1 conjugate. Linear standard curve showed increase in concentration (log C) accompanied by decrease in maximal absorbance (%B/B0) values (%B/B0 = 124.3 ? 45.0 log C) with highly significant negative correlation (?0.973). The middle of the test (50% B/B0) was at 30 ng L?1 with 6.7 ng L?1 the lowest detection limit (90% B/B0). Duplicate analysis of 41 samples showed AFM1 levels below EC permissible limits (50 ng L?1) with maximums of 40, 13, and 7.4 ng L?1 in milk, yogurt and butter, respectively. However, possibility of exceeding limits cannot be excluded, and demands regular monitoring.  相似文献

The level of accumulation of selected essential and non-essential metals, namely; Ca, Cu, Fe, Zn, Mn, Cd, Pb, and Cr have been investigated in the seeds, fruits, and flowers of some medicinal plants utilized for tapeworm treatment in Ethiopia and their respective soil samples. These include seed of Cucurbita maxima (Duba), fruit of Embelia abyssinica (Ankoko), flowers of Hagenia abyssinica (Kosso), and fruits of Rosa abyssinica (Kega) and their respective soil samples. A wet digestion procedure with a mixture of conc. HNO3 and HClO4 for the plant samples and a mixture of conc. HNO3, HCl, and H2O2 for soil samples were used to solubilize the metals. Ca (1280–12,670?mg?kg?1) was the predominant metal followed by Fe (104–420?mg?kg?1), and Zn (18–185?mg?kg?1) in all the plant materials except for Hagenia abyssinica flower from Hirna in which Mn (16–42?mg?kg?1) followed by Fe. Among the non-essential toxic metals, Pb was not detected in Cucurbita maxima of Boji, Gedo and Hirna origins and in Rosa abyssinica of Hirna site. Similarly, Cr was not detected in Rosa abyssinica fruits of Boji and Gedo sites. The sampled soils were found to be between strongly acidic to weakly basic (pH: 4.7–7.1). In the soil samples, Ca (8528–18,900?mg?kg?1) was the most abundant metal followed by Fe (417–912?mg?kg?1), Zn (155–588?mg?kg?1), Mn (54–220?mg?kg?1), Cr (21–105. mg?kg?1), Cu (11–58?mg?kg?1), Pb (13–32?mg?kg?1) and Cd (2.8–4.8?mg?kg?1). The levels of most of the metals determined in the medicinal plants and the respective soil samples are in good agreement with those reported in the literature and the standards set for the soil by various legislative authorities.  相似文献

The extracellular extract obtained from 3 weeks incubation of the soil isolate cyanobacterium strain Nostoc piscinale GT-319 in BG-11 broth medium showed cytotoxic activity against Chinese Hamster Ovary (CHO) cells. Based on comet assay, a concentration of 333 µg mL?1 (IC50) produced DNA breakages in CHO cells. The concentration of 481 mg kg?1 (LD50) produced acute toxicity in mice at 48 h.  相似文献

The gene for the catalytic domain of thermostable endo-β-1,3-glucanase (laminarinase) LamA was cloned from Thermotoga maritima MSB8 and heterologously expressed in a bioengineered Synechococcus sp. PCC 7002. The mutant strain was cultured in a photobioreactor to assess biomass yield, recombinant laminarinase activity, and CO2 uptake. The maximum enzyme activity was observed at a pH of 8.0 and a temperature of 70°C. At a CO2 concentration of 5%, we obtained a maximum specific growth rate of 0.083 h–1, a biomass productivity of 0.42 g?L–1?d–1, a biomass concentration of 3.697 g?L–1, and a specific enzyme activity of the mutant strain of 4.325 U?mg–1 dry mass. All parameters decreased as CO2 concentration increased from 5% to 10% and further to 15% CO2, except enzyme activity, which increased from 5% to 10% CO2. However, the mutant culture still grew at 15% CO2 concentration, as reflected by the biomass productivity (0.26 g?L–1?d–1), biomass concentration (2.416 g?L–1), and specific enzyme activity (3.247 U?mg–1 dry mass).

