Cnidarian–dinoflagellate symbioses are not well understood at the molecular level. Observed specificity between partners during initiation, establishment, and maintenance of the relationship strongly implies a role for chemical signaling. This report presents biochemical and immunocytochemical evidence for potential signaling molecules, as large molecular weight glycoproteins, secreted by Symbiodinium dinoflagellates both in culture and in symbiosis. Polyclonal antibodies directed against recovered exudate from S. microadriaticum, the natural endosymbiont of Cassiopea xamachana, the upside–down jellyfish, were highly specific in recognizing exudates from Symbiodinium species that can successfully induce developmental metamorphosis in the host but did not recognize exudates from Symbiodinium species that do not. Immunoblot analyses showed S. microadriaticum exudate to be protease sensitive. Release of antigenic material by symbiotic S. microadriaticum was demonstrated through light and electron microscopy using immunogold-labeled anti-S. microadriaticum (anti-Sm-XuLg) antibodies as probes. These secreted, symbiont-derived glycoconjugates may be candidates for interspecific molecular signals.  相似文献

以硝普钠(Sodium nitroprusside,SNP)为一氧化氮(Nitric oxide,NO)供体研究了NO对海洋微藻生长的影响.对不同浓度SNP在海水介质中释放NO的过程进行了监测;对所培养的亚心形扁藻(Platymonas subcordiformis)和中肋骨条藻(Skeletonema costatum)进行藻密度测定,观测NO对微藻生长的影响.结果表明:5、10和100μmol·L-1的SNP释放NO浓度大约分别为6×10-9、9×10-9和2×10-7mol·L-1左右,而释放时间分别为4、5.5和7.5h.研究表明,NO对不同微藻有明显不同的作用规律:NO持续作用下,对亚心形扁藻的最佳作用浓度在10-8mol·L-1数量级;对赤潮藻中肋骨条藻的最佳作用浓度在10-9mol·L-1数量级;赤潮藻对NO的响应比非赤潮藻更灵敏,NO可能是海洋生态系中微藻生长重要的调节因子.  相似文献

In view of their possible involvement in ammonium assimilation in the coral/algal symbiosis, we have purified two distinct glutamate dehydrogenase isoenzymes from the symbiotic dinoflagellate Symbiodinium microadriaticum (Freudenthal) extracted from the staghorn coral Acropora formosa collected from Magnetic Island, North Queensland, Australia, in 1986–1987. An NADPH-specific glutamate dehydrogenase (GDH) displayed biphasic kinetics with respect to ammonium as the variable substrate; at low substrate concentrations the apparent K m was below 1 mM, whereas at high substrate concentrations the corresponding value was approximately 200 mM. The NADPH-GDH displayed extremely low activity in the direction of glutamate oxidation; together with the kinetic data this suggests a probable role in ammonium assimilation. A second (NADH-specific) GDH was found to have both amination and deamination activities, and presumably functions in vivo in glutamate oxidation. Kinetic constants are reported for both GDH isoenzymes.  相似文献

一氧化氮(nitric oxide,NO)作为生物活性分子,广泛参与各种生物和非生物胁迫。采用营养液培养,研究Cu胁迫下番茄体内L-精氨酸、NO、多胺代谢对外源NO的响应机制,以期为铜污染区域蔬菜种植提供科学依据和技术支持。结果表明,Cu胁迫下添加外源SNP(硝普钠,外源NO供体)能够调节番茄根系和叶片中精氨酸脱羧酶(Arginine Decarboxylase,ADC)、鸟氨酸脱羧酶(Ornithine Decarboxylase,ODC)及一氧化氮合酶(Nitric Oxide Synthase,NOS)活性。根系中,外源SNP能够上调NOS活性,L-精氨酸代谢向着NO合成方向进行;叶片中,外源SNP能够同时上调ADC、ODC、NOS活性,PA、NO的合成同时进行;此外,Cu胁迫下添加外源SNP能够增加根系和叶片L-精氨酸含量,而作为PA、NO的合成前体,精氨酸含量升高无疑会间接促进PA、NO的合成,从而提高番茄对Cu胁迫的抵抗能力。  相似文献

