Enzymatic decolourization of the azo dye, Direct Yellow (DY106) by Cucurbita pepo (courgette) peroxidase (CP) is a complex process, which is greatly affected by pH, temperature, enzyme activity and the concentrations of H2O2 and dye. Courgette peroxidase was extracted and its performance was evaluated by using the free-CP (FCP) and immobilized-CP (ICP) forms in the decolourization of DY106. Immobilization of peroxidase in calcium alginate beads was performed according to a strategy aiming to minimize enzyme leakage and keep its activity at a maximum value by optimizing sodium alginate content, enzyme loading and calcium chloride concentration. The initial conditions at which the highest DY106 decolourization yield was obtained were found at pH 2, temperature 20℃, H2O2 dose 1 mmol/L (FCP) and 100 mmol/L (ICP). The highest decolourization rates were obtained for dye concentrations 50 mg/L (FCP) and 80 mg/L (ICP). Under optimal conditions, the FCP was able to decolorize more than 87% of the dye within 2 min. While with ICP, the decolourization yield was 75% within 15 min. The decolourization and removal of DY106 was proved by UV-Vis analysis. Fourier transform infrared (FT-IR) spectroscopy analysis was also performed on DY106 and enzymatic treatment precipitated byproduct.  相似文献

The Egyptian Red Sea environment especially along El-Sokhna area to the Suez refineries (Suez) is severely contaminated with organic compounds, as well as overfishing. This may be well contributory to recent serious declines in fish stocks. Fish embryos are also particularly vulnerable to oil exposure, even at extremely low concentrations of less than one part per billion. Consequently, even traces of oil pollution at levels often considered safe for wildlife can cause severe damage to fish. Sixteen polycyclic aromatic hydrocarbons (PAHs) were investigated in ten fish species of aquatic species by high performance liquid chromatography (HPLC). The compositions of PAHs determined in all samples were measured in order to use them as chemical markers for identifying different sources of PAH pollutants in the studied region. The total content of these16 PAHs ranged from 399.616 up to 67,631.779 ng/g wet weight. The data show that these values are considered to be alarmingly high enough to cause lethal toxicity effect by accumulation. All studied aquatic species samples are characterized by relatively high concentrations of the six-membered ring PAHs. The origin of PAHs in the collected samples is either petrogenic, biogenic, or mixed petrogenic and biogenic.  相似文献

With the use of cost-effective natural materials, biosorption is considered as an ecological tool that is applied worldwide for the remediation of pollution. In this study, we proposed Lemna gibba biomass (LGB), a lignocellulosic sorbent material, for the removal of two textile dyes, Direct Red 89 (DR-89) and Reactive Green 12 (RG-12). These azo dyes commonly used in dying operations of natural and synthetic fibres are the most important pollutants produced in textile industry effluents. For this purpose, batch biosorption experiments were carried out to assess the efficacy of LGB on dye treatment by evaluating the effect of contact time, biomass dosage, and initial dye concentration. The results indicated that the bioremoval efficiency of 5 mg?L?1 DR-89 and RG-12 reached approximately 100 % after 20 min of the exposure time; however, the maximum biosorption of 50 mg?L?1 DR-89 and 15 mg?L?1 RG-12 was determined to be about 60 and 47 %, respectively. Fourier transform infrared spectroscopy used to explain the sorption mechanism showed that the functional groups of carboxylic acid and hydroxyl played a major role in the retention of these pollutants on the biomass surface. The modelling results using Freundlich, Langmuir, Temkin, Elovich, and Dubini Radushkevich (D-R) isotherms demonstrated that the DR-89 biosorption process was better described with the Langmuir theory (R 2?=?0.992) while the RG-12 biosorption process fitted well by the D-R isotherm equation (R 2?=?0.988). The maximum biosorption capacity was found to be 20.0 and 115.5 mg?g?1 for DR-89 and RG-12, respectively, showing a higher ability of duckweed biomass for the bioremoval of the green dye. The thermodynamic study showed that the dye biosorption was a spontaneous and endothermic process. The efficacy of using duckweed biomass for the bioremoval of the two dyes was limited to concentrations ≤50 mg?L?1, indicating that L. gibba biomass may be suitable in the refining step of textile effluent treatment.  相似文献

Abstract

The susceptibility of wild mallard ducklings to the delayed neurotoxic effect of the neurotoxic organophosphorus insecticides cyanofenphos and leptophos was evaluated following a daily dosing regimen. Ducklings were treated daily with either cyanofenphos or with leptophos at different dose levels for 90 days, or until they died, or became paralyzed. A control group of ducklings given corn oil at 1 ml/kg daily for 90 days was used for comparison. All treated birds were observed daily for any clinical signs of neurotoxicity during the course of this study. All of the surviving ducklings that were treated with cyanofenphos at 4 mg/kg/day or leptophos at 10 mg/kg/day developed clinical signs of delayed neurotoxicity after 7 to 11 weeks of intoxication. Symptoms included leg weakness, ataxia, severe ataxia and paralysis. The observed clinical signs were confirmed by histological changes found in the spinal cords of the treated birds. These changes were of the type associated with organophosphorus‐induced delayed neuropathy (OPIDN). These results demonstrate that wild mallard ducklings are susceptible to OPIDN and this avian species can be used in screening organophos‐phorus compounds for such effect.  相似文献
