Transgenic or genetically modified plants possess novel genes that impart beneficial characteristics such as herbicide resistance. One of the least understood areas in the environmental risk assessment of genetically modified crops is their impact on soil- and plant-associated microbial communities. The potential for interaction between transgenic plants and plant residues and the soil microbial community is not well understood. The recognition that these interactions could change microbial biodiversity and affect ecosystem functioning has initiated a limited number of studies in the area. At this time, studies have shown the possibility that transgenes can be transferred to native soil microorganisms through horizontal gene transfer, although there is not evidence of this occurring in the soil. Furthermore, novel proteins have been shown to be released from transgenic plants into the soil ecosystem, and their presence can influence the biodiversity of the microbial community by selectively stimulating the growth of organisms that can use them. Microbial diversity can be altered when associated with transgenic plants; however, these effects are both variable and transient. Soil- and plant-associated microbial communities are influenced not only by plant species and transgene insertion but also by environmental factors such as field site and sampling date. Minor alterations in the diversity of the microbial community could affect soil health and ecosystem functioning, and therefore, the impact that plant variety may have on the dynamics of the rhizosphere microbial populations and in turn plant growth and health and ecosystem sustainability, requires further study.  相似文献

Theories suggest that rapid microbial growth rates lead to quicker development of metal resistance. We tested these theories by adding hexavalent chromium [Cr(VI)] to soil, sowing Indian mustard (Brassica juncea), and comparing rhizosphere and bulk soil microbial community responses. Four weeks after the initial Cr(VI) application we measured Cr concentration, microbial biomass by fumigation extraction and soil extract ATP, tolerance to Cr and growth rates with tritiated thymidine incorporation, and performed community substrate use analysis with BIOLOG GN plates. Exchangeable Cr(VI) levels were very low, and therefore we assumed the Cr(VI) impact was transient. Microbial biomass was reduced by Cr(VI) addition. Microbial tolerance to Cr(VI) tended to be higher in the Cr-treated rhizosphere soil relative to the non-treated systems, while microorganisms in the Cr-treated bulk soil were less sensitive to Cr(VI) than microorganisms in the non-treated bulk soil. Microbial diversity as measured by population evenness increased with Cr(VI) addition based on a Gini coefficient derived from BIOLOG substrate use patterns. Principal component analysis revealed separation between Cr(VI) treatments, and between rhizosphere and bulk soil treatments. We hypothesize that because of Cr(VI) addition there was indirect selection for fast-growing organisms, alleviation of competition among microbial communities, and increase in Cr tolerance in the rhizosphere due to the faster turnover rates in that environment.

The aim of this paper was to assess the influence of tamarisk shrubs on soil fertility, salinity and nematode communities in various habitats located in an arid desert-oasis region in northwest China. Three habitats were studied: sand dune, riparian zone and saline meadow, where tamarisk shrubs have been established in recent decades in order to vegetation restoration used as desertification control and saline land rehabilitation projects and become the dominant plant community. The parameters measured include soil organic carbon (SOC), total nitrogen, available phosphorus (P) and potassium (K), pH, salt component, and nematode community characteristics. Enrichment ratios (a comparison of the soil measurements between soils under canopy and in the open interspaces) for soil nutrients and salinity were used to evaluate fertility and salinity islands underneath the tamarisk shrubs. The soil nematode community was used as a biological indicator of soil condition. SOC and available P and K were higher beneath the plant canopy than in the open interspaces outside that canopy. The enrichment ratios for SOC and nutrients were highest for the sand dune habitat and tamarisk shrubs clearly created islands of greater salinity under the canopies. Nematode abundance per 100 g dry soil varied considerably between the locations and habitats, with the highest abundance found in sand dune and the lowest in saline meadow. A significantly higher nematode abundance and a lower trophic diversity were found in soils under the canopy compared to the soils in the open interspaces. With the exception of saline meadow, the abundance of bacterivores increased and fungivores decreased under the canopy relative to the open interspaces, and bacterivores dominated under the canopies in the sand dune and riparian habitats. The enrichment ratios for salinity were higher than for fertility, suggesting that improved soil fertility can not limit the impact of salinization beneath tamarisk shrubs. The adverse effect of salt accumulation on the soil environment should be taken into account when using tamarisk as restoration plant species, especially in saline meadow and controlling of tamarisk density should be considered when undertaking re-vegetation projects in the arid desert oasis regions.  相似文献

