In general, bivalves are not affected by exposure to toxic dinoflagellates that produce paralyzing shellfish poisons (PSP).
After injection with PSP extracted from the Gymnodinium catenatum, Nodipecten subnodosus is paralyzed, indicating that PSP provokes effects similar to what is observed in vertebrates, including paralysis and metabolic
stress. To investigate the processes involved in poisoning by PSP, lions-paw scallops were injected with gonyautoxin (GTX)
2/3 epimers in the adductor muscle. Mild doses provoked adductor muscle contractions and paralysis, mantle retraction, and
incapacity of shell closure, but scallops gradually recovered in a clear, dose-time recovery pattern. With high doses of GTX
2/3, scallops were permanently paralyzed, and hemocytes in hemolymph were reduced. Surprisingly, under these conditions, scallops
continued normal feeding and did not show any microscopic defect in intestine or gills, but hemocytes infiltrated the adductor
muscle and abnormal vitellogenesis and mantle melanization occurred. Paralysis stress was accompanied by negative scallop
responses, based on visible effects, generation of nitric oxide, lipid peroxidation, and changes in antioxidant and hydrolytic
enzymes in hemocytes and tissues. These data can be used to understand potential side effects of PSP in bivalves. 相似文献
As plants constitute the foundation of the food chain, concerns have been raised about the possibility of toxic concentrations of metals and metalloids being transported from plants to the higher food chain strata. In this perspective, the use of important phytotoxicity endpoints may be of utmost significance in assessing the hazardous nature of metals and metalloids and also in developing ecological soil screening levels. The current study aimed to investigate the role of glutathione (GSH) and its associated enzymes in the metabolic adaptation of two grass species namely Eriophorum angustifolium Honck. and Lolium perenne L. to metals and metalloids stress in the vicinity of a chemical industrial complex (Estarreja, Portugal). Soil and plant samples were collected from contaminated (C) and non-contaminated (reference, R) sites, respectively, near and away from the Estarreja Chemical Complex, Portugal. Soils (from 0 to 10 and 10 to 20 cm depths) were analyzed for pH, organic carbon, and metals and metalloids concentrations. Plant samples were processed fresh for physiological and biochemical estimations, while oven-dried plant samples were used for metals and metalloids determinations following standard methodologies. Both soils and plants from the industrial area exhibited differential concentrations of major metals and metalloids including As, Cu, Hg, Pb, and Zn. In particular, L. perenne shoot displayed significantly higher and lower concentrations of Pb and As, respectively at contaminated site (vs. E. angustifolium). Irrespective of sites, L. perenne shoot exhibited significantly higher total GSH pool, oxidized glutathione (GSSG) and oxidized protein (vs. E. angustifolium). Additionally, severe damages to photosynthetic pigments, proteins, cellular membrane integrity (in terms of electrolyte leakage), and lipid peroxidation were also perceptible in L. perenne shoot. Contrarily, irrespective of the sites, activities of catalase and GSH-regenerating enzyme, GSH reductase, and GSH-metabolizing enzymes such as GSH peroxidase and GSH sulfotransferase were significantly higher in shoot of E. angustifolium. Despite the higher total GSH content, L. perenne is vulnerable to multi-metals-induced stress in comparison to E. angustifolium as depicted by increased GSH- and protein oxidation, low reactive oxygen radical-processing potential (exhibited in terms of low catalase activity) and poor GSH pool utilization efficiency (in terms of lower GSH-associated enzymes activities). The outcome of the present study may be significant for understanding vital GSH-mediated metals and metalloids tolerance mechanisms in plants as well as their unsuitability for animal consumption due to higher metals and metalloids burdens. 相似文献
