The use of molecular tools, principally qPCR, versus traditional culture-based methods for quantifying microbial parameters (e.g., Fecal Indicator Organisms) in bathing waters generates considerable ongoing debate at the science–policy interface. Advances in science have allowed the development and application of molecular biological methods for rapid (~2 h) quantification of microbial pollution in bathing and recreational waters. In contrast, culture-based methods can take between 18 and 96 h for sample processing. Thus, molecular tools offer an opportunity to provide a more meaningful statement of microbial risk to water-users by providing near-real-time information enabling potentially more informed decision-making with regard to water-based activities. However, complementary studies concerning the potential costs and benefits of adopting rapid methods as a regulatory tool are in short supply. We report on findings from an international Working Group that examined the breadth of social impacts, challenges, and research opportunities associated with the application of molecular tools to bathing water regulations.  相似文献

Stable isotope probing (SIP) was used to identify microbes stimulated by ethanol addition in microcosms containing two sediments collected from the bioremediation test zone at the US Department of Energy Oak Ridge site, TN, USA. One sample was highly bioreduced with ethanol while another was less reduced. Microcosms with the respective sediments were amended with 13C labeled ethanol and incubated for 7 days for SIP. Ethanol was rapidly converted to acetate within 24 h accompanied with the reduction of nitrate and sulfate. The accumulation of acetate persisted beyond the 7 d period. Aqueous U did not decline in the microcosm with the reduced sediment due to desorption of U but continuously declined in the less reduced sample. Microbial growth and concomitant 13C-DNA production was detected when ethanol was exhausted and abundant acetate had accumulated in both microcosms. This coincided with U(VI) reduction in the less reduced sample. 13C originating from ethanol was ultimately utilized for growth, either directly or indirectly, by the dominant microbial community members within 7 days of incubation. The microbial community was comprised predominantly of known denitrifiers, sulfate-reducing bacteria and iron (III) reducing bacteria including Desulfovibrio, Sphingomonas, Ferribacterium, Rhodanobacter, Geothrix, Thiobacillus and others, including the known U(VI)-reducing bacteria Acidovorax, Anaeromyxobacter, Desulfovibrio, Geobacter and Desulfosporosinus. The findings suggest that ethanol biostimulates the U(VI)-reducing microbial community by first serving as an electron donor for nitrate, sulfate, iron (III) and U(VI) reduction, and acetate which then functions as electron donor for U(VI) reduction and carbon source for microbial growth.  相似文献

Concepts of sustainable development have stimulated innovation in the delivery of environmental management. In particular, new partnership approaches have been developed in recognition of the need to adopt more holistic perspectives and facilitate multi-sectoral and cross-sectoral working. River catchments are complex systems and have been one particular focus of experimentation in environmental management mechanisms. This paper provides a brief overview of river management experience in the UK, charting changing approaches in terms of scope and organisation since the 1970s. This sets the context for a more detailed examination of the Mersey Basin Campaign and, in particular, its River Valley Initiatives. The paper concludes with an evaluation of the merits of this approach in relation to sustainable river management.  相似文献

Introduced and cryptogenic species in Port Phillip Bay, Victoria, Australia   总被引:4,自引:0,他引:4  
Port Phillip Bay (PPB) is a large (1,930 km2), temperate embayment in southern Victoria, Australia. Extensive bay-wide surveys of PPB have occurred since 1840. In 1995/1996 the Commonwealth Scientific and Industrial Research Organization (CSIRO) Centre for Research on Introduced Marine Pests (CRIMP) undertook an intensive evaluation of the region with the aims of developing a comprehensive species list of native and introduced biota and contrasting previous bay-wide assessments with a current field survey in order to detect new incursions and discern alterations to native communities. Two methods were used to meet these aims: a re-evaluation of regional museum collections and published research in PPB to identify and determine the timing of introductions; and field surveys for benthic (infauna, epifauna and encrusting) organisms between September 1995 to March 1996. One hundred and sixty introduced (99) and cryptogenic (61) species were identified representing over 13% of the recorded species of PPB. As expected, the majority of these are concentrated around the shipping ports of Geelong and Melbourne. Invasions within PPB appear to be increasing, possibly due to an increase in modern shipping traffic and an increase in aquaculture (historically associated with incidental introductions); however the records of extensive biological surveys suggest that this may, in part, be an artefact of sampling effort. In contrast to Northern Hemisphere studies, PPB (and Southern Hemisphere introductions in general) have significantly different suites of successfully invading taxa. Early non-invasive fetal sexing is widely available over the Internet, leading to concerns about its possible use for sex selection. The aim of this review is to summarise the results of surveys describing lay attitudes towards sex selection for non-medical reasons to help address or inform such concerns. A search of electronic databases and key journals was supplemented by an Internet search and citation-tracking. Twenty-one quantitative studies were identified. Most were conducted in the US, with five recent studies in the UK and Germany. Most studies found that, overall, people were not in favour of sex selection. However, this varied from 94 to 18%. People may hold more negative attitudes when the method of sex selection is specified, particularly if this involves termination of pregnancy (TOP). Attitudes towards the general availability of sex selection are less negative than those towards personal use. Attitudes were consistently negative in German surveys. Little is known of such attitudes outside the US, the UK and Germany, and how such attitudes may change as new sex selection technologies become available. Studies of use of early non-invasive fetal sexing for sex selection for non-medical reasons are needed to assess the extent to which concerns about usage are justified and to assist in debates on how these might be most appropriately addressed. We report the finding of paternal isodisomy for chromosome 14 in a fetus found to have a der(14;14)(q10;q10) by amniocentesis. The pregnancy was complicated by severe polyhydramnios and elevated amniotic fluid alpha-fetoprotein (AFP). The infant showed features consistent with paternal uniparental disomy (UPD) including postnatal growth retardation, poor respiratory function, feeding difficulties, and evidence of hypertrophic cardiomyopathy. The present case, in addition to other reported cases of UPD involving balanced acrocentric rearrangements, supports testing for UPD in prenatally detected Robertsonian translocations and isochromosomes. We report the prenatal diagnosis of a fetus with a de novo Robertsonian translocation: 45,XY,der(15;15)(q10;q10). Although Robertsonian translocations are common chromosomal rearrangements, those involving homologous chromosomes are infrequent. Since chromosome 15 is imprinted, uniparental disomy (UPD) is a concern when chromosomal rearrangements involving chromosome 15 are identified. In the present case, UPD studies showed normal biparental inheritance. In contrast to the fact that most homologous acrocentric rearrangements are isochromosomes, these results indicate postzygotic formation of a Robertsonian translocation between biparentally inherited chromosomes 15.