A sound multi-species vegetation buffer design should incorporate the species that facilitate rapid degradation and sequestration of deposited herbicides in the buffer. A field lysimeter study with six different ground covers (bare ground, orchardgrass, tall fescue, timothy, smooth bromegrass, and switchgrass) was established to assess the bioremediation capacity of five forage species to enhance atrazine (ATR) dissipation in the environment via plant uptake and degradation and detoxification in the rhizosphere. Results suggested that the majority of the applied ATR remained in the soil and only a relatively small fraction of herbicide leached to leachates (<15%) or was taken up by plants (<4%). Biological degradation or chemical hydroxylation of soil ATR was enhanced by 20 to 45% in forage treatment compared with the control. Of the ATR residues remaining in soil, switchgrass degraded more than 80% to less toxic metabolites, with 47% of these residues converted to the less mobile hydroxylated metabolites 25 d after application. The strong correlation between the degradation of N-dealkylated ATR metabolites and the increased microbial biomass carbon in forage treatments suggested that enhanced biological degradation in the rhizosphere was facilitated by the forages. Hydroxylated ATR degradation products were the predominant ATR metabolites in the tissues of switchgrass and tall fescue. In contrast, the N-dealkylated metabolites were the major degradation products found in the other cool-season species. The difference in metabolite patterns between the warm- and cool-season species demonstrated their contrasting detoxification mechanisms, which also related to their tolerance to ATR exposure. Based on this study, switchgrass is recommended for use in riparian buffers designed to reduce ATR toxicity and mobility due to its high tolerance and strong degradation capacity.  相似文献

White lupine (Lupinus albus L.) was used as a phosphorus (P)-efficient model plant to study the effects of elevated atmospheric CO(2) concentrations on (i) P acquisition, (ii) the related alterations in root development and rhizosphere chemistry, and (iii) the functional and structural diversity of rhizosphere microbial communities, on a P-deficient calcareous subsoil with and without soluble P fertilization. In both +P (80 mg P kg(-1)) and -P treatments (no added P), elevated CO(2) (800 micromol mol(-1)) increased shoot biomass production by 20 to 35% and accelerated the development of cluster roots, which exhibit important functions in chemical mobilization of sparingly soluble soil P sources. Accordingly, cluster root formation was stimulated in plants without P application by 140 and 60% for ambient and elevated CO(2) treatments, respectively. Intense accumulation of citrate and increased activities of acid and alkaline phosphatases, but also of chitinase, in the rhizosphere were mainly confined to later stages of cluster root development in -P treatments. Regardless of atmospheric CO(2) concentrations, there was no significant effect on accumulation of citrate or on selected enzyme activities of C, N, and P cycles in the rhizosphere of individual root clusters. Discriminant analysis of selected enzyme activities revealed that mainly phosphatase and chitinase contributed to the experimental variance (81.3%) of the data. Phosphatase and chitinase activities in the rhizosphere might be dominated by the secretion from cluster roots rather than by microbial activity. Alterations in rhizosphere bacterial communities analyzed by denaturing gradient gel electrophoresis (DGGE) were related with the intense changes in root secretory activity observed during cluster root development but not with elevated CO(2) concentrations.  相似文献

