Limited information is available on the environmental behavior and associated potential risk of manufactured oxide nanoparticles (NPs). In this research, toxicity of nanoparticulate and bulk ZnO, Al2O3 and TiO2 were examined to the nematode Caenorhabditis elegans with Escherichia coli as a food source. Parallel experiments with dissolved metal ions from NPs were also conducted. The 24-h median lethal concentration (LC50) and sublethal endpoints were assessed. Both NPs and their bulk counterparts were toxic, inhibiting growth and especially the reproductive capability of the nematode. The 24-h LC50 for ZnO NPs (2.3 mg L−1) and bulk ZnO was not significantly different, but significantly different between Al2O3 NPs (82 mg L−1) and bulk Al2O3 (153 mg L−1), and between TiO2 NPs (80 mg L−1) and bulk TiO2 (136 mg L−1). Oxide solubility influenced the toxicity of ZnO and Al2O3 NPs, but nanoparticle-dependent toxicity was indeed observed for the investigated NPs.  相似文献

Extensive production and consumption of nanomaterials such as ZnO and TiO2 has increased their release and disposal into the environment. The accumulation of nanoparticles (NPs) in ecosystem is likely to pose threat to non-specific targets such as bacteria. The present study explored the effect of ZnO and TiO2 NPs in a model bacterium, Salmonella typhimurium. The uptake of ZnO and TiO2 bare NPs in nano range without agglomeration was observed in S. typhimurium. TEM analysis demonstrated the internalization and uniform distribution of NPs inside the cells. Flow cytometry data also demonstrates that both ZnO and TiO2 NPs were significantly internalized in the S. typhimurium cells in a concentration dependent manner. A significant increase in uptake was observed in the S. typhimurium treated even with 8 and 80 ng mL−1 of ZnO and TiO2 NPs with S9 after 60 min, possibly the formation of micelles or protein coat facilitated entry of NPs. These NPs exhibited weak mutagenic potential in S. typhimurium strains TA98, TA1537 and Escherichia coli (WP2uvrA) of Ames test underscoring the possible carcinogenic potential similar to certain mutagenic chemicals. Our study reiterates the need for re-evaluating environmental toxicity of ZnO and TiO2 NPs presumably considered safe in environment.  相似文献

Toxicity of nano-scaled aluminum, silicon, titanium and zinc oxides to bacteria (Bacillus subtilis, Escherichia coli and Pseudomonas fluorescens) was examined and compared to that of their respective bulk (micro-scaled) counterparts. All nanoparticles but titanium oxide showed higher toxicity (at 20 mg/L) than their bulk counterparts. Toxicity of released metal ions was differentiated from that of the oxide particles. ZnO was the most toxic among the three nanoparticles, causing 100% mortality to the three tested bacteria. Al2O3 nanoparticles had a mortality rate of 57% to B. subtilis, 36% to E. coli, and 70% to P. fuorescens. SiO2 nanoparticles killed 40% of B. subtilis, 58% of E. coli, and 70% of P. fluorescens. TEM images showed attachment of nanoparticles to the bacteria, suggesting that the toxicity was affected by bacterial attachment. Bacterial responses to nanoparticles were different from their bulk counterparts; hence nanoparticle toxicity mechanisms need to be studied thoroughly.  相似文献

Cell lines of Etroplus suratensis established in our laboratory were evaluated for their potential use as screening tools for the ecotoxicological assessment of tannery effluent. The cytotoxic effect of tannery effluent in three cell lines derived from eye, kidney and gill tissue of E. suratensis was assessed using multiple endpoints such as Neutral Red (NR) assay, Coomassie Blue (CB) protein assay and Alamar Blue (AB) assay. Acute toxicity tests on fish were conducted by exposing E. suratensis for 96 h to tannery effluent under static conditions. The toxic effect of tannery effluent on the survival of fish was found to be concentration and time dependent. The tannery effluent at the concentration of 15% caused 100% mortality at 96 h whereas the lower concentration (0.5%) caused 13.33% mortality. The cytotoxicity of tannery effluent was found to be similar in the three cell lines tested, independent of the toxic endpoints employed. EC50 values, the effective concentration of tannery effluent resulting in 50% inhibition of cytotoxicity parameters after 48 h exposure to tannery effluent were calculated for eye, kidney and gill cell lines using NR uptake, AB and cell protein assays. Statistical analysis revealed good correlation with r2 = 0.95-0.99 for all combinations between endpoints employed. Linear correlations between each in vitro EC50 and the in vivo LC50 data, were highly significant p < 0.001 with r2 = 0.977, 0.968 and 0.906 for AB50, NR50, and CB50, respectively.  相似文献