A micro-scale algal growth inhibition (μ-AGI) test using a common micro-plate based fluorometric detection was used to demonstrate the effects of humic substances (HSs) on the toxicity of tetrabromobisphenol A (TBBPA) and its oxidative decomposition products 2,5-dibromo-1,4-benzoquinone (2,5-DBBQ), 2,5-dibromohydroquinone (2,5-DBHQ), 2,6-dibromobenzoquinone (2,6-DBBQ), and 2,6-dibromophenol (2,6-DBP) to Pseudokirchneriella subcapitata. The EC50 values were: EC50(TBBPA) = 7 mg L?1, EC50(2,5-DBHQ) = 7 mg L?1, EC50(2,5-DBBQ) = 19 mg L?1, EC50(2,6-DBP) = 49 mg L?1, and EC50(2,6-DBBQ) = 13 mg L?1. The toxicity of the chemicals was slightly lower in the presence of HA. The toxicity of TBBPA decomposed by a biomimetic catalytic system consisting of iron (III) 5,10,15,20-tetrakis (p-sulfonatophenyl) porphyrin (Fe(III)-TPPS) and KHSO5 was also evaluated using P. subcapitata and Chlamydomonas reinhardtii.  相似文献

Studies were carried out to determine the toxicity of some selected pesticides on fresh water fish in a tropical environment. The uptake of the pesticides lindane, pentachlorophenol (PCP), and propoxur, which are frequently used by farmers and industrialists were studied in concrete ponds at the University of Cape Coast, in Ghana. The fishes used for the study were Oreochromis niloticus, Clarias gariepinus, and Chrysicthys nigrodigitatus, and were obtained from cultured ponds in the Cape Coast District and Mankessim in the Central Region and Weija Dam, in the Greater Accra region of Ghana. Single high lethal concentration (SD) or acute treatment and cumulative/chronic (or multiple minor) lethal concentration (CD) treatment were employed in administering the pesticides to the fishes via water. Gas chromatograph (GC) Electron Capture Detector (ECD) analysis was done on the dead fishes to see the extent of ingestion. The LC50 values obtained for lindane on the three fish samples were as follows: Chrysicthys?–?0.38?mg?L?1; Oreochromis?–?0.42?mg?L?1, and Clarias?–?1.2?mg?L?1. Mortalities occurred in fish within 3–5 days of application. For the PCP on Chrysicthys, Oreochromis, and Clarias species the LC50 values were 0.42, 0.32 and 0.64?mg?L?1, respectively for over a 2–3 day period. For a three-time influx period of propoxur the LC50 for Chrysicthys, Oreochromis, and Clarias, were 22.0, 30.40, and 45.04 (all in mg?L?1), respectively. The results obtained indicated that the pesticides had adverse effects on the general growth and reproduction of fishes. Gonadosomatic indices also showed that the pesticides affected the development of the body, the gonads, and their reproduction.  相似文献

Organophosphate, dichlorvos (2,2-dichlorovinyldimethylphosphate, DDVP), and diazinon (DZN) as well as carbamates are currently found in contaminated water, dust, soil, agricultural waste, and elsewhere in the environment, and can be harmful after accidental or deliberate exposure. Intoxication by these compounds causes a generalized cholinergic crisis due to the inhibition of acetylcholinesterase (AChE), whose major physiological role in mammalian tissues is in nervous transmission. The half-maximal inhibitory concentration (IC50) was found to be higher for carbaryl than DDVP and DZN, about 3–4 times in liver and about 4–6 times in muscles. The half time (t 1/2) of inhibition (8?µmol?L?1 DDVP or DZN; 40?µmol?L?1 carbaryl) ranged between 4 and 16?min and decreased as follows: pig?>?cattle?>?sheep for liver tissue, and cattle?>?sheep?>?pig for muscle. Rate constants of inhibition (k i) ranged between 43?×?10?3 and 168?×?10?3?min?1 for liver and from 46?×?10?3 to 115?×?10?3?min?1 for muscles. Concentrations of perfluorooctane sulfonate (PFOS) were measured for the first time in major rivers within southwest, Nigeria. PFOS was concentrated from water and sediment using solid-phase extraction, identified and quantified with high-performance liquid chromatography coupled with tandem mass spectrometry. Concentrations of PFOS ranged from 1.71 to 16.19 ng L?1 in water, and from 1.64 to 10.29 ng g?1 in sediments across all locations. Comparatively, the concentrations of PFOS observed in this study were within the range ever measured in the environment. Field-based sediment water distribution coefficients (log Kd, L kg?1) ranged from 2.08 to 3.56. While no correlation was observed between Kd and organic carbon contents, there was significant positive correlation between Kd and salinity (r2 = 0.7867), which suggested that activities capable of increasing salinity can enhance PFOS removal from the environment.  相似文献