Stimulated rhizodegradation of atrazine by selected plant species   总被引:1,自引:0,他引:1
The efficacy of vegetative buffer strips (VBS) in removing herbicides deposited from surface runoff is related to the ability of plant species to promote rapid herbicide degradation. A growth chamber study was conducted to compare C-atrazine (ATR) degradation profiles in soil rhizospheres from different forage grasses and correlate ATR degradation rates and profiles with microbial activity using three soil enzymes. The plant treatments included: (i) orchardgrass ( L.), (ii) smooth bromegrass ( Leyss.), (iii) tall fescue ( Schreb.), (iv) Illinois bundle flower (), (v) perennial ryegrass ( L.), (vi) switchgrass ( L.), and (vii) eastern gamagrass (). Soil without plants was used as the control. The results suggested that all plant species significantly enhanced ATR degradation by 84 to 260% compared with the control, but eastern gamagrass showed the highest capability for promoting biodegradation of ATR in the rhizosphere. More than 90% of ATR was degraded in the eastern gamagrass rhizosphere compared with 24% in the control. Dealkylation of atrazine strongly correlated with increased enzymatic activities of β-glucosidase (GLU) ( = 0.96), dehydrogenase (DHG) ( = 0.842), and fluorescein diacetate (FDA) hydrolysis ( = 0.702). The incorporation of forage species, particularly eastern gamagrass, into VBS designs will significantly promote the degradation of ATR transported into the VBS by surface runoff. Microbial parameters widely used for assessment of soil quality, e.g., DHG and GLU activities, are promising tools for evaluating the overall degradation potential of various vegetative buffer designs for ATR remediation.  相似文献

The present study was conducted in 47 different riparian areas distributed throughout Denmark to investigate diversity and distributional patterns of plant communities along a lowland stream size gradient (first to fifth order). The investigated areas were representative for Danish riparian areas not in use for agricultural production. We investigated plant community richness along a stream size gradient and the influence of eutrophication on the abundance of different plant communities. Vegetation analyses were performed in transects placed perpendicular to the stream channel, with a total of 1798 plots analyzed. Overall, we found a positive relationship between stream mean depth as a measure of stream size and the number of plant community types identified in the riparian areas. We also found that the abundance of the identified communities was positively correlated with their nutrient preference and negatively correlated with their moisture preference. The abundance of alkaline fens and Molinia meadows (protected community types) in riparian areas decreased with increasing size of the stream, whereas the abundance of humid meadows and wet herb fringes increased with increasing size of the stream. Based on our findings, we recommend that wide buffer zones be established along streams with protected habitat types in the associated riparian areas to reduce the direct impact from agriculture. Furthermore, we recommend that wide buffer zones be established along middle-sized and large streams because several community types may develop.  相似文献

A 12-mo greenhouse study was conducted to evaluate the contribution of root death and decay on the dissipation of polycyclic aromatic hydrocarbons (PAHs) in rhizosphere soil. The contaminated soil was previously treated by land-farming, but residual PAHs remained after treatment. Tall fescue (Festuca arundinacea Schreb.) and yellow sweet clover (Melilotus officinalis Lam.) were the target plants. To specifically evaluate the effect of root decay on contaminant dissipation, plants were treated with glyphosate, a broad spectrum herbicide, to induce root decay. Although tall fescue treatments had the highest root and shoot biomass and root surface area, this plant did not result in the highest contaminant degradation rates. Significant differences were noted between treatments for seven PAHs, with the active yellow sweet clover resulting in 60 to 75% degradation of these compounds. Induced root death and decay did not produce a significant enhancement of PAH degradation. The PAH microbial degrader populations in the vegetated treatments were more than 100 times greater than those in the unvegetated control. The phospholipid fatty acid (PLFA) structural group profile shifted over the growing period, indicating a change in the community structure. In conclusion, phytoremediation was shown to be an effective polishing tool for PAH-affected soil previously subjected to biological treatment.  相似文献

Contamination of soil by hazardous substances poses a significant threat to human, environmental, and ecological health. Cleanup of the contaminants using destructive, invasive technologies has proven to be expensive and more importantly, often damaging to the natural resource properties of the soil, sediment, or aquifer. Phytoremediation is defined as the cleanup of contaminated sites using plants. There has been evidence of enhanced polycyclic aromatic hydrocarbons (PAHs) degradation in rhizosphere soils for a limited number of plants. However, research focusing on the degradation of PAHs in the rhizosphere of trees is lacking. The objective of this study was to assess the potential use of trees to enhance degradation of PAHs located in manufactured gas plant-impacted soils. In greenhouse studies with intact soil cores, acenaphthene, anthracene, fluoranthene, naphthalene, and phenanthrene decreased significantly (p < 0.05) in green ash (Fraxinus pennsylvanica Marshall) and hybrid poplar (Populus deltoides x P. nigra DN 34) phytoremediation treatments when compared to the unplanted soil control. Increases in PAH microbial degraders in rhizosphere soil were observed when compared to unvegetated soil controls. In addition, the rate of degradation or biotransformation of PAHs was greatest for soils with black willow (Salix nigra Marshall), followed by poplar, ash, and the unvegetated controls. These results support the hypothesis that a variety of plants can enhance the degradation of target PAHs in soil.  相似文献