Anti-estrogenic activity in wastewater is gaining increased attention because of its endocrine-disrupting function. In this study, the level and removal efficiency by coagulation of anti-estrogenic activity in secondary effluents of domestic wastewater treatment plants were studied. Anti-estrogenic activity was detected in secondary effluent samples at a tamoxifen (TAM) equivalent concentration level of 0.38–0.94 mg-TAM L−1. Dissolved organic matters (DOM) with the molecular weight (MW) less than 3000 Da in hydrophobic acids (HOA) and hydrophobic neutrals (HON) fractions of the secondary effluent were the key fractions related to anti-estrogenic activity. Coagulation with FeCl3 and polyaluminium chloride (PAC) can remove the anti-estrogenic activity of the secondary effluents, but the removal efficiency was limited. The removal efficiency using FeCl3 coagulant was higher than that induced by PAC. Dissolved organic carbon was continuously removed with increased coagulant dose (0–120 mg L−1 FeCl3 or 0–60 mg L−1 PAC). However, the removal of anti-estrogenic activity was not enhanced further when the coagulant concentration was beyond a critical value (30 mg L−1 FeCl3 or 10 mg L−1 PAC). The highest removal of anti-estrogenic activity was about 36% by FeCl3 and 20% by PAC. Size exclusion chromatography results indicated difficulty in removing DOM with MW less than 3000 Da in the secondary effluent during coagulation even at a high coagulant concentration, which led to low removal efficiency of anti-estrogenic activity.  相似文献

The efficiency of ZnO and TiO2 suspensions in the photocatalytic degradation of two fungicides (cyprodinil and fludioxonil) in leaching water was investigated. The experiments were carried out at pilot plant scale using compound parabolic collectors under natural sunlight. The blank experiments for both irradiated compounds solutions showed that both oxides strongly enhanced the removal of the fungicides. The addition of an oxidant (Na2S2O8) to the ZnO or TiO2 increased the rate of photooxidation. The degradation of cyprodinil and fludioxonil followed first order kinetics according to the Langmuir-Hinshelwood model. Complete degradation of both fungicides was achieved within 4 h (t30W = 18 min) when treated with illuminated ZnO. The disappearance time (DT75), when referred to the normalized illumination time (t30W), was lower than 40 and 550 min (t30W = 2 and 40 min) for both fungicides using ZnO or TiO2, respectively. ZnO appeared to be more effective in cyprodinil and fludioxonil oxidation than TiO2 probably due to its nonstoichiometry.  相似文献

A field survey and greenhouse experiments were conducted using Physalis alkekengi L. to investigate strategies of phytoremediation. In addition, ZnO nanoparticles were synthesized using P. alkekengi. P. alkekengi plants grew healthily at Zn levels from 50 to 5000 mg kg−1 in soils. The plants incorporated Zn into their aerial parts (with mean dry weight values of 235-10,980 mg kg−1) and accumulated biomass (with a mean dry weight of 25.7 g plant−1) during 12 weeks. The synthesized ZnO nanoparticles showed a polydisperse behavior and had a mean size of 72.5 nm. The results indicate that P. alkekengi could be used for the remediation of zinc-contaminated soils. Moreover, the synthetic method of synthesizing ZnO nanoparticles from Zn hyperaccumulator plants constitutes a new insight into the recycling of metals in plant sources.  相似文献