The objective of this work was to analyze the effect of the interaction between feeding strategy and COD/sulfate ratio on the removal efficiency of sulfate and organic matter from a synthetic wastewater. An anaerobic sequencing batch reactor with recirculation of the liquid phase and containing immobilized biomass on polyurethane foam (AnSBBR) was used. The AnSBBR with a total volume of 3.7 L, treated 2.0 L synthetic wastewater in 8-h cycles at 30 ± 1 °C and was inoculated with anaerobic biomass from a UASB. Two feeding strategies were assessed: (a) batch and (b) batch followed by fed-batch. In strategy (a) the reactor was fed in 10 min with 2 L wastewater containing sulfate and carbon sources. In strategy (b) 1.2 L wastewater (containing only the sulfate source) was fed during the first 10 min of the cycle and the remaining 0.8 L (containing only the carbon source) in 240 min. The COD/sulfate ratios assessed were 1 and 3. Based on these values and on the concentrations of organic matter (0.5–11.25 gCOD/L) and sulfate (0.5 and 2.5 gSO42?/L), the sulfate and organic matter loading rates applied equaled 1.5 and 4.5 gSO42?/L d for sulfate and 1.5, 4.5 and 13.5 gCOD/L d for organic matter. After stabilization of the system time profiles were run of monitored parameters (COD, sulfate, sulfide and sulfite). In general, the reactor showed to be robust for use in the anaerobic treatment of wastewaters containing sulfate. Gradual feeding (strategy b) of the carbon source favored sulfate reduction, resulting in sulfate removal efficiencies of 84–98% and organic matter removal efficiencies of 48–95%. The best results were observed under COD/sulfate ratio equal to 1 (loading rates of 1.5 and 4.5 gSO42?/L d for sulfate, and 1.5 and 4.5 gCOD/L d for organic matter). When COD/sulfate ratio was 3 (loading rates of 1.5 and 4.5 gSO42?/L d for sulfate, and 4.5 and 13.5 gCOD/L d for organic matter) the effect of feed mode became less significant. These results show that the strategy batch followed by fed-batch is more advantageous for COD/sulfate ratios near the stoichiometric value (0.67) and higher organic matter and sulfate concentrations. 相似文献
The present study assessed the DNA damage in environmentally exposed volunteers living in seven municipalities in an industrial coal region, through the use of the comet assay with blood cells and the micronucleus test with buccal cells. Blood and buccal smears were collected from 320 male volunteers living in seven cities inserted in a coal region. They were ages of 18 and 50 years and also completed a questionnaire intended to identify factors associated with DNA damage through a Poisson regression analysis. The comet assay detected significant differences in DNA damage in volunteers from different municipalities, and neighboring cities (Pedras Altas, Aceguá, and Hulha Negra) had a higher level of DNA damage in relation to control city. Some of the risk factors associated with identified DNA lesions included residence time and life habits. On the other hand, the micronucleus test did not identify differences between the cities studied, but the regression analysis identified risk factors such as age and life habits (consumption of mate tea and low carbohydrates diet). We conclude that there are differences in the DNA damage of volunteers from different cities of the carboniferous region, but the presence of micronuclei in the oral mucosa does not differ between the same cities. Furthermore, we alert that some related factors may increase the risk of genotoxicity, such as residence location and time, and living and food habits. Finally, we suggest the need for continuous biomonitoring of the population, as well as for investing in health promotion in these vulnerable populations.
