摘    要:乌毛蕨的叶片经PBS抽提和硫酸铵沉淀,得到的粗蛋白经活性碳柱处理后,在分子筛层析柱Sephadex G-100上纯化得到一种凝集素(Blechnum Orientale Lectin,简称BOL),BOL的比活性比粗提液提高了32倍,活力回收率达75%,在PAGE上显示一条蛋白染色带,而在SDS-PAGE上显示BOL由两个不同的亚基组成,亚基分子量分别为16600和22910.BOL是一种糖蛋白,其中性糖含量为20.06%.其分子中含有Mg和Ca两种金属元素,含量分别为0.08%和0.70%.BOL对温度变化敏感,其凝血活性能被N-乙酰葡萄糖胺所抑制,用滤纸圆片法检测,BOL在测试的8种菌中对大肠杆菌和玉米大斑病菌的生长有抑制作用,图4表5参13

PARTIAL PROPERTIES OF BLECHNUM ORENTALE LECTIN
Abstract:The crude protein was obtained by extracting the leaves of Blechnum orientale with PBS and precipitating with (NH 4) 2SO 4. The crude protein was purified by Sephadex G-100 filtration after treatment through active carbon column. The agglutinating activity of the purified lectin (BOL) increased 32 times and the activity recovery is 75 %. One single protein band of lectin appeared on PAGE, and SDS-PAGE showed it consisted of two subunits of different molecular weight (16 600 and 22 910). BOL was a glycoprotein with 20.06 % neutral saccharides, containing 0.08 % Mg 2 and 0.70 % Ca 2 . BOL was sensitive to temperature change and its agglutinating activity inhibited by GlcNAc. It could apparently inhibit the growth of Escherichia coli and Helminthosporium turcicum Pass with the eight tested bacteria and plant pathogeny fungi by testing filter paper wafer.
