Studies conducted in animal systems have shown that lead is an immunosuppressive agent at levels far below those causing overt toxicity. Children less than six years of age are the population at highest risk for exposure to environmental lead; however little data were available to assess effects on the developing immune system in this age group. Reported here is the completed Phase I study on 193 children, ages 9 months to 6 years, with blood lead levels from 1 to 50 (g dL–1, recruited from the urban population of Springfield-Greene County, Missouri, through their participation in the WIC (Women, Infants, and Children) and Lead Poisoning Prevention Programs. This portion of the study dealt with enumeration of cells involved with the immune response andin vitro mitogenic stimulation of lymphocytes. The percent lymphocytes, monocytes, granulocytes, T cells (total), B cells, CD4+T's and CD8+T's and CD4+/CD8+ ratios were determined and the data were analysed. No consistent significant differences were seen among the various risk categories currently identified by the CDC. Though two age groups showed some possible effect of lead, none of the various cellular parameters within these age groups showed significant correlation with blood lead. The lymphocyte response toin vitro mitogenic stimulus was studied on 42 children (including 17 in risk classifications IIA and higher) using the mitogens phytohemagglutinin (PHA), Concanavalin A (Con A), and Pokeweed mitogen (PWM). No consistent statistically significant differences were seen among the various risk categories; the effects of lead, if present, are most likely subtle and obscured by the interindividual and time-dependent variation inherent in this type of study.  相似文献

An earlier nationwide reconnaissance study had indicated that lead levels in housedusts in Brighton and Hove were high compared with the general mean for the United Kingdom (1110 g/g versus 507 ug/g). A more rigorous study, with sample selection based on house age, has revealed a lower mean value of 705 g/g. Lead levels in 35 houses from the original study which were revisited had not changed significantly over the intervening 5 year period. The results suggest that the high mean value obtained previously was a reflection of the uneven age distribution in the sample population. The mean lead concentrations in soil and road dust samples, also obtained, were 404 and 794 g/g, respectively. Lead levels in ail three sample types (housedust, soil and road dust) were significantly correlated with each other and all showed a tendency to increase with increasing age of property.  相似文献

Pilot study of sources of lead exposure in Moscow,Russia   总被引:11,自引:0,他引:11  
This preliminary investigation of sources of lead exposure in Moscow, Russia, by Russian and US collaborators measured lead in paint, interior dust, and drinking water in seven day-care centres, and in petrol, soil and canned food. Some paint samples exceeded US regulatory standards for lead in paint on surfaces (0.5%). Dust lead loadings were < 1.7 g cm–2 and below the guidance levels of the US EPA. Drinking water lead concentrations were at or below the US drinking water standard of 15 g L–1. Lead concentrations in petrol from Moscow vehicles and petrol stations were consistent with a regulation banning the sale of leaded petrol within the Moscow City limits. Except for baby food, lead levels were higher in the Russian canned foods (range 6 to 1240 g kg–1, dry weight) compared to corresponding US canned foods, with ratios of Russian to US levels of up to 120:1 for evaporated milk. Lead concentrations in soil generally ranged from 500 to 2000 g g–1, levels that would trigger hazard reduction measures according to US EPA guidance. These findings, together with the use of lead in petrol outside Moscow, indicate multiple sources of lead exposure in Russia. Priorities for future research are discussed including the establishment of interlaboratory quality control programmes.  相似文献

Data collected over the past four years indicate that filets of suckers and some species of sunfish taken from certain sites in Missouri's Old Lead Belt consistently contain elevated lead concentrations which exceed the limit of 0.3 g/g suggested by the World Health Organization. The problem exists in bottom-feeding species taken from some sites along the Big River affected by erosion from abandoned lead tailings piles. Fish from many other sites within the Old Lead Belt and the New Lead Belt, including current wastewater treatment systems, are within the acceptable limits for lead in food. Public health implications are discussed relative to observed lead concentrations and available data regarding lead toxicity to humans.  相似文献

