The nickel and zinc tolerant plant growth promoting Bradyrhizobium sp. (vigna) RM8 was isolated from nodules of greengram, grown in metal contaminated Indian soils. The plant growth promoting (PGP) potentials of strain RM8 was assessed both in the presence and absence of nickel and zinc under in vitro conditions. Strain RM8 tolerated a high level of nickel (300 microg ml(-1)) and zinc (1400 microg ml(-1)) on yeast extract mannitol agar medium. Bradyrhizobium sp. (vigna) strain RM8 produced 13.3 microg ml(-1) of indole acetic acid in Luria Bertani broth at 100 microg ml(-1) of tryptophan which increased to 13.6 microg ml(-1) at 50 microg Ni ml(-1) and 13.5 microg ml(-1) at 300 microg Zn ml(-1). Strain RM8 was positive for siderophore, HCN and ammonia both in the absence and presence of nickel and zinc. The PGP activity of this strain was further evaluated with increasing concentrations of nickel and zinc using greengram as a test crop. The bioinoculant enhanced the nodule numbers by 82%, leghaemoglobin by 120%, seed yield by 34%, grain protein by 13%, root N by 41% and shoot N by 37% at 290 mg Ni kg(-1) soil. At 4890 mg Zn kg(-1) soil, the bioinoculant increased the nodule numbers by 50%, leghaemoglobin by 100%, seed yield by 36%, grain protein by 13%, root N by 47% and shoot N by 42%. The bioinoculant strain RM8 reduced the uptake of nickel and zinc by plant organs compared to plants grown in the absence of bioinoculant. This study suggested that the bioinoculant due to its intrinsic abilities of growth promotion and attenuation of the toxic effects of nickel and zinc could be exploited for remediation of metal from nickel and zinc contaminated sites.  相似文献

Trichoderma atroviride F6, isolated from decaying feather and resistant to 100 mg l(-1) Cd2+ and 250 mg l(-1) Ni2+, was applied for rhizoremediation of Cd, Ni and Cd-Ni combination contaminated soils through association with Brassica juncea (L.) Coss. var. foliosa. The strain significantly alleviated the cellular toxicity of cadmium and nickel to plants. Inoculation of B. juncea (L.) Coss. var. foliosa with T. atroviride F6 resulted a 110%, 40% and 170% increase in fresh weight in Cd, Ni and Cd-Ni contaminated soils, respectively (P<0.05). The translocation factors and metal bioconcentration factors calculated for the inoculated plant were increased compared to the noninoculated plants. The results indicated that the efficiency of phytoextraction for B. juncea (L.) Coss. var. foliosa enhanced after inoculating with T. atroviride F6. The fungal treated plants grown in Cd-Ni combination contaminated soils showed higher phytoextraction efficiency than those in Cd or Ni contaminated soils. Thus, it is suggested that the fungus T. atroviride F6 endowed with organic-degrading capabilities could be exploited for fungi-assisted phytoremediation of mixed organic-metal contaminated soils.  相似文献

Forty-six bacterial cultures, including one culture collection strain, thirty from the rhizosphere of Alyssum murale and fifteen from Ni-rich soil, were tested for their ability to tolerate arsenate, cadmium, chromium, zinc, mercury, lead, cobalt, copper, and nickel in their growth medium. The resistance patterns, expressed as minimum inhibitory concentrations, for all cultures to the nine different metal ions were surveyed by using the agar dilution method. A large number of the cultures were resistant to Ni (100%), Pb (100%), Zn (100%), Cu (98%), and Co (93%). However, 82, 71, 58 and 47% were sensitive to As, Hg, Cd and Cr(VI), respectively. All cultures had multiple metal-resistant, with heptametal resistance as the major pattern (28.8%). Five of the cultures (about of 11.2% of the total), specifically Arthrobacter rhombi AY509239, Clavibacter xyli AY509235, Microbacterium arabinogalactanolyticum AY509226, Rhizobium mongolense AY509209 and Variovorax paradoxus AY512828 were tolerant to nine different metals. The polymerase chain reaction in combination with DNA sequence analysis was used to investigate the genetic mechanism responsible for the metal resistance in some of these gram-positive and gram-negative bacteria that were, highly resistant to Hg, Zn, Cr and Ni. The czc, chr, ncc and mer genes that are responsible for resistance to Zn, Cr, Ni and Hg, respectively, were shown to be present in these bacteria by using PCR. In the case of, M. arabinogalactanolyticum AY509226 these genes were shown to have high homology to the czcD, chrB, nccA, and mer genes of Ralstonia metallidurans CH34. Therefore, Hg, Zn, Cr and Ni resistance genes are widely distributed in both gram-positive and gram-negative isolates obtained from A. murale rhizosphere and Ni-rich soils.  相似文献