Nano-TiO2 enhances the toxicity of copper in natural water to Daphnia magna   总被引:1,自引:0,他引:1  
The acute toxicity of engineered nanoparticles (NPs) in aquatic environments at high concentrations has been well-established. This study demonstrates that, at a concentration generally considered to be safe in the environment, nano-TiO2 remarkably enhanced the toxicity of copper to Daphnia magna by increasing the copper bioaccumulation. Specifically, at 2 mg L−1 nano-TiO2, the (LC50) of Cu2+ concentration observed to kill half the population, decreased from 111 μg L−1 to 42 μg L−1. Correspondingly, the level of metallothionein decreased from 135 μg g−1 wet weight to 99 μg g−1 wet weight at a Cu2+ level of 100 μg L−1. The copper was found to be adsorbed onto the nano-TiO2, and ingested and accumulated in the animals, thereby causing toxic injury. The nano-TiO2 may compete for free copper ions with sulfhydryl groups, causing the inhibition of the detoxification by metallothioneins.  相似文献

The Ni-doped and N-doped TiO2 nanoparticles were investigated for their antibacterial activities on Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) bacteria. Their morphological features and characteristics such as particle size, surface area, and visible light absorbing capacity were compared and discussed. Scanning electron microscopy, X-ray diffraction, and UV–visible spectrophotometry were used to characterize both materials. The inactivation of E. coli (as an example of Gram-negative bacteria) and S. aureus (as an example of Gram-positive bacteria) with Ni-doped and N-doped TiO2 was investigated in the absence and presence of visible light. Antibacterial activity tests were conducted using undoped, Ni-doped, and N-doped TiO2. The N-doped TiO2 nanoparticles show higher antibacterial activity than Ni-doped TiO2. The band gap narrowing of N-doped TiO2 can induce more visible light absorption and leads to the superb antibacterial properties of this material. The complete inactivation time for E. coli at an initial cell concentration of 2.7?×?104 CFU/mL was 420 min which is longer than the 360 min required for S. aureus inactivation. The rate of inactivation of S. aureus using the doped TiO2 nanoparticles in the presence of visible light is greater than that of E. coli. The median lethal dose (LD50) values of S. aureus and E. coli by antibacterial activity under an 18-W visible light intensity were 80 and 350 mg/ml for N-doped TiO2, respectively.  相似文献

In the present study, the photocatalytic degradation of five sulfonylurea herbicides (chlorsulfuron, flazasulfuron, nicosulfuron, sulfosulfuron and triasulfuron) has been investigated in aqueous suspensions of zinc oxide (ZnO), tungsten (VI) oxide (WO3), tin (IV) oxide (SnO2) and zinc sulfide (ZnS) at pilot plant scale under natural sunlight. Photocatalytic experiments, especially those involving ZnO photocatalysis, showed that the addition of semiconductors in tandem with the oxidant (Na2S2O8) strongly enhances the degradation rate of the herbicides in comparisons carried out with photolytic tests. The degradation of the herbicides follows a first order kinetics according to the Langmuir-Hinshelwood model. In our conditions, the amount of time required for 50% of the initial pesticide concentration to dissipate (t½) ranged from 8 to 27 min (t30W = 0.3-1.2 min) for sulfosulfuron and chlorsulfuron, respectively in the ZnO/Na2S2O8 system. None of the studied herbicides was found after 120 min of illumination (except chlorsulfuron, 0.2 μg L−1).  相似文献