Sorption and leaching behavior of hexaconazole in four different soils (alluvial, red, laterite, and black) was studied using a batch equilibration technique. The values of the Freundlich adsorption constant 1/nads ranged from 0.75 to 0.85 for all four soils, showing strong non-linear behavior. Upon stepwise desorption with CaCl2 solution (10 mmol·L?1), release of hexaconazole was maximum with the first elution, the amount decreasing with each subsequent one. The leaching behavior under saturated flow conditions was also studied with soil columns packed in polythene tubes. The mobility of hexaconazole was maximum in sandy loam and minimum in black soil.  相似文献

Controlled laboratory experiments were conducted to examine how photosynthesis and growth occur in Potamogeton wrightii Morong under different photoperiods and nutrient conditions. The experiment was based on a 3×2 factorial design with three photoperiods (16, 12 and 8 h) of 200 μE · m?2·s?1 irradiance and two nutrient conditions, high (90 μmol N · L?1·d?1 and 9 μmol P · L?1·d?1) and low (30 μmol N L?1·d?1 and 3 μmol P · L?1·d?1). After 14, 28, 56 and 70 days of growth, plants were harvested to determine net photosynthesis rate and various growth parameters. Above- and below-ground biomass were investigated on days 56 and 70 only. Plants under low nutrient conditions had greater leaf area, more chlorophyll a, a higher rate of net photosynthesis and accumulated more above- and below-ground biomass than plants in the high nutrient condition. Plants with an 8 h photoperiod in the low nutrient condition had a significantly higher rate of net photosynthesis, whereas 8 h photoperiod plants in the high nutrient condition had a lower rate of net photosynthesis and their photosynthetic capacity collapsed on day 70. We conclude that P. wrightii has the photosynthetic plasticity to overcome the effects of a shorter photoperiod under a tolerable nutrient state.  相似文献

The impact of bisphenol A (BPA) on Gammarus fossarum and Lumbriculus variegatus was studied in four artificial indoor streams (0, 5, 50 and 500?µg?L?1 BPA, nominal) over 103 days in a pulse–dose exposure scenario (weekly BPA application). For G. fossarum populations at day 103, the proportions of juveniles and of breeding females from the highest BPA treatment were in tendency reduced. For individually exposed gammarid pairs an EC10 of 17?µg?L?1 BPA (nominal) for the proportion of reproductive females in the fourth brood was determined. During the first three broods, the largest brood size occurred at the highest BPA concentration, whereas in the fourth brood it decreased concentration-dependently (fourth brood EC10?=?5?µg?L?1 BPA, nominal). Effects on L. variegatus were a reduced population growth (103?d-EC10 of 2?µg?L?1 BPA, nominal) and an increase in dry weight and the number of segments in large, complete worms.  相似文献

The toxic effects of an organophosphorous insecticide, monochrotophos (MCP) were investigated on Paramecium caudatum in static acute toxicity tests (10?min and 2?h). The lethal concentrations (50%) were determined by probit method, for technical monocrotophos as 60 and 40.6?mg?L?1, respectively. We have combined conventional light microscopy and a computerized video tracking system to determine behavioural and morphological changes in paramecium on exposure to MCP. Paramecia exposed to highest concentrations (90–100?mg?L?1) used for 10?min exposure, exhibited initial increase with subsequent decrease in mobility with enormous blebbing, leading to lysis of cells. In the second set of experiments, the cells exposed to lethal concentration (40.6?mg?L?1) for 2?h were under stress, and reduced their locomotor behaviour, i.e. distance travelled per unit time (mm in 6?min) and swimming speed (mm?s?1) with increased time of exposure. In the third set of experiments, the number of generations and generation time in 24?h was evaluated with respect to the different sub lethal concentrations (5, 10, 15 and 20?mg?L?1) of toxicant. The number of generations decreased and generation time extended significantly in a concentration dependent manner. The results indicate that the Paramecium toxicity assay could be used as a complimentary system to rapidly elucidate the cytotoxic potential of insecticides.  相似文献

