The aim of this study was to investigate how the worm Polydora sp., which induces oysters into creating mud blisters in response to an irritation within their shells, could interfere with the oyster Crassostrea gigas physiology and ethology. The impact was characterized by studying two groups of oysters (non-parasitized and parasitized) during a 30 days period: (1) the animal behaviour by analysing their valve activity (valvometry), and (2) the animal respiratory physiology by measuring in vivo the oxygen partial pressure and the specific oxygen consumption in selected tissues (heart, fast and slow adductor muscle). We also researched a putative impact on the expression of several oxidative stress genes at the heart level. Our results show that Polydora sp. is clearly an oyster’s parasite as it induces a decrease in oyster growth according to the infestation intensity. Moreover, it modifies the behaviour and the respiratory physiology of the molluscs. Infested animals opened more frequently but for less time and their level of blood oxygenation was systematically higher than healthy molluscs. These high levels of oxygenation had no effect on the oxidative metabolism of the tissues studied but they induced oxidative stress. Indeed, the superoxide dismutase gene showed a threefold increase in expression in the heart of infested oysters. A putative scenario of the weakening mechanism is proposed.  相似文献

Chlordecone (CLD) is a chlorinated hydrocarbon insecticide, now classified as a persistent organic pollutant. Several studies have previously reported that chronic exposure to CLD leads to hepatotoxicity, neurotoxicity, raises early child development and pregnancy complications, and increases the risk of liver and prostate cancer. In situ chemical reduction (ISCR) has been identified as a possible way for the remediation of soils contaminated by CLD. In the present study, the objectives were (i) to evaluate the genotoxicity and the mutagenicity of two CLD metabolites formed by ISCR, CLD-5a-hydro, or CLD-5-hydro (5a- or 5- according to CAS nomenclature; CLD-1Cl) and tri-hydroCLD (CLD-3Cl), and (ii) to explore the angiogenic properties of these molecules. Mutagenicity and genotoxicity were investigated using the Ames’s technique on Salmonella typhimurium and the in vitro micronucleus micromethod with TK6 human lymphoblastoid cells. The proangiogenic properties were evaluated on the in vitro capillary network formation of human primary endothelial cells. Like CLD, the dechlorinated derivatives of CLD studied were devoid of genotoxic and mutagenic activity. In the assay targeting angiogenic properties, significantly lower microvessel lengths formed by endothelial cells were observed for the CLD-3Cl-treated cells compared to the CLD-treated cells for two of the three tested concentrations. These results suggest that dechlorinated CLD derivatives are devoid of mutagenicity and genotoxicity and have lower proangiogenic properties than CLD.  相似文献

Mechanisms that can influence the tolerance of hypoxia in brackish waters were studied in resting and fed crabs, Carcinus maenas, at 15?°C. Mortality, blood oxygenation, acid-base status and lactate concentration were analysed in fed crabs held in full-strength normoxic seawater (32.5‰?S) and then transferred for 24?h to a partial pressure of oxygen (Po2) of 3?kPa (1.4?mg?l?1) and various salinities (17, 12.5, 10, 8‰?S). At salinity levels >10‰, fed crabs tolerated Po2 values as low as 3?kPa in the ambient water and 0.5?kPa in their arterial blood for 24?h without switching to anaerobic metabolism. Only below 10‰?S did their blood-lactate content rise, leading to their death despite the fact that their blood O2-content was twice the control value measured in full-strength normoxic seawater and their blood Po2 did not decrease below values recorded at higher salinity levels. Addition of CO2 to 8‰?S water (CO2 partial pressure increasing from 0.1 to 0.3?kPa) decreased blood-lactate production and mortality, suggesting that at 10‰?S impairment of the O2 supply is limited by an excessive blood O2-affinity. The results are discussed in terms of the distribution (?10‰?S) of C. maenas along salinity gradients in estuaries and bays.  相似文献
