刺芹侧耳对孔雀石绿的脱色降解及其产物分析   总被引:2,自引:0,他引:2

以白腐真菌刺芹侧耳(Pleurotus eryngii-Co007)为染料脱色菌株,研究三苯甲烷染料孔雀石绿的浓度、脱色p H、脱色温度及脱色时间对染料脱色的影响,并对降解产物进行紫外-可见吸收光谱分析、红外光谱分析、GC-MS分析和植物毒性实验,以揭示孔雀石绿可能的降解路径及其产物毒性.结果表明:在p H 6、30℃条件下,P.eryngii-Co007脱色降解200 mg·L-1孔雀石绿,9 h脱色率可达98.22%;孔雀石绿的降解产物主要包括4-(二甲氨基)二苯甲酮、4-(甲氨基)二苯甲酮和4-氨基二苯甲酮;推测孔雀石绿可能的降解路径为孔雀石绿中心碳的羟基化反应,随后中心碳迅速发生碳-碳键断裂,产生4-(二甲氨基)二苯甲酮,4-(二甲氨基)二苯甲酮经过两个连续的N-去甲基化过程,分别产生4-(甲氨基)二苯甲酮和4-氨基二苯甲酮;植物毒性实验表明,P.eryngii-Co007对孔雀石绿有较好的脱毒作用.综上,P.eryngii-Co007能高效脱色降解高浓度的孔雀石绿,同时可显著降低染料对植物的毒害作用.  相似文献

本研究以城市剩余污泥预处理液作为底物,调节底物不同C/N比,测定了厌氧发酵过程中几种关键酶的酶活及发酵产物乙酸、丙酸及丁酸的浓度.同时,利用Matlab软件通过回归分析和函数拟合构建了C/N比-关键酶活-酸产量之间的多项式函数模型,发现所建立的二元三次多项式模型的拟合优度R2均大于0.9,且残差平方和较小,因此,判定二元三次多项式更适合描述C/N比-关键酶活-酸产量的数学关系.最后,建立了关键酶活、C/N比和产酸量三因素间的曲面模型,发现该模型能够很好地描述污泥厌氧发酵中C/N比条件对关键酶和产酸类型的影响,并能进一步预测C/N比所对应的酶活和产酸发酵类型,可为今后的实验研究及工程放大提供理论参考.  相似文献

污水处理厂污泥产生量日益增加,对环境的影响倍受关注。污泥除了含有大量的葡萄糖、蛋白质等有机物外,还包括大量的微生物,具有厌氧发酵产氢的潜能。通过批式实验系统研究了热处理污泥厌氧发酵产氢情况。研究结果表明,经过适当热处理,可以抑制耗氢菌,同时能保持产氢菌的活性,另外,对污泥还有一定的融胞作用,使污泥中溶解性的糖和蛋白质的含量增加,提高预处理污泥的产氢效率;最佳的热处理温度为75℃,处理后污泥进行厌氧发酵产氢的最大累积产氢量为18.32 mL,比产氢率3.49 mL/g VS。  相似文献

Triclosan is an antimicrobial agent, an endocrine disrupting compound, and an emerging contaminant in the environment. This is the first study investigating triclosan biodegradation potential of four oxygenase-expressing bacteria: Rhodococcus jostii RHA1, Mycobacterium vaccae JOB5, Rhodococcus ruber ENV425, and Burkholderia xenovorans LB400. B. xenovorans LB400 and R. ruber ENV425 were unable to degrade triclosan. Propane-grown M. vaccae JOB5 can completely degrade triclosan (5 mg L−1). R. jostii RHA1 grown on biphenyl, propane, and LB medium with dicyclopropylketone (DCPK), an alkane monooxygenase inducer, was able to degrade the added triclosan (5 mg L−1) to different extents. Incomplete degradation of triclosan by RHA1 is probably due to triclosan product toxicity. The highest triclosan transformation capacity (Tc, defined as the amount of triclosan degraded/the number of cells inactivated; 5.63 × 10−3 ng triclosan/16S rRNA gene copies) was observed for biphenyl-grown RHA1 and the lowest Tc (0.20 × 10−3 ng-triclosan/16S rRNA gene copies) was observed for propane-grown RHA1. No triclosan degradation metabolites were detected during triclosan degradation by propane- and LB + DCPK-grown RHA1. When using biphenyl-grown RHA1 for degradation, four chlorinated metabolites (2,4-dichlorophenol, monohydroxy-triclosan, dihydroxy-triclosan, and 2-chlorohydroquinone (a new triclosan metabolite)) were detected. Based on the detected metabolites, a meta-cleavage pathway was proposed for triclosan degradation.  相似文献

