In recent years, the exposure of humans to phthalate esters through environmental contamination has increased. One among them is di-ethyl phthalate (DEP), which is used as a plastisizer for cellulose ester plastic films and sheets, solid rocket propellants, molded and extruded articles, as a component in insecticide sprays and various other substances, as well as in industrial applications. Release into the environment occurs primarily as a result of production and manufacturing of DEP and during the use and disposal of products containing DEP. Therefore, a study was undertaken to evaluate gender-specific toxicity of DEP in Wistar rats. Rats of both sexes, weighing 125–130?g, were administered 50?ppm (w/v) DEP in water ad libitum for a period of 180 days and were given normal diet. Control animals received normal diet and water ad libitum. During the treatment, rats were weighed every week and water consumption per day was measured. After the completion of treatment, liver weight?:?body weight?1 ratio, liver weight, body weight?1, liver and serum enzymes, and other biochemical parameters of liver and serum were assessed. It was observed that there was no significant change in body weight?1, liver weight, liver weight?: body weight?1 ratio, and water consumption in both sexes. There were significant increases in liver acid phosphatase (ACP) activity and kidney glutathione levels, and nonsignificant changes in liver alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), succinate dehydrogenase (SDH), and lactate dehydrogenase (LDH) activities in DEP-treated male rats, whereas in DEP-treated female rats the liver showed significant decrease in ALP and SDH and nonsignificant changes in AST, ALT, and LDH activities. Serum ACP and LDH levels in DEP-treated male rats were significantly decreased, and in the case of DEP-treated female rats, only serum LDH levels were significantly decreased. There was no significant change in serum ALP, AST, and SDH levels in DEP-treated male and female rats as compared to control rats. Histology of the livers of both male and female rats showed loss of hepatic architecture, degenerative changes in hepatocytes, and vacuolation in hepatocytes in both the centrilobular and periportal areas. It can be concluded from this study that prolonged exposure to DEP at 50?ppm levels can be harmful to animals and humans. This is evident from the present study as certain significant changes in enzyme activities in the liver, serum, and histological alterations in liver were observed.  相似文献

研究了苯氯乙酮(CN)对大白鼠灌胃亚慢性毒性试验和对藻类急性毒性试验,分别获得了苯氯乙酮对大白鼠最小有伤害剂量为2.032mg·kg-1,最大无伤害作用剂量为0.812 mg·kg-1,对藻类半数生长抑制浓度(96 h EC50)为0.360 mg·L-1,并运用EPA水环境质量基准方法及欧盟风险评价技术指南的相关方法,推算出水环境中CN的环境安全阈值为0 36 μg·L-1.  相似文献

为探讨邻苯二甲酸二乙基己酯[di-(2-ethylhexyl)phthalate,DEHP]在诱发哮喘和哮喘发病机理方面的作用,将Wistar大鼠随机分为正常组、卵清白蛋白(OVA)致敏组和DEHP染毒组,每组8只.用OVA致敏加激发的方法制作大鼠哮喘模型.DEHP染毒组分为2个组,每天分别给予0.7mg·kg-1和70mg·kg-1邻苯二甲酸二乙基己酯灌胃,连续30d.OVA致敏组、DEHP染毒组大鼠均在第31d给予1%OVA雾化,诱发哮喘.第38d测定大鼠在不同氯化乙酰甲胆碱(MCH)剂量下的肺功能,进行支气管肺泡灌洗液(BALF)的嗜酸性粒细胞(EOS)计数.结果显示,与OVA组相比,DEHP染毒组气道高反应性增强,嗜酸性粒细胞比例明显增多(P<0.01).因此,邻苯二甲酸二乙基己酯可增强哮喘模型大鼠气道高反应性和嗜酸性粒细胞浸润,可诱导哮喘的发生.  相似文献