Summary. The basic chemical structure of the sex pheromone of the pine sawfly Microdiprion pallipes (Fallén) has earlier been identified as the propionate ester of (2S,3S,7R/S,11R/S)/(2R,3R,7R/S,11R/S)-3,7,11- trimethyl-2-tridecanol. We now report the results from further investigations on the male response to individual stereoisomers and to blends of stereoisomers, both in electroantennographic (EAG) recordings and in field trapping experiments. We also present our attempts to determine the stereochemistry of the compounds present in females of M. pallipes. By comparing gas chromatograms and mass spectra obtained from natural extracts with those from synthetic compounds it was found that the females contain one or more of the four (2S,3S,7R/S,11R/S)-3,7,11-trimethyl-2- tridecanol isomers (SS++-1). The active pheromone component is the corresponding propionate ester 2. In EAG experiments, males responded most strongly to five propionate ester samples, namely two four-isomer blends: SS++-2 and SR++-2, and three individual stereoisomers: SSSR-, SRRR- and SRSR-2. In a series of field trapping experiments it was found that males were attracted to the SR++-2 four-isomer blend and to the individual isomer SSSR-2. Based on the EAG-recordings and field responses of males and the stereoisomers found in the females, we suggest that the propionate ester of (2S,3S,7S,11R)-3,7,11-trimethyl-2- tridecanol (SSSR-2) is used as a main component of the sex pheromone in M. pallipes. Apparently the males react to other stereoisomers in addition to that or those produced by the females.  相似文献

A new method for the assessment of the effects of several contaminants on marine microalgae,Tetraselmis suecica (Kyling) Butcher,Skeletonema costatum (Grev.) Cleve, andProrocentrum lima (Ehrenberg) Dodge was developed in 1990. The method is based on the measurement of cell esterase activity using a fluorimetric stain, fluorescein diacetate (FDA), selected from amongst three stains (FDA, Neutral Red, thiazolyl tetrazolium bromure) for its higher sensitivity. Biochemical (K m,V max) and physiological (growth, specific activity) aspects of the enzymatic activity as revealed by the FDA method are discussed. Different categories of compounds (weed-killers, insecticides, metals) and some water samples from Seine Bay were tested for their toxic effects on microalgae. Experiments were performed on microplates using a fluorimetric microplate reader. The various steps of the experiments and data-processing were controlled by software. Applications of the system to rapid ecotoxicological tests (determination of the IC50, i.e., the concentration at which 50% inhibition of growth occurs) and to the assessment of environment quality by studying the toxic effect of water samples on microalgae are proposed.  相似文献

Sorites orbiculus (Ehrenberg), S. orbitolitoides (Hofker) and Amphisorus hemprichii (Ehrenberg) harbor dinophyceans similar to, but not identical with, Symbiodinium microadriaticum. The symbionts, which bear shortened flagella, are concentrated beneath the well-lit, lateral test wall. The dinoflagellate symbionts (zooxanthellae) present in many reef corals aid in the survival of the symbiotic unit in nitrogen deficient tropical waters by providing additional routes of nitrogen uptake and metabolism. The enzymatic pathway of ammonia assimilation from seawater and the re-assimilation of coral ammonium waste by zooxanthellae was studied by examining the affinity of glutamine synthetase for one of its substrates, ammonia. Glutamine synthetase activity was measured in dinoflagellates of the species Symbiodinium microadriaticum found in symbiotic association with various marine coelenterates. Michaelis-Menten kinetics for the substrate ammonia were determined for freshly isolated dinoflagellates from Condylactis gigantea (apparent NH3 Km=33 M) and for cultured dinoflagellates from Zoanthus sociatus (apparent NH3 Km=60 M). On the basis of the low apparent Kms for NH3, it appears that ammonia assimilation by these symbiotic dinoflagellates occurs via the glutamine synthetase/glutamate synthase pathway. Additionally, the uptake of exogenous ammonium by an intact coelenterate-dinoflagellate symbiosis was strongly inhibited by 0.5 mM methionine sulfoximine, and inhibitor of glutamine synthetase.  相似文献

