In this study, residual concentrations of chlorpyrifos (CPF) were determined in feed (40) and fodder (25) samples collected from various locations of Tarai region of Uttarakhand. For extracting residues, liquid–liquid partition followed by alumina column clean up was used and the detection and quantification of residues was undertaken with the help of high-performance liquid chromatography using C18 column and diode array detector at 220?nm. Of the total 40 feed samples analyzed, 7 (17.5%) samples were found positive for CPF with the mean residual concentration of 0.058?µg?g?1; while out of 25 fodder samples, CPF residues were detected in a single (4%) sample with residual concentration of 0.39?µg?g?1. However, none of the feed or fodder samples contained CPF residues above the prescribed limit.  相似文献

This study investigated whether low-level, long-term in utero and post-natal exposure to organohalogen pollutants disrupts male reproductive organ morphology and testosterone production in Greenland sledge dogs (Canis familiaris), as a model of Arctic top predators feeding on marine mammals. Six male dogs were followed for 1 year and testosterone concentrations, testes/baculum morphology and baculum bone mineral density (BMD) was determined. Three males were exposed to organohalogenated contaminants (OHCs) in utero through maternal dietary intake of minke whale blubber (Balaenoptera acutorostrata), with a post-weaning ∑OHC intake of 10.4–11.7 µg kg?1 day?1 resulting in an adipose tissue range of ∑OHC 4518–5729 ng (g lw)?1 after 1 year. Three control males were exposed to very low concentrations of OHCs through pork fat. No significant differences were seen in plasma testosterone concentrations, baculum weight, BMD, and testicular length in the six male dogs (control, n = 3 and exposed, n = 3) measured at 3, 5, 7, 9, and 12 months of age. Testicular weights were significantly lower in the exposed group (p = 0.015, n = 2). Although this study had a limited number of animals, it was observed that in utero and the following 12 months of chronic exposure to a complex mixture of contaminants in the form of naturally accumulated OHCs does not affects testosterone levels, but possibly affects testicular weights in sledge dogs.  相似文献

Narwhals (Monodon monoceros) from North West Greenland are known to bioaccumulate mercury (Hg) in tissues and internal organs. This is postulated to be a health concern and therefore studies were undertaken to conduct a screening study of Hg concentrations and histopathology of liver and renal tissues in a total of 12 specimens. The sample consisted of two sub-adults (one male, one female) and 10 adult (six males, four females) collected in Qaanaaq (Thule) 2010. In liver, Hg mean ± SD was 11.88 ± 10.47 μg/g ww (range: 0.39?31.8 μg/g ww) while the concentrations in kidneys were 1.85 ± 1.20 μg/g ww (range: 0.41?4.03 μg/g ww). There was no marked difference in Hg concentrations between males and females while sub-adults had significantly lower concentrations. The histological examinations of renal tissue showed glomerular capillary dilatation and basement membrane thickening, dilatation, and hyalinization of Bowman's space/capsule and tubular lesions with hyaline casts accompanied by interstitial fibrosis in the kidneys. In liver tissue, portal cell infiltrates and fibrosis, bile duct proliferation, lipid-filled Ito cells, and steatosis were found. There was no marked difference in histological prevalence between males and females and in Hg concentrations in individuals with or without lesions. Four of seven renal lesions and one liver lesion were found in the two sub-adult whales. Based upon these findings, as well as the nature of the lesions, evidence suggests that histopathological alterations were a result of age but that Hg might be an aggravating co-factor in development of renal lesions in particular.  相似文献

Persistent organic pollutants such as Polychlorinated biphenyls (PCB) and dichloro diphenyl trichloroethane (DDT) are known to exert various adverse health effects in wildlife mammals. The impact from dietary intake of minke whale (Balaenoptera acutorostrata) blubber high in organohalogen and other chemical contaminants on renal morphology and function was investigated via a controlled study on West Greenland sledge dogs (Canis familiaris). Our results showed significantly higher frequencies of glomerular, tubular, and interstitial lesions in the exposed group. Furthermore, higher urine protein?:?creatinine ratio and plasma urea levels were found in the exposed group, which indicated a negative impact on kidney function via tubular and glomerular dysfunctions. The lesions were similar to those observed in top predator marine species–such as seals and polar bears–and humans exposed via diet to persistent environmental pollutants. The lesions may have impacts on the overall health of these animals. It is reasonable to suggest that chronic exposure to organic pollutants via a country diet of high trophic level marine organisms may have an impact on renal morphology and function in Arctic apex predators in general as well as Inuit people.  相似文献