A long term programme to assess the impact of reduced lead emissions in the UK has been established in central London and rural Suffolk. Blood lead concentrations of 5 and 6 year old children and their mothers both show an urban-rural difference of about 1.5 g/dl, which is related to the urban-rural differences in air and dust lead concentrations between areas.This paper is also being published in Proceedings of the Conference on Lead in the Home Environment and is one of several selected from SEGH sponsored conferences for simultaneous publication in this journal.  相似文献

The variability of household dust lead concentration and loadings over the period of about a year has been examined in 10 homes. The overall uncertainty on a single sample is ± 65% for lead concentration and ± 81% for lead loading. Redecoration involving electric sanding and/or blow lamp preparation of painted surfaces is the major cause of variation in lead levels. The effect is short lived, levels returning to normal within 2 months of redecoration ending. There is evidence of a seasonal trend for dust loading and lead loading but not for lead concentration. Data from the Edinburgh Lead Study are used to estimate the respective contributions of water and dust lead to blood lead in 6–9 year old children. Both sources are significantly related to blood lead. An exposure of 100 g/l in kitchen cold water is estimated to be equivalent to 2700 g/g of lead in dust. After treatment with 1.0 and 3.3 g Pb kg-1 for 20 weeks in flowing seawater at ambient salinity and temperature, Crassostrea virginica accumulated as much as 6.57 and 11.42 g g-1 dry weight, respectively, and no mortalities were recorded. Lead uptake was curvilinear; however, the general trend was an increase in lead concentration over time. Dry weight of the oyster had no significant relation with tissue lead concentration (g g-1); however, a significant positive relationship existed between weight and lead content (g). A decrease in lead concentration in the tissues occurred from mid-August to mid-September despite continuous addition of lead to the seawater. Addition of lead to the seawater was terminated after 20 weeks (October), and lead loss was studied for the following 12 weeks. Lead loss was studied well after spawning in an attempt to eliminate any influence this might have on lead concentration in the tissues. After 4 weeks, approximately 54% of the accumulated lead was lost under conditions of a natural temperature decline. The biological half-life of lead in C. virginica tissues was calculated to be 5.5 weeks. In the ensuing 8 weeks, little or no lead loss was observed. Larvae from each treatment developed to the straight-hinged stage within 48 h with no apparent abnormalities. Lead appeared to have no adverse effect on larvae when parents had been treated with 1.0 and 3.3 g kg-1 seawater for 10 weeks.  相似文献

The EPA lead model predicts mean blood lead levels and risk of elevated blood lead levels in children based on lead uptake from multiple sources. In the latest model versions, environmental data from individual homes within a community can be used to predict the overall blood lead distribution and percent risk of exceeding a specific blood lead level (i.e. 10 g dl–1). Recent criteria used by the EPA to evaluate this information include no more than 5% of houses with a greater than 5% lead risk, and a community weighted-average risk below 5%. Environmental (primarily soil) and blood lead data from a residential community near a smelter were used to illustrate recent uses of the model. Scheduled remediation in the community will remove soil for approximately 60% of the houses (i.e. those with lead levels > 1000 mg kg–1). After remediation, the model results indicate a relatively low community risk (0.5–1.9%), although the percentage of houses with lead risks above 5% ranged from 3 to as high as 13%, depending on the variation in blood lead and assuming the model's 7 g dl–1 increase in blood lead with each 1000 mg kg–1 increase in soil lead level. A comparison of the limited blood lead data with soil lead levels below 1000 mg kg–1, however, indicated no apparent relationship. Given these uncertainties, less invasive actions than additional soil removal (e.g. exposure intervention, monitoring conditions, and follow-up as necessary) may be appropriate under the new EPA guidance for lead in soil.  相似文献