Juvenile Pacific giant lions-paw scallops Nodipecten subnodosus were fed the toxic dinoflagellate Gymnodinium catenatum, a producer of paralytic shellfish poison (PSP), supplied with Isochrysis galbana (a nontoxic microalgae). Short-term (<24 h) experiments were performed to determine clearance and ingestion rates of G. catenatum. Kinetics of PSP was examined in longer-term experiments (>2 days). At high food concentrations, juvenile scallops showed production of pseudofeces, partial shell valve closure, and reduction in feeding. According to HPLC analysis, the only toxins present in the dinoflagellate G. catenatum and in the scallops were the gonyautoxins (GTXs), except in the labial palps and digestive gland, where trace amounts of saxitoxin (STX) were present in scallops. These tissues could play an important role in toxin biotransformation. The ranking of toxin concentration in tissues was: digestive gland > labial palps > intestine > gills > mantle > adductor muscle, where the total contribution of viscera was more than 80% of the total toxin body burden. Juvenile scallops exhibited no apparent detrimental physiological responses during the long-term feeding experiment. The dinoflagellate may contribute nutrients to the scallop, in addition to the microalgae I. galbana. The dinoflagellate may enhance cell uptake and byssus production. Once PSP accumulated during the first 12 days, it was slowly eliminated. The limited capacity for accumulating toxins in the adductor muscle favors domestic marketing of scallops.  相似文献

Chlorella autotrophica Shihira and Krauss (clone 580), a euryhaline microalga from the marine coastal environment is subject to large fluctuations in external salinity and nitrogen supply. The alga exhibits maximum growth at salinities lower than 100% ASW (artificial seawater). Cells divide faster and show higher cell yields when the supply of either NH 4 + or NO 3 - is increased above 0.2 mM. Cells growing on NH 4 + show high levels of NADPH-glutamate dehydrogenase (GDH) activity, and the levels of glutamine synthetase (GS) are decreased to very low levels under these conditions. Methionine sulfoximine (MSX), an inhibitor of GS, has little effect on cell division and nitrogen assimilation of cells growing on NH 4 + . Cells growing on NO 3 - , however, show marked inhibition (65%) in nitrogen assimilation in the presence of 5 mM MSX. This MSX concentration also causes growth retardation and a progressive decrease in cell protein and nitrogen content. GS is almost completely inhibited by 5 mM MSX in both NH 4 + and NO 3 - -grown cells. Cells growing on NH 4 + maintain high levels of NADPH-GDH activity in the presence of MSX. NADPH-GDH activity in MSX-treated NO 3 - -grown cells increases, and, in the presence of 5 mM MSX, reaches 40% of the level found in NH 4 + -grown cells. These results are consistent with NADPH-GDH providing an alternate pathway for NH 4 + assimilation by this marine Chlorella species.  相似文献

The Atlantic surfclam, Spisula solidissima (Dillwyn), is broadly distributed in sandy sediments of the western North Atlantic between the Gulf of St. Lawrence and the Gulf of Mexico. In the United States, a substantial commercial fishery between Long Island and Cape Hatteras harvests offshore populations of one subspecies, S. s. solidissima. A smaller coastal form, S. s. similis Say (also known as S. s. raveneli Conrad), has a partially sympatric geographic distribution, but differs in several life-history characteristics. DNA sequence variation in mitochondrial cytochrome oxidase I (COI) and in introns at two nuclear calmodulin loci was examined to measure genetic divergence between the two subspecies and to test for population structure among populations of S. s. solidissima. Surfclams were collected from seven localities between 1994 and 2001. Based on both mitochondrial and nuclear DNA variation, the two subspecies of S. solidissima are reciprocally monophyletic, with a net COI divergence of 13.9%, indicating long-term reproductive isolation. The only significant differentiation among populations of S. s. solidissima (based on an AMOVA analysis of COI sequences) was between the Gulf of St. Lawrence and more southerly populations. A long internal branch in the S. s. solidissima genealogy coupled with low haplotype diversity in the northern-most population suggests that populations north and south of Nova Scotia have been isolated from each other in the past, with gene exchange more recently. Populations of S. s. similis from Atlantic and Gulf of Mexico coasts had a net COI divergence of 9.2%. Thus, diversification of Spisula spp. clams in the western North Atlantic involved an early adaptive divergence between coastal and offshore forms, with later barriers to dispersal emerging in the offshore form from north to south and in the coastal form between Atlantic and Gulf of Mexico populations.  相似文献