Metal oxide nanomaterials have exhibited toxicity to a variety of aquatic organisms, especially microbes and invertebrates. To date, few studies have evaluated the toxicity of metal oxide nanomaterials on aquatic vertebrates. Therefore, this study examined effects of ZnO, TiO2, Fe2O3, and CuO nanomaterials (20-100 nm) on amphibians utilizing the Frog Embryo Teratogenesis Assay Xenopus (FETAX) protocol, a 96 h exposure with daily solution exchanges. Nanomaterials were dispersed in reconstituted moderately hard test medium. These exposures did not increase mortality in static renewal exposures containing up to 1000 mg L−1 for TiO2, Fe2O3, CuO, and ZnO, but did induce developmental abnormalities. Gastrointestinal, spinal, and other abnormalities were observed in CuO and ZnO nanomaterial exposures at concentrations as low as 3.16 mg L−1 (ZnO). An EC50 of 10.3 mg L−1 ZnO was observed for total malformations. The minimum concentration to inhibit growth of tadpoles exposed to CuO or ZnO nanomaterials was 10 mg L−1. The results indicate that select nanomaterials can negatively affect amphibians during development. Evaluation of nanomaterial exposure on vertebrate organisms are imperative to responsible production and introduction of nanomaterials in everyday products to ensure human and environmental safety.  相似文献

We examined the effect of ozone (O3) on Norway spruce (Picea abies) needle epicuticular wax over three seasons at the Kranzberg Ozone Fumigation Experiment. Exposure to 2× ambient O3 ranged from 64.5 to 74.2 μl O3 l−1 h AOT40, and 117.1 to 123.2 nl O3 l−1 4th highest daily maximum 8-h average O3 concentration. The proportion of current-year needle surface covered by wax tubes, tube aggregates, and plates decreased (P = 0.011) under 2× O3. Epistomatal chambers had increased deposits of amorphous wax. Proportion of secondary alcohols varied due to year (P = 0.004) and O3 treatment (P = 0.029). Secondary alcohols were reduced by 9.1% under 2× O3. Exposure to 2× O3 increased (P = 0.037) proportions of fatty acids by 29%. Opposing trends in secondary alcohols and fatty acids indicate a direct action of O3 on wax biosynthesis. These results demonstrate O3-induced changes in biologically important needle surface characteristics of 50-year-old field-grown trees.  相似文献

This research was conducted in the middle Duratón River (Central Spain), in the vicinity of Burgomillodo Reservoir. An industrial effluent enters the river 300 m downstream from the dam. Fluoride and turbidity levels significantly increased downstream from the effluent, these levels being to some extent affected by differential water releases from the dam. The community of submersed macrophytes exhibited slighter responses and, accordingly, lower discriminatory power than the community of benthic macroinvertebrates, this indicating that metrics and indices based on macroinvertebrates may be more suitable for the biological monitoring of water pollution and habitat degradation in dammed rivers receiving industrial effluents. However, in relation to fluoride bioaccumulation at the organism level, macrophytes (Fontinalis antipyretica and Potamogeton pectinatus) were as suitable bioindicators of fluoride pollution as macroinvertebrates (Ancylus fluviatilis and Pacifastacus leniusculus). Fluoride bioaccumulation in both hard and soft tissues of these aquatic organisms could be used as suitable bioindicator of fluoride pollution (even lower than 1 mg F L−1) in freshwater ecosystems. Echinogammarus calvus exhibited a great sensitivity to the toxicity of fluoride ions, with a 96 h LC50 of 7.5 mg F L−1 and an estimated safe concentration of 0.56 mg F L−1. The great capacity of E. calvus to take up and retain fluoride during exposures to fluoride ions would be a major cause of its great sensitivity to fluoride toxicity. It is concluded that the observed fluoride pollution might be partly responsible for the absence of this native amphipod downstream from the industrial effluent.  相似文献

Increasing antibacterial resistance and pathogenicity in the environment is of growing concern due to its potential human risk. In the present study, 236 Escherichia coli isolates were collected from Wenyu River in China on drugless (48 isolates) and quinolone-containing plates (189 isolates). Their minimum inhibitory concentrations (MICs) were determined ranging from 0.125 μg mL−1 to 128 μg mL−1. Mutation points related to fluoroquinolone resistance were observed at S83 to L and D87 to N or Y in the GyrA subunit and S80 to R or I and E84 to G in the ParC subunit. Generally, MICs of LEV and GAT are dependent on the patterns of these mutation points. The profile with three mutation points was related to LEV-resistant E. coli isolates, and the (S83L, D87N + S80I) mutation profile was most prevalent (65.7%) in LEV-resistant isolates, while a large proportion of isolates, even those with three mutation points, were susceptive to GAT. The incidence of virulence factors in LEV-resistant isolates (44.7%, 59/132) was much higher than in nonresistant isolates (23.1%, 24/104) (χ2 = 11.925, 1° of freedom, p < 0.001) indicating that fluoroquinolone-resistant E. coli would pose a potential risk. A similar distribution was also found in isolates resistant to GAT (χ2 = 7.843, 1° of freedom, = 0.0079).  相似文献