We explored the potential relationships between male traits, reproductive success, testosterone levels in the breeding season, and humoral immunocompetence in male red-winged blackbirds (Agelaius phoeniceus). Males responded to a single injection of diphtheria-tetanus vaccine by producing antibodies to both antigens. The primary responses to each antigen were positively correlated with each other, but the relationship was clearly non-linear. No male responded strongly to diphtheria without also responding strongly to tetanus, but many males had strong responses to tetanus and weak responses to diphtheria. Response to tetanus was positively associated with hormone levels, whereas the relationship between testosterone and the response to diphtheria was weakly and nonsignificantly negative. We found no convincing relationships between immune responses and male morphological traits (epaulet size, body blackness, and body size), male success in male-male competition (territory size and tenure), or male reproductive success (number of mates, average clutch size, proportion of offspring sired, or proportion of young fledged). These results do not support testosterone acting as a constraint on immunity. Testosterone could be mediating a condition-dependent trade-off between sexual ornaments and the immune system, but the evidence for this was weak, as neither any male sexual trait nor any measure of performance was associated with immune responses. We could not eliminate the possibility that male sexual traits advertise other aspects of immunity that are not dependent upon condition or hormones.  相似文献

High-precision lead isotope ratios and lead concentrations have been compared statistically and graphically in women of child-bearing age (n = 77) from two smelter communities and one general urban community to evaluate the relative contributions to blood lead of tissue lead stores and lead from the contemporaneous environment (soil, floor dust, indoor airborne dust, water, food). Blood lead (PbB) contents were generally low (e.g. <10 g dL–1). Statistically significant isotopic differences in blood and environmental samples were observed between the three cities although isotopic differences in blood for individual subjects living in close proximity (200 m radius) was as large as the differences within a city. No single environmental measure dominated the biological isotope profile and in many cases the low levels of blood lead meant that their isotopic profiles could be easily perturbed by relatively small changes of environmental exposure. Apportioning of sources using lead isotopes is possibly not feasible, nor cost effective, when blood lead levels are <5 g dL–1. Interpretations based on statistical analyses of city-wide data do not give the same conclusions as when the houses are considered individually. Aggregating data from multiple subjects in a study such as this obscures potentially useful information. Most of the measures employed in this study, and many other similar studies, are markers of only short-to-medium integration of lead exposure. Serial sampling of blood and longer sampling times, especially for household variables, should provide more meaningful information.  相似文献

Two primary lead smelters and one secondary lead smelter have been active in the Socorro, New Mexico (USA) area in the last 110 years: the Billing smelter from 1883 to 1894, the Cuba Road smelter from 1881 to 1900, and Cal West from 1979 to 1984. Samples of plants and surface soil under each plant from all three sites were analysed for lead. The plants consisted of sparse grasses, cacti, creosote bush, snakeweed, mesquite and fourwing saltbush. Lead levels in the plants increased (2-440 g g–1) as the lead in the alkaline soils (25-10000 ng g–1) increased. However, the BAC (biological absorption coefficient), which is the ratio of lead content in the plant to the lead content in the soil, a measure of relative accumulation, decreased by one to two orders of magnitude, except for grasses and snakeweed. At background lead levels, there was little difference between lead in rootsversus foliage. At high lead levels, there was higher lead in roots versus foliage at the Billing and Cuba Road sites. The reverse was noted at Cal West. Because this is a recent operation, the higher lead in foliage may be due to foliar uptake. Plant growth at all sites appeared healthy.  相似文献