采用表达芯片研究铅(Pb2+)致高血压大鼠模型病变主动脉的基因表达谱,以及可能涉及的信号转导途径.结果表明,Pb2+致高血压大鼠和对照组相比,在3463个已知基因和表达序列标签(EST)中,有24个基因和1个EST表达上调幅度≥2倍;有2个基因和1个EST表达下调幅度≥2倍.在差异表达≥2倍的26个基因中,有13个上调的基因与细胞凋亡、局部粘附、细胞生长、增殖、细胞迁移、平滑肌兴奋、胶原重构等生物学过程相关;2个下调基因与能量代谢有关.表达差异基因相对集中的信号途径是与细胞运动、增殖和存活关系密切的局部粘附信号途径.  相似文献

为了研究大鼠吸入甲醛后其肺组织病理结构的改变并探讨其损伤机理,以雌性Wisar大鼠为实验材料,采用动式吸入方式染毒15d(暴露组甲醛浓度32~37mg·m-3,每日暴露4h)后,取肺组织进行光镜及透射电镜观察.光镜观察结果表明,甲醛暴露后,大鼠肺组织主要表现为间质性肺炎和肺泡性肺炎的病理学特点,肺泡壁毛细血管扩张、充血、出血严重,个别区域可见大量炎细胞浸润,肺内细支气管和肺泡腔内见大量血细胞,部分肺泡腔内有透明膜形成,内含脱落的肺泡上皮细胞.电镜观察结果表明,甲醛暴露后,大鼠肺泡上皮细胞损伤严重,线粒体嵴断裂、肿胀、空泡化,部分肺泡上皮细胞胞膜溶解,细胞器丢失,细胞坏死、脱落.推测甲醛吸入可致大鼠肺病理损伤,损伤机理可能与自由基损害和醛糖还原酶(AR)表达变化有关.  相似文献

The joint action of pyrethroids, lambda-cyhalothrin (LC) in combination with organophosphates, fenitrothione (FNT) on antioxidant defense system and lipid peroxidation biomarkers in rat testes was studied. The results suggest that incubation of testes homogenate with different concentrations of insecticide mixture for different time intervals significantly decreased the activity of antioxidant enzymes, like glutathione S-transferase (GST), superoxide dismutase (SOD) and catalase (CAT), and the level of reduced glutathione (GSH). In addition, a significant inhibition in transaminases (AST, ALT), phosphatases (AcP, AlP) activity and protein content were observed. On the other hand, FNT plus LC increased the cellular lipid peroxidation (LPO) level and the activity of lactate dehydrogenase (LDH). In conclusion, the use of insecticides mixture might cause marked oxidative damage in a concentration and time-dependent manner.  相似文献

为了了解常规灭鼠下城市小型兽类的种类组成和数量季节消长,通过分析浙江省杭州、宁波、绍兴、金华、湖州和萧山等6个城市1997-1999年灭鼠情况和3年的监测数据,结果表明,其种类组成啮齿目的的褐家鼠、黄胸鼠、小家鼠、黑线姬鼠和食虫目的臭QuQing;其中袍家鼠占42.9%、小家鼠32.1%、黄胸鼠10.6%、黑线姬鼠6%、臭QuQing8.3%;这些动物的密度季节变化呈现单峰型或双峰型。每年3-4月和9-10月开展两次灭鼠,能有效地控制小型兽类的密度,但应注意灭鼠药的轮换和灭鼠药的发放形式。  相似文献

具有致癌性的抗氧化剂N—苯基—2-萘胺(P2NA)和N—苯基—1-萘胺(PINA)在体外籍新鲜分离的大鼠肝细胞进行生物转化.肝细胞培育后的产物经TLC和HPLC分析表明,二者主要代谢产物为酚类.P2NA生成二个酚类化合物,经与标准样品作平行层析证实为6-羟基P2NA和4'-羟基—P2NA,而P1NA仅生成一个酚类化合物.P2NA经去苯基反应的代谢产物2-萘胺未被检出.  相似文献