The effects of maize (Zea mays L.), genetically modified to express the Cry1Ab protein (Bt), and an insecticide on soil microbial and faunal communities were assessed in a glasshouse experiment. Soil for the experiment was taken from field sites where the same maize cultivars were grown to allow comparison between results under glasshouse conditions with those from field trials. Plants were grown in contrasting sandy loam and clay loam soils, half were sprayed with a pyrethroid insecticide (deltamethrin) and soil samples taken at the five-leaf stage, flowering, and maturity. The main effect on all measured parameters was that of soil type and there were no effects of Bt trait or insecticide on plant growth. The Bt trait resulted in more soil nematodes and protozoa (amoebae), whereas insecticide application increased plant Bt concentration and altered nematode community structure. The only significant effects on soil microbial community structure, microarthropods, and larvae of a nontarget root-feeding Dipteran, were due to soil type and plant growth stage. The results indicate that, although there were statistically significant effects of the Bt trait on soil populations, they were small. The relative magnitude of the effect could best be judged by comparison with the insecticide treatment, which was representative of current best practice. The Bt trait had no greater effect than the insecticide treatment. Results from this glasshouse experiment were in broad agreement with conclusions from field experiments using the same plant material grown in the same soils.  相似文献

The levels of zinc accumulated by roots, stems, and leaves of two plant species, Rubus ulmifolius and Phragmites australis, indigenous to the banks of a stream in a Portuguese contaminated site were investigated in field conditions. R. ulmifolius, a plant for which studies on phytoremediation potential are scarce, dominated on the right side of the stream, while P. australis proliferated on the other bank. Heterogeneous Zn concentrations were found along the banks of the stream. Zn accumulation in both species occurred mainly in the roots, with poor translocation to the aboveground sections. R. ulmifolius presented Zn levels in the roots ranging from 142 to 563 mg kg(-1), in the stems from 35 to 110 mg kg(-1), and in the leaves from 45 to 91 mg kg(-1), vs. average soil total Zn concentrations varying from 526 to 957 mg kg(-1). P. australis showed Zn concentrations in the roots from 39 to 130 mg kg(-1), in the stems from 31 to 63 mg kg(-1), and in the leaves from 37 to 83 mg kg(-1), for the lower average soil total Zn levels of 138 to 452 mg kg(-1) found on the banks where they proliferated. Positive correlations were found between the soil total, available and extractable Zn fractions, and metal accumulation in the roots and leaves of R. ulmifolius and in the roots and stems of P. australis. The use of R. ulmifolius and P. australis for phytoextraction purposes does not appear as an effective method of metal removing, but these native metal tolerant plant species may be used to reduce the effects of soil contamination, avoiding further Zn transfer to other environmental compartments.  相似文献

This study investigated the effects of organic and inorganic nutrients on the microbial degradation of the common soil contaminant pyrene. The material used in this investigation was collected from potted trees that had been growing for over a year in a soil artificially contaminated with polycyclic aromatic hydrocarbons. Soil was removed from the nonroot (bulk) and root (rhizosphere) zones of these pots and used in mineralization studies that tracked microbial degradation of 14C-pyrene. The factors influencing degradation in these zones were then tested by amendment with essential inorganic nutrients or with root-derived materials. As expected, pyrene mineralization was greater in soil removed from the rhizosphere than in bulk soil. The rate of mineralization in rhizosphere soil was inhibited by inorganic nutrient amendment, whereas nutrients stimulated mineralization in the bulk soil. Pyrene mineralization in bulk soil was also increased by the addition of root extracts intended to mimic exudation by living roots. However, amendment with excised fine roots that were allowed to decay over time in soil initially inhibited mineralization. With time, the rate of mineralization increased, eventually exceeding that of unamended bulk soil. Combined, the initial inhibition and subsequent stimulation produced a zero net impact of decaying fine roots on bulk soil mineralization. Our results, in conjunction with known temporal patterns of fine root dynamics in natural systems, support the idea that seasonal variations in nutrient and substrate availability may influence the long-term effect of plants on organic degradation in soil, possibly reducing or negating the beneficial effects of vegetation that are often observed in short-term studies.  相似文献