Bacteria resistant to Cu2+, Ni2+ or Zn2+ were isolated from the rhizosphere of water hyacinth (Eichhornia crassipes (Mart.)) and their metal ion removal capacities (RCs) were determined. The Ni2+ and Zn2+ RCs of the respective metal ion-resistant bacteria were less than 4.1 mg g(-1), while one of the Cu2+-resistant bacteria (Strain CU-1) showed a significant high Cu2+ RC of 10.6 mg g(-1). The effect of inoculating water hyacinth with Strain CU-1 on its Cu2+ RC was further studied. Water hyacinths were treated with an antibiotic, oxytetracycline (OTC), to remove most rhizospheric bacteria of plant roots. Inoculation of Strain CU-1 increased the Cu2+ RC of the plant root by 1.91 (OTC-treated) and 1.56 (OTC-untreated) folds respectively when compared with the control. Results also showed that Strain CU-1 colonized onto the plant root and led to the increase of Cu2+ RC of the roots of water hyacinth.  相似文献

The potential of purple non-sulphur bacteria for bioremediation was assessed by investigating the ability of Rhodobacter sphaeroides strain R26.1 to grow photosynthetically in heavy metal contaminated environments. Bacterial cultures were carried out in artificially polluted media, enriched with the transition metal ions Hg2+, Cu2+, Fe2+, Ni2+, Co2+, MoO4(2-), and CrO4(2-) in millimolar concentration range. For each investigated ion the effect on growth parameters was evaluated. The analysis of concentration-effect curves revealed a differentiated response, indicating that diverse mechanisms of tolerance and/or resistance are involved. Adaptation or selection procedures were not applied, leading to assess intrinsic abilities of coping with these contaminants. The microorganism proved to be highly tolerant to heavy metal exposure, especially towards Co2+, Fe2+ and MoO4(2-). In addition Ni2+ and Co2+ were found to decrease the cellular content of light harvesting complexes. A characteristic behavior was observed with mercuric ions, which produced a significant increase of the lag-phase.  相似文献

This study investigates the ability of the biodynamic model to predict the trophic bioaccumulation of cadmium (Cd), chromium (Cr), copper (Cu), nickel (Ni) and zinc (Zn) in a freshwater bivalve. Zebra mussels were transplanted to three sites along the Seine River (France) and collected monthly for 11 months. Measurements of the metal body burdens in mussels were compared with the predictions from the biodynamic model. The exchangeable fraction of metal particles did not account for the bioavailability of particulate metals, since it did not capture the differences between sites. The assimilation efficiency (AE) parameter is necessary to take into account biotic factors influencing particulate metal bioavailability. The biodynamic model, applied with AEs from the literature, overestimated the measured concentrations in zebra mussels, the extent of overestimation being site-specific. Therefore, an original methodology was proposed for in situ AE measurements for each site and metal.  相似文献

Abandoned industrial sites and mines may constitute possible hazards for surrounding environment due to the presence of toxic compounds that may contaminate soils and waters. The possibility to remove metal contaminants, specifically nickel (Ni), by means of fungi was presented exploiting a set of fungal strains isolated from a Ligurian dismissed mine. The achieved results demonstrate the high Ni(II) tolerance, up to 500 mg Ni l?1, and removal capability of a Trichoderma harzianum strain. This latter hyperaccumulates up to 11,000 mg Ni kg?1, suggesting its possible use in a bioremediation protocol able to provide a sustainable reclamation of broad contaminated areas.  相似文献

The variability of species sensitivity distribution (SSD) due to contaminant bioavailability in soil was explored by using nickel as metal of concern. SSDs of toxicity test results of Avena sativa L. originating from different soils and expressed as total content and available (0.01 M CaCl2) extractable concentration were compared to SSDs for terrestrial plants derived from literature toxicity data. Also the 'free' nickel (Ni2+) concentration was calculated and compared. The results demonstrated that SSDs based on total nickel content highly depend on the experimental conditions set up for toxicity testing (i.e. selected soil and pH value) and thus on metal bioavailability in soil, resulting in an unacceptable uncertainty for ecological risk estimation. The use in SSDs of plant toxicity data expressed as 0.01 M CaCl2 extractable metal strongly reduced the uncertainty in the SSD curve and thus can improve the ERA procedure remarkably by taking bioavailability into account.  相似文献