There is inadequate morphological nomenclature and definition of organ pathology when using wild fish in biomonitoring of environmental pollution. The aim of this investigation was to provide a guide that assesses histology of Shorthorn (Myoxocephalus scorpius) and Fourhorn (Myoxocephalus quadricornis) sculpins obtained from a study using these two fish species as bioindicator organisms to evaluate environmental impact attributed to a dumpsite located in East Greenland. Specific histopathological lesions were quantified on the basis of Bernet et al. and all lesions were photomicrographed, evaluated, and placed into specific categories of five reaction patterns. These were circulatory disturbances, regressive changes, progressive alterations, inflammation, and tumors based on anatomical location, type, and severity. This method is postulated to enable an objective assessment of the histological integrity of fish gills and liver; thus, making it possible to compare sculpin pathology and reaction patterns at different locations. Data suggest that this updated histological guide might be used for identification and quantification of histological lesions when applying sculpins in biomonitoring programs in Greenland and other Arctic regions.  相似文献

The effects of persistent organic pollutants on renal and liver morphology in farmed arctic fox (Vulpes lagopus) were studied under experimental conditions. Control animals received a diet containing pork (Sus scrofa) fat with low amounts of persistent organic pollutants, while the diet of the exposed animals contained whale blubber, 'naturally' contaminated with persistent organic pollutants. Polychlorinated biphenyls (PCB) and organochlorine pesticide (OCP) concentrations in the whale blubber were 488 and 395 ng/g wet weight, respectively. Animals were sacrificed and sampled when they were at their fattest (winter) as well as their lowest body weight (summer). The results show that PCB and OCP exposure causes renal (and probably also liver) lesions in arctic foxes. The prevalence of glomerular, tubular and interstitial lesions was significantly highest in the exposed group (chi-square: all p<0.05). The frequency of liver lesions (steatosis, intravascular granulocyte accumulations, interstitial cell infiltrations, lipid granulomas, portal fibrosis and bile duct hyperplasia) were also highest in the exposed group, although not significantly (chi-square: all p>0.05). The prevalence of lesions was not significantly different between lean (winter) and fat (summer) foxes for any of the lesions (chi-square: all p>0.05). We suggest that wild arctic foxes exposed to an environmental cocktail of persistent organic pollutants, such as PCBs and OCPs, in their natural diet are at risk for developing chronic kidney and liver damage. Whether such lesions may have an impact on age and health of the animals remains uncertain.  相似文献

Long-finned pilot whales (Globicephala melas) from the Faroe Islands are known to be heavily polluted with contaminants, such as mercury (Hg) and organochlorine compounds (OC). This is postulated to exert adverse health effects on whales as well as the human population who rely on its meat and blubber as food sources. It was therefore decided to conduct a screening pilot study to determine contaminant concentrations and histopathology of liver and renal tissues in a total of 14 specimens from this subpopulation. In blubber, the mean ∑OC concentration was 31,887 ng g?1 lw (range: 18,170–47,425 ng g?1 lw) of which ∑PCB concentration was 23,416 ng g?1 lw (range: 13,947–34,543 ng g?1 lw; n = 3). In liver, mean Hg concentration was 138 µg?1 ww (range: 54–351 µg g?1 ww; n = 7) and mean cadmium (Cd) concentration 15 µg g?1 ww (range: 7–31 µg g?1 ww; n = 7). In the kidney, the mean Cd concentration was 36 µg g?1 ww (range: 23–47 µg g?1 ww; n = 7). Of the liver Hg concentrations, two were at the suggested toxic threshold levels of 60 µg g?1 ww and five were 2–6-fold above. Liver selenium (Se) : Hg was on average 1.11 (range: 0.97–1.41; n = 7) indicating that Se was in excess, which decreased the risk of acute Hg poisoning. Histopathological examinations showed high prevalence (>35%) of renal glomerular arteriosclerosis, glomerular capillary dilatation, dilatation and hyalinization of Bowman's space/capsule and tubular hyaline casts. In liver tissue, high prevalence was found for portal cell infiltrates, lipid granulomas, hepatocytic lipid accumulation, bile duct proliferation, lipid-filled Ito cells, and focal necrosis. In a single juvenile male, 4 of 8 renal and 3 of 7 liver lesions were present. Cadmium concentrations increased significantly in the presence of glomerular arteriosclerosis and a similar trend was found for tubular hyaline casts. Based on these findings, as well as the nature of the lesions, data indicate that the histopathological changes were a result of age and that contaminants are likely to be the co-factors in the development in at least three renal and four liver lesions.  相似文献