Transgenic Bt corn expressing the Cry3Bb insecticidal protein active against corn rootworm (CRW) (Diabrotica spp.; Coleoptera: Chrysomelidae) was released for commercial use in 2003 and is expected to be widely adopted. Yet, the direct and indirect risks to soil microorganisms of growing this CRW-resistant Bt corn versus applying insecticides to control the rootworm have not been assessed under field conditions. The effects of CRW Bt corn and the insecticide tefluthrin [2,3,5,6-tetrafluoro-4-methylbenzyl (Z)-(1RS)-cis-3-(2-chloro-3,3,3-trifluoroprop-1-enyl)-2,2-dimethylcyclopropanecarboxylate] on soil microbial biomass, activity (N mineralization potential, short-term nitrification rate, and soil respiration), and bacterial community structure as determined by terminal restriction fragment length polymorphism (T-RFLP) analysis were assessed over two seasons in a field experiment. Bt corn had no deleterious effects on microbial activity or bacterial community measures compared with the non-transgenic isoline. The T-RFLP analysis indicated that amplifiable bacterial species composition and relative abundance differed substantially between years, but did not differ between rhizosphere and bulk soils. The application of tefluthrin also had no effect on any microbial measure except decreased soil respiration observed in tefluthrin-treated plots compared with Bt and non-transgenic isoline (NoBt) plots in 2002. Our results indicate that the release of CRW Bt corn poses little threat to the ecology of the soil microbial community based on parameters measured in this study.  相似文献

Wetland ecosystems are profoundly affected by altered nutrient and sediment loads received from anthropogenic activity in their surrounding watersheds. Our objective was to compare a gradient of agricultural and urban land cover history during the period from 1949 to 1997, with plant and soil nutrient concentrations in, and sediment deposition to, riparian wetlands in a rapidly urbanizing landscape. We observed that recent agricultural land cover was associated with increases in Nitrogen (N) and Phosphorus (P) concentrations in a native wetland plant species. Conversely, recent urban land cover appeared to alter receiving wetland environmental conditions by increasing the relative availability of P versus N, as reflected in an invasive, but not a native, plant species. In addition, increases in surface soil Fe content suggests recent inputs of terrestrial sediments associated specifically with increasing urban land cover. The observed correlation between urban land cover and riparian wetland plant tissue and surface soil nutrient concentrations and sediment deposition, suggest that urbanization specifically enhances the suitability of riparian wetland habitats for the invasive species Japanese stiltgrass [Microstegium vimenium (Trinius) A. Camus].  相似文献

Root activity can modify the chemistry of the rhizosphere and alter phosphorus (P) availability and uptake. However, until recently, relatively little was known about the dynamics of soil solution P at the root surface because of our inability to measure in situ changes in solution P at the plant root. A mini-rhizotron experiment with corn (Zea mays L. cv. Stine 2250), soybean [Glycine max (L.) Merr. cv. Pioneer 3563), cottonwood (Populus deltoids L.), smooth brome (Bromus inermis Leyss.), and switchgrass (Panicum virgatum L.) was conducted to measure the spatial and temporal dynamics of P in the rhizosphere solution of a fine silty, P-rich calcareous soil (solid-phase total P concentration = 62 mg kg(-1), pH = 7.68) from western Iowa. Micro-suction cups were used to collect samples of soil solution from defined segments of the rhizosphere, and capillary electrophoresis (CE) was used to determine the P concentration of the soil solution. At the end of 10 d, a decreasing P concentration gradient in soil solution toward the root was observed in corn, cottonwood, and smooth brome. No clear rhizosphere effect was observed for soybean and switchgrass. Statistical analysis indicated significantly lower solution P concentrations in the rhizospheres of corn (p = 0.05), cottonwood (p = 0.01), and smooth brome (p = 0.01) compared with bulk soil solution. Results indicate that P depletion from rhizosphere soil solution depends on plant species. Under the conditions of this study, corn, cottonwood, and smooth brome were more effective in depleting solution P than soybean and switchgrass.  相似文献