We characterized fluorescent silica nanoparticles (FNPs), which had been applied in many biological systems, in fish embryo rearing media (ERM) solution and evaluated the potential toxicity to the early development of Oryzias latipes embryos. Distribution of FNPs in embryos and larvae of O. latipes was studied by fluorescent and confocal laser scanning microscopic studies. Embryos exposed to three different concentrations of FNPs in stirred or sonicated ERM solutions were observed up to 2 d after hatching. FNPs had a negligible effect on the hatchability of O. latipes embryos; however, compared to controls, more than 30% of eggs were abnormal in 10 and 50 mg FNP L−1 solutions. We found that the toxic effect was increased in sonicated FNP solution, which seems to be related with the dissolution of FNPs in ERM solutions that could be accelerated by sonication. Further study found that the CaCl2 included in ERM solution might enhance the dissolution of the FNPs and the silicate ion released from FNPs partially contributed to larval toxicity. This study showed that some nanoparticles may not be stable in biological fluids even if they are stable in water. Dissolution factors such as sonication and cellular components should be considered in biological application of nanoparticles.  相似文献

Hyperaccumulators contain tubular cellulose and heavy metals, which can be used as the sources of carbon and metals to synthesize nanomaterials. In this paper, carbon nanotubes (CNTs), Cu0.05Zn0.95O nanoparticles, and CNTs/Cu0.05Zn0.95O nanocomposites were synthesized using Brassica juncea L. plants, and the ultraviolet (UV)-light-driven photocatalytic degradations of bisphenol A (BPA) using them as photocatalysts were studied. It was found that the outer diameter of CNTs was around 50 nm and there were a few defects in the crystal lattice. The synthesized Cu0.05Zn0.95O nanocomposites had a diameter of around 40 nm. Cu0.05Zn0.95O nanocomposites have grown on the surface of the CNTs and the outer diameter of them was around 100 nm. The synthesized hybrid carbon nanotubes using B. juncea could enhance the efficiency of photocatalytic degradation on BPA. The complete equilibration time of adsorption/desorption of BPA onto the surface of CNTs, Cu0.05Zn0.95O nanoparticles, and CNTs/Cu0.05Zn0.95O nanocomposites was within 30, 20, and 30 min, and approximately 14.9, 8.7, and 17.4 % BPA was adsorbed by them, respectively. The combination of UV light irradiation (90 min) with CNTs, Cu0.05Zn0.95O nanoparticles, and CNTs/Cu0.05Zn0.95O nanocomposites could lead to 48.3, 75.7, and 92.6 % decomposition yields of BPA, respectively. These findings constitute a new insight for synthesizing nanocatalyst by reusing hyperaccumulators.  相似文献

Solar disinfection (SODIS) of Escherichia coli suspensions in low-density polyethylene bag reactors was investigated as a low-cost disinfection method suitable for application in developing countries. The efficiency of a range of SODIS reactor configurations was examined (single skin (SS), double skin, black-backed single skin, silver-backed single skin (SBSS) and composite-backed single skin) using E. coli suspended in model and real surface water. Titanium dioxide was added to the reactors to improve the efficiency of the SODIS process. The effect of turbidity was also assessed. In addition to viable counts, E. coli injury was characterised through spread-plate analysis using selective and non-selective media. The optimal reactor configuration was determined to be the SBSS bag (t50 = 9.0 min) demonstrating the importance of UVA photons, as opposed to infrared in the SODIS disinfection mechanism. Complete inactivation (6.5-log) was achieved in the presence of turbidity (50 NTU) using the SBSS bag within 180 min simulated solar exposure. The addition of titanium dioxide (0.025 g L−1) significantly enhanced E. coli inactivation in the SS reactor, with 6-log inactivation observed within 90 min simulated solar exposure. During the early stages of both SODIS and photocatalytic disinfection, injured E. coli were detected; however, irreversible injury was caused and re-growth was not observed. Experiments under solar conditions were undertaken with total inactivation (6.5-log) observed in the SS reactor within 240 min, incomplete inactivation (4-log) was observed in SODIS bottles exposed to the same solar conditions.  相似文献