We combined Bakker and Demerouti's spillover–crossover model with Taylor's biobehavioral perspective, tested the comprehensive model, and pursued a set of gender‐related research questions. Negative work interactions were expected to entail two strain responses, high‐ and low‐arousal negative affect. Both should be related to cortisol secretion but transmitted via different social pathways, a positive and a negative one. During a 7‐day ambulatory assessment with 56 couples, we assessed daily variations in the severity of negative social interactions at work and at home along with participants' affect and cortisol levels. Using multilevel structural equation modeling, we found evidence for the three hypothesized processes: strain at work as a consequence of social stress, spillover of strain into the home, and crossover to the partner. On socially more stressful days, participants showed increased high‐ and low‐arousal negative affect at work. Low‐arousal negative affect spilled over into the home. Only for men, high‐arousal negative affect spilled over, and only women showed a tendency for slowed decline of cortisol levels on more socially stressful days (i.e., slower recovery). Surprisingly, high‐arousal negative affect at work tended to be negatively related to partners' high‐arousal negative affect. Commonalities predominated differences between men and women. Enzymes encoded by genes biodegrading microcystins (MCs) can help reveal the function of genes and biodegradation pathway of MCs. Here the first and important gene (USTB-05-A, 1,008 bp) involved in biodegradation of microcystin-RR (MC-RR) was cloned from Sphingopyxis sp. USTB-05 and firstly expressed in Escherichia coli BL21 (DE3) with an expression vector of pGEX4T-1 successfully. The nucleotide sequences of cloned USTB-05-A possessed 92.5% homology to that of mlA reported in Sphingomonas sp. strain ACM-3962. The deduced amino acid sequences containing the cleavage sites of 26th (alanine) and 27th (leucine) showed 83% identical to that of MlrA. The cell-free extract (CE) of recombinant E. coli BL21 (DE3) containing USTB-05-A had high activity for biodegrading MC-RR. Initial MC-RR of 40 mg L−1 was completely biodegraded under total protein of 350 mg L−1 within 0.25 h. A product derived from MC-RR appeared distinctly with the decrease of MC-RR peak on the profile of HPLC. The product (m/z 1056.5) had molecular weight of 18 higher than that of MC-RR (m/z 1038.7). The findings provided the positive evidences that biodegradation of MC-RR began with the breakage of cyclic MC-RR and then it was converted to linear MC-RR as the first product catalyzed by first enzyme of Sphingopyxis sp. An activated carbon (AC) containing a high concentration (374 mg g−1) of Fe was prepared by carbonization of an ion-exchange resin. To examine its chemical reactivity as a catalyst to decompose 2,2′,4,4′,5,5′-hexachlorobiphenyl (PCB-153), the decomposition parameters of temperature and time were varied under air or N2. Decomposition at 350 °C was achieved within 15 min under air and 30 min under N2, and the efficiency of PCB-153 decomposition was 99.7% and 98.0%, respectively. An analysis of inorganic chloride ions revealed that PCB-153 was mineralized effectively during the decomposition. The Brunauer-Emmett-Teller (BET) surface area and pore volume of the AC were measured to assess the adsorption capacity before and after the decomposition. The differences between decomposition under air and N2 reflected the differences in the BET surface and pore volume measurements. A decomposition pathway was postulated, and the reactive characteristics of chlorine atoms loaded on the benzene rings followed the order of para meta ortho, which agrees with the calculated results from a density functional theory study.  相似文献

Abstract

Lactobacillus amylovorus, an amylolytic species, was cultured in increasing concentrations of sweet potato starch to test the effect of this progressive acclimation on lactic acid production. This research is part of a project on the use of the waste stream from a sweet potato cannery to produce lactic acid. The media used for this acclimation was a modified version of the de Man, Rogosa and Sharpe medium, in which glucose was partially or totally substituted with sweet potato starch. The process was done in five steps, starting with 100% glucose in the first step and ending with 100% sweet potato starch in the last one. At each step, the effectiveness of the acclimation was tested by running fermentations with and without pH control for 62?h. The effect of the overall adaptation process was tested by comparing the growth and activity of the acclimated vs non-acclimated bacteria using sweet potato starch as the only source of carbohydrates. Growth and activity assessments indicated that L. amylovorus was able to ferment sweet potato starch into lactic acid. In most cases, pH control resulted in better substrate utilisation and larger amounts of lactic acid. In the comparison study, however, the adaptation process had a major influence on lactic acid production than the effect of pH. For 20?g L–1 sweet potato starch media, adapted L. amylovorus under no pH control yielded 11.20?g L–1 versus the non-adapted bacteria, which yielded 7.10?g L–1. Under controlled pH conditions, 14.80 and 4.20?g L–1 lactic acid were produced by adapted and non-adapted bacteria respectively.  相似文献
