Abundances of virus-like particles (VLPs, mostly bacteriophages) are high in aquatic environments; therefore, techniques for precise enumeration are essential in ecological monitoring. VLPs were determined after staining with SYBR Gold by conventional epifluorescence microscopy and compared to enumerations performed by confocal laser scanning microscopy (CLSM). In order to assess the potential of CLSM for viral direct counts (VDCs), we processed samples from different freshwater and marine systems. Optical sectioning by CLSM and production of an overlay picture of multiple scans enables the often uneven whole investigated filter area to be brought to the plane of focus. This allows for subsequent image analysis of digitally created high-quality images. Another advantage using the CLSM was that the short spot excitation of the stain via laser beam minimized fading of the stain. The VDC results show that there is no significant difference between the two methods. Regarding the known difficulties of viral abundance estimates on particulate material, CLSM was further applied to enumerate VLPs on a small set of marine transparent exopolymeric particles sampled from the Atlantic Ocean. Our data suggest that CLSM is a useful tool to count viruses in water samples as well as attached to certain types of aquatic aggregates.  相似文献

Design for the Environment (DFE) is a systems-oriented approach for designing more ecologically and economically sustainable product systems. It couples the product development cycle used in business with the physical life-cycle of a product. DFE integrates environmental requirements into the earliest stages of design so total impacts caused by product systems can be reduced. With DFE practices, environmental, performance, cost, cultural, and legal requirements are balanced. Concepts such as concurrent design, total quality management, cross-disciplinary teams, and multiattribute decision making are essential elements of DFE.1 In this article, the authors show how these DFE initiatives can provide the design and product development teams with the training, information, decision support tools, and procedures that ensure improved product development decisions and environmental performance.  相似文献

The lifespan of electric and electronic equipment is becoming shorter and the amount of related waste is increasing. This study aimed to contribute to the knowledge about qualitative and quantitative characteristics of such wastes in Greece. Specifically, results are presented from a field survey, which took place in the city of Thessaloniki, Greece, during the year 2002. The survey was conducted with suitable questionnaires in department stores and in households of various municipalities. Household appliances were grouped as follows: (A) large (refrigerator, freezer, washing machine, clothes dryer, electric cooker, microwave oven, electric heater), (B) small (vacuum cleaner, electric iron, hair dryer), (C) information technology and telecommunication equipment (PC, laptop, printer, phone) and (D) consumer equipment (radio, TV, video, DVD, console). The analysis indicated that the lifespan of all new goods is gradually reducing (apart from refrigerators, for which the lifespan was surprisingly found to be increasing) and provided linearized functions for predicting the lifespan, according to the year of manufacture, for certain large appliances.  相似文献