Fullerenes are set to be produced on an industrial scale in anticipation of their wide applications. This calls for research on their environmental and health impacts. This study investigates and compares the cell toxicity of different aqueous fullerene aggregates. Popular C60 dispersal methods were used to prepare four types of nC60 aggregates. These aggregates were tested against the indicator species Escherichia coli (E. coli) AMC 198. With aggregates of around 150 nm in diameter, the THF/nC60 suspension was very toxic and gave rise to a half maximal effective concentration (EC50) of 0.54 mg L−1 in E. coli. By contrast, the Tol/nC60 suspension exhibited a cytoprotective role while the Aqu-N2/nC60 and Aqu-O2/nC60 suspensions enhanced the metabolism of E. coli. Although some toxicants, such as THF and THF-peroxide, were introduced into the THF/nC60 suspension during the dispersion, the toxicity of nC60 itself cannot be neglected.  相似文献

Combination of divergent active principles to achieve broad-spectrum control is gaining popularity to manage the weed menace in intensive agriculture. However, such application could have non-target impacts on the soil processes affecting soil ecology and environmental interactions. A field experiment was conducted to investigate the impact of separate and combined applications of herbicides bensulfuron methyl and pretilachlor on the emission of N2O and CH4, and related soil and microbial parameters in a flooded alluvial field planted to rice cv Lalat. Single application of the herbicide bensulfuron methyl or pretilachlor resulted in a significant reduction of N2O and CH4 emissions while the combination of these two herbicides distinctly increased N2O and CH4 emissions. Cumulative N2O emissions (kg N2O-N) followed the order of bensulfuron methyl (0.35 kg ha−1) < pretilachlor (0.36 kg ha−1) < control (0.45 kg ha−1) < bensulfuron methyl 0.6% + pretilachlor 6.0% single dose (0.49 kg ha−1) < bensulfuron methyl 0.6% + pretilachlor 6.0% double dose (0.54 kg ha−1). Cumulative CH4 emissions (kg CH4), on the other hand, followed the order of bensulfuron methyl (47.89 kg ha−1) < pretilachlor (73.17 kg ha−1) < bensulfuron methyl 0.6% + pretilachlor 6.0% single dose (93.50 kg ha−1) < control (106.54 kg ha−1) < bensulfuron methyl 0.6% + pretilachlor 6.0% double dose (124.67 kg ha−1). The inhibitory effect of separate application of herbicides bensulfuron methyl 0.6% and pretilachlor 6.0% on N2O emission was linked to lower mineral N, lower denitrifying and nitrifying activity and low denitrifier and nitrifier populations. Inhibitory effect on CH4 emission, on the contrary, was linked to prevention in the drop of redox potential, lower readily mineralizable carbon (RMC) and microbial biomass carbon (MBC) contents as well as lower methanogenic and higher methanotrophic bacterial population. Admittedly, stimulatory effect of combined application of herbicides bensulfuron methyl 0.6% and pretilachlor 6.0% at double dose on N2O and CH4 emission was related to reversal of the identified indicators of inhibition. Results indicate that while individual application of herbicides bensulfuron methyl 0.6% or pretilachlor 6.0% can reduce N2O and CH4 emission from flooded soil planted to rice, their combined application at normal dose can keep the emission at a comparatively lower level with significantly higher grain yield as compared to the herbicides applied alone.  相似文献
