This study is part of our investigations about the release of persistent organic pollutants from melting Alpine glaciers and the relevance of the glaciers as secondary sources of legacy pollutants. Here, we studied the melt-related release of polychlorinated biphenyls (PCBs) in proglacial lakes and glacier streams of the catchment of the Silvretta glacier, located in the Swiss Alps. To explore a spatial and temporal distribution of chemicals in glacier melt, we combined two approaches: (1) analysing a sediment record as an archive of past remobilization and (2) passive water sampling to capture the current release of PCBs during melt period. In addition, we determined PCBs in a non-glacier-fed stream as a reference for the background pollutant level in the area. The PCBs in the sediment core from the Silvretta lake generally complied with trends of PCB emissions into the environment. Elevated concentrations during the most recent ten years, comparable in level with times of the highest atmospheric input, were attributed to accelerated melting of the glacier. This interpretation is supported by the detected PCB fractionation pattern towards heavier, less volatile congeners, and by increased activity concentrations of the radioactive tracer 137Cs in this part of the sediment core. In contrast, PCB concentrations were not elevated in the stream water, since no significant difference between pollutant concentrations in the glacier-fed and the non-glacier-fed streams was detected. In stream water, no current decrease of the PCBs with distance from the glacier was observed. Thus, according to our data, an influence of PCBs release due to accelerated glacier melt was only detected in the proglacial lake, but not in the other compartments of the Silvretta catchment.  相似文献

A pregnant woman with indeterminate Duchenne muscular dystrophy (DMD) carrier status, but with DMD diagnosed in her deceased brother (unavailable for study), presented for prenatal diagnosis, intending to continue the pregnancy only if proven unaffected with DMD with near absolute certainty. Creatine kinase (CK) assays to clarify carrier status were inconclusive. Male sex in the fetus was identified, but DNA restriction fragment length polymorphism (RFLP) analysis was not yet available to this centre to investigate the possible transmission of the DMD gene, and the pregnancy was terminated. Tissue histology and dystrophin protein analysis demonstrated the absence of DMD. In a situation with proven maternal carrier status, future fetal inheritance of the opposite maternal X chromosome would indicate the presence of DMD. However, maternal carrier status remained in doubt through a second pregnancy, even with RFLP studies, and was finally established when dystrophin analysis confirmed the presence of DMD in the second fetus. Histologic findings are presented, contrasting features in the two fetuses. The value of dystrophin analysis for establishing the diagnosis of fetal DMD, in this case proving maternal carrier status in a difficult situation, and for demonstrating DMD gene:RFLP haplotype relationships is illustrated.  相似文献

The effect of various pesticides on the biofilm formation by the phytopathogenic bacterium Clavibacter michiganensis ssp. sepedonicus (Cms), the potato ring rot causative agent, was explored for the first time. Systemic herbicides: 2,4-D, diuron, glyphosate, clopyralid, fluorodifen, as well as the commercial preparations “Lazurite,” “Ridomil Gold,” and the mitochondria inhibiting pesticides analog, sodium monoiodoacetate, were studied. These pesticides' effect on the Cms biofilm formation was shown to be distinct and dependent on the agent under question. Cms biofilm formation was reduced when exposed to sodium monoiodoacetate, as well as “Lazurite” preparation, that could be due to the bactericidal effect of these agents. 2,4-D and “Ridomil Gold” preparation stimulated the biofilm formation. Systemic herbicides diuron, glyphosate, clopyralid, fluorodifen did not exert appreciable influence on the process of bacterial biofilm formation.  相似文献