The microalgal community associated with Eudendrium racemosum, a marine hydroid widely distributed in the Mediterranean Sea, was studied during an annual cycle, at monthly frequency, in a coastal station of the Ligurian Sea. Microalgae were represented mainly by diatoms, which exhibited higher abundance and biomass values between autumn and spring (max 46,752 cells mm−2 and 1.94 μg C mm−2, respectively), while during summer a significant decrease was observed (min 917 cells mm−2 and 0.013 μg C mm−2). High levels of abundance of filamentous cyanobacteria were observed in summer. Spatial distribution of epibiontic microalgae showed a markedly increasing gradient from the basal to the apical part of hydroid colonies. Considering the growth forms of diatom communities, motile diatoms (mainly small naviculoid taxa) were the most abundant in all the periods. Adnate (Amphora and Cocconeis spp.) were distributed mainly in the basal and central part of hydroid colonies and showed two peaks (autumn and summer). Erect forms (mainly Tabularia tabulata, Licmophora spp., Cyclophora tenuis) were mainly distributed in the apical part of the colonies and showed their maximum densities in spring–summer. Tube-dwelling (Berkeleya rutilans, Parlibellus sp.) were observed at low densities throughout the study period, without any significant temporal or spatial variability. Comparing the microalgal communities on marine hydroid to those grown on mimic substrata placed in the sampling station during summer, significantly higher values of abundance were observed in the hydroid, suggesting that microalgae may benefit from the polyp catabolites. This fact was particularly evident for the adnate diatoms, whose temporal trend paralleled the cycle of hydroid host.  相似文献

The response of chlorophyll–protein complexes to super- and sub-saturating photon-flux densities, PFD (250?μmol quanta m?2?s?1 and 40?μmol quanta m?2?s?1, respectively) were analyzed for Symbiodinium microadriaticum Freudenthal, the symbiont of the Caribbean jellyfish Cassiopeia xamachana; S. kawagutii Trench and Blank, the symbiont of the Indo-Pacific scleractinian Montipora verrucosa; and S. pilosum Trench and Blank, the symbiont of the Carribbean zoanthid Zoanthus sociatus. The results indicate that each species exhibits a quantitatively distinct chlorophyll (chl) a distribution among its chl–protein complexes when cultured under standardized high and low light conditions. In response to sub-saturating PFD, the three species differentially increased the cellular concentrations of most of the chl–protein complexes. Increases in P700 (reaction center of Photosystem I) under sub-saturating PFD correlate with an increase in the cellular concentrations of the Photosystem I-enriched complexes. Similarly, increases in photosynthetic unit (PSU) size correlate with an increase in the cellular concentrations of the water-soluble peridinin–chl-a–protein (PCP) complexes and the membrane-bound chl a–chl c 2–peridinin–protein (acpPC) complexes, which together represent the light-harvesting components of this group. In S. microadriaticum, acclimation to sub-saturating PFD uniquely includes the preferential enrichment of the dimeric form of PCP. Under super-saturating PFD, an enrichment in photo-protective xanthophylls was detected in acpPC from S. microadriaticum and S. pilosum, but not from S. kawagutii. Each species demonstrated a characteristic photo-acclimatory response which correlates with its distribution as endosymbiont in nature, supporting the concept that different species of symbiotic dinoflagellates are adapted (sensu Björkman 1981) to different photic environments. Summary The sex pheromone of the grain aphid,Sitobion avenae (Fab.), is identified as (4aS,7S,7aR)-nepetalactone, for which specific olfactory cells are located in the secondary rhinaria of the male antenna. The related (1R,4aS,7S,7aR)-nepetalactol was not produced by females in significant amounts (ca 10,000-fold less than the nepetalactone), although cells in the male antenna were found that responded specifically to this compound. Behavioural activity in the linear track ol-factometer showed attraction of males to the nepetalactone, but not to the nepetalactol.  相似文献

Stereoisomers of 4-methyl-3-heptanol (MH) are pheromone components of several Scolytus bark beetles. The elm bark beetle Scolytus laevis (Coleoptera: Scolytidae) has in previous studies been caught in traps baited with commercial MH containing all four stereoisomers, but the lure has been considered a weak attractant. In this study, we addressed the question whether stereospecific responses by S. laevis to stereoisomers of MH might contribute to its niche separation from other sympatric Scolytus species. Using GC–MS, we analyzed extracts of hindguts and abdomens from male and female S. laevis and the sympatric S. triarmatus. We also tested all four MH-stereoisomers individually and in combinations in the field to determine their role for S. laevis. All four stereoisomers were synthesized via a boronic ester method with 1,2-dicyclohexylethanediol as chiral director. In addition, the (3S,4R)-stereoisomer of MH was prepared through enantioselective, lipase-mediated transesterification of a mixture of the four stereoisomers of MH. Females of both species contained small amounts of syn-MH, and males contained trace amounts of anti-MH. The anti stereoisomer (3R,4S)-MH was attractive to male and female S. laevis, whereas the syn stereoisomer (3S,4S)-MH acted as an inhibitor or deterrent and reduced the catch when added to the attractive isomer. The syn isomer is the main aggregation pheromone component of the larger and sympatric S. scolytus and possibly also of S. triarmatus. The avoidance response of S. laevis to the (3S,4S)-stereoisomer may reduce interspecific competition for host trees.  相似文献

