Retention times of treated water in a constructed wetland (CW) with horizontal subsurface flow were determined both in the vegetative and non-vegetative periods of 2005. Tracer experiments were performed using fluorescein, an organic compound detectable at extremely low concentrations. Nominal and tracer retention times were determined and compared. Winter tracer retention time (TRT 194 h) and nominal retention time (nHRT 190 h) were nearly exactly equal, while summer TRT (335 h) was approximately twofold higher than nHRT (158 h). Residence time distribution function (RTD) was used to compare retention times obtained for the vegetative and non-vegetative periods. The obtained results document a significant influence of dense common reed vegetation on retention characteristics of the studied system. Common reed can convert a significant volume of water from liquid to gas via evapotranspiration (ET) and thus prolong water retention times in the system. This is very important both technically and ecologically. Longer retention times mean a longer time for microbiological decay of wastewater. Water converted from liquid to gas causes cooling of the microclimate, which is very important, especially in intensively cultivated areas with a lack of water.  相似文献

A simple, sensitive, convenient and low cost spectrofluorometric technique for determination of quinalphos is described. Quinalphos is decomposed with sodium ethoxide. Resultant sodium diethyl thiophosphate (Na-DETP) is extracted with a cationic fluorescein as ion pair complex into 1-chloro-2-nitrobenzene. The fluorescence of the ion pair, fluorescein-DETP is measured at 530 nm after excitation at 490 nm. This technique is applicable for the determination of quinalphos in water, food grains and vegetable samples up to ng level with Amberlite XAD-4.  相似文献

The most important factor affecting efficacy and drift of pesticide applications is the droplet spectrum. To measure pesticide drift, researchers utilize fluorescent tracers to rapidly quantify spray deposition. Although fluorescent tracers have been used for more than 50 years, no experiments have been performed on the effect they have on the properties of pesticide formulations (density and viscosity) or droplet spectrum, which affect the drift of pesticides. Therefore, we examined the effect of an oil- and water-based tracer on the volume median diameter (VMD), viscosity, and density of oil- and water-based pesticide formulations. In addition, we experimentally fit and demonstrate the utility of using distributions to characterize pesticide droplet spectra. The addition of tracers to both water- and oil-based formulations did not significantly alter the VMD, viscosity, and density. Lognormal distributions provided the best fit for the water- and oil-based formulations with and without tracer. Our results demonstrated that the addition of oil- and water-based tracers do not significantly alter pesticide formulations properties and droplet spectrum, and most likely do not alter the movement of pesticide droplets in the environment.  相似文献

以荧光素作为荧光基团,基于硫化氢(H2S)的还原性,设计合成了一种off-on型H2S荧光探针.该探针本身在乙腈溶液中荧光十分微弱,与H2S反应之后,在520 nm处产生强荧光,同时溶液颜色由无色变成亮黄色,可实现对H2S的可视化检测.随着H2S浓度的逐渐增高,探针的荧光强度和紫外吸收强度也逐渐增强,在0—50μmol·L-1的范围内,二者和H2S浓度之间都具有很好的线性关系,相关系数R2分别为0.9934、0.9921.探针对H2S具有很好的选择性,常见阴离子和还原性硫醇分子共存时,对检测不会产生干扰.该探针对H2S的检测限为10μmol·L-1,具有较高的检测灵敏度.  相似文献

H3PO4活化花生壳炭(750℃)为吸附剂,荧光素钠为吸附质.考察了炭化温度、投加量、pH、离子强度、吸附时间以及初始浓度对花生壳活性炭吸附荧光素钠的影响.重点研究了花生壳活性炭对荧光素钠的等温吸附特征,并利用准一级、准二级动力学模型和颗粒内扩散方程对花生壳活性炭吸附荧光素钠的动力学过程进行了分析.结果表明,本实验条件下,Langmuir等温吸附模型能很好地描述花生壳活性炭对荧光素钠的吸附效果,吸附强度因子a为正值,表明吸附过程在本实验条件下可自发进行.通过带入实验数据校对得出准二级动力学方程能更好地描述花生壳活性炭对水中荧光素钠的吸附动力学过程.  相似文献