Animal bone is able to adsorb copper and nickel ions from their single aqueous solutions. It was noted that a decrease in the sorbent concentration with constant copper or nickel concentration, or an increase in the copper or nickel concentration with a constant sorbent concentration resulted in a higher metal loading per unit weight of the sorbent. Increase in the initial pH of the metal solution resulted in an increase in the metals uptake per unit weight of the sorbent. Freundlich isotherm model was found to be applicable for the experimental data of Cu2+ and Ni2+. The results showed that animals bones can be used for the adsorption of the Cu2+ and Ni2+ with higher affinity toward Cu2+ ions. The new sorbent was able to decrease copper concentration to a limit lower than the limit permitted by the environmental regulations.  相似文献

Inoculation of plants with microorganisms may reduce the toxicity of heavy metals to plants in contaminated soils. In this study, we have shown that the plant growth promoting bacteria Methylobacterium oryzae strain CBMB20 and Burkholderia sp. strain CBMB40 from rice reduce the toxicity of Ni and Cd in tomato and promote plant growth under gnotobiotic and pot culture experiments. The bacterial strains bound considerable amounts of Ni(II) and Cd(II) in their growing and resting cells and showed growth in the presence of NiCl2 and CdCl2. In gnotobiotic assay, inoculation with the bacterial strains reduced the ethylene emission and increased the tolerance index of the seedlings against different concentrations of NiCl2/CdCl2. In pot experiments carried out with non-polluted, Ni and Cd supplemented Wonjo-Mix bed soil, the results clearly demonstrated reduction in the accumulations of Ni(II) and Cd(II) in roots and shoots, with significant increase in the plant growth attributes with bacterial inoculations compared to untreated control. Strain CBMB20 performed better than CBMB40 in reducing the heavy metal accumulations in plants. Our results suggest conclusively, that protection against the heavy metals toxicity is rendered by these bacterial strains by reducing their uptake and further translocation to shoots in plants and promote the plant growth by other PGP characteristics.  相似文献

Adsorption properties of Co2+, Ni2+, and Cr3+ on an Amberlite IRN-77 cation exchange resin were investigated in batch systems. Levels of adsorption rapidly approached an equilibrium state within 1 h. The adsorption characteristics of each metal onto the resin were accurately represented by Langmuir isotherms. Co2+ and Ni2+, which have an equivalent electrovalence, displayed similar levels of adsorption onto the resin when they coexisted in the solution. However, when Cr3+ was added to the solution it competitively replaced Co2+ and Ni2+ ions that had been previously adsorbed onto the resin, resulting in the desorption of these metals into the solution. The result was likely due to a higher adsorption affinity of Cr3+ relative to Co2+ and Ni2+. This implies that interactively competitive adsorption of multi-cations onto the resin should be thoroughly considered when contemplating the efficient operation of an ion exchange process in the treatment of industrial wastewater.  相似文献

Soil samples with a range of chemical and physical properties were collected from 10 different rural regions of China. Trace metals (Ni, Co, Cu, and Pb) in the soils were partitioned by a sequential extraction procedure into Mg(NO(3))(2) extractable (F1), CH(3)COONa extractable (F2), NH(2)OH.HCl extractable (F3), HNO(3)?H(2)O(2) extractable (F4), and residual (F5) fractions. Chemical fractionation showed that F1 fraction of the metals was less than 1% and residue was the dominant form for Cu and Ni in all samples, and for Co in most of the samples. Significant interrelationships of the fractions varied considerably with the different metals. Winter wheat (Triticum aestivum L.) and alfalfa (Medicago sativa L.) had been grown on the soils in a pot-culture experiment under greenhouse conditions for 40 days. Metal availability to the plants was evaluated by simple and multiple regression analysis. The Mg(NO(3))(2) extractable Co (F1) was significantly correlated with Co concentrations in different parts of wheat and in the whole of alfalfa. For the other metals, the independent variables of the multiple regression models, chosen by stepwise selection, were given as: F1 and F2 + F3 + F4 for Ni; F1, F2 + F3 and F4 for Cu; and F3 + F4 for Pb. The results of this study demonstrate that the sequential extraction procedure, in conjunction with multiple regression models using a combination of correlated fractions as an independent variable, may be useful for the prediction of plant absorption of trace metals in soils.  相似文献