We investigated the mechanisms by which glycerol and glucose enter freshly isolated zooxanthellae (FIZ),Symbiodinium microadriaticum Freudenthal, of the mangrove jellyfish,Cassiopea xamachana Bigelow, and the specific sites of host factor interaction. Glucose entry into FIZ is accomplished by a Na+-dependent symport system driven by an electrochemical gradient generated via a Na+-K+ ATPase. Inhibition of glucose uptake by a low molecular weight fraction of host homogenates [mol. wt<2 kilodaltons (kD)] occurs through the interaction of a putative host factor with the carrier protein and not the ATPase. Glycerol entry is apparently accomplished by simple or facilitated diffusion and is not affected by host homogenate fractions.  相似文献

The morphological expressions of imposex in gastropods have been classified into seven stages (S0–S6) with three types (a, b and c). This typical imposex scheme, however, fails to cover some new findings such as specimens with the proximal vas deferens section passing by the vaginal opening (VDSP) and aborted egg capsules. In the present study, Cantharus cecillei was sampled from the coastal waters of China from May to November in 2003. This species showed all the morphological expressions in the typical imposex scheme with the exception of S1b and S2b. In addition, specimens with VDSP occurred in all stages other than stage 4, and were documented for the first time with scanning electron micrographs or in histological sections. Besides VDSP, which was observed exteriorly, excrescent tissue, e.g. the vas deferens, also originated in the ventral channel or the inner wall of the pallial oviduct was extruded. Based on these new findings, the typical imposex scheme was modified to obtain one valid for C. cecillei. Types a*, b* and c* were added as new types corresponding to types a, b and c, respectively. S4* was also rearranged to correspond to S4; S4*, thus, no longer represented the end of imposex development. The updated scheme was not only valid for describing diverse expressions of imposex development in C. cecillei and Nassarius reticulatus, but could also be generalized to define various forms of VDSP in other species. Histological observation indicated that egg capsules would be blocked by the tissues of the vas deferens section or the extrusion of the pallial oviduct wall. This new mechanism, leading to the sterilization of imposex-affected females, was first conclusively revealed and described in the present scheme.Communicated by T. Ikeda, Hakodate  相似文献

Fe-BEA catalysts are active for the NH3-SCR of NO. For industrial application, a binder should be added to the Fe-BEA catalysts to make them tightly adhere to the monoliths. The addition of alumina and zirconia as binders to the Fe-BEA led to a different effect on NO conversion. The catalytic activity of the mixed samples was evaluated by the temperature programmed procedure in a flow-reactor system, and the mechanism was analyzed via SEM, BET, XRD and XPS. It was found that larger iron particles were formed by the migration of parent iron particles in the Fe-BEA catalyst with alumina. This led to the increase of Fe3+ magnitude and iron cluster, enhancing the abilities of NO oxidation and storage. Accordingly, the SCR activity increased slightly in low temperature but decreased sharply in high temperature. For the Fe-BEA with zirconia sample, NO oxidation and storage abilities decreased due to the less iron clusters. The increase of Fe3+ magnitude resulted in higher catalytic oxidation ability, which gave rise to little change in the SCR activity compared with the Fe-BEA.

Rates of oxygen and carbon-dioxide exhange were measured in symbiotic and aposymbiotic specimens of the sea anemone Anthopleura elegantissima while fed and starved under light or dark conditions. Respiratory quotients indicated that fed anemones switched from a carbohydrate to a fat catabolism when starved, with the exception that symbiotic individuals starved in the light showed a pronounced carbohydrate catabolism for over 1 month. The source of the carbohydrate was probably photosynthate translocated by the dinoflagellate Symbiodinium (=Gymnodinium) microadriaticum (Freudenthal) living in the anemones' tissues. The starved symbiotic anemones maintained in the light had lipid levels not significantly different from fed controls and 44 to 61% higher than starved aposymbiotic anemones after 1 month. Thus, the quality and quantity of the metabolic flux from the symbionts to the sea anemone were sufficient to conserve the host's lipid reserves.  相似文献