The physicochemical characteristics of the River Challawa in the Kumbotso Local Government Area of Kano State were studied across four seasons (warm and dry, cold and dry, hot and dry, and warm and wet seasons) between October 2006 and August 2008. Physicochemical parameters like pH, electrical conductivity (EC), total dissolved solids (TDSs), total suspended solids (TSS), alkalinity, turbidity, hardness, dissolved oxygen (DO), biological oxygen demand (BOD), chemical oxygen demand (COD), ammonia, chloride, nitrate, sulfate, and phosphate were analyzed and compared with standard permissible limits. The mean range of parameters determined across the seasons revealed: 8.36–8.59 for pH; 209.57–335.27 µS cm?1 for EC; 134.05–208.5 mg L?1 for TDS; 182.4–273.45 mg L?1 for TSS; 125.3–276.07 mg L?1 for hardness; 16.11–40 mg L?1 of CaCO3 for alkalinity; 2.76–3.78 mg L?1 for DO; 1.9–43.33 mg L?1 for BOD; 231.67–310.06 mg L?1 for COD; 221.67–441.67 mg L?1 for turbidity; 17.98–35.89 mg L?1 for chloride; 5.12–8.89 mg L?1 for nitrate; 37–558.83 mg L?1 for sulfate; and 0.47–0.81 mg L?1 for phosphate. Data showed that all parameters determined were significantly different among the various sampled sites across seasons. With the exception of COD, EC, and phosphate, all other parameters determined were within the WHO and USPH standard limits.  相似文献

The consequence of polycyclic aromatic hydrocarbons (PAHs) in the environment is of great concern. The hydrophobic properties of PAHs significantly impact phase distribution causing limited bioavailability. Enhanced biodegradation has been extensively carried out by surfactants and the redeployment effect was recognized. However, the quantitative relationship concerning the impact of solids was rarely reported. A batch of biphasic tests were carried out by introducing Mycobacterium vanbaalenii PYR-1 and hydroxypropyl-β-cyclodextrin (HPCD) into a mixture of phenanthrene solution and various glass beads (GB37-63, GB105-125, and GB350-500). The comparative results demonstrated that HPCD had little effect on microbial growth and was not degradable by bacterium. A model was proposed to describe the biodegradation process. The regression results indicated that the partition coefficient k d (1.234, 0.726 and 0.448 L·g?1) and the degradation rate k (0 mmol·L?1: 0.055, 0.094, and 0.112; 20 mmol·L?1: 0.126, 0.141, and 0.156; 40 mmol·L?1: 0.141, 0.156 and 0.184 d?1) were positively and negatively correlated with the calculated total surface area (TSA) of solids, respectively. Degradation enhanced in the presence of HPCD, and the enhancing factor f was calculated (20 mmol·L?1: 15.16, 40.01, and 145.5; 40 mmol·L?1: 13.29, 37.97, and 138.4), indicating that the impact of solids was significant for the enhancement of biodegradation.  相似文献

Biodegradation of N,N-bis(carboxymethyl)-L-glutamate and its utilization by micro-organisms in marine environments were studied in Organization of Economic Development and Cooperation (OECD) screening tests and with pure bacterial cultures. Biodegradation in excess of 60% was achieved in closed bottle tests in less than 60 days, suggesting complete degradation. The bacterial strain isolated from marine sandy sediment and utilizing N,N-bis(carboxymethyl)-L-glutamate as sole source of carbon, nitrogen, and energy was identified as Rhizobium radiobacter. The strain was capable to mineralize N,N-bis(carboxymethyl)-L-glutamate in seawater and in synthetic media containing 0–60 g L?1 sea salts. Growth and adaptation studies indicated that N-carboxymethyl-L-glutamate and L-glutamate are main metabolites.  相似文献