The noxious effects of low or effective dose exposure to single or mixed pesticides on macrophage activity and the lymphohematopoietic organs were investigated. Male Wistar rats were orally exposed to dichlorvos, dicofol, endosulfan, dieldrin and permethrin, either as single or combined mixtures during a 28-day study containing eight groups: one group received a semipurified diet (non-treated); two groups received a semipurified diet containing low dose mixture (dieldrin 0.025 mg/kg, endosulfan, 0.6 mg/kg, dicofol 0.22 mg/kg, dichlorvos 0.23 mg/kg, permethrin 5 mg/kg) or an effective dose mixture (dichlorvos 2.3 mg/kg, dicofol 2.5 mg/kg, endosulfan 2.9 mg/kg, dieldrin 0.05 mg/kg and permethrin 25.0 mg/kg), respectively; the other five groups received a semipurified diet containing each single pesticide in effective doses. At sacrifice, the thymus, spleen, mesenteric lymph nodes, Payer's patches and bone marrow were removed for histological analysis. Peritoneal macrophages were obtained to determine the phagocytosis and spreading indexes and tumoral necrosis factor alpha (TNF-α), nitric oxide (NO) and H2O2 production. Exposure to pesticide mixtures did not alter the percentage of macrophage phagocytosis and spreading, TNF-α production or the NO and H2O2 release when compared to the non-treated group. Neither was there any apparent evidence that a pesticide mixture at low or effective doses altered the histological structure of the lymphohematopoietic organs. The findings indicate that short-term treatment with pesticide mixtures did not induce an apparent immunotoxic effect in male Wistar rats. 相似文献
The objectives of this work were to develop biodegradable trays from cassava starch, sugarcane fibers and Na-montmorillonite (Na-MMT) using a baking process and to study the effects of these components on the physical properties, photo- and bio-degradation of the trays. The sample F20 (produced with 20 g fiber/100 g formulation) showed the maximum yield production (100 %). All formulations resulted in well-shaped trays with densities between 0.1941 and 0.2966 g/cm3. The addition of fibers and Na-MMT resulted in less dense and less rigid trays compared to control samples (only starch). The studied processing conditions resulted in good nanoclay dispersion, leading to the formation of an exfoliated structure. The evaluation of the photo-degradation stability of the trays under UV exposure for 336 h showed that a sample produced with a specific combination of fiber and nanoclay (20 g fiber and 5 g nanoclay/100 g formulation) had the highest loss in stress at break (91 %). Biodegradation assays showed that Control trays (starch) and F20 (20 g fiber/100 g formulation) lost a greater percentage of their weight after 90 days of incubation in soil, with losses of up to 85.50 and 82.70 %, respectively. 相似文献
Natural and contaminated waters of the final reaches of the Chubut River (Patagonia, Argentina) were studied to obtain information about river organic matter and effects of domestic and industrial discharges (fishery effluents and sewages). Fluorescence Excitation–Emission Matrices (EEMs) were obtained from samples only filtered (0.45 μm) and diluted, if necessary, to avoid the inner filter effect. In addition, physicochemical parameters were measured to know the quality of the water and the effluents. Results show that EEMs allow a rapid and simple control of the effluents from fisheries and domestic sewage in Chubut River estuary, necessary to take management decisions. 相似文献
Persistent harmful scenarios associated with disposal of radioactive waste, high-background radiation areas and severe nuclear accidents are of great concern regarding consequences to both human health and the environment. Of particular concern is the extracellular DNA in aquatic environments contaminated by radiological substances. Strand breaks induced by radiation promote decrease in the transformation efficiency for extracellular DNA. The focus of this study is the quantification of DNA damage following long-term exposure (over one year) to low doses of natural uranium (an alpha particle emitter) to simulate natural conditions, since nothing is known about alpha radiation induced damage to extracellular DNA. A high-resolution Atomic Force Microscope was used to evaluate DNA fragments. Double-stranded plasmid pBS as a model for extracellular DNA was exposed to different amounts of natural uranium. It was demonstrated that low concentrations of U in water (50 to 150 ppm) produce appreciable numbers of double strand breaks, scaling with the square of the average doses. The importance of these findings for environment monitoring of radiological pollution is addressed. 相似文献
In the present study, the photocatalytic degradation of five sulfonylurea herbicides (chlorsulfuron, flazasulfuron, nicosulfuron, sulfosulfuron and triasulfuron) has been investigated in aqueous suspensions of zinc oxide (ZnO), tungsten (VI) oxide (WO3), tin (IV) oxide (SnO2) and zinc sulfide (ZnS) at pilot plant scale under natural sunlight. Photocatalytic experiments, especially those involving ZnO photocatalysis, showed that the addition of semiconductors in tandem with the oxidant (Na2S2O8) strongly enhances the degradation rate of the herbicides in comparisons carried out with photolytic tests. The degradation of the herbicides follows a first order kinetics according to the Langmuir-Hinshelwood model. In our conditions, the amount of time required for 50% of the initial pesticide concentration to dissipate (t½) ranged from 8 to 27 min (t30W = 0.3-1.2 min) for sulfosulfuron and chlorsulfuron, respectively in the ZnO/Na2S2O8 system. None of the studied herbicides was found after 120 min of illumination (except chlorsulfuron, 0.2 μg L−1). 相似文献
