Various pretreatments methods including sonication and grinding were performed on red seaweed Gelidium amansii for the subsequent extraction of agarose. The agarose products are usually extracted from agar powder products from seaweeds. In this study, the agarose was extracted using a direct polyethylene glycol (PEG) method without the need to first process the agar from seaweed. The agar extract was frozen then thawed and mixed directly with PEG solution to precipitate the agarose. The quality of agarose obtained was evaluated through physico-chemical properties analysis which includes spectral technique (FTIR), melting and boiling point, gel strength and sulfate content. These properties were compared with a non-pretreated sample and it was found that the addition of pretreatment steps improved the quality of agarose but gave a slightly lower yield. The gel strength of pretreated samples was much higher and the sulfate content was lower compared to non-pretreated samples. The best pretreatment method was sonication which gave gel strength of 742 g cm-2 and sulfate content of 0.63%. The extraction of agarose can be further improved with the use of different neutralizing agents. Pretreating the seaweed shows potential in improving the quality of agarose from seaweed and can be applied for future extraction of the agarose.

Influence of ultrasonication on anaerobic bioconversion of sludge.   总被引:1,自引:0,他引:1  
The influence of ultrasonication on hydrolysis, acidogenesis, and methanogenesis in anaerobic decomposition of sludge was investigated. The sonicated sludge exhibited prehydrolysis and preacidogenesis effects in the anaerobic decomposition process. First-order hydrolysis rates increased from 0.0384 day(-1) in the control digester to 0.0672 day(-1) in the digester fed, with sludge sonicated at 0.52 W/mL. The sonication appeared to be ineffective in relation to acidogenesis reaction rates, but it provided a better buffering capacity to diminish the adverse effect of acidification. Digesters fed with sonicated sludge demonstrated enhanced methanogenesis over the control unit. Determination by coenzyme F420 verified that sonication is able to promote the growth of methanogenic biomass and facilitate a positive methanogenic microbial development in suppressing the initial methanogenesis limitation. The results suggest that ultrasonication could enhance anaerobic decomposition of sludge, resulting in an accelerated bioconversion, improved organics degradation, improved biogas production, and increased methane content.  相似文献

Numerous studies have demonstrated that anaerobic packed-bed reactors (APBRs) containing media such as clay chip, small stone, and porous plastic modules are effective for treating a variety of organic wastes. Most of these studies focused on the treatment capabilities of the processes, yet little work was done about the factors affecting their designs and performance. This paper presents the results of a study on a laboratory scale APBR system examining the significance of media factors such as media roughness, specific surface area, porosity and pore size on treatment performance. The results showed that media roughness and pore size were more important over surface area on the reactor performance. Within the same media porosity, pore size plays a more significant role than surface area in term of COD removal efficiency. The study indicates that the ability of a support medium to retain biomass either as suspended growth entrapped in the void space, or by attachment to the media surface is a significant consideration in an upflow APBR. A testing protocol for measuring methane gas produced by the suspended biomass using a batch serum bottle technique was developed and presented in this study. The method involved measurements of hourly methane production and instantaneous TOC concentration from the suspended biological solids extracted from various reactor heights. The proportion of methane production associated with the suspended biomass was as high as 58% at organic loading rate of 16 g COD/L.day. The results suggest that media pore size and porosity play a significant role in the performance of upflow APBRs.  相似文献

The formation of gas hydrates is a major issue during the operation of oil and gas pipelines, because gas hydrates cause plugging, thereby disrupting the normal oil and gas flows. A solution is to inject gas hydrate inhibitors such as ionic liquids. Contrary to classical inhibitors, ionic liquids act both as thermodynamic inhibitors and hydrate inhibitors, and as anti-agglomerates. Imidazolium-based ionic liquids have been found efficient for the inhibition of CO2 and CH4 hydrates. For CO2 gas hydrates, N-ethyl-N-methylmorpholinium bromide showed an average depression temperature of 1.72 K at 10 wt% concentration. The induction time of 1-ethyl-3-methyl imidazolium bromide is 36.3 h for CO2 hydrates at 1 wt% concentration. For CH4 hydrates, 1-ethyl-3-methyl-imidazolium chloride showed average depression temperature of 4.80 K at 40 wt%. For mixed gas hydrates of CO2 and CH4, only quaternary ammonium salts have been studied. Tetramethyl ammonium hydroxide shifted the hydrate liquid vapour equilibrium to 1.56 K at 10 wt%, while tetrabutylammonium hydroxide showed an induction time of 0.74 h at 1 wt% concentration.

ABSTRACT: Field data collected from the Feitsui Reservoir in Taiwan indicate that the water is at a stage between mesotrophic and eutrophic. Recent measurements such as total phosphorus, chlorophyll a and Secchi depth levels suggest that the water quality has been declining. A two‐dimensional, laterally averaged, finite difference hydrodynamic and water quality model was used to simulate the water quality in the Feitsui Reservoir in an effort to determine sound water quality management strategies. The model was calibrated and verified using data collected in 1996 and 1997. The calibrated model was used to simulate algal biomass (in terms of chlorophyll a) levels under various wasteload reduction scenarios. Model results show that 50 percent reduction of the total phosphorus load will improve the existing water quality, shifting the trophic status from eutrophic/mesotrophic to oligotrophic. The modeling effort has yielded valuable information that can be used by decision makers for the evaluation of different management strategies of reducing watershed nutrient loads.  相似文献