Concentrations of organochlorine (OC) contaminants and histomorphology of liver, kidney, and thyroid tissues were studied in nine adult and one subadult glaucous gulls (Larus hyperboreus) collected at Svalbard on 2 August 2011. Concentrations of liver polychlorinated biphenyls (PCB; range: 150–2820?ng?g?1 ww), dichlorodiphenyltrichloroethane (DDT; range: 58–724?ng?g?1 ww), and chlordanes (CHL; range: 11–126?ng?g?1 ww) dominated the OC profile followed by hexachlorobenzene (HCB; range: 11–42?ng?g?1 ww), mirex (range: 2–52?ng?g?1 ww), and β-hexachlorocyclohexane (β-HCH; range: 1–7?ng?g?1 ww). Histological examination of the liver showed mononuclear cell infiltrations and granulomas in 10 and 6 gulls, respectively, while intense intrahepatic lipid accumulation (steatosis) was found in two and focal necrosis in one gull. In kidney, glomerular sclerosis and adhesions was found in five and one gull, respectively. Thickening of the glomerular basement membranes and tubular necrosis was found in four and seven gulls, respectively, while mononuclear cell infiltrations were found in two individuals. In the thyroid gland, a high density of small follicles accompanied by follicular epithelial cell proliferation was observed in five glaucous gulls. Gulls with hepatic steatosis had significantly higher ΣDDT levels than the gulls without hepatic steatosis and a similar trend was found for ΣPCB. When normalizing OC concentrations for lipid content in liver, gulls with lipid granulomas had significantly lower β-HCH and significantly higher mirex levels, respectively, than gulls without lipid granulomas. Also; gulls with thickening of the glomerular basement membranes had non-significantly higher ΣPCB levels than gulls without. The histological findings were similar to those of controlled laboratory studies and OC-contaminated wildlife (e.g., polar bears; Ursus maritimus) and the data of this study therefore suggest that OC exposure may be a co-factor in the development of organ alterations in glaucous gulls. However, other environmental factors such as age, element exposure, and infectious micropathogens cannot be ruled out as co-factors, and it is uncertain if the tissue changes found exert adverse health effects on the population of Svalbard glaucous gulls.  相似文献

The associations between blood organohalogen contaminant (OHC) concentrations and thyroid gland histology were studied in 10 adult female glaucous gulls (Larus hyperboreus) from the Norwegian Arctic (Bjørnøya) during the incubation period. This histological investigation was undertaken as previous glaucous gull studies from the same area reported negative relationships between circulating OHC concentrations and thyroid hormone levels. Organohalogen concentrations have previously been associated with altered blood plasma thyroid hormone concentrations, as a result of parenchymal thyroid gland alterations and perturbation of the hypothalamic-pituitary-thyroid (HPT)-axis. In this study, PCB (range: 186–1027 ng g?1ww), DDT (77–203 ng g?1ww) and chlordane (18–65 ng g?1ww) concentrations dominated the blood plasma OHC profile in incubating female glaucous gulls. High density of small follicles accompanied by follicular epithelial cell proliferations was seen in thyroid glands in seven of 10 gulls. Focal thyroiditis and nodular hyperplasia were found in two birds. No significant differences in plasma OHC concentrations were noted between gulls exhibiting high density of small follicles and cell proliferations and those birds not showing histological changes. Based on these findings, data suggest that the histological changes in thyroid glands of OHC-contaminated female glaucous gulls may be due to natural variance, although an OHC-induced thyroid stimulating hormone (TSH) perturbation resulting in epithelial cell hyperplasia and increased follicular density cannot be ruled out and remains to be verified. Hence, a large-scale histological study is required, in order to elaborate the potential linkage between changes in thyroid gland histology, OHC exposure and regulation of the HPT-axis in the Arctic-breeding glaucous gull.  相似文献