The concentrations of Cd, Co, Cu, Ni, Pb, Zn, Fe and Mn in different inorganic fertilizers (urea, calcium superphosphate, iron sulphate and copper sulphate) and in pesticides (two herbicides and one fungicide) are evaluated together with the contribution of these metals in soils from their use. The study was made in rice farming areas to the north of Albufera Natural Park (Valencia, Spain). The results obtained show that superphosphate is the fertilizer that contains the highest concentrations of Cd, Co, Cu and Zn as impurities. Copper sulphate and iron sulphate have the most significant concentrations of Pb, and are the only fertilizers in which Ni was detected. The three pesticides analysed show similar Cd contents and the highest levels of Fe, Mn, Zn, Pb and Ni are found in the herbicides. The most significant additions of heavy metals as impurities that soil receives from agricultural practices, are Mn, Zn, Co and Pb. Three contamination indexes have been applied to provide a basis for comparison of potential heavy metal toxicity. These results denote the potential toxicity of heavy metals in the studied soils.  相似文献

Effect of soil on microbial responses to metal contamination   总被引:9,自引:0,他引:9  
An experiment was conducted to investigate microbial responses to metal inputs in five soils with varying clay and organic contents; one soil had also a higher pH. These soils were treated with a low metal, sewage sludge control or with this sludge contaminated to achieve Cu=112, Ni=58 and Zn=220 mg kg(-1) in medium and Cu=182, Ni=98 and Zn=325 mg kg(-1) in high metal soils. CO(2) evolution rates were measured at 1 week and at 4-5-day intervals thereafter until the end of the incubation (7 weeks). Extractable metals (CaCl(2) and water), biomass C, metabolic quotient, ergosterol, bacterial-fungal phospholipid fatty acid (PLFA-3 weeks only) ratio and mineral N were measured at 3 and 7 weeks. Metal inputs caused a marked increase in metal availability in the slightly acidic sandy loams, a smaller increase in slightly acidic clays and had little effect in the alkaline loam. After an initial increase in CO(2) evolution with metal inputs in all soils, the high metal treatment alone caused a significant decrease at later stages, mainly in sandy loams. Although biomass C and metabolic quotient decreased in all soils with higher metal inputs, the effect was more pronounced in the sandy loams. Metal inputs increased ergosterol and decreased bacterial-fungal PLFA ratios in most soils. Larger mineral N contents were found in all high metal soils at 3 weeks but, after 7 weeks, metals caused a significant decrease in sandy loams. CaCl(2) and water-extractable Cu, Ni and Zn contents were closely correlated with microbial indices in sandy loam but not in clay soils. Overall, the effect of treatments on microbial and extractable metal indices was greater in loams. Within a single series, higher organic soils showed less pronounced responses to metal inputs, although this trend was not always consistent.  相似文献

Biomineralization dependent on bacterial activity has been described for struvite which is formed in soils, guano, putrescent matter and sediments. A new biomineral containing nickel instead of magnesium, Ni(NH4)(PO4) . 6H2O, has been identified. It was formed by nickel resistant Streptomyces acidiscabies E13, and putatively named nickel struvite. The mineral formation is dependent on biological activity since non-viable bacterial cells are not capable to induce formation of Ni-struvite under identical conditions. Formation of Ni-struvite was observed on colony surfaces upon prolonged incubation of solid minimal or complex media containing elevated concentrations of 8-15mM NiCl2. The formation of magnesium containing crystals was not observed although Mg2+ is present in the medium. However, the process was not depending on desiccation since small crystals attached to the mycelial biomass of the bacteria were observed microscopically also in liquid cultures of nickel supplemented minimal and complex media after two weeks of incubation. The capacity to induce biomineralization of a nickel containing mineral is postulated to constitute a resistance factor, allowing the soil bacterium to withstand high nickel concentrations. The strain shows nickel resistance as an adaption to its habitat, since this bacterium was isolated from a former uranium mining site in Eastern Thuringia, Germany, where nickel concentrations of up to 2000ppm (translating to appr. 30mM) occur as a result of former mining activities.  相似文献

Some plants have high ability to absorb heavy metals in high concentrations. In this study, Halimione portulacoides was tested in conjunction with citric acid, in order to evaluate the possible use of this plant in phytoremediation processes in salt marshes. Two different concentrations of chelator were used combined with two heavy metal concentrations. When 25microM of citric acid was applied, Cd uptake and translocation was enhanced while for Ni these processes were almost inhibited. Increasing citric acid concentration to 50microM, Ni absorption decreased by the roots while for Cd there was still an increase in root uptake. Analysing translocation with this concentration of chelator, a decreased metal content in the upper organs for both metals was observed. While for Cd an optimal concentration of 25microM of citric acid was observed for phytoremediative processes, for nickel neither concentrations of chelator showed advantages for application in this remediative method.  相似文献