Effects of Recreational Impacts on Soil Microbial Communities   总被引:51,自引:0,他引:51
/ The functional diversity of soil microbial communities in heavilyimpacted subalpine campsites and adjacent undisturbed areas was comparedusing the Biolog method of carbon utilization profiles. Principal componentsanalysis of patterns and level of microbial activity indicate that microbialcommunities differentiate in response to disturbance in the top 6 cm of soil, while below 6 cm there were no recognizable differences between disturbed andundisturbed soil communities. Analysis of the factors that differentiate theupper microbial communities between disturbed and undisturbed sites revealedthat the percent of total carbon sources utilized was significantly less inthe disturbed (54%) than in undisturbed areas (95%). Carbonsubstrates important in the discrimination between soil communities includeplant, invertebrate, and microbial derivatives that could not be metabolizedby microbial communities from disturbed sites. Comparisons of totalculturable actinomycetes, bacteria, and fungi reveal no difference in overallnumber of colony forming units (CFU) on disturbed and undisturbed sites, buta marked decrease in actinomycetes on disturbed sites. Biolog andspread-plate data combined indicate a shift in the structure and function ofthe microbial community in campsite soils, which may be a useful indicator ofsoil community disturbance.KEY WORDS: Microbial functional diversity; Anthropogenic disturbance;Recreational impacts; Carbon source profile; Subalpine  相似文献

Supplying freshwater is one of the important methods to help restore degraded wetlands. Changes in soil properties and plant community biomass were evaluated by comparing sites with freshwater treatment versus reference sites following freshwater addition to wetlands of the Yellow River Delta for 7 years. The results indicated that soil organic carbon (SOC) was significantly increased in all wetland sites that were treated with freshwater compared to the reference sites. The treatment wetlands had greater total nitrogen (TN), lower pH and electrical conductivity and higher water content in the soil compared to the reference wetlands. In general, the upper soil layer (0-20 cm) had greater SOC than the lower soil layer (20-40 cm). The increase of SOC in the freshwater reintroduction wetlands was higher in the Suaeda salsa plant community (mean ± standard error) (6.89 ± 0.63 g/kg) and Phragmites communis plant community (4.11 ± 0.12 g/kg) than in the Tamarix chinensis plant community (1.40 ± 0.31 g/kg) in the upper soil layer. The differences were especially marked between the treated and reference wetlands for SOC and TN in the P. communis plant communities. The C:N ratio of the soil was significantly greater in the treated compared to the reference wetlands for the S. salsa plant community. Although the C: N ratios increased after treatment, they were all <25 suggesting that N availability was not limiting soil organic matter decomposition. Our results indicate that freshwater addition and the concomitant increase in soil moisture content enhances the accumulation of SOC in the Yellow River Delta.  相似文献

Denitrification potential in urban riparian zones   总被引:3,自引:0,他引:3
Denitrification, the anaerobic microbial conversion of nitrate (NO3-) to nitrogen (N) gases, is an important process contributing to the ability of riparian zones to function as "sinks" for NO3- in watersheds. There has been little analysis of riparian zones in urban watersheds despite concerns about high NO3- concentrations in many urban streams. Vegetation and soils in urban ecosystems are often highly disturbed, and few studies have examined microbial processes like denitrification in these ecosystems. In this study, we measured denitrification potential and a suite of related microbial parameters (microbial biomass carbon [C] and N content, potential net N mineralization and nitrification, soil inorganic N pools) in four rural and four urban riparian zones in the Baltimore, MD metropolitan area. Two of the riparian zones were forested and two had herbaceous vegetation in each land use context. There were few differences between urban and rural and herbaceous and forest riparian zones, but variability was much higher in urban than rural sites. There were strong positive relationships between soil moisture and organic matter content and denitrification potential. Given the importance of surface runoff in urban watersheds, the high denitrification potential of the surface soils that we observed suggests that if surface runoff can be channeled through areas with high denitrification potential (e.g., stormwater detention basins with wetland vegetation), these areas could function as important NO3- sinks in urban watersheds.  相似文献