The final clean-up of residential lead abatement projects in federally-supported housing, as well as in other housing in a number of states, must meet surface dust lead clearance levels expressed as g of lead per square foot. These clearance levels were established because hand-to-mouth ingestion of lead-contaminated dust is recognised as a major pathway through which many children are exposed. A dilemma exists because many floors in housing undergoing abatement are carpeted and the established clearance levels are generally not recommended for use on carpets. These clearance levels are also used as 'action levels' to determine whether exposure reduction activities are needed. The US Environmental Protection Agency is currently in the process of issuing standards for hazardous levels of lead in interior dust and bare soil under Title X of the Housing and Community Development Act of 1992, The Residential Lead-Based Paint Hazard Reduction Act of 1992. An effort to develop a potential surface dust lead clearance level for carpets was made using an existing vacuum dust collection method that has previously been shown to be a reliable indicator of childhood lead exposure. This method was designed for use on carpeted and non-carpeted surfaces. Using data from the Cincinnati Soil Lead Abatement Demonstration Project, the suggested floor-dust lead level where an estimated 95% of the population of children would be expected to have blood lead values below the national goal of 10 g dL–1, was more than an order of magnitude lower than the current floor-dust lead clearance level of 1080 g m–2 (100 g ft–2). Further comparisons of blood lead and carpet lead levels in other parts of the country should be performed before a risk-based lead loading clearance level is established.  相似文献

Levels of Pb, Zn, Cu, Cr, Ni and Cd were determined in topsoil and vegetation in the vicinity of a factory manufacturing lead- batteries in Ibadan, Nigeria. The samples were collected along five transects in different directions, and varying distances up to 1000m from the factory. Soil lead levels were found to be elevated around the factory, with average levels of about 2000mgkg–1 close to the fence that declined gradually to about 50mgkg–1 some 750m away. Soil-lead level around a primary school located about 500m from the factory was as high as 1450mgkg–1. Lead levels were equally elevated in the vegetation, though average levels in vegetation were slightly lower than in the soil. Cadmium concentrations in soil and vegetation, though low, were more positively correlated with lead levels than any of the other metals are with lead.  相似文献

The background levels of lead in Jamaica in soils and sediments, estimated at 37 mg kg–1, are relatively high compared with world averages. Several areas have values in excess of this due to mineralisation and pollution. One such is the residential Hope Flats/Kintyre area in which levels of lead up to 2.5% are found in the soils and up to 8 g kg–1 in the water of the nearby Hope River. The blood lead levels of a sample of children were in the range 5.7–57 g dl–1. The high lead levels suggest a potential health risk, particularly for the children. This can be minimised by programmes which include community education, case management and abatement to reduce the lead exposure.  相似文献

In order to assess the intake of lead and cadmium by consumers of home grown vegetables in urban areas, replicated experimental plots of uniform size, comprising summer and winter crops, were established in 94 gardens and allotments in nine towns and cities in England.The geometric mean lead and cadmium concentrations for the soils (n = 94) were 217 g g–1 (ranging from 27 to 1,676 g g–1) and 0.53 g g–1 (<0.2–5.9 g g–1), respectively. Compared with agricultural soils, the garden and allotment soils contained elevated levels of lead but not cadmium.Lead concentrations in the vegetables ranged from <0.25 g g–1 to 16.7 g g–1 dry weight and cadmium concentrations ranged from <0.025 g g–1 to 10.4 g g–1 dry weight. Lead concentrations were higher than reported background levels, although <1% exceeded the statutory limit for saleable food in the UK (1 g g–1 fresh weight). Cadmium concentrations were generally similar to background levels.  相似文献