The use of two EDTA concentrations for enhancing the bioavailability of cadmium, chromium, and nickel in three natural soils (Ohio, New Mexico and Colombia) was investigated. The resulting uptake, translocation and selectivity with Helianthus annuus after mobilization were also examined. In general, plants grown in the sandy-loam Ohio soil had a higher uptake that resulted in a selectivity and total metal content of Cd>Cr>Ni and 0.73 mg and Cr>Cd>Ni and 0.32 mg for 0.1 and 0.3 g kg-1 EDTA, respectively. With the silty-loam New Mexico soil, although the total metal uptake was not statistically different the EDTA level did alter the selectivity; Cd>Cr>Ni (0.1 g kg-1 EDTA) and Cd>Cr>Ni (0.3 g kg-1 EDTA). Conversely, with the Colombian (sandy clay loam) soil increasing the EDTA level resulted in a higher total metal uptake (0.62 mg) than the 0.1 g kg-1 (0.59 mg) treatment. For all three soils, the translocation of Cd was limited. Evaluating the mobile metal fraction with and without EDTA determined that the chelator was capable of overcoming mass transfer limitations associated with the expandable clay fraction in the soils. Root wash results and root biomass concentrations indicated that Cd sorption was occurring. Therefore limited Cd translocation was attributed to insufficient phytochelatin levels.  相似文献

Radionuclide sorption by natural and modified clays is extensively accepted to be an important process from the radioactive waste point of view. This work focused on modification of natural attapulgite with a layered double hydroxide to produce a novel chemisorbent for Sr2+, Ni2+, and Co2+ removal from multicomponent solution. The structural and surface characteristics of both attapulgite (ATP) and modified attapulgite (LDH-ATP) were investigated using XRD, FTIR, SEM, and thermal analysis. Comparison of sorption features of Sr2+, Ni2+, and Co2+ onto ATP and LDH-ATP was achieved; the results indicated that LDH-ATP was the most efficient sorbent for Sr2+, Ni2+, and Co2+. Kinetic studies established that the sorption is fast and reaching >90% within 30 min. The sorption of Sr2+, Ni2+, and Co2+ are well defined by non-linear pseudo-second-order model and controlled by an intra-particle diffusion mechanism. The diffusivity was determined using homogeneous surface diffusion (HSDM) model and found in the order 10−13 m2/min; this confirmed that the sorption of the three ions is chemisorption process. LDH-ATP can be employed as a candidate chemisorbent for the removal of some metal ions from waste solution.  相似文献

Gallipoli is a small town on the European side of the Dardanelles Strait, in Turkey. More than 450,000 soldiers gave their lives on the Gallipoli Peninsula (Gallipoli Campaign) over a nine-month period during World War I. In that war, many naval and landing bombs were exploded. The ingredients of these bombs may cause heavy metal contamination in soils. An environmental geochemical investigation has been carried out around Gallipoli Peninsula in order to determine the extent of chemical pollution in the soil. The preliminary data reveal that soils in the area are, in general, not contaminated. The concentrations of many heavy metals, such as arsenic (As), cadmium (Cd), copper (Cu), chromium (Cr), cobalt (Co), manganese (Mn), nickel (Ni), lead (Pb), strontium (Sr), vanadium (V) and zinc (Zn) are in the range of those of average agricultural soils. These low concentrations are explained by the good leaching characteristics of Oligocene–Miocene rocks, Pleistocene terrace material, alluvium sandy-silty soils, and surface run-off.  相似文献

In the urban-rural transitional area of Hangzhou, China, 74 topsoil samples were collected from vegetable fields to measure the contents of arsenic (As), copper (Cu), cobalt (Co), cadmium (Cd), chromium (Cr), mercury (Hg), manganese (Mn), nickel (Ni), lead (Pb) and zinc (Zn). The combination of multivariate statistical and geostatistical methods successfully separated the contaminating elements (As, Cd, Cu, Hg, Pb and Zn) from uncontaminated elements (Co, Cr, Ni and Mn). A significant correlation was found between these uncontaminated elements and total Al2O3, Fe2O3, and SiO2 of the soils, indicating that the source of these elements was mainly controlled by soil-forming factors. On the other hand, these contaminating elements showed relatively weaker correlation and higher spatial variability, indicating that their enrichment and spatial heterogeneity were mostly affected by anthropic inputs. Through the pollution evaluation, it was found that only 30.8% of the study area did not suffer from moderate or severe pollution.  相似文献