Screening by gas chromatography with flame ionization detection and gas chromatography-mass spectrometry has been carried out on sludge extracts of wastewater treatment basins. Soxhlet extraction with trichlorotrifluoroethane was applied. The yields for petroleum hydrocarbons and PAH recovery were high, usually in excess of 90%. Understanding the processes leading to population declines in fragmented landscapes is essential for successful conservation management. However, isolating the influence of disparate processes, and dispersal in particular, is challenging. The Grey Shrike-thrush, Colluricincla harmonica, is a sedentary woodland-dependent songbird, with learned vocalizations whose incidence in suitable habitat patches falls disproportionally with decline in tree cover in the landscape. Although it has been suggested that gaps in tree cover might act as barriers to its dispersal, the species remains in many remnants of native vegetation in agricultural landscapes, suggesting that it may have responded to habitat removal and fragmentation by maintaining or even increasing dispersal distances. We quantified population connectivity of the Grey Shrike-thrush in a system fragmented over more than 120 years using genetic (microsatellites) and acoustic (song types) data. First, we tested for population genetic and acoustic structure at regional and local scales in search of barriers to dispersal or gene flow and signals of local spatial structuring indicative of restricted dispersal or localized acoustic similarity. Then we tested for effects of habitat loss and fragmentation on genetic and acoustic connectivity by fitting alternative models of mobility (isolation-by-distance [the null model] and reduced and increased movement models) across treeless vs. treed areas. Birds within -5 km of each other had more similar genotypes and song types than those farther away, suggesting that dispersal and song matching are limited in the region. Despite restricted dispersal detected for females (but not males), populations appeared to be connected by gene flow and displayed some cultural (acoustic) connectivity across the region. Fragmentation did not appear to impact greatly the dispersal of the Grey Shrike-thrush: none of the mobility models fit the genetic distances of males, whereas for females, an isolation-by-distance model could not be rejected in favor of the models of reduced or increased movement through treeless gaps. However, dissimilarities of the song types were more consistent with the model of reduced cultural connectivity through treeless areas, suggesting that fragmentation impedes song type sharing in the Grey Shrike-thrush. Our paper demonstrates that habitat fragmentation hinders important population processes in an Australian woodland bird even though its dispersal is not detectably impacted.  相似文献

Parental feeding effort is an important determinant of chick development in birds. Quality and amount of food provided to each nestling, however, might affect the development of the immune system. Food availability around the nest site may account for part of the difference in body condition and immune capacity of nestlings. We tested this idea in the socially monogamous serin (Serinus serinus), investigating growth and immune capacity of nestlings in relation to food availability around the nest site. We used a set of immunological and serological assays, including antibody responses to sheep red blood cells (SRBCs), the T lymphocyte cell-mediated immune response (PHA), granulocyte:lymphocyte ratio (G:L), haematocrit and sedimentation rate. The results suggest that serins raised under poor environmental conditions were less immunocompetent. The immune response to PHA and SRBCs significantly increased with food availability. Haematocrit values were positively, sedimentation rate and the G:L ratio negatively correlated with food availability. Similar results were obtained when examining chick development in relation to the immunological and serological variables. However, there was a negative relationship between parental feeding rates and food availability, as well as between parental feeding rates and chick development. We conclude that body condition and immune capacity of nestlings are strongly correlated with the availability of food around the nest site.  相似文献

An algorithm for stepwise approximation to the most reliable source of contamination is proposed. The concept of the algorithm is based on discrimination rather than on similarity. Gradually, by means of more complicated and higher distinguishing power approaches, the differences between the spilled sample and the suspected source are checked. Capillary gas chromatography with flame-ionization and mass spectrometric detection was applied to gather the necessary data. Fingerprint, diagnostic parameter approaches and pattern recognition methods were used for reliably distinguishing the analytical results. If the compared samples are not discriminated by any means, they are considered as identical.  相似文献

The drifting sea ice has been suggested as important in the transport and concentration of organic matter and pollutants in the Arctic. We collected sea ice-associated amphipods in the marginal ice zone north of Svalbard and in the Fram Strait in September 1998 and 1999 to assess contaminant accumulation in ice-associated organisms. Organochlorine concentrations increased from the more herbivorous Apherusa glacialis to the more carnivorous Gammarus wilkitzkii and the more necrophagous Onisimus spp. The relative contribution of compound classes to the sum of organochlorines differed between the amphipod families, with a higher relative contribution of hexachlorocyclohexanes (HCHs) in A. glacialis. The composition of the compound classes HCHs. chlordanes and dichlorodiphenyltrichloroethanes (DDTs) was similar between the amphipod families, whereas the profiles of polychlorinated biphenyls (PCBs) differed. The occurrence of organochlorines differed spatially, with higher alpha-HCH concentrations in amphipods from the Fram Strait in comparison with amphipods collected north of Svalbard. This could be related to the sea ice drift route, since sea ice in the Fram Strait had a drift route across the central Arctic Ocean, while the sea ice north of Svalbard had a western drift route to the sampling stations. Even though marine invertebrates have direct uptake by passive diffusion of contaminants across their gills. our results imply that the species' ecology such as diet is important in the bioaccumulation process of organic pollutants. In addition, the results show that sea ice drift route influences the concentrations of organochlorine pollutants in ice-associated organisms.  相似文献