The quantity and particle size characteristics of lead in dust released during three different paint removal techniques was determined under controlled conditions and in situ in a dwelling. Air-lead and deposited dust-lead levels were highest after sanding but burning-off and hot-air removal methods also produced significant contamination. Large‐scale poisoning events are common to scavenging bird species that forage communally, many of which are in decline. To reduce the threat of poisoning and compensate for other persistent threats, management, including supplemental feeding, is ongoing for many reintroduced and endangered vulture populations. Through a longitudinal study of lead exposure in California condors (Gymnogyps californianus), we illustrate the conservation challenges inherent in reintroduction of an endangered species to the wild when pervasive threats have not been eliminated. We evaluated population‐wide patterns in blood lead levels from 1997 to 2011 and assessed a broad range of putative demographic, behavioral, and environmental risk factors for elevated lead exposure among reintroduced California condors in California (United States). We also assessed the effectiveness of lead ammunition regulations within the condor's range in California by comparing condor blood lead levels before and after implementation of the regulations. Lead exposure was a pervasive threat to California condors despite recent regulations limiting lead ammunition use. In addition, condor lead levels significantly increased as age and independence from intensive management increased, including increasing time spent away from managed release sites, and decreasing reliance on food provisions. Greater independence among an increasing number of reintroduced condors has therefore elevated the population's risk of lead exposure and limited the effectiveness of lead reduction efforts to date. Our findings highlight the challenges of restoring endangered vulture populations as they mature and become less reliant on management actions necessary to compensate for persistent threats. This study examines the recent soil Lead Abatement Strategy (LAS) in Boolaroo, New South Wales, Australia, that was designed to “achieve a reduction in human exposure to lead dust contamination in surface soils”. The abatement programme addressed legacy contamination of residential areas following closure of lead smelting operations in 2003 at the Pasminco Cockle Creek Smelter (PCCS). The principal objective of the LAS was to “cap and cover” lead-contaminated soils within the urban environment surrounding the PCCS. Soil lead concentrations of 2500–5000 mg/kg were scheduled for removal and replacement, while concentrations between 1500 and 2500 mg/kg were replaced only under limited circumstances. To date, there has been no industry, government or independent assessment of the clean-up programme that involved >2000 homes in the township of Boolaroo. Thus, by measuring post-abatement soil lead concentrations in Boolaroo, this study addresses this knowledge gap and evaluates the effectiveness of the LAS for reducing the potential for lead exposure. Soil lead concentrations above the Australian residential soil health investigation level value for residential soils (300 mg/kg) were identified at all but one of the residential properties examined (n = 19). Vacuum dust samples (n = 17) from the same homes had a mean lead concentration of 495 mg/kg (median 380 mg/kg). Bio-accessibility testing revealed that lead in household vacuum dust was readily accessible (% bio-accessible) (mean = 92 %, median = 90 %), demonstrating that the risk of exposure via this pathway remains. Assessment of a limited number of properties (n = 8) where pre-abatement soil lead levels were available for comparison showed they were not statistically different to post-abatement. Although the LAS did not include treatment of non-residential properties, sampling of community areas including public sports fields, playgrounds and schools (n = 32) was undertaken to determine the contamination legacy in these areas. Elevated mean soil lead concentrations were found across public lands: sports fields = 5130 mg/kg (median = 1275 mg/kg), playgrounds and schools = 812 mg/kg (median = 920 mg/kg) and open space = 778 mg/kg (median = 620 mg/kg). Overall, the study results show that the LAS programme that was dominated by a “cap and cover” approach to address widespread lead contamination was inadequate for mitigating current and future risk of lead exposures.  相似文献

To establish allowable daily intakes for humans from animal bioassay experiments, benchmark doses corresponding to low levels of risk have been proposed to replace the no-observed-adverse-effect level for non-cancer endpoints. When the experimental outcomes are quantal, each animal can be classified with or without the disease. The proportion of affected animals is observed as a function of dose and calculation of the benchmark dose is relatively simple. For quantitative responses, on the other hand, one method is to convert the continuous data to quantal data and proceed with benchmark dose estimation. Another method which has found more popularity (Crump, Risk Anal 15:79–89; 1995) is to fit an appropriate dose–response model to the continuous data, and directly estimate the risk and benchmark doses. The normal distribution has often been used in the past as a dose–response model. However, for non-symmetric data, the normal distribution can lead to erroneous results. Here, we propose the use of the class of beta-normal distribution and demonstrate its application in risk assessment for quantitative responses. The most important feature of this class of distributions is its generality, encompassing a wide range of distributional shapes including the normal distribution as a special case. The properties of the model are briefly discussed and risk estimates are derived based on the asymptotic properties of the maximum likelihood estimates. An example is used for illustration.