FDA-PI双色荧光法检测蓝藻细胞活性的研究   总被引:11,自引:0,他引:11  
对FDA-PI双色荧光法检测水华鱼腥藻和铜绿微囊藻的活性进行了研究,用荧光显微镜对染色结果进行测定.结果表明,在蓝色光激发下(495nm),活细胞被双醋酸荧光素FDA染成亮绿色,死亡细胞被碘化丙锭PI染成红色.染色效率与原植体类型和细胞密度有关.对细胞密度为6×107—7×109个·l-1的铜绿微囊藻,FDA染色效率可达94%以上.对细胞密度为4×107—5×108个·l-1的水华鱼腥藻,FDA染色效率可达91%以上,但细胞密度增大到5×109个·l-1时,由于藻丝体易卷曲在一起,FDA染色率下降到67%.对死亡细胞,PI染色率基本都可达到100%.因此,用FDA-PI检测活细胞和死亡细胞混合的细胞悬液,可根据细胞所发出的不同荧光而判断细胞活性.  相似文献

The main aim of this study was to assess the impact of pesticidal residues on soil microbial and biochemical parameters of the tea garden soils. The microbial biomass carbon (MBC), basal (BSR) and substrate induced respirations (SIR), β -glucosidase activity and fluorescein diacetate hydrolyzing activity (FDHA) of six tea garden soils, along with two adjacent forest soils (control) in West Bengal, India were measured. The biomass and its activities and biochemical parameters were generally lower in the tea garden soils than the control soils. The MBC of the soils ranged from 295.5 to 767.5 μ g g? 1. The BSR and SIR ranged from 1.65 to 3.08 μ g CO2-C g? 1 soil h? 1 and 3.08 to 10.76 μ g CO2-C g? 1h? 1 respectively. The β -glucosidase and FDHA of the soils varied from 33.3 and 76.3 μ g para-nitrophenol g? 1 soil h? 1 and 60.5 to 173.5 μ g fluorescein g? 1h? 1respectively. The tea garden soils contained variable residues of organophosphorus and organochlorine pesticides, which negatively affected the MBC, BSR, SIR, FDHA and β -glucosidase activity. Ethion and chlorpyriphos pesticide residues in all the tea garden soils varied from 5.00 to 527.8 ppb and 17.6 to 478.1 ppb respectively. The α endosulfan, β endosulfan and endosulfan sulfate pesticide residues in the tea garden soils ranged from 7.40 to 81.40 ppb, 8.50 to 256.1 ppb and 55 to 95.9 ppb respectively. Canonical correlation analysis shows that 93% of the total variation was associated with the negative impact of chlorpyriphos, β and α endosulfan and endosulfan sulfate on MBC, BSR and FDHA. At the same time ethion had negative impact on SIR and β -glucosidase. Data demonstrated that the pesticide residues had a strong impact on the microbial and biochemical components of soil quality.  相似文献

在pH 3.5~4.4的醋酸盐缓冲溶液中, Ag+与Cl-离子反应形成AgCl。当Ag+离子适当过量时,AgCl能与Ag+结合形成[AgCl·Ag]+阳离子,它能借静电引力和疏水作用力与荧光素一价阴离子(HL-)反应形成离子缔合物[(AgCl·Ag)HL],该疏水性的离子缔合物能在水相挤压作用和范德华力的作用下彼此靠近而进一步聚集,形成平均粒径约为20 nm的纳米微粒[(AgCl·Ag)HL]n。此时仅能引起吸收光谱和荧光光谱的微小变化,但能导致倍频散射(FDS)和二级散射(SOS)等共振非线性散射(RNLS)的显著增强,其最大FDS和SOS波长分别位于350和560 nm处。两种散射增强(ΔIFDS和ΔISOS)在一定范围内均与氯离子浓度成正比,均可用于氯离子的测定。其中以FDS最灵敏,对于氯离子的检测,其线性范围是0.04~1.22 μg/mL, 检出限为10.9 ng/mL;在环境空气或无组织排放废气HCl的检测中,当采气体积为60L时,其线性范围是0.007~0.21 mg/m3,检出限为1.9×10-3 mg/m3;在有组织排放废气样品中,当采气体积为10 L时,其线性范围是0.04~1.25 mg/m3,检出限为1.1×10-2 mg/m3。该文研究了纳米微粒对吸收、RNLS光谱的影响、反应的适宜条件及影响因素,考察了共存物质的影响,表明方法有良好的选择性,据此利用上述反应发展了一种用SOS和FDS技术高灵敏度、高选择性和简便、快速测定环境空气和废气中HCl及环境水样中氯化物的新方法。文中还对反应机理进行了讨论。  相似文献
